Composting of date palm (Phoenix dactylifera L.) residues contaminated with Fusarium f.sp oxysporum albedinis, causal agent of the vascular wilt (Bayoud) of the date palm, has been achieved. The effect of the aeration of the piles by manual turning has been studied. The maintenance of an adequate humidity of 60%-70%, necessary to the good progress of the composting process, required the contribution of 11.4 L of water/kg of the dried residues. The evolution of the temperatures in the three piles presents the same phases. A latency phase, followed after 2-3 d of composting by a thermophilic phase, which lasts about 24 d, where the temperature remains elevated between 50 and 70 degrees C. Then a cooling phase that takes about 15 d, during which the temperatures fall to values between 25 and 35 degrees C, near room temperature. Fusarium f.sp oxysporum albedinis is eliminated completely during the thermophilic phase of composting, and increasing frequencies of turning accelerate its disappearance to a certain extent. On the other hand, pH remained steady and relatively basic oscillating between 8.2 and 8.7. Ninety percent (90%) of the the date palm residues are composed exclusively of organic matters. The total nitrogen represents only 0.4%. The contribution of manure decreases the ratio of carbon to nitrogen (C/N) from 115 to 48 in the initial mixture. After 80 d of composting and according to the frequency of return up, there is a reduction of the granulometry of the substratum, the C/N ratio (from 29% to 44%), the organic matter (from 15% to 23%), the total volume (from 25% to 35%), and of the dry weight of the swaths (from 16% to 24%). On the other hand there is an increase in total nitrogen rate (from 20% to 40%) and in the mineral matter (from 23% to 35%).  相似文献

The inoculation of the roots of resistant (BSTN) and susceptible (JHL) cultivars of date palm seedlings byFusarium oxysporum f. sp.albedinis (Foa) induces an increase in activity of phenylalanine ammonia-lyase (E.C. 4. 3. 1. 5., PAL). The post-infectional response in the PAL activity in the resistant cultivar roots was faster and higher than that in the susceptible cultivar. However, the elicitation of the seedlings by the hyphal wall preparation (HWP) ofFoa induces an identical PAL response in the resistant and the susceptible cultivars. The elicitor activity of HWP was dose-dependent, the optimal concentration which induces a maximum PAL activity was 10 mg of mycelium per mL. The elicitor present in the HWP was thermostable since its elicitor activity was maintained after heat treatment (121 °C for 45 min). The treatment of the HWP with protease (Pronase E) does not have an effect on the HWP elicitor activity. However, the treatment of the HWP with sodium periodate inhibits its elicitor activity. This data suggests that the HWP elicitor is a carbohydrate compound. In addition, the HWP elicitor is non-specific since it induces identical responses of the PAL activity in two cultivars showing different behaviors to the pathogen. The absence of specificity of HWP elicitors and the differential response of the PAL activity to the infection byFoa and to the elicitation by the HWP are discussed. An explanation of the general interactions between plant and parasite is proposed.  相似文献

The bayoud disease, vascular fusariosis of date palm tree (Phoenix dactylifera L.), is caused by the pathogenic fungus Fusarium oxysporum f. sp. albedinis. The characteristic symptoms of the bayoud disease were elicited on detached leaves of F. oxysporum f. sp. albedinis‐susceptible cultivars of date palm trees, which were treated either with the FII (F. oxysporum f. sp. albedinis) fraction purified from the organic extracts of a F. oxysporum f. sp. albedinis liquid culture, or with a solution of fusaric acid. Enniatins, which are secreted by several Fusarium species, were tested at different concentrations and were not capable of inducing symptoms on such detached leaves. The FII (F. oxysporum f. sp. albedinis) fraction was unable to induce necrosis of potato slices, which indicates that it does not contain significant amounts of enniatins. The high‐performance liquid chromatography (HPLC) profiles of the FII (F. oxysporum f. sp. albedinis) fraction showed toxic peaks different from fusaric acid. A fraction, named FII (AZ4), was obtained from culture filtrates of a saprophytic Fusarium strain maintained in the same cultural conditions as for the F. oxysporum f. sp. albedinis. The HPLC profile of the FII (AZ4) fraction did not show the characteristic phytotoxic peaks present in the FII (F. oxysporum f. sp. albedinis) fraction. This finding well agrees with the fact that the FII (AZ4) fraction is not toxic to detached date palm leaves. Moreover, the HPLC profiles of FII fractions obtained from other special forms of F. oxysporum are different the FII (F. oxysporum f. sp. albedinis) profile. The phytotoxic compounds purified from the FII (F. oxysporum f. sp. albedinis) fraction are probably new molecules that may help in understanding the pathogenesis of bayoud disease.  相似文献

Alfalfa (Medicago sativa L.) cell suspension cultures of Verticillium albo-atrum resistant and susceptible genotypes were established from leaf callus tissues. Treatment of cultures with conidia and heat-released elicitors of V. albo-atrum induced a large increase in the activity of phenylalanine ammonia-lyase, only in the cells of the resistant genotypes with a maximum after 12 h. In co-cultivation with the fungal conidia and resistant cell lines, the production of spores were inhibited. Fusarium oxysporum albedinis was grown in the presence of 20 different sugars or their derivatives. The results obtained were correlated with the stereochemical structure of the main oses. Osmotic phenomena as well as active and selective transport were used to explain differences in utilization of oses of very near structure. The fusion of protoplasts from the cycloheximide-resistant mutant FOL(C) of Fusarium oxysporum f. sp. lycopersici (FOL) and the mycostatin-resistant mutant FORL(M) of F. oxysporum f. sp. radicis-lycopersici (FORL), produced hybrids which expressed significant differences from the parents in their pathogenicity and growth and in the electrophoretic separation patterns of their proteins, enzymes and isoenzymes. The results suggest a transformed genetic basis for these altered expressions and the feasibility of using protoplast fusion technology for examining the biology of pathogenicity genes and for elucidating the disease and virulence potential for new races from within hybridisable taxa of Fusarium spp. Such information would be useful for the design and development of long-term control systems for Fusarium diseases, particularly in breeding programs for disease resistance in crops.  相似文献

Fusarium oxysporum f. sp. albedinis, the bayoud disease agent of date palm, grows on a mineral medium containing the cell walls of date palm roots as a sole carbohydrate source. The growth and development of pathogen under these conditions was related to the production of extracellular cell wall-degrading enzymes (CWDE): cellulases, polygalacturonases, polygalacturonate transeliminases, and pectinmethylesterases. The mycelial growth and the sporulation of pathogen were higher in the presence of cell walls of susceptible cultivars (BFG, JHL, BSK) than in the presence of those of resistant cultivars (IKL, SLY, BSTN). After 8 d of fungal culture, the activity of CWDE was equal whatever is the origin of the cell walls (resistant or susceptible cultivars). After 16 d of culture, the activity of these CWDE was higher when the parasite was cultivated on the cell walls of the susceptible cultivars than on those of the resistant cultivars. A positive correlation was observed between CWDE activities and the growth and the sporulation of F. oxysporum after 16 d of culture. These results clearly show a relation between the susceptibility/resistance of the cell walls of the roots of the date palm to the parasitic CWDE and the susceptibility/resistance of the cultivars.  相似文献

These studies are concerned with the isolation and identification of secondary metabolites produced by Fusarium oxysporum f. sp. albedinis (F. o. a.), the causal agent of bayoud, the wilt disease of the date palm (Phoenix dactylifera L.). Fungal secondary metabolites are chemical compounds identified in a limited number of species. They consist of toxins, antibiotics and antifungal agents. Among the metabolites we could isolate from the pathogen grown in a liquid medium, and then identify by gas chromatography coupled with mass spectrometry (GC-MS), phenylacetic acid has been distinguished. This compound is widely described in the literature as having antimicrobial, antifungal, phytotoxic properties and also endowed with hormonal activity similar to that of indole acetic acid (IAA). The genomes of many filamentous fungi consist of a ‘core’ part containing conserved genes essential for normal development as well as conditionally dispensable (CD) or lineage‐specific (LS) chromosomes. In the plant‐pathogenic fungus Fusarium oxysporum f. sp. lycopersici, one LS chromosome harbours effector genes that contribute to pathogenicity. We employed flow cytometry to select for events of spontaneous (partial) loss of either the two smallest LS chromosomes or two different core chromosomes. We determined the rate of spontaneous loss of the ‘effector’ LS chromosome in vitro at around 1 in 35 000 spores. In addition, a viable strain was obtained lacking chromosome 12, which is considered to be a part of the core genome. We also isolated strains carrying approximately 1‐Mb deletions in the LS chromosomes and in the dispensable core chromosome. The large core chromosome 1 was never observed to sustain deletions over 200 kb. Whole‐genome sequencing revealed that some of the sites at which the deletions occurred were the same in several independent strains obtained for the two chromosomes tested, indicating the existence of deletion hotspots. For the core chromosome, this deletion hotspot was the site of insertion of the marker used to select for loss events. Loss of the core chromosome did not affect pathogenicity, whereas loss of the effector chromosome led to a complete loss of pathogenicity.  相似文献

The pathogenicity of different isolates of Fusarium oxysporum obtained from plants of Gerbera (Gerbera jamesonii), Chrysanthemum (Chrysanthemum morifolium), Paris daisy (Argyranthemum frutescens) and African daisy (Osteospermum sp.), all in the family Asteraceae, was tested on different cultivars of these hosts, to assess their pathogenicity. The reactions were compared with those of isolates of F. oxysporum f. sp. chrysanthemi and of f.sp. tracheiphilum obtained from the American Type Culture Collection. We found that isolates of F. oxysporum f. sp. chrysanthemi can be distinguished as three physiological races on the basis of their pathogenicity to the panel of differential cultivars. Sequencing of the intergenic spacer (IGS) region of ribosomal DNA (rDNA) and phylogenetic analysis showed that the Fusarium races fell into three phylogenetic groups, which coincided with those observed in pathogenicity tests. Analysis of the IGS sequences revealed a high degree of similarity among strains from Italy and Spain from different host species, suggesting that recent outbreaks in these ornamentals were probably caused by introduction of infected nursery material from a common origin.  相似文献

Fungus gnat adults transported Fusarium oxysporum f.sp. radicis-lycopersici from Petri dish culture and infected host plants to the roots and hypocotyls of healthy tomato and bean plants. The source of the fungus did not affect the ability of fungus gnats to transport the fungus to healthy hosts. The presence of fungus gnat larvae in media in which young tomato plants were grown did not increase the incidence of plant infection by the pathogen. Fungus gnat adults appear to aid in the dissemination of F. oxysporum f.sp. radicis-lycopersici.  相似文献

This study set out to build a model identifying areas where a positive Net Present Value (NPV) could be obtained from date palm (Phoenix dactylifera) using CLIMEX and six parameters including (a) suitable soil taxonomy and physicochemical soil properties, (b) slopes of less than 10°, (c) land uses suitable for date palm cultivation, (d) availability of roads, (e) availability of water, and (f) low risk of the lethal disease caused by Fusarium oxysporum f. spp. in the years 2030, 2050, 2070, and 2100 in Iran. Here, we utilized the A2 scenario and two global climate models (GCMs): CSIRO-Mk3.0 (CS) and MIROC-H (MR). Economic feasibility was estimated based on the assumption that the decision to plant date palms by landholders is motivated by a desire to maximize their return to land. Our results indicate that only 5450 km2 of southern Iran will be highly profitable for cultivation of date palm, with NPV > 10,000, while profitable (with NPV between 4200 and 10,000) and moderately profitable (with NPV between 0 and 4200) areas would cover only 500 and 50 km2, respectively, in future. A comparison of mean outputs from the two chosen GCMs and those of the economic and CLIMEX output combination indicates that only about 0.01% of areas from both GCMs will be highly economically viable for cultivation of date palm. In this study we ensure that the predictions become robust, rather than producing hypothetical findings, limited purely to publication.  相似文献

以3株黄瓜尖镰孢菌(Fusarium oxysporum f.sp.cucumarinum)、23株镰孢菌属(Fusariumspp.)真菌和分离自土壤的20株真菌、6株细菌和7株放线菌为材料,采用化学裂解法提取总DNA,进行PCR-RFLP和巢式PCR检测,试验证明PCR-RFLP程序不能完全区分Fusarium属内不同种,而巢式PCR对黄瓜尖镰孢菌具有特异性.运用优化的PCR-RFLP和巢式PCR检测程序对染病黄瓜组织进行了检测,结果表明,两种方法均可在接种发病早期(未显症时)检测出黄瓜枯萎病菌,PCR-RFLP在感病品种接种后3d即可检测到病原菌,而巢式PCR在接种后5d才能检测到病原菌.  相似文献

Populations of Fusarium oxysporum f. sp. albedinis, the causal agent of Bayoud disease of date palm, are derivatives of a single clonal lineage and exhibit very similar Fot 1 hybridization patterns. In order to develop a sensitive diagnostic tool for F. oxysporum f. sp. albedinis detection, we isolated several DNA clones containing a copy of the transposable element Fot 1 from a genomic library of the date palm pathogen. Regions flanking the insertion sites were sequenced, and these sequences were used to design PCR primers that amplify the DNA regions at several Fot 1 insertion sites. When tested on a large sample of Fusarium isolates, including 286 F. oxysporum f. sp. albedinis isolates, 17 other special forms, nonpathogenic F. oxysporum isolates from palm grove soils, and 8 other Fusarium species, the primer pair TL3-FOA28 allowed amplification of a 400-bp fragment found only in F. oxysporum f. sp. albedinis. Sequence analysis showed that one of the Fot 1 copies was truncated, lacking 182 bp at its 3′ terminus. The primer pair BI03-FOA1 amplified a 204-bp fragment which overlapped the Fot 1 truncated copy and its 3′ site of insertion in the F. oxysporum f. sp. albedinis genome and identified 95% of the isolates. The primer pairs BIO3-FOA1 and TL3-FOA28 used in PCR assays thus provide a useful diagnostic tool for F. oxysporum f. sp. albedinis isolates.  相似文献

The roots of date palm contain four cell wall‐bound phenolic acids identified as p‐hydroxybenzoic, p‐coumaric, ferulic and sinapic acids. The ferulic acid represents the major phenolic compound since it constitutes 48.2–55.8% of cell wall‐bound phenolic acids. All these phenolic acids were present in the resistant cultivar (BSTN) and the susceptible cultivar (JHL). However, the pre‐infection contents of p‐coumaric, ferulic and sinapic acids were greater in the resistant cultivar than in the susceptible one. For the contents of p‐hydroxybenzoic acid, there was no significant difference between the resistant cultivar and the susceptible cultivar. Similarly, the pre‐infection contents of lignin were approximately equal for both cultivars. Inoculation of the date palm roots by Fusarium oxysporum f. sp. albedinis induced important modifications to the contents of the cell wall‐bound phenolic compounds and lignin, which made it possible to distinguish between resistant and susceptible cultivars. The post‐infection contents of cell wall‐bound phenolic compounds underwent a rapid and intense increase with a maximum accumulation on the tenth day for p‐hydroxybenzoic acid (1.54 μmol/g), p‐coumaric acid (2.77 μmol/g) and ferulic acid (2.64 μmol/g) and on the fifteenth day for sinapic acid (1.85 μmol/g). The maximum contents accumulated in the resistant cultivar were greater than those in the susceptible cultivar, namely, 11 times for p‐hydroxybenzoic acid, 2.6 times for p‐coumaric acid, 1.8 times for ferulic acid and 12.3 times for sinapic acid. In the susceptible cultivar, p‐coumaric acid and ferulic acid contents also increased after inoculation although they did not reach the pre‐infection contents of the resistant cultivar. The contents of p‐hydroxybenzoic acid in the susceptible cultivar roots did not present post‐infection modification and those of sinapic acid decreased instead. The lignin contents increased in both cultivars with a maximum accumulation on the fifteenth day. However, the maximum contents accumulated in the resistant cultivar roots were 1.5 times greater than those of the susceptible cultivar. These results showed clear differences between the resistant BSTN and the susceptible JHL cultivars. The implication of cell wall‐bound phenolic compounds and lignin in the resistance of date palm to F. oxysporum f. sp. albedinis appears to be dependent on the speed and intensity of their accumulation with greater contents in the first stage of infection.  相似文献

Bayoud, a vascular wilt of date palm caused by Fusarium oxysporum f. sp. albedinis (Foa), is the most devastating disease in palm groves of north Africa. Although Foa is able to induce resistance mechanisms in its host plant, no data are available on the early responses of the root cells. Fusaric acid (FA) is the main toxin found in culture filtrates from Foa aggressive strains. This phytotoxin induces modifications of membrane permeability or membrane potential in various cell types and could thus be involved in the early steps of signal exchange between the pathogen and the plant. We showed an early differential behaviour of the disease‐resistant and ‐susceptible cultivars from date palm when challenged by FA. This response could be due to a differential sensitivity of H+‐ATPases to FA.  相似文献

Root rot severity of asparagus plants grown in sterilized field soil inoculated with Fusarium oxysporum f . sp . asparagi (Foa) was reduced by more than 50% when the soil was precolo nized by each of 13 non - pathogenic (np) isolates of F. oxysporum originating from asparagus roots or field soils . In a greenhouse experiment , application of six np isolates to naturally infested field soil was followed by a 23 - 49% decrease of disease severity , depending on the isolate . One of them , Fo47 originating from Fusarium suppressive soil in France , was applied to field plots infested with Foa . Foa root rot was not suppressed in asparagus plants grown for 1 year in these plots . Pathogenic and np isolates extensively colonized the root surface and isolates of both types infected the roots of asparagus plants grown in sterilized field soil , with significant differences among the np isolates . Inoculation of sterilized field soil with np isolates reduced germination of Foa chlamydospores by 43 - 64% depending on the isolate used . It is concluded that np isolates of F. oxysporum can suppress asparagus root rot caused by Foa in naturally infested field soil . The differences for root colonization capacity among the np isolates imply that selection for this trait might reveal isolates that perform better under field conditions .  相似文献

Seventy isolates of Fusarium oxysporum f.sp. ciceris (Foc) causing chickpea wilt representing 13 states and four crop cultivation zones of India were analysed for their virulence and genetic diversity. The isolates of the pathogen showed high variability in causing wilt incidence on a new set of differential cultivars of chickpea, namely C104, JG74, CPS1, BG212, WR315, KWR108, GPF2, DCP92‐3, Chaffa and JG62. New differential cultivars for each race were identified, and based on differential responses, the isolates were characterized into eight races of the pathogen. The same set of isolates was used for molecular characterization with four different molecular markers, namely random amplified polymorphic DNA, universal rice primers, simple sequence repeats and intersimple sequence repeats. All the four sets of markers gave 100% polymorphism. Unweighted paired group method with arithmetic average analysis grouped the isolates into eight categories at genetic similarities ranging from 37 to 40%. The molecular groups partially corresponded to the states of origin/chickpea‐growing region of the isolates as well as races of the pathogen characterized in this study. The majority of southern, northern and central Indian populations representing specific races of the pathogen were grouped separately into distinct clusters along with some other isolates, indicating the existence of variability in population predominated by a single race of the pathogen. The present race profiling for the Indian population of the pathogen and its distribution pattern is entirely new. The knowledge generated in this study could be utilized in resistance breeding programme. The existence of more than one race, predominated by a single one, in a chickpea cultivation zone as supported by the present molecular findings is also a new information.  相似文献

Polyamine oxidase and lipoxygenase enzymes are key players for hyper sensitive reaction (HR) during incompatible interaction of host-pathogen. Thus, the role of lipoxygenase and polyamines was studied in the wilt pathogen infected and non infected tissues of resistant and susceptible genotypes of castor at 0 days after infection (DAI), 5 DAI and 10 DAI (30 days after sowing). The lipoxygenase (LOX) and polyamine oxidase (PAO) activities were higher in the incompatible interaction at all the stages of analysis. The constitutive level of malondyaldehyde (MDA) content, a product of lipid peroxidation was higher in susceptible genotypes (VP-1 and VI-9), while induced level was higher in resistant genotypes (48–1 and SKP-84) at 5 DAI and 10 DAI . Polyamine profiling using HPTLC showed higher spermidine and spermine content in resistant genotypes at 10 DAI. Furthermore, spermidine was detected only in the roots of resistant genotypes at 10 DAI. These results suggest the role of high titers of polyamines, LOX and PAO in disease resistance possibly through HR induction.  相似文献
