The geographic distribution of haplotype diversity in the rodent Calomys musculinus, sampled from 16 wild populations of Argentina, was analysed on two geographical scales. The species is the natural reservoir of the Junin virus, the etiological agent of the Argentine haemorrhagic fever (AHF). In all, 24 composite haplotypes were recognised in the mtDNA D-loop region. Haplotypes 1 and 2, internal in the network, were the most frequent and were present in almost all populations. The absence of large genetic gaps between widely distributed haplotypes, the existence of exclusive haplotypes in more than 50% of the sampled populations and the absence of isolation by distance at a macrogeographical scale are in support of the hypothesis of a recent range expansion of the populations of the Humid Pampa, with low to moderate current gene flow. The dispersal of this opportunistic species would have been favoured by the explosive increments in density after agriculture was introduced. When only nearby populations within the endemic area of AHF were considered, a pattern of isolation by distance was detected. At present, genetic drift appears to be the main force acting to randomly differentiate C. musculinus populations, which would also lead to random differentiation of Junin virus strains and a reduction in the virulence of the pathogen in 'historic' AHF areas. The knowledge of migration patterns of the reservoir populations facilitates reliable prediction of the potential spread of the human disease.  相似文献

Random amplified polymorphic DNA polymerase chain reaction (RAPD-PCR) markers were used to evaluate the relative contribution of gene flow as a determinant of the population genetic structure of the wild rodent Calomys musculinus (the reservoir of Argentine hemorrhagic fever [AHF]) in central Argentina. One hundred eighty-seven individuals from 13 populations (9 of them from the endemic zone of AHF and 5 from areas outside it) were analyzed using 78 polymorphic RAPD loci. Genetic variation within each population was high; each individual was characterized by a unique RAPD phenotype. C. musculinus populations showed a moderate to high genetic subdivision and a random pattern of differentiation. Populations separated by the same geographic distance showed very different degrees of genetic divergence. The results indicate that populations of C. musculinus have colonized their present ranges relatively recently and differentiation by genetic drift has proceeded faster than homogenization by gene flow at the macrogeographic scale analyzed (10-700 km).  相似文献

In order to assess the reliability of RAPD markers in the estimation of the genetic structure of natural populations of the murid rodent Calomys musculinus (reservoir of Junin virus, ethiological agent of Argentine Hemorrhagic Fever), we have analyzed the heritability of RAPD bands in 10 parents and their offspring (33 individuals). Fourteen out of a total of 119 bands obtained were absent in the parental patterns, but consistently amplified in offspring from some families. These bands can be eliminated from analyses. Overall degree of band sharing between individuals, including non-parental bands, correctly grouped members of a family in the same cluster in a UPGMA tree, with a high bootstrap percentage. Results support the usefulness of RAPDs as hereditable markers. One hundred polymorphic RAPD loci were identified in three natural populations of C. musculinus. Mean expected heterozygosity in three natural populations ranged from 0.206 to 0.220. Allele frequency based and phenotype based measures of genetic differentiation among natural populations of C. musculinus gave similar results (Weir and Cockerham's theta = 0.133; Excoffier et al.'s phi = 0.127). These values were considerably higher than those found previously using allozymes as genetic markers, and are compatible with moderate to low levels of gene flow among populations.  相似文献

胡黄连为特产中国-喜马拉雅特有高山植物,作为常用中、藏药材,受到灭绝性采挖,作为濒危和二级保护植物亟待科学的保护。该研究以云南和西藏7个野生居群91个个体为材料,基于 cpDNA trnL-F 非编码序列测序分析胡黄连的遗传多样性和遗传结构,分析显著进化单元,确立优先保护居群并提出科学的保护策略。结果表明:胡黄连 trnL-F 序列长度为871~876 bp,根据序列的核苷酸变异共鉴定出5个单倍型,西藏占有2个单倍型,云南占有3个单倍型,西藏和云南2个地区的所有单倍型均不共享。胡黄连具有较低的单倍型多样性(Hd =0.43419)和核苷酸多样性(Dij =0.00466)。种群间分化度(Fst =0.864520)和基因流(Nm =0.04)、居群间的遗传分化水平(GST =0.916)、AMOVA 分析(0.78%的遗传变异发生在居群内,60.97%的遗传变异发生在地区内居群间,38.25%的遗传变异发生在地区间)均表明,胡黄连居群间存在明显遗传分化。多数一致性树将胡黄连划分为3个进化分支(Ⅰ、Ⅱ、Ⅲ),这3个分支均与地理相关,分支Ⅰ分布于横断山区的4个居群,分支Ⅱ是分布于东喜马拉雅的一个居群,分支Ⅲ是分布于喜马拉雅中段的2个居群。3个分支分属于3个“进化显著单元(ESU)”。这3个 ESU 中白马雪山、茨中、定日、波密、聂拉木五个居群都需要保护,建议现阶段应优先保护的居群是云南白马雪山和西藏波密居群,以就地保护为主。  相似文献

油松(Pinus tabulaeformis)是我国特有广布树种。秦岭是我国亚热带和暖温带气候的重要分界线,贺兰山是我国西北干旱区与半干旱区的分界线。旨在以分布于秦岭、贺兰山和晋陕两省的10个天然油松种群为研究对象,通过对其线粒体DNA(nad1和matR内含子)的序列,探讨地理阻隔对油松遗传结构的影响。结果表明,10个天然油松种群的100个个体中共检测到27个单倍型,多态位点172个,简约信息位点35个,单一多态性位点137个。其中1个单倍型为全部种群共有,4个单倍型为2-5个种群共有,其余22个单倍型为各个种群所独有。在这10个种群中,有8个种群分别具有1-4个独有单倍型。尽管秦岭南侧种群和北侧种群分别具有两个和一个独有单倍型; 贺兰山东侧种群和西侧种群均具有3个独有单倍型;但是与晋陕种群(即油松分布区中心种群)相比,其种群独有单倍型平均数目则明显较少。与此同时,不仅秦岭南北两侧或贺兰山东西两侧种群的独有单倍型的进化关系往往较近,而且秦岭北侧或贺兰山东侧种群的独有单倍型与油松分布区中心种群的某些独有单倍型的关系亦较近。从而导致秦岭南北两侧或贺兰山东西两侧种群与油松分布区中心种群之间表现出比较复杂的聚类关系。由此可见,油松的遗传结构与山脉屏障的存在没有明显关系。  相似文献

Breeding structure of Glossina morsitans submorsitans Newstead was evaluated by using genetic markers in mitochondrial DNA where diversity was scored at two loci in five natural populations from The Gambia and two populations in Ethiopia (form ugandensis Vanderplank), countries separated by c. 5450 km. Twenty six haplotype combinations were found, of which 17 were shared among two or more populations. Nine haplotypes were found in The Gambia and 23 haplotypes in Ethiopia. There were 12 unique haplotypes. Only six haplotypes were shared between the two countries. Populations in The Gambia (he = 0.26 +/- 0.04) showed less than a third of the diversity of populations in Ethiopia (he = 0.84 +/- 0.03). This suggests recovery from an earlier reduction in population. In a nested analysis of molecular variance of haplotype frequencies, 65% of the variance was due to differences within populations, 34% to differences between populations grouped by country, and only 1% was due to differences among populations within countries. In terms of gene flow, the fixation index FST = 0.35, which leads to an estimate by Wright's island model of less than one reproducing migrant per generation exchanged between the eastern and western submorsitans populations. Nei's genetic similarity measure showed a deep division between Gambian and Ethiopian populations.  相似文献

Identification and characterization of the self-incompatibility genes in Brassicaceae species now allow typing of self-incompatibility haplotypes in natural populations. In this study we sampled and mapped all 88 individuals in a small population of Arabidopsis lyrata from Iceland. The self-incompatibility haplotypes at the SRK gene were typed for all the plants and some of their progeny and used to investigate the realized mating patterns in the population. The observed frequencies of haplotypes were found to change considerably from the parent generation to the offspring generation around their deterministic equilibria as determined from the known dominance relations among haplotypes. We provide direct evidence that the incompatibility system discriminates against matings among adjacent individuals. Multiple paternity is very common, causing mate availability among progeny of a single mother to be much larger than expected for single paternity.  相似文献

【目的】为了明确我国甜菜夜蛾Spodoptera exigua地理种群间的遗传分化及基因流,阐明该种害虫在我国的种群历史动态。【方法】本研究对采自我国20个地理种群的529头甜菜夜蛾样品进行线粒体COI基因序列测定与分析,利用DnaSP 5.0和Arlequin 3.11软件分析种群间遗传多样性、遗传分化、基因流水平及分子变异,构建了单倍型系统发育树与单倍型网络图。【结果】在所分析的所有529个序列样本中,共检测出10个单倍型,其中Hap_1为所有种群所共享。总群体遗传多样性较低(Hd=0.257±0.025,Pi=0.0007±0.0001,Kxy=0.323),群体间遗传分化较小(FST=0.211),基因流水平较高(Nm=1.870)。AMOVA分析表明,甜菜夜蛾遗传变异主要来自种群内,种群间变异水平较低。各种群间遗传分化程度与地理距离无显著相关性(R2=0.005,P>0.05)。各单倍型相互散布在不同种群中,未形成明显系统地理结构。中性检验(Tajima’s D=-2.177, P<0.05; Fu’s FS=-8.629, P<0.05)与错配分布分析表明,我国甜菜夜蛾种群曾经历种群近期扩张。【结论】研究结果揭示,甜菜夜蛾各种群间的基因交流并未受到地理距离的影响,验证了甜菜夜蛾具有高度的迁飞能力。  相似文献

In a previous phylogeographic study of the rodent Calomys musculinus, 24 haplotypes of the mitochondrial DNA D-loop region were detected using the restriction fragment length polymorphism technique (PCR-RFLP). Seven percent of the individuals showed patterns in which the sum of the sizes of the restriction fragments exceeded the size of the original PCR product. In the present paper we analyze possible causes of these atypical haplotypes. PCR products were cloned, and two or three different clones from a single individual were detected by their RFLP patterns. Nine clones with different restriction patterns were selected for sequence analyses. A maximum parsimony phylogenetic analysis revealed two well-supported paraphyletic groups. One group comprised sequences showing low nucleotide divergence compared with the most common haplotypes detected in the phylogeographic study. The other group was basal to the three species of Calomys other than C. musculinus included in the study; the mutations in the short portion of the cytochrome b gene amplified corresponded to 12 amino acid substitutions. The results suggest that two independent insertions of mtDNA sequences into the nucleus occurred; these sequences would co-amplify in the PCR procedure. Identification of pseudogenes is crucial to obtain reliable reconstruction of the intraspecific genealogy in phylogeographic studies.  相似文献

为了合理有效地保育天然臭柏(Juniperus sabina L.)种质资源,追溯和阐释其分布格局的历史成因,本文对我国内蒙古自治区、陕西省、甘肃省和青海省共10个天然臭柏居群388个个体的核糖体内转录间隔区(ITS)序列片段进行测序分析。结果显示:臭柏ITS序列总长度为1089 bp,共含有25个变异位点,定义32个单倍型,其中H4和H6单倍型为共有单倍型;分子变异分析(AMOVA)显示,臭柏居群变异主要来源于居群内,遗传变异为95.04%,而居群间遗传变异仅4.96%,居群间差异水平极显著(F ST=0.0496,P<0.001);Network单倍型网络分析表明,H4和H6为古老单倍型,其他单倍型是由他们衍生而来;遗传分化系数N ST(0.072)0.05)。推测臭柏起源于第三纪中新世(Miocene)中期约12.38 Mya,在第四纪冰期可能存在多个小型避难所。沙埋产生不定根的扩繁能力和较好的有性更新环境可能是沙地居群遗传多样性高于山地居群的决定性因素。  相似文献

Recent work has suggested that rates of introgression should be inversely related to levels of gene flow because introgressed populations cannot be 'rescued' by intraspecific gene flow if it is too low. Mitochondrial and chloroplast DNA (mtDNA and cpDNA) experience very different levels of gene flow in conifers due to their contrasted maternal and paternal modes of transmission, hence the prediction that mtDNA should introgress more readily than cpDNA in this group. Here, we use sequence data from both mtDNA and cpDNA to test this hypothesis in a group of closely related spruces species, the Picea asperata complex from China. Nine mitochondrial and nine chloroplast haplotypes were recovered from 459 individuals in 46 natural populations belonging to five species of the Picea asperata complex. Low variation was found in the two mtDNA introns along with a high level of differentiation among populations ( G ST = 0.90). In contrast, we detected higher variation and lower differentiation among populations at cpDNA markers ( G ST = 0.56), a trend shared by most conifer species studied so far. We found that cpDNA variation, although far from being fully diagnostic, is more species-specific than mtDNA variation: four groups of populations were identified using cpDNA markers, all of them related to species or groups of species, whereas for mtDNA, geographical variation prevails over species differentiation. The literature suggests that mtDNA haplotypes are often shared among related conifer species, whereas cpDNA haplotypes are more species-specific. Hence, increased intraspecific gene flow appears to decrease differentiation within species but not among species.  相似文献

【目的】通过对四纹豆象Callosobruchus maculatus Fabricius不同地理种群mtDNA-Cytb和COⅠ基因部分序列进行比较, 分析其不同地理种群间的遗传分化情况, 为揭示其与生物入侵的关系及入侵过程中种群系统发育地理格局与演变机制提供依据。【方法】用PCR产物直接测序法对分别来自中国海南、喀麦隆、韩国和泰国的四纹豆象4个地理种群的mtDNA-Cytb和COⅠ序列进行测序, 运用软件MEGA3.1对四纹豆象不同地理种群mtDNA-Cytb和COⅠ序列进行序列分析, 以绿豆象C. chinensis为外群构建了不同单倍型的分子系统树。【结果】34条420 bp Cytb序列中共检测到14个多态位点和5种单倍型, 33条822 bp COⅠ序列中检测到28个多态位点和9种单倍型, 其中4种单倍型为独享单倍型, 其余为全部或部分种群的共享单倍型。AMOVA分析结果显示, 四纹豆象4个地理种群间的遗传结构差异并不明显, 遗传差异主要发生在地理种群内。对4个地理种群进行了Fst值和基因流动统计, 结果表明4个地理种群间既存在着一定数量的基因交流, 也存在一定程度的遗传分化。【结论】根据单倍型分布格局初步推测, 中国不可能是四纹豆象的原产地, 而喀麦隆有可能是原产地之一, 并且喀麦隆种群与泰国种群之间的基因交流比较充分, 而中国种群与其他种群之间的遗传分化相对较大。  相似文献

利用叶绿体DNA三个片段(trnK-matK、trnL-trnF、rpl32-trnL)对中国大陆广布的水生植物穗状狐尾藻(Myriophyllum spicatum L.)的遗传多样性进行了初步分析,以探讨其自然居群的遗传结构及具有广泛分布格局的可能机制。AMOVA分析显示,穗状狐尾藻8个居群间的遗传变异为84.97%,而居群内的遗传变异为15.03%,居群遗传分化系数(Fst)为0.85,表明穗状狐尾藻具有较高的遗传多样性(Hd=0.83)且主要存在于居群间,奠基者效应可能导致了最初的遗传差异,而隔离障碍(Nm=0.09)又进一步导致了居群间的遗传分化。基于17个单倍型构建的系统发育树和网络关系图均显示,单倍型H5和H6在居群中的分布范围最广且出现频率最高,表明H5和H6可能为最古老的祖先单倍型。Mantel检验表明居群间的遗传距离与地理距离之间不存在显著的相关性,失配分布检测结果显示穗状狐尾藻在历史上曾发生过扩张事件,而Tajima’s、FuLi’s D*和F*检测发现,该物种不存在明显的谱系地理格局,这可能与穗状狐尾藻种子的长距离扩散有关。  相似文献

Abstract.  Variation in the mtDNA 16S ribosomal RNA gene in populations of Triatoma infestans (Klug) was surveyed. DNA sequence comparisons yielded 18 haplotypes among 130 individuals from 16 localities that represent a large proportion of the range of T. infestans in Argentina. The most common genotype in all populations was found in 76.9% of individuals and two other haplotypes were shared among different populations. The remaining 15 haplotypes were present exclusively in one of the populations, suggesting currently low levels of genetic exchange. Analysis of mtDNA 16S sequences uncovered substantial genetic variation among T. infestans populations. Haplotype and nucleotide diversities varied among populations, from 0% to 0.84% and 0% to 0.29%, respectively. It appears that this locus has a low mutation rate. Uncorrected pairwise differences of T. infestans haplotypes ranged from 0% to 1.2%. The molecular phylogeny supported the monophyly of T. infestans haplotypes and clustered two different pairs of haplotypes with a moderate degree of bootstrap support (∼ 60%). Mitochondrial DNA phylogeographic differentiation was not evident, suggesting a recent rapid spread of the species. Analysis of molecular variance showed hierarchical structure in the data. Considerably less variation was found among T. infestans populations from the northwest and northeast regions than among those belonging to the central area. Such a lack of variation may be indicative of one or more past population bottlenecks.  相似文献

BACKGROUND: Evolution depends on natural selection acting on phenotypic variation, but the genes responsible for phenotypic variation in natural populations of vertebrates are rarely known. The molecular genetic basis for plumage color variation has not been described in any wild bird. Bananaquits (Coereba flaveola) are small passerine birds that occur as two main plumage variants, a widespread yellow morph with dark back and yellow breast and a virtually all black melanic morph. A candidate gene for this color difference is the melanocortin-1 receptor (MC1R), a key regulator of melanin synthesis in feather melanocytes. RESULTS: We sequenced the MC1R gene from four Caribbean populations of the bananaquit; two populations of the yellow morph and two populations containing both the yellow morph and the melanic morph. A point mutation resulting in the replacement of glutamate with lysine was present in at least one allele of the MC1R gene in all melanic birds and was absent in all yellow morph birds. This substitution probably causes the color variation, as the same substitution is responsible for melanism in domestic chickens and mice. The evolutionary relationships among the MC1R haplotypes show that the melanic alleles on Grenada and St. Vincent had a single origin. The low prevalence of nonsynonymous substitutions among yellow haplotypes suggests that they have been under stabilizing selection, whereas strong selective constraint on melanic haplotypes is absent. CONCLUSIONS: We conclude that a mutation in the MC1R is responsible for the plumage polymorphism in a wild bird population and that the melanic MC1R alleles in Grenada and St. Vincent bananaquit populations have a single evolutionary origin from a yellow allele.  相似文献

The Magadi tilapia (Alcolapia grahami, formerly Oreochromis alcalicus grahami) is a remarkable example of teleost life in an extreme environment. Typical conditions include water pH=10, titration alkalinity>300 mM, osmolality=525 mOsm, temperatures ranging from 23 degrees to 42 degrees C, and O(2) levels fluctuating diurnally between extreme hyperoxia and anoxia. A number of relatively small tilapia populations are present in various thermal spring lagoons around the margin of the lake separated by kilometers of solid trona crust (floating Na(2)CO(3)) underlain by anoxic water. Despite the apparent isolation of different populations, annual floods may provide opportunities for exchange of fish across the surface of the trona and subsequent gene flow. To assess the question of isolation among Lake Magadi populations, we analyzed the variable control region of the mitochondrial DNA (mtDNA) from six lagoons. A total of seven mtDNA haplotypes, including three common haplotypes, were observed in all six populations. Several of the Lake Magadi populations showed haplotype frequencies indicative of differentiation, while others showed very little. However, differentiation among lagoon populations was discordant with their geographical distribution along the shoreline. All populations exhibited the unusual trait of 100% ureotelism but specialized morphological and physiological characteristics were observed among several of the lagoon systems. In addition, distinct differences were observed in the osmolality among the lagoons with levels as high as 1,400-1,700 mOsm kg(-1), with corresponding differences in the natural levels of whole-body urea. These levels of osmotic pressure proved fatal to fish from less alkaline systems but remarkably were also fatal to the fish that inhabited lagoons with this water chemistry. Upon more detailed inspection, specific adaptations to differential conditions in the lagoon habitat were identified that allowed survival of these cichlids. Additional evidence against potential for gene flow among lagoons despite the sharing of common mtDNA haplotypes was that the osmolality of floodwaters following a heavy rain showed lethal levels exceeding 1,700 mOsm kg(-1). In isolation, different mtDNA haplotypes would be predicted to go to fixation in different populations due to rapid generation times and the small effective population sizes in a number of lagoons. We propose a model of balancing selection to maintain common mtDNA sequences through a common selection pressure among lagoons that is based on microhabitats utilized by the tilapia.  相似文献

毛茛叶报春(Primula ranunculoides Chen)是我国特有的珍稀濒危花卉,其野生种质资源已十分稀少。为了揭示其谱系地理结构,本文对11个居群114个个体的叶绿体trnL-F和trnS-G基因片段进行了测序分析。结果共检测到10个单倍型,除YJPA和YJPB两居群共享一个单倍型(H9)外,其余各居群均仅有一个特有单倍型,居群内不存在单倍型多样性。毛茛叶报春具高分化的谱系地理结构,居群间的总平均遗传距离达0.015,单倍型间的平均突变次数高达31.2步,两者均显著高于同属的近缘种。其种子散布能力弱、生境片断化和地理隔离明显及两年生的生活史可能是导致其高分化谱系地理结构产生的主要原因。Samova、Structure以及TCS分析均支持将毛茛叶报春11个居群的遗传结构分为6组,它们与地形(山脉)关系密切对应,彼此间分化明显,因此建议不同山体的居群应视为不同的保护单元进行遗传管理。  相似文献

黄胫小车蝗Oedaleus infernalis Saussure是一种在我国分布广泛、 对农牧业生产危害严重的经济害虫。本文对黄胫小车蝗10个地理种群的线粒体COI基因序列进行测序和分析, 利用DnaSP 5.0和Arlequin 3.5.1.2软件对该蝗虫种群间的遗传多样性、 遗传分化程度、 基因流水平及分子变异进行了分析, 建立了单倍型贝叶斯系统发育进化树和单倍型网络图。结果表明: 在所分析的144个序列样本中, 共检测到21种单倍型, 其中1种单倍型为10个地理种群所共享。总群体的单倍型多样性指数为0.653, 各地理种群单倍型多样度范围在0.423~0.790之间。总群体和各种群的Tajima’s D检验结果皆不显著, 说明该种害虫在较近的历史上未经历群体扩张。总群体的遗传分化系数Gst为0.04436, 固定系数Fst为0.05255, 基因流Nm为9.01。AMOVA分子方差分析结果表明, 黄胫小车蝗的遗传分化主要来自种群内部, 种群间的遗传变异水平较低。各地理种群的遗传距离的大小与其地理距离间没有显著的相关性。贝叶斯系统发育进化树与单倍型网络图显示, 黄胫小车蝗各地理种群中的单倍型散布在不同的分布群中, 分布格局较为混杂, 未形成明显的系统地理结构。研究结果揭示, 黄胫小车蝗各种群间的基因交流并未受到地理距离的影响。  相似文献

Angiotensin-I-converting enzyme (ACE) is known to be associated with human cardiovascular and psychiatric pathophysiology. We have undertaken a global survey of the haplotypes in ACE gene to study diversity and to draw inferences on the nature of selective forces that may be operating on this gene. We have investigated the haplotype profiles reconstructed using polymorphisms in the regulatory (rs4277405, rs4459609, rs1800764, rs4292, rs4291), exonic (rs4309, rs4331, rs4343), and intronic (rs4340; Alu [I/D]) regions covering 17.8 kb of the ACE gene. We genotyped these polymorphisms in a large number of individuals drawn from 15 Indian ethnic groups and estimated haplotype frequencies. We compared the Indian data with available data from other global populations. Globally, five major haplotypes were observed. High-frequency haplotypes comprising mismatching alleles at the loci considered were seen in all populations. The three most frequent haplotypes among Africans were distinct from the major haplotypes of other world populations. We have studied the evolution of the two major haplotypes (TATATTGIA and CCCTCCADG), one of which contains an Alu insertion (I) and the other a deletion (D), seen most frequently among Caucasians (68%), non-African HapMap populations (65?C88%), and Indian populations (70?C95%) in detail. The two major haplotypes among Caucasians are reported to represent two distinct clades A and B. Earlier studies have postulated that a third clade C (represented by the haplotypes TACATCADG and TACATCADA) arose from an ancestral recombination event between A and B. We find that a more parsimonious explanation is that clades A and B have arisen by recombination between haplotypes belonging to clade C and a high-frequency African haplotype CCCTTCGIA. The haplotypes, which according to our hypothesis are the putative non-recombinants (PuNR), are uncommon in all non-African populations (frequency range 0?C12%). Conversely, the frequencies of the putative recombinant haplotypes (PuR) are very low in the Africans populations (2?C8%), indicating that the recombination event is likely to be ancient and arose before, perhaps shortly prior to, the global dispersal of modern humans. The global frequency spectrum of the PuR and the PuNR is difficult to explain only by drift. It appears likely that the ACE gene has been undergoing a combination of different selective pressures.  相似文献

With the recent technical advances in molecular biology, chloroplast DNA (cpDNA) has become a marker used for the study of cytoplasmic differentiation of natural populations of plants. As chloroplasts are maternally inherited in most plant species, the seed component of gene flow is thus made accessible. We present here a study of cpDNA polymorphism within the maritima subspecies of the gynodioecious Beta vulgaris in which we try to assess the impact of such a reproductive system on seed flow. One hundred and eighty-eight wild beets were sampled from 20 hermaphroditic and 20 gynodioecious (i.e. containing both hermaphroditic and female plants) populations from the Atlantic coast of Europe. cpDNA variability in these populations was characterized with a rapid restriction fragment length polymorphism (RFLP) method. Eight cpDNA haplotypes were found. Strong differentiation among populations was observed ( F ST = 0.43) and was consistent with isolation by distance, although most of the cpDNA haplotypes were ubiquitous. Gynodioecy seems to affect the distribution of cpDNA diversity: gynodioecious populations of Beta vulgaris ssp. maritima contained a greater number of cpDNA types but were less differentiated among themselves than hermaphroditic ones.  相似文献
