A 120 min incubation period with sulfhydryl reagents, such as p-chloromercuribenzoic acid, shows greater than 50% loss of electron-transport activity in Photosystem (PS) II of spinach chloroplasts. Since p-chloromercuriphenylsulfonic acid, a nonpenetrating sulfhydryl reagent, and 4,4′-dithiopyridine, a bifunctional sulfhydryl reagent, show greater inhibition of 3-(3,4-dichlorophenyl)-1,1-dimethylurea-insensitive silicomolybdate reduction than of dibromothymoquinone-insensitive indophenol reduction, it is postulated that two different sulfhydryl reagent-sensitive sites are involved in the PS II electron-transport chain of spinach chloroplasts.  相似文献

We have studied the recovery of the photochemical activity of Photosystem I after the charge separation induced by a flash under conditions where the secondary donors are in the reduced form.

The rate-limiting steps are on the donor side. The first step is completed within 400 μs. The second step is much slower (half time 1 ms) and corresponds to the transfer of electrons from plastoquinone. Under our conditions, only one intermediate is involved in electron transfer between the centers and the plastoquinone pool.

Electron exchange between the System I centers has been demonstrated.  相似文献

Diphenylamines with highly electronegative substituents are effective inhibitors of photosynthetic electron transport and photophosphorylation. They inhibit only Photosystem II- and not Photosystem I-dependent photoreductions. As judged from the missing tetramethylphenylenediamine-bypass, displacement experiments with [14C]metribuzin, and measurements of oxygen evolution in trypsinated chloroplasts, diphenylamines act neither as dibromomethylisopropylbenzoquinone- nor as dichlorophenyldimethylurea-type inhibitors. All of the diphenylamines tested were found to function as ADRY-type reagents, (Renger, G. (1972) Biochim. Biophys. Acta 256, 428–439) which modify the stability of redox equivalents stored within the water-splitting enzyme system Y.The site of inhibition of diphenylamines is assumed to be located at the reducing side of Photosystem II or the reaction center itself. The inhibitory effect could involve a modification of cytochrome b-559 or its surrounding. In an assay for herbicidal activity, diphenylamines showed more pronouncing effect on mono- than on dicotyledonous plants.  相似文献

The ratio of Photosystem (PS) II to PS I electron-transport capacity in spinach chloroplasts was compared from reaction-center and steady-state rate measurements. The reaction-center electron-transport capacity was based upon both the relative concentrations of the PS IIα, PS IIβ and PS I centers, and the number of chlorophyll molecules associated with each type of center. The reaction-center ratio of total PS II to PS I electron-transport capacity was about 1.8:1. Steady-state electron-transport capacity data were obtained from the rate of light-induced absorbance-change measurements in the presence of ferredoxin-NADP+, potassium ferricyanide and 2,5-dimethylbenzoquinone (DMQ). A new method was developed for determining the partition of reduced DMQ between the thylakoid membrane and the surrounding aqueous phase. The ratio of membrane-bound to aqueous DMQH2 was experimentally determined to be 1.3:1. When used at low concentrations (200 μM), potassium ferricyanide is shown to be strictly a PS I electron acceptor. At concentrations higher than 200 μM, ferricyanide intercepted electrons from the reducing side of PS II as well. The experimental rates of electron flow through PS II and PS I defined a PS II/PS I electron-transport capacity ratio of 1.6:1.  相似文献

The midpoint potential of the primary electron acceptor of Photosystem I in spinach chloroplasts was titrated using the photooxidation of P700 at −196 °C as an index of the amount of primary acceptor present in the oxidized state. The redox potential of the chloroplast suspension was established by the reducing power of hydrogen gas (mediated by clostridial hydrogenase and 1,1′-trimethylene-2,2′-dipyridylium dibromide) at specific pH values at 25 °C. Samples were frozen to −196 °C and the extent of the photooxidation of P700 was determined from light-minus-dark difference spectra. This titration indicated a midpoint potential of −0.53 V for the primary electron acceptor of Photosystem I.  相似文献

The reaction center chlorophyll of Photosystem I in spinach chloroplasts was highly enriched. Preparations having 5–9 chlorophylls per 1 P700 were obtained by treating the Photosystem I particles prepared by digitonin treatment of chloroplasts with wet diethyl ether. All P700 present in the extracted particles was found to be photoactive, undergoing oxidation upon illumination.  相似文献

Chlorpromazine, phenothiazine and trifluoperazine, known as calmodulin antagonists, inhibit electron transport in Photosystem II of spinach chloroplasts in concentrations from 20–500 μM. The inhibition site is located on the diphenyl carbazide to indophenol pathway in Tris-treated chloroplasts, indicating that water oxidation is not affected by these drugs. Ca2+ ions, bound to chloroplast membranes before the addition of calmodulin antagonists, can protect against inhibition up to 25% of the electron transport rate. In presence of A23187, the Ca2+-specific ionophore, Ca2+ ions provide less protection against inhibition by the 3 calmodulin antagonists used. A possible role of a calmodulin-like protein in spinach chloroplasts is postulated.  相似文献

Stoichiometries of electron transport complexes in spinach chloroplasts   总被引:9,自引:0,他引:9  
The stoichiometric relationship among photosystem II complexes, photosystem I complexes, cytochrome b/f complexes, high-potential cytochrome b-559, and chlorophyll in spinach chloroplasts has been determined. Two features of this data stand out, in contrast to currently proposed stoichiometries in which the ratio of photosystem II to photosystem I is reported to be 2:1 and the chlorophyll to reaction center ratio to be as low as 260:1. Using a variety of techniques it was found that the stoichiometry of photosystem II:photosystem I:cytochrome b/f complex was 1:1:1, within 10%, and that the ratio of total chlorophyll to these components was 600:1, also within 10%. A ratio of two high-potential cytochrome b-559 molecules per 640 chlorophyll, or two molecules per photosystem II reaction center, was found. These ratios were remarkably constant regardless of the time of year or the source of the spinach. The concentration of photosystem II complexes was determined using a pH electrode to measure the flash-induced proton release resulting from water oxidation. The photosystem I reaction center concentration was measured by two different techniques that compared favorably. In the first method a pH electrode was used to measure the amount of flash-induced proton consumption associated with the 3-(3,4-dichlorophenyl)-1,1-dimethylurea-insensitive oxidation of N,N,N',N'- tetramethylphenylenediamine , resulting in the production of hydrogen peroxide. In the second method the amount of P700 oxidized by far-red light was determined using dual-wavelength spectroscopy. The concentration of the cytochrome b/f complex was determined assuming 1 mol of cytochrome f per complex. The concentration of cytochrome f was measured spectroscopically by its light-induced turnover and by chemical difference spectra. The concentration of high-potential cytochrome b-559 was determined by chemical difference spectra. In addition to these studies, the light-induced absorbance change exhibiting a peak at 323 nm that has been attributed to the reduction of the primary quinone acceptor of photosystem II has been investigated. This measurement frequently has been used to quantitate the photosystem II to chlorophyll ratio. However, in view of these results it is argued that this technique significantly overestimates the photosystem II concentration.  相似文献

The Rubisco activase amino acid sequences of spinach and tobacco are 79% identical, yet the tobacco protein does not facilitate the activation of the uncarbamylated, ribulose bisphosphate bound form of spinach ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco, EC 4.1.1.39) and vice versa. In contrast, combinations of the spinach Rubisco activase with Rubisco from non-Solanaceae species and combinations of tobacco Rubisco activase with Rubisco from other Solanaceae species are almost as effective as the analogous combination. To examine the basis of the preference of an activase protein for either Solanaceae or non-Solanaceae Rubisco, several recombinant chimeric proteins were obtained by combining regions from the cDNAs of spinach and tobacco activase and expression in Escherichia coli. The chimeric proteins were analyzed for ATP hydrolysis and ability to activate spinach and tobacco Rubisco. Comparisons of Rubisco preference with composition of the various activase chimeras indicate that the major determinants of Rubisco preference seem to be localized in the carboxyl-terminal region.  相似文献

The electroluminescence induced by external electric fields in blebs prepared from chloroplasts consists of two kinetically different phases, rapid (R) and slow (S), which were shown to be linked to Photosystem I (PS I) and Photosystem II (PS II) activities, respectively (Symons, M., Korenstein, R. and Malkin, S. (1985) Biochim. Biophys. Acta 806, 305–310). In this report we describe conditions involving heat treatment of broken chloroplasts, which make it possible to observe R phase electroluminescence essentially devoid of any contribution by the S phase. This allowed the precise measurement of the emission spectrum of PS I electroluminescence. The emission spectrum of PS II electroluminescence was obtained using regular broken chloroplasts, which show only S-type emission. The latter emission spectrum is identical to the one obtained for ordinary prompt fluorescence, peaking at 685 nm with a bandwidth of about 25 nm. The PS I emission spectrum is symmetric around 705 nm and is much broader, about 60 nm.  相似文献

The effect of monogalactosyl diacylglycerol and digalactosyl diacylglycerol on reconstitution of Photosystem I activity in heptane-extracted and galactolipase-treated spinach chloroplasts was investigated.2. Both galactolipids, in a molar ratio with chlorophyll of 2.5, partially restored Photosystem I activity in heptane-extracted chloroplasts. An addition o saturating amounts of plastocyanin caused complete reactivation of Photosystem I.3. Similarly, with galactolipase-treated chloroplasts, both galactolipids partially restored Photosystem I activity and additional amounts of plastocyanin were required for complete reactivation.4. The action of galactolipids on partial reconstitution of Photosystem I supports the suggestion of their structural role in the restoration of thylakoid membranes.  相似文献

The fluorescence yield of chloroplasts reflects the redox state of the electron acceptor of the Photosystem II reaction center, with increasing yield as the acceptor is reduced. Chemical reductive titrations of fluorescence yield in chloroplasts at room temperature indicate two distinct midpoint potentials, suggesting the possibility of Photosystem II electron acceptor heterogeneity. We have carried out a potentiometric titration of the fluorescence decay kinetics in spinach chloroplasts using a continuous mode-locked dye laser with low-intensity excitation pulses and a picosecond-resolution single-photon timing system. At all potentials the fluorescence decay is best described by three exponential components. As the potential is lowered, the slow phase changes 30-fold in yield with two distinct midpoint potentials, accompanied by a modest (3-fold) increase in the lifetime. The titration curve for the slow component of the fluorescence decay of spinach chloroplasts is best characterized by two single-electron redox reactions with midpoint potentials at pH 8.0 of +119 and ?350 mV, with corresponding relative contributions to the fluorescence yield of 49 and 51%, respectively. There is little change in the fast and middle components of the fluorescence decay. We found that the oxidized form of the redox mediator 2-hydroxy-1,4-naphthoquinone preferentially quenches the fluorescence, causing an anomalous decrease in the apparent midpoint of the high-potential transition. This effect accounts for a significant difference between the midpoint potentials that we observe and some of those previously reported. The selective effect of reduction potentials on particular fluorescence decay components provides useful information about the organization and distribution of the Photosystem II electron acceptor.  相似文献

A model for the photochemical apparatus of photosynthesis is presented which accounts for the fluorescence properties of Photosystem II and Photosystem I as well as energy transfer between the two photosystems. The model was tested by measuring at ?196 °C fluorescence induction curves at 690 and 730 nm in the absence and presence of 5 mM MgCl2 which presumably changes the distribution of excitation energy between the two photosystems. The equations describing the fluorescence properties involve terms for the distribution of absorbed quanta, α, being the fraction distributed to Photosystem I, and β, the fraction to Photosystem II, and a term for the rate constant for energy transfer from Photosystem II to Photosystem I,kT(II→I). The data, analyzed within the context of the model, permit a direct comparison of α andkT(II→I) in the absence (?) and presence (+) of Mg2+:α/?α+= 1.2andk/?T(II→I)k+T(II→I)= 1.9. If the criterion thatα + β = 1 is applied absolute values can be calculated: in the presence of Mg2+,a+ = 0.27 and the yield of energy transfer,φ+T(II→I) varied from 0.065 when the Photosystem II reaction centers were all open to 0.23 when they were closed. In the absence of Mg2+? = 0.32 andφT(II→I) varied from 0.12 to 0.28.The data were also analyzed assuming that two types of energy transfer could be distinguished; a transfer from the light-harvseting chlorophyll of Photosystem II to Photosystem I,kT(II→I), and a transfer from the reaction centers of Photosystem II to Photosystem I,kt(II→I). In that caseα/?α+= 1.3,k/?T(II→I)k+T(II→I)= 1.3 andk/?t(II→I)k+(tII→I)= 3.0. It was concluded, however, that both of these types of energy transfer are different manifestations of a single energy transfer process.  相似文献

Partition in an aqueous Dextran-polyethylene glycol two-phase system has been used for the separation of chloroplast membrane vesicles obtained by press treatment of a grana-enriched fraction after unstacking in a low salt buffer.

The fractions obtained were analysed with respect to chlorophyll, photochemical activities and ultrastructural characteristics. The results reveal that the material partitioning to the Dextran-rich bottom phase consisted of large membrane vesicles possessing mainly Photosystem II properties with very low contribution from Photosystem I. Measurements of the H2O to phenyl-p-benzoquinone and ascorbate-Cl2Ind to NADP+ electron transport rates indicate a ratio of around six between Photosystem II and I.

The total fractionation procedure could be completed within 2–3 h with high recovery of both the Photosystem II water-splitting activity and the Photosystem I reduction of NADP+.

These data demonstrate that press treatment of low-salt destabilized grana membranes yields a population of highly Photosystem-II enriched membrane vesicles which can be discriminated by the phase system. We suggest that such membrane vesicles originate from large regions in the native grana membrane which contain virtually only Photosystem II.  相似文献

Rapid light-induced transients in EPR Signal IIf (F?+) are observed in 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU)-treated, Tris-washed chloroplasts until the state F P680 Q? is reached. In the absence of exogenous redox mediators several flashes are required to saturate this photoinactive state. However, the Signal IIf transient is observed on only the first flash following DCMU addition if an efficient donor to Signal IIf, phenylenediamine or hydroquinone, is present. Complementary polarographic measurements show that under these conditions oxidized phenylenediamine is produced only on the first flash of a series. The DCMU inhibition of Signal IIf can be completely relieved by oxidative titration of a one-electron reductant with E08.0 = +480 mV. At high reduction potentials the decay time of Signal IIf is constant at about 300 ms, whereas in the absence of DCMU the decay time is longer and increases with increasing reduction potential.A model is proposed in which Q?, the reduced Photosystem II primary acceptor, and D, a one-electron 480 mV donor endogenous to the chloroplast suspension, compete in the reduction of Signal IIf (F?+). At high potentials D is oxidized in the dark, and the (Q? + F?+) back reaction regenerates the photoactive F P680 Q state. The electrochemical and kinetic evidence is consistent with the hypothesis that the Signal IIf species, F, is identical with Z, the physiological donor to P680.  相似文献

In spinach chloroplasts it has been shown that (1) the size of the proton gradient under phosphorylating conditions is smaller than under non-phosphorylating conditions; (2) ADP, ATP or Dio-9, added under non-phosphorylating conditions, decrease the rate of electron transport but increase the size of the proton gradient; (3) ADP, ATP or Dio-9 inhibit not only electron transport but also the rate of decay of the proton gradient; (4) the H+/e ratio under non-phosphorylating conditions is 1.0. It is not affected by ADP, ATP or Dio-9.

These results show that protons pass out of the thylakoids at the site of ATP synthesis and that this leakage is inhibited by ADP, ATP or Dio-9, compounds that interact with the site of ATP synthesis. As these compounds do not alter the H+/e ratio the formation of the proton gradient must be an intermediate between electron transport and ATP synthesis. These data are in support of the chemiosmotic theory of coupling of electron transport to ATP synthesis.  相似文献

By density gradient centrifugation of the 80000 × g supernatant of digitonintreated spinach chloroplasts two main green bands and one minor green band were obtained. The purification and properties of the particles present in the main bands, which were shown to be derived from Photosystem I and Photosystem II, have been described previously; those of the particles in the minor fraction will be described in the present paper.

After purification, these particles show Photosystem II activity but are devoid of Photosystem I activity. They have a high chlorophyll a/chlorophyll b ratio and are enriched in β-carotene and cytochrome b559. At liquid nitrogen temperature, photoreduction of C550 and photooxidation of cytochrome-b559 can be observed. At room temperature, cytochrome b559 undergoes slight photooxidation.

These properties indicate that this particle may be the reaction-center complex of Photosystem II. It is suggested that, in vivo, the Photosystem II unit is made up of a reaction-center complex and an accessory complex, the latter being found in one of the main green bands of the density gradient.  相似文献
