In the natural populations +Tüb, +Prov, and +Rov, similar Adh F allele frequencies occur (q F=0.11, 0.18, and 0.08, respectively). However, there is a discrepancy in that the Adh F allele in +Tüb is closely linked to the lethal factor 1(2)Stm, which reduces relative fitness of the F phenotype to zero. In spite of this, polymorphism is maintained also in +Tüb, because the heterozygotes are superior to the homozygous S type (relative fitness=0.88). Under laboratory culture conditions, in +Tüb the relative fitness of the S genotype further decreases to 0.6. After outcrossing the lethal factor, relative fitnesses for S, FS, and F become 0.6, 1, and 0.48, respectively, implying that fitness for S remains the same. Relative values for S, FS, and F in +Prov, not affected by the lethal factor, are calculated by the maximum average fitness method to be 1, 1.2, and 0.2 under the assumption that heterozygous FS are similarly superior to S as in the natural +Tüb population and all allele frequencies found are stable equilibrium values.  相似文献

Lines homozygous for the Adh S and Adh F alleles were extracted from a French and an African natural population of D. melanogaster. The lines from each geographic region were then crossed and the Mendelian F2 constituted the first generation subjected to selection for an increase in adult survival in the presence of 2% ethanol.The responses to selection were similar in the two strains, in spite of their different ethanol tolerance. In less than 10 generations, life span with ethanol increased from about 7 to more than 12 days, with realized heritabilities of 0.14 and 0.18. This extension of longevity was also observed with other concentrations of ethanol but not with water alone. The increase in life span in presence of alcohol appears to result from unknown metabolic processes, which are not obligatorily related to the capacity of the flies to tolerate starvation, nor to their size, their lipid content of their ethanol tolerance. In the two lines, however, the Adh S allele was quickly eliminated, suggesting a selective advantage for the F allele.  相似文献

A further polymorphic rat esterase with broad tissue expression and restricted substrate specificity is described and tentatively called Es-6. Inbred rat strains have either fixed allele Es-6F or fixed allele Es-6S. Es-6 is not linked to the established esterase cluster consisting of the eight esterase loci Es-1, Es-2, Es-3M, Es-4M, Es-4W, Es-5 (=Es-3W), Es-7, and Es-8 in LG V of the rat or to RT1, Gc, c, a, and h. 【背景】西南高山葡萄酒产区的甘孜州产区,具有生产优质葡萄酒的自然禀赋。【目的】研究四川甘孜州葡萄酒产区真核微生物种类多样性、本土酿酒酵母遗传多样性,以及商业酵母对本土酵母多样性的影响。【方法】利用ITS高通量测序技术对赤霞珠接种发酵和自然发酵过程中的微生物进行多样性分析,并利用Interdelta指纹图谱分析法,对经过26S rRNA基因鉴定的野生酿酒酵母基因型进行分类。【结果】ITS测序结果显示,接种发酵和自然发酵各时期均注释到7个科7个属的酵母,通过Interdelta指纹图谱分析发现甘孜州产区的酿酒酵母共有5种基因型。该产区酿酒酵母的6株代表菌株与我国其他产区109株酿酒酵母的进化树分析结果显示,均与来自北京产区的酿酒酵母菌株亲缘关系更近。【结论】甘孜州葡萄酒子产区酵母资源丰富,表现出较高的微生物多样性和中等程度的本土酿酒酵母基因型多样性,为后续优良本土酵母菌株的筛选奠定基础。  相似文献

The mineralization of 14C-phenanthrene, sorbed to porous synthetic amberlite sorbents, i.e., IRC50, XAD7-HP, and XAD2, by three phenanthrene-degrading Mycobacterium soil isolates, i.e., strains VM552, VM531, and VM451 and three phenanthrene-degrading Sphingomonas soil isolates, i.e., strains LH162, EPA505 and LH227, was compared. In P-buffer and in the presence of IRC50, for all strains the maximum rate of mineralization of 14C-phenanthrene was significantly higher (1.1–1.9 ng ml−1 h−1) than the initial abiotic desorption rate (0.2 ng ml−1 h−1), indicating that both Mycobacterium and Sphingomonas utilize sorbed phenanthrene with a higher rate than can be explained by abiotic desorption. Because all Mycobacterium and Sphingomonas strains belonged to different species, it can be suggested that this feature is intrinsic to those genera rather than a specific feature of a particular strain. The final mineralization extent in P-buffer in the presence of IRC50 was about a factor of two higher for the Mycobacterium strains compared to the Sphingomonas strains. Moreover, a significantly higher normalized phenanthrene mineralization ratio in the presence of IRC50 to the control (without IRC50) was found for the Mycobacterium strains compared to the normalized ratio found for the Sphingomonas strains. Addition of minimal nutrients had a more beneficial effect on phenanthrene mineralization by Sphingomonas compared to Mycobacterium, resulting into similar mineralization extents and rates for both types of strains in the presence of IRC50. Our results show that Mycobacterium is better adapted to utilization of sorbed phenanthrene compared to Sphingomonas, especially in nutrient-poor conditions.  相似文献

Keeping Drosophila cultures at 28 C results in elimination of all minor multiple ADH bands, thought to be due to conformational change. Thus in diploid and triploid adults heterozygous for the Adh F and Adh Salleles, relative staining intensities are found for the three bands which were in conformity with the assumption that both alleles are equally expressed. Among all polymorphic strains derived from natural Central European and Mediterranean populations, the strain +Tüb is unique in that its Adh Fallele is closely linked to a new recessive lethal factor, named 1(2)Stm. All Adh F 1/AdhF 1 pupae are unable to emerge, and die. The lethal effect is obvious 50 hr earlier by retarded eye, bristle, and body wall pigmentation. Although all pupae of the phenotype F die, Adh F allele frequency scarcely seems to be lowered in this natural population.  相似文献

A fitness function (function maximized under natural selection) is studied in a population model in which the growth of a population is suppressed by crowding, density-independent continuous mortality (by euryphagous predators) and periodic disturbances. The dynamics of the population density between occurrence of disturbance can be expressed as,dN/dt=(F(N/K)−D)N, whereN is the population density,K is the carrying capacity,D is the density-independent continuous mortality, andF is the growth regulation factor described as a function of crowding (N/K). The period of disturbance isS. The survival rate under disturbance isu. It is concluded that the fitness function is (approximately) a product of competitive ability (C), carrying capacity, and degree of saturation, and is given byCKF −1(D−(lnu)/S). The degree of saturation is the inverse function of regulation factor (F) at the death rate due to predators and disturbance. I assume a population in which density is regulated only through survival. In this case, a low survival rate at the critical age-group means a high value ofCKF −1(D−(lnu)/S). Therefore, the reciprocal of the density-dependent survival rate at critical age-group is a measure of the fitness function. Using this measure, I predict the optimal age (body size) at first reproduction of a species of salamander. I also found that fitness calculated from observed values ofl(x) andm(x) includes a tautology. When the concept of fitness function is compared with the ESS method, the latter is more flexible. However, there is a possibility that an ESS is at the minimum of fitness function.  相似文献

Cysteine-conjugated volatile thiols are powerful aromatic compounds that contribute to the fruity notes of many white wines and especially Sauvignon Blanc. Genetic selection programs of wine yeast starters able to produce more volatile thiols constitute, therefore, an important goal for the wine industry. Recent investigations on yeast metabolism suggested that the ß-lyase Irc7p and the control of its gene expression by nitrogen catabolite repression constitute a rational way for yeast genetic improvement. This work demonstrates that the use of a natural ure2 mutation can be used to design wine starters with an enhanced capacity of volatile thiols production. By applying backcrosses driven by molecular markers, this allelic form was introduced in different starter backgrounds. Our investigations demonstrate that the ure2 inheritance is able to enhance the production of 4MMP (recently renamed 4MSP) and 3MH (recently renamed 3SH). For 4MMP, this effect depends of the presence of the allele IRC7 LT encoding a long form of the Irc7 protein. Moreover, a correlation in between the expression level of this allelic form and 4MMP production was found within industrial starters. All together, these results emphasised the use of molecular breeding for improving quantitative traits of industrial strains without the use of genetically modifying strategies.  相似文献

【背景】商业酵母的使用造成葡萄酒同质化问题严重,发掘优良本土酿酒酵母具有十分重要的意义。【目的】从168株宁夏本土酿酒酵母菌株中筛选出性能优良、具有出色葡萄酒发酵能力的菌株。【方法】基于杜氏管发酵试验和乙醇、高糖等耐受性试验分析产H2S能力及生长曲线测定的方法,筛选出发酵力好、耐受性强、低产H2S的本土酿酒酵母进行赤霞珠葡萄酒发酵试验,测定葡萄酒样基础理化指标、酚类物质和挥发性成分,探究筛选出的酿酒酵母发酵特性。【结果】初步筛选出发酵快速,能适应13%乙醇、350 g/L葡萄糖、250 mg/L SO2、pH 1.0的生存环境且低产H2S的4株本土酿酒酵母YC-E8、QTX-D17、QTX-D7、YQY-E18。菌株YC-E8产甘油能力强,所发酵酒样香气与商业酵母XR、F33最为接近,适用于赤霞珠葡萄酒的发酵。菌株QTX-D17发酵酒样中酒精、单宁、总酚和花色苷含量最高,表现出本土酿酒酵母优良的发酵特性。菌株QTX-D7所发酵酒样香气中乙酸乙酯、辛酸乙酯、1-壬醇等物质含量较高,赋予了葡萄酒香蕉味、苹果味、菠萝味、椰子味等愉悦花果香。【结论】最终筛选出3株优良本土酿酒酵母QTX-D17...  相似文献

The effects of genotype at the alcohol dehydrogenase (ADH) locus on developmental time and fecundity were studied using three strains of Drosophila mercatorum. All three strains had similar genetic backgrounds and two were homozygous for the same electrophoretic allele (F for fast). The third strain was homozygous for the slow allele (S). The life history traits of all possible progeny of these strains were studied. The results showed that for both mean developmental time and pattern of eclosion, strong maternal genotype and background effects were present. Fecundity among genotypes was not different, although there was an effect on the total number of offspring suggesting differences in egg-to-adult survivorship.  相似文献

Summary Properdin factor B phenotypes were determined in 7 Bantu speaking Negroid populations, 1 Indian, and 1 Colored population of South Africa in a total of 1258 individuals. In the Negroid populations allele frequencies were: BfF 0.655, BfS 0.282, BfRARE 0.063, in the Indian population: BfF 0.322, BfS 0.645, BfRARE 0.033, and in the Colored population: BfF 0.513, BfS 0.435, BfRARE 0.052. The Adh1-C mallele and each gene in the Adh1-FC mduplication have been cloned and restriction-mapped. Of the C mallele 6 kb was sequenced. A single amino acid substitution of aspartate for tyrosine at residue 52 accounts for the altered enzymatic properties of the C mprotein. Comparison of the nucleotide sequence to that of Adh1-1F and Adh1-1S shows structural and restriction site polymorphisms in the 3 flanking DNA. C mlacks the insertion sequence present in 1F and 1S and contains a complex sequence composed of two direct repeats and an inverted repeat. The two genes of the duplication allele have similar restriction maps to C mand each other.  相似文献

The Adh and αGpdh allozyme loci (both located on the second chromosome) showed considerable fluctuations in allele frequencies in a seminatural population of Drosophila melanogaster during 1972–97. Both long-term and short-term fluctuations were observed. The short-term fluctuations occurred within almost all years and comparison of allele frequencies between winters and summers showed significantly higher AdhS (P < 0.001) and αGpdhF (P < 0.01) allele frequencies in summers. Frequencies of these alleles were significantly positively correlated with environmental temperature, suggesting the adaptive significance of these allozyme polymorphisms. Frequency changes of the Odh locus (located on the third chromosome) showed no seasonal pattern and were not correlated with environmental temperature. Almost all short-term and long-term increases in AdhS frequency were accompanied by a corresponding decrease in αGpdhS frequency (r = –0.82, P < 0.001) and vice versa. Further analysis showed that gametic disequilibria between the Adh and αGpdh loci, which frequently occurred, were due to the presence of inversion In(2L)t located on the same chromosome arm and In(2L)t frequencies were positively correlated with environmental temperature. Gametic disequilibria between Adh and Odh and between Odh and αGpdh were hardly observed. Because In(2L)t is exclusively associated with the AdhS/αGpdhF allele combination, the observed correlated response in Adh/αGpdh allele frequencies is (at least partly) explained by hitchhiking effects with In(2L)t. This means that the adaptive value of the allozyme polymorphisms has been overestimated by ignoring In(2L)t polymorphism. Fluctuations in Adh allele frequencies are fully explained by selection on In(2L)t polymorphism, whereas we have shown that αGpdh frequency fluctuations are only partly explained by chromosomal hitchhiking, indicating the presence of selective differences among αGpdh genotypes in relation with temperature and independent of In(2L)t. Frequency fluctuations of αGpdh and In(2L)t are consistent with their latitudinal distributions, assuming that temperature is the main environmental factor varying with latitude that causes directly or indirectly these frequency distributions. However, the results of the tropical greenhouse population show no correlation of Adh (independent of In(2L)t) and Odh allele frequencies with environmental temperature, which may indicate that the latitudinal distribution in allele frequencies for these loci is not the result of selection on the F/S polymorphism in a direct way.  相似文献

A quantitative trait locus (QTL) analysis was performed on the size and shape of the mandible in F2 mice between KK-A y and C57BL/6 J strains and the effect of the A y allele on the morphology of the mandible was analyzed. A total of 13 measurements were taken on each right mandible. By means of discriminant and canonical discriminant analyses, KK-A y males and KK males were exactly discriminated from each other. In contrast to its known effects on body weight, the A y allele reduced the overall size of the mandible. QTL analysis of the 13 measurements and on three principal components extracted from these measurements identified multiple QTLs. When F2 a/a and F2 A y /a were analyzed separately, 11 significant main-effect QTLs were identified in F2 a/a, whereas only two QTLs were identified in F2 A y /a. Although four significant interactions were identified, all were in F2 a/a. The A y allele thus made the difference in the size and shape of the mandible between strains obscure. Among mandible QTLs, those on Chrs 5 (Mssq6 and Mssq7) and 15 (Mssq14) were important. Mssq6 had an effect on the height of the posterior mandible. Mssq7 had an effect on mandible length. Mssq14 had an effect on the height of the anterior and posterior mandible. Mssq7 and Mssq14 also had an effect on the overall size. Thus, mandible QTLs have distinct and characteristic sites of action. Therefore, mandible morphology will be determined largely by the combination of these QTLs.  相似文献

The polymorphism of Gc (group-specific components) has been investigated for a series of 3,160 individual samples from 11 Mongoloid populations in Asia and North and South America by isoelectric focusing on polyacrylamide gels. The samples fall into six Gc phenotypes which can be explained by the three common alleles, Gc1F, Gc1S, and Gc2, together with several variant phenotypes explained as the heterozygotes for the three common alleles. The distribution of Gc1F suballele appears to be considerably different from population to population among Mongoloids, ranging from 0.105 (Machiguenga Indans, Peru) to 0.609 (Kadazan, Borneo). A clear geographic cline from Southeast Asia into South America in Gc1F allele was observed in the populations. In general, Gc1F allele frequencies are lower in European populations and higher in African populations. The range of variability in the Gc1F values observed among the Asiatic populations is between the Africans and the Europeans.  相似文献

InBrassica, self-incompatibility (SI) can be overcome by CO2 application, an effective method for obtaining numerous inbred lines for F, commercial seed. We previously reported two different S-alleles ofBrassica campestris, S733 and S734, with extremely different degrees of susceptibility to this gas. In the current study, we raised a cross-population between those two genetic lines, and analyzed their reaction level of self-incompatibility to CO2 (RLSICO2). Here, all 40 of our progeny from the F1 cross-population were susceptible, maintaining high values of RLSICO2. This suggests that the susceptible line, S734, is dominant to the insusceptible line, S733. We also generated an F2 selfing-population of each crossed progeny, S733♀ S734♂ and S733♂ S734♀, to assess the RLSICO2 of each individual. PCR-RFLP analysis was performed to determine the S-genotype of the F2 population. The S734 allele segregated in a theoretical ratio of the dominant trait, and the RLSICO2 was consistent with the dominance relationship. Therefore, we have now demonstrated that high RLSICO2 in β.campestris is controlled by a dominant gene. Both authors contributed equally to this work  相似文献
