Damage to the ozone layer has led to increased levels of ultraviolet radiation at the earth’s surface. Increased ultraviolet radiation can affect macroalgae in many important ways, including reduced growth rate, changes in cell biology and ultrastructure. Kappaphycus alvarezii is a red macroalga of economic interest due to its production of kappa carrageenan. In this study, we examined two strains of K. alvarezii (green and red) exposed to ultraviolet B radiation (UVBR) for 3 h per day during 28 days of cultivation in vitro. UVBR caused changes in the ultrastructure of cortical and subcortical cells, which included increased thickness of the cell wall and plastoglobuli, reduced intracellular spaces, changes in the cell contour, and destruction of chloroplast internal organization. While the green strain exposed to photosynthetically active radiation (PAR) showed growth rates of 6.75% day−1, the red strain grew only 6.35% day−1. Upon exposure to PAR + UV-B, a decreasing trend in growth rates was observed for both strains, with the green strain growing 3.0% day−1 and the red strain growing 2.77% day−1. Significant differences in growth rates between control and UV-B-exposed algae were also found in both strains. Furthermore, compared with control algae, phycobiliprotein contents (phycoerythrin, phycocyanin, and allophycocyanin) were observed to decrease in both strains after PAR + UV-B exposure. However, while the chlorophyll a levels increased in both strains, the green strain showed no significant differences in chlorophyll a levels. Taken together, these findings strongly suggested that UVBR negatively affects the ultrastructure, growth rates, and photosynthetic pigments of intertidal macroalgae and, in the long term, their economic viability.  相似文献

We have studied the effects of nitrate supply under photosynthetic active radiation (PAR) plus ultraviolet radiation (UVR) exposure on photosynthetic pigments (chlorophyll a and carotenoids), photoprotective UV screen mycosporine-like amino acids (MAAs), and photosynthetic parameters, including the maximum quantum yield (F v/F m) and electron transport rate (ETR) on the red agarophyte Gracilaria tenuistipitata. Apical tips of G. tenuistipitata were cultivated under ten different concentrations of NO3 for 7 days. It has been shown that G. tenuistipitata cultured under laboratory conditions has the ability to accumulate high amounts of MAAs following a nitrate concentration-dependent manner under PAR + UVR. Two MAAs were identified, shinorine and porphyra-334. The relative concentration of the first increased under high concentrations of nitrate, while the second one decreased. The presence of antheraxanthin is reported for the first time in this macroalgae, which also contains zeaxanthin, lutein, and β-carotene. The accumulation of pigments, photoprotective compounds, and photosynthetic parameters of G. tenuistipitata is directly related to N availability. All variables decreased under low N supplies and reached constant maximum values with supplements higher than 0.5 mM NO3. Our results suggest a high potential to acclimation and photoprotection against stress factors (including high PAR and UVR) directly related to N availability for G. tenuistipitata.  相似文献

We report the effect of UV-B radiation (0.8 ± 0.1 mW cm−2) and UV-B radiation supplemented with low-intensity PAR (∼80 μmol photons m−2 s−1) on the photosynthesis, photosynthetic pigments, phosphoglycolipids, oxidative damage, enzymatic antioxidants, and UV-absorbing compounds in Phormidium tenue, a marine cyanobacterium. UV-B radiation resulted in a decline in photosynthesis and photosynthetic pigments leading to lower biomass. P. tenue synthesized UV-absorbing compounds like mycosporine-like amino acids (MAAs) and scytonemin in response to UV-B radiation. Quantity of MAAs and scytonemin was higher when UV-B was supplemented with low-level PAR. UV-B treatment also resulted in quantitative changes in phosphoglycolipids of the membrane. The UV-B treatment resulted in a slight increase in the level of peroxidation of cell membrane and very little increase in the activity of superoxide dismutase (SOD). Results indicate that UV-B affected photosynthesis and that the main protective system was the synthesis of MAAs and scytonemin-like compounds rather than antioxidant enzymes such as SOD.  相似文献

Abies faxoniana is a key species in reforestation processes in the southeast of the Qinghai-Tibetan Plateau of China. The changes in growth, photosynthesis and nutrient status of A. faxoniana seedlings exposed to enhanced ultraviolet-B (UV-B), nitrogen supply and their combination were investigated. The experimental design included two levels of UV-B treatments (ambient UV-B, 11.02 KJ m−2 day−1; enhanced UV-B, 14.33 KJ m−2 day−1) and two nitrogen levels (0; 20 g N m−2). The results indicated that: (1) enhanced UV-B significantly caused a marked decline in growth parameters, net photosynthetic rate (Pn), photosynthetic pigments and F v/F m, (2) supplemental nitrogen supply increased the accumulation of total biomass, Pn, photosynthetic pigments and F v/F m under ambient UV-B, whereas supplemental nitrogen supply reduced Pn, and not affect biomass under enhanced UV-B, (3) enhanced UV-B or nitrogen supply changed the concentration of nutrient elements of various organs.  相似文献

The activity of Na+/H+ exchanger to remove toxic Na+ is important for growth of organisms under high salinity. In this study, the halotolerant cyanobacterium Aphanothece halophytica was shown to possess Na+/H+ exchange activity since exogenously added Na+ could dissipate a pre-formed pH gradient, and decrease extracellular pH. Kinetic analysis yielded apparent K m (Na+) and V max of 20.7 ± 3.1 mM and 3,333 ± 370 nmol H+ min−1 mg−1, respectively. For cells grown under salt-stress condition, the apparent K m (Na+) and V max was 18.3 ± 3.5 mM and 3,703 ± 350 nmol H+ min−1 mg−1, respectively. Three cations with decreasing efficiency namely Li+, Ca2+, and K+ were also able to dissipate pH gradient. Only marginal exchange activity was observed for Mg2+. The exchange activity was strongly inhibited by Na+-gradient dissipators, monensin, and sodium ionophore as well as by CCCP, a protonophore. A. halophytica showed high Na+/H+ exchange activity at neutral and alkaline pH up to pH 10. Cells grown at pH 7.6 under high salinity exhibited higher Na+/H+ exchange activity than those grown under low salinity during 15 days of growth suggesting a role of Na+/H+ exchanger for salt tolerance in A. halophytica. Cells grown at alkaline pH of 9.0 also exhibited a progressive increase of Na+/H+ exchange activity during 15 days of growth.  相似文献

Kappaphycus alvarezii is a seaweed of great economic importance for the extraction of kappa carrageenan from its cell walls. The most common strains are dark red, brown, yellow, and different gradations of green. It is known that ultraviolet radiation (UVR) affects macroalgae in many important ways, including reduced growth rate, reduction of primary productivity, and changes in cell biology and ultrastructure. Therefore, we examined the brown strain of K. alvarezii exposed to ultraviolet-B radiaton (UVBR) for 3 h per day during 28 days of cultivation. The control plants showed growth rates of 7.27% d−1, while plants exposed to UVBR grew only 4.0% d−1. Significant differences in growth rates and in phycobiliproteins between control and exposed plants were also found. Compared with control plants, phycobiliprotein contents were observed to decrease after UV-B exposure. Furthermore, the chlorophyll a (Chl a) contents decreased and showed significant differences. UVBR also caused changes in the ultrastructure of cortical and subcortical cells, which included increased thickness of the cell wall and number of plastoglobuli, reduced intracellular spaces, changes in the cell contour, and destruction of chloroplast internal organization. Reaction with Toluidine Blue showed an increase in the thickness of the cell wall, and Periodic Acid-Schiff stain showed a decrease in the number of starch grains. By the significant changes in growth rates, photosynthetic contents and ultrastructual changes observed, it is clear that UVBR negatively affects intertidal macroalgae and, by extension, their economic viability.  相似文献

The long-term performance and stability of Pseudomonas putida mt-2 cultures, a toluene-sensitive strain harboring the genes responsible for toluene biodegradation in the archetypal plasmid pWW0, was investigated in a chemostat bioreactor functioning under real case operating conditions. The process was operated at a dilution rate of 0.1 h−1 under toluene loading rates of 259 ± 23 and 801 ± 78 g m−3 h−1 (inlet toluene concentrations of 3.5 and 10.9 g m−3, respectively). Despite the deleterious effects of toluene and its degradation intermediates, the phenotype of this sensitive P. putida culture rapidly recovered from a 95% Tol population at day 4 to approx. 100% Tol+ cells from day 13 onward, sustaining elimination capacities of 232 ± 10 g m−3 h−1 at 3.5 g Tol m−3 and 377 ± 13 g m−3 h−1 at 10.9 g Tol m−3, which were comparable to those achieved by highly tolerant strains such as P. putida DOT T1E and P. putida F1 under identical experimental conditions. Only one type of Tol variant, harboring a TOL-like plasmid with a 38.5 kb deletion (containing the upper and meta operons for toluene biodegradation), was identified.  相似文献

Combined effect of light intensity and glucose concentration on Arthrospira platensis growth and photosynthetic response was evaluated using a 32 factorial design. This design was carried out with light levels of 50, 100, and 150 μmol photons m−2 s−1 and glucose concentrations of 0.5, 1.5, and 2.5 g L−1. Results from the response surface methodology were that the highest level of light intensity and glucose concentration improved biomass (1.33 g L−1), maximum specific growth rate (0.49 day−1), and net photosynthetic rate (139.89 μmol O2 mg Chl−1 h−1). Furthermore, the interaction of both factors showed that at low light, glucose had a low effect on maximum biomass and maximal net photosynthetic rate. However, at the highest light levels, the effect of glucose was more sensitive and the increase of glucose concentration increased the levels of all responses. The rates of the instantaneous relative growth, net photosynthesis, and dark respiration of growth cultures showed two different phases in mixotrophic condition. The first was distinguished by the preponderance of the photoautotrophic mode; the second was based mainly on photoheterotrophy.  相似文献

The zooxanthellate octocoral Sinularia flexibilis is a producer of potential pharmaceutically important metabolites such as antimicrobial and cytotoxic substances. Controlled rearing of the coral, as an alternative for commercial exploitation of these compounds, requires the study of species-specific growth requirements. In this study, phototrophic vs. heterotrophic daily energy demands of S. flexibilis was investigated through light and Artemia feeding trials in the laboratory. Rate of photosynthetic oxygen by zooxanthellae in light (≈200 μmol quanta m−2 s−1) was measured for the coral colonies with and without feeding on Artemia nauplii. Respiratory oxygen was measured in the dark, again with and without Artemia nauplii. Photosynthesis–irradiance curve at light intensities of 0, 50, 100, 200, and 400 μmol quanta m−2 s−1 showed an increase in photosynthetic oxygen production up to a light intensity between 100 and 200 μmol quanta m−2 s−1. The photosynthesis to respiration ratio (P/R > 1) confirmed phototrophy of S. flexibilis. Both fed and non-fed colonies in the light showed high carbon contribution by zooxanthellae to animal (host) respiration values of 111–127%. Carbon energy equivalents allocated to the coral growth averaged 6–12% of total photosynthesis energy (mg C g 1 buoyant weight day 1) and about 0.02% of the total daily radiant energy. “Light utilization efficiency (ε)” estimated an average ε value of 75% 12 h 1 for coral practical energetics. This study shows that besides a fundamental role of phototrophy vs. heterotrophy in daily energy budget of S. flexibilis, an efficient fraction of irradiance is converted to useable energy.  相似文献

Bifidobacterium longum grew at 65 L pilot scale of the membrane bioreactor (MBR), externally fitted with ceramic membrane (0.7 m2). Cell mass at the MBR reached 22.18 g L−1 as dry cell weight in 12 h, which is 8.44 times higher than cell mass attained at the vial culture. The growth rate in the vial culture was μ = 0.385 h and at the batch culture was μ = 1.13 h in the exponential period and μ = 0.31 h−1 in the stationary period. In the fed-batch mode was μ = 1.102 h−1 for 6 h with inoculation and declined to μ = 0.456 h−1 with feeding of feed medium. The growth rate at the MBR was μ = 0.134 h−1. The number of viable cells was 6.01 × 1012 cfu L−1 at the batch culture, but increased to 1.15 × 1014 cfu L−1 at the MBR culture. The specific growth rate of viable cell number (colony-forming units per liter, per hour) improved by 6.01 times from the batch to the MBR culture. The wall shear stress mainly generated by the pump, and the membrane incorporated into the MBR was controlled during the cultivation at the MBR. The viability of B. longum declined to under 10% in the first 2 weeks of the 4-week stability test (40°C) as B. longum was exposed to over wall shear stress 713 Pa, but the viability improved to 30–40% in wall shear stress of 260 Pa or STR culture. The loss in the cell viability can be saved by managing with wall shear stress during the cultivation at the MBR.  相似文献

To improve the production of Kappaphycus plantlets in tissue culture, optimum media concentrations of an Ascophyllum nodosum extract (Acadian Marine Plant Extract Powder, AMPEP), plant growth regulators (PGR), pH–temperature combinations, and explant density were determined. Kappaphycus alvarezii var. tambalang purple (PUR), kapilaran brown (KAP), vanguard brown (VAN), adik-adik (AA), tungawan green (TGR), and K. striatum var. sacol green (GS) were used as explants. Based on the shortest period for shoot emergence and the economical use of AMPEP, the optimum enriched media was 3.0 mg L−1 AMPEP and 0.1 mg L−1 AMPEP + PGR 1 mg L−1 each phenylacetic acid (PAA) and zeatin for PUR, 1.0 mg L−1 AMPEP + PGR for KAP and GS, 0.1 mg L−1 AMPEP + PGR for VAN, and 3.0 mg L−1 AMPEP and 0.001 mg L−1 AMPEP + PGR for AA and TGR. Results showed that the addition of PGR to low concentrations of AMPEP hastened shoot formation. pH–temperature combinations for the most rapid shoot formation were determined for the brown (KAP) and purple (PUR) color morphotypes of K. alvarezii var. tambalang and the green morphotype of K. striatum var. sacol (GS) cultured in 1.0 mg L−1 AMPEP + PGR. The brown morphotype produced the most number of shoots at pH 7.7 at 20°C after as little as 20 days. Purple K. alvarezii showed an increased shoot formation at pH 6.7 at 25°C and the green K. striatum morphotype at pH 8.7 at 25°C. The optimum number of explants added to the culture media was also determined for tungawan green (TGR), brown (KAP), and tambalang purple (PUR) varieties of K. alvarezii in 1.0 mg L−1 AMPEP + PGR. The number of explants and the volume of the culture media combination were also tested. The highest average number of shoots formed occurred in two explants:1 mL culture media (2:1) for KAP and PUR (35.00% and 16.67%, respectively) and 1 explant: 2 mL culture media for the TGR (100.00%) with a range of 0.5–3.0 mm shoot length after 40 days in culture. The earliest shoot formation was observed after 21 days for the brown and 9 days for both the green and purple color morphotypes of Kappaphycus, in all densities investigated. This indicated that within the range tested, the density of explants did not have a significant effect on the rate of shoot formation but did influence the average number generated from the culture. The rate of production of new and improved Kappaphycus explants for a commercial nursery stock was improved through the use of AMPEP with optimized culture media pH, temperature, and density conditions.  相似文献

Three varieties of Kappaphycus alvarezii (Kapilaran, KAP), Tambalang purple (PUR), Adik-adik (AA), and one variety of Kappaphycus striatum var. sacol (green sacol (GS) were used to determine the efficiency of Acadian marine plant extract powder (AMPEP) as a culture medium at different concentrations, for the regeneration of young plants of Kappaphycus varieties, using tissue culture techniques for the production of seed stock for nursery and outplanting purposes for the commercial cultivation of carrageenophytes. A shorter duration for shoot formation was observed when the explant was treated with AMPEP + Plant Growth Regulator (PGR = PAA + zeatin at 1 mg L−1) compared to AMPEP when used singly. However, four explants responded differently to the number of days required for shoot formation. The KAP variety took 46 days to form shoots at 3–4 mg L−1 AMPEP + PGR; while PUR required 21 days at 3–5 mg L−1 AMPEP and 3–4 mg L−1 AMPEP + PGR. AA required 17 days at 3–5 mg L−1 AMPEP and AMPEP + PGR; and GS 25 days at 1 mg L−1 AMPEP + PGR. It was observed that among the four explants used, PUR and AA initiated shoot formation with the use of AMPEP only at higher concentrations (3–5 mg L−1) after a shorter period. Only PUR responded positively to ESS/2 for shoot initiation. The use of AMPEP alone and/or in combination with PGR as a culture medium in the propagation of microplantlets using tissue culture technique is highly encouraging.  相似文献

The tissue distribution and ontogeny of Na+/K+-ATPase has been examined as an indicator for ion-regulatory epithelia in whole animal sections of embryos and hatchlings of two cephalopod species: the squid Loligo vulgaris and the cuttlefish Sepia officinalis. This is the first report of the immunohistochemical localization of cephalopod Na+/K+-ATPase with the polyclonal antibody α (H-300) raised against the human α1-subunit of Na+/K+-ATPase. Na+/K+-ATPase immunoreactivity was observed in several tissues (gills, pancreatic appendages, nerves), exclusively located in baso-lateral membranes lining blood sinuses. Furthermore, large single cells in the gill of adult L. vulgaris specimens closely resembled Na+/K+-ATPase-rich cells described in fish. Immunohistochemical observations indicated that the amount and distribution of Na+/K+-ATPase in late cuttlefish embryos was similar to that found in juvenile and adult stages. The ion-regulatory epithelia (e.g., gills, excretory organs) of the squid embryos and paralarvae exhibited less differentiation than adults. Na+/K+-ATPase activities for whole animals were higher in hatchlings of S. officinalis (157.0 ± 32.4 μmol gFM−1 h−1) than in those of L. vulgaris (31.8 ± 3.3 μmol gFM−1 h−1). S. officinalis gills and pancreatic appendages achieved activities of 94.8 ± 18.5 and 421.8 ± 102.3 μmolATP gFM−1 h−1, respectively. High concentrations of Na+/K+-ATPase in late cephalopod embryos might be important in coping with the challenging abiotic conditions (low pH, high pCO2) that these organisms encounter inside their eggs. Our results also suggest a higher sensitivity of squid vs. cuttlefish embryos to environmental acid-base disturbances.  相似文献

Recently recruited urchins from the same brood, but with different initial sizes, may respond differently to similar environmental factors. The aim of this study was to assess and compare the effects of starvation and diet on the survival, growth rates in size and weight, and gonad index among small and large sub-adult purple sea urchins, Strongylocentrotus purpuratus. Small urchins ranged from 7.3 to 7.8 mm and large urchins from 11.8 to 14.1 mm (test diameters). Two independent experiments were performed. In the first experiment, sea urchins were fed during 22 weeks on Egregia menziesii (ad libitum) and for only 1 day month−1 (starved condition). Feeding regime significantly affected survival, somatic growth rate in size and weight, and gonad index, with higher means in the ad libitum treatments than in starving conditions. A recurrent cannibalism event by conspecifics occurred in small sea urchins under starving conditions. In the second experiment, sea urchins were fed during 13 weeks ad libitum with four diets: kelp (E. menziesii), coralline algae (Bossiella orbigniana), eelgrass (Phyllospadix scouleri) and a mixed diet of the three species. Survival was not affected by diet or urchin size, but diet significantly affected somatic growth rate in size and weight and gonad index. Kelp promoted the highest growth rate (2.23 ± 0.21 mm month−1), the mixed diet produced an intermediate growth (1.26 ± 0.21 mm month−1), while the lowest values corresponded to coralline algae and the eelgrass (0.30 ± 0.12 and 0.10 ± 0.03 mm month−1, respectively, means ± SE). The mean growth rate of small urchins (averaging all diets) was higher than in large specimens (1.17 ± 0.37 and 0.77 ± 0.28 mm month−1, respectively).  相似文献

As a source of agar, the red macroalga Gelidium floridanum is a seaweed of great economic importance. However, it grows in a region exposed to high ultraviolet B radiation (UVBR). Therefore, to study the in vitro effect of UVBR on this plant, apical segments of G. floridanum were cultivated and exposed to photosynthetically active radiation (PAR) at 80?μmol photons m?2?s?1 and PAR + UVBR at 1.6?W?m?2 at 3?h per day for 7?days. The samples were processed for electron microscopy, and agar yield, growth rates, mitochondrial activity, protein levels, chlorophyll a, phycobiliproteins, carotenoids and phenolic compounds, and photosynthetic performance were examined. After 7?days of exposure to PAR + UVBR, G. floridanum experienced ultrastructural damage that was primarily observed in the internal organization of chloroplasts, increased cell wall thickness, as well as increased volume of plastoglobuli and free ribosomes. Moreover, this exposure might have caused photodamage and photoinhibition of photosynthetic pigments (chlorophyll a and phycobiliproteins), leading to a decrease in photosynthetic efficiency, relative electron transport rate, and maximum photosynthetic rate. These phenomena were matched with a corresponding decrease in growth rates and depigmentation, combined with partial necrosis of the apical segments exposed to PAR + UVBR. Additionally, the UVBR-induced damage elicited a marked cellular antioxidant response, possibly as a consequence of free radical generation.  相似文献

The biodegradation potential of an innovative enclosed tubular biofilm photobioreactor inoculated with a Chlorella sorokiniana strain and an acclimated activated sludge consortium was evaluated under continuous illumination and increasing pretreated (centrifuged) swine slurry loading rates. This photobioreactor configuration provided simultaneous and efficient carbon, nitrogen, and phosphorous treatment in a single-stage process at sustained nitrogen and phosphorous removals efficiencies ranging from 94% to 100% and 70–90%, respectively. Maximum total organic carbon (TOC), NH4 +, and PO4 3− removal rates of 80 ± 5 g C mr −3 day−1, 89 ± 5 g N mr −3 day−1, and 13 ± 3 g P mr −3 day−1, respectively, were recorded at the highest swine slurry loadings (TOC of 1,247 ± 62 mg L−1, N–NH4 + of 656 ± 37 mg L−1, P–PO4 3+ of 117 ± 19 mg L−1, and 7 days of hydraulic retention time). The unusual substrates diffusional pathways established within the phototrophic biofilm (photosynthetic O2 and TOC/NH4 + diffusing from opposite sides of the biofilm) allowed both the occurrence of a simultaneous denitrification/nitrification process at the highest swine slurry loading rate and the protection of microalgae from any potential inhibitory effect mediated by the combination of high pH and high NH3 concentrations. In addition, this biofilm-based photobioreactor supported efficient biomass retention (>92% of the biomass generated during the pretreated swine slurry biodegradation).  相似文献

A β-1,4-endoglucanase (Cel5A) was cloned from the genomic DNA of saccharolytic thermophilic eubacterium Thermoanaerobacter tengcongensis MB4 and functionally expressed in Escherichia coli. Substrate specificity analysis revealed that Cel5A cleaves specifically the β-1,4-glycosidic linkage in cellulose with high activity (294 U mg−1; carboxymethyl cellulose sodium (CMC)). On CMC, kinetics of Cel5A was determined (K m 1.39 ± 0.12 g l−1; k cat/K m 1.41 ± 0.13 g−1 s−1). Cel5A displays an activity optimum between 75 and 80 °C. Residues Glu187 and Glu289 were identified as key catalytic amino acids by sequence alignment. Interestingly, derived from a non-halophilic bacterium, Cel5A exhibits high residual activities in molar concentration of NaCl (3 M, 49.3%) and KCl (4 M, 48.6%). In 1 M NaCl, 82% of Cel5A activity is retained after 24 h incubation. Molecular Dynamics studies performed at 0 and 3 M NaCl, correlate the Cel5A stability to the formation of R-COO···Na+ ···OOC-R salt bridges within the Cel5A tertiary structure, while activity possibly relates to the number of Na+ ions trapped into the negatively charged active site, involving a competition mechanism between substrate and Na+. Additionally, Cel5A is remarkably resistant in ionic liquids 1-butyl-3-methyllimidazolium chloride (1 M, 54.4%) and 1-allyl-3-methylimidazolium chloride (1 M, 65.1%) which are promising solvents for cellulose degradation and making Cel5A an attractive candidate for industrial applications.  相似文献

Controlled cultivation of marine macroalgal biomass such as Ulva species, notably Ulva lactuca, is currently studied for production of biofuels or functional food ingredients. In a eutrophic environment, this macrophyte is exposed to varying types of nutrient supply, including different and fluctuating levels of nitrogen sources. Our understanding of the influences of this varying condition on the uptake and growth responses of U. lactuca is limited. In this present work, we examined the growth response of U. lactuca exposed to different sources of nitrogen (NH4+; NO3; and the combination NH4NO3) by using photo-scanning technology for monitoring the growth kinetics of U. lactuca. The images revealed differential increases of the surface area of U. lactuca disks with time in response to different N-nutrient enrichments. The results showed a favorable growth response to ammonium as the nitrogen source. The NH4Cl and NaNO3 rich media (50 μM of N) accelerated U. lactuca growth to a maximum specific growth rate of 16.4 ± 0.18% day−1 and 9.4 ± 0.72% day−1, respectively. The highest biomass production rate obtained was 22.5 ± 0.24 mg DW m−2·day−1. The presence of ammonium apparently discriminated the nitrate uptake by U. lactuca when exposed to NH4NO3. Apart from showing the significant differential growth response of U. lactuca to different nitrogen sources, the work exhibits the applicability of a photo-scanning approach for acquiring precise quantitative growth data for U. lactuca as exemplified by assessment of the growth response to two different N-sources.  相似文献

The recombinant Pichia pastoris harboring an improved methionine adenosyltransferase (MAT) shuffled gene was employed to biosynthesize S-adenosyl-l-methionine (SAM). Two l-methionine (l-Met) addition strategies were used to supply the precursor: the batch addition strategy (l-Met was added separately at three time points) and the continuous feeding strategies (l-Met was fed continuously at the rate of 0.1, 0.2, and 0.5 g l−1 h−1, respectively). SAM accumulation, l-Met conversion rate, and SAM productivity with the continuous feeding strategies were all improved over the batch addition strategy, which reached 8.46 ± 0.31 g l−1, 41.7 ± 1.4%, and 0.18 ± 0.01 g l−1 h−1 with the best continuous feeding strategy (0.2 g l−1 h−1), respectively. The bottleneck for SAM production with the low l-Met feeding rate (0.1 g L−1 h−1) was the insufficient l-Met supply. The analysis of the key enzyme activities indicated that the tricarboxylic acid cycle and glycolytic pathway were reduced with the increasing l-Met feeding rate, which decreased the adenosine triphosphate (ATP) synthesis. The MAT activity also decreased as the l-Met feeding rate rose. The reduced ATP synthesis and MAT activity were probably the reason for the low SAM accumulation when the l-Met feeding rate reached 0.5 g l−1 h−1.  相似文献

Hypokinesia (HK) induces electrolyte losses in electrolyte-deficient tissue, yet the mechanisms of electrolyte losses in electrolyte-deficient tissue remain unknown. Mechanisms of electrolyte deposition could be involved. To determine the effect of prolonged HK on potassium (K+) deposition were measured muscle K+ content and K+ losses. Studies were conducted on 20 physically healthy male volunteers during 30 days pre-experimental period and 364 days experimental period. Subjects were equally divided into two groups: control subjects (CS) and experimental subjects (ES). The CS group was run average distances of 9.8 ± 1.7 km day−1 and the ES group was walked average distances of 2.7 ± 0.6 km day−1. Muscle K+ content decreased (p < 0.05) and plasma K+ concentration, and K+ losses in urine and feces increased (p < 0.05) in the ES group compared to their pre-experimental level and the values in their respective CS group. Muscle K+ content, plasma K+ level, and urine and fecal K+ losses did not show any changes in the CS group compared to their pre-experimental values. The conclusion was that K+ losses in K+-deficient muscle of healthy subjects could have been attributable to the less efficient K+ deposition inherently to prolonged HK.  相似文献
