Abstract Lysogeny is widespread in the lactic acid bacteria. The majority of lysogens can be induced by UV irradiation or treatment with mitomycin C, but indicator strains which allow lytic growth of the induced phage are often not easy to identify. A few temperate phages have been shown to transduce chromosomal and/or plasmid markers. Information about the molecular biology of the temperate phages from lactic acid bacteria is sparse and needs significant supplementation in order that these potentially valuable phages might be utilized more efficiently as tools for improving existing starter strains in dairy fermentations.  相似文献

考察谷氛酸产生菌在缺氧条件下积累L-乳酸和琥珀酸的情况.结果表明:在缺氧条件下,嗜乙酰乙酸棒杆菌(Corynebacterium acetoacidophilum)ATCC 13870积累有机酸的浓度随菌体密度的增大而增加,其中琥珀酸和乳酸积累的最适pH分别为7.5和8.0,最高质量浓度分别为22.5和60g/L.碳酸盐是影响产酸与有机酸分布的主要因素.比较ATCC 13870在NaHC03浓度为40和400mmol/L时的代谢通量,发现后者合成琥珀酸的代谢通量比前者提高了214.1%,合成乳酸的代谢通量降低了61.8%,说明PEP节点处的代谢通量分配明显受NaHCO3浓度的影响,而PYR节点受环境因素的影响不明显.  相似文献

The bacterial secretion of glutamate was studied through plasma membrane fluidity, measured by anisotropy using the fluorophore TMA-DPH incorporated in the lipid part of the cell membrane. Cells of Brevibacterium sp. ATCC 13869 (wild type) were switched from the biotin-limited, producing state to the biotin-supplemented, non-producing state, and back. The following conclusions could be drawn: 1. It was not possible to detect any change in anisotropy by switching the cells from biotin-limited biotin-supplemented, as well as from biotin-supplemented, to biotin-limited, media. 2. The anisotropy value in the glutamic acid fermentation remains constant during the lag, exponential, growth, production and stationary phases. 3. The treatment of cells with a neutral synthetic polyester of ethylene-and propyleneoxide with soya oil-fatty acids increased the anisotropy values, indicating incorporation of the surfactant. 4. Glutamate secretion is not coupled with membrane fluidity, so a leak providing a general fluidization of the membrane could not be detected.  相似文献

Summary Rod-shaped inclusions in the nuclei and/or cytoplasm of strains ofEuglena spirogyra, Lepocinclis ovum, Strombomonas conspersa andTrachelomonas oblonga var.punctata have been identified as living populations of bacteria by reference to size, shape and ultrastructure. Most strains contain 20–200 bacteria per nucleus. Each bacterial cell is surrounded by a mucilaginous sheath and consists of a double-membraned envelope, dense cytoplasm with small ribosomes, and a filamentous DNA-containing centre. The bacteria multiply to keep pace with euglenoid division but there is no evidence that the euglenoid cells benefit from or are harmed by the association. Attempts to transfer the endonuclear bacteria to uninfected strains and species have been unsuccessful, as have attempts to isolate the bacterium into pure culture. The infected strain ofEuglena spirogyra cannot fix nitrogen. For the second-generation asymmetric synthesis of the trans-tris(homoglutamic) acids via Strecker reaction of chiral ketimines, the cyanide addition as the key stereodifferentiating step produces mixtures of diastereomeric alpha-amino nitrile esters the composition of which is independent of the reaction temperature and the type of the solvent, respectively. The subsequent hydrolysis is exclusively achieved with concentrated H(2)SO(4) yielding diastereomeric mixtures of three secondary alpha-amino alpha-carbamoyl-gamma-esters and two diastereomeric cis-fused angular alpha-carbamoyl gamma-lactams as bicyclic glutamic acid derivatives, gained from in situ stereomer differentiating cyclisation of the secondary cis-alpha-amino alpha-carbamoyl-gamma-esters. Separation was achieved by CC. The pure secondary trans-alpha-amino alpha-carbamoyl-gamma-esters cyclise on heating and treatment with concentrated H(2)SO(4), respectively, to diastereomeric cis-fused angular secondary alpha-amino imides. Their hydrogenolysis led to the enantiomeric cis-fused angular primary alpha-amino imides. The configuration of all compounds was completely established by NMR methods, CD-spectra, and by X-ray analyses of the (alphaR,1R,5R)-1-carbamoyl-2-(1-phenylethyl)-2-azabicyclo[3.3.0]octan-3-one and of the trans-alphaS,1S,2R-2-ethoxycarbonylmethyl-1-(1-phenylethylamino)cyclopentanecarboxamide.  相似文献
