Sloths are unusual mobile ecosystems, containing a high diversity of epibionts living and growing in their fur as they climb slowly through the canopies of tropical forests. These epibionts include poorly studied algae, arthropods, fungi, and bacteria, making sloths likely reservoirs of unexplored biodiversity. This review aims to identify gaps and eliminate misconceptions in our knowledge of sloths and their epibionts, and to identify key questions to stimulate future research into the functions and roles of sloths within a broader ecological and evolutionary context. This review also seeks to position the sloth fur ecosystem as a model for addressing fundamental questions in metacommunity and movement ecology. The conceptual and evidence-based foundation of this review aims to serve as a guide for future hypothesis-driven research into sloths, their microbiota, sloth health and conservation, and the coevolution of symbioses in general.

Background

Sloths are slow-moving arboreal mammals inhabiting tropical rainforests in Central and South America. The six living species of sloths are occasionally reported to display a greenish discoloration of their pelage. Trichophilus welckeri, a green algal species first described more than a century ago, is widely believed to discolor the animals fur and provide the sloth with effective camouflage. However, this phenomenon has not been explored in any detail and there is little evidence to substantiate this widely held opinion.

We studied the diet, activity budget, vertical ranging, and postural behaviour in relation to weather of the three-toed sloth (Bradypus variegatus flaccidus) in disturbed montane forest remnants (1150 m asl) in northern Venezuela. Sloths spent most (72.9%) of their time resting and had a nearly exclusive (99.4%) leaf diet. While resting they assumed a sitting – not hanging – posture mostly (90.2% of observations). Species of three families, Clethraceae, Cecropiaceae, and Clusiaceae accounted for 77% of feeding records. Young leaves (67.2%) accounted for most of the leaf diet. Activity and posture were dependent on weather conditions. Sloths fed more often during mid-day hours and tended to rest more at dawn and dusk. In northern Venezuela sloths tended to use more frequently the upper strata of the canopy, while in warmer lowland sites they use intermediate levels more often. They adopted postures that maximized exposure of ventral surfaces to incident solar radiation when sunny, but minimized their surface area by huddling when cloudy, foggy or rainy. We propose that sunning behaviour of sloths may speed up their fermentation rate, and ultimately, might have been an important selective factor in the evolution of derived upside-down posture of sloths.

The spatial ecology of sloths was studied in an agricultural landscape in Limón Province, Costa Rica. Two sloth species, the three-toed sloth (Bradypus variegatus) and the two-toed sloth (Choloepus hoffmanni), actively used and traveled through a cacao agroforest and its contiguous living fence rows and riparian forests. This agroecosystem was embedded in an agricultural landscape dominated by banana and pineapple plantations and pastures with dispersed trees. The two-toed sloth (C. hoffmanni) was found in 101 tree species and used 34 for food; the three-toed sloth (B. variegatus) was found in 71 tree species and used 15 for food. Choice of preferred species differed between the two sloth species. Trees commonly used by sloths for food and/or refuge in the cacao agroforest included Erythrina poeppigiana, Cecropia obtusifolia, Leucaena leucocephala; in the living fence rows, Cordia alliodora, Erythrina poeppigiana, Ocotea sinuata and Trophis racemosa; in the riparian forests, Coussapoa villosa, Cecropia obtusifolia, Hura crepitans, Pterocarpus officinalis and Spondias mombin; and in the pastures with dispersed trees, Cordia alliodora, Coussapoa villosa, Erythrina poeppigiana, Ocotea sinuata and Hura crepitans. This study demonstrates the importance of the cacao agroforest as well as arboreal elements in other land uses in providing resources for sloth conservation in a larger agricultural landscape.

Sloths have a dense coat on which insects, algae and fungi coexist in a symbiotic relationship. This complex ecosystem requires different levels of controls; however, most of these mechanisms remain unknown. We investigated the bacterial communities inhabiting the hair of two- (Choloepus Hoffmanni) and three-toed (Bradypus variegatus) sloths and evaluated their potential for producing antibiotic molecules capable of exerting control over the hair microbiota. The analysis of 16S rRNA amplicon sequence variants revealed that the communities in both host species are dominated by Actinobacteriota and Firmicutes. The most abundant genera were Brevibacterium, Kocuria/Rothia, Staphylococcus, Rubrobacter, Nesterenkonia and Janibacter. Furthermore, we isolated nine strains of Brevibacterium and Rothia capable of producing substances that inhibited the growth of common mammalian pathogens. The analysis of the biosynthetic gene clusters of these nine isolates suggests that the pathogen-inhibitory activity could be mediated by the presence of siderophores, terpenes, beta-lactones, Type III polyketide synthases, ribosomally synthesized and post-translationally modified peptides, non-alpha poly-amino acids like e-Polylysine, ectoine or non-ribosomal peptides. Our data suggest that Micrococcales that inhabit sloth hair could have a role in controlling microbial populations in that habitat, improving our understanding of this highly complex ecosystem.

Sloths are morphologically specialized in suspensory quadrupedal locomotion and posture. During steady-state locomotion they utilize a trot-like footfall sequence. Contrasting the growing amount of published accounts of the functional morphology and kinematics of sloth locomotion, no study concerned with the dynamics of their quadrupedal suspensory locomotion has been conducted. Brachiating primates have been shown to travel at low mechanical costs using pendular mechanics, but this is associated with considerable dynamic forces exerted onto the support. To test whether sloth locomotion can be described by simple connected pendulum mechanics, we analyzed the dynamics of sloth locomotion with use of a mechanical segment link model. The model integrates the body segment parameters and is driven by kinematic data with both segment parameters and kinematic data obtained from the same sloth individual. No simple pendular mechanics were present. We then used the model to carry out an inverse dynamic analysis. The analysis allowed us to estimate net limb joint torques and substrate reaction forces during the contact phases. Predominant flexing limb joint torque profiles in the shoulder, elbow, hip, and knee are in stark contrast to published dominant extensor torques in the limb joints of pronograde quadrupedal mammals. This dissimilarity likely reflects the inverse orientation of the sloth towards the gravity vector. Nevertheless, scapular pivot and shoulder seem to provide the strongest torque for progression as expected based on unchanged basic kinematic pattern previously described. Our model predicts that sloths actively reduce the dynamical forces and moments that are transmitted onto the support. We conclude that these findings reflect the need to reduce the risk of breaking supports because in this case sloths would likely be unable to react quickly enough to prevent potentially lethal falls. To achieve this, sloths seem to avoid the dynamical consequences of effective pendular mechanics.

The extraordinary biological diversity of tropical forests harbors a rich chemical diversity with enormous potential as a source of novel bioactive compounds. Of particular interest are new environments for microbial discovery. Sloths – arboreal mammals commonly found in the lowland forests of Panama – carry a wide variety of micro- and macro-organisms on their coarse outer hair. Here we report for the first time the isolation of diverse and bioactive strains of fungi from sloth hair, and their taxonomic placement. Eighty-four isolates of fungi were obtained in culture from the surface of hair that was collected from living three-toed sloths (Bradypus variegatus, Bradypodidae) in Soberanía National Park, Republic of Panama. Phylogenetic analyses revealed a diverse group of Ascomycota belonging to 28 distinct operational taxonomic units (OTUs), several of which are divergent from previously known taxa. Seventy-four isolates were cultivated in liquid broth and crude extracts were tested for bioactivity in vitro. We found a broad range of activities against strains of the parasites that cause malaria (Plasmodium falciparum) and Chagas disease (Trypanosoma cruzi), and against the human breast cancer cell line MCF-7. Fifty fungal extracts were tested for antibacterial activity in a new antibiotic profile screen called BioMAP; of these, 20 were active against at least one bacterial strain, and one had an unusual pattern of bioactivity against Gram-negative bacteria that suggests a potentially new mode of action. Together our results reveal the importance of exploring novel environments for bioactive fungi, and demonstrate for the first time the taxonomic composition and bioactivity of fungi from sloth hair.

Recent phylogenetic analyses imply a distant relationship and long separated evolution of two-toed sloths (Choloepus) and three-toed sloths (Bradypus). No known fossil sloth is interpreted to have been suspensory. As a consequence, the suspensory posture and locomotion of the extant genera likely evolved convergently in both lineages, forming a new framework for the analysis of functional aspects of the locomotor apparatus of extant tree sloths. The suspensory posture and locomotion has altered functional demands from the phylogenetically plesiomorphic non-suspensory pronograde situation. Here, anatomical traits that have been argued to be of adaptive significance for quadrupedal suspensory locomotion are reviewed and the evolution of these traits is discussed in light of the new framework. Experimental data are largely limited to Choloepus, but help to deduce functional aspects of the anatomy in Bradypus as well. The most important adaptive traits are hands and feet modified into relatively rigid hook-like appendages, great mobility of all joints proximal to the midcarpal and transverse tarsal joints, relatively long arms with a relatively short scapula, a rounded thorax with a small diameter, a highly mobile sterno-clavicular articulation, and emphasis on powerful flexion in the proximal limb joints via advantageous lever arms. Despite these changes, patterns of limb kinematics remained conservative during the course of evolution in the lineages leading to extant tree sloths, and it is suggested here that this also applies to the pattern of neuromuscular control of limb movements during locomotion. Morphological ‘solutions’ to altered functional demands posed by inversed orientation of the body differ in the two genera of extant tree sloths, thereby corroborating the proposed diphyly. Convergent evolution in tree sloths may be attributed to functional constraints posed by fossorial adaptations in early Xenarthra that canalized sloths to adopt a suspensory posture and locomotion in the arboreal habitat.

The information on ecological behavior of wild sloths is very scarce. In this study we determined the home ranges and resources used by three adult female three-toed sloths (Bradypus variegatus) and their four young in an agricultural matrix of cacao (Theobroma cacao), pasture, riparian forests and living fencerows in Costa Rica. Births occurred during November-December and the young became independent at five to seven months of age. Initially, mothers remained fixed in one or a few trees, but expanded their use of resources as young sloths became independent from them. Mothers initially guided the young to preferred food and cover resources, but they gradually left their young in small nucleus areas and colonized new areas for themselves. Home range sizes for young sloths (up to seven months of age) varied between 0.04-0.6 hectares, while home range sizes for mothers varied from 0.04-25.0 hectares. During the maternal care period, 22 tree species were used, with the most common being Cecropia obtusifolia (30.9%), Coussapoa villosa (25.6%), Nectandra salicifolia (12.1%), Pterocarpus officinalis (5.8%) and Samanea saman (5.4%). However, young sloths used only 20 tree species, with the most common being C. villosa (18.4%), S. saman (18.5%) and N. salicifolia (16.7%). The cacao agroforest was used only by mother sloths and never by their young following separation. However, in the riparian forest, both mother sloths and young used the tree species. A total of 28 tree species were used by the mother sloth; including the food species: C. obtusifolia, C. villosa, N. salicifolia and P. officinalis. However, the young used 18 trees species in this habitat with N. salicifolia and S. saman most commonly used, although they rested and fed during the day in C. obtusifolia, C. villosa and O. sinnuata. The cacao agroforest with adjacent riparian forests and fencerows provides an important habitat type that links the smaller secondary forests and other patches.

As human population, food consumption, and demand for forest products continue to rise over the next century, the pressures of land‐use change on biodiversity are projected to intensify. In tropical regions, countryside habitats that retain abundant tree cover and structurally complex canopies may complement protected areas by providing suitable habitats and landscape connectivity for a significant portion of the native biota. Species with low dispersal capabilities are among the most at risk of extinction as a consequence of land‐use change. We assessed how the spatial distribution of the brown‐throated sloth (Bradypus variegatus), a model species for a vertebrate with limited dispersal ability, is shaped by differences in habitat structure and landscape patterns of countryside habitats in north‐central Costa Rica using a multi‐scale framework. We quantified the influence of local habitat characteristics and landscape context on sloth occurrence using mixed‐effects logistic regression models. We recorded 27 sloths within countryside habitats and found that both local and landscape factors significantly influenced their spatial distribution. Locally, sloths favored structurally complex habitats, with greater canopy cover and variation in tree height and basal area. At the landscape scale, sloths demonstrated a preference for habitats with high proportions of forest and nearby large tracts of forest. Although mixed‐use areas and tree plantations are not substitutes for protected forests, our results suggest they provide important supplemental habitats for sloths. To promote the conservation and long‐term viability of sloth populations in the tropical countryside, we recommend that land managers retain structurally complex vegetation and large patches of native habitat.

Background and Aims

Cyanolichens are usually stated to be bipartite (mycobiont plus cyanobacterial photobiont). Analyses revealed green algal carbohydrates in supposedly cyanobacterial lichens (in the genera Pseudocyphellaria, Sticta and Peltigera). Investigations were carried out to determine if both cyanobacteria and green algae were present in these lichens and, if so, what were their roles.

Methods

The types of photobiont present were determined by light and fluorescence microscopy. Small carbohydrates were analysed to detect the presence of green algal metabolites. Thalli were treated with selected strengths of Zn2+ solutions that stop cyanobacterial but not green algal photosynthesis. CO2 exchange was measured before and after treatment to determine the contribution of each photobiont to total thallus photosynthesis. Heterocyst frequencies were determined to clarify whether the cyanobacteria were modified for increased nitrogen fixation (high heterocyst frequencies) or were normal, vegetative cells.

Key Results

Several cyanobacterial lichens had green algae present in the photosynthetic layer of the thallus. The presence of the green algal transfer carbohydrate (ribitol) and the incomplete inhibition of thallus photosynthesis upon treatment with Zn2+ solutions showed that both photobionts contributed to the photosynthesis of the lichen thallus. Low heterocyst frequencies showed that, despite the presence of adjacent green algae, the cyanobacteria were not altered to increase nitrogen fixation.

Conclusions

These cyanobacterial lichens are a tripartite lichen symbiont combination in which the mycobiont has two primarily photosynthetic photobionts, ‘co-primary photobionts’, a cyanobacterium (dominant) and a green alga. This demonstrates high flexibility in photobiont choice by the mycobiont in the Peltigerales. Overall thallus appearance does not change whether one or two photobionts are present in the cyanobacterial thallus. This suggests that, if there is a photobiont effect on thallus structure, it is not specific to one or the other photobiont.

Sloths are mammals renowned for spending a large proportion of time hanging inverted. In this position, the weight of the abdominal contents is expected to act on the lungs and increase the energetic costs of inspiration. Here, we show that three-fingered sloths Bradypus variegatus possess unique fibrinous adhesions that anchor the abdominal organs, particularly the liver and glandular stomach, to the lower ribs. The key locations of these adhesions, close to the diaphragm, prevent the weight of the abdominal contents from acting on the lungs when the sloth is inverted. Using ventilation rate and body orientation data collected from captive and wild sloths, we use an energetics-based model to estimate that these small adhesions could reduce the energy expenditure of a sloth at any time it is fully inverted by almost 13%. Given body angle preferences for individual sloths in our study over time, this equates to mean energy saving of 0.8–1.5% across individuals (with individual values ranging between 0.01 and 8.6%) per day. Given the sloth''s reduced metabolic rate compared with other mammals and extremely low energy diet, these seemingly innocuous adhesions are likely to be important in the animal''s energy budget and survival.

Algal farms maintained by different species of territorial herbivorous damselfishes vary in size, algal biomass, and species composition. To determine the factors that affect the structure of these farms, we compared farming behavior and intensity between two sympatric damselfish species, Stegastes nigricans (Lacepède) and S. obreptus (Whitley), which maintain two different types of farms in the Okinawa Islands, southern Japan. By weeding of indigestible algae and prompt exclusion of herbivorous fishes and grazing sea urchins, S. nigricans manage relatively small farms largely dominated by one algal crop species, Polysiphonia sp. In contrast, by delayed exclusion of herbivores, S. obreptus maintain larger farms invaded by diverse indigestible algae, such as Chondria sp. and Padina sp. As a result of intensive management, S. nigricans attains a higher algal biomass per area than does S. obreptus. This study provides a new perspective, as it suggests that damselfishes practise both intensive and extensive farming. The former results in small and highly productive monoculture farms of one palatable alga, while the latter leads to large, species-rich, mixed-culture farms.

Free-ranging Hoffmann's two-toed sloths (Choloepus hoffmanni; n=26) and brown-throated three-toed sloths (Bradypus variegatus; n=15) were manually captured and immobilized with 2.5 mg/kg ketamine + 0.02 mg/kg medetomidine administered intramuscularly. Physical examinations were conducted on each sloth 10 min after initial injection, and blood, fecal, and ectoparasite samples were collected. Heart rate, respiratory rate, body temperature, indirect systolic blood pressure, and indirect peripheral oxygen saturation were monitored every 5 min for the duration of anesthesia. After 45 min, atipamazole (0.1 mg/kg) was administered intramuscularly, as an antagonist to medetomidine, in order to facilitate recovery. All recoveries were smooth, rapid, and uneventful. Physiologic parameters were compared across time, gender, and species. All sloths, regardless of species and gender, demonstrated a time-dependent decrease in heart rate and blood pressure, and an increase in respiratory rate, during the course of anesthesia. Peripheral oxygen saturation was similar for all sloths over time. There were significant species differences for heart rate (Choloepus > Bradypus), respiratory rate (Choloepus > Bradypus), and systolic blood pressure (Bradypus > Choloepus), while there were significant gender differences for body temperature (males > females) and blood pressure (males > females). Results of this study suggest that the ketamine-medetomidine mixture, as described above, is a safe and effective anesthetic combination in free-ranging two- and three-toed sloths, although peripheral blood pressure should be monitored during anesthesia.

In many temperate estuaries, mats of opportunistic macroalgae accumulate on intertidal flats and in lower elevations of salt marshes, perhaps playing a role in linking water column nitrogen (N) supply to these benthic habitats. Using a flow-through seawater system and tidal simulator, we varied densities (equivalent to 0, 1, 2, or 3 kg m−2 wet mass) of 15N-labelled macroalgae (Enteromorpha intestinalis) on estuarine sediments in microcosms with/without pickleweed (Salicornia virginica) to assess N transfers from algae. In the 6-week experiment, macroalgal biomass increased from initial levels in the lower density treatments but all algae lost N mass, probably through both leakage and decomposition. With all densities of algae added, sediments and pickleweed became enriched in 15N. With increasing mat density, losses of algal N mass increased, resulting in stepwise increases in 15N labeling of the deeper sediments and pickleweed. While we did not detect a growth response in pickleweed with macroalgal addition during the experiment, N losses from algal mats that persist over many months and/or recur each year could be important to the mineral nutrition of N-limited marsh plants. We conclude that N dynamics of intertidal sediments and lower salt marsh vegetation are linked to the N pools of co-occurring macroalgae and that further study is needed to assess the magnitude and importance of N transfers.

Promiscuous mating strategies are much more common than previously appreciated. So much so, that several authors have proposed that promiscuity is the “rule” rather than the exception in vertebrate mating systems. Decreasing species mobility and increasing habitat fragmentation have both been suggested to reduce the “polygyny potential” of the environment and promote other mating strategies like promiscuity in females. We explored the social and genetic mating system for one of the most sedentary extant mammals, the brown-throated three-toed sloth (Bradypus variegatus), within a highly fragmented Neotropical habitat. Surprisingly, we found that three-toed sloths were strongly polygynous, with males excluding male competitors from their core ranges, and exhibiting strong reproductive skew. Indeed, only 25% of all resident adult males sired offspring and one individual sired half of all sampled juveniles. Paradoxically, a sedentary life-history strategy seems to facilitate polygyny in fragmented landscapes because multiple females can persist within small patches of habitat, and be monopolized by a single male. Our work demonstrates that strong polygyny can arise in systems in which the polygyny potential should be extremely low, and other strategies, including promiscuity, would be favoured. Mating systems can be influenced by a multitude of factor and are dynamic, varying among taxa, over time, and across habitats; consequently, mating systems remain difficult to predict based on general ecological principles.

The occasion of the Xenarthra Symposium during the ICVM 9 meeting allowed us to reflect on the considerable advances in the knowledge of sloths made by the “X-community” over the past two decades, particularly in such aspects as locomotion, mastication, diet, dental terminology, intraspecific variation, sexual dimorphism, and phylogenetic relationships. These advancements have largely been made possible by the application of cladistic methodology (including DNA analyses) and the discovery of peculiar forms such as Diabolotherium, Thalassocnus, and Pseudoglyptodon in traditionally neglected areas such as the Chilean Andes and the Peruvian Pacific desert coast. Modern tree sloths exhibit an upside-down posture and suspensory locomotion, but the habits of fossil sloths are considerably more diverse and include locomotory modes such as inferred bipedality, quadrupedality, arboreality or semiarboreality, climbing, and an aquatic or semi-aquatic lifestyle in saltwater. Modern tree sloths are generalist browsers, but fossil sloths had browsing, grazing, or mixed feeding dietary habits. Discovery of two important sloth faunas in Brazil (Jacobina) and southern North America (Daytona Beach and Rancho La Brea) have permitted evaluation of the ontogenetic variation in Eremotherium laurillardi and the existence of possible sexual dimorphism in this sloth and in Paramylodon harlani. A new dental terminology applicable to a majority of clades has been developed, facilitating comparisons among taxa. An analysis wherein functional traits were plotted onto a phylogeny of sloths was used to determine patterns of evolutionary change across the clade. These analyses suggest that megatherioid sloths were primitively semiarboreal or possessed climbing adaptations, a feature retained in some members of the family Megalonychidae. Pedolateral stance in the hindfoot is shown to be convergently acquired in Mylodontidae and Megatheria (Nothrotheriidae + Megatheriidae), this feature serving as a synapomorphy of the latter clade. Digging adaptations can only be securely ascribed to scelidotheriine and mylodontine sloths, and the latter are also the only group of grazing sloths, the remainder being general browsers.

Background and Aims

Many terrestrial orchids have an obligate requirement for mycorrhizal associations to provide nutritional support from germination to establishment. This study will investigate the ability of orchid mycorrhizal fungi (OMF) to utilize a variety of nutrient sources in the nutrient-impoverished (low organic) soils of the Southwest Australian Floristic Region (SWAFR) in order to effectively compete, survive and sustain the orchid host.

Methods

Mycorrhizal fungi representing key OMF genera were isolated from three common and widespread species: Pterostylis recurva, Caladenia flava and Diuris corymbosa, and one rare and restricted species: Drakaea elastica. The accessibility of specific nutrients was assessed by comparing growth including dry biomass of OMF in vitro on basal CN MMN liquid media.

Key Results

Each of the OMF accessed and effectively utilized a wide variety of nutrient compounds, including carbon (C) sources, inorganic and organic nitrogen (N) and inorganic and organic phosphorus (P). The nutrient compounds utilized varied between the genera of OMF, most notably sources of N.

Conclusions

These results suggest that OMF can differentiate between niches (micro-niche specialization) in a constrained, highly resource-limited environment such as the SWAFR. Phosphorus is the most limited macronutrient in SWAFR soils and the ability to access phytate by OMF indicates a characterizing functional capacity of OMF from the SWAFR. Furthermore, compared with OMF isolated from the rare D. elastica, OMF associating with the common P. recurva produced far greater biomass over a wider variety of nutritional sources. This suggests a broader tolerance for habitat variation providing more opportunities for the common orchid for recruitment and establishment at a site.
