Phylogenetic analysis of 4 cyanobacterial strains isolated from hot springs in Rajgir, India, was carried out using the 16S rRNA gene (1400 bp). These strains were identified as members of Chroococcales ( Cyanothece sp. strain HKAR-1) and Nostocales ( Nostoc sp. strain HKAR-2, Scytonema sp. strain HKAR-3, and Rivularia sp. strain HKAR-4). Furthermore, we evaluated the presence of ultraviolet-screening and (or) photoprotective compounds, such as mycosporine-like amino acids (MAAs) and scytonemin, in these cyanobacteria by using high-performance liquid chromatography. Well-characterized MAAs, including the critical and highly polar compounds shinorine, porphyra-334, and mycosporine-glycine, as well as several unknown MAAs, were found in these hot-spring-inhabiting microorganisms. The presence of scytonemin was detected only in Scytonema sp. strain HKAR-3 and Rivularia sp. strain HKAR-4. The results indicate that hot spring cyanobacteria, namely Cyanothece, Nostoc, Scytonema, and Rivularia, belonging to different groups possess various photoprotective compounds to cope up with the negative impacts of damaging radiations. 相似文献

Previously we improved the neurotensin (NT)-polyplex by the coupling of HA2 fusogenic peptide (FP) and Vp1 SV40 karyophilic peptide (KP). We now report the proportion of [(125)I]-NT, [(3)H]-FP, and poly-l-lysine (PLL) in the NT-polyplex, and some of its biophysical properties. We concluded that the most efficient NT-polyplex comprised 1 NT, 4 FP, and 2 PLL molecules. Electrophoresis revealed that high acidity is detrimental for NT-polyplex stability. Electron microscopy and electrophoresis studies showed that 6 muM KP and 1% serum condensed the plasmid DNA (pDNA) before the appearance of toroid structures. Four plasmids were used to evaluate the transfection efficiency. In vitro, maximum expression was produced at molar ratios (pDNA : [(125)I]-NT-[(3)H]-FP-PLL conjugate) of 1:34 for pEGFP-N1 and 1:27 for pECFP-Nuc. Cotransfection of those plasmids was attained at their optimum molar ratios. In vivo, maximum expression of the pDAT-BDNF-flag in dopamine neurons was produced at a 1:45 molar ratio, whereas that of pDAT-EGFP was at 1:20. The NT-polyplex in the presence of 1 muM SR-48692, an NT-receptor specific antagonist, and untargeted polyplex did not cause transfection in vivo demonstrating the specificity of gene transfer via NT-receptor endocytosis. Excess light is harmful for photosynthetic organisms. The cyanobacterium Synechocystis PCC 6803 protects itself by dissipating the excess of energy absorbed by the phycobilisome, the water-soluble antenna of Photosystem II, into heat decreasing the excess energy arriving to the reaction centers. Energy dissipation results in a detectable decrease of fluorescence. The soluble Orange Carotenoid Protein (OCP) is essential for this blue-green light induced mechanism. OCP genes appear to be highly conserved among phycobilisome-containing cyanobacteria with few exceptions. Here, we show that only the strains containing a whole OCP gene can perform a blue-light induced photoprotective mechanism under both iron-replete and iron-starvation conditions. In contrast, strains containing only N-terminal and/or C-terminal OCP-like genes, or no OCP-like genes at all lack this light induced photoprotective mechanism and they were more sensitive to high-light illumination. These strains must adopt a different strategy to longer survive under stress conditions. Under iron starvation, the relative decrease of phycobiliproteins was larger in these strains than in the OCP-containing strains, avoiding the appearance of a population of dangerous, functionally disconnected phycobilisomes. The OCP-containing strains protect themselves from high light, notably under conditions inducing the appearance of disconnected phycobilisomes, using the energy dissipation OCP-phycobilisome mechanism. 相似文献

We present a model of the S-state mechanism, as well as an improved eigenvalue analysis, that integrate into a coherent ensemble several features found since the S-state model was initially developed. These features include the presence of S–1, deactivations in the dark interval between flashes, and the change in the number of active PS II centers by photoinhibition or photoactivation. A new feature is the capacity to predict the steady-state distribution of S-states under conditions of steady photoinhibition or photoactivation. The improved eigenvalue analysis allowed the calculation of the initial S-state distribution. In addition, the model resolved true photochemical misses from apparent misses due to deactivations in the dark interval between flashes. The model suggested that most of the misses that are commonly reported are due to deactivations, and not to an intrinsic inefficiency of the photochemical mechanism of PS II. Because models that allow double-hits encompassing the S2 to S3 transition often predict negative initial quantities of S2 in cyanobacteria, our proposed model specifically prohibited them. The model accounts for inhomogeneous misses and a steady-state distribution of the type (S2)(S1)>(S3)(S0). This 5-step model uses only 4 probabilities, and is therefore easy to handle. The use of this model is critical for the analysis of several cyanobacterial strains, as well as for any species that show non-negligible deactivations in the dark interval between flashes. 相似文献

A water-soluble chlorophyll-binding protein (WSCP) is the single known instance of a putative chlorophyll (Chl) carrier in green plants. Recently the photoprotective function of WSCP has been demonstrated by EPR measurements; the light-induced singlet-oxygen formation of Chl in the WSCP tetramer is about four times lower than that of unbound Chl. This paper describes the crystal structure of the WSCP-Chl complex purified from leaves of Lepidium virginicum (Virginia pepperweed) to clarify the mechanism of its photoprotective function. The WSCP-Chl complex is a homotetramer comprising four protein chains of 180 amino acids and four Chl molecules. At the center of the complex one hydrophobic cavity is formed in which all of the four Chl molecules are tightly packed and isolated from bulk solvent. With reference to the novel Chl-binding mode, we propose that the photoprotection mechanism may be based on the inhibition of physical contact between the Chl molecules and molecular oxygen. 相似文献

A network of genes regulated by light in cyanobacteria 总被引:1,自引:0,他引:1
Oxygenic photosynthetic organisms require light for their growth and development. However, exposure to high light is detrimental to them. Using time series microarray data from a model cyanobacterium, Synechocystis 6803 transferred from low to high light, we generated a gene co-expression network. The network has twelve sub-networks connected hierarchically, each consisting of an interconnected hub-and-spoke architecture. Within each sub-network, edges formed between genes that recapitulate known pathways. Analysis of the expression profiles shows that the cells undergo a phase transition 6-hours post-shift to high light, characterized by core sub-network. The core sub-network is enriched in proteins that (putatively) bind Fe-S clusters and proteins that mediate iron and sulfate homeostasis. At the center of this core is a sulfate permease, suggesting sulfate is rate limiting for cells grown in high light. To validate this novel finding, we demonstrate the limited ability of cell growth in sulfate-depleted medium in high light. This study highlights how understanding the organization of the networks can provide insights into the coordination of physiologic responses. 相似文献

Hypothesis of coherent vibration states in biological systems based on nonlinear interaction between longitudinal elastic and electric polarization fields with metabolic energy supply was formulated by Frohlich. Conditions for excitation of coherent states and generation of electromagnetic fields are satisfied in microtubules which form electrical polar structures. Numerical models are used for analysis of Frohlich's vibration states in cells. Reduction of activity and of energy production in mitochondria, and disintegration of cytoskeleton structures by phosphorylation on the pathway of cancer trasformation can diminish excitation of the Frohlich's vibration states and of the generated electromagnetic field, which results in disturbances of the interaction forces between cells. Interaction forces between cancer cells may be smaller than interaction forces between healthy cells and cancer cells as follows from numerical models. Mechanism of malignity, i.e. local invasion, detachment of cancer cells, and metastasis, is assumed to depend on the electromagnetic field. 相似文献

Prolonged exposure of plants to high fluxes of solar radiation as well as to other environmental stressors disturbs the balance between absorbed light energy and capacity of its photochemical utilization resulting in photoinhibition of and eventually in damage to plants. Under such circumstances, the limiting of the light absorption by the photosynthetic apparatus efficiently augments the general photoprotective mechanisms of the plant cell, such as reparation of macromolecules, elimination of reactive oxygen species, and thermal dissipation of the excessive light energy absorbed. Under stressful conditions, plants accumulate, in different cell compartments and tissue structures, pigments capable of attenuation of the radiation in the UV and visible parts of the spectrum. To the date, four principle key groups of photoprotective pigments are known: mycosporine-like amino acids, phenolic compounds (including phenolic acids, flavonols, and anthocyanins), alkaloids (betalains), and carotenoids. The accumulation of UV-absorbing compounds (mycosporine-like amino acids and phenolics in lower and higher plants, respectively) is a ubiquitous mechanism of adaptation to and protection from the damage by high fluxes of solar radiation developed by photoautotrophic organisms at the early stages of their evolution. Extrathylakoid carotenoids, betalains, and anthocyanins play an important role in long-term adaptation to the illumination conditions and in protection of plants against photodamage. A prominent feature of certain plant taxa lacking some classes of photoprotective pigments (such as anthocyanins) is their substitution by other compounds (e.g. keto-carotenoids or betalains) disparate in terms of chemical structure and subcellular localization but possessing close spectral properties. 相似文献

The present study characterizes the dynamic interfacial properties of calf lung surfactant (CLS) and samples reconstituted in a stepwise fashion from phospholipid (PL), hydrophobic apoprotein (HA), surfactant apoprotein A (SP-A), and neutral lipid fractions. Dipalmitoylphosphatidylcholine (DPPC), the major PL component of surfactant, was examined for comparison. Surface tension was measured over a range of oscillation frequencies (1-100 cycles/min) and bulk phase concentrations (0.01-1 mg/ml) by using a pulsating bubble surfactometer. Distinct differences in behavior were seen between samples. These differences were interpreted by using a previously validated model of surfactant adsorption kinetics that describes function in terms of 1) adsorption rate coefficient (k1), 2) desorption rate coefficient (k2), 3) minimum equilibrium surface tension (gamma*), 4) minimum surface tension at film collapse (gammamin), and 5) change in surface tension with interfacial area for gamma < gamma* (m2). Results show that DPPC and PL have k1 and k2 values several orders of magnitude lower than CLS. PL had a gammamin of 19-20 dyn/cm, significantly greater than CLS (nearly zero). Addition of the HA to PL restored dynamic interfacial behavior to nearly that of CLS. However, m2 remained at a reduced level. Addition of the SP-A to PL + HA restored m2 to a level similar to that of CLS. No further improvement in function occurred with the addition of the neutral lipid. These results support prior studies that show addition of HA to the PL markedly increases adsorption and film stability. However, SP-A is required to completely normalize dynamic behavior. 相似文献

Amorphous protein aggregates are oligomers that lack specific, high-order structures. Soluble amorphous aggregates are smaller than ~1 µm. Despite their lack of high-order structure, amorphous protein aggregates exhibit specific biophysical properties such as reversibility of formation, density, conformation, and biochemical stability. Our mutational analysis using a Solubility Controlling Peptide (SCP) tag strongly suggests that amorphous aggregation of small globular proteins can significantly increase in vivo immune response and that the magnitude of enhanced immune responses depends on the aggregates’ biophysical and biochemical properties. We propose that SCP tags might help develop subunit (protein) adjuvant-free (immunostimulant-free) vaccines by controlling the aggregation propensity of target proteins. 相似文献

Ferroptosis is an oxidative and iron-dependent form of regulated cell death (RCD) recently described in eukaryotic organisms like animals, plants, and parasites. Here, we report that a similar process takes place in the photosynthetic prokaryote Synechocystis sp. PCC 6803 in response to heat stress. After a heat shock, Synechocystis sp. PCC 6803 cells undergo a cell death pathway that can be suppressed by the canonical ferroptosis inhibitors, CPX, vitamin E, Fer-1, liproxstatin-1, glutathione (GSH), or ascorbic acid (AsA). Moreover, as described for eukaryotic ferroptosis, this pathway is characterized by an early depletion of the antioxidants GSH and AsA, and by lipid peroxidation. These results indicate that all of the hallmarks described for eukaryotic ferroptosis are conserved in photosynthetic prokaryotes and suggest that ferroptosis might be an ancient cell death program. 相似文献

We use an advanced fluorescence method of Nonlinear Laser Fluorimetry in combination with Fluorescence Induction and Relaxation technique to study the influence of excess‐light conditions on the physiological state of the green alga Chlorella pyrenoidosa. We demonstrate that zeaxanthin‐dependent non‐photochemical quenching leads to a significant increase in the rate constant of singlet‐singlet annihilation of chlorophyll a excited state, which suggests profound conformational changes in the light‐harvesting complexes of photosystem II.
