Populations of three salt tolerant forage grasses (Cynodon dactylon, Imperata cylindrica, and Sporobolus arabicus) were collected from the salt-affected soils of the Salt Range and normal non-saline soils of the Faisalabad region to assess their mechanism of adaptation to saline stress by determining ion relations and some specific anatomical modifications. The population of S. arabicus from the Salt Range showed increased growth (root and shoot length, and root and shoot dry weights) under saline conditions. Salt tolerance in this species was related to structural modifications such as increased area of root, stem, leaf blade, and leaf sheath for toxic ion accumulation, increased vesicular hair density in leaves and aerenchyma formation in leaf sheath for ion exclusion. Uptake of toxic ions was high in the Salt Range population of C. dactylon and salt tolerance was related to ion exclusion through specific leaf structural modifications such as vesicular hairs. Salt tolerance in the Salt Range population of I. cylindrica was mainly associated with restricted uptake of toxic Na+ and Cl at root level, and accumulation of toxic ions via increased succulence in leaf blades and leaf sheaths in addition to some excretion of toxic ions through leaf sheath aerenchyma.  相似文献

Photosynthetic rates, chlorophyll content, and activities of several photosynthetic enzymes were determined per cell, per unit DNA, and per unit leaf area in five ploidal levels of the C4 dicot Atriplex confertifolia. Volumes of bundle sheath and mesophyll protoplasts were measured in enzymatic digestions of leaf tissue. Photosynthetic rates per cell, contents of DNA per cell, and activities of the bundle sheath enzymes ribulose 1,5-bisphosphate carboxylase (RuBPC) and NAD-malic enzyme per cell were correlated with ploidal level at 99% or 95% confidence levels, and the results suggested a near proportional relationship between gene dosage and gene products. There was also a high correlation between volume of mesophyll and bundle sheath cells and the ploidal level. Contents of DNA per cell, activity of RuBPC per cell, and volumes of cells were correlated with photosynthetic rate per cell at the 95% confidence level. The mesophyll cells did not respond to changes in ploidy like the bundle sheath cells. In the mesophyll cells the chlorophyll content per cell was constant at different ploidal levels, there was less increase in cell volume than in bundle sheath cells with an increase in ploidy, and there was not a significant correlation (at 95% level) of phosphoenolpyruvate carboxylase activity or content and pyruvate,Pi dikinase activity with increase in ploidy. The number of photosynthetic cells per unit leaf area progressively decreased with increasing ploidy from diploid to hexaploid, but thereafter remained constant in octaploid and decaploid plants. Numbers of cells per leaf area were not correlated with cell volumes. The mean photosynthetic rates per unit leaf area were lowest in the diploid, similar in 4×, 6×, and 8×, and highest in the decaploid. The photosynthetic rate per leaf area was highly correlated with the DNA content per leaf area.  相似文献

The association with and digestion of intact leaf sections of cool- and warm-season grasses by cattle rumen protozoa were investigated by light and scanning electron microscopy and by in vitro dry matter disappearance studies. Within extensively degraded areas of mesophyll tissue in cool-season forages, almost all protozoa were Epidinium ecaudatum form caudatum, with maximum numbers at 4 to 10 h of incubation. However, few protozoa were found inside warm-season forage leaves. In in vitro dry matter disappearance studies of a series of incubations with and without 1.6 mg of streptomycin per ml, which inhibited the cellulolytic activity of the bacteria, and in comparison with uninoculated controls, rumen protozoa degraded 11.0 and 3.7 percentage units of orchardgrass and bermuda-grass, respectively. Scanning electron microscopy showed that the tissues degraded in orchardgrass consisted of large amounts of mesophyll and portions of the parenchyma bundle sheath and epidermis; no tissue loss due to the protozoa was observed in bermudagrass. The relationship of these observations to forage digestion is discussed.  相似文献

Arundinella hirta L. is a C4 plant having an unusual C4 leaf anatomy. Besides mesophyll and bundle sheath cells, A. hirta leaves have specialized parenchyma cells which look morphologically like bundle sheath cells but which lack vascular connections and are located between veins, running parallel to them. Activities of phosphoenolpyruvate and ribulose-1,5-bisphosphate carboxylases and phosphoenolpyruvate carboxykinase, NADP-and NAD-malic enzymes were determined for whole leaf extracts and isolated mesophyll protoplasts, specialized parenchyma cells, and bundle sheath cells. The data indicate that A. hirta is a NADP-malic enzyme type C4 species. In addition, specialized parenchyma cells and bundle sheath cells are enzymatically alike. Compartmentation of enzymes followed the C4 pattern with phosphoenolpyruvate carboxylase being restricted to mesophyll cells while ribulose-1,5-bisphosphate carboxylase and decarboxylating enzymes were restricted to bundle sheath and specialized parenchyma cells.  相似文献

To examine anatomical adaptations in a potential forage grass, Imperata cylindrica (L.) Raeuschel, a population was collected from the natural salt-affected soils of the Salt Range, Pakistan. Using a hydroponic system, the degree of salt tolerance in terms of structural modifications in the Salt Range ecotype was compared with that in an ecotype collected from a normal non-saline habitat of the Faisalabad region. The Salt Range ecotype was superior to the Faisalabad ecotype in biomass production under saline conditions. High salt tolerance of the Salt Range ecotype was associated with increased succulence in root and leaf (mainly midrib), formation of aerenchyma in leaf sheath, increased vascular bundle area, metaxylem area and phloem area, highly developed bulliform cells on leaves and increased sclerification in root and leaf. Furthermore, both stomatal density and stomatal area were considerably reduced under high salinities in the Salt Range ecotype.  相似文献

The morphology, anatomy and vasculature of Pinus leaves was studied. The results indicate that the Pinus leaf is always supplied with only one single vascular bundle, which is surrounded by a prominent bundle sheath. In several taxa, especially of subgenus Pinus, the vascular bundle may however be subdivided by longitudinal parenchymatic bands in the middle part of the leaf. As a result, the single bundle gets the appearance of two individual bundles, which are surrounded by a common bundle sheath. The general rule that a bundle sheath does sheath only one single bundle, as in other gymno- and angiospermous seed plants, applies therefore also for Pinus. The morphological and anatomical similarities between cladodes of Sciadopitys (Sciadopityaceae) and Pinus leaves are based on a completely different bauplan. The idea of “hidden cladodes” in some Pinaceae is therefore obsolete.  相似文献

The species Arundinella hirta L. posseses a striking variation of the leaf anatomy that is characteristic of C4 grasses. In addition to a sheath of large, bright green cells around the vascular bundles, there are strands of large parenchyma cells which appear identical to the bundle sheath cells and which run parallel to the vascular bundles, but which are not associated with any vascular tissue. This species may be useful for studying the cellular compartmentalization associated with the C4 pathway and should provide interesting material for determining the role of translocation in the functioning of the C4 system.  相似文献

利用石蜡片法,对台湾草(Zoysia tenuifoia Willd.ex Trin.)、海雀稗(Paspalum vaginatum Sw.)、狗牙根[Cnodon dactylon(L.)Pers.]三种细叶暖季型草坪草的叶片有茎的解剖结构进行了研究。结果表明:叶片及茎的解剖结构与植物的耐旱性、耐践踏性和弹性等坪用特性有着密切联系。三种植物中,台湾草因叶片表皮细胞、泡状细胞、维管束鞘、机械组织有茎中纤维带的特征,而在耐旱性、耐踏踏性和弹性等坪用特性上表现出优于其他二种草坪草;狗牙根的耐旱性较强,耐踏踏性和弹性一般,海雀稗则在这几项坪用特征上均较弱。  相似文献

The localization of enzymes responsible for nitrate assimilation and the generation of NADH for nitrate reduction were studied in corn (Zea mays L.) leaf blades. The techniques used effectively separated mesophyll and bundle sheath cells as judged by microscopic observations, enzymic assays, chlorophyll a/b ratios and photochemical activities. Nitrate reductase, nitrite reductase, and the nitrate content of leaf blades were localized primarily in the mesophyll cells, although some nitrite reductase was found in the bundle sheath cells. Glutamine synthetase, NAD-malate dehydrogenase, NAD-glyceraldehyde-3-phosphate dehydrogenase, and NADP-glutamate dehydrogenase were found in both types of cells, however, more NADP-glutamate dehydrogenase was found in the bundle sheath cells than in the mesophyll cells. These data indicate that the mesophyll cells are the major site for nitrate assimilation in the leaf blade because they contained an ample supply of nitrate and the enzymes considered essential for the assimilation of nitrate into amino acids. Because the specific activity of nitrate reductase was severalfold lower than the other enzymes involved in nitrate assimilation, nitrate reduction is indicated as the rate-limiting step in situ. A sequence of reactions is proposed for nitrate assimilation in the mesophyll cells of corn leaves as related to the C-4 pathway of photosynthesis.  相似文献

The Sucrose export defective1 (Sxd1) gene of maize was cloned and shown to encode a novel protein conserved between plants and cyanobacteria. The structure of the Sxd1 locus was determined in wild-type plants and two independent sxd1 alleles. Expression analysis demonstrated that the gene was transcribed in all green tissues, with highest levels in maturing leaf blades. In situ hybridization studies revealed high levels of Sxd1 mRNA in bundle sheath cells, with lower levels within the mesophyll. The SXD1 protein was localized to chloroplasts, in both bundle sheath and mesophyll cells. Levels of sucrose, glucose, and fructose were compared between wild-type and sxd1 plants. Mutant plants were fully capable of producing sucrose and accumulated all three sugars at concentrations above those measured in wild-type plants. Despite these increased sugar concentrations, photosynthetic gene expression was not significantly downregulated in affected areas of sxd1 leaf blades. These results are consistent with photosynthate being trapped within anthocyanin-accumulating regions of sxd1 leaves due to plasmodesmal occlusion at the bundle sheath-vascular parenchyma boundary of the minor veins. A model for SXD1 function is proposed in which the protein is involved in a chloroplast-to-nucleus signaling pathway necessary for proper late-stage differentiation of maize bundle sheath cells, including the developmentally regulated modification of plasmodesmata.  相似文献

Intercellular distribution of enzymes involved in amino nitrogen synthesis was studied in leaves of species representing three C4 groups, i.e. Sorghum bicolor, Zea mays, Digitaria sanguinalis (NADP malic enzyme type); Panicum miliaceum (NAD malic enzyme type); and Panicum maximum (phosphoenolpyruvate carboxykinase type). Nitrate reductase, nitrite reductase, glutamine synthetase, and glutamate synthase were predominantly localized in mesophyll cells of all the species, except in P. maximum where nitrite reductase had similar activity on a chlorophyll basis, in both mesophyll and bundle sheath cells. NADH-glutamate dehydrogenase was concentrated in the bundle sheath cells, while NADPH-glutamate dehydrogenase was localized in both mesophyll and bundle sheath cells. The activities of nitrate-assimilating enzymes, except for nitrate reductase, were high enough to account for the proposed in vivo rates of nitrate assimilation.  相似文献

Aiming to complement the group’s taxonomy, the objective of this work is to describe and characterise, searching for morpho-anatomical patterns, the leaf anatomy of the Manilkara species occurring in the northeast region of Brazil. The leaves of all Manilkara species were analysed, using herbaria specimens and collections made in the field by the authors. The characterisation of the leaf blades was performed according to the usual protocols in vegetal anatomy. As diagnosed by anatomical characters, the Manilkara Adans. genus presents petioles with a thick cuticle, a uniseriate epidermis, a sclerenchymatous sheath involving the vascular bundle, and laticifers on the cortical and medullar regions. The anatomical data increase the M. elata and M. huberi synonymisation and show the importance of the anatomy as a complementary tool to the taxonomy based on external morphology.  相似文献

Aizoaceae is one of the most important and widespread succulent plant families in both tropical/subtropical regions and arid zones. In this study, 27 species were collected from various floristic regions in Egypt, Kingdom of Saudi Arabia and cactus farms (Kalupia – Egypt). The morphological characteristics of every taxon were recorded. The important morphological features included: the number of leaves per plant; leaf types; leaf position (cauline or radical; the latter indicates leaves arising from, or near, the roots); leaf arrangement; petiolate or sessile leaves; leaf sheath present or absent; leaf shape; leaf margin; leaf tip; presence of leaf ‘window area’; leaf texture; and presence of white or dark miniscule dots (white miniscule dots from calcium carbonate and dark miniscule dots from tannin sacs). The investigated anatomical features were as follows: shape of the transverse section; the type of epidermal cells; the presence of large epidermal cells (bladder cells); presence of papilla and simple hairs; presence of tannin sacs; shapes of calcium oxalate crystals; shape of the xylem vessels; and the presence of Kranz unit (the unit that constitutes the vascular bundle/s, parenchyma sheath, and surrounding mesophyll) or collenchyma sheath. All data were recorded in a data matrix (as either text or numerical data), which was used to construct the identification key and phylogeny tree using a multi-variate statistical package. The results of our analysis may open the possibility of using the morphological and anatomical features of leaves to distinguish between the subfamilies, genera, and species of Aizoaceae.  相似文献

The leaf anatomy of the rhizomatous Iris species with ensiform leaves and the related genera Pardanthopsis and Belamcanda is described. Their isobilateral leaves may or may not have a pseudo-dorsiventral structure. Variable characters of their leaf blades include: outline in transverse section, height and shape of papillae, form and structure of stomata, transverse section outline of marginal fibre strands and sclerenchy matous inner bundle sheath at phloem and xylem poles, forms of mesophyll arrangement, mesophyll structure and air canals, vascular bundle arrangement and the detailed structure of the larger vascular bundles, distribution of tannin, size and distribution of crystals. The taxonomic and ecological significance of these characters has been evaluated.
The anatomical characteristics of 25 supraspecific taxa in three genera are presented and compared in tables. The relationships and evolutionary position of these taxa are discussed. Each of the three subgroups within Iris appears to be correlated with a syndrome of anatomical characters. Some species currently of uncertain taxonomic position are discussed, and their classification based on anatomical data is suggested.

The location and some morphological, anatomical, and functional aspects of the gravity-sensitive pulvini of a selected number of grass shoots are examined. There are two distinct gravity-sensitive regions near the nodal regions of Gramineae. One, the leaf sheath pulvinus, is located at the base of the sheathing leaf bases, and is characteristic of the subfamily Festucoideae. The other, the internodal pulvinus, is located at the base of the internode, a little above the nodal joint. Most members of the Panicoideae possess internodal pulvini, in addition to more or less developed leaf sheath pulvini. Three members of the Oryzoideae examined possess leaf sheath pulvini only, while Phragmites australis (Arundinoideae) possesses both leaf sheath and internodal pulvini. Leaf sheath pulvini of some grasses develop hairs, cork-silica cell pairs or stomatal apparatuses over the epidermis while many others are devoid of any such idioblasts. Both the leaf sheath and internodal pulvini of all grasses examined preferentially exclude, or accumulate very little silica, whereas the regions of the shoot immediately above and below the pulvini in these same grasses accumulate large quantities of silica. Pulvini remain unsilicified or poorly silicified throughout their life and even after several days following geotropic bending. Pulvini are also distinguished from the regions above and below them by the lack of lignin in the bundle cap cells. Lignin is found only in the xylem vascular tissue, and this consists of annular and helical vessel elements only. The bundle cap cells are rich in pectin and are described as collenchymatous. All pulvini possess specialized zones of cells containing starch statoliths. In response to horizontal displacement of the shoots, the lower side of the pulvini grows by cell elongation only. The collenchymatous cells stretch in a manner that results in alternately thin and thick regions of cell wall.  相似文献
