Summary The aim of this project was to see whether 5-hydroxytryptamine (5-HT) believed to be present in the skin glands of anuran amphibians can be demonstrated histochemically. The Periodic acid-Schiff technique (P.A.S.) and three histochemical methods known to demonstrate 5-HT in enterochromaffin cells (Pontana's, diazonium and Schmorl's) were applied to the dorsal skin of three species—Xenopus laevis, Rana angolensis and Bufo regularis.Mucous glands were identified by their P.A.S. reactivity in all three species. Poison glands were identifiable in Bufo regularis and Xenopus laevis only. The secretory granules of the latter glands have strong positive reactions with all three histochemical techniques used.It is concluded that the poison glands of anuran amphibians contain 5-HT demonstrable by histochemical means.  相似文献

In amphibians, secretions of toxins from specialized skin poison glands play a central role in defense against predators. The production of toxic secretions is often associated with conspicuous color patterns that warn potential predators, as it is the case of many dendrobatid frogs, including Ameerega picta. This species resembles the presumably nontoxic Leptodactylus lineatus. This study tests for mimicry by studying the morphology and distribution of skin glands, components of skin secretion, and defensive behavior. Dorsal skin was studied histologically and histochemically, and skin secretions were submitted to sodium dodecyl sulfate polyacrylamide gel electrophoresis, reversed phase high performance liquid chromatography and assays for proteolytic activity. We found that poison glands in A. picta are filled with nonprotein granules that are rich in carbohydrates, while L. lineatus glands present protein granules. Accordingly, great amounts of proteins, at least some of them enzymes, were found in the poison of L. lineatus but not in that of A. picta. Both species differ greatly on profiles of gland distribution: In L. lineatus, poison glands are organized in clusters whose position coincides with colored elements of the dorsum. These regions are evidenced through a set of displays, suggesting that poison location is announced to predators through skin colors. In contrast, A. picta presents lower densities of glands, distributed homogeneously. This simpler profile suggests a rather qualitative than quantitative investment in chemical defense, in agreement with the high toxicity attributed to dendrobatids in general. Our data suggest that both species are toxic or unpalatable and transmit common warning signals to predators, which represents a case of Müllerian mimicry. J. The strategies used by ectotherms to minimise the detrimental effects of suboptimal thermal environments on physiological performance are often related to whether they inhabit a terrestrial or aquatic environment. Most terrestrial ectotherms use thermoregulatory strategies to maintain body temperature within an optimal range, while many aquatic ectotherms utilise thermal acclimation to maintain performance over varying seasonal temperatures. This study aimed to elucidate the relative contributions of acclimation and behavioural thermoregulation to maintaining whole-animal performance over varying seasonal temperatures in the semi-terrestrial Lamington spiny crayfish (Euastacus sulcatus). Crayfish activity and surface temperatures were determined by means of radio tracking and behavioural observations. Field studies demonstrated that E. sulcatus is exposed to stable daily temperatures, varying only between seasons from 10°C in late winter to over 20°C in summer. Also, terrestrial behaviour corresponded to a small portion of crayfish time (1.13%), much lower than predicted, indicating that E. sulcatus has limited opportunity for behavioural thermoregulation. We also tested the effect of acclimation to either 10 or 20°C on chela strength and stamina. We found acclimation had a more marked effect on chela stamina than maximum strength measures; however, overall acclimatory capacity was limited in E. sulcatus. Thus, we found that the semi-terrestrial crayfish E. sulcatus used neither thermoregulatory behaviours nor physiological strategies to deal with seasonal changes in environmental temperature.  相似文献

1. Continual stimulation of frog skin epithelium and the salivary glands of the insects Calliphora and Musca with 5-hydroxytryptamine (5-HT) leads to desensitisation, i.e. the tissue fails to respond to the application of further 5-HT. 2. Incubation of desensitised frog skin and Musca salivary glands with either N-acetyl neuraminic acid or inositol partially restored the 5-HT responses whilst incubation with a combination of N-acetyl neuraminic acid and inositol gave additive effects on the recovery of the 5-HT responses. 3. Incubation of desensitised salivary glands of Calliphora with inositol totally restored the 5-HT response whilst incubation with N-acetyl neuraminic acid had no effect. 4. It is concluded that desensitisation involves depletion of secondary messenger from the tissues coupled with receptor degradation and that considerable differences exist in the turnover of 5-HT receptors, the receptors in Musca salivary glands being highly labile, those in Calliphora salivary glands highly stable and those of frog skin epithelium being intermediate in their stability.  相似文献

Endemic New Zealand frogs of the genus Leiopelma are from a basal lineage of extant anurans that release defensive secretions onto their skin when disturbed. Here, we characterize the gross anatomy and microscopic structure of the skin of L. archeyi, L. hochstetteri, and L. pakeka using stereoscopic, light and transmission electron microscopy. The terrestrial L. archeyi and L. pakeka possess dimorphic granular glands, categorized as type I and II, based on their frequency and morphological traits, whereas the semi‐aquatic L. hochstetteri lacks type I glands. This is the first report of differential dimorphism in anurans of the same genus. This dimorphism could be interpreted as an adaptation to different physiological or ecological needs of these species. However, species within this ancient genus share similar general gland morphology with other anurans, namely, a secretory unit containing storage granules ensheathed by myoepithelial cells. Type I glands are ellipsoid, large and contain a homogeneous mass of electron‐dense granules (1.8 ± 0.08 μm in diameter). Type II glands are round and contain larger heterogeneous granules (4.06 ± 0.16 μm) of varying densities. Exposure to noradrenaline causes the contraction of myoepithelial cells, resulting in bulk discharge of type I glands through the epidermal duct onto the skin surface. Differential release of secretions from dimorphic glands may be indicative of their functional specialisation in antipredatory or regulative roles. Mass spectrometric techniques were used to de novo sequence peptides present in the skin secretions of Leiopelma species. A total of 30 previously undescribed peptides from Leiopelma species were fully or partially sequenced. These peptides exhibited no similarity to any known compounds. J. The distribution of biogenic amines in the young pluteus of Psammechinus miliaris Gmelin was examined by means of the histochemical fluorescence method of Falck and Hillarp. Two different systems where monoamines occur were detected. Catecholamines were mainly found in a presumably impulse propagating neuron-like system, while 5-HT (serotonin) was predominantly localized in the endoderm wall. The results are discussed in relation to the ontogeny and phylogeny of sea urchins.  相似文献

Summary The effect of electrical field stimulation on noradrenaline (NA), dopamine (DA) and 5-hydroxytryptamine (5-HT) nerve terminals in rat brain slicesin vitro was investigated. Slices prepared from the cerebral cortex or the neostriatum were incubated in physiologic buffer for 30 min and then superfused by buffer and stimulated by an electrical field (biphasic pulses, 10 Hz, 12 mA, 2 ms) for various time periods. The effect of the stimulation was studied at the cellular level with the histochemical fluorescence technique of Falck and Hillarp. The transmitter overflow into the superfusing buffer caused by the stimulation was studied with isotope technique. Cerebral Cortex NA Nerve Terminals. Stimulation caused release of NA from cortical NA nerve terminals. Already after 2 min stimulation a slight decrease of the fluorescence intensity of the nerve terminals could be found. Stimulation for 15 to 30 min greatly reduced the fluorescence intensity. In slices preincubated with3H-NA the stimulation-induced overflow of tritium during 2 min stimulation was about 15% (i.e. 15% of the tissue tritium content was overflowing into the superfusing buffer in response to stimulation for 2 min). During prolonged stimulation there was a considerable decline of the tritium efflux. Cerebral Cortex 5-HT Nerve Terminals. The 5-HT-analogue 6-hydroxytryptamine (6-HT) which is readily taken up into 5-HT nerve terminals was used to permit good visualization of these nerve terminals. Uptake of 6-HT into cortical NA nerve terminals was prevented by preincubation with 6-hydroxydopamine (6-OH-DA) or protriptyline. Stimulation for 15 to 30 min reduced the fluorescence intensity of the 5-HT nerve terminals. In slices preincubated with3H-5-HT the stimulation-induced overflow of tritium during 2 min stimulation was about 5%. The tritium efflux slowly decreased during continuous stimulation. Neostriatal DA Nerve Terminals. In slices frozen directly after preparation an intense diffuse fluorescence could be seen. After incubation in drug-free buffer at 37° C the fluorescence was localized in the varicosities of the DA nerve terminals. The central parts of the slices almost completely lacked specific fluorescence, while the outer zones were brightly fluorescent. No clear reduction of the fluorescence intensity of the DA nerve terminals in the outer zone could be observed after stimulation for 30 min. In slices preincubated with3H-DA the stimulation-induced overflow of tritium during 2 min stimulation was about 2%. The tritium efflux slowly decreased during continuous stimulation.It is suggested that the differences in release between the various nerve terminal systems foundin vitro reflect differences in transmitter release occurringin vivo. The occurrence and release of thyrotrophin-releasing hormone (TRH) and 5-hydroxytryptamine (5-HT) from amphibian skin have been described by previous investigators. In the present study, the precise location and site of release of TRH and 5-HT from the skin of Rana pipiens and Xenopus laevis have been examined using a combination of procedures including immunohistochemistry, HPLC, and radioimmunoassay. The results indicate that TRH is located specifically within the dermal glands of these species, and that both TRH and 5-HT are discharged from these glands following adrenergic stimulation. The origin and functional significance of these substances in amphibian skin granular glands are discussed.  相似文献

Summary The granular glands in Xenopus laevis skin are known to contain large quantities of biogenic amines and bioactive peptides which closely resemble mammalian brain-gut peptides. We studied the development of glands producing 5-hydroxytryptamine (5-HT) and caerulein using immunohistochemistry, HPLC-fluorometric systems and RIA. The immunoreactivities of 5-HT and caerulein were first detected in the spherical gland rudiments in the stratum spongiosum at St. 58 (Nieuwkoop and Faber stage), or at the beginning of metamorphosis. Both immunoreactivities appeared in the same rudiment at the same time. Some of the gland rudiments have a small lumen filled with both immunoreactive materials at St. 58–59. During the rest of the metamorphic period, the glands grow in size, accumulating immunoreactive materials in the lumen. The concentrations of 5-HT and caerulein in the skin of tadpoles were below 1 ng per mg wet tissue at St. 58–59, increased as metamorphosis proceeded and reached 63 and 134 ng per mg wet tissue at St. 66, or at the end of metamorphosis, respectively. The amphibian granular glands where large quantities of biogenic amines and hormone-like peptides are rapidly synthesized may provide a useful model for the study of the development of amine- and peptide-producing cells including neurons and paraneurons.  相似文献

Summary The formaldehyde-induced fluorescence technique had shown 5-hydroxytryptamine-containing enteric neurons in the intestine of the teleost Platycephalus bassensis, but did not reveal such neurons in the intestine of Tetractenos glaber or Anguilla australis. Re-examination of these animals with 5-hydroxytryptamine immunohistochemistry showed immunoreactive enteric neurons in the intestine of all three teleost species. The 5-hydroxytryptamine-containing enteric neurons showed essentially the same morphology in all species examined: the somata were situated in the myenteric plexus, extending down into the circular muscle layer, but none were found in the submucosa; processes were found in the myenteric plexus, the circular muscle layer and the lamina propria. It was concluded that the neurons may innervate the muscle layers or the mucosal epithelium, but were unlikely to be interneurons. In a range of teleosts, enterochromaffin cells were found in the intestine of only those species in which the formaldehyde technique did not visualize neuronal 5-hydroxytryptamine. Available evidence suggests that, in vertebrates, 5-HT-containing enterochromaffin cells are lacking only where there is an innervation of the gut mucosa by nerve fibres containing high concentrations of 5-HT.  相似文献

Summary Immunocytochemical techniques were used to investigate the distribution and co-localization of neuronspecific enolase (NSE) and serotonin (5-HT) in the skin of the conger eel, Conger conger. NSE and 5-HT immunoreactivity were found in Merkel cells; these cells were also identified at the electron-microscope level by the presence of characteristic granules and their association with an intraepithelial nerve ending. For the first time, it was demonstrated that Merkel-cell granules of vertebrate skin exhibit in immunoreaction with 5-HT. The production of amines may indicate that the Merkel cells of C. conger have both secretory capabilities and transduction functions.However, immunocytochemical investigation of the synaptic zones at the electron microscope level will be necessary to confirm this hypothesis.The present histochemical results suggest that NSE and 4-HT may be marker substances for Merkel cells, and that immunocytochemistry is a useful tool for the light-microscopic localization of these cells.  相似文献

There are at least 40,000 species of microalgae in the aquatic environment. Fifteen species of marine dinoflagellates and freshwater cyanobacteria are known to produce paralytic shellfish toxins (PSTs) and represent a threat to human and/or livestock health. Although known toxic species are regularly monitored, the wider cross‐section of microalgae has not been systematically tested for PSTs. Advances in rapid screening techniques have resulted in the development of highly sensitive and specific methods to detect PSTs, including the sodium channel and saxiphilin binding assays. These assays were used in this study in 96‐well formats to screen 234 highly diverse isolates of Australian freshwater and marine microalgae for PSTs. The screening assays detected five toxic species, representing one freshwater cyanobacterium (Anabaena circinalis Rabenhorst) and four species of marine dinoflagellates (Alexandrium minutum Halim, A. catenella Balech, A. tamarense Balech, and Gymnodinium catenatum Graham). Liquid chromatography‐fluorescence detection was used to identify 14 saxitoxin analogues across the five species, and each species exhibited distinct toxin profiles. These results indicate that PST production is restricted to a narrow range of microalgal species found in Australian waters.  相似文献

Males of the two species of Mertensiella (M. caucasica and M. luschani) possess a tubercle projecting from the skin of the dorsal tail base, the single morphological character that defines the genus. The dorsal tail tubercle functions during courtship, and its role is similar in both species. The tubercle is inserted into the cloaca of the female during ventral amplexus, shortly before the male deposits a spermatophore. Histological examination, however, revealed that the dorsal tubercles differ structurally between the two species. In M. caucasica, the tubercle consists primarily of elongate mucous glands, with granular glands occurring only at the base. Both mucous and granular glands of the tubercle are larger than those in typical skin. Unlike typical skin, however, mucous glands are larger than granular glands. In M. luschani, mucous glands and granular glands occur throughout the tubercle, and the granular glands are larger than the mucous glands, although both types are larger than those in typical skin. —The injection of 50 μg of 5,6-dihydroxytryptamine (5,6-HT) into a lateral ventricle of the rat depleted the spinal cord and various regions of the brain of indoleamines (presumably 5-HT) and 5-hydroxyindole acetic acid. The concentrations of 5-HT were measured by two different methods: the formation of a fluorescent derivative with o-phthalaldehyde, and the native fluorescence in hydrochloric acid. When the results of both methods were compared on the pons and medulla 4 days after injecting 5,6-HT, the loss in indoleamine appeared to be greater when o-phthalaldehyde was used. This suggests that the two methods may be measuring different compounds. According to both methods, the loss of 5-HT persisted for several days after the injection of 5,6-HT, but by 2 months 5-HT concentrations (measured only by the native fluorescence procedure), had recovered to near-normal values. The depletion of 5-HT was most pronounced in regions adjacent to the ventricular system and in the spinal cord. Initially, caudate and septum were more affected on the side of the injection, and later showed some permanent atrophy. The injection of up to 50 μg of 5,6-HT did not lead to any significant loss of noradrenaline or dopamine from the brain, or to any reduction in the activity of the enzyme tyrosine hydroxylase. The drug was a potent inhibitor of the uptake of [3H]5-HT by brain slices, but was less effective in inhibiting catecholamine uptake systems. These observations suggest a preferential action on tryptaminergic neurones. Larger doses of 5,6-HT caused a loss of catecholamines and tyrosine hydroxylase from the brain, and were severely toxic.  相似文献

Summary The effect of pH, temperature, and l-lactate on the O2 bindign properties of haemocyanin (Hc) from three talitroidean species i.e., the aquatic Apohyale pugettensis, the semi-terrestrial Megalorchestia californiana, and the semi-/euterrestrial Traskorchestia traskiana was studied. The Hc of A. pugettensis was characterized by a higher O2 affinity (and more pronounced Bohr shift) than the Hc of either M. californiana or T. traskiana. Apohyale was the only species that possessed He that was sensitive to temperature change. Resuspending Hc from each of the three species in a stock Ringer's solution (based on the ionic composition of the haemolymph of T. traskiana) showed that the persistence of the difference in Bohr shift between Apohyale and the other two species was due to differences in the haemocyanins themselves and not attributable to their respective ionic environments. An inverse relationship was found between the cooperativity (n 50) and pH of Hc from T. traskiana and A. pugettensis but not for M. californiana. Summary Reproductive isolation in termites is not well known. Our study carried out on two sympatric species from northern Vietnam, Macrotermes annandalei and M. barneyi, showed that dispersal flights and sex pheromones were two important factors in their reproductive isolation. These fungus-growing termites were isolated, partially due to the timing of their respective dispersal flights. M. annandalei flew the first day after rain, while the flights of M. barneyi occurred the second day after rain. However, the flights can also be simultaneous in the two species. Sex pheromones of M. annandalei and M. barneyi were shown to be species-specific. In both species, they were secreted by females from two glandular sources, from tergal glands located on tergite 6 to 10 in M. annandalei and tergite 5 to 10 in M. barneyi, and from posterior sternal glands located on sternite 6 and 7 in both species. These posterior sternal glands, found for the first time in the Termitidae, were sex-specific glands. Although not fully identified, sex pheromones of M. annandalei and M. barneyi were clearly different from the trail-following pheromone secreted by the sternal gland stricto sensu located on the sternite 5. These results show that in termites, the sexual behaviour, the glandular origin of sex pheromones and their role in reproductive isolation greatly vary depending on the species and deserve to be more extensively studied.Received 8 April 2003; revised 1 September 2003; accepted 10 September 2003.  相似文献

The purpose of the present study was to demonstrate visually and localize the presence of serotonin (5-HT) in the ovary and oviduct of the domestic hen using a histochemical Falck-Hillarp method. Experiments were carried out on White Leghorn laying hens with no egg in the shell gland. The specific yellow fluorescence, indicating the presence of 5-HT, was found both in the ovary and all examined oviductal parts. The strongest fluorescence was present in the ovarian stroma containing small follicles with a diameter under 4 mm. In the wall of the largest preovulatory follicle a very strong fluorescence was located mainly in the theca layer. In the oviductal parts, the intensity of 5-HT fluorescence in the infundibulum and magnum was fairly strong, whereas in the isthmus and shell gland it was weak. Fluorescence seen in the infundibulum, magnum, and isthmus was primarily localized along the luminal borders of the fold surface epithelium. In the shell gland 5-HT fluorescence was found within the uterine folds, especially in the tubular glands. Moreover, the presence of an egg in the definite oviductal segment (infundibulum or isthmus) increased the intensity of yellow fluorescence in this part.  相似文献

Exotic species can invade and establish new habitats both as a result of their own traits, and as a result of the characteristics of the environment they invade. Here, we show that the abundance of the invasive submerged aquatic plant, Myriophyllum spicatum (Eurasian watermilfoil) is highly dependent on the conditions of the environment in a mesocosm experiment. M. spicatum is allelopathic towards epiphytic algae, and in the absence of algivorous snails, we found that the abundance of both algae and M. spicatum significantly increased with experimentally increased nutrient loading, while the abundance of native submerged macrophytes declined. However, when snails were present, snail biomass increased with increasing nutrient loading, and M. spicatum biomass was consistently low while native submerged macrophyte biomass was consistently high. Our results stress the importance of the interaction between species traits and environmental conditions when considering the invasiveness of certain exotic species and the invasibility of certain environments.  相似文献

The morphology and histomorphology of the tongue and the histochemistry of the lingual glands of eight specimens from four species of Ligurian Sea odontocetes (Stenella coeruleoalba, Tursiops truncatus, Grampus griseus, and Ziphius cavirostris) were studied. The shape of tongues and the appearance of their dorsal surfaces differed between species. The lingual glands differed in size, distribution, and histochemistry by species. In S. coeruleoalba and G. griseus, a strong alcianophilic mucous material was detected in the lingual glands, while neutral and acidic mucous substances were observed in the most proximal secretory acini. In G. griseus, small simple alveolar apocrine glands were also found, and the duct of the serous lingual glands in Z. cavirostris is of apocrine type. Numerous mechanoreceptors were observed. Only the tongue of the young specimens showed marginal papillae: their histomorphological composition is consistent with the hypothesis that they create a tight seal between the tongue and the roof of the cavity in order to create suction. This comparative study suggests that differences in tongue morphology and in the morphology and histochemistry of lingual glands might be related to feeding habits.  相似文献
