The settling behaviour of Aphis craccivora Koch, the vector of cowpea aphid-borne mosaic virus (CAMV) in cowpea, Vigna unguiculata (L.) Walp., on aphid-resistant, aphid-tolerant and aphid-susceptible cowpea lines was investigated. It was found that although apterae counts on the aphid-susceptible and aphid-tolerant lines were higher than on the aphid-resistant ones, apterae were more widely dispersed among the latter. Whereas there was a positive significant correlation between alatae numbers and incidence of CAMV in the aphid-susceptible and aphid-tolerant lines, this correlation was negative in the aphid-resistant ones. However, the incidence of CAMV was not significantly different from each other in all the lines. This indicated that aphid activity (e.g. wide dispersal) was more important in the spread of CAMV than the absolute number of viruliferous alatae, an effect which was manifested on the aphid-resistant lines.Aphids acquire CAMV more readily from aphid-susceptible and aphid-tolerant source plants than from aphid-resistant ones. The significance of this in relation to secondary spread of CAMV and the effect of noncolonising aphids are discussed.
Le comportement d'atterissage d'Aphis craccivora et l'acquisition du virus de la mosaïque transmis par puceron chez les variétés de niébe résistantes au puceron
Résumé Le comportement d'atterissage d'Aphis craccivora, vecteur du virus de la mosaique transmis par puceron (CAMV) sur Vigna unguiculata, a été examiné sur des variétés résistantes, sensibles ou tolérantes aux pucerons. On a observé que bien que les dénombrements d'aptères sur variétés sensibles ou tolérantes aient été plus élevés que sur variétés résistantes, les aptères étaient plus largement dispersés sur ces dernières. Tandis qu'il y avait une corrélation positive significative entre les nombres d'ailés et la présence de CAMV sur les variétés sensibles et tolérantes aux pucerons, cette corrélation était négative sur les variétés résistantes. Cependant, la fréquence de CAMV n'était pas significativement différente parmi toutes ces variétés. Ceci a montré l'activité des pucerons (c'est à dire leur dispersion) étail plus importante dans la diffusion du CAMV que le nombre absolu d'ailés virosés, effet qui était manifeste sur les variétés résistantes aux pucerons.Les pucerons contaminés au CAMV le sont plus par des plantes sensibles ou tolérantes aux pucerons que par des plantes résistantes. La discussion porte sur la signification de ceci en relation avec la diffusion secondaire de CAMV, et de l'effet des pucerons non colonisateurs.

The feeding activity of Aphis craccivora (Koch) was monitored on cowpea plants of aphid-susceptible (Vita 7) and aphid-resistant (TVu 801) cultivars, using an AC electronic feeding monitoring system. Waveforms corresponding to salivation, phloem ingestion and non-phloem ingestion were observed. Aphids probing on the resistant cultivar showed a significantly reduced ingestion of phloem sap compared with those aphids which fed on the susceptible cultivar. The insect also made more brief and repeated probes on TVu 801. In addition, the duration of non-probing activities and non-phloem ingestion was shorter on the susceptible than on the resistant cultivar.  相似文献

Electrical penetration graph recordings using direct current (DC-EPGs) were used to analyze aspects of the probing behavior of cowpea aphid,Aphis craccivora Koch, on intact plants and on hexane, ethyl acetate, and methanol extracts of leaves of aphid-resistant (ICV-12) and aphid-susceptible (ICV-1) cultivars of cowpeaVigna unguiculata (L.) Walp. In one set of experiments, recordings were done on plants with or without parafilm wrapping, or on plants painted with raw leaf juice and extracts of the two cultivars. In another study, recordings were done on leaf extracts homogenized in water or in 0.5M sucrose solution and then placed in parafilm membrane sachets. Electrodes were inserted into soil mix for the experiments on potted plants or into extract fractions and raw juice for the membrane feeding experiments on leaf extracts in parafilm sachets. Waveform signals were recorded from resistance fluctuations from interactions between aphids and substrates, and electromotive forces generated within each preparation. ICV-12 plants with or without parafilm wrapping, and ethyl acetate extracts and raw juice of that cultivar significantly (P≤0.05) reduced stylet penetration behavior. Thus, antixenosis as manifested by disruption of aphid stylet activity on host substrates, appeared to be a governing modality of aphid resistance in ICV-12.  相似文献

Impact of initial density of cowpea aphid Aphis craccivora Koch (Aphididae) at infestation on the growth and yield of aphid-susceptible cowpea cultivar ICV-1 and aphid-resistant cultivar ICV-12, was investigated. Plants at the seedling, flowering and podding stages of development were infested with five aphid densities consisting of 0, 2, 5, 10 and. 20 aphids per plant and maintained for 22 days. Extended leaf heights of plants and aphid counts were recorded at 7, 12, 17 and 22 days after infestation. Two crop growth parameters (biomass duration and leaf area duration), and two plant yield parameters (number of pods per plant and number of seeds per pod) were recorded. Due to the occurrence of parthenogenesis and changes in population dynamics during infestations, aphid densities were converted into cumulative cowpea aphid-days, to facilitate data analyses and interpretation. ANOVA indicated that there was significant (P=s 0.05) difference in aphid-day accumulations between the two cultivars when infested at the seedling stage. Accumulations on cv. ICV-1 were greater than on cv. ICV-12. However, no such differences between the cultivars were detected when plants were infested at flowering and podding stages. Therefore, the seedling stage was used for comparisons of the impact of cowpea aphid-days on the growth and yield parameters of the two cultivars. At the 95% confidence intervals, ICV-12 plants were consistently taller than ICV-1 plants. Infested ICV-1 seedlings showed stunting and other growth deformities which were not observed on ICV-12 plants. Regression analyses revealed substantial reductions in the growth and yield parameters of ICV-1 relative to ICV-12. Overall, cowpea aphid-days provided a convenient and reliable method for studying the aphid population dynamics and the subsequent impact on plant growth and yield performance.  相似文献

The effects of two synthetic pyrethroids, cypermethrin and deltamethrin, on the feeding behaviour of Aphis craccivora and its transmission of the nonpersistent cowpea aphid-borne mosaic virus (CAMV) were compared to those of an organophosphate (dimethoate) and carbamate (pirimicarb) in greenhouse and laboratory tests. Cypermethrin at 100, 150 and 200 mg a.i. litre-1 and deltamethrin at 75, 100 and 150 mg a.i. litre-1 restricted the acquisition and inoculation of CAMV, whereas dimethoate and pirimicarb had no significant effect. Cypermethrin protected against transmission of CAMV longer after application than deltamethrin and there was a strong interaction between The duration of the first probe was shorter on pyrethroid-treated plants than on dimethoate-or pirimicarb-treated plants, and on pyrethroid-treated plants aphids probably did not probe long enough to acquire or inoculate CAMV. It is concluded that the use of synthetic pyrethroids is potentially valuable in controlling CAMV in the field until resistance against the virus has been bred into cultivars.  相似文献

The ability of populations of adult apterous Aphis craccivora, A. gossypii and A. citricola to transmit the cowpea aphid-borne virus (CAMV) and cucumber mosaic virus (CMV) to cowpea was investigated. CMV was more readily transmitted than CAMV by all three aphid species, but was most readily transmitted by A. craccivora and A. citricola, with transmissions ranging from 64–71 %. With CAMV, the infection level with A. gossypii was higher than with A. craccivora and both were more efficient than A. citricola, although the differences were not significant. With mixed infections of CAMV and CMV, there was a higher level of CAMV transmission using A. craccivora from sequential feeding when CMV-infected plants were fed on first followed by CAMV-infected plants, suggesting that cowpea plants were more prone to infection by CAMV when already infected by CMV.  相似文献

Biological control and soybean cultivars bred for increased resistance to the soybean aphid (Aphis glycines) are two approaches used to manage this serious pest of soybeans in North America. However, as with many other pest systems, the compatibility of these two pest management approaches has not been studied in detail. The aphidiine wasp Aphidius colemani is one of several candidate species for biological control of the soybean aphid in soybean in North America. Resistance to the soybean aphid in the USDA soybean cultivar Dowling is largely controlled by a single dominant gene Rag1, which is the focus of plant breeding programs directed against the soybean aphid. In this study, we measured developmental and behavioral differences in the parasitic wasp A. colemani when it attacked soybean aphids feeding on either the aphid-resistant Dowling or aphid-susceptible Glenwood cultivars of soybean. We used a combination of choice and no-choice experiments to examine the effects of host plant cultivar on the number of parasitized aphids formed and the sex ratio and body weights of adult offspring produced. Significantly more aphids were parasitized when they fed on Glenwood compared to Dowling and these offspring were larger when they developed in aphids that fed on Glenwood soybeans. To distinguish between effects on foraging decisions and offspring survivorship, we conducted an additional experiment that followed the oviposition decisions and fate of each parasitized aphid. Foraging female A. colemani spent less time handling individual aphids and encountered and attacked aphids at a higher rate when they fed on aphids feeding on Glenwood soybeans than aphids feeding on Dowling soybeans. Furthermore, wasp survivorship in aphids was greater on Glenwood than Dowling. Taken together, aphid-resistance in soybeans has negative effects on foraging behavior and offspring fitness of A. colemani raising concerns about the compatibility of these two pest management approaches.  相似文献

We examine how the distribution of a leafgalling aphid (Pemphigus betae) affects other species associated with natural stands of hybrid cottonwoods (Populus angustifolia x P. fremontii). Aphid transfers on common-garden clones and RFLP analysis show that resistance to aphids in cottonwoods is affected by plant genotype. Because susceptible trees typically support thousands of galls, while adjacent resistant trees have few or none, plant resistance traits that affect the distribution of this abundant herbivore may directly and/or indirectly affect other species. We found that the arthropod community of aphid-susceptible trees had 31% greater species richness and 26% greater relative abundance than aphid-resistant trees. To examine direct and indirect effects of plant resistance traits on other organisms, we experimentally excluded aphids and found that abundances and/or foraging behavior of arthropods, fungi, and birds were altered. First, exclusion of gall aphids on susceptible trees resulted in a 24% decrease in species richness and a 28% decrease in relative abundance of the arthropod community. Second, exclusion of aphids also caused a 2- to 3-fold decrease in foraging and/or presence of three taxa of aphid enemies: birds, fungi, and insects. Lastly, aphidexclussion resulted in a 2-fold increase in inquilines (animals who live in abodes properly belonging to another). We also found that fungi and birds responded to variation in gall density at the branch level. We conclude plant resistance traits affect diverse species from three trophic levels supporting a bottom-up influence of plants on community structure.  相似文献

Summary Inheritance of resistance to cowpea aphid, Aphis craccivora Koch, in three resistant cultivars of cowpea, Vigna unguiculata (L.) Walp, was studied. The parents, F1 and F2 population were grown in an insect-proof screenhouse. Each 3-day-old seedling was infested with 10 apterous adult aphids. Seedling reaction was recorded when the susceptible check was killed. The segregation data revealed that the resistance of ICV11 and TVU310 is governed by single dominant genes. All the F2 seedlings of the cross ICV10xTVU310 were resistant, indicating that they have the same gene for resistance. However, the F2 populations from the crosses ICV10xICV11 and ICV11xTVU310 segregated in a ratio of 151, indicating that the dominant genes in ICV11 and TVU310 are non-allelic and independent of each other. The resistance gene of ICV10 and TVU310 is designated as Ac1 and that of ICV11 as Ac2.  相似文献

The soybean aphid (Aphis glycines Matsumura) is the most damaging insect pest of soybean [Glycine max (L.) Merr.] in North America. New soybean aphid biotypes have been evolving quickly and at least three confirmed biotypes have been reported in USA. These biotypes are capable of defeating most known aphid resistant soybean genes indicating the need for identification of new genes. Plant Introduction (PI) 567301B was earlier identified to have antixenosis resistance against biotype 1 and 2 of the soybean aphid. Two hundred and three F7:9 recombinant inbred lines (RILs) developed from a cross of soybean aphid susceptible cultivar Wyandot and resistant PI 567301B were used for mapping aphid resistance genes using the quantitative trait loci (QTL) mapping approach. A subset of 94 RILs and 516 polymorphic SNP makers were used to construct a genome-wide molecular linkage map. Two candidate QTL regions for aphid resistance were identified on this linkage map. Fine mapping of the QTL regions was conducted with SSR markers using all 203 RILs. A major gene on chromosome 13 was mapped near the previously identified Rag2 gene. However, an earlier study revealed that the detached leaves of PI 567301B had no resistance against the soybean aphids while the detached leaves of PI 243540 (source of Rag2) maintained aphid resistance. These results and the earlier finding that PI 243540 showed antibiosis resistance and PI 567301B showed antixenosis type resistance, indicating that the aphid resistances in the two PIs are not controlled by the same gene. Thus, we have mapped a new gene near the Rag2 locus for soybean aphid resistance that should be useful in breeding for new aphid-resistant soybean cultivars. Molecular markers closely linked to this gene are available for marker-assisted breeding. Also, the minor locus found on chromosome 8 represents the first reported soybean aphid-resistant locus on this chromosome.  相似文献

The effectiveness of the synthetic pyrethroids deltamethrin and lambda-cyhal-othrin in preventing (i) aphid colonisation of four cowpea cultivars with different levels of aphid resistance and (ii) the introduction and subsequent spread of cowpea aphid-borne mosaic virus and cucumber mosaic virus was investigated under tropical field conditions. Sprays of these pyrethroids eight days apart prevent aphid colonisation and within crop spread of virus by the colonising Aphis craccivora. However, neither deltamethrin nor lambda-cyhalothrin prevented the initial introduction of virus into the cowpea crop and, when incoming alate incidence was high, virus incidence was higher in the sprayed than in the unsprayed plots. In addition, the degree of aphid resistance of each cultivar affected secondary virus spread within the crop, with greatest spread in the most resistant cultivar.  相似文献

A study to determine yield response to the Russian wheat aphid, Diuraphis noxia (Mordvilko), was conducted during the 1997-1998 and 1998-1999 growing seasons at three eastern Colorado locations, Akron, Fort Collins, and Lamar, with three wheat lines containing either Russian wheat aphid-resistant Dn4 gene, Dn6 gene, or resistance derived from PI 222668, and TAM 107 as the susceptible control. Russian wheat aphids per tiller were greater on TAM 107 than the resistant wheat lines at the 10x infestation level at Fort Collins and Akron in 1999. Yield, seed weight, and number of seeds per spike for each wheat line were somewhat affected by Russian wheat aphid per tiller mainly at Fort Collins. The infested resistant wheat lines harbored fewer Russian wheat aphids and yielded more than the infested susceptible wheat lines. Wheat lines containing the Dn4, Dn6, and PI 222668 genes contain different levels of antibiosis or antixenosis and tolerance. Although differences existed among sites and resistance, there is a benefit to planting resistant wheat when there is a potential for Russian wheat aphid infestations.  相似文献

The tripartite interactions in a pathosystem involving wheat (Triticum aestivum L.), the Barley yellow dwarf virus (BYDV), and the BYDV vector aphid Sitobion avenae were studied under field conditions to determine the impact of these interactions on aphid populations, virus pathology and grain yield. Wheat varietal resistance to BYDV and aphids varied among the three wheat varieties studied over two consecutive years. The results demonstrated that (1) aphid peak number (APN) in the aphid + BYDV (viruliferous aphid) treatment was greater and occurred earlier than that in the non-viruliferous aphid treatment. The APN and the area under the curve of population dynamics (AUC) on a S. avenae-resistant variety 98-10-30 was significantly lower than on two aphid-susceptible varieties Tam200(13)G and Xiaoyan6. (2) The production of alatae (PA) was greater on the variety 98-10-30 than on the other varieties, and PA was greater in the aphid + BYDV treatment on 98-10-30 than in the non-viruliferous aphid treatment, but this trend was reversed on Tam200(13)G and Xiaoyan6. (3) The BYDV disease incidence (DIC) on the variety 98-10-30 was greater than that on the other two varieties in 2012, and the disease index (DID) on Tam200(13)G was lower than on the other varieties in the aphid + BYDV and BYDV treatments in 2012, but not in 2011 when aphid vector numbers were generally lower. (4) Yield loss in the aphid + BYDV treatment tended to be greater than that in the aphid or BYDV alone treatments across varieties and years. We suggested that aphid population development and BYDV transmission tend to promote each other under field conditions. The aphids + BYDV treatment caused greater yield reductions than non-viruliferous aphids or virus treatment. Wheat varietal resistance in 98-10-30 affects the aphid dispersal, virus transmission and wheat yield loss though inhibits aphid populations from increasing.  相似文献

In a series of laboratory and climate chamber tests we compared the growth and behaviour of Aphis craccivora on one susceptible (ICV-1) and two aphid-resistant (ICV-11 and ICV-12) cowpea lines. The aphids' growth rates were much lower on the resistant cowpea lines than on the susceptible one, indicating strong antibiosis. In addition, the aphids invariably settled in higher numbers on the susceptible line than on either of the resistant. Compared to ICV-1, damaged leaves of the resistant line ICV-12 were settled upon to a higher degree than undamaged leaves, and leaf discs from the same line were even less resistant.On resistant lines individual aphids waited a significantly longer time before making their first test probe. Total probing time as well as the time preceding a decision to stay or leave was also longer.These results are discussed in relation to the possible mechanisms involved, and we also consider the effects of previous leaf feeding on the expression of resistance in the field.
Résumé Au cours d'expériences au laboratoire et en chambres climatisées nous avons comparé la croissance et le comportement de A. craccivora sur une lignée sensible (ICV-1) et deux lignées résistantes (ICV-11 et ICV-12) de V. unguiculata. Les vitesses de croissance des pucerons ont été beaucoup plus lentes sur les lignées résistantes que sur la lignée sensible, ce qui révèle une forte antibiose. De plus, les pucerons atterrissent invariablement en plus grand nombre sur la variété sensible. Par comparaison avec ICV-1, les atterrissages sur lignée résistante ICV-12 étaient plus nombreux sur les feuilles endommagées que sur les feuilles intactes; les disques de feuilles de cette même lignée étaient encore moins résistants.Les pucerons ont séjourné individuellement un temps plus long sur les lignées résistantes avant de faire leur premier sondage. Le temps consacré aux sondages ainsi que le temps précédant de choix entre départ ou maintien sur la feuille étaient plus longs avec les lignées résistantes.Ces résultats ont été discutés en fonction des mécanismes impliqués. Nous avons aussi examiné les effets de la consommation antérieure sur les manifestations de la résistance dans la nature.

Abstract. Two laboratory studies were conducted to investigate effects of treatments for direct "current electrical penetration graph (DC-EPG) recordings or 'tether effect', on behaviour and reproductive performance of cowpea aphid Aphis craccivora Koch. The experiments constituted a control study in application of DC-EPGs to analyse cowpea aphid feeding behaviour and host plant resistance mechanisms. Resistant (ICV-12) and susceptible (ICV-1) cultivars of cowpea Vigna unguiculata (L.) Walp were used. EPG treatments included two groups of aphids: tethered aphids that were exposed to DC electricity via an attachment of a thin, flexible gold wire on their dorsum using a droplet of adhesive silver paint, and 'free' (untethered) aphids with a dorsal spot of silver paint only. EPGs of the tethered aphids were recorded continuously for c. ! h, whereas from the 'free' aphids recordings were done only for brief periods of 2–5 min, by temporarily contacting a gold wire to the spot of silver paint. Waveform signals generated from resistance fluctuations and electromotive forces, and representing aphid stylet penetration behaviour were recorded. A separate experiment was conducted to investigate effects of EPG treatments on aphid survivorship and population growth. Overall, EPG treatments did not significantly affect aphid stylet penetration behaviour or life-table parameters. However, effects of crop cultivar on those characteristics were significant. Waveform E2, which denotes aphid ingestion in phloem sieve elements, and non-penetration behaviour were important indicators of aphid resistance in ICV-12. Also, apart from the number of aphid generations, other life-table parameters were useful indicators of ICV-12 resistance. Thus, DC-EPGs provided a reliable technique for studying aphid stylet behaviour, and investigation of aphid resistance in cowpeas.  相似文献

The feeding behavior of cowpea aphid, Aphis craccivora Koch (Homoptera: Aphididae) was examined on seedlings of narrow leafed lupin, Lupinus angustifolius L., and yellow lupin, L. luteus L., using electronic monitoring of insect feeding behavior (EMIF). Aphid feeding behavior was first compared between resistant (cv. Kalya) and susceptible (cv. Tallerack) varieties of narrow-leafed lupin. Aphids spent significantly more time in non- penetration and stylet pathway activities, and significantly less time in the sieve element phase on Kalya than on Tallerack, suggesting that feeding deterrence is an important component of aphid resistance in Kalya. Aphid feeding on a susceptible yellow lupin variety (cv. Wodjil) was then compared with that on two resistant lines, one (Teo) with high and the other (94D024-1) with low seed alkaloid content. There were no consistent differences in aphid feeding behavior between Wodjil and Teo. Total, mean and percentage sieve element phase times were significantly lower, and total and percentage times in non-phloem phase were greater on 94D024-1 than on Wodjil, suggesting the possibility of phloem-based deterrence in 94D024-1.  相似文献

Two tomato inbred backcross line (IBL) populations, derived from crosses between aphid-susceptible Lycopersicon esculentum Mill. 'Peto 95-43' X resistant wild L. pennellii Corr (D'arcy) accession LA716, and Peto 95-43 X resistant wild L. hirsutum f. glabratum Mull accession LA407, were evaluated in replicated field experiments for resistance to potato aphid, Macrosiphum euphorbiae (Thomas), and green peach aphid, Myzus persicae (Sulzer). Aphid infestation scores for each IBL and control (LA716, LA407, Peto 95-43, and susceptible 'Alta') plot were recorded weekly for 5 and 9 wk during the summers of 2000 and 2001, respectively. Aphid infestation scores from leaflets were used to calculate area under the infestation pressure curve (AUIPC), a measure of aphid infestation throughout the growing season, for each IBL and control. Score AUIPC was highly correlated with actual aphid count AUIPC, indicating that scores accurately reflected aphid infestation. Score AUIPC was also highly correlated across both years (2000 and 2001) and locations. Low score AUIPC was significantly correlated with larger plant size and sprawling, indeterminate plant growth habit. Seven IBLs, LA716, and LA407 were significantly more resistant to aphids (lower score AUIPC) than susceptible parent Peto 95-43 in both years. Two IBLs, 1034 and 1051, were not significantly different from resistant LA407 for score AUIPC in both years. The seven aphid-resistant IBLs identified here can be useful as donor parent material for resistance breeding efforts in cultivated tomato.  相似文献

Aphis gossypii is the main virus vector in muskmelon crops. The melon gene Vat confers resistance to non‐persistent virus transmission by this aphid. The mechanism of this resistance is not well understood, but no relationship has been detected between resistance and the probing behaviour of aphids on resistant plants. Results presented here suggest that temporary blockage of aphid stylet tips preventing virus particle release may explain the resistance conferred by Vat gene. We performed experiments in which viruliferous aphids were allowed to probe different sequences of resistant (Vat‐bearing) and/or susceptible melon plants. The results demonstrated that A. gossypii inoculates Cucumber mosaic virus (CMV) efficiently in susceptible plants having previously probed resistant plants, showing that the resistance mechanism is reversible. Furthermore, the infection rate obtained for susceptible plants was the same (25%) regardless of whether the transmitting aphid had come directly from the CMV source or had subsequently probed on resistant plants. This result suggests that virus is not lost from stylet to plant during probing of resistant plants, supporting the temporary blockage hypothesis. We also found that the ability of Myzus persicae to transmit CMV is noticeably reduced after probing on resistant plants, providing evidence that this aphid species also responds to the presence of the Vat gene. Finally, we also found that in probes immediately after virus acquisition M. persicae inoculates resistant plants with CMV more efficiently than susceptible plants, perhaps because the Vat gene product induces increased salivation by this aphid.  相似文献

Plant viruses and aphids can interact via contest competition for plant resources and induce changes in plant physiology, which can have effects on a third trophic level. The aim of this study was to determine how the interactions between a circulative plant virus and its aphid vector may affect the performance of an endoparasitoid and how parasitism may affect the efficiency of virus transmission by its aphid vector. The timing when parasitized aphids were transferred to virus-infected lettuce leaves was critical for the performance of A. ervi. Higher parasitoid larvae mortality, longer developmental times and lower percentages of mummification were detected on viruliferous/parasitized aphid nymphs when the time lag between parasitism and exposure to the virus was less than 24 h.
