Propranolol caused a contractile response in the isolated rabbit ear artery (EA). The concentration of propranolol causing a threshold contraction was 1.76 × 10?6M while that causing a maximal contraction of 2.2 ± 0.18 g was 3 × 10?5M. Higher concentrations caused tissue relaxation. Phentolamine, 10?7 and 10?6M reduced the propranolol-induced contractions by 50% and 90%, respectively while prazosin, 10?8, 10?7 and 10?6M caused reductions of 54, 74 and 88%, respectively. Reserpinization of the rabbit with 5 mg/kg 24 hours before use eliminated the EA contractile response to tyramine but had no effect on that to propranolol. Desmethylimipramine plus deoxycorticosterone acetate enhanced the submaximal contraction of the EA to propranolol. In vitro denervation with 6-hydroxydopamine (6-OHDA) decreased the response of the EA to tyramine and propranolol by 96% and 85% respectively but increased that to norepinephrine (NE) by 11%. Rabbit thoracic aorta (TA) did not respond to propranolol. In EA contracted with vasopressin o or 30 mM potassium, propranolol 10?4 and 3 × 10?4M caused a 20% and 100% relaxation, respectively. It is concluded that propranolol elicits a contractile response in the EA, at least in part, by direct activation of postsynaptic alpha adrenoceptors.  相似文献

Abstract The peptides proctolin, crustacean cardioactive peptide (CCAP) and FMRFamide, which are known to modulate insect muscle contractions, were assayed for their action on oviduct contractions in Hylobius abietis. A video microscopy technique and computer‐based method of data acquisition and analysis were used to investigate the effects of theses peptides on spontaneous contractions of continuously perfused oviducts. All three peptides tested stimulate spontaneous contraction activity of the pine weevil oviduct, increasing the frequency and amplitude of phasic contractions in a dose‐dependent manner. Proctolin is more potent as a stimulator than CCAP. For proctolin a threshold response of oviduct muscles is at concentration of peptide 10?11–10?10 mol/L and for CCAP at concentration range 10?10–10?9 mol/L. FMRFamide exerts a weak stimulatory effect on the oviduct, and at higher concentrations of the peptide (above 10?8 mol/L). The peptides exert different responses on oviduct contractions and they may play a role as functional regulators in such processes as egg movement and oviposition.  相似文献

Possible involvement of cAMP-dependent mechanisms in the development of both phasic and tonic contractions induced by oxytocin — OT (25 nM and 25 µM, respectively), as well as of KCl-induced contracture, was studied on the myometrium of estradiol-dominated rats using the myometrial strips with suppressed spontaneous mechanical activity. The intracellular cAMP level was modulated by furosemide that had been previously shown to decrease cAMP content in the rat myometrium tissue. When added to the medium in the pulse mode together with 25 nM OT, furosemide (0.02 mM) increased contraction amplitude by 224%, whereas higher, 0.2 and 20 mM, furosemide concentrations suppressed the response by 286% or totally removed it, respectively. Being present in the bath permanently, 0.2 mM furosemide progressively decreased the amplitude of OT-induced phasic contractions. Under such conditions, 0.02 mM furosemide exerted biphasic effect on the responses, so that the initial enhancement was replaced by the progressive inhibition. Dibutyryl-cAMP (dbcAMP) at a proper concentration restored the responsiveness of the tissue to OT in the presence of furosemide in the saturating concentration. Contractile responses induced by 25 µM OT comprised both phasic and tonic components. In a Ca2+-free medium, the OT-induced contractions seemed to be associated with Ca2+ release from intracellular stores. Permanent presence of furosemide in the CaCl2-containing medium inhibited OT-induced responses in the same manner as omission of Ca2+ from the medium, i.e., furosemide did not affect the responses caused by Ca2+ release but inhibited those mediated via acceleration of the Ca2+ influx. The furosemide-sensitive component of responses to OT was combined with a persistent contraction caused by KCl depolarization; there was a moderate decrease in amplitude of the KCl-induced contracture due to furosemide action. The decrease could be prevented by dbcAMP addition. A fluorescent chelate probe and a Millipore filtration technique have been used to study the effects of β-bungarotoxin (β-toxin) on passive and active Ca++ uptake and ATPase in fragmented sarcoplasmic reticulum (SR) of rabbit skeletal muscle. β-Toxin at 3 × 10?6 M did not affect ATPase activity. In the absence of ATP, β-Toxin increased the passive uptake of Ca++; in the presence of ATP, active Ca++ uptake was inhibited. The effect of β-toxin in SR can be detected at concentrations as low as 10?9 M. The results suggest that β-toxin induces Ca++ leakage in SR membranes.  相似文献

Explants of fetal mouse spinal cord and cerebral cortex generate organotypic slow-wave and repetitive-spike discharges in vitro which can be abolished by agents which reduce the concentration of Ca++ available to the tissue. Synaptically mediated discharges are rapidly blocked in Ca++-free balanced salt solution (BSS), or in regular BSS after addition of 10?3 M EGTA, 5–10 × 10?3 M Mg++, or 10?4 M xylocaine, but simple spike potentials can still propagate. When low concentrations of cyclic AMP or dibutyryl cyclic AMP (2 × 10?6 M) are added to the Ca ++-free BSS or Ca++-antagonist-BSS, a temporary (1–20 min) restoration of characteristic complex bioelectric activities occurs (or the onset of depression is delayed if cyclic AMP is initially added). Phosphodiesterase inhibitors, e.g. 10?3 M caffeine, are also effective in restoring these blockades, whereas 5′AMP and ATP are not. Application of 10?6 M cyclic AMP or 10?3 M caffeine in regular BSS greatly enhances excitability of some CNS explants, resembling convulsive effects observed in CNS in situ. The data suggest that cyclic AMP can mobilize Ca++ from membranebound Ca pools within neurons in CNS explants so as to permit Ca++-dependent release of neurotransmitter during Ca++ deficits. Thus, it may also be that under normal conditions, cyclic AMP can regulate the availability of Ca++ for synaptic transmission in the central nervous system, thereby modulating the efficacy of synaptic functions.  相似文献

1. The longitudinal muscle of the body wall (LMBW) of the holothurian, Isostichopus badionotus contracted when treated with acetylcholine (ACh). The threshold concentration for initiating a contraction was 10−8M ACh.2. Inward calcium (Ca2+) current blockers, diltiazem and verapamil, blocked contractions induced by ACh suggesting that Ca2+ channels are involved. Verapamil caused small rhythmic contractions to occur in some muscle preparations.3. Caffeine initiated contractions only at the high concentration of 10 mM and caused rhythmic contractions in otherwise non-spontaneously beating muscle. Leukotriene (LTC4) is one of the components of Slow Reacting Substance of Anaphylaxis (SRS-A) and is a potent constrictor of guinea pig ilea. The contraction is likely to be a receptor-mediated process. Here we report the existence of specific binding sites for 3H-LTC4 in a crude membrane preparation from guinea pig ileal longitudinal muscle.At 4°C in the presence of 20 mM Serine-borate, binding increases linearly with protein concentration, reaches equilibrium in 10 minutes, and is reversible upon addition of 3 × 10?5M unlabelled LTC4. The dissociation curve is consistent with the existence of more than one class of binding site. Ca++ and Mg++ greatly enhance the binding of 3H-LTC4 at equilibrium. In the presence of 5mM CaCl2 and MgCl2 not only LTC4 (IC50 10?7M), but also LTD4 (albeit with much lower affinity, IC50 = 6 × 10?5M) and the SRS-A antagonist FPL 55712 (IC50 = 10?5M) can compete with 3H-LTC4 for its binding sites. FPL 55712 only displaces 60–70% of the total amount bound, while LTC4 displaces 90–95%.These studies indicate that multiple classes of binding sites exist for 3H-LTC4 in guinea pig ileal longitudinal muscle, and that at least part of these binding sites might be related to the ability of LTC4 to contract guinea pig ilea.  相似文献

The coupling of ion binding to the single strand helix—coil transition in poly (A) and poly(C) is used to obtain information about both processes by ion titration and field-jump relaxation methods. Characterisation of the field-jump relaxation in poly(C) at various concentrations of monovalent ions leads to the evaluation of a stability constant K = 71 M?1 for the ion binding to the polymer. The rate constant of helix formation is found to be 1.3 × 107 s?1, whereas the dissociation rate is 1.0 × 106 s?1. Similar data are presented for poly (A) and poly (dA).The interaction of Mg++ and Ca++ with poly (A) and poly (C) is measured by a titration method using the polymer absorbance for the indication of binding. The data can be represented by a model with independent binding “sites”. The stability constants increase with decreasing salt concentration from 2.7 × 104 M?1 at medium ionic strengths up to 2.7 × 107 M?1 at low ionic strength. The number of ions bound per nucleotide residue is in the range 0.2 to 0.3. Relaxation time constants associated with Mg++ binding are characterised over a broad range of Mg++ concentrations from 5 μM to 500 μM. The observed concentration dependence supports the conclusion on the number of binding places inferred from equilibrium titrations. The rate of Mg++ and Ca++ association to the polymer is close to the limit of diffusion control (kR = 1 × 1010 to 2 × 1010 M?1 s?1). This high rate demonstrates that Mg++ and Ca++ ions do not form inner-sphere complexes with the polynucleotides. Apparently the distance between two adjacent phosphates is too large for a simultaneous site binding of Mg++ or Ca++, and inner sphere complexation at a single phosphate seems to be too weak. The data support the view that the ions like Mg++ and Ca++ surround the polynucleotides in the form of a mobile ion cloud without site binding.  相似文献

Longitudinal and radial movements during spontaneous contractions of isolated segments of terminal ileum of the brushtail possum, a species of arboreal folivore, were studied using high definition spatiotemporal maps. Segments obtained from specimens were continuously perfused with solutions of various apparent viscosities at 3 cm and 5 cm hydrostatic pressure. A series of sustained tetrodotoxin-sensitive peristaltic events occurred during perfusion. The leading edge of each peristaltic event progressed by a succession of rhythmic surges of circular contraction with concerted concurrent phasic longitudinal contractions. Three types of peristaltic event were observed, with differing durations of occlusion and patterns of cyclic, in phase, circular and longitudinal contractions. Each peristaltic event was preceded by a change of shade on the D map that indicated circumferential dilatation. Differences in the slopes of these phasic shade changes from those occurring during peristalsis indicate that this distension is passive and likely results from aboral displacement of fluid. Tetradotoxin insensitive longitudinal contraction waves of frequency 9.2 min−1 occurred during and in the absence of peristalsis, originating at a variety of sites, and propagating either in an orad or aborad direction but predominantly in the latter. Perfusion with 1% guar gum, at 5 cm hydrostatic pressure caused the lumen to become distended and the generation of peristaltic events to cease pending reduction of the hydrostatic head to 3 cm but longitudinal contractile activity was preserved. Neither the frequencies nor the rates of progression of circular and longitudinal contractile events, nor the temporal coordination between these events, varied with the apparent viscosity of the perfusate or altered in a manner that could facilitate mixing.  相似文献

Actin filament bundles isolated from Limulus sperm were used for quantitative electron microscope studies of F-actin assembly. The assembly rate constants were calculated. In addition, the critical concentrations (Cos) for both filament ends were directly determined. In 75 mM KCI and 1–5 mM Mg++, the Cos were 0.1 μM and 0.5 μM for the barbed and pointed ends, respectively. Substitution of Ca++ (20–200 μM) for Mg++ resulted in Cos of 0.4 μM for both filament ends. Consistent with these findings, filament growth occurred only from the barbed ends of Limulus bundles “seeded” into F-actin solutions in KCI and Mg++. Finally, filaments originally grown from the pointed filament ends of Limulus bundles were gradually lost as the actin solution reached steady state. These results demonstrate that actin filaments can “treadmill” under physiological conditions, albeit at very slow rates.  相似文献

Guinea pig vas deferens responds to externally applied acetylcholine (ACh) or noradrenaline (NA) by a small rapid contraction (phasi phase) and then a large contraction (tonic phase). The phasic phase was not affected by removal of external Ca2+, but tonic phase depended on external Ca2+. At lower temperatures the two components became larger and detectable separately. The tonic phase induced by ACh at low temperature (at 20°C) was greatly depressed by brief treatment with colchicine (0.5 μM – 5 μM), although the tonic phase at high temperature (at 37°C) was not affected. Na-induced contraction (phasic or tonic phase) was not changed by the colchicine-treatment. High K+ (40 mM)-contracture, which in many cases consisted of a single phase and depended on external Ca2+, was also not affected by brief treatment with colchicine. Culture of vas deferens for 3 days in the presence of colchicine, increased the phasic phase of ACh- and NA-induced contractions significantly, but reduced the tonic phase of contractions induced by ACh and NA. Colchicine also reduced high K+-contracture, the decrease depending on the period of culture with colchicine. Organ culture with colchicine did not affect the amounts of m-ACh and α-Ad receptors or the IC50 value of ACh and NA on 3H-ligand binding. These results suggest that colchicine specifically interacts with some steps in m-ACh and α-Ad receptor-responsor (e.g. ionophore) coupling without affecting the receptor number or affinity of the receptors for agonists. The mechanisms of action of colchicine are discussed in relation to m-ACh and α-Ad receptor functions.  相似文献

Summary Motile models ofAmoeba proteus prepared by extraction at –20 °C with a 50% buffered glycerol solution showed remarkable contraction on addition of Mg-ATP with retainment of Ca++ sensitivity. The initial contraction around the nucleus occurred on addition of Mg-ATP independently of the Ca++ concentration, and was followed by contractions of three different patterns. In 10–8M Ca++, the granuloplasm contracted as a whole and separated from the membrane leaving no detectable particles in the anterior region. In 10–6M Ca++, the granuloplasm contracted leaving some granules which showed active and vigorous two-directional streaming similar to that in the living cell. Sometimes a part of the plasma membrane twitched. The streaming lasted up to 20 minutes. In 10–4M Ca++, after the initial contraction around the nucleus, no significant movement was observed. All these movements in the models were inhibited by cytochalasin B or N-ethylmaleimide. The relationship between the Mg-ATP and the Ca++ concentrations was examined.  相似文献

Binding of [65Zn++] and [45Ca++] to the acetylcholine (ACh)-receptor, purified from the Torpedo electric organ, was studied by equilibrium dialysis. Whereas [65Zn++] bound to 56 nmoles of sites per mg protein with a dissociation constant of 2.5 × 10−6M, no binding of [45Ca++] at concentrations up to 10−3M could be detected with this method. However, the binding of [acetyl-3H]choline to the receptor was blocked equally by very high Zn++ or Ca++ concentrations, and the Ki for this low affinity binding was 7 × 10−3M. The high affinity binding of [65Zn++] to the receptor was blocked best by Cd++ then Co++ and Mn++, but least by Mg++ and Ca++. When the purified ACh-receptor itself was analyzed for the presence of cations by atomic absorption, it was discovered that 4.7% of its weight was due to bound Ca++ that could not be removed even by extensive dialysis. When Ca++-free solutions (containing 1 mM EDTA) were used during purification, 0.6% of the molecular weight of the receptor was still due to bound Ca++. This was equivalent to 15 moles of Ca++ for each mole of ACh bound at saturation. It is suggested that the source of this Ca++ is endogenous, and that it is tightly bound to the ACh-receptor molecule.  相似文献

Rho-kinase-dependent Ca2+ sensitization is an essential process for contraction of mammalian vascular smooth muscle but the information about its effects in non-mammalian vessels is scarce. We aimed to investigate, using the Rho-kinase inhibitor hydroxyfasudil, the potential role of the Rho-kinase pathway of Ca2+ sensitization in depolarization- and agonist-mediated contraction of chicken embryo (at day 19 of the 21 days of incubation) femoral arteries. Contraction elicited by KCl (125 mM) comprised two phases (phasic and tonic contraction), both of which were abolished in the absence of extracellular Ca2+. Hydroxyfasudil (10 μM) left the initial phasic component nearly intact but abolished the tonic component. Hydroxyfasudil also induced a marked impairment of the contractions elicited by phenylephrine (PE), the thromboxane A2 mimetic U46619, and endothelin-1. In contrast, inhibition of protein kinase C (PKC) by chelerythrine did not affect KCl- or PE-induced contractions, indicating lack of participation of PKC-mediated Ca2+ sensitization. Incubation under chronic hypoxia (15% O2 from day 0) impaired embryonic growth but did not significantly affect hydroxyfasudil-mediated relaxation. In summary, our findings are indicative of a role for Rho-kinase activity in depolarization- and agonist-induced force generation in chicken embryo femoral arteries.  相似文献

Vanadate inhibits the Ca++-ATPase of sarcoplasmic reticulum from pig heart half maximally at about 10?5 M. Mg++ promotes this inhibition by vanadate whereas increasing Ca++-concentrations protect the enzyme against vanadate inhibition. Keeping the ratio Mg++ATP constant there was no influence of ATP on the vanadate inhibition at concentrations up to 5 × 10?3 M ATP. Whenever the ratio Mg++ATP was higher than 1:1 the inhibitory effect of vanadate on the Ca++-ATPase was increased.  相似文献

Concentrations of prostaglandin E1 (PGE1; 10?7 M) that do not elicit tension responses in aortic strips potentiate contractions induced by submaximal concentrations (10?8 ? 10?7 M) of norepinephrine (NE) or angiotensin III (Ang III) but not those of high K+ depolarization or maximal NE or Ang III concentrations. Higher concentrations of PGE1 (10?6 M and above) initiate contractions which are additive with submaximal responses to NE and Ang III but not to K+. These same concentrations of PGE1 also decrease 45Ca retention at high affinity La+++-resistant sites in a manner similar to but not additive with NE and Ang III. Uptake of 45Ca at low affinity La+++-resistant sites (which is increased by high K+-depolarization) is not altered by 10?6 M PGE1. The effects of PGE1 are not altered by decreased extracellular Ca++ (0.1 mM), decreased temperature, phentolamine or meclofenamate. Thus, PGE1 does not appear to increase uptake of extracellular Ca++ in this smooth muscle tissue. Instead, PGE1 increases mobilization of Ca++ from the same high affinity La+++-resistant sites affected by Ang III and NE and, in this manner, may increase responses to these two stimulatory agents.  相似文献

1. Leucomyosuppressin (LMS) inhibited neurally evoked contractions of the hindgut of the cockroach Leucophaea maderae. The threshold for this inhibition of LMS was in the range of 1 × 10−10 M.2. LMS caused a sharp reduction in both l-glutamate and proctolin induced contractions. Dose-response profiles of the effect of LMS (held constant at 10−8M) on variable amounts of proctolin showed an inhibitory effect at 10−9 M proctolin and below, but at 5 × 10−9 M proctolin and above, LMS caused no inhibition.3. Potassium (158 mM) depolarized hindguts treated with LMS (10−8 M) showed a marked reduction (76% ± 2.1) in the proctolin (10−8 M) response.4. When calcium depleted preparations were returned to normal calcium levels (2 mM) in the presence of proctolin (10 −8 M) a contraction occurred that was 45% ± 4 of the maximum in normal saline solution. However, LMS (10−8 M) reduced this response to only 28% ± 2 of the maximum.5. Proctolin (10−8 M) induced contractions in the presence of the manganous ions (2mM) fell to 63% ± 4 of the maximum but on the addition of LMS (10−8M), such responses fell to only 16% ± 5 of the maximum.6. These results offer evidence for a non-synaptic site of action for LMS and a perturbation of key calcium dependent events in the excitation-contraction coupling sequence of visceral muscle by this peptide.  相似文献

The effect of morphine on ATPase of synaptic plasma membranes (SPM) and synaptic vesicles isolated from the mouse brain was studied. The activity of synaptic vesicle Mg++-dependent ATPase from mice rendered morphine tolerant and dependent by pellet implantation was 40% higher than that from placebo implanted mice. However, the activities of Mg++-dependent ATPase and Na+, K+ activated ATPase of SPM of tolerant and nontolerant mice were not significantly different. The activity of synaptic vesicular Mg++-dependet ATPase was dependent on the concentration of Mg++ but not of Ca++; maximum activity was obtained with 2 mM MgCl2. On the other hand, Mg++-dependent ATPase activity of SPM was dependent on both Mg++ and Ca++, activity being maximum using 2 mM MgCl2 and 10?5 M CaCl2. It is suggested that this stimulation of ATPase activity may alter synaptic transmission and may thus be involved in some aspects of morphine tolerance and dependence.  相似文献

Previous studies from this laboratory have indicated that tricyclohexyltin hydroxide (Plictran) is a potent inhibitor of both basal- and isoproterenol-stimulated cardiac sarcoplasmic reticulum (SR) Ca2+-ATPase, with an estimated IC-50 of 2.5 × 10?8M. The present studies were initiated to evaluate the mechanism of inhibition of Ca2+-ATPase by Plictran. Data on substrate and cationic activation kinetics of Ca2+-ATPase indicated alteration of Vmax and Km by Plictran (1 and 5×10?8M), suggesting a mixed type of inhibition. The beta-adrenergic agonist isoproterenol increased Vmax of both ATP- and Ca2+-dependent enzyme activities. However, the Km of enzyme was decreased only for Ca2+ Plictran inhibited isoproterenol-stimulated Ca2+-ATPase activity by altering both and Vmax and Km of ATP as well as Ca2+-dependent enzyme activities, suggesting that after binding to a single independent site, Plictran inhibits enzyme catalysis by decreasing the affinity of enzyme for ATP as well as for Ca2+ Preincubation of enzyme with 15 μM cAMP or the addition of 2mM ATP to the reaction mixture resulted in slight activation of Plictran-inhibited enzyme. Pretreatment of SR with 5 × 10?7M propranolol and 5 × 10?8M Plictran resulted in inhibition of basal activity in addition to the loss of stimulated activity. Preincubation of heart SR preparation with 5 × 10?5M coenzyme A in combination with 5 × 10?8M Plictran partly restored the beta-adrenergic stimulation. These results suggest that some critical sites common to both basal- and beta-adrenergic-stimulated Ca2+-ATPase are sensitive to binding by Plictran, and the resultant conformational change may lead to inhibition of beta-adrenergic stimulation.  相似文献

Prostacyclin (PGI2) produced a biphasic response in canine isolated basilar arteries. In low doses (1 × 10?8M?1 × 10?7M) PGI2 caused a slight but consistent relaxation of resting muscle tone. In low concentrations (1 × 10?8M?1 × 10?6M) PGI2 antagonized muscle contractions caused by serotonin or prostaglandin (PG) F. This relaxant effect with low doses of PGI2 on the isolated cerebral artery contrasts with findings obtained with other PGs and supports the hypothesis that PGI2 is a mediator of vasodilatation. However, in 1 × 10?5M concentrations PGI2 contracted the arterial muscle and did not antagonize contractions induced by serotonin or PGF.  相似文献
