The effect of NH4+ addition to NO3?-growing cells of the non-N2-fixing cyanobacterium Phormidium laminosum (Agardh) Gomont (strain OH-1-pCl1) on photo-synthetic and respiratory electron transport as well as on the intracellular levels of amino acids and some organic acids was studied. Addition of ammonium to nitrate-growing cells resulted in substantial increases in the pool size of most amino acids and a transient decrease in the pool size of organic acids. The high demand for organic acids was partially overcome by degradation of stored carbohydrates, more than by newly fixed carbon, as indicated by the large stimulation of the respiration rate upon ammonium addition. Following ammonium addition, the photosynthetic yield of the in vivo noncyclic electron transport decreased, and the sensitivity of photosystem II to photodamage increased. Results indicate that cells balance their photosynthetic and respiratory activities depending on nitrogen availability and point to an important involvement of respiration in providing energy for ammonium assimilation until adaptation of bioenergetic processes to the new nitrogen source is complete.  相似文献

Heterotrophic growth of the microalga Chlorella vulgaris Beij. in synthetic as well as sterilized municipal wastewater of a nonindustrialized city was measured. The city wastewater contained high levels of ammonium and nitrate, medium levels of phosphate, and low levels of nitrite and organic molecules and could not support heterotrophic growth of C. vulgaris. Evaluation of 11 known carbon sources for this microalga that were added to standard synthetic wastewater containing the same levels of nitrogen and phosphorus as the municipal wastewater revealed that the best carbon sources for heterotrophic growth were Na‐acetate and d ‐glucose. These provided the highest growth rates and the largest removal of ammonium. Growth increased with concentration of the supplement to an optimum at 0.12 M Na‐acetate. This carbon source was consumed completely within 10 d of incubation. Higher concentrations inhibited the growth of C. vulgaris. The microalgal populations under heterotrophic growth conditions were one level of magnitude higher than that under autotrophic growth conditions that served as a comparison. No growth occurred in the dark in the absence of a carbon source. Na‐acetate was superior to d ‐glucose. In municipal wastewater, when Na‐acetate or d ‐glucose was added, C. vulgaris significantly enhanced ammonium removal under heterotrophic conditions, and its capacity was equal to ammonium removal under autotrophic growth conditions. This study showed that sterilized wastewater can be treated by C. vulgaris under heterotrophic conditions if supplemented with the appropriate organic carbon source for the microalgae.  相似文献

The combined effects of ammonium concentration and UV radiation on the red alga Porphyra columbina (Montagne) from the Patagonian coast (Chubut, Argentina) was determined using short‐term (less than a week) experimentation. Discs of P. columbina were incubated with three ammonium concentrations (0, 50, and 300 μM NH4Cl) in anilluminated chamber (PAR=300 μmol photons·m?2·s?1, UVA=15 W·m?2, UVB=0.7 W·m?2) at 15°C. Algae incubated at 300 μM ammonium showed a significant increase (P<0.05) in the concentration of mycosporine‐like amino acids (MAAs) compared with the initial value or with the other ammonium treatments. The increase of MAAs was, however, a function of the quality of irradiance received, with a higher increase in samples exposed to UVA compared with UVB (29% and 5% increase, respectively). However, UVB radiation was more effective in inducing MAA synthesis per unit energy received by the algae. Samples exposed to PAR only had an intermediate increase in MAA concentration of 16%. Chl a concentration decreased through the incubation with the greatest decrease at high ammonium concentration. Phycobiliprotein (BP) decreased through time with the smallest decrease occurring at high ammonium concentration. Photoinhibition (as a decrease of optimal quantum yield) was significantly greater under nitrogen‐deprived conditions than that under replete ammonium levels. Maximal gross photosynthesis (GPmax), as oxygen evolution, and maximal electron transport rate (ETRmax), as chl fluorescence, increased with the ammonium concentration. Positive relationships between maximal GP or ETR and pigment ratios (BP/chl a and MAAs/chl a) and negative relationships with chl a concentration were found.  相似文献

In batch cultures of four Mediterranean strains (from France, Italy, and Spain) of Alexandrium catenella (Whedon et Kof.) Balech growing on a daily light cycle, ammonium and urea uptake were estimated by the 15N tracer technique. Ammonium uptake could be described by Michaelis–Menten kinetics along a substrate gradient of 0.1–10 μgat N · L?1 for the four strains, while two different patterns were observed for urea uptake with Michaelis–Menten kinetics for one strain and linear kinetics for the others. In all cases, an increase in uptake rates with time was noted over the daylight period. This trend led to a net increase in the maximum uptake rate (Vmax; for saturable kinetics) and in the initial slope α. For ammonium, Vmax increased by a factor of 2–10 depending on the strain, and, for urea, the maximal uptake rates measured increased by a factor of 2–18. Temporal variations of half‐saturation constants (Ks) for both nutrients did not show a clear trend. Increases in Vmax and α showed an acclimation of the cells’ uptake system over time to a N pulse, which may be explained by the light periodicity. For two strains, extensive ammonium release was observed during urea assimilation. This mechanism removes urea from the medium, so it is no longer available to other potential competitors, but supplies N back to the medium in the form of ammonium. From a methodological point of view, the phenomenon leads to considerable underestimates of the contribution of urea to phytoplankton growth.  相似文献

The incorporation of 15NH4+ as a function of time and concentration was used to estimate ammonium uptake by Caulerpa cupressoides (West) C. Agardh, taken from its habitat on the sediments of Tague Bay lagoon, St. Croix, U.S. Virgin Islands. 15N-based uptake rates followed Michaelis-Menten type saturation kinetics; the maximum uptake rate was 8.7 ± 3.0 μmol N/g dry wt·h at 26° C and the half-saturation constant was 48 ± 10 μM (X?± SE). The high half-saturatiaon constant reflects the dependence of Caulerpa on sediment pore waters as a nitrogen source. Calculations of uptake from isotope time course data were compared to estimates made from ammonium depletion. More ammonium disappeared than could be accounted for by the incorporation of 15N in Caulerpa, and isotope dilution of the ammonium pool is shown not to be responsible for underestimates of uptake, primarily because large 15N additions (40–300 μM) were used. It is suggested that either (1) a secondary ammonium sink such as wall sorption or bacterial uptake significantly influenced ammonium concentrations, or (2) 15N was lost as labelled dissolved organic nitrogen or volatilized during 15N sample preparation.  相似文献

Chlamydomonas reinhardtii was grown in medium with different carbon (acetate, CO2, or both), and nitrogen (ammonium chloride, peptone, urea) sources and under light of different spectral composition. The light-dark cycles were found more suitable for mixotrophic growth than continuous irradiation. Both blue (BR) and red (RR) radiations decreased photosynthetic capacity of mixotrophic cells compared to “white light” (WL). Effect of RR was associated with photon distribution favouring photosystem 1 (PS1) suggesting increased cyclic phosphorylation. Mixotrophic growth in 10 mM NH4Cl increased photosynthetic oxygen evolution compared to standard concentration of 5 mM NH4Cl used for growing C. reinhardtii. Autotrophic growth stimulated the photosynthetic capacity compared to mixotrophic one. However, higher photosynthetic capacity was achieved for mixotrophic cells by growing them at high NH4 +/K+ ratio and high phosphate concentration. Marine phytoplankton and macroalgae acquire important resources, such as inorganic nitrogen, from the surrounding seawater by uptake across their entire surface area. Rates of ammonium and nitrate uptake per unit surface area were remarkably similar for both marine phytoplankton and macroalgae at low external concentrations. At an external concentration of 1 μM, the mean rate of nitrogen uptake was 10±2 nmol·cm?2·h?1 (n=36). There was a strong negative relationship between log surface area:volume (SA:V) quotient and log nitrogen content per cm2 of surface (slope=?0.77), but a positive relationship between log SA:V and log maximum specific growth rate (μmax; slope=0.46). There was a strong negative relationship between log SA:V and log measured rate of ammonium assimilation per cm2 of surface, but the slope (?0.49) was steeper than that required to sustain μmax (?0.31). Calculated rates of ammonium assimilation required to sustain growth rates measured in natural populations were similar for both marine phytoplankton and macroalgae with an overall mean of 6.2±1.4 nmol·cm?2·h?1 (n=15). These values were similar to maximum rates of ammonium assimilation in phytoplankton with high SA:V, but the values for algae with low SA:V were substantially less than the maximum rate of ammonium assimilation. This suggests that the growth rates of both marine phytoplankton and macroalgae in nature are often constrained by rates of uptake and assimilation of nutrients per cm2 surface area.  相似文献

Effects of ammonium on the photosynthetic recovery of Nostoc flagelliforme Berk. et M. A. Curtis were assayed when being rehydrated in low‐K+ or high‐K+ medium. Its photosynthetic recovery was K+ limited after 3 years of dry storage. The potassium absorption of N. flagelliforme reached the maximum after 3 h rehydration in low‐K+ medium but at 5 min in high‐K+ medium. The K+ content of N. flagelliforme rehydrated in high‐K+ medium was much higher than that in low‐K+ medium. The maximal PSII quantum yield (Fv/Fm) value of N. flagelliforme decreased significantly when samples were rehydrated in low‐K+ medium treated with 5 mM NH4Cl. However, the treatment of 20 mM NH4Cl had little effect on its Fv/Fm value in high‐K+ medium. The relative Fv/Fm 24 h EC50 (concentration at which 50% inhibition occurred) value of NH4+ in high‐K+ medium (64.35 mM) was much higher than that in low‐K+ medium (22.17 mM). This finding indicated that high K+ could alleviate the inhibitory action of NH4+ upon the photosynthetic recovery of N. flagelliforme during rehydration. In the presence of 10 mM tetraethylammonium chloride (TEACl), the relative Fv/Fm 24 h EC50 value of NH4+ was increased to 46.34 and 70.78 mM, respectively, in low‐K+ and high‐K+ media. This observation suggested that NH4+ entered into N. flagelliforme cells via the K+ channel. Furthermore, NH4+ could decrease K+ absorption in high‐K+ medium.  相似文献

The effects of ammonium assimilation on photosynthetic carbon fixation and O2 exchange were examined in two species of N-limited green algae, Chlorella pyrenoidosa and Selenastrum minutum. Under light-saturating conditions, ammonium assimilation resulted in a suppression of photosynthetic carbon fixation by S. minutum but not by C. pyrenoidosa. These different responses are due to different relationships between cellular ribulose bisphosphate (RuBP) concentration and the RuBP binding site density of ribulose bisphosphate carboxylase/oxygenase (Rubisco). In both species, ammonium assimilation resulted in a decrease in RuBP concentration. In S. minutum the concentration fell below the RuBP binding site density of Rubisco, indicating RuBP limitation of carboxylation. In contrast, RuBP concentration remained above the binding site density in C. pyrenoidosa. Compromising RuBP regeneration in C. pyrenoidosa with low light resulted in an ammonium-induced decrease in RuBP concentration below the RuBP binding site density of Rubisco. This resulted in a decrease in photosynthetic carbon fixation. In both species, ammonium assimilation resulted in a larger decrease in net O2 evolution than in carbon fixation. Mass spectrometric analysis shows this to be a result of an increase in the rate of mitochondrial respiration in the light.  相似文献

The effects of nitrate and ammonium addition on net and gross photosynthesis, CO2 efflux and the dissolved inorganic carbon compensation point of nitrogen-limited Selenastrum minutum Naeg. Collins (Chlorophyta) were studied. Cultures pulsed with nitrate or ammonium exhibited a marked decrease in both net and gross photosynthetic carbon fixation. During this period of suppression the specific activity of exogenous dissolved inorganic carbon decreased rapidly in comparison to control cells indicating an increase in the rate of CO2 efflux in the light. The nitrate and ammmonium induced rates of CO2 efflux were 31.0 and 33.8 micromoles CO2 per milligram chlorophyll per hour, respectively, and represented 49 and 48% of the rate of gross photosynthesis. Nitrate addition to cells at dissolved inorganic carbon compensation point caused an increase in compensation point while ammonium had no effect. In the presence of the tricarboxylic acid cycle inhibitor fluoroacetate, the nitrate-induced change in compensation point was greatly reduced suggesting the source of this CO2 was the tricarboxylic acid cycle. These results are consistent with the mechanism of N-induced photosynthetic suppression outlined by Elrifi and Turpin (1986 Plant Physiol 81: 273-279).  相似文献

The possible origin of carbon skeletons for ammonium assimilation in Cyanidium caldarium (Tilden) Geitler was investigated. N-sufficient cells assimilated ammonium at a rate of 182 ± 18 μmol·mL packed cell volume (pcv)-1· h-1. Removal of CO2 or darkening almost immediately prevented ammonium assimilation. N-limited cells in light assimilated ammonium at a rate of 493 ± 45 μmol · mL pcv-1· h-1 in the presence of CO2 and at a lower rate of 168 ± 17 μmol · mL pcv-1· h-1 in the absence of CO2. In darkness they assimilated ammonium at a rate of 293 ± 29 μmol · mL pcv-1 h-1 in the presence of CO2, only 60% of the assimilation rate in light. In the absence of CO2, ammonium was assimilated at a similar rate of 325 ± 14 μmol · mL pcv-1· h-1. Under the latter conditions, however, assimilation was inhibited after 40 min and ceased after 70 min; it resumed upon resupply of CO2. We suggest that N-sufficient cells of C. caldarium obtain carbon skeletons for ammonium assimilation exclusively by photosynthetic reactions. Upon N-limitation they develop the ability, apparently through derepression or activation of regulatory enzyme system(s), to obtain a consistent quantity of additional carbon skeletons and ATP from mobilization of carbon reserves. This enables the N-limited cell to assimilate ammonium not only in light but also in darkness, and at a higher rate than N-sufficient cells. The fact that ammonium assimilation in light occurs at a higher rate than in darkness suggests that ammonium assimilation in light is the sum of both light and dark ammonium assimilation, which implies separate metabolic reactions for the two processes. These results suggest the existence of two distinct and differently controlled pathways in N-limited cells, but not in N-sufficient cells, through which carbon skeletons for ammonium assimilation originate. An important role for dark CO2 fixation in dark or light ammonium assimilation is also indicated.  相似文献

Inorganic phosphate (Pi)-limited chemostat cultures of the green alga Selenastrum minutum were employed to investigate interactions between Pi assimilation, respiration and photosynthetic processes. Changes in net and gross gas exchange rates indicated that O2 evolution decreases during photosynthetic Pi assimilation. Room temperature and 77K Chi a fluorescence measurements revealed that this photosynthetic suppression is correlated with a transition from state 1 to state 2. Substantial photosynthetic Pi uptake rates occur in the presence of DCMU and KCN. Additionally, the cellular ratio of ATP:NADPH increases following Pi enrichment, suggesting that the ratio of cyclic to linear electron flow is enhanced in response to the high energy requirements of Pi uptake. Net starch degradation was observed during photosynthetic Pi assimilation and the cellular pool size of 3-phosphoglycerate increased; however, gross gas exchange parameters and cellular metabolite pool sizes indicated that mitochondrial respiration plays a smaller role during Pi assimilation in the light than it does in the dark. These observations were used to formulate a model depicting possible interactions between photosynthetic electron flow, photosynthetic and respiratory carbon metabolism and metabolite exchange between the chloroplast, cytosol and mitochondrion during photosynthetic Pi assimilation.  相似文献

The ammonium analogue, methylamine, is taken up rapidly from dilute solution by Macrocystis pyrifera (L.) C. A. Agardh. 14C-methylamine was used to characterize the transport system, with respect to dependence on external concentration, temperature, pH and substrate specificity. The results suggest that methylamine enters the algal tissue via a specific mediated transport system. Uptake of methylamine showed no consistent relation to the N content of the plant tissue, but was highly dependent on the portion of plant sampled and severely affected by cutting the tissue. The strong inhibition of methylamine uptake by ammonium and lesser inhibition by other alkylamines suggests that the uptake system functions as an “ammonium permease”. Uptake of 14C-methylamine can be used as a highly sensitive measure of NH4+ uptake activity and should be a useful tool for studying NH4+ uptake in the laboratory and field.  相似文献

After a 3-year exposure to elevated CO2, young trees of Sitka spruce (Picea sitchensis (Bong.) Carr.) were planted in native, nutrient-deficient forest soil and grown for two more years with three CO2 treatments in open-top chambers, and with two nutrient treatments (with and without supplied N). Elevated CO2 resulted in larger fresh mass, dry mass, leaf area and leaf thickness in two-year old needles, but had no effect on one-year old and current needles. Tree height, basal diameter and biomass production also increased, regardless of N supply. In trees without added N, elevated CO2 resulted in higher root-to-shoot and absorbing roots-to-stump ratios. Regardless of N supply, trees grown in elevated CO2 had lower photosynthetic rates on a leaf area basis. Photosynthesis reduction was accompanied by a decline in Rubisco activity and leaf N concentration. Under elevated CO2, added N elevated photosynthesis and Rubisco activity, suggesting a dependence on N availability of the photosynthetic response to elevated CO2. Stomatal conductance of trees grown with added N decreased in response to elevated CO2. This may account for the larger reduction in intercellular CO2 concentration, and hence photosynthesis, in the trees supplied with N than in those without N supply.  相似文献

关于氮素对高大气CO2浓度下C3植物光合作用适应现象的调节机理已有较为深入的研究, 但对其光合作用适应现象的光合能量转化和分配机制缺乏系统分析。该文以大气CO2浓度和施氮量为处理手段, 通过测定小麦(Triticum aestivum)抽穗期叶片的光合作用-胞间CO2浓度响应曲线以及荧光动力学参数来测算光合电子传递速率和分配去向, 研究了长期高大气CO2浓度下小麦叶片光合电子传递和分配对施氮量的响应。结果表明, 与正常大气CO2浓度处理相比, 高大气CO2浓度下小麦叶片较多的激发能以热量的形式耗散, 增施氮素可使更多的激发能向光化学反应方向的分配, 降低光合能量的热耗散速率; 大气CO2浓度升高后小麦叶片光化学淬灭系数无明显变化, 高氮叶片的非光化学猝灭降低而低氮叶片明显升高, 施氮促进PSII反应中心的开放比例, 降低光能的热耗散; 高大气CO2浓度下高氮叶片通过PSII反应中心的光合电子传递速率(JF)较高, 而且参与光呼吸的非环式电子流速率(J0)显著降低, 较正常大气CO2浓度处理的高氮叶片下降了88.40%, 光合速率增加46.47%; 高大气CO2浓度下小麦叶片JF-J0升高而J0/JF显著下降, 光呼吸耗能被抑制, 更多的光合电子分配至光合还原过程。因此, 大气CO2浓度增高条件下, 小麦叶片激发能的热耗散速率增加, 但增施氮素后小麦叶片PSII反应中心开放比例提高, 光化学速率增加, 进入PSII反应中心的电子流速率明显升高, 光呼吸作用被抑制, 光合电子较多地进入光化学过程, 这可能是高氮条件下光合作用适应性下调被缓解的一个原因。  相似文献

Petalonia fascia (Müller) Küntze and Laminaria saccharina (L.) Lamouroux cultured in N-free medium exhibited transient depressions in light-limited and light-saturated gross O2 evolution within 0.3 h following ammonium resupply. Recovery of photosynthesis to pre-resupply rates required 24 h. After 6-9 d of ammonium resupply, photo synthetic rates of N-deprived algae were similar to those of control plants grown with nitrogen. This study demonstrates that N-deprived macroalgae exhibit suppressed photosynthesis upon nitrogen resupply similar to many microalgal species. The apparent slowness of short-term recovery from nitrogen-elicited photo synthetic suppression may have ecological consequences for macroalgae under conditions of patchy nutrient supply.  相似文献

Microcystis aeruginosa Kütz. 7820 was cultured at 350 and 700 μL·L ? 1 CO2 to assess the impacts of doubled atmospheric CO2 concentration on this bloom‐forming cyanobacterium. Doubling of CO2 concentration in the airflow enhanced its growth by 52%–77%, with pH values decreased and dissolved inorganic carbon (DIC) increased in the medium. Photosynthetic efficiencies and dark respiratory rates expressed per unit chl a tended to increase with the doubling of CO2. However, saturating irradiances for photosynthesis and light‐saturated photosynthetic rates normalized to cell number tended to decrease with the increase of DIC in the medium. Doubling of CO2 concentration in the airflow had less effect on DIC‐saturated photosynthetic rates and apparent photosynthetic affinities for DIC. In the exponential phase, CO2 and HCO3 ? levels in the medium were higher than those required to saturate photosynthesis. Cultures with surface aeration were DIC limited in the stationary phase. The rate of CO2 dissolution into the liquid increased proportionally when CO2 in air was raised from 350 to 700 μL·L ? 1, thus increasing the availability of DIC in the medium and enhancing the rate of photosynthesis. Doubled CO2 could enhance CO2 dissolution, lower pH values, and influence the ionization fractions of various DIC species even when the photosynthesis was not DIC limited. Consequently, HCO3 ? concentrations in cultures were significantly higher than in controls, and the photosynthetic energy cost for the operation of CO2 concentrating mechanism might decrease.  相似文献

Ammonium is assimilated in algae by the glutamine synthetase (GS)–glutamine:2‐oxoglutarate aminotransferase pathway. In addition to the assimilation of external ammonium taken up across the cell membrane, an alga may have to reassimilate ammonium derived from endogenous sources (i.e. nitrate reduction, photorespiration, and amino acid degradation). Methionine sulfoximine (MSX), an irreversible inhibitor of GS, completely inhibited GS activity in Ulva intestinalis L. after 12 h. However, assimilation of externally derived ammonium was completely inhibited after only 1–2 h in the presence of MSX and was followed by production of endogenous ammonium. However, endogenous ammonium production in U. intestinalis represented only a mean of 4% of total assimilation attributable to GS. The internally controlled rate of ammonium uptake (Vi) was almost completely inhibited in the presence of MSX, suggesting that Vi is a measure of the maximum rate of ammonium assimilation. After complete inhibition of ammonium assimilation in the presence of MSX, the initial or surge (Vs) rate of ammonium uptake in the presence of 400 μM ammonium chloride decreased by only 17%. However, the amount that the rate of ammonium uptake decreased by was very similar to the uninhibited rate of ammonium assimilation. In addition, the decrease in the rate of ammonium uptake in darkness (in the absence of MSX) in the presence of 400 μM ammonium chloride matched the decrease in the rate of ammonium assimilation. However, in the presence of 10 μM ammonium chloride, MSX completely inhibited ammonium assimilation but had no effect on the rate of uptake.  相似文献

Oxygen inhibition of photosynthesis was studied with intact spinach (Spinacia oleracea L.) chloroplasts which exhibited very high rates of photosynthetic CO2 reduction and were insensitive to additions of photosynthetic intermediates when CO2 was available at saturating concentrations. Photosynthetic rates were measured polarographically as O2 evolution, and the extent of the reduction of substrate was estimated from the amount of O2 evolved. With CO2 as substrate, inhibition of photosynthesis by O2 was dependent on pH. At pH values above 8, rates of O2 evolution were strongly inhibited by O2 and only a fraction of the added bicarbonate was reduced before O2 evolution ceased. The extent of O2 evolution declined with increasing O2 concentration and decreasing initial bicarbonate concentration. At pH 7.2, the initial photosynthetic rate was inhibited about 30% at high O2 levels, but the extent of O2 evolution was unaffected and most of the added bicarbonate was reduced. Photosynthetic O2 evolution with 3-phosphoglycerate as substrate was similarly dependent on pH and O2 concentration. In contrast, there was little effect of O2 and pH on oxaloacetate-dependent oxygen evolution. Acid-base shift experiments with osmotically shocked chloroplasts showed that ATP formation was not affected by O2. The results are discussed in terms of a balance between photosynthetic O2 evolution and O2 consumption by the ribulose diphosphate oxygenase reaction.  相似文献

We measured maximum ammonium uptake rates of the green alga Scenedesmus quadricauda (Turpin) Brébisson and the blue-green alga Microcystis novacekii (Kom.) Comp. grown in nitrogen (ammonium)–limited chemostats. Maximum uptake rates per cellular carbon were larger in S. quadricauda than in M. novacekii. These rates increased with increased specific growth rates. Maximum uptake rates per cellular nitrogen were also larger in S. quadricauda than in M. novacekii. The maximum uptake rates per cellular nitrogen were nearly constant against increased cellular N:C ratios under nitrogen-limited conditions. The higher maximum uptake rates indicate that S. quadricauda had higher uptake abilities for ammonium than M. novacekii when grown under nitrogen limitation. We examined the competition between both species under two distinct nutrient supply modes, using measured maximum uptake values and computer simulations. Microcystis novacekii prevailed in the small-pulse, high-frequency nutrient supply mode, whereas S. quadricauda became competitively superior in the large-pulse, low-frequency nutrient supply mode. These results indicate that we could control nuisance blooms of blue-green algae in lakes and reservoirs by changing the nutrient supply modes.  相似文献
