Abstract

High-field nuclear magnetic resonance measurements were carried out on substance P fragments SP4–11 [pGlu5]-SP5–11 and [pGlu6]SP6–11 both at 400 and at 500 MHz. A spectral simulation was carried out on two of these peptides and the coupling constants were interpreted in terms of the conformations. The JNH-CHa coupling constants are all ~8 Hz, with the exception of glycine, indicating no preferred conformation for the backbone. For the amino acids other than p-Glu, a comparison of the coupling constant data suggests the same relative rotamer populations for the side chains. Proton longitudinal relaxation time data were measured for all three peptides and support the above conclusions.  相似文献

Analogs of Ac-[Nle4]-α-MSH4–11-NH2 and Ac-[Nle4, D -Phe7]-α-MSH4–11-NH2 were prepared with D -isomeric replacements at the His6, Arg8, and Trp9 residues. The requirement for an indole moiety at position 9 also was evaluated by replacement with L -leucine in both parent fragment analogs. D -isomeric replacements at positions 6 and 8 in either series were detrimental to biological potency in frog (Rana pipiens) and lizard skin (Anolis carolinensis) in vitro melanotropic assays. However, Ac-[Nle4, D -Trp9]-α-MSH4–11-NH2 and Ac-[Nle4, D -Phe7, D -Trp9]-α-MSH4–11-NH2 were equipotent and 10 × more potent than Ac-[Nle4]-α-MSH4–11-NH2, respectively, in the lizard skin bioassay, and 30 and 1900 times more potent in the frog skin bioassay. Ac-[Nle4, D -Phe7, D -Trp9]-α-MSH4–11-NH2 was 3 × more potent than α-MSH in the frog skin bioassay. Proton nmr studies in aqueous solution revealed a marked preservation of the backbone conformation of these linear analogs. Chemical-shift variations due to the through-space anisotropic influence of the core aromatic amino acid residues permitted evaluation of side-chain topology. The observed topology was consistent with nonhydrogen-bonded β-like structure (? = ?139°, ψ = +135° for L -amino acids; ? = +139°, ψ = ?135° for D -amino acids) as the predominant solution conformation. The biological and conformational data suggest that high melanotropic potency requires a close spatial arrangement of the His6, Phe7, and Arg8 side chains.  相似文献

Abstract: The regulation of adenylate cyclase activity by adrenocorticotropin/α-melanocyte–stimulating hormone (ACTH/MSH)-like peptides was investigated in rat brain slices using a superfusion method. Adenylate cyclase activity was concentration-dependently increased by ACTH-(1–24), α-MSH (EC50 values 16 and 6 nM, respectively), and [Nle4,D-Phe7]α-MSH (EC50 value 1.6 nM), in the presence of forskolin (1 μM, optimal concentration). 1-9-Dideoxy-forskolin did not augment the response of adenylate cyclase to ACTH-(1–24). Various peptide fragments were tested for their ability to enhance [3H]cyclic AMP production. [Nle4,D-Phe7]α-MSH increased [3H]cyclic AMP formation with a maximal effect of 30% and was more potent than ACTH-(1–24), ACTH-(1–16)-NH2, α-MSH, ACTH-(1–13)-NH2, [MetO4]α-MSH, [MetO24,D-Lys8,Phe9]ACTH-(4–9), ACTH-(7–16)-NH2, ACTH-(1–10), and ACTH-(11–24), in order of potency. This structure–activity relationship resembles that found for the previously described peptide-induced display of excessive grooming. ACTH-(1–24) stimulated adenylate cyclase activity in both striatal (maximal effect, ?20%) and septal slices (maximal effect, ?40%), but not in hippocampal or cortical slices. Lesioning of the dopaminergic projections to the striatum did not result in a diminished effect of [Nle4,D-Phe7]α-MSH on [3H]cyclic AMP accumulation, which indicates that the ACTH/MSH receptor–stimulated adenylate cyclase is not located on striatal dopaminergic terminals. ACTH-(1–24) did not affect the dopamine D1 or D2 receptor–mediated modulation of adenylate cyclase activity. Based on the present data, we suggest that the binding of endogenous ACTH or α-MSH to a putative ACTH/MSH receptor in certain brain regions leads to the activation of a signal transduction pathway using cyclic AMP as a second messenger.  相似文献

A highly potent and selective agonist to the tachykinin NK-3 receptor, [pGlu6, N-MePhe8, Aib9] substance P (6–11) ( I ), was synthesized via the solid phase method. The ED50 of I was 4n M in the guinea pig ileum in the absence of atropine (NK-1 + NK-3 receptors) and this agonist was 5000-fold less potent in the presence of atropine (NK-1 receptor). The analogue was virtually inactive in the rat vas deferens (NK-2 receptor). A detailed analysis of the solution conformation of this analogue in DMSO-d6 and in a DMSO-d6 H2O cryornixture was carried out by a combination of 1H-nmr 2D techniques (DQF-COSY, TOCSY, NOESY and ROESY) and model building based on empirical energy calculations. Peptide I exists as a mixture of isomers containing cis and trans Phe-N-MePhe peptide bonds. The main isomer, containing a cis Phe-N-MePhe peptide bond, shows a preferred folded conformation characterized by a type VI β-turn with Phe and N-MePhe in the i + 1 and i + 2 positions. The turn is followed by a helical segment extending to the C-terminal. This conformation is compared to previously reported conformations of other selective tachykinin agonists and may be a promising lead for the design of novel NK-3 agonists with additional conformational constraints. © 1993 John Wiley & Sons, Inc.  相似文献

α-Melanotropin (α-MSH) retains less than 1% of its original activity after a 60 min incubation with 10% rat brain homogenate. [Nle4, D-Phe7]-α-MSH is nonbiodegradable in rat serum (240 min incubation) and still maintains 10% of its original activity in 10% rat brain homogenate (240 min incubation). The related fragment analogue, Ac-[Nle4, D-Phe7]-α-MSH4–10-NH2, retains 50% of its activity after a 240 min incubation in rat brain homogenate, whereas Ac-[Nle4, D-Phe7]-α-MSH4–11-NH2 is totally resistant to inactivation by rat brain homogenate. Both [Nle4, D-Phe7]-fragments are resistant to degradation by rat serum, but [Nle4]-α-MSH, Ac-[Nle4]-α-MSH4–10-NH2 and Ac-[Nle4]-α-MSH4–11-NH2 are rapidly inactivated under both conditions. The cyclic melanotropin, [ ]-α-MSH, is inactivated in rat brain homogenate as is the shorter Ac-[ ]-α-MSH4–10-NH2 analogue, but neither cyclic melanotropin is inactivated upon incubation in serum from rats. Ac-[ ]-α-MSH4–10-NH2 is resistant to inactivation by either rat serum or a brain homogenate. Some of these melanotropin analogues may provide useful probes for the localization and characterization of putative melanotropin receptors in both the central nervous system and peripheral tissues.  相似文献

Syntheses are described of the nociceptin (1–13) amide [NC(1–13)-NH2] and of several analogues in which either one or both the phenylalanine residues (positions 1 and 4), the arginine residues (positions 8 and 12) and the alanine residues (positions 7 and 11) have been replaced by N-benzyl-glycine, N-(3-guanidino-propyl)-glycine and β-alanine, respectively. The preparation is also described of NC(1–13)-NH2 analogues in which either galactose or N-acetyl-galactosamine are β-O-glycosidically linked to Thr5 and/or to Ser10. Preliminary pharmacological experiments on mouse vas deferens preparations showed that Phe4, Thr5, Ala7 and Arg8 are crucial residues for OP4 receptor activation. Manipulation of Phe1 yielded peptides endowed with antagonist activity but [Nphe1] NC(1–13)-NH2 acted as an antagonist still possessing weak agonist activity. Introduction of the βAla residue either in position 7 or 11 of the [Nphe1] NC(1–13)-NH2 sequence, abolished any residual agonist activity and [Nphe1, βAla7] NC(1–13)-NH2 and [Nphe1, βAla11] NC(1–13)-NH2 acted as competitive antagonists only. Modification of both Ala7 and Ala11 abolished the antagonist activity of [Nphe1]NC(1–13)-NH2 probably by hindering receptor binding. Changes at positions 10 and 11 gave analogues still possessing agonist activity. [Ser(βGal)10] NC(1–13)-NH2 displayed an activity comparable with that of NC(1–13)-NH2, [Ser(βGalNAc)10] NC(1–13)-NH2 and [βAla11] NC(1–13)-NH2 were five and 10 times less active, respectively.The α-amino acid residues are of the l-configuration. Alpha-melanotropin (alpha-melanocyte stimulating hormone, alpha-MSH) is a hormone produced by the pituitary gland of most vertebrate animals. This melanotropic peptide, Ac-Ser-Tyr-Ser-Met-Glu-His-Phe-Arg-Trp-Gly-Lys-Pro-Val-NH2, regulates melanin pigmentation of the skin of some mammals. Although MSH may be absent from the human pituitary gland, this peptide can stimulate pigment formation in human skin. We have synthesized several analogues of alpha-MSH, which are superpotent, prolonged-acting, and resistant to inactivation by serum enzymes. One such analogue, [NLe4, D-Phe7]alpha-MSH, has proven particularly useful in a number of physiological studies. In addition, some [Nle4, D-Phe7]-substituted fragment analogues of MSH are even more active than the native hormone, alpha-MSH. For example, these analogues are 100–1,000 times more active than alpha-MSH in stimulating S-91 mouse melanoma tyrosinase activity in vitro. We have successfully labeled one such peptide to high specific activity; this melanotropin, [3H]-Ac-[Nle4, D-Phe7]alpha-MSH4–11NH2, has been shown by others to bind to B16 melanoma cells. We have also conjugated several ligands (fluorescein and biotin) to [Nle4, D-Phe7]alpha-MSH. These melanotropin conjugates might prove useful for melanotropin receptor studies and for the clinical localization of metastatic melanoma. We have demonstrated that [Nle4, D-Phe4]alpha-MSH can be topically applied and transdermally delivered across the skin of mice and humans in vitro, as determined by bioassay and RIA. Initial toxicologic studies indicate that the analogue is nontoxic to mice and is not mutagenic. Studies are underway to determine whether this analogue may prove useful as a “tanning hormone” for increasing the pigmentation of light-skinned individuals or possibly even for treating people with certain hypopigmentary disorders.  相似文献

Electrical stimulation of the isolated rabbit iris sphincter muscle in the presence of atropine gives rise to a contraction that can be blocked by tachykinin antagonists. The ability of a series of novel tachykinin antagonists to inhibit the contractile effect of SP on the guinea-pig taenia coli and to suppress the electrically evoked contraction of the atropinized rabbit iris sphincter was tested. Several of the novel antagonists were found to be more potent in terms of pA2 and pIC50 values than the two previously described analogs, [d-Pro2, d-Trp7,9]SP-(1–11) and [d-Arg1, d-Trp7,9, Leu11]SP-(1–11) (Spantide). Apart from d-Trp in positions 7 and 9 the characteristic features of the potent novel antagonists were d-Cl2Phe (or d-Cys(Bzl)) in position 5, Asn in position 6 and Nle in position 11. In addition Pal in position 3 seemed to offer an enhanced potency.  相似文献

13NO3 and 13NH4+ compartmental analyses were carried out in seedling roots of trembling aspen (Populus tremuloides Michx.), lodgepole pine (Pinus contorta Dougl. ex Loud. var. latifolia Engelm.) and interior Douglas-fir (Pseudotsuga menziesii var. glauca [Beissn.] Franco) at 0·1 and 1·5 mol m–3 external NO3 or NH4+ concentrations ([NO3]o or [NH4+]o, respectively). At the lower [NO3]o, the capacities and efficiencies of acquisition and accumulation of NO3, based upon NO3 fluxes and cytoplasmic NO3 concentrations ([NO3]c), were in the order aspen >> Douglas-fir > pine. At 1·5 mol m–3[NO3]o, the NO3 influx increased 18-fold in pine, four-fold in Douglas-fir and approximately 1·4-fold in aspen; in fact, at 1·5 mol m–3[NO3]o, the NO3 influx in pine was higher than in aspen. However, at high [NO3]o, efflux also increased in the two conifers to a much greater extent than in aspen. In aspen, at both [NO3]o, approximately 30% of the 13N absorbed was translocated to the shoot during 57 min of 13N loading and elution, compared with less than 10% in the conifers. At 0·1 mol m–3[NH4+]o, influx and net flux were in the order: aspen > pine > Douglas-fir but the differences were much less than in NO3 fluxes. At 1·5 mol m–3[NH4+]o, NH4+ influx, efflux and [NH4+]c greatly increased in aspen and Douglas-fir and, to a much lesser extent, in pine. In aspen, 29 and 12% of the 13N absorbed was translocated to the shoot at 0·1 and 1·5 mol m–3[NH4+]o, respectively, compared with 5 to 7% in the conifers at either [NH4+]o. These patterns of nitrogen (N) uptake, particularly in the case of NO3, and the observed concentration responses of NO3 uptake, reflect the availability of N in the ecological niches, to which these species are adapted.  相似文献

The redox and second-order nonlinear optical (NLO) properties of organic–inorganic hybrid polyoxoanions [LNbOEMe3]3 ? , [LNbOEPh3]3 ?  (L = α-{PW11O39}7 ? , E = Si, Ge, Sn, Pb) are investigated by density functional theory method. The element substitution effects on the molecular nonlinear response have been analysed. The results show that the computed β0 values depend on both E (E = Si < Ge < Sn < Pb) and the organic groups connected with E. For [LNbOEPh3]3 ?  (L = α-{PW11O39}7 ? , E = Si, Ge, Sn, Pb), the analysis of major contributions to β0 value suggests that the charge transfer from organic group to Keggin polyanion plays the key role in NLO response; the polyanion acts as a donor, whereas the organic group acts as an acceptor in [LNbOEPh3]3 ? . Our results show that this kind of organic–inorganic hybrid compound possesses larger molecular second-order polarisability and might be a potential NLO material.  相似文献

Abstract: Primary cultures of chromaffin cells from bovine adrenal medulla were used to evaluate the ability of several opiates to reduce the release of catecholamines induced by stimulation of nicotinic receptors. Etorphine, β-endorphin, Met-enkephalin[Arg6,Phe7], and the synthetic peptide [d -Ala2,Me-Phe4,Met(O)s-ol]enkephalin inhibited the acetylcholine-induced release of catecholamines with an IC30 varying from 10?7 to 1 × 10?6M. The effect was stereospecific because levorphanol (IC30= 7.5 × 10?7M) was approximately two orders of magnitude more potent than dextrorphan. Morphine (μ-receptor agonist), [d -Ala2, d -Leu5]enkephalin (δ-receptor agonist), ethylketazocine (k -receptor agonist), and N-allylnormetazocine (σ-receptor agonist) were at least 100–1000 times less potent than etorphine. Diprenorphine (IC50= 5 × 10?7M) and naloxone (IC50= 10?6M) antagonized the effect of etorphine. High-affinity, saturable, and stereospecific binding sites for [3H]etorphine, [3H]dihydromorphine, [3H-d -Ala2,d -Leu5]enkephalin, [3H]ethylketazocine, and for [3H]N-allylnormetazocine, [3H]diprenorphine, and [3H]naloxone were detected in chromaffin cell membranes and in membranes obtained from adrenal medulla homogenates. However, the number of binding sites for [3H]etorphine and [3H]diprenorphine was 10–70 times higher than the number of sites measured with the other 3H ligands. The rank order of potency of these compounds for the displacement of [3H]etorphine binding correlates (r = 0.90) with the rank order of potency of the same compounds for the inhibition of acetylcholine-induced catecholamine release. These data suggest that a stereoselective opiate receptor (different from the classic μ-, δ-, k -, or σ-receptor) with high affinity for etorphine, diprenorphine, β-endorphin, and Met-enkephalin[Arg6,Phe7] modulates the function of the nicotinic receptor in adrenal chromaffin cells.  相似文献

This study contributes to the investigation related to guest–host interactions between the chemotherapeutic agent cisplatin and a functionalised silica matrix in order to improve and find new materials such as drug carriers. The adsorption of cisplatin and its complexes, cis-[PtCl(NH3)2]+ and cis-[Pt(NH3)2]2+, on a SH-functionalised SiO2(111) surface has been studied by the atom superposition and electron delocalisation method. The adiabatic energy curves for the adsorption of the drug and its products on the delivery system were considered. The electronic structure and bonding analysis were also performed. The molecule and their complex are adsorbed on the functionalised surface resulting in a major absorption of the cis-[Pt(NH3)2]2+ complex. The molecule–surface interactions are formed via –SH group. The molecule/complexes SH electron-donating effect plays an important role in the catalytic reaction. The more important drug–carrier interactions occur through the Cl–H bond for the adsorption of cis-[PtCl2(NH3)2] and cis-[PtCl(NH3)2]+, and through the Pt–S and Pt–H interactions for cis-[Pt(NH3)2]2+ adsorption. When the new interactions are formed, the functionalised carrier maintains their matrix properties while the molecule is the most affected after adsorption. The Pt atomic orbitals present the most important changes during adsorption.  相似文献

Electrical field stimulation of guinea-pig isolated hilus bronchi induced tetrodotoxin-sensitive contractions of which only a minor part could be inhibited by atropine. The remaining non-cholinergic bronchonstriction was antagonized by a heptapeptide and an undecapeptide substance P (SP) analogue (Arg5, D-Trp7, 9) SP5–11, IC50 = 24.0 μM and (D-Pro2, D-Trp7, 9)SP, IC50 = 10.0 μM. Of the exogenously added tachykinins, both eledoisin (8 times) and physalaemin (3 times) were more potent bronchoconstrictors than SP. Pretreatment with the SP-analogues shifted concentration-response curves to the tachykinins to the right, eledoisin being most readily antagonized. (Arg5, D-Trp7, 9)SP5–11 also antagonized the neural response more readily than that of SP. In addition, in the frog isolated sciatic nerve preparation the two SP-analogues were found to possess potent lidocaine-like neurodepressant actions which further complicated the interpretation of the neural inhibitory effects of these compounds. It is concluded that if a tachykinin contributes to non-cholinergic bronchoconstriction, an eledoisin-like peptide is a more likely candidate than SP itself. Since SP-antagonists may have local anaesthetic properties their value as tools in neurophysiology seems limited. Inferentially, the non-cholinergic bronchoconstrictive neurotransmitter remains to be identified.  相似文献

The fate of 6α- and 6β-hydrogens of lathosterol during the transformation into 20-hydroxyecdysone was chased by feeding [3α,6β-2H2]- and [3α,6α-2H2]-lathosterols to hairy roots of Ajuga reptans var. atropurpurea. The behavior of 6β-hydrogen, which mostly migrated to the C-5 position of 20-hydroxyecdysone, was in agreement with that of C-6 hydrogen of cholesterol. The results strongly supported the view that cholesterol and lathosterol are first metabolized into 7-dehydrocholesterol, which is then converted into 20-hydroxyecdysone via 7-dehydrocholesterol 5α,6α-epoxide in the hairy roots.  相似文献

Copper(I) and silver(I) complexes of formulae [Cu(NCCH3)4]+[A] ([A] = [B(C6F5)4] (1), {B[C6H3(CF3)2]4} (2), [(C6F5)3B–C3H3N2–B(C6F5)3] (3), and [Ag(NCCH3)4]+[B(C6F5)4] (4) are examined with particular emphasis on the strength of their M–N bond and its influence on the catalytic performance of these complexes in cyclopropanation and aziridination. To examine the strength of the M–N interactions, vibrational spectra of the related hydrogenated and deuterated species [Cu(NCCH3)4]+, [Cu(NCCD3)4]+, [Ag(NCCH3)4]+, and [Ag(NCCD3)4]+ are also determined. It is found that the metal–nitrile bond strength is an important factor for the catalytic activity of the respective complexes.  相似文献

DNA photolyases (EC 4.1.99.3) are enzymes that catalyze photoreversion of cis,syn-thymine photodimer (T[c,s]T), which is one of major photolesion products in DNA, by utilizing UV light. In this work, we have designed and synthesized Zn2+–1,4,7,10-tetraazacyclododecane complexes bearing a lumiflavin and l-tryptophan (ZnL3) or l-phenylalanine (ZnL4) as artificial DNA photolyases. We have found that (ZnL3)red, whose flavin unit was reduced in situ by Na2S2O4, accelerates the photoreversion of T[c,s]T utilizing near-UV light in aqueous solution at pH 7.6 and 11. Interestingly, more efficient photoreversion of T[c,s]T was achieved by UV irradiation of an oxidized form of ZnL3 [(ZnL3)ox] in the presence of an excess amount of Et3N at pH 11. UV–vis and fluorescence measurements and action spectra showed that an oxidized form of flavin of (ZnL3)ox was photoreduced by Et3N into its reduced form (ZnL3)red, which promoted the photoreduction of T[c,s]T. Comparison of the photochemical properties of ZnL3 with those of ZnL4 suggested that a tryptophan unit in ZnL3 contributed to the stabilities of the flavin through intramolecular photoinduced electron transfer.Electronic supplementary material Supplementary material is available in the online version of this article at and is accessible for authorized users.  相似文献

The reference standard methyl (2-amino-5-(benzylthio)thiazolo[4,5-d]pyrimidin-7-yl)-d-leucinate (5) and its precursor 2-amino-5-(benzylthio)thiazolo[4,5-d]pyrimidin-7-yl)-d-leucine (6) were synthesized from 6-amino-2-mercaptopyrimidin-4-ol and BnBr with overall chemical yield 7% in five steps and 4% in six steps, respectively. The target tracer [11C]methyl (2-amino-5-(benzylthio)thiazolo[4,5-d]pyrimidin-7-yl)-d-leucinate ([11C]5) was prepared from the acid precursor with [11C]CH3OTf through O-[11C]methylation and isolated by HPLC combined with SPE in 40–50% radiochemical yield, based on [11C]CO2 and decay corrected to end of bombardment (EOB). The radiochemical purity was >99%, and the specific activity (SA) at EOB was 370–1110 GBq/μmol with a total synthesis time of ~40-min from EOB. The radioligand depletion experiment of [11C]5 did not display specific binding to CX3CR1, and the competitive binding assay of ligand 5 found much lower CX3CR1 binding affinity.  相似文献

The interaction of a series of biologically active gastrin fragments with calcium ions has been investigated by CD in trifluoroethanol. It was found that the gastrin octapeptide pGlu10,Nle15-HG[10–17] binds one calcium ion per molecule. The hypothesis is made that the binding involves the C-terminal, biologically important tetrapeptide. When the chain is elongated to the gastrin nonamer pGlu9,Nle15-HG[9–17], a second binding site is available, which is most likely situated at the N-terminal part of the molecule. Further elongation of the peptide chain up to the dodecapeptide pGlu6,Nle15-HG[6–17] does not provide any additional binding site. Saturation of the two sites in the shorter peptides produces different changes in the chiroptical properties in the near- and far-uv. As the chain is elongated, this difference tends to disappear. This result is consistent with an increased conformational order of the longer peptides. In the shorter fragments, the strength of this second binding is appreciably lower than that of the first, while in the longer peptides, the strength of the two bindings is comparable. On the assumption that the variation of the CD properties is proportional to the extent of binding, the constant for the binding of the second ion was determined to be of the order of 5 × 105 L/mol for the nonapeptide.  相似文献

Gomesin (Gm) was the first antimicrobial peptide (AMP) isolated from the hemocytes of a spider, the Brazilian mygalomorph Acanthoscurria gomesiana. We have been studying the properties of this interesting AMP, which also displays anticancer, antimalarial, anticryptococcal and anti‐Leishmania activities. In the present study, the total syntheses of backbone‐cyclized analogues of Gm (two disulfide bonds), [Cys(Acm)2,15]‐Gm (one disulfide bond) and [Thr2,6,11,15,d ‐Pro9]‐Gm (no disulfide bonds) were accomplished, and the impact of cyclization on their properties was examined. The consequence of simultaneous deletion of pGlu1 and Arg16‐Glu‐Arg18‐NH2 on Gm antimicrobial activity and structure was also analyzed. The results obtained showed that the synthetic route that includes peptide backbone cyclization on resin was advantageous and that a combination of 20% DMSO/NMP, EDC/HOBt, 60 °C and conventional heating appears to be particularly suitable for backbone cyclization of bioactive peptides. The biological properties of the Gm analogues clearly revealed that the N‐terminal amino acid pGlu1 and the amidated C‐terminal tripeptide Arg16‐Glu‐Arg18‐NH2 play a major role in the interaction of Gm with the target membranes. Moreover, backbone cyclization practically did not affect the stability of the peptides in human serum; it also did not affect or enhanced hemolytic activity, but induced selectivity and, in some cases, discrete enhancements of antimicrobial activity and salt tolerance. Because of its high therapeutic index, easy synthesis and lower cost, the [Thr2,6,11,15,d ‐Pro9]‐Gm analogue remains the best active Gm‐derived AMP developed so far; nevertheless, its elevated instability in human serum may limit its therapeutic potential. New α,β-unsaturated ketones 4a,b; 5a–c; and 6a,b; as well as 4-H pyran 7; pyrazoline 8a,b; isoxazoline 9; pyridine 10–11; and quinoline-4-carboxylic acid 12a,b derivatives were synthesized and evaluated for in vitro antitumour activity against HepG2, MCF-7, HeLa, and PC-3 cancer cell lines. Antioxidant activity was investigated by the ability of these compounds to scavenge the 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) radical cation (ABTS?+). Compounds 6a, 6b, 7, and 8b exhibited potent antitumour activities against all tested cell lines with [IC50] ?5.5–18.1 µΜ), in addition to significantly high ABTS?+ scavenging activities. In vitro EGFR kinase assay for 6a, 6b, 7, and 8b as the most potent antitumour compounds showed that; compounds 6b, and 7 exhibited worthy EGFR inhibition activity with IC50 values of 0.56 and 1.6?µM, respectively, while compounds 6a and 8b showed good inhibition activity with IC50 values of 4.66 and 2.16?µM, respectively, compared with sorafenib reference drug (IC50?=?1.28?µM). Molecular modelling studies for compounds 6b, 7, and 8b were conducted to exhibit the binding mode towards EGFR kinase, which showed similar interaction with erlotinib.  相似文献
