The problem of the functioning specificity of sex chromosomes during the early stages of embryogenesis in man and the associated problem of the sex ratio in spontaneous and induced abortions, as well as in newborns, remains open. We have conducted a cytogenetic examination of 342 spontaneous abortions divided into three clinical groups on the basis of the severity of the developmental disturbances of the embryo: spontaneous abortionssensu stricto with a developed embryo without any significant intrauterine delay of development (n=100), nondeveloping pregnancies (n=176), and anembryonic fetuses (n=66). The frequency of chromosomal mutations in these groups was 22.0, 48.3, and 48.5%, respectively. Statistical analysis has demonstrated significant differences between the studied groups in the frequencies of the normal and abnormal karyotypes: the major contributions to these differences were associated with autosomal trisomy, triploidy, and the 46.XY karyotype. The presence of 46.XY may reflect the specific genetic mechanisms of the prenatal mortality of embryos with the normal karyotype, associated with sex and/or with the imprinting of X-chromosomes. The sex ratio in spontaneous abortions with the normal karyotype was as follows: 0.77 for spontaneous abortions with well-developed embryos without any significant intrauterine delay of development; 0.60 for nondeveloping pregnancies; and 0.31 for anembryonic fetuses. An analysis of DNA from the embryos and their parents has demonstrated a low probability of contamination of cell cultures with mother cells as a possible source of the prevalence of embryos with the 46.XX karyotype among spontaneous abortions. Nondeveloping pregnancies and anembryonic fetuses showed statistically significant differences in the sex ratio from the control group consisting of medical abortions (1,11). Differences in the sex ratio were due to an increasingly lower proportion of embryos with karyotype 46.XY (relative to the expected one) among the fetuses with an increased severity of developmental disturbances. The statistical “chances ratio” index also provided evidence that embryos with the 46.XY karyotype had a higher propensity to produce a well-formed fetus as compared with the female embryos. We propose that the expression of genes of the maternal X-chromosome in XY embryos supports a more stable development during early embryogenesis as compared with XX embryos. In the latter case, normal development is coupled with the operation of an additional mechanism for compensation of the dose of X-linked genes. Operation of this mechanism increases the probability of disturbances in female embryos. A higher viability of XY embryos during the early stages of ontogenesis in man appears to explain their underrepresentation in samples of spontaneously aborted embryos and appears to be the major factor responsible for the deviation of the sex ratio from the theoretically expected value.  相似文献

In the present study, using immunohistochemistry, we studied the expression of CD30 and CD30-L in 35 deciduas obtained from women following elective abortion during normal physiological gestation and in 60 deciduas obtained from women after spontaneous abortion with or without signs of inflammation. The main difference was noticed in the first trimester of gestation in which was found a decrease in CD30/CD30-L-positive decidual glandular and stromal cells in a greater number of cases of spontaneous abortions with respect to cases of physiological pregnancies (70% vs 50%, p<0.05). In addition, deciduas from spontaneous abortions with inflammation and without inflammation reacted similarly. The reduced expression of CD30 and CD30-L and their cellular pattern detected in the deciduas from spontaneous abortions suggest that the CD30/CD30-L system is crucial for preventing abortions in the first trimester. Furthermore, the distinctive expression of CD30/CD30-L in deciduas from physiological pregnancies may indicate that the CD30/CD30-L system exerts its main role in the first trimester.  相似文献

1. Twenty-five solutions which contained KCl (0.0, 0.2, 0.4, 0.6, and 0.8 gm. per liter), in combination with CaCl2 (0.0, 0.2, 0.4, 0.6, and 0.8 gm. per liter), 10.0 gm. of NaCl, and 0.2 gm. of NaHCO3 per liter of solution were tested in order to determine satisfactory KCl/CaCl2 ratios in an insect physiological salt mixture for the maintenance of muscular activity by the isolated crop of the American roach. Satisfactory activity products (0.390 to 0.549) were obtained in seven mixtures with KCl/CaCl2 ratios of 0.2/0.2, 0.4/0.4, 0.6/0.6, 0.8/0.8, 0.2/0.4, 0.4/0.6, and 0.6/0.8, expressed as gram per liter. These ratios lie between 0.50 and 1.00. In solutions which contained calcium, but no potassium, approximately 50 per cent of the crops exhibited an initial tone increase and were arrested in rigor. See Fig. 2. In solutions which contained potassium, but no calcium, all crops showed an initial loss of tone and arrest in relaxation. See Fig. 2. 2. Seven KCl/CaCl2 ratios (see paragraph 1 above) were tested with eight NaCl concentrations (1.0, 1.2, 1.3, 1.4, 1.5, 1.6, 1.7, and 1.8 per cent) at a pH of 8.0. In these mixtures, the ones with KCl/CaCl2 ratios of less than 1.0 produced higher activity products than those with ratios equal to 1.00. The highest average activity product (0.849) was obtained in the solutions with 0.2 gm. of KCl and 0.4 gm. of CaCl2 per liter. 3. Four KCl/CaCl2 ratios (0.2/0.2, 0.4/0.4, 0.2/0.4, and 0.4/0.6 gm. per liter) were tested with 1.4, 1.5, and 1.6 per cent NaCl at a pH of 7.5. When analyzed with data from comparable solutions at a pH of 8.0, it was found that 1.4 per cent NaCl afforded an optimum environment for isolated crop activity. 4. Effects of hydrogen and hydroxyl ion concentrations were studied at pH values of 6.8, 7.5, 8.0, and 8.9. The highest average activity product, 1.011, was produced at a pH of about 8.0. 5. A satisfactory physiological salt solution for the isolated foregut of the American roach, Periplaneta americana, would contain 14.0 gm. of NaCl, 0.4 gm. of CaCl2, 0.2 gm. of KCl, and 0.2 gm. of NaHCO3 per liter of solution. This mixture should have a pH value between 7.8 and 8.2. 6. Durations of crop activity extending over periods as long as 25 hours were quite common, and several crops maintained contractions for more than 30 hours. The greatest longevity was for crop 814, from a female, which continued activity for slightly more than 47 hours. 7. A significant difference between the activity products of the crops from males and the crops from females was recorded. Although there was not a significant difference in the amount of food ingested by males and females, 12 hours after feeding there was more food in the females'' crops, and the food progressed more rapidly through the males'' crops than through the females''. In addition, crops from the two sexes reacted differently to the effects of day old solutions. This sex difference is apparently related to an inherently increased activity of the crop from the male roach.  相似文献

The Dead Sea is a hypersaline water body. Its total dissolved salts content is on the average 322.6 gm/liter. The dominant cation is Mg (40.7 gm/liter), followed by Na (39.2 gm/liter), Ca (17 gm/liter) and K (7 gm/liter). The major anion is Cl (212 gm/liter), followed by Br (5 gm/liter); SO4 and HCO3, are very minor. The lake contains a limited variety of microorganisms and no higher organisms. The number of recorded species is very low, but the total biomass is reasonably high (about 105 bacteria/ml and 104 algal cells/ml). The indigenous flora is comprised mainly of obligate halophylic bacteria, such as the pink, pleomorphicHalobacterium sp., aSarcina-like coccus, and the facultative halophilic green alga,Dunaliella. Sulfate reducers can be isolated from bottom sediments. Recently a unique obligate magnesiophile bacteria was isolated from Dead Sea sediment. Several of the Dead Sea organisms possess unusual properties. TheHalobacterium sp. has extremely high intercellular K+ concentration (up to 4.8M) and extraordinary specificity for K+ over Na. TheDunaliella has very high intracellular concentration of glycerol (up to 2.1M). The microorganisms exert marked influence on some biogeochemical processes occurring in the lake, such as the control of the sulfur cycle and the formation and diagenesis of organic matter in the sediments. The Dead Sea is an excellent example of the development of two different mechanisms for adjusting to a hostile environment. The algae adjust to the high salinity by developing a mechanism for the exclusion of salts from the intracellular fluid and using glycerol for osmotic regulation. On the other hand, the bacteria adapt to the environment by adjusting their internal inorganic ionic strength, but not composition, to that of the medium. The problem of population dynamics and limiting factors for algal and bacterial productivity are discussed in view of the total absence of zooplankton and other consumers other than bacteria.  相似文献

Summary A simple and rapid procedure for direct organogenesis from root nodulelike structures of alfalfa (Medicago sativa L.) line SGg, spontaneously induced on growth regulator-free Gamborg (B5) medium, was developed. Prolific adventitious shoot initiation was obtained using a combination of 1.0 mg/liter TIBA and 0.5 mg/liter 2iP. Transfer of shoots to a medium containing 0.5 mg/liter ABA and reduced concentration of TIBA (0.5 mg/liter) before rooting markedly stimulated shoot development. Regenerated shoots rooted easily and revealed the early appearance of nodulelike structures on basal medium (B5) lacking growth regulators. Analysis of endogenous growth regulator levels of SGg roots maintained on growth regulators free media, showed that spontaneous shoot appearances was correlated with high cytokinin-to-auxin ratios.  相似文献

Objectives

The aim of this study was to determine whether the concentration of disintegrin and metalloprotease protein12 (ADAM12) in first trimester maternal serum can be used as a marker for first-trimester complete spontaneous abortions, missed abortions, ectopic pregnancies and hydatidiform moles.

Methods

The maternal serum concentrations of ADAM12 were measured in the range of 5–9+6 weeks of gestation using an automated AutoDelfia immunoassay platform in 9 cases of complete spontaneous abortion, 27 cases of missed abortions, 56 cases of ectopic pregnancies, 12 cases of hydatidiform moles, and 100 controls. Logistic regression analysis was used to determine significant factors for predicting adverse pregnancy outcomes in early pregnancy. Screening performance was assessed using receiver operating characteristic curves.

Results

Two hundred and four women were enrolled in the study. In the control group, the level of ADAM12 increased with gestational age. The median ADAM12 levels in the spontaneous abortion (0.430 MoM), ectopic pregnancy (0.460 MoM) and hydatidiform mole (0.037 MoM) groups were lower than that in the control group, while the median ADAM12 level in the missed abortion group (1.062 MoM) was not significant from the controls (1.002 MoM). Logistic regression analysis demonstrated that the level of ADAM12 in maternal serum facilitated the detection of ectopic pregnancies (OR = 0.909; 95% CI = 0.841∼0.982) and complete spontaneous abortion (OR = 0.863; 95% CI = 0.787∼0.946).

Conclusions

In complete spontaneous abortion and ectopic pregnancy, ADAM12 maintained at low levels in early pregnancies, and there were significant differences compared to normal pregnancies. ADAM12 is a promising marker for the diagnosis of complete spontaneous abortion and ectopic pregnancy in symptomatic women, and under certain conditions, ADAM12 can diagnose ectopic pregnancy and spontaneous abortion before an ultrasonographic detection of the conditions.  相似文献

Summary Elementary Na+ currents were recorded at 19°C in cell attached and inside-out patches from cultured neonatal rat cardiocytes in order to study the effect of cAMP and other 6-aminopurines.The treatment of the cardiocytes with db-cAMP (1×10–3 mol/liter) led to a decline of reconstructed macroscopic peakI Na to 62±7.6% of the initial control value. This reduction in NP0 was mostly accompanied by a decrease in burst activity. Openstate kinetics were preserved even in DPI-modified, noninactivating Na+ channels. Since the stimulator of the adenylate cyclase, forskolin (1×10–6 mol/liter), evoked a similar pattern of response, the NP0 decrease can be considered as the functional correlate of Na+ channel phosphorylation brought about by cAMP-dependent protein kinase. As found in inside-out patches, cAMP (1×10–3 mol/liter) remained effective under cell-free conditions and reduced reconstructed macroscopic peakI NA to about 50% of the initial control value when the absence of Mg-ATP at the cytoplasmic membrane surface prevents phosphorylation reactions. A very similar response developed in the cytoplasmic presence of other 6-aminopurines including ATP (1×103 mol/liter), adenosine (1×10–4 mol/liter), adenine (1×10–5 mol/liter) and hypoxanthine (1×10–5 mol/liter). This susceptibility to adenine suggests that cardiac Na+ channelsin situ could sense intracellular fluctuations of adenine nucleotides, most likely of ATP.  相似文献

Summary By using rifampicin to increase the rate of and synthesis in a heterodiploid strain of E. coli carrying the mutation rif pr (Km7), which codes for a rifampicin sensitive RNA polymerase to which the drug binds weakly, and the dominant mutation rif D R , which codes for a rifampicin resistant RNA polymerase, the concentration of these subunits in the cell was increased 1.6 fold. Measurements made after removal of rifampicin from the cells showed that the excess and subunits did not reduce the rate of their own synthesis below normal.  相似文献

We documented normal pregnancy and the rate of pregnancy failure in female baboons by measuring chorionic gonadotropin (bCG) and progesterone (P) levels in 162 mated cycles of 70 baboon females on days 10, 12, and 14 postovulation. Females were mated with males during turgescene. The presence of pregnancy was defined by bCG levels >20 μg/ml by day 14 postovulation and/or documentation of a gestational sac using ultrasonography. Of the 162 cycles, 75 were fertile. Of these animals, 33 were used in other studies and thus were not included in these analyses. The analyses are based on 43 pregnancies from 91 cycles that were untreated throughout their gestations. Twenty-six of these pregnancies had abnormal bCG and/or progesterone levels in early pregnancy. All of those pregnancies with abnormal endocrine parameters terminated with spontaneous abortion (60%). Certain abnormal bCG patterns were repeatedly observed in some animals and were correlated with repeated spontaneous abortions. Of 17 pregnancies with normal bCG and P patterns, 15 (88%) continued to term with a normal fetal outcome. In this study, a pregnancy rate per mated cycle of 47% was observed, yet 60% of untreated pregnancies abortyed spontaneously. Overall 16% of the mated cycles had continuing pregnancies with normal outcome. These studies demonstrate that a high rate of early abortions occurs in the baboon and that a single bCG determination is insufficient to define the presence of a “normal” pregnancy which might be expected to carry to term with a normal outcome.  相似文献

This study investigates oxidative stress and antioxidants in normal human pregnancy and post-partum period. Thirty-seven healthy women with normal pregnancies were included. Both urinary and serum samples were collected throughout the pregnancy and post-partum period. Oxidative stress was estimated by measuring the reliable in vivo marker, namely 8-iso-prostaglandin F2α (8-iso-PGF2α, an F2-isoprostane) and antioxidant status was evaluated by measuring α- and γ-tocopherol in serum samples. Pregnancy was associated with successively increased levels of 8-iso-PGF2α with advancing gestational age. The median post-partum value corresponded to the values observed in early gestation and a significant decrease was observed from late pregnancy to the post-partum period. Lipid-adjusted α- and γ-tocopherol levels decreased with advancing gestational age. This longitudinal study suggests that mild oxidative stress is involved in normal human pregnancy.  相似文献

For better operational control of the completely mixed oxygen activated sludge process (CMOAS), a study concerning the kinetics, performance, and operational stability of the Ramanathan-Gaudy model was conducted. Short-term experiments were conducted at various dilution rates (1/9, 1/6, 1/3, 1/1.5, and 1/1.0 hr?1) by using two recycle solids concentration values (5000 and 10,000 mg/liter). The influent substrate was an actual industrial organic wastewater (soft drink waste) and its concentration was maintained at 1000 mg/liter COD. The hydraulic recycle ratio, α, was maintained at 0.30. It was found that for CMOAS system with constant recycle cell concentration, a “steady state” with respect to reactor biological solids and effluent COD at different dilution rates could be attained. No appreciable dilute-out of reactor biological solids and substrate was observed up to the dilution rate of 1 hr?1 for both systems of different XR (5000 and 10,000 mg/liter). For the system of XR = 5000 mg/liter, except the dilution rate of hr?1, the effluent filtrate COD was lower than 100 mg/liter, the aerator biological solids concentration was about 1550 mg/liter, and the COD removal efficiency was higher than 90% for all dilution rates. For the system of XR = 10,000 mg/liter, the effluent filtrate COD was lower than 71 mg/liter, the aerator biological solids concentration was about 2750 mg/liter, and the COD removal efficiency was higher than 90% throughout all the dilution rates selection in the present study. The value of the Sludge Volume Index (SVI) was the range of 37.0 to 58.5 and provided good settleability of sludge. The sludge yield was 0.53 for the system of XR = 5000 mg/liter and 0.57 for the system of XR = 10,000 mg/liter. The carbohydrate and the protein content of the cells were 10.1–21.6% and 35.6–50.6%, respectively. For predicting the reactor biological solid and effluent COD of the CMOAS system by using the Ramanathan-Gaudy model, two sets of values for the biological kinetic constants should be considered since it provided the best fit of predicted values of the observed values. In the present study, μm = 0.4 hr?1, ks = 92 mg/liter for 1/3 ? D ? 1, and μm = 0.05 hr?1, ks = 11.1 mg/liter for 1/9 ? D < 1/3 were used to calculate the predicted values of reactor biological solid and effluent filtrate COD.  相似文献

An F1-ATPase-defective mutant, TBLA-1, was constructed by the transduction of a defective gene for the a subunit of F1-ATPase, atpA401, into Escherichia coli W1485lip2, a lipoic acid-requiring pyruvic acid producer. The pyruvic acid production of the strain TBLA-1 was found to be improved markedly compared with that of strain W1485lip2. In cultures using a jar fermentor, the strain W1485lip2 consumed 50 g/liter of glucose and produced 25 g/liter of pyruvic acid after culture for 32 h, while strain TBLA-1 consumed the same amount of glucose, and produced more than 30 g/liter of pyruvic acid in a 24-h culture. A revertant, No. 63–1, derived from the strain TBLA-1, had a normal level of F1-ATPase activity, and showed a similar pattern of pyruvic acid production to that of strain W1485lip2.  相似文献

Summary We report in this paper the effect of metabolic depletion on several modes of furosemide-sensitive (FS) Na and K transport in human red blood cells. The reduction of ATP content below 100 mol/liter cells produced a marked decrease in the maximal activation (V max) of the outward. FS transport of Na and K into choline medium in the presence of ouabain (0.1 mM) and 1 mM MgCl2. TheK 0.5 for internal Na to activate the FS Na efflux was not altered by metabolic depletion. However, metabolic depletion markedly decreased the K i for external K (K o ) to inhibit the FS Na efflux into choline medium (from 25 to 11 mM). Repletion of ATP content by incubation of cells in a substraterich medium recovered control levels ofV max of the FS Na and K fluxes and of K i for external K to inhibit FS Na efflux. TheV max of FS Na and K influxes was also markedly decreased when the ATP content dropped below 100 mol/liter cells. This was mainly due to a decrease in the inward-coupled transport of K and Na (Na o -stimulated K influx and the K o -stimulated Na influx). The FS K i /K o exchange pathway of the Na–K cotransport, estimated from the FS K influx from choline-20 mM K o medium into cells containing 22 mmol Na/liter cells, was also reduced by starvation. Starvation did not inhibit the FS Na i /Na o exchange pathway, estimated as FS Na influx from a medium containing 130 mM NaCl into cells containing 22 mmol Na/liter cells. The unidirectional FS22Na efflux and influx were also measured in control and starved cells containing 22 mmol Na/liter cells, incubated in a Na medium (130 mM) at varying external K (0 to 20 mM). In substrate-fed cells, incubated in the absence of external K, FS Na efflux was larger than Na influx. This FS net Na extrusion (400 to 500 mol/liter cells·hr) decreased when external K was increased, approaching zero around 15 mM K o . In starved cells the net Na extrusion was markedly decreased and it approached zero at lower K o than in substrate-fed cells. Our results indicate that the FS Na and K fluxes, and their major component, the gradient driven Na–K–Cl cotransport system, are dependent on the metabolic integrity of the cells.  相似文献

During human pregnancy the placenta produces a variety of proteins like steroid hormones and their receptors that are responsible for the establishment and ongoing of the feto-placental unit. Inhibins are dimeric glycoproteins, composed of an α-subunit and one of two possible β-subunits (β A or β B). Aims of the present study were the determination of the frequency and tissue distribution patterns of the inhibin/activin subunits in human placental tissue of normal pregnancies and pregnancies complicated with fetal growth restriction (IUGR). Slides of paraffin embedded placental tissue were obtained after delivery from patients diagnosed with IUGR (n = 6) and normal term placentas (n = 8). Tissue samples were fixed and incubated with monoclonal antibodies inhibin/activin-subunits -α, -β A, -β B. Intensity of immunohistochemical reaction on the slides was analysed using a semi-quantitative score and statistical analysis was performed (P<0.05). A significant lower expression of the inhibin-α subunit in IUGR extravillous trophoblast compared to normal pregnancies was observed, while the inhibin-α immunostaining was significantly upregulated in syncytiotrophoblast. Additionally, a significant down-regulation of inhibin-β B subunit in extravillous trophoblast cells in IUGR syncytiotrophoblast cells was demonstrated. A co-localisation of inhibin-α and the β-subunits was also observed, suggesting a production and secretion of intact inhibin A and inhibin B. Although the precise role of these inhibin/activin subunits in human placenta and IUGR pregnancies is still unclear, they could be involved in autocrine/paracrine signalling, contributing to several aspects like angiogenesis and tissue remodelling.  相似文献

Summary In order to investigate the question whether ammonium reabsorption in the thick ascending limb of Henle's loop (TALH) proceeds via the Na+,K+,Cl-cotransporter, plasma membrane vesicles were prepared from TALH cells isolated from rabbit kidney outer medulla and the effect of NH 4 + on their transport properties was investigated. It was found that, in the presence of a 78-mmol/liter NaCl gradient, 5 mmol/liter NH 4 + inhibited bumetanide-sensitive rubidium flux by 86%; a similar decrease was observed for 5 mmol/liter, K+. Inhibition of bumetanide-sensitive rubidium uptake by NH 4 + was competitive and an apparentK i of 1.9 mmol/liter was found Bumetanide-sensitive sodium uptake measured in the presence of a 83 mmol/liter KCl gradient was not inhibited by 5 mmol/liter NH 4 + . A 100-mmol/liter NH4Cl gradient was, however, capable of stimulating bumetanide-sensitive sodium uptake to the same extent as a KCl gradient. These data suggest that NH 4 + is accepted by the K+ site of the Na+,K+,Cl-cotransport system and that the transporter can function in a Na+, NH 4 + ,2Cl mode. Background

Preterm birth is a leading cause of perinatal mortality, yet the evolutionary history of this obstetrical syndrome is largely unknown in nonhuman primate species.

Methodology/Principal Findings

We examined the length of gestation during pregnancies that occurred in a captive chimpanzee colony by inspecting veterinary and behavioral records spanning a total of thirty years. Upon examination of these records we were able to confidently estimate gestation length for 93 of the 97 (96%) pregnancies recorded at the colony. In total, 78 singleton gestations resulted in live birth, and from these pregnancies we estimated the mean gestation length of normal chimpanzee pregnancies to be 228 days, a finding consistent with other published reports. We also calculated that the range of gestation in normal chimpanzee pregnancies is approximately forty days. Of the remaining fifteen pregnancies, only one of the offspring survived, suggesting viability for chimpanzees requires a gestation of approximately 200 days. These fifteen pregnancies constitute spontaneous abortions and preterm deliveries, for which the upper gestational age limit was defined as 2 SD from the mean length of gestation (208 days).

Conclusions/Significance

The present study documents that preterm birth occurred within our study population of captive chimpanzees. As in humans, pregnancy loss is not uncommon in chimpanzees, In addition, our findings indicate that both humans and chimpanzees show a similar range of normal variation in gestation length, suggesting this was the case at the time of their last common ancestor (LCA). Nevertheless, our data suggest that whereas chimpanzees'' normal gestation length is ∼20–30 days after reaching viability, humans'' normal gestation length is approximately 50 days beyond the estimated date of viability without medical intervention. Future research using a comparative evolutionary framework should help to clarify the extent to which mechanisms at work in normal and preterm parturition are shared in these species.  相似文献

At least two spontaneous abortions were karyotyped in 273 women during cytogenetic surveys in New York City and Honolulu. These pairs were analyzed using maximum-likelihood logistic-regression analysis to adjust for maternal age and location. There was a significantly increased risk for a chromosomally normal spontaneous abortion after a previous abortion with a normal karyotype. There was no increased risk for trisomy in a second spontaneous abortion following either a previous trisomic abortion or an abortion with another abnormal karyotype. This is unexpected, given the increased risk for trisomy found among live births and at prenatal diagnosis in young women with a previous trisomic birth. The most likely explanation is that the increased recurrence risk for trisomy is restricted to trisomy for only one or a few chromosomes, for reasons such as parental trisomy mosaicism. These data predict no increased risk of chromosome abnormality in future pregnancies after either (1) spontaneous abortions with trisomies of a kind that are always lethal in utero or (2) multiple early abortions in the presence of normal parental karyotypes.  相似文献
