Summary AnEscherichia coli strain constitutive for -galactosidase was immobilized onto cotton cloth. The resultingE.coli film was used as a resident inoculum in repeated batch fermentations for 30 days in the presence ofBrevibacterium ammoniagenes added as a contaminant. Analysis of -galactosidase production shows that contamination did not decrease the capacity of the film to generateE.coli cells, or decrease theE.coli population on the film.  相似文献

Summary Cultures of Streptomyces flavogriseus produced considerable amounts of xylanase when grown on xylan containing media. Comparatively lower yields of this enzyme were obtained when hay or avicel served as main carbon source, -xylosidase was synthesized intracellularly and appeared less dependent on the fermentation substrate. The strain produced simultaneously various enzymes of the cellulase complex and the xylose induced glucose isomerase.  相似文献

Summary The enzyme production and localization ofThielavia terrestris strains C464 and NRRL 8126 were compared to determine their optimum temperature and pH for cellulase activity. High levels of intracellular -glucosidase activity were detected in the former strain. The intracellular -glucosidase of both strains were more thermostable than the extracellular enzyme; the half life ofT.terrestris (C464) endoglucanase activity at 60°C was greater than 96 hrs.  相似文献

Summary Strain Bacillus subtilis MS was constructed with 12–22 fold increase of -amylase production, caused by presence of multiple -amylase gene copies in the chromosome of industrial strain Bacillus subtilis CCM2722, as demonstrated by DNA hybridization. The enhanced production is a result of multiple integration of plasmid pTVA1, carrying a temperature sensitive origin of replication from pE194, and containing the -amylase gene and a modified transposon Tn917.  相似文献

A comparison of the rate of carboxymethyl cellulase (CMCase), avicelase, xylanase, -glucosidase and -xylosidase production and rates of growth by 4 different strains ofCellulomonas revealed a wide range of behaviour; with some strains producing more CMCase, avicelase, xylanase, -glucosidase and -xylosidase from complex lignocellulosic (LC) biomass (from saline land) and CMC while some others producing small amounts of these enzymes. One strain,C. biazotea, was better with respect to enzyme production potential and growth behaviour than most of the other strains and has been chosen as a starting strain for genetic improvement for producing enzymes of the cellulase complex.  相似文献

Summary L-Sorbose, which is known as an inhibitor of -1,3-glucan synthesis in fungi, induces the production of cellulases in strains belonging to Trichoderma reesei. Especially, mutant strains PC-3–7 and X-31, which were obtained by several steps of mutation from QM 9414, have the most effective cellulase inducibility by L-sorbose comparing with other mutants of Trichoderma reesei. They synthesized cellulases effectively in liquid culture, whenever the alkaline treated sugarcane bagasse was used as a main carbon source for lowering the cost of cellulase production.  相似文献

Summary Cellulase production by Trichoderma reesei mutant RUT-C30, immobilized on 4% -carrageenan beads, was monitored in continuous culture for 13 days. Cellulase production averaged 26.0 Filter Paper Units (FPU)/l/hr; carbon and nitrogen requirements per FPU produced were reduced to 1/4-1/2 those of conventional continuous culture.  相似文献

Crown gall initiation on Mentha × piperita var. citrata (Ehrh.) Briq. (mint) was investigated using a range of wild type and mutant strains of Agrobacterium tumefaciens. Axenic transformed shoot cultures of Mentha citrata were established on plant stems inoculated with the nopaline strain T37 of Agrobacterium tumefaciens. The presence of T-DNA in the transformed tissues and the absence of bacterial contamination was established by Southern Blot hybridisation, using 32P labelled fragments of the T-DNA and virulence region of the Ti plasmid as probes. The shoot cultures synthesised a mint oil fraction which contained the major terpenes characteristic of the parent plant in quantities similar to those found in intact tissue. Summary The production of -carotene by the biomass ofRhodotorula strain var.glutinis, during the stationary phase of growth and in non-proliferating conditions was assayed. Summary Synergistic coculture of an amylolytic yeast (Saccharomycopsis fibuligera) andS. cerevisiae, a non-amylolytic yeast, fermented unhydrolyzed starch to ethanol with conversion efficiencies over 90% of the theoretical maximum. Fermentation was optimal between pH 5.0 to 6.0. Using a starch concentration of 10% (w/v) and a 5% (v/v) inoculum ofS. fibuligera, increasingS. cerevisiae inoculum from 4% to 12% (w/v) resulted in 35–40% (w/v) increase in ethanol yields. Anaerobic or limited aerobic incubation almost doubled ethanol yields.  相似文献

Summary An investigation of plasmid stability in aSaccharomyces cerevisiae double mutant has been performed. The host was a double recombinantura3 furl mutant containing a plasmid bearing the yeast URA3+ allele and an expression cassette for human 1-antitrypsin. The mutant was grown in continuous culture employing a semi-defined medium containing added uracil to provide non selective growth conditions. After 150 generations of continuous growth, no cured cells had been detected: the specific expression level of 1-antitrypsin remained constant throughout the experiment.  相似文献

Summary The production of hyaluronate lyase (E.C.4.2.2.1.), HL, byStreptococcus equisimilis strain SHL-03 has been studied under repeated batch culture conditions with fixed harvest volume (discharged fraction, =0.875). The productivity of this system was 49.2% higher than that of a classical batch culture. The process could be repeated at least for 15 cycles (245 hours) with a productivity of 33.77 U/ml h and a yield of 12,800-U/g Glu.These results suggest that this system could provide a useful procedure to increase the productivity of HL production.  相似文献

Summary The -mating factor pre-pro-leader sequence under the regulation of theGAL10 promoter was used to direct the secretion of echistatin by recombinant yeasts. Optimization of the culture medium and host strain increased the productivity of shake flask cultures twenty-fold to 8 mg/L. Summary The yeastsP. stipitis NRRL Y-7124 andP. tannophilus NRRL Y-2460 were entrapped in -carrageenan beads and used for repeated batch fermentation of D-xylose, in a series of four reactors. The operating conditions finally chosen gave an oxygen coefficient (KLa) of 0.83 min–1, as measured by the sulphite method. Ethanol yields were 0.40 g/g forP. stipitis and 0.36 g/g forP. tannophilus (respectively 78.4% and 70.5% of the theoretical yields). In spite of its lower retention by the gel,P. stipitis exhibited greater fermenting capacities thanP. tannophilus.  相似文献

Summary Starting from anileu auxotroph ofFusarium graminearum producing high levels of the mycotoxin zearalenone, selection after UV irradiation gave low-producing mutants of essentially normal morphology,zea,ileu. Heterokaryons betweenzea,ileu strains and an auxotrophic strainlz,inos derived from the lazy morphological mutant ofFusarium graminearum, which has abnormal morphology and also produces little or n zearalenone, produced significant levels (over 50% of the wild-type level) of mycotoxin. Summary Exopolysaccharide production byPseudomonas sp. strain EPS-5028 increased under conditions of high C/N ratios at the pH range 6.7–8 and 25°C. The polymer was formed from a variety of carbon substrates and contains D-glucose, D-galactose and uronic acid in the approximate molar ratio of 325. The polysaccharide produced is of high viscosity (1,600 cP; 1% polymer in D. I. water).  相似文献

Summary Polyene-resistant mutants ofPenicillium chrysogenum Wis. 54–1255 have been obtained by stepwise selection in increasing concentrations of polyene antibiotics. From the parent strain, sensitive to 10 g/ml of polyene antibiotics, mutants resistant to fungimycin (1.3 mg/ml), amphotericin B (0. 5 mg/ml), or nystatin (167 g//ml) were obtained. Their penicillin production is different from that of the parent strain and in particular some of the fungimycin-resistant mutants produce higher levels of penicillin.  相似文献

Summary Approximately 850 fungus and yeast strains were tested for their ability to hydroxylateN-acetyl-o-toluidine in the 4-position.
