A protein fraction containing prolactin activity from the pituitary tissue of a teleost fish, Tilapia mossambica, has been purified by a combination of ion-exchange and exclusion chromatographic procedures. The purified Tilapia prolactin was characterized by disc gel electrophoresis, amino-terminal group identification, and amino acid analysis. Its amino acid composition was found to be similar to ovine prolactin. The purified fish prolactin was found to be 40–50 times more potent than ovine prolactin in the Tilapia sodium-retaining bioassay. However, it was found to be devoid of Gillichthys yellow pigment-dispersing activity which was previously thought to be a property of teleost prolactin.  相似文献

The protozoan Tetrahymenapyriformis was found to be resistant to the toxic action of ricin invivo. Isolated Tetrahymena ribosomes were strongly resistant to the A subunit of ricin when tested in a cell free protein synthesis system under different conditions and also lacked the ability to bind A chain stoichometrically. This suggests that Tetrahymena is resistant invivo because it contains a ribosome which is not susceptible to the toxic action of ricin.  相似文献

Temperature and light interact to modify the chemical and biochemical composition of a nitrogen-limited marine diatom, Thalassiosira allenii Takano, grown at a constant dilution rate in continuous culture and under a light:dark cycle.The percent of the total 14C incorporated into protein, polysaccharide and lipid, the N/C ratio and the C/cell varied with temperature in a markedly non-linear manner. The N/cell was negatively correlated to temperature. The Chl aC ratio was positively correlated with temperature under saturating light and non-saturating light for temperatures > 25 °C, but was constant under non-saturating light conditions for temperatures < 25 °C.Productivity index (PI) was negatively correlated to temperature under saturating light conditions, but did not vary under low light. In each case, the variation in PI with temperature was governed by the variation in Chl aC.The dark carbon loss rate was exponentially related to temperature and independent of light. Variation in the percent of the total 14C incorporated into protein and polysaccharide, the NC ratio and C/cell was primarily due to the effects of N-limitation < 20 °C and primarily due to the effects of temperature > 20 °C. Variation in N/cell was primarily due to the effects of temperature over the entire range of temperature studied. Variation in Chl aC was caused by the interaction of temperature and light effects.In most cases, temperature and nutrient effects interacted to govern how a particular parameter varied with temperature while light affected the range of values over which the parameter varied.The percent of the total 14C incorporated into protein exhibited a significant linear relationship with NC.The dark carbon loss rate, NC ratio and Chl aC ratio data were used to test the applicability of a model for the physiological adaptation of unicellular algae. The model, with parameters derived from a non-linear least-squares fit of the dark carbon loss rate data, adequately described the NC ratio between 15 and 25 °C at 290 and 137 μE · m?2 · s?1, but failed to describe the data at 28 °C and at 48 μE · m?2 · s?1. The Chl aC ratio was adequately described by the model under all light and temperature conditions.  相似文献

Scattering of green light and chlorophyll fluorescence by spinach leaves kept in a stream of air or nitrogen were compared with leaf adenylate levels during illumination with blue, red or far-red light. Energy charge and ATP-ADP ratios exhibited considerable variability in different leaves both in the dark and in the light. Variability is explained by different possible states of the reaction oxidizing triose phosphate or reducing 3-phosphoglycerate. Except when oxygen levels were low, there was an inverse relationship between light scattering and chlorophyll fluorescence during illumination with blue or red light. When CO2 was added to a stream of CO2-free air, chlorophyll fluorescence increased, sometimes after a transient decrease, and both light scattering and leaf ATPADP ratios decreased. Similar observations were made when air was replaced by nitrogen under blue or high-intensity red light. Under these conditions, over-reduction caused inhibition of electron transport and phosphorylation in chloroplasts. However, when air was replaced by nitrogen during illumination with low-intensity red light or far-red light, light scattering increased instead of decreasing. Under these light conditions, ATPADP ratios were maintained in the light. They decreased drastically only after darkening. Although ATPADP ratios responded faster than light scattering or the slow secondary decline of chlorophyll fluorescence due to illumination, it appeared that in the steady state, light scattering and chlorophyll fluorescence are useful indicators of the phosphorylation state of the leaf adenylate system at least under aerobic conditions, when chloroplast and extrachloroplast adenylate systems can effectively communicate.  相似文献

The amplification effect of 19-norandrost-4-ene-3, 17-dione (19-nor-A-dione) on aldosterone in normal and sodium-loaded conditions was evaluated using adrenalectomized rats fed a normal or high sodium diet. The administration of 19-nor-A-dione in normal or sodium-loaded conditions did not cause any significant change in urinary NaK ratio. The simultaneous administration of subthreshold doses of aldosterone and 19-nor-A-dione in normal conditions also did not cause any significant change in urinary NaK ratio. However, the simultaneous administration of subthreshold doses of aldosterone and 19-nor-A-dione in sodium-loaded conditions caused a significant decrease in urinary NaK ratio. The decrease in urinary NaK ratio was caused by a decrease in urinary Na excretion. These results demonstrate that 19-nor-A-dione, which did not amplify the action of aldosterone in normal conditions, amplified the action of subthreshold doses of aldosterone in sodium-loaded conditions. 19-Nor-A-dione is considered to be an amplifier of aldosterone which works only in sodium-loaded conditions.  相似文献

Isolated rat liver cells were exposed invitro to ageing in an aerobic condition and to the effects of anoxia, thermal and p-chloromercuribenzene sulfonic (PCMBS) acid treatment, The survival of the cells was studied at normal and acidic pH using vital dye staining, intracellular concentration of potassium and ATP as indicators of viability. In aerobic conditions at pH 7.4, control cells lost their viability within approximately 6 hours. Extracellular acidosis not only prolonged the life span of isolated control hepatocytes against mechanical and other possible stress factors associated with the incubation and the absence of substrates in the medium, but also protected the cells significantly against various other superimposed injurious effects. These observations augment our previous observations on Ehrlich ascites tumore cells and lend further support to the hypothesis that extracellular acidosis, a common phenomenon associated with cell injury, is not harmful to cell survival invitro evidently prolongs it. The mechanism(s) behind this effect is not fully understood but it is suggested that extracellular acidosis stabilizes, and thereby protects cellular membrane systems making them more resistant to various deteriorating effects.  相似文献

We have investigated the action of high doses of androgens in Gobius niger L., a marine teleostean fish, by characterizing specific steroid receptors in liver and by assaying the plasma vitellogenin concentration under different hormonal treatments. Estrogen and androgen receptors were characterized in the liver nuclear extracts according to their binding specificity. The maximum binding capacity was 25 fmoles/mg protein for the estrogen and androgen receptors. In vivo, high doses of DHT(·) increased the concentration of plasmatic vitellogenin as assayed by immunodiffusion while low doses were inefficient. In spite of a similar number of estrogen and androgen nuclear receptor sites (25 fmoles/mg protein), DHT was at least 70 fold less active than et on yolk protein and vitellogenin induction both in male and female Gobius niger. In addition, the antiestrogen tamoxifen, which was inactive by itself, inhibited the e2 and the DHT induced accumulation of vitellogenin. Progesterone (2 mg/fish) was also totally inactive in inducing vitellogenin. We conclude that the induction of vitellogenin by DHT is mediated by the estrogen receptor rather than by the androgen receptor.In addition to the estradiol induced protein in rat uterus and to other estrogenic responses obtained by androgens in mammary cancer, fish vitellogenin is another estrogen regulated protein which can be induced by high doses of androgens. Hepatic microsomal membranes, prepared under various conditions that yield either ‘intact’ or ‘disrupted’ microsomal vesicles, have been labeled via the sulfhydryl groups of intrinsic membrane proteins using nitroxide analogs of N-ethylmaleimide. Electron paramagnetic resonance spectra revealed the presence of two dominant classes of bound label corresponding to differing degrees of immobilization, the ratio of which were quantitated using a parameter designated the ‘W/S’ ratio. For latent microsomes, the value of this parameter was determined to be 0.65 ± 0.02 and was influenced by factors such as label/protein ratio, incubation period, nitroxide structure, temperature and pH. The W/S ratio was also sensitive to the degree of membrane integrity as revealed by the latency of mannose 6-phosphate activity of glucose-6-phosphohydrolase. In addition, membrane disruption resulted in a corresponding decrease in the order parameter for nitroxide-labeled fatty acids intercalated within the lipid bilayer. The W/S ratio was observed to be dependent upon the method of microsome preparation yielding values of 1.02 ± 0.02 for ‘hypertonically disrupted’ vesicles and 1.28 ± 0.02 for ‘mechanically disrupted’ vesicles. Microsomal marker enzymes such as cytochrome P-450 and FAD-containing monooxygenase retained significant levels of functionally following nitroxide incorporation.  相似文献

A possibility of a heme-heme interaction between the heme c and heme d moieties in Pseudomonas nitrite reductase was examined by using magnetic and natural circular dichroism. The MCD of the heme c moiety in the ferric enzyme was similar to that of mammalian ferricytochrome c in shape and intensity, whereas in the reduced state the MCD intensity was considerably smaller than that of ferrocytochrome c. When the heme d moiety was perturbed by the complex formation with CO, imidazole or cyanide as well as by pH changes, the depressed MCD was restored to the MCD level of mammalian ferrocytochrome c, accompanying conformational changes around the prosthetic groups. Thus, it was concluded that the heme-heme interaction exists only in the reduced enzyme and that this interaction is released under appropriate conditions.  相似文献

Upon illumination of suspensions of intact chloroplasts, fluorescence of 9-aminoacridine was quenched, light scattering was increased, chlorophyll fluorescence was decreased after an initial increase, and chloroplast ATPADP ratios were increased. The response of 9-aminoacridine fluorescence quenching and light scattering to light intensity, anaerobiosis and inhibition of electron transport by DCMU was similar to that shown by chloroplast ATPADP ratios. It is discussed under what conditions 9-aminoacridine fluorescence quenching or light scattering can be used to monitor changes in the phosphorylation state of the chloroplast adenylate system.  相似文献

Plasma membranes from heart (sarcolemma) were prepared by the method of Kidwai, A.M. ((1975) Methods in Enzymology (Fleischer, S. and Packer, L., eds.), Vol. XXXIA, pp. 134–144, Academic Press, New York). On many occasions the sarcolemmal fraction identified by the enzyme markers such as (Na+ + K+)-ATPase banded at heavier densities (3 > 1.25 g/ml) than expected for plasma membrane (d < 1.15 g/ml). Radio-iodination of the membrane was added as an independent marker and conditions for the reproducible preparation of the sarcolemma were studied. Cultured heart cells were enzymatically iodinated under conditions which did not affect viability and labeled primarily the sarcolemma. The distribution of radioactivity in homogenates of cultured cells on the density gradient corresponded to that of the enzymes' activity. The best sarcolemma preparation was obtained with 0.3 M KCl extraction of heart homogenates in the presence of 0.05 M pyrophosphate, especially if the salt was also present during the fractionation by density gradient centrifugation. Alterations in the density were also observed with erythrocytes and cultured liver cells' plasma membrane. The data suggests a meta-stable state of the plasma membranes due to handling or storage which could cause alterations of some of their physical properties (e.g. density).  相似文献

Thermotropic properties of purified cytochrome c1 and cytochrome c have been studied by differential scanning calorimetry under various conditions. Both cytochromes exhibit a single endothermodenaturation peak in the differential scanning calorimetric thermogram. Thermodenaturation temperatures are ionic strength, pH, and redox state dependent. The ferrocytochromes are more stable toward thermodenaturation than the ferricytochromes. The enthalpy changes of thermodenaturation of ferro- and ferricytochrome c1 are markedly dependent on the ionic strength of the solution. The effect of the ionic strength of solution on the enthalpy change of thermodenaturation of cytochrome c is rather insignificant. The formation of a complex between cytochromes c and c1 at lower ionic strength causes a significant destabilization of the former and a slight stabilization of the latter. The destabilization of cytochrome c upon mixing with cytochrome c1 was also observed at high ionic strength, under which conditions no stable complex was detected by physical separation. This suggests formation of a transient complex between these two cytochromes. When cytochrome c was complexed with phospholipids, no change in the thermodenaturation temperature was observed, but a great increase in the enthalpy change of thermodenaturation resulted.  相似文献

The food conversion efficiency of Fundulus heteroclitus (L.) was measured in terms of calories and nitrogen for three diets: Palaemonetes sp., Nereis sp., and Uca pugnax. Fish were allowed to feed ad libitum for 30–60 min each day. Growth, ingestion, egestion, and excretion were measured for fish fed each diet. The growth rates of Fundulus heteroclitus fed two of the single item diets of invertebrates were higher than growth rates previously estimated for mummichogs from the natural population in Canary Creek marsh. In comparison with other fish species, F. heteroclitus was found to have a higher than average assimilation efficiency (X? = 87% for all three diets) and a lower than average gross growth efficiency (≈12% for two of the diets). Metabolic costs accounted for an average of 69% of ingested energy. Excretion rates were also large, with excretion accounting for more energy than egestion in two of the three diets.  相似文献

The factors influencing the overall mobility of the major proteins of the acetylcholine receptor-rich membranes from Torpedo marmorata have been investigated by saturation transfer ESR spectroscopy and the lateral distribution of these proteins has been studied by electron microscopy. A spin-labelled derivative of maleimide, 3-maleimido-2,2,5,5-tetramethyl-1-pyrrolidinyloxyl (MSL), was used under various conditions of incubation, enabling us to attach it mainly to either an extrinsic protein of 43 kdaltons, or an intrinsic protein (40 kdaltons) bearing the α-toxin-binding site. (1) The direct reaction of MSL with the membrane fragments resulted in almost exclusive labelling of the 43 kdalton protein, an extrinsic protein located on the inner face of the receptor-rich membranes. (2) After the free SH groups were blocked with N-ethylmaleimide and the disulfide bridges opened with the reducing agent dithiothreitol, MSL reacted with both the 40 and 43 kdalton proteins (6.0±0.6MSL molecules per α-toxin-binding site). (3) After the latter labelling procedure membranes were exposed to pH 11, resulting in extraction of the 43 kdalton protein and leaving 2.2 ± 0.4MSL molecules per α-toxin-binding site; sodium dodecyl sulfate polyacrylamide gel electrophoresis performed with N-[14C]ethylmaleimide suggested that MSL was bound mainly to the 40 kdalton polypeptide chain of the acetylcholine receptor. The following conclusions were made with the native and alkaline-treated membranes: In the native membranes, saturation transfer ESR does not reveal any significant protein rotational diffusion (itrotational correlation time τc > 1 ms). Temperature variations and/or lipid modifications obtained by fusion of exogenous lipids and/or cholesterol exchange have little influence on the saturation transfer ESR spectra. Electron microscopy reveals that upon lipid addition, proteins remain in the form of clusters while areas depleted of proteins appear. On the other hand, alkaline treatment strikingly enhances the motion of the MSL-labelled proteins in the membrane (100 ? τc ? 120 μs). Furthermore, the rotational diffusion of the MSL-labelled proteins (mainly the 40 kdalton protein) becomes sensitive to temperature, lipid composition and the lipid-to-protein ratio. Electron microscopy shows that alkaline extraction does not cause large reorganization of the acetylcholine receptor in the plane of the membrane. However, when phospholipids are added to pH 11 treated membranes, a dispersion of the receptor rosettes is observed. In contrast, cholesterol enrichment of the latter membranes induces clustering of the receptor and immobilization as judged by saturation transfer ESR. Upon reassociation of the pH 11 soluble proteins with the alkaline-treated membranes, the restriction of the acetylcholine receptor rotational mobility is also restored (τc ? 1 ms).  相似文献

The binding of inhibitors to site I of rabbit muscle phsphorylase b has beenstudied kinetically and thermodynamically for caffeine, adenine and adenosine. The effect of ligands on the tertiary structure has been investigated by studying the protection against 5,5′-dithiobis (2-nitrobenzoic acid) (DTNB) titration of the slow-reacting sulphydryl groups of the enzyme. Calorimetric and cysteinyl protection data taken together suggest that these inhibitors bind to both sites N and I even under conditions of saturation by glucose. Calorimetric results show that inhibitor binding to sites I and N at 25°C is driven enthalpically, although both ΔH and ΔS of interaction are significant. We conclude that attractive dispersion forces ought to be the main ones responsible for inhibitor binding to site I. AMP-activated phosphorylase b is inhibited by both caffeine and adenine by cooperative and exclusive binding to the inactive T conformation. The binding of the substrate (phosphate) and AMP when adenine is present was found to be exlusive to the active R conformation, whereas non-exclusive binding of the activator was observed when caffeine was added.  相似文献

Hen globin 9S mRNA complexes efficiently with mouse sarcoma 18S rRNA, and to a lesser extent with 28S rRNA, but not with tRNA. The mRNA-18S rRNA complex is dissociated under conditions that lead to disruption of hydrogen bonds, and exhibits a biphasic thermal denaturation curve with Tms at ca. 39° and 58° in 0.15 M NaCl-0.03 M Tris-HCl, pH 7.5. Hen globin mRNA also interacts with 18S rRNAs from various other eukaryotes, and the melting profile and Tm of the complex formed with hen 18S rRNA is very similar to that of the complex formed with mouse sarcoma 18S rRNA.  相似文献

Messenger ribonucleic acid isolated from angler fish (Lophiusamericanus) islets of Langerhans was translated in the wheat germ cell-free protein synthesizing system containing different combinations of radioactive amino acids. Preproinsulin (~ 11,000 daltons) was identified amongst the translation products, by sodium dodecyl sulfate gel electrophoresis, and subjected to microsequencing techniques. The fish preproinsulin was found to possess an NH2-terminal signal peptide of 24 amino acids, with regions of homology to human, rat and chicken preproinsulin signal sequences.  相似文献
