以淡色库蚊Culex pipiens pallens为试材,初步进行了典型光活化杀虫剂α-三噻吩(α-T)的电子自旋共振(ESR)分析,并研究其对昆虫保护酶系统的影响。ESR分析表明,近紫外光和药剂α-T均可促进活性氧自由基浓度升高,且两者具有协同关系,近紫外光能显著促进α-T增加自由基的作用,处理3 h时试虫自由基总浓度增加了99.6%。近紫外光和药剂α-T对保护酶系统的影响随酶的种类而有所不同,但在近紫外光存在的条件下,α-T对超氧物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活体活性均有抑制作用,且光照3 h时,抑制作用最强。  相似文献

The unicellular blue-green alga Anacystis nidulans was repeatedlytreated with X-rays and radiophosphate (32P) during successivesubcultures. The strains so obtained were characterized by comparisonwith the untreated control strain, with respect to their resistanceto ultraviolet light, X-rays, streptomycin, and isoniazid. The32P-treated strain was found to be relatively more resistantto streptomycin than the untreated strain and the X-rayed strainwas found to be relatively rnore sensitive to isoniazid thanthe control. In old cultures, cells of the X-rayed strain weresignificantly smaller than those of the untreated strain. The effects of X-rays on cell- and heterocyst dimensions ofthe nitrogen-fixing alga Chlorogloea fritschii Mitra were studied.In the irradiated material the cell diameter and heterocystbreadth were greater, rather than smaller, than in unirradiatedmaterial.  相似文献

Effect of UV-B Radiation on Leaf Optical Properties Measured with Fibre Optics   总被引:12,自引:0,他引:12  
Changes in the internal light microenvironment in leaves ofplants of Brassica campestris L. cv. Emma, B. carinata L., andMedicago saliva L. cv. Armour in response to exposure to UV-B(UV-B, 280–320 nm) radiation were measured using a fibreopticmicroprobe. Plants were exposed for 2 weeks either to high visiblelight or to supplemental ultraviolet-B radiation. The spectral regime (400–700 nm; PAR) was measured eithermidway through the leaf palisade or the spongy mesophyll. Afterexposure to UV-B radiation leaves of Brassica campesiris attenuatedtransmitted light more than the controls. At the same time bothforward and back scattered light increased in the palisade andspongy mesophylls. In contrast, UV-treatment of Medicago salivaleaves increased light transmission into the palisade, whilethe back scattered component showed little change. Leaves ofcariiwla showed little change in response to UV. Other responsesto UV-B radiation included increases in leaf thickness, decreasedtotal chlorophyll content, and changes in UV-B screening pigmentsand chlorophyll fluorescence induction kinetics. Brassica campestriswas most sensitive to exposure to enhanced levels of UV-B radiation,whereas leaves of B. carinata were the least sensitive. Ourdata indicate that exposure to UV-B radiation altered the lightmicroenvironment within leaves of the species different ways.These changes appeared to be caused by alterations in pigmentcontent and leaf anatomy. In turn, the altered distributionof PAR within the leaf could influence photosynthesis. Solar ultraviolet (UV) radiation poses a threat to most livingorganisms. Aquatic organisms have evolved three basic mechanismsto cope with harmful levels of radiation. Two mechanisms, avoidance(e.g. vertical migration) and photoprotection (e.g. productionof photoprotective compounds that act as filters, antioxidants,etc.), serve to minimize the dose of UV radiation that reachesthe organism's vital structures (DNA, membranes, etc.). Thethird mechanism, repair (e.g. dark repair mechanisms, such asnucleotide excision repair; or photoreactivation mechanisms,such as photoenzymatic repair), serves to repair the damagefollowing UV exposure. Here, we compare the vulnerability toUV-B radiation of three copepod species (Boeckella brevicaudata,Boeckella gibbosa, and Boeckella gracilipes) that occur in lakesthat differ in UV-B penetration and depth. Our aim was to gaininsight into the significance of each of the three mechanismsin different UV-B environments. Results from a 3-day ‘insitu’ incubation in ultra-oligotrophic Lake Toncek showedthat B.gracilipes is highly vulnerable to UV-B and UV-A radiation.In contrast, virtually no mortality was observed in B.gibbosaand B.brevicaudata during the same period. In order to discriminatethe contribution of photoprotection and photoreactivation, thethree species were subsequently exposed in the laboratory toan artificial source of UV-B radiation, both in the presenceand absence of visible radiation (recovery radiation). The photoprotectionpotential (i.e. resistance to UV-B in the absence of recoveryradiation) of B.gracilipes and B.gibbosa was lower than thatof B.brevicaudata. On the other hand, photoreactivation (higherresistance to UV-B in the presence of recovery radiation) wasobserved in B.brevicaudata and B.gibbosa, but not in B.gracilipes.To cope with damaging UV-B levels in nature, B.gracilipes dependsexclusively on the attenuation by the external media (i.e. avoidance).Although B.gibbosa tends to avoid the surface waters of lakes,it also occurs in shallow transparent pools. Most likely itsability to survive in these shallow, high UV environments isdue to its photoreactivation potential. Finally, despite itsoccurrence in highly turbid lakes, B.brevicaudata seems extremelywell suited to cope with UV-B radiation thanks to a combinationof photoreactivation and photoprotection.  相似文献

Vastus lateralismuscle biopsies were obtained from endurance-trained (running ~50km/wk) and untrained (no regular physical exercise) men, and theexpression of an array of insulin-signaling intermediates wasdetermined. Expression of insulin receptor and insulin receptorsubstrate-1 and -2 was decreased 44% (P < 0.05), 57%(P < 0.001), and 77% (P < 0.001),respectively, in trained vs. untrained muscle. The downstream signalingtarget, Akt kinase, was not altered in trained subjects. Components ofthe mitogenic signaling cascade were also assessed. Extracellularsignal-regulated kinase 1/2 mitogen-activated protein kinase expressionwas 190% greater (P < 0.05), whereas p38 mitogen-activatedprotein kinase expression was 32% lower (P < 0.05), intrained vs. untrained muscle. GLUT-4 protein expression was twofoldhigher (P < 0.05), and the GLUT-4 vesicle-associatedprotein, the insulin-regulated aminopeptidase, was increased 4.7-fold(P < 0.05) in trained muscle. In conclusion, the expressionof proteins involved in signal transduction is altered in skeletalmuscle from well-trained athletes. Downregulation of early componentsof the insulin-signaling cascade may occur in response to increasedinsulin sensitivity associated with endurance training.

The responses of two ataxia-telangiectasia (A-T) cell strains to the lethal effects of monochromatic far, mid and near ultraviolet radiations have been determined and compared with the responses of three normal human cell strains. Our results confirm a previous observation that the A-T cell strain AT4BI is abnormally sensitive to the lethal effects of mid u.v. (313 nm) radiation. After far u.v. (254 nm) radiation the strain AT4BI exhibits a small but statistically significant increase in sensitivity compared to the normal strains. Of most interest, in terms of a mechanistic interpretation of the sensitivity of A-T strains, the survival responses of neither A-T strain tested to near u.v. (365 nm) radiation differed significantly from the mean response of the normal strains, although it is of interest that one normal strain (48BR) was found to be significantly more resistant to near u.v. radiation than any of the other strains tested. The results are discussed in terms of the possible induction of radiogenic lesions in DNA by ultraviolet radiations and the possible mechanisms of radiation sensitivity in ataxia-telangiectasia.  相似文献

Summary A mutant strain of the blue-green alga Anacystis nidulans showing a 1000-fold increased resistance to streptomycin over the parental strain was isolated following treatment with diethyl sulphate. The mutant strain was found to show profuse filamentation, partial requirement of streptomycin for pigment production, and greater resistance to ultraviolet radiation. It also showed greater sensitivity to dimethyl sulphate than the parental strain. It seems that the streptomycin-resistant strain is a composite one comprising at least four kinds of mutations, namely streptomycin-resistance, filamentation, u.v.-resistance and partial dependence on streptomycin for pigment accumulation. Further. the streptomycin-resistant mutant seems nuclear rather than extra-nuclear in nature.Treatment with diethyl sulphate also conferred a slight increase in resistance to penicillin in this alga.  相似文献

Mutants of bacteriophage T4 which exhibit increased sensitivity to ultraviolet radiation specifically at high temperature were isolated after mutagenesis with hydroxylamine. At 42 °C the mutants are twice as sensitive to ultraviolet light as T4D, whereas at 30 °C they exhibit survival curves almost identical to that of the wild-type strain. Complementation tests revealed that the mutants possess temperature-sensitive mutations in the v gene.Evidence is presented to show that T4 endonuclease V produced by the mutants is more thermolabile than the enzyme of the wild-type. (1) Extracts of cells infected with the mutants were capable of excising pyrimidine dimers from ultraviolet irradiated T4 DNA at 30 °C, but no selective release of dimers was induced at 42 °C. (2) Endonuclease V produced by the mutant was inactivated more rapidly than was the enzyme from T4D-infected cells when the purified enzymes were incubated in a buffer at 42 °C. From these results it is evident that the v gene is the structural gene for T4 endonuclease V, which plays an essential role in the excision-repair of ultraviolet light-damaged DNA.The time of action of the repair endonuclease was determined by using the mutant. Survival of a temperature-sensitive v mutant, exposed to ultraviolet light, increased when infected cells were incubated at 30 °C for at least ten minutes and then transferred to 42 °C. It appears that repair of DNA proceeds during an early stage of phage development.  相似文献

The effect of ultraviolet (UV-B) radiation on the accumulation and tissue localization of phenolic compounds in two strains of callus cultures of tea plant (Camellia sinensisL.) were investigated. The strains differed in their morphological and physiological characteristics and biosynthetic capacity. UV-B radiation hampered culture growth, decreased the size of callus-forming cells and promoted the accumulation of soluble and, to a lesser extent, polymeric forms of phenolic compounds, such as lignin. This accumulation was accompanied by an increase in the phenolic compound deposition in cell walls and intercellular space and by deposition of a lignin-like material on the surface of callus cultures. The strain characterized by an increased formation of phenolic compounds was more resistant to UV-B radiation as compared to that with lower phenolic productivity.  相似文献

以兰州吐鲁沟公园的二点叶螨Tetranychus urticae 为敏感品系,分别用氧乐果、甲氰菊酯、四螨嗪喷雾处理15次,其抗性水平分别为38.5、479.8和67.3倍。将抗性品系分别与敏感品系进行杂交和回交的结果表明:抗氧乐果品系的显性系数DRS(R♀×S)为0.4700、DSR(S♀×R)为0.4749;抗甲氰菊酯品系的DRS(R♀×S)为0.5155, DSR为0.5237;抗四螨嗪品系的DRS为0.3134, DSR为0.2466。表明二点叶螨对这3种药剂的抗性均是由单个不完全显性基因所控制。在连续10个月不接触药剂的情况下,3个抗性品系的抗药性都有下降,抗氧乐果品系的抗性下降最快,只有敏感品系的抗性倍数的3.6倍;抗甲氰菊酯种群的抗性下降较慢,为敏感品系的95.9倍。再经甲氰菊酯、氧乐果、四螨嗪分别连续15次喷雾处理后,3个抗性种群的抗性水平又再度回升,抗甲氰菊酯品系回升较快,抗性为敏感品系的523.5倍,抗四螨嗪品系次之,抗氧乐果的品系抗性恢复最慢。  相似文献

A mutant of micrococcus radiodurans which is deficient in recombination has been isolated after treatment of the wild type with N-methyl-N'-nitro-N-nitrosoguanidine. We have called this mutant Micrococcus radiodurans rec30. The efficiency of recombination in this mutant, as measured by transformation, is less than 0.01% that of the wild type. It is 15 times more sensitive to the lethal action of ultraviolet radiation, 120 times more sensitive to ionizing radiation, and 300 times more sensitive to mitomycin C (MMC) than the wild type. It is probably inactivated by a single MMC-induced deoxyribonucleic acid cross-link per genome. The excision of ultraviolet-induced pyrimidine dimers is normal. There is no radiation-induced degradation of deoxyribonucleic acid. All spontaneous revertants selected for resistance to low levels of MMC had wild-type resistance to radiation and MMC, and the same efficiency of recombination as the wild type, suggesting that the recombination deficiency of the strain is due to a single mutation. Deoxyribonucleic acid from this mutant can transform M. radiodurans UV17 presumed deficient in an exr type gene to wild type.  相似文献

AIMS: To isolate a bacterium that produces high yield of melanin and to examine the effect of this bacterial pigment on the efficacy of a bioinsecticide. METHODS AND RESULTS: A novel melanin-producing bacterium, designated as strain WS, was isolated from the East Lake, Wuhan, China. Taxonomic studies of this strain indicate that it belongs to Aeromonas media. Physicochemical analysis of the pigment produced by strain WS (melanin WS) suggests that it is the authentic 3,4-dihydroxyphenylalanine (DOPA)-melanin. This melanin and that produced by Pseudomonas maltophilia P28 (melanin P28) share many biophysical properties, but the yield of the melanin WS is significantly higher than that of the melanin P28. In addition, the melanin WS appears to be more effective in the protection of a bioinsecticide against ultraviolet (UV) or solar radiation. At the concentration of 10 ppm, the melanin P28 exhibited no photoprotective effect on the bioinsecticide against UV radiation; in contrast, 5 ppm of melanin WS displayed an obvious protective effect. Similarly, the melanin WS displayed more protective effect on the bioinsecticide against solar radiation than the melanin P28 did over a 4-day period, with the effect being more dramatic for the last 2 days. CONCLUSIONS: We have isolated a novel bacterial strain of A. media that produces high levels of melanin. The melanin produced by this strain offers effective photoprotection of a commercial bioinsecticide BTI against UV and solar radiation. SIGNIFICANCE AND IMPACT OF THE STUDY: Our study suggests that the melanin produced by our newly isolated A. media strain has the potential to be used as a general photoprotective agent for bioinsecticides.  相似文献

A field experiment was carried out to analyse the growth oflettuce, onion and red beet in terms of: (a) canopy architecture,radiation interception and absorption; (b) efficiency of conversionof absorbed radiation into biomass; and (c) dry matter partitioning.Growth analysis, total solar radiation interception, PAR interceptionand absorption by the crop canopy, ground cover, maintenancerespiration of onion bulbs and red beet storage roots were measured.Models for different leaf angle distribution and ground coverwere used to simulate light transmission by the crop canopy. The three crops are shown to have contrasting growth patternsfrom both a morphological and a physiological point of view.Lettuce showed very high light interception and growth afterthe early growth stages but, throughout the growth cycle, thisleafy crop showed the lowest radiation use efficiency due tothe respirational cost of the high leaf area. Onion showed alower early relative growth rate than lettuce and red beet.This was due partly to the low light interception per unit leafarea in the later stages of growth and partly to the low initialradiation use efficiency compared with the other two crops.On the other hand, thanks to more uniform distribution of theradiation inside the canopy, to the earlier termination of leafdevelopment and to the very low level of bulb respiration, onionshowed high radiation use efficiency and was able to producea large amount of dry matter. Red beet leaf posture and canopystructure resulted in high light interception and absorption.Its radiation use efficiency was lower than that of onion, partlyperhaps because of the more adverse distribution of the interceptedradiation fluxes within the canopy and partly because of thehigh respiration cost of a continuous dry-matter allocationto the leaves. However, this crop can accumulate a very largeamount of dry matter as leaf blade development and storage rootgrowth can both continue almost indefinitely, providing continuouslyavailable sinks. Ground cover gave a good estimate of the PAR interception onlyat low values of light interception but, in general, it underestimatedPAR interception in all three crops. Ratios between attenuationcoefficients established by considering PAR or total solar radiationand LAI or ground cover were calculated. Radiation Interception, Partitioning and Use in Grass -Clover Mixtures   总被引:2,自引:0,他引:2  
Mixed swards of perennial ryegrass /white clover were grownin competition under controlled environmental conditions, attwo temperatures and with different inorganic nitrogen supplies.The swards were studied after canopy closure, from 800 to 1200°C d cumulative temperatures. Clover contents did not varysignificantly during the period. A simulation model of lightinterception was used to calculate light partitioning coefficientsand radiation use efficiencies for both components of the mixturein this controlled environment experiment. Additionally, thissame radiative transfer model was applied to the field datafrom Woledge (1988) (Annals of Applied Biology112: 175 –186)and from Woledge, Davidson and Dennis (1992) (Grass and ForageScience47: 230 –238). The measured and simulated valuesof light transmission, at different depths in the mixed canopy,were highly correlated (P<0.001) with more than 80% of thetotal variance explained. The daily average of photosyntheticallyactive radiation (PAR) interception in a natural environmentwas estimated from simulations, for the field and controlledenvironment data. Under these conditions, white clover capturedsignificantly more light per unit leaf area than perennial ryegrassat low, but not at high, nitrogen supply. In the controlled environment experiment, the radiation useefficiency of the legume was lower than that of its companiongrass. For both species, radiation use efficiency was negativelycorrelated with the mean irradiance of the leaf. The role ofa compensation between light interception and light use forstabilizing the botanical composition of dense grass –cloverswards is discussed. 室内筛选小菜蛾中抗品系(SRP)、高抗品系(RP)、以敏感小菜蛾幼虫(SP)为寄主的菜蛾绒茧蜂SRC品系和以SRP幼虫为寄主的菜蛾绒茧蜂RSC品系对氰戊菊酯的抗性。分别筛选了13、14、14和13代。小菜蛾SRP和RP品系分别获得了68.9和605.8倍的抗性,菜蛾绒茧蜂SRC和RSC品系分别获得了4.3和11.0倍的抗性。上述结果表明通过施药于体内有寄生蜂的小菜蛾幼虫筛选寄生蜂抗性,可以获得具有抗性的寄生蜂。以SRP为寄主的RSC品系的抗性水平高于以SP为寄主的SRC品系的抗性水平,表明和抗性较高的寄主同步筛选,寄生蜂的抗性发展更快。小菜蛾SP、SRP和RP三个品系幼虫的多功能氧化酶(MFO)活性比为1∶1.15∶1.50;菜蛾绒茧蜂SC、SRC和RSC三个品系幼虫的MFO活性比为1∶1.10∶1.49,成蜂的MFO活性比为1∶1.18∶1.54;而每种昆虫不同品系的羧酸酯酶(CarE)、总酯酶(Es)活性水平与其抗性水平变化不一致,表明抗性与MFO活性升高有关,而与CarE和Es的活性无关。  相似文献

Heat shock induces resistance to killing by ultraviolet light inEscherichia coli JE1011. When cells growing at 30°C were transferred to 42°C, maximum resistance to ultraviolet radition was reached after 30–45 min, but no change in heat resistance occurred. The effect was dependent on growth or protien synthesis. In contrast,E. coli B becomes more sensitive to the radiation and more heat resistant after a similar treatment. Thus, ultraviolet resistance and thermal resistance are not induced together in these two strains and may arise by independent mechanisms. It is also possible that thelon gene is involved in the effect of heat shock on ultraviolet resistance.  相似文献

Stress relaxation measurements were conducted on stage IVb Phycomycessporangiophores in order to correlate the effect of imposedstress on cell wall growth. It was found that the cell wallshowed maximum growth when subjected to maximum stress. Growthunder stress decreased as the stress decreased. This techniquewas used to measure the response of the sporangiophore to alight stimulus; the response is measured directly from the stressrelaxation curve. Stress/strain measurements were also conducted on the stageIVb Phycomyces sporangiophores in order to further characterizethe mechanical properties of the growing zone. It was foundthat the stress/strain ratio was invariant to the strain ratewithin the ranges tested but the stress/strain ratio did increasewith larger loads, i.e., the stress/strain ratio shows non-linearbehaviour.  相似文献
