Summary The authors review ten essential amino acids with regard to their metabolic, physiologic and therapeutic effects throughout the human body. Physical properties of these biologically active compounds are discussed as a foundation for their diverse roles in special nitrogen containing products, neurotransmitters, and as alternative energy sources. Both normal and abnormal amino acid metabolism are considered in the areas of digestion, elimination of metabolic products, metabolic intermediates, and defects in these systems. Recent developments in therapeutic applications are further examined for clinical utility and as an economical alternative to traditional clinical treatment modalities.  相似文献

L-asparaginase EC 3.5.1.1 was purified to homogeneity from Thermus thermophilus. The apparent molecular mass of L-asparaginase by SDS-PAGE was found to be 33 kDa, whereas by its mobility on Sephacryl S-300 superfine column was around 200 kDa, indicating that the enzyme at the native stage acts as hexamer. The purified enzyme showed a single band on acrylamide gel electrophoresis with pI = 6.0. The optimum pH was 9.2 and the Km for L-asparagine was 2.8 mM. It is a thermostable enzyme and it follows linear kinetics even at 77°C. Chemical modification experiments implied the existence of histidyl, arginyl and a carboxylic residues located at or near active site while serine and mainly cysteine seems to be necessary for active form.  相似文献

A flexible loop of amino acids (loop II) closes over the active site of hypoxanthine phosphoribosyltransferase (HPRT) as the enzyme approaches the transition state [Biochemistry 37 (1998) 17120]. Formerly, the deletion of much of loop II from the HPRT of Trypanosoma cruzi resulted in a 2-3 order of magnitude reduction in k(cat) values with relatively modest changes in the Michaelis constants for substrates [Biochim. Biophys. Acta 1537 (2001) 63-70]. However, the contributions of individual loop II residues to catalysis remained poorly understood or have been disputed. Herein, saturation mutagenesis was used to generate relatively random sets of mutations in the 12 residues of active site loop II in the HPRT from T. cruzi and steady-state kinetics was used to investigate reactions catalyzed by the mutants. The results of analyses of 18 different mutations in an evolutionarily invariant Ser-Tyr dipeptide are consistent with interactions, between main chain nitrogen atoms of these residues and the O1A atom of phosphoribosylpyrophosphate (PRPP) or pyrophosphate (PPi), being essential for efficient enzyme chemistry. The results of analyses of 55 mutations in the nine other amino acids in loop II are inconsistent with these residues participating directly in enzyme chemistry, but are consistent with several of their side chains influencing loop flexibility and folding, as well as the efficiency for nucleotide formation relative to pyrophosphorolysis.  相似文献

The rapid increase in the amount of protein sequence data has created a need for automated identification of sites that determine functional specificity among related subfamilies of proteins. A significant fraction of subfamily specific sites are only marginally conserved, which makes it extremely challenging to detect those amino acid changes that lead to functional diversification. To address this critical problem we developed a method named SPEER (specificity prediction using amino acids' properties, entropy and evolution rate) to distinguish specificity determining sites from others. SPEER encodes the conservation patterns of amino acid types using their physico-chemical properties and the heterogeneity of evolutionary changes between and within the subfamilies. To test the method, we compiled a test set containing 13 protein families with known specificity determining sites. Extensive benchmarking by comparing the performance of SPEER with other specificity site prediction algorithms has shown that it performs better in predicting several categories of subfamily specific sites.  相似文献

Whole body and egg essential amino acid (EAA) composition of silver pomfret, Pampus argenteus, was determined to estimate their dietary requirements using A/E ratios as an indicator. Ten adult fish (sex ratio 1 : 1) were sampled each month from Kuwaiti waters for a 1‐year period (November 2007 to October 2008). There was no significant difference (P > 0.05) between the whole body amino acid (AA) composition of males and females except for glutamic acid and serine. There were no significant differences (P > 0.05) between the contents of leucine, methionine, phenylalanine and threonine in whole body and eggs. However, arginine, isoleucine and valine contents in eggs were significantly higher than those of whole body, while lysine and histidine in whole body was significantly higher than that of eggs. Among non‐essential AAs, cystine, tyrosine, alanine, and serine content in eggs were significantly (P < 0.05) higher than those in whole body. A high correlation between the whole body A/E ratios of silver pomfret with the A/E ratios for P. argenteus (r2 = 0.79), P. punctatissimus (r2 = 0.90), sea bream (r2 = 0.81), sea bass (r2 = 0.69) and Japanese flounder (r2 = 0.95) was observed. Based on a lysine requirement value of 4.5 g per 100 g protein for Asian sea bass, the EAA requirement values of histidine, leucine, lysine and threonine for silver pomfret estimated using whole body EAA composition was slightly higher than those of eggs values. On the other hand, arginine, isoleucine, Met + Cys, Phe + Tyr, and valine values estimated using egg composition in silver pomfret were slightly higher than those estimated by whole body composition. However, a higher correlation was observed with the whole body EAA composition and estimated EAA requirement values (r2 = 0.81) compared to that with eggs (r2 = 0.72). Until dose‐response experiments are carried out to determine the EAA requirements precisely, the estimated EAA values using whole body EAA as proposed in this study could be used when formulating diets for silver pomfret.  相似文献

The significance of thermal polyamino acids (proteinoids) as abiotic predecessors of proteins is reviewed on the basis of new experimental results. Most proteinoids yield only 50% to 80% amino acid upon acid hydrolysis. They contain 40% to 60% less peptide links than typical proteins, whereas their average nitrogen content is like that of proteins. The arrangement of amino acid residues is nonrandom. The degree of nonrandomness is difficult to determine because unusual crosslinks disturb most of the sequencing methods typically applied, in protein chemistry. The products obtained in a polymerization experiment are heterogeneous. They can be separated into a limited number of related fractions by chromatography or electrophoresis and other separation methods applied in protein chemistry. Their molecular weights are typically between 400 and 10 000. The number of free NH2-groups, is usually smaller than in comparable proteins A significant fraction of NH2-groups yields imidazole-type bases during the thermal polymerization. Optically active amino acids racemize during the same process. So far no helicity could be detected. Proteinoids are thus clearly distinct from proteins However, many of them exhibit weak catalytic activities and tend to undergo self-assembly into microstructures. Their properties of which only a few have been mentioned still support their role as possible candidates for ancestors of first proteins.  相似文献

The symbiotic bacteria Buchnera contribute to the nutrition of pea aphids, Acyrthosiphon pisum, through the provision of essential amino acids which are lacking in the diet. However, chemically defined diets, containing nutritionally adequate amounts of essential amino acids, fail to rescue aposymbiotic aphids, in which the bacteria have been disrupted with antibiotics. In this study the injection of a mixture of essential amino acids into the haemocoel of aposymbiotic aphids was shown to alleviate, at least partially, the impact of symbiont loss. Specifically, the total amino acid content in the tissues of aposymbiotic aphids was reduced by approximately 40% to levels comparable with symbiotic insects, and there was a 1.7-fold increase in the number of embryos, suggesting that the availability of essential amino acids promotes aphid protein synthesis by rejuvenating the free amino acid pool of aposymbiotic aphids. In addition, a similar effect on the total amino acid content was observed when phenylalanine alone, but not glutamine, lysine or tryptophan, was injected into the haemocoel of aposymbiotic aphids, and there was also a significant increase in the number of embryos following injection of phenylalanine or tryptophan alone. The impact of amino acid injection on the embryo complement of aposymbiotic aphids was limited to an increase in the number of embryos, with no increase in basal embryo size. It is proposed that older embryos may rely on their own complement of symbiotic bacteria for essential amino acid provisioning. Taken together, the data highlight the importance of bacterial provisioning of essential amino acids, particularly the aromatic amino acids, in the intact symbiosis.  相似文献

Three species of the cosmopolitan genus Ulva (U. paschima , U. chaugulii and U. ohnoi ) from the Persian Gulf were identified using morphological and molecular markers, and were analyzed for total lipids, fatty acids, proteins and amino acid profiles. Our results show that the studied Ulva species have high protein content (9?25% dry weight), contain essential fatty acids and nearly all essential amino acids. This suggests Ulva could have potential as a form of human nutrition and animal feed with a low carbon footprint.  相似文献

Abstract

Lignocellulosic biomass (LB) is the renewable feedstock for the production of fuel/energy, feed/food, chemicals, and materials. LB could also be the versatile source of the functional oligosaccharides, which are non-digestible food ingredients having numerous applications in food, cosmetics, pharmaceutical industries, and others. The burgeoning functional food demand is expected to be more than US$440 billion in 2022. Because of higher stability at low pH and high temperature, oligosaccharides stimulate the growth of prebiotic bifidobacteria and lactic acid bacteria. Xylooligosaccharides (XOS) are major constituents of oligosaccharides consisting of 2–7 xylose monomeric units linked via β-(1,4)-linkages. XOS can be obtained from various agro-residues by thermochemical pretreatment, enzymatic or chemoenzymatic methods. While thermochemical methods are fast, reproducible, enzymatic methods are substrate specific, costly, and produce minimum side products. Enzymatic methods are preferred for the production of food grade and pharmaceutically important oligosaccharides. XOS are potent prebiotics having antioxidant properties and enhance the bio-adsorption of calcium and improving bowel functions, etc. LB can cater to the increasing demand of oligosaccharides because of their foreseeable amount and the advancements in technology to recover oligosaccharides. This paper summarizes the methods for oligosaccharides production from LB, classification, and benefits of oligosaccharides on human health.  相似文献

Protein quality mainly depends on the essential amino acid (EAA) profile, but also on its bioavailability, because EAA digestibility is generally lower than the analyzed amounts. This information is needed in the aquaculture industry for aquafeed formulation. For this purpose, the apparent digestibility coefficients of dry matter, protein, and essential amino acids of eight feedstuffs of terrestrial origin were determined for the juvenile whiteleg shrimp Litopenaeus vannamei (15-19 g), using 1% chromic oxide as an inert marker. A reference diet was formulated and produced in the laboratory. Eight experimental diets were prepared each with 30% of one of the experimental ingredients added to the reference diet: casein, porcine byproduct meal poultry byproduct meal, corn meal, wheat gluten meal, soybean paste, sorghum meal, and wheat meal. The experiment consisted of a single-factor, completely randomized design with three replicates per treatment. Samples of ingredients, diets and feces were analyzed for nitrogen and amino acids. For amino acid assay, we used reverse-phase high performance liquid chromatography. To avoid partial loss of methionine and cystine, samples of ingredients, diets, and feces were oxidized with performic acid to methionine sulfone and cysteic acid prior to acid hydrolysis. The apparent dry matter and protein digestive utilization coefficients varied from 68% to 109% and from 70% to 103%, respectively. Apparent digestibility of protein for casein, soy paste, wheat meal and wheat gluten were very high (over 90%), corn gluten and poultry byproducts meal showed high protein digestibility (over 80%), but porcine byproducts meal and sorghum meal had low digestibility (76% and 70%, respectively). There was a reasonable, but not total, correspondence between apparent protein digestibility and average essential amino acid digestibility coefficients, except for arginine in corn gluten, phenylalanine and leucine in sorghum meal, phenylalanine in soy paste and lysine in wheat meal and poultry by-product meal. The most digestible feed ingredients for whiteleg shrimp were: wheat gluten, wheat meal and soy paste; poultry byproduct meal and corn gluten were less digestible and the lowest digestibility occurred in porcine byproduct meal and sorghum meal. Feedstuffs exhibited great variability in dry matter, protein and amino acid digestive utilization coefficients, which should be considered when formulating shrimp feeds.  相似文献

Histidine, arginine and lysine residues are essential for the multifunctional 2,3-bisphosphoglycerate synthase-phosphatase purified from pig skeletal muscle. The synthase, phosphatase and phosphoglycerate mutase activities of the enzyme are concurrently lost upon treatment with diethylpyrocarbonate, phenylglyoxal and trinitrobenzenesulfonate. The phosphatase activity shows hyperbolic kinetics. In contrast, the synthase activity shows a nonhyperbolic pattern which fits to a second-degree polynomial. The Km values for glycerate 1,3-P2, glycerate 3-P and glycerate 2,3-P2 are similar to those of the enzyme from mammalian erythrocytes.  相似文献

黄荆精油对玉米象的杀虫活性成分、毒力及作用机制   总被引:1,自引:1,他引:1  
为明确黄荆精油及其主要成分作为储粮保护剂的开发利用价值,采用水蒸气蒸馏法从泰山黄荆Vitex negundo Linn.叶片中提取精油,利用GC-MS技术进行了成分分析鉴定,研究了黄荆精油及其主要成分桉树脑和α-蒎烯对玉米象 Sitophilus zeamais Motschulsky 成虫的毒力及作用机制。结果表明: 黄荆精油中含量大于0.3%的成分有31种,其中石竹烯、桉树脑和α-蒎烯的含量分别为35.97%,8.21%和0.69%。桉树脑、α-蒎烯和黄荆精油对玉米象成虫的综合杀虫毒力都较高,并以桉树脑的毒力最高,LC50为0.7171 g·kg-1;在2.0 g·kg-1剂量下,桉树脑、α-蒎烯和黄荆精油对玉米象种群的抑制率分别达100%,85.45%和89.73%;3种药剂对玉米象72 h触杀LC50分别为0.2690,0.7529 和0.2969 mg·cm-2,毒力都很高;3种药剂在4 g·kg-1剂量下,对玉米象的驱避率分别为96.49%,84.26%和90.61%,防虫作用大;3种药剂对玉米象72 h的熏蒸LC50分别为14.053, 28.648 和21.429 μL·L-1,熏蒸杀虫毒力较高,其中以桉树脑的毒力最高。3种药剂对玉米象有干扰呼吸的作用,对离体乙酰胆碱酯酶、过氧化氢酶和羧酸酯酶都有抑制作用。这些结果明黄荆精油及其主要成分的作用机制具有多样性。  相似文献

Sarcopenia, the age-related loss of muscle mass and strength, is a fundamental cause of frailty, functional decline and disability. In the year 2000, $18.5 billion in health care costs were directly attributable to sarcopenia. This economic burden will increase dramatically as the elderly population grows over the next decade. The primary causes of sarcopenia include a sedentary lifestyle and malnutrition. While resistance training appears to be a promising intervention, older individuals exhibit a blunted hypertrophic response to exercise stimuli. It has been posited that this decrement in regenerative capacity may be due to the loss of postprandial anabolism as well as an increase in reactive oxygen species. As such, a combination of resistance training and nutritional interventions may be a promising candidate in combating sarcopenia. Nevertheless, the mechanisms by which the manipulation of dietary variables may improve the sarcopenic condition are not well understood. To address this gap in extant knowledge, this review will examine the effects of protein, amino acid and/or antioxidant intake on sarcopenia both at rest and following resistance training exercise.  相似文献

Two redox-asymmetric amide-bridged bis(terpyridine)ruthenium(II) complexes (3a, 3b) have been prepared by amide coupling of a carboxylic acid functionalized complex with an amine substituted complex and they were fully characterized by spectroscopic analyses. They emit at 692 and 750 nm at room temperature in fluid solution with quantum yields larger than 10−3 and 3MLCT lifetimes of 22 ns. Ru···Ru distances were estimated from DFT models as 17.7 and 13.4 Å for 3a and 3b, respectively. Cyclic voltammetry gives two oxidation potentials for the differently substituted ruthenium sites with splittings of 0.10 and 0.23 V for 3a and 3b, respectively. Oxidation of 3b with CeIV ions gives the corresponding mixed-valent RuII-RuIII system which is valence-localized according to NIR spectroscopic and theoretical analyses.  相似文献

An epimerization-free system for coupling N-protected peptides with free amino acids was developed. A number of inorganic substances were tested as epimerization suppressant additives during the coupling by various methods (carbodiimide plus additives, uronium salts, Woodward's reagent-K, isobutyl-chloroformate, etc.). Some of them (ZnCl2, RbClO4, LiCl, SnCl4, AlCl3, etc.) in combination with some coupling methods can guarantee coupling with minimal epimerization (D-epimer < 1%). But only a simultaneous use of 1-hydroxybenzotriazole and Cu2+ ions as additives in carbodiimide-mediated peptide couplings appeared to give a standard result (D-epimer < 0.1%). There was no epimerization even in the case when N-methyl amino acid (sarcosine) was used as an amino component, while in the absence of Cu2+ ions an unacceptable level of epimerization was observed (D-epimer, 22% for carbodiimide with the 1-hydroxybenzotriazole method). So far it has been considered that Cu2+ ions prevent obtaining peptides in high yields (< 90%) by various coupling methods. We have found that the use of 1-hydroxybenzotriazole, CuCl2 and 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide instead of dicyclohexylcarbodiimide provides a possible method for obtaining the desired peptides in 90-99% yields without epimerization. All these results were shown by employing several model peptide couplings with free amino acids as amino components dissolved in an effective solvent system which readily dissolved them.  相似文献
