The microbiome associated with brown planthopper (BPH) plays an important role in mediating host health and fitness. Characterization of the microbial community and its structure is prerequisite for understanding the intricate symbiotic relationships between microbes and host insect. Here, we investigated the bacterial and fungal communities of BPH at different developmental stages using high‐throughput amplicon sequencing. Our results revealed that both the bacterial and fungal communities were diverse and dynamic during BPH development. The bacterial communities were generally richer than fungi in each developmental stage. At 97% similarly, 19 phyla and 278 genera of bacteria were annotated, while five fungal phyla comprising 80 genera were assigned. The highest species richness for the bacterial communities was detected in the nymphal stage. The taxonomic diversity of the fungal communities in female adults was generally at a relatively higher level when compared to other developmental stages. The most dominant phylum of bacteria and fungi at each developmental stage all belonged to Proteobacteria and Ascomycota, respectively. A significantly lower abundance of bacterial genus Acinetobacter was recorded in the egg stage when compared to other developmental stages, while the dominant fungal genus Wallemia was more abundant in the nymph and adult stages than in the egg stage. Additionally, the microbial composition differed between male and female adults, suggesting that the microbial communities in BPH were gender‐dependent. Overall, our study enriches our knowledge on the microbial communities associated with BPH and will provide clues to develop potential biocontrol techniques against this rice pest.  相似文献

Bacterial communities are known to play important roles during the developmental stages of insects, but current knowledge of bacteria associated with the midgut of Apis dorsata, the giant Asian honeybee, is limited. Using polymerase chain reaction‐denaturing gradient gel electrophoresis analysis (PCR‐DGGE) and 16S rRNA sequencing, the aim of this study was to determine the dynamics of bacterial community structure across four A. dorsata life stages in different geographical locations. The results reveal that bacterial diversity increased as the bee progressed through larval stage to newly emerged worker and old worker. However, in the pupal stage, no bands identified as bacteria could be observed. Overall, 2 bacterial phyla (Proteobacteria and Firmicutes) and 4 classes (Alphaproteobacteria, Betaproteobacteria, Gammaproteobacteria, and Bacilli) were identified, but the frequency varied among the different stages and locations. The classes of Gammaproteobacteria and Bacilli dominated among larval, newly emerged worker and old worker developmental stages.  相似文献

为了解大豆根际细菌群落结构多样性及根际细菌群落结构的变化,该研究以大豆苗期和成熟期的根际土壤为材料,采用Illumina高通量测序技术测定细菌16S rRNA V3+V4区序列,探究大豆不同生育期根际土壤细菌群落结构的变化。对原始数据进行拼接、过滤、去除嵌合体序列和聚类分析等数据处理,并对OTU进行分类学注释。在此基础上运用ANOVA分析物种组成变化,Alpha多样性指数研究细菌多样性变化。结果表明:细菌丰富度和多样性在不同生育期有显著变化,其中成熟期土壤中的细菌丰富度和多样性指数均明显高于苗期; 变形菌、放线菌、酸杆菌是大豆根际的优势菌门,其含量在不同生育期也有显著变化; 假诺卡氏菌属、糖丝菌属、鞘氨醇单胞菌属是大豆根际的优势菌属,这些菌属中的部分菌群属于根际促生菌,具有潜在的促生效应。这些结果证实大豆的生育期对根际土壤细菌群落结构有重要影响。  相似文献

We investigated microbial succession on lake sturgeon (Acipenser fulvescens) egg surfaces over the course of their incubation period as a function of simulated stream flow rate. The primary objective was to characterize the microbial community assembly during succession and to examine how simulated stream flow rate affect the successional process. Sturgeon eggs were reared under three flow regimes; high (0.55 m/s), low (0.18 m/s), and variable (0.35 and 0.11 m/s alternating 12 h intervals). Eggs were collected from each flow regime at different egg developmental stages. Microbial community DNA was extracted from egg surface and the communities were examined using 16S rRNA gene-based terminal restriction fragment length polymorphism and 454 pyrosequencing. Analysis of these datasets using principal component analysis revealed that microbial communities were clustered by egg developmental stages (early, middle, and late) regardless of flow regimes. 454 pyrosequencing data suggested that 90–98 % of the microbial communities were composed of the phyla Proteobacteria and Bacteroidetes throughout succession. β-Protebacteria was more dominant in the early stage, Bacteroidetes became more dominant in the middle stage, and α-Proteobacteria became dominant in the late stage. A total of 360 genera and 5,826 OTUs at 97 % similarity cutoff were associated with the eggs. Midway through egg development, the egg-associated communities of the low flow regime had a higher diversity than those communities developed under high or variable flow regimes. Results show that microbial community turnover occurred during embryogenesis, and stream flow rate influenced the microbial succession processes on the sturgeon egg surfaces.  相似文献

Blacklegged ticks (Ixodes scapularis) are one of the most important pathogen vectors in the United States, responsible for transmitting Lyme disease and other tick‐borne diseases. The structure of a host's microbial community has the potential to affect the ecology and evolution of the host. We employed high‐throughput sequencing of the 16S rRNA gene V3‐V4 hypervariable regions in the first study to investigate the tick microbiome across all developmental stages (larvae, nymphs, adults). In addition to field‐collected life stages, newly hatched laboratory‐reared larvae were studied to determine the baseline microbial community structure and to assess transovarial transmission. We also targeted midguts and salivary glands due to their importance in pathogen maintenance and transmission. Over 100 000 sequences were produced per life stage replicate. Rickettsia was the most abundant bacterial genus across all sample types matching mostly the Ixodes rickettsial endosymbionts, and its proportion decreased as developmental stage progressed, with the exception of adult females that harboured a mean relative abundance of 97.9%. Laboratory‐reared larvae displayed the lowest bacterial diversity, containing almost exclusively Rickettsia. Many of the remaining bacteria included genera associated with soil, water and plants, suggesting environmental acquisition while off‐host. Female organs exhibited significantly different β‐diversity than the whole tick from which they were derived. Our results demonstrate clear differences in both α‐ and β‐diversity among tick developmental stages and between tick organs and the tick as a whole. Furthermore, field‐acquired bacteria appear to be very important to the overall internal bacterial community of this tick species, with influence from the host bloodmeal appearing limited.  相似文献

【目的】黑果枸杞是一种耐盐植物,是我国西北干旱区盐渍土改良的优良植物物种,其根际土壤细菌群落结构在不同生长时期的变化特征尚不清楚。【方法】本研究采用Illumina MiSeq高通量测序研究了黑果枸杞3个生长阶段的根际土壤细菌群落结构的动态变化。【结果】所有样品中共获得317467条序列,对应于7028个细菌/古细菌OTUs。根际土壤细菌群落的α多样性显著高于非根际土壤。衰老期根际细菌的多样性和丰富度明显低于营养生长期和花/果期。变形菌门和酸杆菌门的相对丰度随生长时期的演变而逐渐降低,而蓝细菌门则相反。厚壁菌门的丰度在衰老期明显高于营养生长期和花/果期。优势属的组成也随生长期的演变而改变,营养生长期、花/果期、衰老期的优势属数量分别为17、16、4,且组成也具有差异。相似性分析表明营养生长期和花/果期的根际细菌群落具有很高的相似性,衰老期根际细菌群落组成与生长期和花/果期具有很高差异,然而与非根际土壤的群落结构具有较高的相似性。【结论】根际土壤细菌群落多样性和组成随生长期的改变而表现出明显的动态变异性,表明黑果枸杞生长时期对根际土壤细菌群落结构具有重要的影响。  相似文献

In the present study, we determined the critical stages and the key factors of mortality for Planococcus citri (Hemiptera: Pseudococcidae) in Brazilian coffee plantations using a life table to understand the role of natural biological control on its population. Predators, parasitoids, rainfall, sunlight, physiological disturbances and fungal diseases were collectively responsible for 98.79% in the total mortality of P. citri. Predators belonging to the Chrysopidae, Syrphidae, Dolichopodidae and Coccinellidae families were the most important mortality factors in the early developmental stages of P. citri (i.e. eggs and 1st and 2nd instar nymphs), whereas predators belonging to the Coccinellidae and Chrysopidae families were the most important mortality factors for the last instars (i.e. 3rd instars and adults) for P. citri. The generalist predators Harmonia axyridis, Chrysoperla genanigra and Chrysoperla externa were the key mortality factors for P. citri. The third nymph stage was considered the critical life stage (i.e. the life stage that most influences population size). Our results show that generalist predators and climatic factors are important sources of natural mortality of P. citri governing the population dynamics of this pest in the field.  相似文献

The life cycle and several life parameters of the zoophytophagous predator Engytatus varians (Distant) (Heteroptera: Miridae), including nymphal growth according to Dyar’s rule, were examined in the laboratory. The egg, nymph (five instars), and adult stages were 9.20, 17.36, and 19.02?d in length, respectively. The growth ratio for nymphs was consistent with Dyar’s rule based on the lengths of the femora of the forelegs, the tibiae and femora of the middle legs, and the antennae. Some biological characteristics of E. varians were also evaluated when the mirid was fed three different diets (B. cockerelli third instars, Sitotroga cerealella Olivier [Lepidoptera: Gelechiidae] eggs, and a mixture of both instars and eggs). The length of the nymphal stage was three days longer on a diet of only S. cerealella eggs than when the mirids were fed the third instars of B. cockerelli only or a mixture of both. The sex ratio was not affected by the type of diet. Nymphs of E. varians consumed B. cockerelli nymphs (80–85) when fed third instars only and third instars?+?S. cerealella eggs, respectively. The potential use of this predator as a biological control agent of B. cockerelli is discussed.  相似文献

[目的]研究连作条件下番茄青枯病不同发病时期的非根际土壤微生物群落差异,明确土壤微生物对青枯病发病时期的响应机制。[方法]本研究对16S rRNA V4-V5区进行实时荧光定量PCR和高通量测序,综合分析了连续种植第1、3、5和7季的发病高峰期和发病末期的番茄非根际土壤的病原菌数量、细菌群落多样性、群落组成、标志细菌类群和群落构建机制。[结果]发病高峰期的青枯菌数量(1.28×107 copies/g)要高于发病末期(1.77×106 copies/g)。随着连作时间的增加,发病时期对细菌群落多样性的影响逐渐增加。第3季和第5季不同发病时期之间的细菌群落alpha和beta多样性存在显著差异。LEfSe分析发现,番茄青枯病发病高峰期和发病末期的非根际土壤样品有其各自不同的标志细菌类群。此外,随连作时间的延长协助青枯菌致病的细菌逐渐累积。发病高峰期是微杆菌属(Microbacterium)和亚硝化螺菌属(Nitrosospira)协助青枯菌致病,而在发病末期,则由鞘脂菌属(Sphingobium)、norankf...  相似文献

中华大刀螳Tenodera sinensis Saussure既是传统的中药材,又是重要的天敌昆虫,因而具有重要的经济价值.为了充分开发和利用中华大刀螳资源,在人工饲养条件下,对其形态学特征和生物学特性进行了研究.结果表明:中华大刀螳为渐变态昆虫,一生经历卵、若虫和成虫3个虫态.室内1年完成1个世代,且无越冬现象.若虫共8龄,以1龄若虫发育历期最短,平均为11.73 d,8龄若虫发育历期最长,平均为24.75 d;3龄若虫存活率最低,为81.08%,5龄若虫存活率最高,为100%;雌、雄成虫阶段的平均历期为99.36 d和72.86 d.此外,本文介绍了中华大刀螳各虫态的形态特征和生物学特性.  相似文献

Total lipid and the fatty acid compositions of phospholipid and triacylglycerol fractions, prepared from eggs, 3rd instars of larvae, pupae, male and female adults of Lertha sheppardi, were analyzed by gas chromatography and gas chromatography-mass spectrometry. The effect of diet (adults’ nutrition) on fatty acid composition of L. sheppardi adults was also investigated. Total lipid of L. sheppardi considerably increased in adults compared with immature stages. There was a significant decrease in total lipid level in larval stage in contrast with egg stage. Qualitative analysis revealed the presence of 14 fatty acids during all stages. The major components were C16 and C18 saturated and unsaturated components which are ubiquitous to most animal species. In addition to these components, one odd-chain (C17:0) and prostaglandin precursor fatty acids were found. The fatty acid profiles of phospholipids and triacylglycerols were substantially different. In phospholipid fraction, monounsaturated fatty acids were the major proportion of fatty acids in both sex of adults and pupae, whereas polyunsaturated fatty acids were the most dominant fatty acids in eggs and 3rd instars. Results of triacylglycerol fraction revealed that fatty acid composition of eggs had higher level of C16:1, C18:0 and C18:3n-3 content than that of 3rd instars and pupae, which suggests accumulation of energetic and structural reserve materials during embryonic development. At more advanced developmental stages, mainly in adult females, the amount of C16:1 increased once again, which may be related to the need for accumulation of sufficient energy and of carbon reservoir in the developing new vitellum. Percentages of C18:1 were significantly high in adult stages compared to other stages. These findings indicate that the accumulation and consumption of fatty acids fluctuate through different development stages. Diet did not effect the fatty acid composition of L. sheppardi adults.  相似文献

【目的】通过检测郫县豆瓣在不同发酵阶段细菌的种类、丰度及数量,探究郫县豆瓣的不同发酵产品发酵过程中细菌的动态变化情况。【方法】采用16S rRNA基因测序对郫县豆瓣4个发酵阶段中细菌种类及丰度进行分析,利用荧光定量PCR(quantitative real-time PCR,qPCR)方法检测不同发酵阶段的细菌总数。【结果】郫县豆瓣在初期的发酵过程中细菌群落处于动态稳定,在不同发酵阶段细菌群落组成相对丰富,从郫县豆瓣整个初期的发酵过程来看,细菌群落多样性呈现升高的趋势,Shannon指数从1.25升高到3.50;在郫县豆瓣初期发酵过程中细菌群落的数量以及多样性与发酵环境息息相关,不同发酵阶段细菌群落的多样性也有所不同,其中在干辣椒发酵阶段中泛菌属(Pantoea)为最优势菌属,占比为20%;在蚕豆瓣发酵阶段中葡萄球菌属(Staphylococcus)的相对丰度最高,占比为38%;混合发酵后,在红油豆瓣发酵阶段的最优势菌属是乳酸杆菌属(Lactobacillus),占比达到27%,郫县豆瓣发酵阶段的最优势菌属是乳酸杆菌属(Lactobacillus),占比为62%。【结论】推断在郫县豆瓣不同发酵阶段初期相对丰度较大的菌属对郫县豆瓣的质量以及产量可能会产生重大影响。  相似文献

【目的】探讨盐角草根部内生细菌群落多样性特征,揭示内生细菌群落结构在宿主关键发育期动态变化规律。【方法】通过罗氏454高通量测序获得内生细菌16S r RNA片段,然后进行生物信息分析。【结果】共获得20363条16S r RNA基因序列。各样品中可操作分类单元(operational taxonomic units,OTUs)在552–941之间。根部内生细菌群落主要包括4个门,其中Proteobacteri门占主导地位,其余依次是Firmicutes,Actinobacteria,Bacteroidetes。在Proteobacteria门中,Gammaproteobacteria是第一大纲,其后是Betaproteobacteria纲。宿主5个发育时期共同拥有7个细菌属,包括Azomonas,Serratia,Pantoea,Serpens,Pseudomonas,Halomonas,Kushneria。整体上看,Gammaproteobacteria纲在宿主5个发育时期呈现增长趋势。优势菌属在5个发育期存在差异,分别为Delftia,Kushneria,Serratia,Pantoea,Erwinia。所有文库总共含2108个特异OTUs,共同拥有5个相同OTUs。花期OTUs数量最多,结种期内生细菌多样性降低。在宿主的5个发育时期中,土壤p H、月均温和土壤盐含量这3个环境因子组成的集合对其内生细菌群落变化具有显著影响。【结论】盐角草内生细菌群落多样性丰富,宿主发育期决定了内生细菌群落结构。  相似文献

The function of “cuticular hydrocarbons” (CHCs) in insects is to protect the body as well as chemical communication and recognition between species. Gas chromatography and mass spectrometry were used to compare the composition of CHCs in eggs, all nymph instars, and male and female adults of the bean bug, Riptortus pedestris (Hemiptera: Alydidae). CHC carbon chain numbers of each developmental stage ranged from 14 to 19 in eggs to the fourth nymph and were 24 in the last nymph. CHCs' carbon chain numbers in male and female adults increased from 16 to 23 and 16 to 27 until 6 days of age, respectively. Major components consisted of n-nonacosane in the larval stages, which changed to n-hentriacontane, n-heptacosane and 13,17-;15,19-demethyltritriacontane in adults. The major component from eggs was not identified. The major component from newly emerged adults was n-heptacosane. CHCs in different developmental stages of the bean bug consisted of n-alkanes with saturated hydrocarbons (27–63%), followed by mono or di-methyl alkanes. However, the CHCs of newly emerged female and male adults consisted mostly of methyl-alkanes. The major constituents of CHCs in the developmental stages of R. pedestris were proportionally different but showed only slight differences in composition.  相似文献

[目的]揭示巨菌草种植前及其不同生长时期对砒砂岩地区土壤基本理化性质及细菌群落结构的影响,为更好应用巨菌草改良砒砂岩地区土壤提供理论依据。[方法]运用Illumina MiSeq高通量测序技术,在门、属水平上比较砒砂岩地区巨菌草不同生长时期对土壤细菌结构及多样性的影响。[结果]巨菌草种植后,三块样地的含水量在巨菌草分蘖期最低,在成熟期达到最高,土壤有机含量有所增加,土壤pH值变化不明显。细菌菌群结构:各样地的土壤细菌在巨菌草种植前及其生长的四个时期,门水平上的优势菌群均为放线菌门(Actinobacteria)、变形菌门(Proteobacteria);在属水平上,含量最高的是未分类(unidentified)的细菌。Alpha多样性及群落组成丰度分析表明,三块样地种植巨菌草后,其细菌多样性和物种丰度均比种植前高,农田地土壤细菌多样性和物种丰度在巨菌草拔节期达到峰值,在分蘖期土壤细菌数量最大,比种植前土壤细菌数量高出8倍多,达2.09×107 CFU/g干土,山顶地土壤细菌多样性和物种丰度在巨菌草分蘖期达到峰值,河滩地土壤细菌多样性在巨菌草苗期达到峰值,其丰富度在拔节期达到最大。[结论]砒砂岩地区土壤细菌群落结构在巨菌草不同生长时期相似,巨菌草能明显促进砒砂岩地区土壤细菌菌群多样性和丰富度,能有效提高土壤有机质含量,但砒砂岩地区各样地土壤细菌多样性、数量和有机质含量达到峰值的时期不同。  相似文献

Crop production may benefit from plant growth-promoting bacteria. The knowledge on bacterial communities is indispensable in agricultural systems that intend to apply beneficial bacteria to improve plant health and production of crops such as canola. In this work, the diversity of root bacterial communities associated to two different developmental phases of canola (Brassica napus L.) plants was assessed through the application of new generation sequencing technology. Total bacterial DNA was extracted from root samples from two different growth states of canola (rosette and flowering). It could be shown how bacterial communities inside the roots changed with the growing stage of the canola plants. There were differences in the abundance of the genera, family, and even the phyla identified for each sample. While in both root samples Proteobacteria was the most common phylum, at the rosette stage, the most common bacteria belonged to the family Pseudomonadaceae and the genus Pseudomonas, and in the flowering stage, the Xanthomonadaceae family and the genus Xanthomonas dominated the community. This implies in a switch in the predominant bacteria in the different developmental stages of the plant, suggesting that the plant itself interferes with the associated microbial community.  相似文献

[目的]陆生植物根际环境与土壤中的微生物菌群关系密切,其根际微生物群落动态极可能直接影响着植物健康及养分高效利用。虽然根际益生菌已被证实可用于提高作物生产力,但由于缺乏对这些菌群组成动态变化规律的认识了解,它们的开发受到限制。研究马铃薯全生育期根际菌群的动态变化规律,探讨根际菌群变化与马铃薯发育时期的相关性,为针对马铃薯不同生长时期开发专用生物益生菌肥奠定理论基础。[方法]本研究着眼于马铃薯田间全生命周期微生物组动态变化,通过Illumina MiSeq高通量测序技术对不同时间点马铃薯根际细菌16S rRNA基因V3-V4区和真菌ITS区测序并对操作分类单位(OTU)进行聚类,分析样品间微生物群落的多样性特征,并通过机器学习的方法建立模型,将根际菌群与田间马铃薯发育时间相关联。[结果]根际菌群在马铃薯各个发育阶段随时间变化明显,营养生长阶段的微生物群落结构发生了显著变化,随着结薯期的开始逐渐稳定,直到块茎成熟后期根际菌群再次出现较大变化,且在不同施肥处理间呈现较大差异。进一步基于模型挖掘了与马铃薯发育时间相关联的22个特征细菌类群和16个特征真菌类群,其中苗期和结薯末期的特征类群分别为梭菌(Clostridium)和放线菌(Actinobacteria)。[结论]马铃薯的生长发育时期是影响根际微生物群落组成的主要因素,益生菌肥的添加主要影响马铃薯结薯末期的细菌微生物菌群结构。  相似文献

Changes in temperature, pH, moisture, C/N ratio, and bacterial community were monitored in Istanbul full-scale composting plant. C/N ratio steadily decreased during composting and final mature compost products had a C/N ratio of less than 20. During the composting process, temperature was mostly above 55 °C and decreased to mesophilic conditions in the matured stages. Different types of bacteria were dominant in every stage of composting and bacterial diversity changed mainly by temperature. Bacillus species were dominant in early stages of composting while Acinetobacter and Sphingobacterium strains were detected in thermophilic and maturing stages. Bacteria were mainly active in the degradation of cellulose and toxic organics while some strains had denitrification ability. Generally, thermophilic stages were more rich in bacterial diversity than mesophilic and hyperthermophilic conditions significantly changed the bacterial community.  相似文献

【目的】意大利蝗Calliptamus italicus是新疆草原的主要优势危害种,以卵在土壤中越冬。呼吸代谢可反映蝗卵的生理状态,呼吸蛋白对于呼吸系统不完善的蝗卵尤为重要。本研究旨在明确意大利蝗卵发育过程中血蓝蛋白基因的表达情况。【方法】采用实时荧光定量PCR方法检测不同发育阶段的蝗卵以及1龄蝗蝻的血蓝蛋白2个亚基基因Hc1和Hc2的表达量。【结果】根据解剖形态观察,将意大利蝗越冬卵的整个发育过程分为10个阶段,包括9个卵发育阶段(C-Ⅰ-C-Ⅹ)和1龄蝗蝻阶段(C-Ⅹ)。Hc1和Hc2在越冬蝗卵各发育阶段以及1龄蝗蝻中均有表达。其中,在蝗卵早期发育阶段(C-Ⅰ, C-Ⅱ和C-Ⅲ),Hc1表达量逐渐增加,C-Ⅲ阶段表达量显著高于C-Ⅰ和C-Ⅱ阶段;滞育阶段(C-Ⅳ, C-Ⅴ和C-Ⅵ),胚胎发育停滞,Hc1表达量较C-Ⅲ,C-Ⅶ和C-Ⅷ阶段低;滞育后发育阶段(C-Ⅶ和C-Ⅷ),蝗卵解除滞育,快速发育,Hc1表达量较早期发育阶段和滞育阶段高,其中,C-Ⅷ阶段Hc1表达量最高(212.3156±10.5470),显著高于其他所有阶段;1龄蝗蝻(C-Ⅹ)的Hc1表达量最低,为0.4017±0.1010。Hc2表达量在C-Ⅴ阶段最高(679.7511±54.5719),显著高于其他所有阶段;除C-Ⅴ阶段外,其他各阶段之间Hc2表达量差异均不显著。Hc1在蝗卵滞育后阶段高表达,而Hc2在蝗卵滞育阶段高表达。【结论】血蓝蛋白亚基基因Hc1和Hc2在整个意大利蝗卵发育过程均有表达,且具有阶段特异性。Hc1与Hc2协同作用为蝗卵发育供氧,其中,Hc1主要负责蝗卵滞育后发育期间的氧气运载,而Hc2主要维持滞育期间的氧气运载,且载氧效率较低。研究结果可为进一步探讨意大利蝗卵的抗逆机制提供科学依据。  相似文献
