Intact cells of the unicellular cyanobacterium Synechococcus UTEX 625 degraded exogenously supplied cyanate (as KOCN) to CO2 and NH3 in a light-dependent reaction. NH3 release to the medium was as high as 80 mol(mgChl)-1h-1 and increased 1.7-fold in the presence of methionine sulfoximine, a glutamine synthetase inhibitor. Cyanate also supporte photosynthetic O2 evolution to a maximum rate of 188 mol O2(mgChl)-1h-1 at pH 8 and 30°C. Cyanate decomposition in cell-free extracts, measured by mass spectrometry as 13CO2 production from KO13CN, occurred in the soluble enzyme fraction, but not in the thylakoid/carboxysome fraction, and was enhanced by HCO3 and inhibited by the dianion oxalate. CO2, rather, than HCO3 , was a product of cyanate decomposition. The ability to decompose cyanate was not dependent upon pre-exposure of cells to cyanate to induce activity. The collective results indicate that Synechococcus UTEX 625 possesses a constitutive, cytosolic cyanase (EC 4.3.99.1), similar in mechanism to that found in some species of heterotrophic bacteria. The reaction catalyzed was: OCN+HCO3+2H+2CO2+NH3. In intact cells, the CO2 produced by the action of cyanase on OCN- was either directly fixed by the Calvin cycle enzyme ribulose-1,5-bisphosphate carboxylase/oxygenase, leading to O2 evolution, or leaked into the medium where it was returned to the cell by the active CO2/HCO3 transport systems for fixation. The relaxation of the non-photochemical quenching of chlorophyll fluorescence has been investigated in cells of the green alga Dunaliella following illumination. The relaxation after the addition of DCMU or darkening was strongly biphasic. The uncoupler NH4Cl induced rapid relaxation of both phases, which were therefore both energy-dependent quenching, qE. The proportion of the slow phase of qE increased at increasing light intensity. In the presence of the inhibitors rotenone and antimycin the slow phase of qE was stabilised for in excess of 15 min. NaN3 inhibited the relaxation of almost all the qE. The rate of glycolate excretion in Euglena gracilis Z and some microalgae grown at the atmospheric level of CO2 was determined using amino-oxyacetate (AOA). The extracellular O2 concentration was kept at 240 M by bubbling the incubation medium with air. Glycolate, the main excretion product, was excreted by Euglena at 6 mol·h-1·(mg chlorophyll (Chl))-1. Excretion depended on the presence of AOA, and was saturated at 1 mM AOA. A substituted oxime formed from glyoxylate and AOA was also excreted. Bicarbonate added at 0.1 mM did not prevent the excretion of glycolate. The excretion of glycolate increased with higher O2 concentrations in the medium, and was competitively inhibited by much higher concentrations of bicarbonate. Aminooxyacetate also caused excretion of glycolate from the green algae, Chlorella pyrenoidosa, Scenedesmus obliquus and Chlamydomonas reinhardtii grown on air, at the rates of 2–7 mol·h-1·(mg Chl)-1 in the presence of 0.2–0.6 mM dissolved inorganic carbon, but the cyanobacterium, Anacystis nidulans, grown in the same way did not excrete glycolate. Chloroplasts with high rates of photosynthetic O2 evolution (up to 120 mol O2· (mg Chl)-1·h-1 compared with 130 mol O2· (mg Chl)-1·h-1 of whole cells) were isolated from Chlamydomonas reinhardtii cells grown in high and low CO2 concentrations using autolysine-digitonin treatment. At 25° C and pH=7.8, no O2 uptake could be observed in the dark by high- and low-CO2 adapted chloroplasts. Light saturation of photosynthetic net oxygen evolution was reached at 800 mol photons·m-2·s-1 for high- and low-CO2 adapted chloroplasts, a value which was almost identical to that observed for whole cells. Dissolved inorganic carbon (DIC) saturation of photosynthesis was reached between 200–300 M for low-CO2 adapted chloroplasts, whereas high-CO2 adapted chloroplasts were not saturated even at 700 M DIC. The concentrations of DIC required to reach half-saturated rates of net O2 evolution (Km(DIC)) was 31.1 and 156 M DIC for low- and high-CO2 adapted chloroplasts, respectively. In C4 grasses belonging to the NADP-malic enzyme-type subgroup, malate is considered to be the predominant C4 acid metabolized during C4 photosynthesis, and the bundle sheath cell chloroplasts contain very little photosystem-II (PSII) activity. The present studies showed that Flaveria bidentis (L.), an NADP-malic enzyme-type C4 dicotyledon, had substantial PSII activity in bundle sheath cells and that malate and aspartate apparently contributed about equally to the transfer of CO2 to bundle sheath cells. Preparations of bundle sheath cells and chloroplasts isolated from these cells evolved O2 at rates between 1.5 and 2 mol · min–1 · mg–1 chlorophyll (Chl) in the light in response to adding either 3-phosphoglycerate plus HCO 3 or aspartate plus 2-oxoglutarate. Rates of more than 2 mol O2 · min–1 · mg–1 Chl were recorded for cells provided with both sets of these substrates. With bundle sheath cell preparations the maximum rates of light-dependent CO2 fixation and malate decarboxylation to pyruvate recorded were about 1.7 mol · min–1 · mg–1 Chl. Compared with NADP-malic enzyme-type grass species, F. bidentis bundle sheath cells contained much higher activities of NADP-malate dehydrogenase and of aspartate and alanine aminotransferases. Time-course and pulse-chase studies following the kinetics of radiolabelling of the C-4 carboxyl of C4 acids from 14CO2 indicated that the photosynthetically active pool of malate was about twice the size of the aspartate pool. However, there was strong evidence for a rapid flux of carbon through both these pools. Summary The apparent energy of activation (E a), Michaelis-Menten constant (K mfor oxaloacetate), V max/K mratios and specific activities of NADP+-malate dehydrogenase (NADP+-MDH; EC 1.1.1.82) were analyzed in plants of Barnyard grass from Québec (QUE) and Mississippi (MISS) acclimated to two thermoperiods 28/22°C, 21/15°C, and grown under two CO2 concentrations, 350 l l-1 and 675 l l-1. E avalues of NADP+-MDH extracted from QUE plants were significantly lower than those of MISS plants. K mvalues and V max/K mratios of the enzyme from both ecotypes were similar over the range of 10–30°C but reduced V max/K mratios were found for the enzyme of QUE plants at 30 and 40°C assays. MISS plants had higher enzyme activities when measured on a chlorophyll basis but this trend was reversed when activities were expressed per fresh weight leaf or per leaf surface area. Activities were significantly higher in plants of both populations acclimated to 22/28°C. CO2 enrichment did not modify appreciably the catalytic properties of NADP+-MDH and did not have a compensatory effect upon catalysis or enzyme activity under cool acclimatory conditions. NADP+-MDH activities were always in excess of the amount required to support observed rates of CO2 assimilation and these two parameters were significantly correlated. The enhanced photosynthetic performance of QUE plants under cold temperature conditions, as compared to that of MISS plants, cannot be attributed to kinetic differences of NADP+-malate dehydrogenase among these ecotypes.  相似文献

Two characteristic temperatures were identified from measurements of the temperature dependence of O2 evolution by Chlorella vulgaris and Anacystis nidulans: T1, the threshold temperature for inhibition of O2 evolution under saturating light conditions, and T2, the upper temperature limit for O2 evolution. Measurement of delayed light emission from photosystem II (PSII) showed that it passed through a maximum at T1 and was virtually eliminated on heating the samples to T2. Related changes were observed in low-temperature (77K) fluoresence emission spectra. Heat-stress had little effect on the absorption properties of the cells at temperatures below T1 but incubation at higher temperatures, particularly under high-light conditions, resulted in extensive absorption losses. An analysis of these measurements suggests that this increased susceptibility to photobleaching is triggered by an inhibition of the flow of reducing equivalents from PSII that normally serves to protect the light-harvesting apparatus of the cells from photo-oxidation. We report here the first measurements on chlorophyll (Chl) a fluorescence characteristics of photoautotrophic soybean cells (cell lines SB-P and SBI-P). The cell fluorescence is free from severe distortion problems encountered in higher plant leaves. Chl a fluorescence spectra at 77 K show, after correction for the spectral sensitivity of the photomultiplier and the emission monochromator, peaks at 688, 696 and 745 nm, representing antenna systems of photosystem II-CP43 and CP47, and photosystem I, respectively. Calculations, based on the complementary area over the Chl a fluorescence induction curve, indicated a ratio of 6 of the mobile plastoquinone (including QB) to the primary stable electron acceptor, the bound plastoquinone QA. A ratio of one between the secondary stable electron acceptor, bound plastoquinone QB, and its reduced form QB - was obtained by using a double flash technique. Owing to this ratio, the flash number dependence of the Chl a fluorescence showed a distinct period of four, implying a close relationship to the S state of the oxygen evolution mechanism. Analysis of the QA - reoxidation kinetics showed (1) the halftime of each of the major decay components ( 300 s fast and 30 ms slow) increases with the increase of diuron and atrazine concentrations; and (2) the amplitudes of the fast and the slow components change in a complementary fashion, the fast component disappearing at high concentrations of the inhibitors. This implies that the inhibitors used are able to totally displace QB. In intact soybean cells, the relative amplitude of the 30 ms to 300 s component is higher (40:60) than that in spinach chloroplasts (30:70), implying a larger contribution of the centers with unbound QB. The progeny of Herpes simplex virus type 1 (HSV-1) grown in ricin-resistant 14 cells (RicR14) lackingN-acetylglucosaminyltransferase I was released in the extracellular medium at a very low rate. By using a monoclonal antibody immobilized on Sepharose we purified from HSV-1-infected RicR14 cells a viral glycoprotein (gC), which carries bothN-andO-linked oligosaccharides. Glycopeptides obtained from [3H]mannoselabeled gC by Pronase digestion were entirely susceptible to endo--N-acetylglucosaminidase H, and the major oligosaccharide released was Man4GlcNAc. The accumulation of this high-mannose species was related to the enzymic defect of the host cells and to the long retention of the viral glycoprotein within the cells. Raising the pO2 reduced nitrogenase activity (C2H2 reduction) of Anabaena cylindrica for both glyoxylate-treated (5 mM) and untreated cells. The stimulation caused by glyoxylate, however, increased with increases of pO2 from 2 to 99 kPa. As the pO2 increased the net CO2 fixation was lowered (Warburg effect) while the CO2 compensation point increased. Glyoxylate partly relieved this sensitivity of net photosynthesis to oxygen and reduced the compensation point considerably. The cells used were preincubated in the dark to exhaust photosynthetic pools. A more pronounced reduction in sensitivity of nitrogenase to oxygen for glyoxylate-treated cells was evident when a preincubation in air with reduced pCO2 (13 l l-1) was used. This was, however, not evident until after a 10-h incubation in air. Before this point 2 kPa O2 sustained the highest nitrogenase activity. Addition of 0.5 and 5 mM of HCO 3 - to Anabaena cultures preincubated at low CO2 levels (29 l l-1) abolished the stimulatory effect of glyoxylate on the nitrogenase. Thus, the results sustain the suggestion that glyoxylate may act as an inhibitor of photorespiratory activities in cyanobacteria and can be used as a means of increasing their nitrogen and CO2 fixation capacities.Abbreviation RuBP ribulose 1,5-bisphosphate  相似文献

Exchange of CO2 and O2 and chlorophyll fluorescence were measured in the presence of 360 1 · 1–1 CO2 in nitrogen in Helianthus annuss L. leaves which had been preconditioned in the dark or at a photon flux density (PFD) of 24 mol · m–2 · s–1 either in 21 or 0% O2. An initial light-dependent O2 outburst of 6 mol · m–2 was measured after aerobic dark incubation. It was attributed to the reduction of electron carriers, predominantly plastoquinone. The maximum initial rate of O2 evolution at PFD 8000 mol · m–2 · s–1 was 170 mol · m–2 · s–2 or about four times the steady CO2-and light-saturated rate of photosynthesis. Fluorescence measurements showed that the rate was still acceptor-limited. Fast O2 evolution ceased after electron carriers were reduced in the dark-adapted leaf, but continued for a short time at the lower rate of 62 mol · m–2 · s–1 in the light-adapted leaf. The data are interpreted to show that enzymes involved in 3-phosphoglycerate reduction are dark-inhibited, but were fully active in low light. In a dark-adapted leaf, respiratory CO2 evolution continued under nitrogen; it was partially inhibited by illumination. Prolonged exposure of a leaf to anaerobic conditions caused reducing equivalents to accumulate. This was shown by a slowly increasing chlorophyll fluorescence yield which indicated the reduction of the PSII acceptor QA in the dark. When the leaf was illuminated, no O2 evolution was detected from short light pulses, although transient O2 production was appreciable during longer light pulses. Trichodesmium sp., isolated from the Great Barrier Reef lagoon, was cultured in artificial seawater media containing a range of Fe concentration. Fe additions stimulated growth, N2 fixation, cellular chlorophyll a content, light-saturated chlorophyll a-specific gross photosynthetic capacity (Pm chla) and the dark respiration rate (Rd chla). Cell yields only doubled for 9 nM Fe relative to zero added Fe, whereas N2 fixation increased 11-fold considerably for 450 nM Fe. The results suggest that N2 fixation of Trichodesmium is more sensitive to Fe limitation than are the cell yields.  相似文献

The role of oxygen in the photoinactivation of the photosynthetic apparatus of Spinacia oleracea L. was investigated. Moderate irradiation (1200 mol photons m-2s-1) of spinach leaves in an atmosphere of pure nitrogen caused strong inhibition of subsequently measured net CO2 assimilation, whereas considerably less photoinhibition was observed in the presence of low partial pressures (10–20 mbar) of O2. The decrease in activity caused by anaerobiosis in the light was not based on stomatal closure; the decline of assimilation represents a photoinhibition, as activity was not impaired by low irradiation (80 mol photos m-2s-1). In contrast, gassing with pure N2 in the dark caused strong inhibition. Electron-transport rates and chlorophyll-fluorescence data of thylakoids isolated from photoinhibited leaves indicated damage to the electron-transport system, in particular to photosystem II reaction centers. In vitro, photoinhibition in isolated thylakoid membranes was also strongly promoted by anaerobiosis. Photoinhibition of electron-transport rates under anaerobic conditions was characterized by a pronounced increase in the initial fluorescence level, F0, of chlorophyll-fluorescence induction, in contrast to photoinhibition under aerobic conditions. Methanobacterium thermoautotrophicum was grown in continuous culture in a fermenter gassed with H2 and CO2 as sole carbon and energy sources, and in a medium which contained either NH4Cl or gaseous N2 as nitrogen source. Growth was possible with N2. Steady states were obtained at various gas flow rates with NH4Cl and with and the maintenance coefficient varied with the gas input and with the nitrogen source. Factors influencing the rate of superoxide (O 2 - ) production by thylakoids were investigated to determine if increased production of the radical was related to injury induced by chilling at a moderate photon flux density (PFD). Plants used were Spinacia oleracea L., Cucumis sativus L. and Nerium oleander L. grown at either 200° C or 45° C. Superoxide production was determined by electron-spin-resonance spectroscopy of the (O 2 - )-dependent rate of oxidation of 2-ethyl-1-hydroxy-2,5,5-trimethyl-3-oxazolidine (OXANOH) to the corresponding oxazolidinoxyl radical, OXANO ·. For all plants, the steady-state rate of O 2 - production by thylakoids, incubated at 25° C and 350 mol photon · m–2 · s–1 (moderate PFD) with added ferredoxin and NADP, was between 7.5 and 12.5 mol · (mg chlorophyll)–1 · h–1. Incubation at 5° C and a moderate PFD, decreased the rate of O 2 - production 40% and 15% by thylakoids from S. oleracea and 20° C-grown N. oleander, chillinginsensitive plants, but increased the rate by 56% and 5% by thylakoids from C. sativus and 45° C-grown N. oleander, chilling-sensitive plants. For all plants, the addition of either ferredoxin or methyl viologen increased the rate of O 2 - -production at 25° C by 75–100%. With these electron acceptors, lowering the temperature to 5° C caused only a slight decrease in O 2 - production. In the absence of added electron acceptors, thylakoids produced O 2 - at a rate which was about 45% greater than that when ferredoxin and NADP were present. The addition of 3-(3,4-dichlorophenyl)-1,1-dimethylurea reduced O 2 - production under all conditions tested. The results show that the rate of O 2 - production increases in thylakoids when the rate of electron transfer to NADP is reduced. The intracellular concentration of cAMP in the green alga Chlorella fusca was in the range of 2 · 10-9 to 10-8 moles/g dry weight and was strongly dependent on the growth conditions. The cAMP level was high with high light intensity, low nitrate or glucose concentration. Intracellular cAMP increased only by factor of 2 when high amounts (up to 10-3 M) of cAMP were added to the medium. Most of the given cAMP was converted to 5-AMP.Addition of cAMP had little effect on the chlorophyll content of the cells, only at 10-6 M some enhancement in photoautotrophic cultures was observed. On the other hand high amounts of cAMP in the medium increased the growth rate. DBcAMP* showed a positive effect on chlorophyll synthesis and growth rate at much lower concentrations compared to cAMP.Stimulation effects of exogenous cAMP on the synthesis of chlorophyll were also observed in mixotrophic cultures with a high glucose/nitrate ratio, conditions where chlorophyll synthesis is repressed. Light-energy absorption by Microcystis aeruginosa with and without gas vacuoles was observed, respectively by using an integrating sphere photometer. As far as the concentration of cell suspension of the order of 1067 cells/ml in this work was concerned, the performance of gas vacuoles to shield incident light was most unlikely. Referring to a correlation secured by the integrating sphere photometer between light absorption and cell concentration of the suspension, a turbidostat culture of the blue-green alga demonstrated that the growth efficiency, Y kJ defined as g cells harvested per kJ of light energy absorbed by the cells was nearly 0.004. In the cyanobacterium Synechococcus UTEX 625, the extent of expression of carboxysomes appeared dependent on the level of inorganic carbon (CO2+HCO inf3 sup- ) in the growth medium. In cells grown under 5% CO2 and in those bubbled with air, carboxysomes were present in low numbers (<2 · longitudinal section-1) and were distributed in an apparently random manner throughout the centroplasm. In contrast, cells grown in standing culture and those bubbled with 30 l CO2 · 1-1 possessed many carboxysomes (>8 · longitudinal section-1). Moreover, carboxysomes in these cells were usually positioned near the cell periphery, aligned along the interface between the centroplasm and the photosynthetic thylakoids. This arrangement of carboxysomes coincided with the full induction of the HCO inf3 sup- transport system that is involved in concentrating inorganic carbon within the cells for subsequent use in photosynthesis. Immunolocalization studies indicate that the Calvin cycle enzyme ribulose bisphosphate carboxylase was predominantly carboxysome-localized, regardless of the inorganic carbon concentration of the growth medium, while phosphoribulokinase was confined to the thylakoid region. Internodal and whorl (branch) cells of the green alga,Chara corallina Klein ex Willd., em. R.D.W., were studied with the extracellular vibrating probe for measuring transmembrane ion currents, and with an extracellular pH microprobe for measuring the surface pH profile. Bands of positive inward current (OH- efflux) 1–3 mm wide were separated by wider bands of outward current (HCO 3 - influx) along the length of the cell. The measured peaks of inward current ranged from 20 to 60 A cm-2 (98 m from the cell surface) which would correspond to a surface ionic flux of 270–800 pmol cm-2 s-1. The peaks of outward current (HCO 3 - influx) ranged from 10 to 30 A cm-2 which would correspond to a surface ionic flux of 140–400 pmol cm-2 s-1. The inward current bands matched the regions of surface alkalinity very well. The outward current (HCO 3 - influx) was reduced at least 10-fold in low-HCO 3 - medium, with a commensurate readjustment in the strength and pattern of inward current (OH- efflux). (Although these experiments involved a manipulation of the external pH, it is felt that the main adjustment in current patterns was in response to the reduction in exogenous HCO 3 - ). The presence of the vibrating probe perturbed the inward current region when vibrating with a 26-m amplitude, but this perturbation was eliminated when a 7-m amplitude was used. The perturbation was usually observed as a reduction in the number of inward current peaks with an increase (approximate doubling) in the amplitudes of the one or two remaining peaks. Both the inward and outward currents were light-dependent, falling off within seconds of light removal.  相似文献
