A novel two-step fermentation process was developed to enhance arachidonic acid (ARA) production by Mortierella alpina ME-1 in a 5 l fermentor. Agitation speed and aeration rate were adjusted from 180 to 40 rpm and from 0.6 to1 vvm, respectively, after 5 days cultivation, to decrease physical damage to the mycelia and to extend the stationary phase. Moreover, 3% (w/v) and 2% (w/v) ethanol were fed after 5 and 7 days cultivation, respectively, to enhance ARA content of total lipid. Eventually, an ARA yield of 19.8 g/l was achieved, which was 1.7 times higher than that of a one-step fed-batch cultivation.  相似文献

Traditional mycelium aging technology was improved to enhance arachidonic acid (ARA) production by Mortierella alpina ME-1. Filtration step was skipped and additional carbon and nitrogen sources were fed during aging. The levels of the significant factors (time, temperature, ethanol, and KNO3) affecting ARA production during improved aging process were also optimized by applying response surface methodology (RSM), and the maximum ARA yield of 19.02 g/l was achieved in a 5 l fermentor at 5.6 days, temperature 13.7 °C, ethanol 42.44 g/l, and KNO3 2.62 g/l. This yield was 1.55 times higher than that of traditional aging technology. The improved mycelium aging technology is considered to be a useful strategy for enhancing ARA production.  相似文献

Effective production of arachidonic acid (ARA) using Mortierella alpina was conducted in a 30-L airlift bioreactor. Varying the aeration rate and temperature significantly influenced cell morphology, cell growth, and ARA production, while the optimal aeration rate and temperature for cell growth and product formation were quite different. As a result, a two-stage aeration rate control strategy was constructed based on monitoring of cell morphology and ARA production under various aeration rate control levels (0.6–1.8 vvm). Using this strategy, ARA yield reached 4.7 g/L, an increase of 38.2% compared with the control (constant aeration rate control at 1.0 vvm). Dynamic temperature-control strategy was implemented based on the fermentation performance at various temperatures (13–28°C), with ARA level in total cellular lipid increased by 37.1% comparing to a constant-temperature control (25°C). On that basis, the combinatorial fermentation strategy of two-stage aeration rate control and dynamic temperature control was applied and ARA production achieved the highest level of 5.8 g/L.  相似文献

A growth-inhibiting, aspirin-containing medium was developed to select arachidonic-acid-(ARA)-producing Mortierella species and to determine the fatty-acid content of 87 Mortierella strains. ARA was detected in 66 strains from 33 species and its production may prove useful for systematic studies on Mortierella spp. The ARA content of the 66 producing strains tested ranged from 4% to 55% of total lipids. Most of the ARA-producing strains of Mortierella (59 strains) grown on potato/glucose/agar synthesized <20% ARA. Four strains produced between 20% and 25% ARA and three produced >40%. An inverse relationship was observed between ARA and oleic-acid contents.Names used in this article are necessary to report factually on available data; however, the USDA neither guarantees nor warrants the standard of the product, and the use of the name by USDA implies no approval of the product to the exclusion of others that may also be suitable.  相似文献

Penicillium citrinum produced a glycolipid with emulsifier properties during cultivation on mineral medium with 1% (v/v) olive oil as carbon source. The emulsifier production was growth-associated and reached maximal activity at 60 h of cultivation. The production yield (Y p/s) was 0.54 and the best emulsifying activity was observed for xylene and diesel oil when compared to other carbohydrates tested. The emulsifier was shown to be stable to a wide range of pH and temperature values and was shown to contain D-galactose, D-glucose and D-xylose (8.2:1.0:5.3) with a total carbohydrate content of 43%. The presence of salts stimulated the emulsification activity, suggesting potential for its application in industrial waste or marine remediation.  相似文献

The production of water-soluble single-sugar glucuronic acid-based oligosaccharides (WSOS) by a cellulose producing strain Gluconacetobacter hansenii PJK was studied in a periodically recycled and fed-batch cultivations using glucose/ethanol or glucose only. Fermentations were carried out in a 2 L jar fermenter equipped with a turbine impeller with 6 flat blades. WSOS were produced constantly but the bacterial cellulose (BC) production stopped at 48 h of cultivation in a periodically recycled culture using the exhausted medium supplemented with glucose and ethanol. Tremendous quantities of WSOS were obtained in fed-batch cultivations using glucose/ethanol (35.6 g/L at 132 h of cultivation) or glucose only (86 g/L after 240 h of cultivation) as the nutritional source. However, the BC production yield under these nutritional conditions decreased significantly in comparison to previous studies about the BC production by the same strain. The overall results revealed that G. hansenii is capable of producing enormous quantities of WSOS compared to those reported previously for compounds of a related chemical nature. Moreover, the WSOS production was found to be dependent on the pH of the culture broth.  相似文献

The purpose of this study was to optimize the solid state cultivation ofMonascus ruber on sterile rice. A single-level-multiple-factor and a single-factor-multiple-level experimental design were employed to determine the optimal medium constituents and to optimize carbon and nitrogen source concentrations for lovastatin production. Simultaneous quantitative analyses of the β-hydroxyacid form and β-hydroxylactone for of lovastatin were performed by the high performance liquid chromatography (HPLC) method with a UV photodiode-array (PDA) detector. The total lovastatin yield (4≈6 mg/g, average of five repeats) was achieved by adding soybean powder, glycerol, sodium nitrate, and acetic acid at optimal composition of the medium increased by almost 2 times the yield observed prior to optimization. The experimental results also indicated that the β-hydroxylactone form of lovastatin (LFL) and the β-hydroxyacid form of lovastatin (AFL) simultaneously existed in solid state cultures ofMonascus ruber, while the latter was the dominant form in the middle-late stage of continued fermentation. These results indicate that optimized culture conditions can be used for industrial production of lovastatin to obtain high yields.  相似文献

Arachidonic acid (ARA, 5,8,1l,14-cis-eicosatetraenoic acid) is widely used in medicine, pharmaceutics, cosmetics, dietary nutrition, agriculture, and other fields. Microbiological production of ARA is of increased interest since the natural sources (pig liver, adrenal glands, and egg-yolk) cannot satisfy its growing requirements. Mechanisms for ARA biosynthesis as well as the regulation of enzymes involved in this process are considered. Review summarizes literature data concerning individual stages of microbiological ARA production, methods for screening of active strains-producers, physiological regulation of ARA synthesis in micromycetes (the effect of growth phase, medium composition, pH, temperature, and aeration), and effective technologies of fermentation and the product recovery. Information on the whole biotechnological process from strain selection to the ARA yield improvement and purification of the end product is presented.  相似文献

Mortierella alpina LPM 301, a producer of arachidonic acid (ARA), was found to possess a unique property of intense lipid synthesis in the period of active mycelium growth. Under batch cultivation of this strain in glucose-containing media with potassium nitrate or urea, the bulk of lipids (28–35% of dry biomass) was produced at the end of the exponential growth phase and remained almost unaltered in the stationary phase. The ARA content of lipids comprised 42–50% at the beginning of the stationary phase and increased continuously after glucose depletion in the medium due to the turnover of intracellular fatty acids; by the end of fermentation (189–210 h), the amount of ARA reached 46–60% of the total fatty acids (16–19% of dry mycelium). Plausible regulatory mechanisms of the growth-coupled lipid synthesis in microorganisms are discussed.  相似文献

The nitrogen-fixing grey alder,Alnus incana (L.) Moench, has a potential use in forest soil restoration and as part of energy forestry plantations. As a first step to estimate nitrogen fixation byA. incana under field conditions we performed studies on nitrogenase activity and its possible relation to abiotic factors and growth of the alders. Nitrogenase activity was measured as acetylene reduction activity (ARA) on eleven 1-year-old seedlings ofA. incana inoculated with a local source ofFrankia and planted in an experimental plot located in Umeå, northern Sweden. Each alder was planted into an open-ended cylinder which was closed with a gas tight lid around the stem base to serve as cuvette during ARA measurements. Propane served as tracer gas. ARA was measured in the middle of the day at 15 occasions during 26 June to 29 September 1987. Growth was recorded as leaf area and top shoot length at each ARA measurement until the end of August. Weather conditions were recorded for the whole growing season.Maximal ARA was recorded in late July or early August and ranged from 1.86 to 106mol C2H4plant–1h–1. Final leaf area ranged from 0.022 to 0.124 m2. A relationship between ARA and the number of hours of sunshine during the same day was observed. ARA in relation to soil temperature increased during the study period, except for the last measurements. ARA in relation to leaf area was initially high but decreased later on. It is suggested that as leaves got older their contribution to photosynthesis per unit leaf area decreased and their potential to deliver nitrogen for retranslocation within plant increased. Both of these events would cause reduced ARA per unit leaf area. The data on ARA, growth, and abiotic factors taken together supported the view that sunshine and weather conditions affected photosynthesis and thereby delivery of assimilates to the nodules.  相似文献

The production of an extracellular polysaccharide, EPS-R, from the marine bacteriumHahella chejuensis was investigated at various aeration rates in a batch culture. Higher aeration rate resulted in enhanced EPS production and increased the viscosity of the culture broth. At an aeration rate of 1.5 vvm, EPS-R (12.2 g/L) was obtained with a yield (Y P/S) of 0.6 from the STN medium after 72h of cultivation. TheH. chejuensis cells changed their rod morphology to a short-rod form in the stationary growth phase.  相似文献

Application of 0, 30, 60, 90 and 120 kg N ha–1 of urea (U) in split doses with (and without)Azolla pinnata, R. Brown was studied for three consecutive seasons under planted field condition. Fresh weight (FW), acetylene reduction activity (ARA) and N yield of Azolla were found to be maximum 14 days after inoculation (DAI). Among the different treatments, maximum Azolla growth was recorded in no N control. The FW, ARA and N yield of Azolla were inhibited increasingly with the increase in N levels. Irrespective of season, FW and N yield of Azolla were inhibited only a small extent with 90 kg N ha–1 U, beyond which the inhibition was pronounced. ARA was inhibited only slightly up to 60 kg N ha–1 of U. Grain yield and crop N uptake of rice increased significantly up to 90 kg N ha–1 of U (alone or in combination with Azolla) in the dry seasons (variety IR 36) and up to 60 kg N ha–1 U in the wet season (variety CR 1018).  相似文献

【背景】工厂化栽培中,杏鲍菇是不定点出菇,通常采用人工疏蕾的方法来实现对子实体数目的控制,但目前关于人工疏蕾保留子实体的数目无具体的研究。【目的】通过人工疏蕾的方式,研究保留不同子实体数目对杏鲍菇产量及品质的影响。【方法】对各处理组进行基质利用情况测定,对采收后各组子实体进行产量、形态指标及质构特性的测定。【结果】保留3—4个子实体的基质利用率较高,在生产实际中对成本的浪费较少;保留不同子实体数目对子实体形态指标、单菇重量及单包产量都有显著影响,保留1个子实体时,单菇重量最大但单包产量最低,保留4个子实体时,单包产量较高且子实体形态一致;从质构特性来看,保留子实体数目为4个时,杏鲍菇子实体品质最好、口感最佳。【结论】在杏鲍菇工厂化生产的人工疏蕾环节,保留子实体数目为3—4个时可以减少对成本的浪费,获得产量较高、品质较好的产品。  相似文献

The yellow-green alga Trachydiscus minutus (Eustigmatophyceae, Heterocontophyta) was cultivated in a standard medium and under nitrogen- and phosphorus-starvation and its triacylglycerols were analyzed by RP-HPLC/MS-APCI. The molecular species of triacylglycerols included a total of 74 triacylglycerols having at least one polyunsaturated fatty acid. Polyunsaturated triacylglycerols were identified for the first time in a yellow-green alga. N-starvation brought about a nearly 50% drop in TAGs containing EPA, and also decreased TAGs containing ARA, while P-starvation had a sizable effect on those TAGs that contain two or three arachidonic acids. In four TAGs containing PUFA, i.e. EEE, EEA, EAA and AAA, N-starvation caused a rapid fivefold increase in ARA content and the ratio of TAGs containing ARA, i.e. AEE to AAA increased tenfold relative to control. Regioisomeric characterization of triacylglycerols containing palmitic, arachidonic (ARA) and eicosapentaenoic acids (EPA) showed that the proportion of positional isomers is affected by N- and P-starvation. N- and P-starvation also changed the ratio of symmetrical to asymmetrical TAGs. Positional isomers exhibited identical ratios of symmetrical and asymmetrical TAGs irrespective of the type of FAs. In control cultivation the major TAGs with a single PUFA were symmetrical ones (PEP or PAP) whose ratio to asymmetrical counterparts (PPE or PPA) was about 3:1, whereas N- and P-starvation yielded opposite ratios, 1:3–1:5. The control cultivation yielded ∼90% asymmetrical TAGs with two PUFAs (i.e. PEE and PAA), whereas with N- and P-starvation the ratio of symmetrical to asymmetrical TAGs increased to 2:1 and 3:2, respectively.  相似文献

Since 1960s, modern techniques for nori (Porhyra) cultivation, artificial seeding of conchospores, floatingnet cultivation and freeze storage of nets, have been introduced into Japan, Korea and China. Owing to recent developments of these techniques, mechanization of processing and adaptation of fast-growing varieties of Porphyra, annual yield of nori have increased rapidly. Individual problems have, however, arisen in each country. In Japan, as the result of overproduction of nori, the farmers' profit level has, in general, decreased. As a consequence, the number of nori farmers, especially among those who would enter the industry, has declined markedly, and this is currently a serious problem. In Korea, there is a pressing need to improve the quality of nori, which is more important than increasing production. In China, the technical level of producing high quality Porphyra through cultivation and processing high quality nori is relatively undeveloped. There is thus a critical requirement to develop basic techniques in both cultivation and processing of nori in China.  相似文献

Summary The effect of reduced solar radiation on associative N2-fixation and plant parameters was studied in three field experiments (1978–80). Gahi-3 pearl millet (Pennisetum americanum (L.) K. Monch.) field plots were shaded with saran shade cloth that reduced solar radiation by 50% and 75%. Acetylene reduction activity (ARA) was reduced by shading in one of the three experiments. The two non-responding experiments were conducted on a wall-drained, low-activity site (ARA means ranging 17–68 n moles ethylene core–1 h–1), the responding experiment was conducted on a poorly drained, high-ARA site.Shading affected the plants drastically, reducing fresh weight and dry matter yields up to 46% (50% shade) and 57% (75% shade). Shading also reduced dry matter percentage from 19.6 (no shade) to 15.3 (75% shade) and increased nitrogen content from 0.6% (no shade) to 1.53% (75% shading). However, shading did not affect protein yield. Inoculation withAzospirillum brasilense had no measurable effect on yield or acetylene reduction in the first two experiments.In the third experiment, shading reduced mean ARA of inoculated plots over 100% but had no significant effect on control plots. Inoculation significantly increased ARA in the nonshaded plots but not in shaded plots. Acetylene reduction activity was high, with means ranging between 208 and 465 n moles ethylene evolved core–1 h–1. Soil moisture and millet growth stage also affected acetylene reduction activity.  相似文献

【背景】丝状真菌是一类重要的工业发酵生产宿主菌,如何进行高通量纯菌培养和高效检测筛选性能优异的菌株是工业丝状真菌研究的重要方向。【目的】研究建立丝状真菌的高通量培养技术并测试应用效果。【方法】通过对丝状真菌培养过程中的制种、接种、培养和检测研究,建立基于孔板的高通量培养技术,并以嗜热毁丝霉为例对该技术进行验证。【结果】与传统的平板制种和摇瓶接种培养方式相比,高通量孔板的培养方式将制种通量提高24倍,单位面积产孢子能力提高350%,液体培养转接效率提高10-40倍,并建立96孔板测定乙醇含量的高通量检测技术。【结论】将丝状真菌的培养和检测通量提高1-2个数量级,为快速检测丝状真菌改造过程产生的大量性状不同菌株并获得目标菌株奠定基础,为丝状真菌高通量筛选研究提供应用指导价值。  相似文献

Abstract

Fungal arachidonic acid (ARA)-rich oil is an important microbial oil that affects diverse physiological processes that impact normal health and chronic disease. In this article, the historic developments and technological achievements in fungal ARA-rich oil production in the past several years are reviewed. The biochemistry of ARA, ARA-rich oil synthesis and the accumulation mechanism are first introduced. Subsequently, the fermentation and downstream technologies are summarized. Furthermore, progress in the industrial production of ARA-rich oil is discussed. Finally, guidelines for future studies of fungal ARA-rich oil production are proposed in light of the current progress, challenges and trends in the field.  相似文献
