The changes in the activity of antioxidant enzymes such as superoxide dismutase (SOD: EC 1.15.1.1), catalase (CAT: EC 1.11.1.6), peroxidase (POX: EC 1.11.1.7), ascorbate peroxidase (APOX: EC 1.11.1.11) and glutathione reductase (GR: EC 1.6.4.2), free proline content, and the rate of lipid peroxidation level in terms of malondialdehyde (MDA) in roots of two rice cultivars (cvs.) differing in salt tolerance were investigated. Plants were subjected to three salt treatments, 0, 60, and 120 mol m−3 NaCl for 7 days. The results showed that activated oxygen species may play a role in cellular toxicity of NaCl and indicated differences in activation of antioxidant defense systems between the two cvs. The roots of both cultivars showed a decrease in GR activity with increase in salinity. CAT and APOX activities increased with increasing salt stress in roots of salt-tolerant cultivar Pokkali but decreased and showed no change, respectively, in roots of IR-28 cultivar. POX activity decreased with increasing NaCl concentrations in salt-tolerant Pokkali but increased in IR-28. SOD activity showed no change in roots of both cultivars under increasing salinity. MDA level in the roots increased under salt stress in sensitive IR-28 but showed no change in Pokkali. IR-28 produced higher amount of proline under salt stress than in Pokkali. Increasing NaCl concentration caused a reduction in root fresh weight of Pokkali and root dry weight of IR-28. The results indicate that improved tolerance to salt stress in root tissues of rice plants may be accomplished by increased capacity of antioxidative system.  相似文献

We have isolated and characterized a putative rice MAPK gene (designated OsMAPK44) encoding for a protein of 593 amino acids that has the MAPK family signature and phosphorylation activation motif, TDY. Alignment of the predicted amino acid sequences of OsMAPK44 showed high homology with other rice MAPKs. Under normal conditions, the OsMAPK44 gene is highly expressed in root tissues, but relatively less in leaf and stem tissues of the japonica type rice plant (O. sativa L. Donggin). mRNA expression of the gene is highly inducible by salt and drought treatment, but not by cold treatment. Moreover, the mRNA level of the OsMAPK44 is up-regulated by exogenously applied Abscisic acid (ABA) and H2O2. When we compared the OsMAPK44 gene expression level between a salt sensitive indica cultivar (IR64) and a salt resistant indica cultivar (Pokkali), they showed some difference in expression kinetics with the salt treatment. OsMAPK44 gene expression in Pokkali was slightly up-regulated within 30 min and then disappeared rapidly, while IR64 maintained its expression for 1 h following down-regulation. Under the salinity stress, OsMAPK44 overexpression transgenic rice plants showed less damage and greater ratio of potassium and sodium than OsMAPK44 suppressed transgenic lines did, suggesting that OsMAPK44 may have a role to prevent damages due to working for favorable ion balance in the presence of salinity.  相似文献

Water deficit for rice is a worldwide concern, and to produce drought-tolerant varieties, it is essential to elucidate molecular mechanisms associated with water deficit tolerance. In the present study, we investigated the differential responses of nonenzymatic antioxidants ascorbate (AsA), glutathione (GSH), and their redox pool as well as activity levels of enzymes of ascorbate–glutathione cycle in seedlings of drought-sensitive rice (Oryza sativa L.) cv. Malviya-36 and drought-tolerant cv. Brown Gora subjected to water deficit treatment of ?1.0 and ?2.1 MPa for 24–72 h using PEG-6000 in sand cultures. Water deficit caused increased production of reactive oxygen species such as O2??, H2O2, and HO? in the tissues, and the level of production was higher in the sensitive than the tolerant cultivar. Water deficit caused reduction in AsA and GSH and decline in their redox ratios (AsA/DHA and GSH/GSSG) with lesser decline in tolerant than the sensitive seedlings. With progressive level of water deficit, the activities of monodehydroascorbate reductase, dehydroascorbate reductase, ascorbate peroxidase (APX), and glutathione transferase increased in the seedlings of both rice cultivars, but the increased activity levels were higher in the seedlings of drought-tolerant cv. Brown Gora compared to the sensitive cv. Malviya-36. Greater accumulation of proline was observed in stressed seedlings of tolerant than the sensitive cultivar. In-gel activity staining of APX revealed varying numbers of their isoforms and their differential expression in sensitive and tolerant seedlings under water deficit. Results suggest that an enhanced oxidative stress tolerance by a well-coordinated cellular redox state of ascorbate and glutathione in reduced forms and induction of antioxidant defense system by elevated activity levels of enzymes of ascorbate–glutathione cycle is associated with water deficit tolerance in rice.  相似文献

The potential involvement of activated oxygen species by submergence stress was studied in two Malaysian rice cultivars, MR219-4 and MR219-9, and cultivar FR13A that is known to be tolerant to submergence. Seedlings of these three rice cultivars were subjected to different submergence periods (4, 8, and 12 days). Under 8 days of complete submergence, FR13A cultivar showed higher lipid peroxidation in terms of malondialdehyde level and activities of antioxidative enzymes, superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), and glutathione reductase (GR) when compared to the MR219-4 and MR219-9 cultivars. MR219-9 showed higher SOD, APX, and GR activities after 12 days of submergence. The levels of SOD activity indicated that detoxification of O2·− to H2O2 was maintained at a stable level throughout the submergence stress until up to 8 days and increased rapidly at 12 days of submergence. The results indicated that tolerance to submergence in rice is associated until 8 days submergence for MR219-4 and FR13A cultivars. These findings suggested that tolerance to submergence stress in rice might be proven by increased the capacity of antioxidative system. In addition, CAT activity has much higher affinity for scavenges H2O2 than APX. Therefore, ascorbate glutathione cycle might be more efficient to scavenge H2O2.  相似文献

Salt-tolerance was studied in transgenic potato. It was conferred by overexpression of ascorbate pathway enzyme (d-galacturonic acid reductase, GalUR). As genetic engineering of the GalUR gene in potato enhances its ascorbic acid content (l-AsA), and subsequently plants suffered minimal oxidative stress-induced damage, we now report on the comprehensive aptness of this engineering approach for enhanced salt tolerance in transgenic potato (Solanum tuberosum L. cv. Taedong Valley). Potatoes overexpressing GalUR grew and tuberized in continuous presence of 200 mM of NaCl. The transgenic plants maintained a higher reduced to oxidized glutathione (GSH:GSSG) ratio together with enhanced activity of glutathione dependent antioxidative and glyoxalase enzymes under salinity stress. The transgenics resisted an increase in methylglyoxal that increased radically in untransformed control plants under salinity stress. This is the first report of genetic engineering of ascorbate pathway gene in maintaining higher level of GSH homeostasis along with higher glyoxalase activity inhibiting the accumulation in methylglyoxal (a potent cytotoxic compound) under salt stress. These results suggested the engineering of ascorbate pathway enzymes as a major step towards developing salinity tolerant crop plants.  相似文献

The present investigation evaluated the ability of an antioxidative defense system in terms of the tolerance against salinity-induced oxidative stress and also explored a possible relationship between the status of the components of an antioxidative defense system and the salt tolerance in Indica rice (Oryza sativa L.) genotypes. When the seedlings of a salt-sensitive cultivar was grown in sand cultures containing different NaCl concentrations (7 and 14 dS m?1) for 5–20 days, a substantial increase was observed in the rate of superoxide anion (O 2 ·? ) production, elevated levels of H2O2 and thiobarbituric acid reactive substances (TBARS) which indicated an enhancement in lipid peroxidation. A declination in the level of thiol clearly indicated an increase in the protein oxidation as well as a decline in the reduced forms of ascorbate (AsA) and glutathione (GSH) and the ratios of their reduced to oxidized forms occurred in the salt-sensitive seedlings. Similar treatment caused a very little alteration or no change in the levels of these components in the seedlings of salt-tolerant cultivar. The activity of antioxidative enzymes superoxide dismutase (SOD), its isoform Cu/Zn-SOD and ascorbate peroxidase (APX) increased in both the cultivars against salinity. In salt-sensitive seedlings, the activity of the various enzymes, guaiacol peroxidase (GPX), catalase (CAT), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR), and glutathione reductase (GR) increased at moderate salinity treatment of 7 dS m?1 NaCl while the activities of these enzymes declined with higher salinity level of 14 dS m?1 NaCl. However, a consistent increase was observed in the activities of these enzymes of salt-tolerant seedlings with an increase in the duration and the level of the salinity treatment. The results suggest that a higher status of antioxidants (AsA and GSH) and a coordinated higher activity of the enzymes (SOD, CAT, GPX, APX, and GR) can serve as the major determinants in the model for depicting salt tolerance in Indica rice seedlings.  相似文献

The research was conducted to investigate comparative oxidative damage including probable protective roles of antioxidant and glyoxalase systems in rice (Oryza sativa L.) seedlings under salinity stress. Seedlings of two rice genotypes: Pokkali (tolerant) and BRRI dhan28 (sensitive) were subjected to 8 dSm−1 salinity stress for seven days in a hydroponic system. We observed significant variation between Pokkali and BRRI dhan28 in phenotypic, biochemical and molecular level under salinity stress. Carotenoid content, ion homeostasis, antioxidant enzymes, ascorbate and glutathione redox system and proline accumulation may help Pokkali to develop defense system during salinity stress. However, the activity antioxidant enzymes particularly superoxide dismutase (SOD), catalase (CAT) and non-chloroplastic peroxidase (POD) were observed significantly higher in Pokkali compared to salt-sensitive BRRI dhan28. Higher glyoxalase (Gly-I) and glyoxalase (Gly-II) activity might have also accompanied Pokkali genotype to reduce potential cytotoxic MG through non-toxic hydroxy acids conversion. However, the efficient antioxidants and glyoxalase system together increased adaptability in Pokkali during salinity stress.  相似文献

Salinity stress is a major limiting factor in cereal productivity. Many studies report improvements in salt tolerance using model plants, such as Arabidopsis thaliana or standard varieties of rice, e.g., the japonica rice cultivar Nipponbare. However, there are few reports on the enhancement of salt tolerance in local rice cultivars. In this work, we used the indica rice (Oryza sativa) cultivar BR5, which is a local cultivar in Bangladesh. To improve salt tolerance in BR5, we introduced the Escherichia coli catalase gene, katE. We integrated the katE gene into BR5 plants using an Agrobacterium tumefaciens-mediated method. The introduced katE gene was actively expressed in the transgenic BR5 rice plants, and catalase activity in T1 and T2 transgenic rice was approximately 150% higher than in nontransgenic plants. Under NaCl stress conditions, the transgenic rice plants exhibited high tolerance compared with nontransgenic rice plants. T2 transgenic plants survived in a 200 mM NaCl solution for 2 weeks, whereas nontransgenic plants were scorched after 4 days soaking in the same NaCl solution. Our results indicate that the katE gene can confer salt tolerance to BR5 rice plants. Enhancement of salt tolerance in a local rice cultivar, such as BR5, will provide a powerful and useful tool for overcoming food shortage problems.  相似文献

To keep pace with ever growing global population, progressive and sustained increase in rice production is necessary, especially in areas with extremely variable climatic conditions, where rice crop suffers from numerous abiotic stresses including salinity. Designing an effective phenotyping strategy requires thorough understanding of plant survival under stress. The investigation was carried out with four rice cultivars namely FR13A, IR42, Rashpanjor, and Pokkali that differed in salinity tolerance. The study showed that a genotype with initial vigour had some advantage in preserving shoot biomass under salt stress. Though both FR13A and IR42 showed sensitivity to salinity, FR13A with higher initial biomass maintained greater dry weight under saline condition. Increase of Na+:K+ ratio under salinity, due to accelerated absorption of Na+ and lesser absorption of K+ compared to control, was considerably higher in susceptible (118–200 %) than in tolerant (33–48 %) genotypes. While Na+ concentration in shoot increased significantly in both tolerant and susceptible genotypes, decrease in shoot K+ content was noticed only in susceptible genotypes. The imbalance of Na+ and K+ contents led to increased H2O2 production, causing greater peroxidation of membrane lipids and reduction in chlorophyll content and CO2 photosynthetic rate. Certain chlorophyll fluorescence parameters could distinguish between salinity tolerant and sensitive genotypes. To protect the plant from oxidative damage, several enzymatic and nonenzymatic antioxidants such as ascorbate were involved. The genotypes with capacity to assemble antioxidant enzymes in time could detoxify the reactive oxygen species more efficiently, leading to greater protection and reduced impact of salt stress.  相似文献

The effect of Al stress on H2O2 production of rice (Oryza sativa L.) seedlings and difference in responses of antioxidant enzymes between Al-tolerant variety (Azucena) and Al-sensitive rice one (IR 64) were investigated. Aluminum-induced H2O2 production and malondialdehyde (MDA) content were more pronounced for IR 64 than for Azucena. In the presence of 2 mM Al, addition of 10 mM imidazole (inhibitor of NADPH oxidase) and 1 mM azide (inhibitor of peroxidase) significantly decreased H2O2 production by 16% and 43% for Azucena, and 21% and 68% for IR 64, respectively. Under Al treatment, the Al-tolerant variety Azucena had significantly higher activities of catalase, ascorbate peroxidase, dehydroascorbate reducase, glutathione peroxidase and glutathione reductase, and higher concentrations of reduced glutathione than the Al-sensitive one IR 64. Treatment with buthionine sulfoximine, a specific inhibitor of GSH synthesis, significantly increased H2O2 production in both varieties in the presence and absence of Al. In contrast, the treatment with GSH significantly decreased the production of H2O2 induced by Al stress. Results suggest that GSH may play an important role in scavenging H2O2 caused by Al stress.  相似文献

Characteristics of chlorophyll fluorescence and antioxidative system were investigated in rice (Oryza sativa L.) super-hybrid Liangyoupeijiu (LYPJ), maternal cultivar Peiai64s, and paternal cultivar indica rice 9311 under chilling stress. During 6-d chilling treatment, chlorophyll content of all three genotypes was gradually declined. However, the decrease in photosystem 2 (PS 2) maximum photochemical efficiency (Fv/Fm) and quantum yield of PS 2 (ΦPS2) was less expressive in LYPJ than in parental cultivars The electrolyte leakage and malondialdehyde content in all cultivars increased after chilling treatment, but LYPJ exhibited the least increasing tendency. Activities of superoxide dismutase (SOD), ascorbate peroxidase (APX) and glutathione reductase (GR) were higher in LYPJ than in parental cultivars. The results demonstrated that tolerance to chilling stress in LYPJ might be adopted mostly from its maternal cultivar.  相似文献

The effect of exogenously applied glycinebetaine (GB) on the alleviation of damaging effects of NaCl treatment was studied in view of relative water content (RWC), malondialdehyde content, and the activity of some antioxidant enzymes in two rice (Oryza sativa L.) cultivars differing in salt tolerance (salt-tolerant Pokkali and--sensitive IR-28), comparatively. Both cultivars took up exogenously applied GB through their roots and accumulated it to considerable levels. Leaf RWC of both cultivars under salt treatment showed an increase with GB application. The activities of superoxide dismutase (SOD), ascorbate peroxidase (AP), catalase (CAT), and glutathione reductase (GR) increased in leaves of Pokkali, but peroxidase (POX) activity decreased under salinity. In IR-28, the activities of SOD, AP and POX increased, whereas CAT and GR decreased upon exposure to salt treatment. When compared to the salt-treated group alone, GB application decreased the activities of SOD, AP, CAT, and GR in Pokkali, whereas it increased the activities of CAT and AP in IR-28 under salinity. However, the activity of POX in IR-28 under salinity showed a decrease with GB application compared to the NaCl group. In addition, lipid peroxidation levels of both cvs. under salt treatment showed a decrease with GB treatment. Therefore, we conclude that GB protects both rice seedlings from salinity-induced oxidative stress.  相似文献

Crop yield is severely affected by soil salinity, as salt levels that are harmful to plant growth occur in large terrestrial areas of the world. The present investigation describes the studies of enzymatic activities, in-gel assays, gene expression of some of the major antioxidative enzymes, tocopherol accumulation, lipid peroxidation, ascorbate and dehydroascorbate contents in a salt-sensitive rice genotype PB1, and a relatively salt-tolerant cultivar CSR10 in response to 200 mM NaCl. Salt solution was added to the roots of hydroponically grown 5-day-old etiolated rice seedlings, 12 h prior to transfer to cool white fluorescent?+?incandescent light (100 μmol photons m?2 s?1). Total tocopherol and ascorbate contents declined in salt-stressed rice seedlings. Among antioxidative enzymes, an increase in the activities of superoxide dismutase (EC 1.15.1.1), catalase (EC 1.11.1.6), ascorbate peroxidase (EC 1.11.1.11), glutathione reductase (EC 1.6.4.2), and their gene expression was observed in both cultivars in response to salt stress. The salt-tolerant cultivar CSR10 resisted stress due to its early preparedness to combat oxidative stress via upregulation of gene expression and enzymatic activities of antioxidative enzymes and a higher redox status of the antioxidant ascorbate even in a non-stressed environment.  相似文献

根据营养液培养试验从水稻Azucena×IR64 发展的一双单倍体(DH) 群体中筛选出抗铁毒与敏感品系。在铁毒害处理后,各品系的抗坏血酸过氧化物酶(AP) 、脱氢抗坏血酸还原酶(DR) 、谷胱甘肽还原酶(GR) 的活性均有明显提高;水稻受铁毒后的生物量递减量与AP、DR、GR 活性呈负相关。说明抗坏血酸过氧化物酶H2O2 清除系统在水稻抗铁毒中起着十分重要的作用。  相似文献

Salinity poses a major threat for agriculture worldwide. Rice is one of the major crops where most of the high-yielding cultivars are highly sensitive to salinity. Several studies on the genetic variability across rice cultivars suggest that the activity and composition of root plasma membrane transporters could underlie the observed cultivar-specific salinity tolerance in rice. In the current study, it was found that the salt-tolerant cultivar Pokkali maintains a higher K+/Na+ ratio compared with the salt-sensitive IR20 in roots as well as in shoots. Using Na+ reporter dyes, IR20 root protoplasts showed a much faster Na+ accumulation than Pokkali protoplasts. Membrane potential measurements showed that root cells exposed to Na+ in IR20 depolarized considerably further than those of Pokkali. These results suggest that IR20 has a larger plasma membrane Na+ conductance. To assess whether this could be due to different ion channel properties, root protoplasts from both Pokkali and IR20 rice cultivars were patch-clamped. Voltage-dependent K+ inward rectifiers, K+ outward rectifiers, and voltage-independent, non-selective channels with unitary conductances of around 35, 40, and 10 pS, respectively, were identified. Only the non-selective channel showed significant Na+ permeability. Intriguingly, in both cultivars, the activity of the K+ inward rectifier was drastically down-regulated after plant growth in salt but gating, conductance, and activity of all channel types were very similar for the two cultivars.  相似文献

Nitric oxide (NO) has emerged as a key molecule involved in many physiological events in plants. To characterize roles of NO in tolerance of tomato (Lycopersicom esculentum Mill.) to salt stress, the protective effects of NO against salt-induced oxidative stress in the leaves of tomato cultivar Hufan1480 (salt-tolerant) and Hufan2496 (salt-sensitive) were evaluated. Under salt stress, Hufan1480 showed higher biomass accumulation, and less oxidative damage when compared with the Hufan2496. Application of exogenous sodium nitroprusside, a NO donor, dramatically alleviated growth suppression induced by salt stress in two tomato ecotypes, reflected by decreased malondialdehyde and O2·− production. Furthermore, the antioxidant enzymes superoxide dismutase, guaiacol peroxidase, catalase and ascorbate peroxidase, the antioxidant metabolites ascorbate and reduced glutathione, and the osmosis molecules proline and soluble sugar were increased in both ecotypes in the presence of NO under salt stress. Therefore, the protective effect of NO against salt-induced oxidative damages in tomato seedlings is most likely mediated through stimulation of antioxidant system.  相似文献

The present study investigates the possible regulatory role of exogenous nitric oxide (NO) in antioxidant defense and methylglyoxal (MG) detoxification systems of wheat seedlings exposed to salt stress (150 and 300 mM NaCl, 4 days). Seedlings were pre-treated for 24 h with 1 mM sodium nitroprusside, a NO donor, and then subjected to salt stress. The ascorbate (AsA) content decreased significantly with increased salt stress. The amount of reduced glutathione (GSH) and glutathione disulfide (GSSG) and the GSH/GSSG ratio increased with an increase in the level of salt stress. The glutathione S-transferase (GST) activity increased significantly with severe salt stress (300 mM). The ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR), catalase (CAT) and glutathione peroxidase (GPX) activities did not show significant changes in response to salt stress. The glutathione reductase (GR), glyoxalase I (Gly I), and glyoxalase II (Gly II) activities decreased upon the imposition of salt stress, especially at 300 mM NaCl, with a concomitant increase in the H2O2 and lipid peroxidation levels. Exogenous NO pre-treatment of the seedlings had little influence on the non-enzymatic and enzymatic components compared to the seedlings of the untreated control. Further investigation revealed that NO pre-treatment had a synergistic effect; that is, the pre-treatment increased the AsA and GSH content and the GSH/GSSG ratio, as well as the activities of MDHAR, DHAR, GR, GST, GPX, Gly I, and Gly II in most of the seedlings subjected to salt stress. These results suggest that the exogenous application of NO rendered the plants more tolerant to salinity-induced oxidative damage by enhancing their antioxidant defense and MG detoxification systems.  相似文献

Rice being an important cereal crop is highly sensitive to salinity stress causing growth retardation and loss in productivity. However, certain rice genotypes like Nonabokra and Pokkali show a high level of tolerance towards salinity stress compared to IR64 variety. This differential response of tolerant varieties towards salinity stress may be a cumulative effect of genetic and epigenetic factors. In this study, we have compared the salinity-induced changes in chromatin modifications at the OsBZ8 locus in salt-tolerant Nonabokra and salt-sensitive IR64 rice varieties. Expression analysis indicates that the OsBZ8 gene is highly induced in Nonabokra plants even in the absence of salt stress, whereas in IR64, the expression significantly increases only during salt stress. Sequence analysis and nucleosomal arrangement within the region ?2000 to +1000 of OsBZ8 gene show no difference between the two rice varieties. However, there was a considerable difference in histone modifications and DNA methylation at the locus between these varieties. In Nonabokra, the upstream region was hyperacetylated at H3K9 and H3K27, and this acetylation did not change during salt stress. However, in IR64, histone acetylation was observed only during salt stress. Moreover, the upstream region of OsBZ8 gene has highly dynamic nucleosome arrangement in Nonabokra, compared to IR64. Furthermore, loss of DNA methylation was observed at OsBZ8 locus in Nonabokra control plants along with low H3K27me3 and high H3K4me3. Control IR64 plants show high DNA methylation and enriched H3K27me3. Collectively these results indicate a significant difference in chromatin modifications between the rice varieties that regulates differential expression of OsBZ8 gene during salt stress.  相似文献

The effects of aluminum (Al) on root elongation, lipid peroxidation, hydrogen peroxide (H2O2) accumulation, antioxidant levels, antioxidant enzymatic activity, and lignin content in the roots of the Al-tolerant rice variety azucena and the Al-sensitive variety IR64 were investigated. Treatment with Al induced a greater decrease in root elongation and a greater increase in H2O2 and lipid peroxidation as determined by the total thiobarbituric acid-reactive substance (TBARS) level in IR64 than in azucena. Azucena had significantly higher levels of superoxide dismutase, ascorbate peroxidase, glutathione reductase, and glutathione peroxidase GSH POD activity compared with IR64. The concentrations of reduced glutathione (GSH) and ascorbic acid, and the GSH/GSSG ratio (reduced vs. oxidized glutathione) were also higher in azucena than in IR64 in the presence of Al. The addition of 1 mg/L GSH improved root elongation in both varieties and decreased H2O2 production under Al stress. By contrast, treatment with buthionine sulfoximine, a specific inhibitor of GSH synthesis, decreased root elongation in azucena and stimulated H2O2 production in both varieties. Moreover, Al treatment significantly increased the cytoplasmic activity of peroxidase (POD) as well as the levels of POD bound ionically and covalently to cell walls in the Al-sensitive variety. The lignin content was also increased. Treatment with exogenous H2O2 also increased the lignin content and decreased root elongation in IR64. Imbibitional heat and chilling stress caused disruption of redox-homeostasis and oxidative damage to newly assembled membrane system by aggravating membrane lipid peroxidation and protein oxidation [measured in terms of thiobarbituric acid reactive substances (TBARS), free carbonyl content (C=O groups) and membrane protein thiol level (MPTL)] along with concomitant increase in accumulation of reactive oxygen species (superoxide and hydrogen peroxide) and significant reduction of antioxidative defense (assessed in terms of total thiol content and activities of superoxide dismutase, catalase, ascorbate peroxidase and glutathione reductase) in both the salt sensitive (Ratna) and resistant (SR 26B) germinating tissues of rice cultivars. When compared, salt resistant cultivar SR 26B found to suffer significantly less oxidative membrane damage as compared to salt sensitive cultivar Ratna. Treatment with low titer of hydrogen peroxide caused significant reversal in oxidative damages to the newly assembled membrane system imposed by imbibitional heat and chilling stress (evident from the data of TBARS, C=O, MPTL, ROS accumulation, membrane permeability status, membrane injury index and oxidative stress index) in seedlings of both the cultivars of rice (Ratna and SR 26B). Imbibitional H2O2 pretreatment also caused up-regulation of antioxidative defense (activities of superoxide dismutase, catalase, ascorbate peroxidase, glutathione reductase and total thiol content) in the heat and chilling stress-raised seedlings of experimental rice cultivars. When the parameters of early growth performances were assessed (in terms of relative growth index, biomass accumulation and vigor index), it clearly exhibited significant improvement of early growth performances of both the rice cultivars. Better response towards H2O2-mediated acclamatory performances and restoration of redox- homeostasis under extremes of temperature were noticed in salt sensitive rice cultivar Ratna compared to salt resistant SR 26B. Taken as a whole, the result suggests the significance of the role of ‘inductive pulse’ of H2O2 in acclimatizing adverse temperature stress by restoration of redox-homeostasis and mitigation of oxidative membrane protein and lipid damages during the recovery phase of post-germination event.  相似文献
