1. The conversion of cholest-5-en-3beta-ol (cholesterol) into cholesta-5,7-dien-3beta-ol by axenic Calliphora erythrocephala larvae was demonstrated. 2. The transformation is probably direct (Delta(5)-->Delta(5,7)) and does not involve a Delta(0) intermediate (Delta(5)-->Delta(0)-->Delta(7)--> Delta(5,7)). 3. Delta(7)-bond formation involves the stereospecific elimination of the 7beta hydrogen atom. 4. The relative amounts of free and esterified sterols were determined in larvae grown on cholesterol as sole sterol source and on 5alpha-cholestan-3beta-ol supplemented with minimal amounts of cholesterol. 5. The significance of the results is assessed in relation to the probable role of cholesta-5,7-dien-3beta-ol as an intermediate in the biosynthesis of ecdysones.  相似文献

The conversion of beta-carotene into retinal was studied in vitro with enzyme preparations from homogenates of hog intestinal mucosa. The hog mucosal enzyme was purified about 27-fold by precipitation with ammonium sulphate, chromatography on DEAE-Sephadex and gel filtration on Sephadex G-200. The reaction displayed a narrow optimum pH range (approx. 7.8-8.2). The enzyme was stimulated strongly by the addition of thiols, and was inhibited by thiol inhibitors and by the chelating agents alphaalpha'-bipyridyl and o-phenanthroline. The reaction required the addition of an appropriate detergent (or bile salt); maximal activity was obtained by addition of an appropriate combination of detergents and lipid (specifically Tween 40, sodium glycocholate and sphingomyelin). The reaction displayed Michaelis kinetics with K(m)1.3x10(-6)m and V(max.)1.1nmole of retinal formed/hr. (for 0.7mg. of enzyme protein). The properties of the hog enzyme are similar to those previously reported for a less purified rat enzyme preparation.  相似文献

Labelled 10α-cucurbita-5,24-dien-3β-ol, the simplest tetracyclic triterpene with a cucurbitane skeleton, was transformed into cucurbitacin C in Cucumis sativus seedlings. This transformation has been previously postulated, but this is the first time it has been demonstrated to operate in plant tissues. Two other potential precursors of cucurbitacins, cycloartenol and parkeol, were incubated under the same conditions. Cycloartenol gave only the expected phytosterols whereas parkeol was recovered unchanged.  相似文献

1. The syntheses of Δ7-[4-14C]cholestenol (XVI, Scheme 3) and Δ7-[6α-3H]-cholestenol (XII, Scheme 2) are described. 2. The metabolism of doubly labelled Δ7-cholestenol (II, Scheme 1) by rat-liver homogenates was studied. 3. During the enzymic conversion of Δ7-cholestenol into cholesterol (IV, Scheme 1) the 6α-hydrogen atom of the former is lost and the overall reaction corresponds to a cis-elimination. 4. In the light of these results various mechanisms for the conversion of Δ7-cholestenol into cholesterol are discussed.  相似文献

Ergosta-5,23-dien-3β-ol and ergosta-7,23-dien-3β-ol were identified for the first time in maize etiolated coleoptiles. They represent more than 11 % of the total 4-desmethyl sterol fraction. It is suggested that they could play some role in the biosynthesis of 24-methyl sterols of this material.  相似文献

The photosensitized oxidation of cholest-4-en-3β-ol in which singlet molecular oxygen is implicated yielded cholest-4-en-3-one and the isomeric epoxides 4α,5-epoxy-5α-cholestan-3-one and 4β,5-epoxy-5β-cholestan-3-one, the epoxides being formed in the ratio 3 : 1. Oxidation of cholest-4-en-3-one by alkaline hydrogen peroxide likewise yielded the isomeric 4,5-epoxides but in the ratio 1 : 7.4. Attempted use of cholest-4-en-3β-ol to intercept singlet molecular oxygen putatively generated in the disproportionation of hydrogen peroxide gave a very complex product mixture of over 50 components from which only cholest-4-en-3-one could be identified. However, neither isomeric 4,5-epoxycholestan-3-one was detected among the products. These data establish that it is unwarranted to infer the action of single molecular oxygen in systems containing cholest-4-en-3β-ol merely by product analysis where the product 4α,5-epoxy-5α-cholestan-3-one is formed.  相似文献

Marked variations in the 3β-hydroxysterol content of hamster spermatozoa were observed as they progress through the epididymis. Cholesterol is the major sterol of caputal spermatozoa while the concentration of precursors of cholesterol was higher than that of cholesterol in caudal spermatozoa. One of these precursors has been identified as desmosterol. A second sterol has now been identified as 5α-cholesta-7, 24-dien-3β-ol by GLC-MS and by NMR. Its concentration is approximately 3-fold higher than that of cholesterol. This 3β-hydroxysterol is also found in epididymal tissue.  相似文献

1. A major component of the lipids in aqueous (pH7.5) homogenates of tuber tissue from Solanum tuberosum was isolated and characterized as 9-(nona-1',3'-dienoxy)non-8-enoic acid. 2. This novel unsaturated ether fatty acid derivative, which contains a butadienylvinyl ether function, has the structure: [Formula: see text] and is formed from linoleic acid by a sequence of enzymic reactions. 3. A precursor of the unsaturated ether derivative is 9-d-hydroperoxyoctadeca-10,12-dienoic acid, formed by the action of S. tuberosum lipoxygenase on linoleic acid. 4. An enzyme that converts the fatty acid hydroperoxide into the unsaturated ether derivative was isolated from S. tuberosum. The pH optimum of this enzyme is approx. 9, although the overall conversion of linoleic acid into the ether derivative is maximal at pH7.5. 5. An unusual feature of this pathway is the insertion of an oxygen atom into the alkyl chain of a fatty acid. 6. This novel mechanism may play a role in the breakdown of polyunsaturated fatty acids to volatile products in plants.  相似文献

Treatment of pregnant rats with AY-9944, a drug interfering with the last steps of cholesterol biosynthesis, accumulated cholesterol precursors in brain and liver of newborn animals. Different sterol profiles were found in these organs. Along with cholesterol and cholesta-5,7-dien-3β -ol, present in both tissues, liver was found to contain a hitherto unreported sterol, absent in brain. The structure of cholesta-5,8-dien-3β -ol was attributed to this compound by mass spectrometric, 1H, and 13C NMR analysis.  相似文献

Labelled 10α-cucurbita-5,24-dien-3β-ol was obtained from [3-3H]squalene-2,3-epoxide incubated with microsomes of Cucurbita maxima seedlings. By contrast the lanostane triterpenoids [2-3H]cycloartenol, [2-3H]parkeol and their corresponding derivatives [2,12-3H]11-ketocycloartenol and [2-3H]24,25-dihydro-90α,11a-epoxyparkeol, incubated under the same conditions, gave no rearranged products with a cucurbitane skeleton. These results suggest that biosynthesis of cucurbitane triterpenoids from squalene-2,3-epoxide occurs through direct cyclization without the intermediacy of lanostane-type triterpenoids such as cycloartenol or parkeol. Labelled cycloartenol, α-amyrin, β-amyrin and 24-methylenecycloartanol were also obtained from [3-3H]squalene-2,3-epoxide in the same enzymatic system yielding labelled 10α-cucurbitadienol.  相似文献

[6-3H1] (24S)-24-Ethylcholesta-5,22,25-trien-3β-ol added to the growth medium of a culture of Trebouxia sp. 213/3 was efficiently taken-up by the cells and converted into (24R)-24-ethylcholesta-5,22-dien-3β-ol (poriferasterol) which is one of the major sterols of this alga. A cell-free homogenate was obtained from Trebouxia which catalysed the NADPH-dependent reduction of [6-3H1] (24S)-24-ethylcholesta-5,22,25-trien-3β-ol to yield poriferasterol. The δ25-sterol reductase was found to be mainly localized in the microsomal fraction of the homogenate.  相似文献

The synthesis and structural elucidation, by 1D and 2D NMR and X-ray diffraction techniques, of novel E/Z 16-azolylmethylene-17-oxoandrostanes 2-9 prepared from the Vilsmeier-Hack reaction product 17-chloro-16-formylandrosta-5,16-dien-3β-yl acetate 1 is reported. The reaction proceeds with pyrrole and pyrrole-alike nitrogen heterocycles such as 7-azaindole, indole, and 3-methylindole, in DMF, at 80 °C, in the presence of K2CO3, and allowed the attachment of privileged heterocyclic moieties, through the nitrogen atom to the steroid core at C16 via a methine carbon bridge, which is unprecedented in the literature and of potential synthetic and biological interest. Considerations on the possible reaction mechanism are included. All the synthesized compounds are new and are currently being tested for biological activities.  相似文献

The structures of two 4α-methylsterols is isolated from Cucumis sativus(Cucurbitaceae) seeds were determined based mainly on their 13CNMR spectra as 24β-ethyl-31-norlanosta-8,25(27)-dien-3β-ol and 24β-ethyl-25(27)- dehydrolophenol, respectively, of which the former is a new sterol from natural sources. These two 4α-methylsterols were identified in the seeds of two other Cucurbitaceae species, Lagenaria leucantha var. Gourda and Citrullus battich. The probable biogenetic significance of the two 4α-methylsterols is discussed. Other 4α-methylsterols identified in the seeds of the three Cucurbitaceae species were obtusifoliol, cycloeucalenol and gramisterol.  相似文献

When [(14)C]cholesterol was incubated with rat liver mitochondria, radioactive 26-hydroxycholesterol, 3beta-hydroxychol-5-enoic acid and other bile acids were isolated from the incubation mixture. 2. In the absence of added 26-hydroxycholesterol, the specific radioactivity of the 26-hydroxycholesterol formed from [(14)C]cholesterol during the incubation was higher than that of the 3beta-hydroxychol-5-enoic acid. Addition of increasing amounts of 26-hydroxycholesterol led to a progressive fall in the specific radioactivity, and to a progressive increase in the mass, of the 3beta-hydroxychol-5-enoic acid recovered at the end of the incubation. 3. It is concluded that 26-hydroxycholesterol is an intermediate in the formation of 3beta-hydroxychol-5-enoic acid from cholesterol. 4. Comparison of the specific radioactivity of the 26-hydroxycholesterol formed in the incubation mixture with that of the added [(14)C]cholesterol indicates that endogenous cholesterol in mitochondria is accessible to cholesterol 26-hydroxylase.  相似文献
