The relative efficacy of a mechanical (Prokopack) collection method vs. manual aspiration in the collection of resting mosquitoes was evaluated in northern Tanzania before and after an intervention using indoor residual spraying and longlasting insecticide‐treated nets. In smoke‐free houses mosquitoes were collected from the roof and walls, but in smoky houses mosquitoes were found predominantly on the walls. Anopheles gambiae (Diptera: Culicidae) constituted 97.7% of the 312 An. gambiae complex specimens identified before but only 19.3% of the 183 identified after the intervention. A single sampling with the Prokopack collected a third of the available insects. Anopheles gambiae completed its gonotrophic development indoors, whereas Anopheles arabiensis did so outdoors. In both species gonotrophic development took 2 days. Most unfed resting An. arabiensis collected outdoors were virgins, whereas the majority of engorged insects were parous (with well‐contracted sacs). Daily survival was estimated to be 80.0%. Only 9.4% of the engorged An. arabiensis collected outdoors and 47.1% of those collected indoors had fed on humans. Using the Prokopack sampler is more efficient than manual methods for the collection of resting mosquitoes. Malaria transmission may have been affected by a change in vector composition resulting from a change in feeding, rather than reduced survival. Monitoring the proportions of members of the An. gambiae complex may provide signals of an impending breakdown in control.  相似文献

A histological technique was used to detect the multiple blood-feeding of female mosquitoes, within the same gonotrophic cycle, in field populations of the malaria vectors Anopheles culicifacies and An. subpictus (Diptera: Culicidae) at a village in the low country of Sri Lanka, during 1994-96. Among 3306 An. culicifacies and 871 An. subpictus engorged females examined, respectively, 34.4% and 30.4% were multiple-fed. In these two species, double meals accounted for 92.7% and 89.5%, and triple meals for 7.3% and 10.5% of multiple meals, respectively. More females of An. culicifacies (53.7%) than An. subpictus (44.5%) multiple-fed on different nights. Multiple feeding rates in the two species were independent of sample size and field abundance. In An. culicifacies, but not An. subpictus, these rates were proportional to ambient temperature but not to humidity. Bloodmeal ELISA showed that 8.3% of 242 An. culicifacies were human-fed and 80% of these (i.e. 6.6%) were concurrently bovid-fed. Combining the histological and ELISA data, it was estimated that multiple feeding involving a human host had occurred in 7.2% of the blood-feeding An. culicifacies population at this study site. Only 1.2% of 406 An. subpictus were human-fed and all of these were concurrently bovid-fed. Multiple bloodfeeding within the same gonotrophic cycle was attributed to a local 'frequent feeding strategy' in these primarily zoophagic and endophilic malaria vectors. A test of host resistance to infestation by the tickRhipicephalus appendiculatus was repeated 5 times, at weekly intervals, on 2 initially susceptible calves. One hundred nymphs (NN) were used each time. Pairs of tick-susceptible rabbits served as controls. The engorged NN were weighed, then allowed to moult individually, and the sex of each was noted after emergence of the adult. During the course of the experiment there was a significant reduction in mean engorged weight (especially in the pre-male NN) and also in the length of the feeding period, and a downward trend in the percentage of NN which engorged. There was a significant increase in the percentage of engorged NN that moulted. Pre-male NN were significantly heavier than pre-female NN after feeding on susceptible hosts, but though mean feeding times were the same, they took 1–2 days longer to moult than pre-females at 28°C. It was concluded that the test itself could induce resistance if applied to fully susceptible calves.  相似文献

Sexual attractiveness of unengorged, semiengorged and fully engorged females of both field and laboratory populations of Ixodes ricinus was studied under laboratory conditions by means of a computerised video tracking system. A male and a female were allowed to walk freely in a glass arena during 1 h and their behavioural interactions were observed. Obtained results revealed that the feeding status of I. ricinus females affects their sexual attractiveness. The highest attractiveness was observed in engorged females, the lowest in unengorged females of the field population. Copulation occurred in all experimental groups with the highest frequency observed in the unengorged virgin laboratory females. Despite engorged females being highly attractive for males on distance, the frequency of copulation was low. Differences between field and laboratory populations were found in both behavioural parameters and frequency of copulation. Background

Mosquito biting frequency and how bites are distributed among different people can have significant epidemiologic effects. An improved understanding of mosquito vector-human interactions would refine knowledge of the entomological processes supporting pathogen transmission and could reveal targets for minimizing risk and breaking pathogen transmission cycles.

Methodology and principal findings

We used human DNA blood meal profiling of the dengue virus (DENV) vector, Aedes aegypti, to quantify its contact with human hosts and to infer epidemiologic implications of its blood feeding behavior. We determined the number of different people bitten, biting frequency by host age, size, mosquito age, and the number of times each person was bitten. Of 3,677 engorged mosquitoes collected and 1,186 complete DNA profiles, only 420 meals matched people from the study area, indicating that Ae. aegypti feed on people moving transiently through communities to conduct daily business. 10–13% of engorged mosquitoes fed on more than one person. No biting rate differences were detected between high- and low-dengue transmission seasons. We estimate that 43–46% of engorged mosquitoes bit more than one person within each gonotrophic cycle. Most multiple meals were from residents of the mosquito collection house or neighbors. People ≤25 years old were bitten less often than older people. Some hosts were fed on frequently, with three hosts bitten nine times. Interaction networks for mosquitoes and humans revealed biologically significant blood feeding hotspots, including community marketplaces.

Conclusion and significance

High multiple-feeding rates and feeding on community visitors are likely important features in the efficient transmission and rapid spread of DENV. These results help explain why reducing vector populations alone is difficult for dengue prevention and support the argument for additional studies of mosquito feeding behavior, which when integrated with a greater understanding of human behavior will refine estimates of risk and strategies for dengue control.  相似文献

A study of the age structure of a population of adult female Aedes cantans Meigen was undertaken using the ovarian oil injection technique to determine parity. Results showed that, contrary to popular opinion, ovariolar sacs do not form gonotrophic dilatations and cannot be used for physiological age determination. Dilatations were formed only by follicles which degenerate at a very early stage in the gonotrophic cycle. The proportion which consistently do degenerate (the gonotrophic diagnostic index) decreases in each successive cycle. Results showed that the mean age of the population during the study was 2.04 parous, which was one gonotrophic cycle less than expected, possibly due to scarcity of hosts in the area.  相似文献

Contrary to current opinion, fully engorged and detached larvae and nymphs of some ixodid ticks consistently take up substantial amounts of atmospheric water vapour and thereby display their regulative capacity for maintaining water balance in subsaturated air. Net uptake of vapour generally begins some days after detachment and the capability persists until shortly after initiation of apolysis, a period which in diapausing specimens may extend up to several months. This was shown forIxodes ricinus, Haemaphysalis punctata, and the North AmericanI. dammini. Apparently, some other engorged ixodid immatures fail to exhibit net vapour uptake, as was shown for both larvae and nymphs ofDermacentor marginatus and nymphs ofHyalomma anatolicum excavatum. But there is some evidence for engorged nymphs ofD. marginatus that active uptake of vapour does occur, masked by spiracular transpiration. Net uptake of vapour is apparently not possible during the pharate phases. InI. ricinus both teneral nymphs and adults are capable of achieving net water gains by active vapour uptake on the first day following ecdysis. There is new evidence from fully engorgedI. ricinus immatures for the decisive role of agranular alveoli in the production of the salivary secretion involved in vapour uptake.  相似文献

ABSTRACT. Females of Aedes aegypti (L.) (Diptera, Culicidae) were not receptive to mating 1 h after the initial insemination or at later times throughout the first gonotrophic cycle. A significant proportion of these females (75–90%) were inseminated again prior to the second gonotrophic cycle. Insemination was determined by the detection in females of sperm labelled with 3 H(G)-adenine or 32PO4 after matings with radiolabeled males. Females deprived of males after the first insemination displayed an abnormal reduction in fertility throughout successive gonotrophic cycles.  相似文献

Tick-borne diseases are a major epidemiological problem worldwide. The aim of this study was to investigate the bacterial composition of saliva obtained from engorged adult Rhipicephalus microplus females. Saliva samples collected from partially or fully engorged adult female ticks were analysed using an ultra-high-throughput Illumina HiSeq 2500 sequencing system. To elucidate the possible routes of bacterial transmission, the bacterial flora from whole ticks were also investigated. Proteobacteria, Firmicutes, and Actinobacteria were the predominant phyla in all samples, and Acinetobacter, Rickettsia, Escherichia and Coxiella were the major genera. Microbial diversity in saliva samples from partially engorged ticks was more complex than that of samples from fully engorged individuals. The comparison of saliva and whole-tick samples suggests that bacteria in saliva also colonize the tick’s body. We believe that some bacterial genera, such as Dermacoccus, Achromia, SMB53, Sutterella, Providencia, Mycoplana, Oscillospira, and Agrobacterium, were found and reported in ticks for the first time. The Coxiella and Rickettsia detected in this study might be tick-borne pathogens, suggesting health risks associated with exposure to R. microplus in humans and animals. These findings may serve as the basis for developing strategies to control ticks and tick-borne diseases.  相似文献

Anopheles darlingi is an important vector of human malaria in the Amazon. Adult females of this mosquito species require a blood meal to develop eggs, preferring humans to other blood sources. Although gonotrophic concordance has been described as the norm for An. darlingi, here we report An. darlingi female mosquitoes taking two or more blood meals within their first gonotrophic cycle. Only half of field‐captured adult females fed one blood meal developed follicles to Christophers' stage V. This outcome is dependent on larval nutrition, as 88% of laboratory‐raised well‐nourished females completed the first gonotrophic cycle with only one blood meal, while less nourished females needed additional blood meals. Half of the field‐captured blood‐seeking An. darlingi females had follicles in intermediate (IIIa and IIIb) and final (V) stages of the gonotrophic cycle, supporting the conclusion that An. darlingi blood feed more than once during a gonotrophic cycle. Additionally, we observed females attempting to blood feed a second time during the same day. Additional studies of An. darlingi biting behavior are necessary to accurately estimate Plasmodium sp. entomologic inoculation rates throughout the An. darlingi vast geographical distribution.  相似文献

Pre-existing energy reserves may play an important role in regulating the utilization of blood meal proteins in female anautogenous mosquitoes. Determining the fate of reserves derived from the sugar meal and larval food during the first gonotrophic cycle would help to elucidate the relative contributions of larval and adult nutrition to survival and reproduction. We measured the allocation of pre-blood-meal reserves to egg production or energy production during the first gonotrophic cycle by using [14C]-labeled female Aedes aegypti mosquitoes. Feeding adults [3,4-14C]-glucose labeled the glycogen and sugar stores (approximately 50%), lipid stores (approximately 25%), and protein and amino acid stores (approximately 25%). During the first gonotrophic cycle, about 60% of the glycogen and sugar stores were metabolized and all were used for energy production. About 33% of the labeled protein and 72% of the labeled amino acid stores were metabolized, with about 9% being transferred to the eggs and the rest oxidized. About 30% of the lipid was metabolized, with about 65% being transferred to the eggs and the rest oxidized. Feeding [1-14C]-oleic acid to larvae effectively labeled adult lipid stores with about 75% of the label in lipid stores and 16% in proteins and 6% in glycogen. During the first gonotrophic cycle, about 35% of the labeled lipid stores were metabolized, with equal amounts being oxidized and transferred to the eggs. None of the other maternal stores labeled by fatty acid were metabolized during the first gonotrophic cycle. These results show that carbohydrate reserves are a critical source of energy during the first gonotrophic cycle, while lipid reserves are used equally for energy production and provisioning the eggs.  相似文献

Seasonal development was investigated in the pigeon tick, Argas reflexus (F.) over a 5-year period. The ticks were kept in desiccators at two deposition sites with different temperature conditions: a warmer attic and a cooler outdoor aviary. The life cycle of A. reflexus consists of the egg, larva, a variable number of two to four nymphal instars and the adult stage. In the cooler aviary, the ticks passed, on average, fewer nymphal instars than in the attic. At both locations, ecdysis of the nymphs and adults occurred only during the summer months, with peak numbers of ticks finishing the moult in August. This consistent pattern was evident irrespective of the feeding date of the preceding developmental stage or the year of observation. The results strongly suggest that nymphs II, nymphs I and larvae fed later than in mid-July, August or September, respectively, entered a state of diapause and, thus, overwintered in the engorged state. Argas reflexus nymphs II from a laboratory stock that were deposited inside the attic showed a remarkably different seasonal pattern of development, even more than 1 year after their deposition. This suggests that a circannual rhythm may be involved in the ticks' seasonal timing. Mortality of the engorged ticks (from repletion to ecdysis of the following stage/instar) was below 1.5% in most cases, irrespective of the season and the location. Unfed larvae survived for a maximum of one year inside the attic, whereas the median survival period of unfed nymphs was at least 3 years at the same location. Based on the present results, the generation time from (F1) egg deposition to oviposition in the F2 generation might be 3-11 years in Central European A. reflexus, depending on the course of development (two or three nymphal instars) and the number of gonotrophic cycles (probably up to six) of the F1. Gonotrophic relations in low and higher bloodsucking Diptera are fundamentally different that results from an initial type of feeding and trophic behaviour. Trophic behaviour of low dipterous hematophages and gonotrophic harmony peculiar to them can be traced from entomophagy, that is from the predatory mode of life on account of small insects. By the type of the trophic behaviour low bloodsucking Diptera are predators with a typical moment contact with the prey. More primitive is a type of gonotrophic harmony characteristic of hunters for diffusely spread prey (incomplete blood portion provides the maturation of incomplete portion of eggs). Hunting for diffusely spread prey is characteristic of entomophages too. The appearance of gregarious ruminants facilitates the possibility of repeated contacts with prey and blood satiation threshold increases. This is a higher type of gonotrophic harmony providing a maximum realization of potential fecundity. The initial saprophagy of higher Diptera is associated with another type of trophic behaviour (long contact with food substratum) that is a prerequisite for quite a different way of evolution of host-parasite relationships in higher Diptera. This leads to more close connections with the host and excludes gonotrophic harmony. Females were the first to begin the exploitation of vertebrate animals. This is connected with the peculiarities of their behaviour during egg laying such as the stay near animals for laying eggs into fresh dung. Autogeneity, nectarophagy and aphagia are homologous phenomena which reflect the loss of an animal component of food or both components at the level of non-specialized saprophagy rather than secondary loss of bloodsucking. Abstract It has been shown previously that infection with Plasmodium yoelii nigeriensis reduces the number of eggs produced by female Anopheles stephensi. Here we examine the mechanism underlying fecundity reduction. Ovaries from infected and uninfected (control) female mosquitoes were examined12, 24 or 36 h after blood-feeding during the first gonotrophic cycle (replicated) or the second gonotrophic cycle (unreplicated). Follicular development was assessed according to Christophers' stages and the proportions of developing and resorbing follicles per ovary were determined. Resorption of some follicles commenced within 12h of blood-feeding, affecting significantly more follicles in the infected females: 1.1% v. 3.2%. The difference was greatest 36h after blood-feeding: 25% reduction (10 v. 35%) in the first cycle; 16% reduction (9 v. 25%) in the second gonotrophic cycle. The mean speed of oogenesis was also found to be significantly retarded in infected mosquitoes. During the second gonotrophic cycle, for example, only 92–94% of follicles reached stage III by 24 h and stage IV by 36 h in infected females, whereas all the developing follicles of uninfected females reached these stages more or less synchronously in the time specified.  相似文献

Estimates of seasonal abundance of larvae, nymphs and females of the paralysis tick, Ixodes holocyclus, were obtained by collecting ticks that engorged on small mammals and birds trapped in two localities in southeastern Queensland: Brisbane (wet sclerophyll forest) and Tamborine Mountain (cleared rain forest). The long-tailed short-nosed bandicoot, Isoodon macrourus, was the most common mammal trapped but small numbers of other marsupials, rodents and ground frequenting birds were also captured. Small numbers of five other tick species were also collected. In both habitats there was clearly one dominant generation of I. holocyclus;7er year, although the presence of all stages at most times of the year indicated overlapping of smaller cohorts. Females were most abundant in spring and early summer, larvae in summer-autumn, and nymphs in autumn-winter. I. holocyclus was abundant on I. macrourus and rare on most other mammals and birds captured. At the peak of abundance of each instar, each bandicoot dropped from 500 to 2000 engorged larvae, 100 to 200 engorged nymphs, and four to six engorged females. Life tables were compiled for the tick in both habitats and these indicate that there was relatively high survival from engorged larva to engorged nymph and thence to engorged female and that most mortality occurred between detaching of the engorged female and the detaching of the engorged larva. The tick was more abundant on bandicoots from cleared rain forest and rain forest edge, than on those from sclerophyll woodland. The survival of engorged larvae and nymphs of I. holocyclus and the larval productivity of engorged females were examined in a warm moist climate where the tick was abundant (Tamborine Mountain) and in a hotter dryer climate where the tick was rare or absent (Amberley). In both localities engorged larvae and nymphs survived to the next instar in all seasons of the year. On most occasions engorged females produced eggs which hatched. Mature bandicoots from tick infested areas showed little or no resistance to infestation with larvae or nymphs of I. holocyclus, whereas other small mammals from the same area showed an appreciable degree of resistance to the immature stages of the tick. Feeding larvae and nymphs exposed to normal light-dark cycles in the laboratory detached during the afternoon and early evening. This behaviour and host resistance are discussed in relation to the daily activity cycles of host species, their habitat preferences, and their role as hosts for I. holocyclus.  相似文献

Two haemolytic bacterial strains of Bacillus pumilus (CU1A, CU1B) and one blood‐utilizing strain of Bacillus licheniformis (CU2B) were isolated from relatively low numbers of field‐collected females of Culicoides oxystoma and Culicoides peregrinus (Diptera: Ceratopogonidae). A total of 36 females, including 18 of each of C. oxystoma and C. peregrinus (consisting of one and a pool of eight blood‐engorged specimens, and one and a pool of eight non‐engorged specimens for each species), were tested. In C. oxystoma, all three strains of bacteria were isolated from the one non‐engorged, the pool of non‐engorged and the pool of blood‐engorged females tested, but CU1A and CU2B were not found in the one blood‐engorged female tested. In C. peregrinus, all three strains were present in the pool of blood‐engorged females. However, the strain CU2B was not found in the pool of non‐engorged females. In the one blood‐engorged and one non‐engorged female tested, CU1A and CU2B were detected. The bacterial strains were identified based on Gram staining, enzyme activity (amylase and protease) and alignment of the 16S rRNA partial gene sequence to that available in the National Center for Biotechnology Information (NCBI) database GenBank. The functional role and significance of these haemolytic and blood‐digesting bacteria within the genus Culicoides remain to be determined.  相似文献
