The antimicrobial effect of 5 naphthoquinones was tested against the phytopathogenic bacteria Erwinia carotovora. Disk diffusion tests and determination of minimal inhibitory concentrations (MIC) indicate that the compound naphthazarin (NTZ) has the best antibacterial activity among the naphthoquinones tested. Studies on the mode of action indicate the effect of NTZ was bactericidal at 10 microg/mL. When cultivation was done in the presence of sodium ascorbate, the restoration of E. carotovora growth was observed with 3 microg/mL NTZ, but not when a 10 microg/mL dose was used. The incubation of NTZ with bacterial suspension of E. carotovora resulted in important changes in the absorption spectra of this naphthoquinone, indicating that a redox reaction takes place. These results may suggest that NTZ induces an increase of reactive oxygen species that are toxic to the cell. The compound NTZ was also effective in preventing E. carotovora growth on potato tubers, inhibiting the soft rot development at a concentration of 2 mg/mL.  相似文献

Growth conditions are described for optimum production of extracellular protease in batch cultures of the soft rot pathogen Erwinia carotovora subsp. carotovora . This protease was inhibited by approximately 96% by 1 mmol/1 EDTA and by 55–6% by 10 mmol/l cysteine thereby classifying it as a metalloprotease. It was not inhibited by a chymotrypsin inhibitor extracted from potato tubers. This evidence suggests that the potato chymotrypsin inhibitor is not associated with resistance of potatoes to E. carotovora subsp. carotovora .  相似文献

The soft rot bacteria Erwinia carotovora and Erwinia chrysanthemi are important pathogens of potato and other crops. However, the taxonomy of these pathogens, particularly at subspecies level, is unclear. An investigation using amplified fragment length polymorphism (AFLP) fingerprinting was undertaken to determine the taxonomic relationships within this group based on their genetic relatedness. Following cluster analysis on the similarity matrices derived from the AFLP gels, four clusters (clusters 1 to 4) resulted. Cluster 1 contained Erwinia carotovora subsp. carotovora (subclusters 1a and 1b) and Erwinia carotovora subsp. odorifera (subcluster 1c) strains, while cluster 2 contained Erwinia carotovora subsp. atroseptica (subcluster 2a) and Erwinia carotovora subsp. betavasculorum (subcluster 2b) strains. Clusters 3 and 4 contained Erwinia carotovora subsp. wasabiae and E. chrysanthemi strains, respectively. While E. carotovora subsp. carotovora and E. chrysanthemi showed a high level of molecular diversity (23 to 38% mean similarity), E. carotovora subsp. odorifera, E. carotovora subsp. betavasculorum, E. carotovora subsp. atroseptica, and E. carotovora subsp. wasabiae showed considerably less (56 to 76% mean similarity), which may reflect their limited geographical distributions and/or host ranges. The species- and subspecies-specific banding profiles generated from the AFLPs allowed rapid identification of unknown isolates and the potential for future development of diagnostics. AFLP fingerprinting was also found to be more differentiating than other techniques for typing the soft rot erwinias and was applicable to all strain types, including different serogroups.  相似文献

Isolates of Erwinia carotovora sub sp. atroseptica, E. carotovora sub sp. carotovora and E. chrysanthemi were compared for their ability to rot potato tubers and to elicit the accumulation of phytoalexins. With three incubation temperatures (15, 22 and 30°C) tubers were more susceptible at higher temperatures and mean rot weights were inversely correlated with mean rishitin concentrations. However, within treatment variation between tubers in the extent of rotting was not correlated with rishitin concentration suggesting that other factor(s) also affect resistance. Phytoalexins, including 15-dihydrolubimin, 3-hydroxylubimin, phytuberin and solavetivone, were measured by high pressure liquid chromatography using a silica column.  相似文献

The soft rot coliform bacteria Erwinia carotovora subsp. carotovora and E. carotovora subsp. atroseptica were isolated by an enrichment method from the rhizosphere of many weed species and crop plants, collected in commercial potato fields either currently in potatoes or in a different crop as part of the rotation. Erwinia carotovora was isolated from 24 plant species in Colorado and 47 species in Scotland. Weeds contaminated with E. carotovora were found in fields growing other crops in which potatoes had not been grown for 1–2 and sometimes much longer. Weeds collected from virgin land in Colorado were not contaminated with E. carotovora but in Scotland virgin soils containing weed roots yielded E. carotovora subsp. carotovora . In general, the numbers of contaminated weeds rose from nil or low levels in spring and early summer to considerably higher levels during mid-season, and fell to progressively lower levels later. Erwinia carotovora subsp. carotovora was the predominant organism recovered from the rhizosphere, but E. carotovora subsp. atroseptica was less common, especially in Scotland, and its incidence varied in different seasons depending on factors such as temperature and moisture conditions. The bacteria could apparently persist in the root zone for an extended period of time and may be a source of inoculum to contaminate soft rot erwinia-free seed potato stocks; the origin of the bacteria was uncertain.  相似文献

采用魔芋Amorphophallus spp.块茎点种、注射、灌根接种及田间调查等方法,对国内普遍栽种的珠芽红魔芋A. bulbifer、珠芽金魔芋A. muelleri、花魔芋A. konjac和白魔芋A. albus等12个种质材料进行抗软腐病鉴定、比较和评价,以分析珠芽魔芋对抗细菌性软腐病的抗病水平。结果表明,供试材料对软腐病抗性差异较大,珠芽金魔芋种质对细菌性软腐病均有免疫性(I),德宏及临沧珠芽红魔芋种质为高抗病品种(HR);缅甸珠芽红魔芋为抗病品种(R);富源花魔芋、楚雄花魔芋、日本农林2号、鄂魔芋1号、秦魔1号、昭通白魔芋、丽江白魔芋均属易感品种(S),与田间抗性调查情况基本相符。  相似文献

Seven new genes controlled by the quorum-sensing signal molecule N-(3-oxohexanoyl)-L-homoserine lactone (OHHL) have been identified in Erwinia carotovora subsp. carotovora. Using TnphoA as a mutagen, we enriched for mutants defective in proteins that could play a role in the interaction between E. carotovora subsp. carotovora and its plant hosts, and identified NipEcc and its counterpart in E. carotovora subsp. atroseptica. These are members of a growing family of proteins related to Nep1 from Fusarium oxysporum which can induce necrotic responses in a variety of dicotyledonous plants. NipEcc produced necrosis in tobacco, NipEca affected potato stem rot, and both affected virulence in potato tubers. In E. carotovora subsp. carotovora, nip was shown to be subject to weak repression by the LuxR family regulator, EccR, and may be regulated by the negative global regulator RsmA.  相似文献

Pectin lyase (PNL) and the bacteriocin carotovoricin (CTV) were induced in Erwinia carotovora subsp. carotovora 71 by the DNA-damaging agents mitomycin C, nalidixic acid, and UV light. To determine whether the recA product was involved in the expression of these damage-inducible phenotypes, we cloned the E. carotovora subsp. carotovora recA+ gene, inactivated it by Tn5 insertion, and constructed an E. carotovora subsp. carotovora recA::Tn5 strain by gene replacement via homologous recombination. The RecA- strain was more sensitive to methyl methanesulfonate, nitroquinoline oxide, and UV light than its RecA+ parent. The recA mutation did not affect the production of pectate lyase, polygalacturonase, cellulase, and protease or the ability to cause soft rot of potato tubers. With this mutant, unlike with the RecA+ parent strain, PNL and CTV were not induced by mitomycin C or detected in potato tuber tissue. The RecA+ phenotype, including the inducibility of PNL and CTV, could, however, be restored in the mutant in trans by the recA+ gene from either E. carotovora subsp. carotovora or Escherichia coli. We conclude that, in E. carotovora subsp. carotovora, the recA product is required in the induction of PNL and CTV.  相似文献

Erwinia carotovora subsp. carotovora is a gram-negative bacterium that causes soft rot disease of many cultivated crops. When a collection of E. carotovora subsp. carotovora isolates was analyzed on a Southern blot using the harpin-encoding gene hrpN as probe, several harpinless isolates were found. Regulation of virulence determinants in one of these, strain SCC3193, has been characterized extensively. It is fully virulent on potato and in Arabidopsis thaliana. An RpoS (SigmaS) mutant of SCC3193, producing elevated levels of secreted proteins, was found to cause lesions resembling the hypersensitive response when infiltrated into tobacco leaf tissue. This phenotype was evident only when bacterial cells had been cultivated on solid minimal medium at low pH and temperature. The protein causing'the cell death was purified and sequenced, and the corresponding gene was cloned. The deduced sequence of the necrosis-inducing protein (Nip) showed homology to necrosis- and ethylene-inducing elicitors of fungi and oomycetes. A mutant strain of E. carotovora subsp. carotovora lacking the nip gene showed reduced virulence in potato tuber assay but was unaffected in virulence in potato stem or on other tested host plants.  相似文献

Potato can be attacked by several economically important pathogens. From the various diseases, in our experiment we dealt with the bacterial soft rot of potato caused by Erwinia species. In the experiments back cross progenies (BC1, BC2, BC3 and BC4) of Solanum brevidens + Solanum tuberosum somatic hybrids produced by the Potato Research Centre, Keszthely were tested to the infection of E carotovora ssp. atroseptica (Eca), E. carotovora ssp. carotovora (Ecc) and E. chrysanthemi (Echr). All together 11 BC genotypes pre selected from several hundred breeding lines based on their preferred agronomical appearance and virus resistance characters as well as 4 Hungarian potato cultivar (Rioja, Desiree, White lady and Hópehely) as controls were involved into the experiments. Tuber slices from each genotype were artificially infected with bacteria suspension Ecc strain D3, and Echr strain CHR 1492, and Eca strain BN3) and incubated at 27 degrees C with 100% relative air humidity for 48 h before evaluation. Dry matter and starch content of tubers were determined right before the tests. Volume of rotted tuber tissue was determined in mm3 and used for comparison of the level of resistance or susceptibility of the genotypes. Relationship between the reaction to the bacteria strains and dry matter content was examined also. Tested genotypes showed the highest resistance to Eca, while the highest susceptibility to Echr. By the increase of BC level the susceptibility of the genotypes significantly increased as well regardless of the tested bacteria. No direct correlation was found between the dry matter content of tubers and their reaction to tested bacteria.  相似文献

Soft rot and wilt symptoms were noticed on 2-year-old seed cabbage plants grown in field crops. Soft rot Erwinia was isolated from the diseased tissues. Pathogenicity tests showed that the bacterium effectively colonised different vegetable species and sunflower, indicating a wide host range.
Pathogenicity, morphological, biochemical and physiological characteristics showed that the investigated strains are of Erwinia carotovora subsp. carotovora (Jones) Bergey et al.  相似文献

Potato is the world's largest non-cereal crop. Potato late blight is a pandemic, foliar wasting potato disease caused by Phytophthora infestans, which has become highly virulent, fungicide resistant, and widely disseminated. Similarly, fungicide resistant isolates of Phytophthora erythroseptica, which causes pink rot, have also become an economic scourge of potato tubers. Thus, an alternate, cost effective strategy for disease control has become an international imperative. Here we describe a strategy for engineering potato plants exhibiting strong protection against these exceptionally virulent pathogens without deleterious effects on plant yield or vigor. The small, naturally occurring antimicrobial cationic peptide, temporin A, was N-terminally modified (MsrA3) and expressed in potato plants. MsrA3 conveyed strong resistance to late blight and pink rot phytopathogens in addition to the bacterial pathogen Erwinia carotovora. Transgenic tubers remained disease-free during storage for more than 2 years. These results provide a timely, sustainable, effective, and environmentally friendly means of control of potato diseases while simultaneously preventing storage losses.  相似文献

Four media were tested for their ability to detect the soft rot potato pathogens Erwinia chrysanthemi (Ech) and Erwinia carotovora ssp. atroseptica (Eca) in potato tubers by means of automated conductance measurements. The specificity of the conductimetric assays was determined by testing a set of different Erwinia spp. and potato-associated saprophytes, including the genera Pseudomonas, Bacillus, Enterobacter and Flavobacterium. All bacteria tested produced conductance responses in Special Peptone Yeast Extract, whereas in minimal medium with L-asparagine only Erwinia spp. and Pseudomonas spp. were able to generate large conductance responses. In minimal medium supplemented with glucose and trimethylamine- N -oxide only Enterobacteriaceae, Erwinia spp. included, generated conductance responses, while with pectate as sole carbon source only Erwinia spp. produced distinct conductance responses. The pectate medium proved to be particularly useful for specific automated conductimetric detection of Erwinia spp. in potato peel extracts. Within 48 h, the detection threshold of the conductimetric assay for Eca varied between 102 and 103 cfu per ml peel extract at both incubation temperatures of 20° and 26°C. Ech was detected at concentrations of 104–105 or 103–104 cfu ml-1 at 20° and 26°C, respectively. To eliminate 'false'-positive reactions in conductimetry caused by Erwinia carotovora ssp. carotovora , results of the conductance measurements have to be confirmed by other techniques, like serology or DNA assays.  相似文献

Current identification methods for the soft rot erwinias are both imprecise and time-consuming. We have used the 16S-23S rRNA intergenic transcribed spacer (ITS) to aid in their identification. Analysis by ITS-PCR and ITS-restriction fragment length polymorphism was found to be a simple, precise, and rapid method compared to current molecular and phenotypic techniques. The ITS was amplified from Erwinia and other genera using universal PCR primers. After PCR, the banding patterns generated allowed the soft rot erwinias to be differentiated from all other Erwinia and non-Erwinia species and placed into one of three groups (I to III). Group I comprised all Erwinia carotovora subsp. atroseptica and subsp. betavasculorum isolates. Group II comprised all E. carotovora subsp. carotovora, subsp. odorifera, and subsp. wasabiae and E. cacticida isolates, and group III comprised all E. chrysanthemi isolates. To increase the level of discrimination further, the ITS-PCR products were digested with one of two restriction enzymes. Digestion with CfoI identified E. carotovora subsp. atroseptica and subsp. betavasculorum (group I) and E. chrysanthemi (group III) isolates, while digestion with RsaI identified E. carotovora subsp. wasabiae, subsp. carotovora, and subsp. odorifera/carotovora and E. cacticida isolates (group II). In the latter case, it was necessary to distinguish E. carotovora subsp. odorifera and subsp. carotovora using the alpha-methyl glucoside test. Sixty suspected soft rot erwinia isolates from Australia were identified as E. carotovora subsp. atroseptica, E. chrysanthemi, E. carotovora subsp. carotovora, and non-soft rot species. Ten "atypical" E. carotovora subsp. atroseptica isolates were identified as E. carotovora subsp. atroseptica, subsp. carotovora, and subsp. betavasculorum and non-soft rot species, and two "atypical" E. carotovora subsp. carotovora isolates were identified as E. carotovora subsp. carotovora and subsp. atroseptica.  相似文献

Using a Casella High Volume Airborne Bacteria Sampler, the soft rot coliform bacteria Erwinia carotovora var. carotovora and E. carotovora var. atroseptica were caught from the open air during rainfall in mid to late summer, autumn and early winter, but not in late winter, spring or early summer. They were not found every time it rained, and never when the weather was dry. The bacteria were caught close to potato crops, but there were several occasions when they were found at sites where there were no such crops or at times of year after potato crops had been harvested. The sources of the organisms are uncertain, but those obtained close to potato crops may have originated there. The results are considered to provide further evidence that airborne spread and subsequent deposition of viable bacteria could cause contamination of Erwinia -free potato stocks, and suggest that rainfall is a major generator of the general atmospheric bacterial aerosol.  相似文献

Plant defense responses to pathogen infection involve the production of active oxygen species, including hydrogen peroxide (H2O2). We obtained transgenic potato plants expressing a fungal gene encoding glucose oxidase, which generates H2O2 when glucose is oxidized. H2O2 levels were elevated in both leaf and tuber tissues of these plants. Transgenic potato tubers exhibited strong resistance to a bacterial soft rot disease caused by Erwinia carotovora subsp carotovora, and disease resistance was sustained under both aerobic and anaerobic conditions of bacterial infection. This resistance to soft rot was apparently mediated by elevated levels of H2O2, because the resistance could be counteracted by exogenously added H2O2-degrading catalase. The transgenic plants with increased levels of H2O2 also exhibited enhanced resistance to potato late blight caused by Phytophthora infestans. The development of lesions resulting from infection by P. infestans was significantly delayed in leaves of these plants. Thus, the expression of an active oxygen species-generating enzyme in transgenic plants represents a novel approach for engineering broad-spectrum disease resistance in plants.  相似文献

The influence of soil moisture and mean day temperature of July to September on pectin structure and soft rot resistance of potato tubers was investigated. Tuber samples of middle early and middle late potato cultivars respectively were tested from 1976 to 1979. Cultivar characteristics as well as growth conditions effect the pectin structure. The tuber tissue of soft rot resistant cultivars contains relative high amounts of esterified pectin. The pectin esterification was promoted by drought during vegetation periods (1976, 1978). A late rise of metabolic activity caused by rainfall at the end of the vegetation period (1978) increased the amount of soluble pectin in the cell wall. This lack of pectin stabilization facilitates tissue maceration by soft rot bacteria. Increased pectin solubility was also caused by unfavorable storing conditions of potato tubers. A low degree of esterification and high solubility of the cell wall pectin affect the relative resistance of potato tubers against soft rot caused by Erwinia carotovora var. atro-septica (van Hall) Dye adversely.  相似文献

An anaerobic liquid enrichment method followed by plating on a selective medium revealed that the soft rot coliform bacterium Erwinia carotovora subsp. carotovora was generally present in water from drains, ditches, streams, rivers and lakes (including reservoirs) in southern Scotland and in Colorado, United States, in mountainous, upland and arable areas through the year. Many sites were remote from susceptible or diseased crops. Erwinia carotovora subsp. atroseptica was isolated much less frequently and no Erwinia bacteria were isolated from underground waters. Erwinia bacteria were also found in rain-water in Scotland, in winter snow from mountain passes in Colorado, and in sea water from the west and east coasts of Scotland and from the coasts of Oregon, California, Texas, Louisiana and Florida. The significance of the occurrence of these bacteria in water is discussed in relation to the control of blackleg and soft rot diseases of potato by production of Erwinia -free stocks.  相似文献

对黑龙江省成熟大白菜[Brassica pekinensis(Lour.)Rupr.]生产田采集的软腐病菌进行分离、纯化,并根据形态学特征分析了大白菜软腐病菌的种群组成。结果表明,引起黑龙江省秋季大白菜软腐病的主要致病菌是胡萝卜软腐欧文氏菌胡萝卜软腐亚种[Erwinia carotovora(Jones)Bergey et al.subsp、carotovora](Ecc);利用20个Ecc菌株的混合菌对来源于不同生态型及不同地区的大白菜品种进行接种,筛选出5个鉴别寄主,以此将20个Ecc菌株划分为5个致病力类型,其中V型为优势致病菌,其分布广且致病力强。  相似文献

Soft rot Erwinia spp., like other closely related plant pathogens, possess a type III secretion system (TTSS) (encoded by the hrp gene cluster) implicated in disease development. We report the sequence of the entire hrp gene cluster and adjacent dsp genes in Erwinia carotovora subsp. atroseptica SCRI1039. The cluster is similar in content and structural organization to that in E. amylovora. However, eight putative genes of unknown function located within the E. carotovora subsp. atroseptica cluster do not have homologues in the E. amylovora cluster. An arrayed set of Tn5 insertional mutants (mutation grid) was constructed and pooled to allow rapid isolation of mutants for any given gene by polymerase chain reaction screening. This novel approach was used to obtain mutations in two structural genes (hrcC and hrcV), the effector gene dspE/A, and the helper gene hrpN. An improved pathogenicity assay revealed that these mutations led to significantly reduced virulence, showing that both the putative E. carotovora subsp. atroseptica TTSS-delivered effector and helper proteins are required for potato infection.  相似文献
