Total DNA was extracted from 25 species (8 genera) of the subfamily Lamioideae sensu Erdtman (family Labiatae). Using rbcL-specific primers, the rbcL gene was amplified by polymerase chain reaction (PCR) and 1420 bp were sequenced directly, or after cloning. RbcL sequences were evaluated with character-state (maximum parsimony; PAUP) and distance methods (neighbour-joining; MEGA). In agreement with classical systematics all taxa studied cluster outside the Nepetoideae. The following clades are apparent: I - Westringia/Prostanthera, II - Ajuga/Teucrium and III - Scutellaria, Stachys, Lamium and Marrubium. Lamium album, L. amplexicaule and L. garganicum appear as a sister clade to Marrubium. Both share a common ancestor with a second Lamium clade (including L. purpureum, L. maculatum and L. galeobdolon). The phylogenetic relationships are discussed with regard to the systematic classification of the subfamilies Lamioideae and Nepetoideae (sensu Erdtman). 相似文献

Canavanine is a nitrogen-rich non-protein amino acid, with a supposedly dual role of food storage and defence in seeds and seedlings. It seems to be unique to Leguminosae subfamily Papilionoideae although it does not occur in all species or genera. This report extends the previous surveys for canavanine to 1200 species in 240 genera (including first records for 70 genera) and assesses the systematic and ecological significance of its distribution. 相似文献

Alkaloid profiles of extracts of fourteen papilionaceous species in the tribes Thermopsideae and Genisteae (sensu Polhill) have been determined. Altogether, eighteen quinolizidine alkaloids, representative of four structural groups, and one dipiperidine alkaloid were identified among the legumes studied. Quinolizidine alkaloids were surveyed in 22 plant samples, representing nineOrmosia species and up to five different plant parts per species, using combined gas chromatography and mass spectroscopy. The detected

Summary Explants of axillary buds excised from mature adult stems of Swainsona formosa (G. Don) J. Thompson (syn. Clianthus formosus) were cultured on Murashige and Skoog medium supplemented with a range of auxins, cytokinins, and sucrose concentrations.
Auxins did not increase shoot or bud numbers above controls, and 2,4-dichlorophenoxyacetic acid was the only auxin to significantly
increase callus production. Benzyladenine or thidiazuron incorporated into the medium at 0.1 μM stimulated shoot and bud production, and shoot growth occurred following removal of cytokinins from the medium after 4 wk.
Shoot number increased linearly with sucrose concentration up to 40 g l−1, but shoot height and the number of cytokinin-induced buds were optimal at sucrose levels of 20–30 g l−1. Roots were initiated in vitro following treatment of cuttings with 0.1% indole-3-butyric acid and 0.1% α-naphthaleneactic acid. Plantlets were successfully

The subtribal classification of the tribe Phaseoleae has drifted into disorder since Bentham's (1865) scheme in the Genera Plantarum. A revised classification is here proposed, which for the first time accounts for all of the currently recognized genera. This classification has seven subtribes: Cajaninae, Diocleinae, Kennediinae, Phaseolinae, Glycininae, Ophrestiinae, and Erythrininae, of which the Ophrestiinae is new. A survey for the free amino acid, canavanine, shows that it is most often present in the Diocleinae and Kennediinae and frequently absent in the rest of the tribe. 相似文献

The presence of alkaloids in six species of Brongniartia and three species of Harpalyce is reported. This survey revealed remarkable qualitative differences in the alkaloid profiles of these two genera. B. discolor, B. lupinoides, B. sousae and B. intermedia showed a typical -pyridone pattern, with cytisine, anagyrine and baptifoline as major alkaloids. In leaves of the first three species ormosanine-type alkaloids occurred additionally. B. flava and B. vazquezii are devoid of -pyridones, but accumulate lupanine, hydroxylated lupanines and ester alkaloids. All three species of Harpalyce were similar in accumulating -pyridones, but H. formosa differed from H. brasiliana and H. pringlei in the presence of epilupinine. In general the alkaloid profiles of Brongniartia and Harpalyce show similarities to those of the Australian genera Hovea, Lamprolobium, Plagiocarpus and Templetonia and support therefore the actual concept of the enlarged tribe Brongniartieae. 相似文献

基于叶绿体trnL_F序列单独分析以及trnL_F和rbcL序列联合分析重建了木通科的分子系统发育。本研究的系统发育拓扑结构与覃海宁和塔赫他间的族划分系统非常一致。猫儿屎族和串果藤族在系统发育树上位于本科的基部。由分布于南美的勃奎拉藤属和拉氏藤属组成的拉氏藤族得到了trnL_F序列分析 (10 0 % )和联合序列分析 (99% )的很好支持。木通族在两个分析里都得到了 10 0 %的靴带支持率。新建立的长萼木通属在trnL_F树上嵌套在木通属内 ;然而 ,在联合分析的树上 ,它与木通属形成姐妹群并得到很高的支持率。在系统发育上关系密切的 3个属 :牛藤果属、八月瓜属和野木瓜属之间的关系仍未解决。牛藤果与八月瓜在两个分析里都形成姐妹群 ,但支持率低。小花鹰爪枫嵌套在野木瓜属内 ,并与西南野木瓜形成姐妹群。木通族内这 3个属可能都不是单系 ,它们的属间界限和属的界定需要更多的分子和形态数据的进一步研究。 相似文献

Phylogenetic relationships in the tribe Millettieae and allies in the subfamily Papilionoideae (Leguminosae) were reconstructed from chloroplast trnK/matK sequences. Sixty-two accessions representing 57 traditionally recognized genera of Papilionoideae were sampled, including 27 samples from Millettieae. Phylogenies were constructed using maximum parsimony and are well resolved and supported by high bootstrap values. A well-supported "core Millettieae" clade is recognized, comprising the four large genera Millettia, Lonchocarpus, Derris, and Tephrosia. Several other small genera of Millettieae are not in the core Millettieae clade. Platycyamus is grouped with Phaseoleae (in part). Ostryocarpus, Austrosteenisia, and Dalbergiella are neither in the core Millettieae or Phaseoleae clade. These taxa, along with core Millettieae and Phaseoleae, form a monophyletic sister group to Indigofereae. Cyclolobium and Poecilanthe are close to Brongniartieae. Callerya and Wisteria belong to a large clade that includes all the legumes that lack the inverted repeat in their chloroplast genome, which confirms previous rbcL and phytochrome gene family phylogenies. The evolutionary history of four characters was examined in Millettieae and allies: the presence of canavanine, inflorescence types, the dehiscence of pods, and the presence of winged pods. trnK/matK sequence analysis suggests that the presence of a pseudoraceme or pseudopanicle and the accumulation of nonprotein amino acids are phylogenetically informative for Millettieae and allies with only a few exceptions. 相似文献

Internal transcribed spacer (ITS) sequences of nuclear ribosomal DNA from 44 taxa of the genus Lupinus and five outgroup taxa were used for phylogenetic analysis. Lupinus appears as a strongly supported monophyletic genus, which is unambiguously part of the Genisteae. The lupines are distributed into five main clades in general accordance with their geographical origin. In the Old World, almost all the recognized taxonomic units are well resolved. The ITS data reveal an unexpectedly close relationship between the diverse sections Angustifoli and Lutei. The ITS results suggest a geographical division between the western New World lupines and the eastern ones. They also indicate the presence of some moderately to strongly supported groups of taxa, such as the Microcarpi-Pusilli group, the L. spariflorus-L. arizonicus group, the L. mexicanus-L. elegans group in the western New World, and the notable L. multiflorus-L. paraguariensis group in the eastern New World. The latter group strongly suggests that the eastern South American compound- and simple-leaved perennial lupines derive from a common ancestor. However, apart from some exceptions, relationships within the genus still remain largely unresolved based on ITS data. The lack of resolution at the base of the genus is suggestive of a rapid initial radiation of the lupines subsequent to the dispersal of their common ancestor. Relative rate tests demonstrate the presence of rate heterogeneity of ITS sequences within Lupinus. In many pairwise comparisons between taxa, substitution rate inequalities are correlated with the habit (annual, perennial), suggesting some role for the generation time effects in the evolutionary history of lupines. 相似文献

Floral ontogeny of Lespedeza thunbergii was studied with the use of scanning electron microscopy (SEM). The ontogeny varies in all whorls from the undirectional mode, which has been long held to be the rule in Leguminosae. In the sepal whorl, the lateral and the adaxial sepals are formed simultaneously, which is interpreted as a tendency towards whorled organ formation. Whorled organ formation is shown in the petal whorl. The antesepalous stamen whorl varies least from the unidirectional mode. Here, the adaxial stamens are formed successively. This is seen as a remnant of an original helical organ formation in Papilionoideae. Within the antepetalous stamen whorl, the two abaxial stamens and the adaxial stamen are formed first, followed by the two lateral stamens. This is a rarely found phenomenon, which is hard to interpret at the present state of knowledge. Concerning the mature flower, it is shown that nectar stomata are found in a distinct area on the adaxial side of the flower. The presented new characteristics should be an initial step toward further work on taxa of the tribe Desmodieae. These studies will broaden the data set and enable a detailed phylogenetic analysis. 相似文献

The 18S rDNA was used to infer oral ciliature patterns of evolution within the family Ophryoscolecidae, with the addition of five new sequences of ciliates from the genus Ostracodinium. Our data confirmed the monophyly of the subfamilies Entodiniinae and Ophryoscolecinae, but more analysis would be required for the definition of the status of the subfamily Diplodiniinae. The oral infraciliature patterns reflect evolutionary divergence in the family Ophryscolecidae, observing monophyly on Entodinium‐type, Diplodinium‐type, Ostracodinium‐type, Epidinium‐type, and Ophryoscolex‐type. The ancestral infraciliature of Entodinium‐type cannot be proven, however, the position of Entodinium‐type showed closer of Diplodinium‐type than Ophryoscolex‐type, corroborating previous studies using morphological characters. The observed inconsistencies reflect the need to increase the number of 18S rDNA sequences to family Ophryoscolecidae and investigate the evolution of this group using other molecular markers. 相似文献

Phylogenetic analysis of phytochrome (PHY) genes reveals the identity and relationships of four PHY loci among papilionoid Leguminosae. A phylogenetic analysis of loci combined according to species suggests that most of the tribe Millettieae belongs to one of two monophyletic clades: the Derris–Lonchocarpus or the Tephrosia clade. Together these two form a monophyletic group that is sister to a lineage represented by Millettia grandis of Millettia sect. Compresso-gemmatae. Collectively, this large monophyletic group is referred to as the Millettieae-core group, which based on our sampling, includes species of Millettieae that do not accumulate the nonprotein amino acid canavanine and that mostly have pseudoracemose or pseudopaniculate inflorescences. This new phylogenetic framework assists in targeting additional taxa for future sampling. For example, the “American Derris” (Deguelia), which accumulate canavanine, might not be members of the Millettieae core group. Afgekia is also predicted not to be a member because it accumulates canavanine and has an inflorescence of terminal racemes. PHY gene analysis specifically reveals that certain genera traditionally classified in Millettieae are actually distantly related to the Millettieae core group, such as Austrosteensia, Callerya, Craibia, Cyclolobium, Fordia, Platycyamus, Poecilanthe, and Wisteria. 相似文献

To obtain additional information on the phylogenetic relationships within the family Trypanosomatidae (order Kinetoplastida), we have sequenced the small subunit ribosomal RNA genes from the endosymbiont containing species Herpetomonas roitmani TCC080, Herpetomonas sp. TCC263, Crithidia oncopelti ATCC 12982 and a partial large subunit rRNA gene from H. roitmani. The small subunit sequences in the two isolates of Herpetomonas are very similar but not identical, and so are their restriction digest profiles of kinetoplast DNA. The size of minicircles in both isolates is 4.2 kilobases. The inferred ribosomal RNA phylogenetic trees shows the genera Herpetomonas and Crithidia as polyphyletic. Endosymbiont-bearing herpetomonads cluster with the endosymbiont-bearing crithidias and a blastocrithidia to form a monophyletic clade, whereas the endosymbiont-free members of these genera are found elsewhere in the tree. These data support the hypothesis of a monophyletic origin of endosymbiosis in trypanosomatid evolution and also suggest that a taxonomic revision is needed in order to better describe the natural affinities in this family. 相似文献
