The killing efficiency of tritium disintegrations in frozen mammalian cells labeled with tritiated uridine, histidine, and lysine was compared with the killing efficiency of incorporated tritiated thymidine. In each case, the distribution of tritium in the cells was determined by chemical fractionation as well as by radio-autography. Of all tritium disintegrations, by far the most effective were those occurring in DNA molecules within frozen cells; such incorporated tritium has a killing efficiency of 0.006. When cells were incubated with tritiated uridine for 10 min to label nuclear RNA, the killing efficiency was 0.0015. When the cells were pulse labeled with tritiated uridine and permitted to grow in nonradioactive media for 10 hr before freezing in order to incorporate tritium into cytoplasmic RNA, the killing efficiency was reduced to 0.0010. The results suggest that decay of tritium in nuclear RNA is more effective than that in cytoplasmic RNA. When the cells were labeled with tritiated histidine or lysine for 30 min, tritium atoms were found mainly in the acid soluble rather than in the protein fraction and the killing efficiency in each case was approximately 0.0007. The results of these suicide experiments indicate that the killing efficiency of tritium disintegrations depends on where tritium is located within the cells. Tritium disintegrations in the nucleus are more effective in killing the cell than that in cytoplasm; and tritium disintegrations on DNA in the nucleus is more effective in killing the cell than that of nuclear RNA.  相似文献

Filtrates from microbial heterotrophic activity assay experiments in which tritiated glucose was used as a radioactively labeled substrate were collected, and their radioactivities were determined. These filtrates were subsequently evaporated to dryness to remove tritiated water generated by metabolism of the labeled glucose; the residue was suspended to original volume with distilled water, and the tritium levels were again assayed. In the water samples assayed, the amount of tritium label retained by the microorganism was about 75% of that respired.  相似文献

Samples of D-glucose and starch were labeled by tritium-atom bombardment. Up to 51% incorporation into D-glucose as non-labile tritium was achieved for crystalline, anhydrous D-glucose and 41% for the amylose-butyl alcohol complex. Distribution of tritium in the carbon skeleton of D-glucose was calculated by comparing the specific molar activity of D-glucose with that of its derivatives. Derivatives prepared were D-gluconic acid, D-arabino-hexulose phenylosotriazole, 4-formyl-2-phenyltriazole, 2-phenyltriazole-4-carboxylic acid, D-arabino-hexulose phenylflavazole, 3-formyl-1-phenylflavazole, and formaldehyde dimedone. The tritium distribution showed definite structural effects. Generally, the products from films of D-glucose and the amylose-butyl alcohol complex had nearly uniform distribution of tritium in D-glucose. The product from crystalline α-D-glucose monohydrate had zero tritium at C-2 and twice the expected amount of tritium at C-5, and that from starch granules had zero or near zero tritium at C-3 and close to twice the expected amount of tritium at C-2.  相似文献

Autoradiography was used to provide evidence for the transfer of Rhizobium produced moieties to the host nuclei of young clover nodule cells. Cells of Rhizobium trifolii Dangeard were labeled with 3H-adenine or 4,5-3H-l -leucine and 3,4-3H-l .-proline, washed free of external label, and allowed to nodulate young seedlings of white clover (Trifolium repens L. cv. White Dutch). Sections (0.5–1.0 μm) of young nodules up to four days old were autoradiographed using the dipping technique. Grain counts indicated movement of tritium from the leucine-proline labeled rhizobia to the polyploid nuclei of two day old clover nodule cells. Acetylene reduction was not detected until approximately 24 hours after the transfer of tritium was observed. No transfer of tritium was observed with 3H-adenine labeled rhizobia. It is hypothesized that nodulating rhizobia may induce clover nodule cells to initiate leghemoglobin synthesis by the transfer of a bacterially produced inducer.  相似文献

The synthesis of N-methanesulfonyl 16-phenoxy-ω-tetranor PGE2 carboxamide (sulprostone — CP-34,089/ZK-57,671) labeled with tritium and carbon-14 is described. Sulprostone labeled with tritium in the phenoxy moiety by means of catalytic hydrogenolysis was obtained in a 17% radiochemical yield with a specific activity of 1.0 Ci/mmol. The methanesulfonamide-14C derivative of sulprostone was prepared from methyl-14C iodide in an 11.8% radiochemical yield having a specific activity of 18.8 mCi/mmol.  相似文献

Light-and electron-microscopic autoradiography have been used to study fibroblast transformation into endothelial cells in the formation of new blood vessels during wound healing in rabbit ear chambers. When cultured fibroblasts labeled with tritium thymidine were transplanted autologously into the chambers, newly formed blood vessels contained endothelial cells labeled with tritium thymidine. This result suggests that fibroblasts play a pivotal role in angiogenesis, as progenitors of endothelial cells in newly formed blood vessels.  相似文献

The resolution obtainable in electron microscopic autoradiographs, using a photographic emulsion consisting of a monolayer of silver bromide crystals, was investigated theoretically and experimentally. The expected distribution of exposed crystals around a point source was calculated from the geometry of the preparation and from the range distribution of the beta particles emitted by tritium. From such a distribution an autoradiographic resolution of the order of 1000 A can be predicted. From the point source distribution, the expected distribution of grains around bacteriophages labeled with tritium was calculated. This distribution was also measured experimentally in electron microscopic autoradiographs of bacteriophages T-2 labeled with thymidine-H3. The two distributions agreed closely. It was also verified, using the nuclear region in thin cross-sections of Bacillus subtilis labeled with thymidine-H3, that resolutions of the same order were obtained for extended sources. It was concluded that an autoradiographic resolution of 1000 A could be achieved with a presently available commercial emulsion, although emulsions with finer grains might be desirable in some circumstances.  相似文献

Microenvironments of the three histidine residues located at the positions 18, 26, and 33 from the amino terminus in bovine heart cytochrome c were analysed in solution by the hydrogen-tritium exchange titration method, which has been developed in this laboratory. Histidine-18, which is liganded to the heme iron, and histidine-26 did not incorporate tritium in native state, indicating that the two are located in solvent inaccessible hydrophobic regions. Histidine-33 was labeled with tritium to an appreciable extent and seemed to be partially buried in the molecule. The pKa value estimated for histidine-33 was 6.1 at 37 degrees by the tritium exchange titration, suggesting that the residue interacts very weakly with a neighboring cationic group. These results seem to be compatible with the tertiary structure of the protein deduced from the X-ray crystallographic analysis.  相似文献

Radioimmunoassays were established for the modified nucleosides N-[9-(beta-D-ribofuranosyl)purin-6-ylcarbamoyl]-L-threonine, t6A, and 2-methylthioadenosine, ms2A. The assays depended on the production of antisera specific for t6A and ms2A that have not been previously reported. The nitrocellulose membrane filtration and saturated ammonium sulfate RIA techniques were compared for efficiency. Various radioactive antigens were employed to establish which type of antigen would give the best binding. The tritium post-labeling procedure of Randerath and Randerath was used to obtain labeled nucleosides of high enough specific activity to be useful for RIAs when the labeled nucleoside was not available commerically. The specificity of the antibodies toward nucleosides and purified tRNAs is reported. Although the titer of the t6A antiserum was low, the specificity was very sharp. An interesting finding was that threonine, a major structural component of the side-chain modification of t6A, was completely infective as an inhibitor.  相似文献

The metabolism of estradiol-17β is primarily an oxidative process at either carbon-2 or carbon-16 in the human. The objective of this study was to determine the relative importance of these two oxygenation pathways in the chimpanzee. The rate of oxidation of estradiol-17β at each position was determined by measuring the release of tritium into body water from carbon-2 or carbon-16. [2-3H]-Estradiol-17β or [16-3H]-estradiol-17β was injected intravenously into three adult male chimpanzees, and blood samples were obtained at several time intervals between 1 and 48 hr. The blood was lyophilized, and the release of tritium from the specifically labeled estrogens into the body fluid pool was determined. The release of tritium from the 16α-position was very low and did not exceed 3% in any animal. The release of tritium from the carbon-2 was much faster, amounting to 29%, 34%, and 35%, respectively, by 24 hr. The ratio of tritium released from carbon-2:carbon-16 was 5.0, 13.2 and 16.9, respectively, at 24 hr after injection of the specifically labeled estradiol-17β. These results demonstrate clearly that the major pathway for oxidative metabolism of estradiol-17β in the chimpanzee is via oxygenation at carbon-2, with the formation of catechol estrogens, as in the human.  相似文献

Lysozyme has been tritiated by the free-radical interceptor method, which involves reaction of the carbon free radicals, resulting from gamma-irradiation of the lyophilized protein in a vacuum, with tritiated hydrogen sulfide. This technique resulted in a high yield of tritiated enzyme which was chromatographically indistinguishable from the native protein. To characterize this material with respect to its tritium distribution, it was first reduced, carboxymethylated, and digested with chymotrypsin. The peptides were separated and purified by ion exchange chromatography and electrophoresis and then analyzed to determine the specific activities of 78 of the 129 residues in the protein chain. The tritium was found to be broadly distributed, with only 12 residues apparently devoid of label. The relative specific activity, defined as the ratio of the specific activity of a given residue to the average specific activity for all residues of the same kind within the protein, did not exceed the value of 3.73 for isoleucine at position 88. A distinct spatial relationship is seen among the more heavily labeled residues when these results are related to the x-ray diffraction model of lysozyme. Thus, residues 16, 18, 19, 21, 23, 29, 31, and 32 may be grouped with 105, 110, 113, and 115; these highly labeled residues are close to a strongly hydrophobic region. Another heavily labeled area involves residues 50, 54, 57. 83, 84, and 88. In addition, residues 1 and 6 at the amino end of the chain are heavily labeled, as are 118, 121, and possibly 119 at the carboxyl end. This distribution of tritium appears to reflect the secondary free-radical distribution in the irradiated protein. These observations confirm previous indications that the secondary distribution is influenced by the conformation of the protein molecule.  相似文献

Two methods were investigated for studying the binding of radiolabeled hybridoma T cells to antigen (Ag) and H-2 products for which they bore receptors. In both cases hybridoma T cells were labeled with tritiated thymidine. In one method labeled cells were added to adherent splenic cells prepulsed with antigen, and the mixture was incubated overnight at 37 degrees C before nonadherent cells were gently washed away. The percent of adherent hybridoma T cells was then estimated by harvesting the adherent monolayers and measuring tritium counts bound. In a second method radiolabeled hybridoma T cells were added to adherent antigen-pulsed B cell lymphomas or hybridomas for between 15 min and 1 hr at 37 degrees C before removal of nonadherent cells and harvesting of the adherent monolayers. In both cases binding was both antigen- and I-region specific. In the second case binding was also rapid; significant binding could be measured after 15 min incubation. These techniques were used to study subclones of one of our T cell hybridomas that were thought by a functional assay (interleukin 2 release) to have lost receptors for Ag/H-2. It was found that subclones of the hybridoma that no longer secreted interleukin 2 in response to Ag/H-2, even though they continued to secrete interleukin 2 in response to concanavalin A, also no longer bound specifically to Ag-pulsed monolayers of the appropriate H-2 type. This confirmed the idea that these subclones had lost the ability to synthesize receptors for Ag/H-2. It is hoped that assays of this type will be useful in the future for the study of Ag/H-2 receptors on T cells.  相似文献

Biologically active peptides evenly labeled with tritium were used for studying the in vitro and in vivo biodegradation of the peptides. Tritium-labeled peptides with a specific radioactivity of 50–150 Ci/mmol were obtained by high temperature solid phase catalytic isotope exchange (HSCIE) with spillover tritium. The distribution of the isotope label among all amino acid residues of these peptides allows the simultaneous determination of practically all possible products of their enzymatic hydrolysis. The developed analytical method includes extraction of tritium-labeled peptides from organism tissues and chromatographic isolation of individual labeled peptides from the mixture of degradation products. The concentrations of a peptide under study and the products of its biodegradation were calculated from the results of liquid scintillation counting. This approach was used for studying the pathways of biodegradation of the heptapeptide TKPRPGP (Selank) and the tripeptide PGP in blood plasma. The pharmacokinetics of Selank, an anxiolytic peptide, was also studied in brain tissues using the intranasal in vivo administration of this peptide. The concentrations of labeled peptides were determined, and the pentapeptide TKPRP, tripeptide TKP, and dipeptides RP and GP were shown to be the major products of Selank biodegradation. The study of the biodegradation of the heptapeptide MEHFPGP (Semax) in the presence of nerve cells showed that the major products of its biodegradation are the pentapeptide HFPGP and tripeptide PGP. The enkephalinase activity of blood plasma was studied with the use of evenly tritium labeled [Leu]enkephalin. A high inhibitory effect of Semax on blood plasma enkephalinases was shown to arise from its action on aminopeptidases. The method, based on the use of evenly tritium-labeled peptides, allows the determination of peptide concentrations and the activity of enzymes involved in their degradation on a μg scale of biological samples both in vitro and in vivo.  相似文献

A method for the calibration of plastic tritium standards for use with LKB Ultrofilm is described and validated. This method uses 12-microns cryostat slices of frozen liver which have been labeled with [3H]-formaldehyde and extracted with chloroform-methanol to remove lipids. Quantitative autoradiographic measurement of 3H radioactivity in the liver slices was underestimated by 35% when lipids were not extracted. Plastic sections impregnated with tritium were calibrated in terms of lipid-extracted, tissue-equivalent radioactivity content. Calibrated standard curves for these plastic standards were closely fit (p = 0.99) by second order polynomial equations for exposures of 1, 4, 7, 13, 28, and 104 days. The equations are generally useful for any plastic tritium standards.  相似文献

Tritiated adenosylcobalamin, labeled at the exchangeable position, has been used to investigate the partitioning of tritium between substrate and product in the reaction catalyzed by glutamate mutase. The isotope partitions between glutamate and methylaspartate in nearly 1:1 ratio, regardless of the direction in which the overall reaction is proceeding. This is consistent with a free-energy profile in which the interconversion of the intermediate glutamyl and methylaspartyl radicals is rapid relative to the transfer of tritium from 5'-deoxyadenosine to either substrate or product. Initial velocity measurements have been used to measure the tritium isotope effects for the transfer of tritium from adenosylcobalamin to product in each direction. The isotope effect is 21 for the formation of glutamate and 19 for the formation of methylasparate. The large magnitude of these isotope effects makes it likely that the rate-determining step may be altered by the substitution of tritium for hydrogen in the reaction. The results of these experiments are compared with previous isotope effect measurements made on other adenosylcobalamin-dependent enzymes.  相似文献

Release of 3H2O from regiospecifically labeled estradiol was measured during 2-hydroxylation of this estrogen by rat liver microsomes. The amount of tritium remaining in the isolated catechol estrogen was also determined. Virtually all the tritium was removed from C-2 during the reaction confirming the absence of an NIH shift. About 20% of the tritium at C-1 was also lost without any such change occurring at C-4 or C-6,7 of the steroid molecule. These findings provide evidence for the formation of an arene oxide or o-semiquinone intermediate during the conversion of estradiol to 2-hydroxyestradiol. No indication of adduct formation at either C-1 or C-4 during this biotransformation was obtained although the 2-hydroxylated product was able to react with a nucleophile such as glutathione. The different regiospecificity of tritium loss in the generation of catechol estrogens and in their subsequent reaction leads to the important conclusion that the reactive intermediates in the two processes must be different. The possible role of catechol estrogens in neoplastic transformation is discussed.  相似文献

For deeper understanding the roles of the mRNA cap structure in cellular processes isotopically labeled dinucleotide cap analogues have been synthesized as tools for NMR and in vivo studies. Tritium or carbon C-13 labeled methyl iodide was used as a source of the isotope material. In order to minimize the number of steps during the radioisotopic synthesis the methylation with tritium labeled methyl iodide was performed with Gp(3)G as a substrate. The C-13 isotope was introduced into the cap dinucleotide by methylation of GDP with C-13 Methyl iodide, followed by coupling the product with guanosine 5'-phosphorimidazolide in DMF with zinc chloride as a catalyst.  相似文献
