Netrium digitus is a representative of the species-rich class Zygnematophyceae (Streptophyta). Its intensive extracellular polysaccharide (EPS) production makes this alga interesting for biotechnological applications with a focus on cosmetics and food additives. Quantitative data on growth and EPS production in suspension and, for the first time, in immobilized culture using lab-scale porous substrate bioreactors, so-called Twin-Layer (TL) systems, is presented. It is shown that the cell as well as the EPS dry weight content is increased at least sixfold in immobilized compared to suspension culture. Due to the high amount of EPS, the biofilms reach a thickness of more than 8 mm after 27 days at 70 μmol photons m?2 s?1 and with 1.5% CO2 supply. Frequent exchange of the growth medium results in a linear cell biomass increase of 2.02?±?0.09 g m?2 growth area day?1 compared to 2.99?±?0.09 g m?2 day?1, when the medium is not exchanged. Under this mode of cultivation, the EPS production is lower and a final concentration of 12.18?±?1.25 g m?2 compared to 20.76?±?0.85 g m?2, when medium was exchanged, is reached. It is clearly demonstrated that the relatively slow growing, but excessively EPS producing, microalgal species N. digitus can be grown in porous substrate bioreactors and that this culturing technique is a promising alternative to suspension culture for the Zygnematophyceae.  相似文献

Gordonia polyisoprenivorans CCT 7137 was isolated from groundwater contaminated with leachate in an old controlled landfill (São Paulo, Brazil), and cultured in GYM medium at different concentrations of sugarcane molasses (2%, 6%, and 10%). The strain growth was analyzed by monitoring the viable cell counts (c.f.u. mL?1) and optical density and EPS production was evaluated at the end of the exponential phase and 24 h after it. The analysis of the viable cell counts showed that the medium that most favored bacterial growth was not the one that favored EPS production. The control medium (GYM) was the one that most favored the strain growth, at the maximum specific growth rate of 0.232 h?1. Differences in bacterial growth when cultured at three different concentrations of molasses were not observed. Production of EPS, in all culture media used, began during the exponential phase and continued during the growth stationary phase. The highest total EPS production, after 24 h of stationary phase, was observed in 6% molasses medium (172.86 g L?1) and 10% (139.47 g L?1) and the specific total EPS production was higher in 10% molasses medium (39.03 × 10?11 g c.f.u.?1). After the exponential phase, in 2%, 6%, and 10% molasses media, a higher percentage of free exopolysaccharides (EPS) was observed, representing 88.4%, 62.4%, and 64.2% of the total, respectively. A different result was observed in pattern medium, which presented EPS made up of higher percentage of capsular EPS (66.4% of the total). This work is the first study on EPS production by G. polyisoprenivorans strain in GYM medium and in medium utilizing sugarcane molasses as the sole nutrient source and suggests its potential use for EPS production by G. polyisoprenivorans CCT 7137 aiming at application in biotechnological processes.  相似文献

Studies were performed on the effect of CaCO3 and CaCl2 supplementation to fermentation medium for ethanol production from xylose, glucose, or their mixtures using Scheffersomyces (Pichia) stipitis. Both of these chemicals were found to improve maximum ethanol concentration and ethanol productivity. Use of xylose alone resulted in the production of 20.68 ± 0.44 g L?1 ethanol with a productivity of 0.17 ± 0.00 g L?1 h?1, while xylose plus 3 g L?1 CaCO3 resulted in the production of 24.68 ± 0.75 g L?1 ethanol with a productivity of 0.21 ± 0.01 g L?1 h?1. Use of xylose plus glucose in combination with 3 g L?1 CaCO3 resulted in the production of 47.37 ± 0.55 g L?1 ethanol (aerobic culture), thus resulting in an ethanol productivity of 0.39 ± 0.00 g L?1 h?1. These values are 229 % of that achieved in xylose medium. Supplementation of xylose and glucose medium with 0.40 g L?1 CaCl2 resulted in the production of 44.84 ± 0.28 g L?1 ethanol with a productivity of 0.37 ± 0.02 g L?1 h?1. Use of glucose plus 3 g L?1 CaCO3 resulted in the production of 57.39 ± 1.41 g L?1 ethanol under micro-aerophilic conditions. These results indicate that supplementation of cellulosic sugars in the fermentation medium with CaCO3 and CaCl2 would improve economics of ethanol production from agricultural residues.  相似文献

The fatty acid composition, the effect of different concentrations of nitrogen (16.5-344 mg ?L?1), phosphorus (9–45 mg? L?1), iron (9–45 mg? L?1) and salinity levels (0–20 psu) on lipid production in the green microalga Scenedesmus dimorphus KMITL, a new strain isolated from a tropical country, Thailand, were studied. The alga was isolated from a freshwater fish pond, and cultured in Chlorella medium by varying one parameter at a time. The main fatty acid composition of this strain was C16–C18 (97.52 %) fatty acids. A high lipid content was observed in conditions of 16.5 mg? L?1-N, or 22 mg ?L?1-P, or 45 mg ?L?1-Fe, or 5 psu salinity, which accumulated lipids to 20.3?±?0.4, 19.4?±?0.2, 24.7?±?0.5, and 14.3?±?0.2 % of algal biomass, respectively. Increasing lipid content and lipid productivity was noted when the alga was cultured under high iron concentration and high salinity, as well as under reduced phosphorus conditions, whereas nitrogen limitation only resulted in an increased lipid content.  相似文献

A new kind of extracellular polysaccharide (EPS) from Pseudomonas fluorescens PGM37 was obtained and culture media was optimized using the statistical methods single factor experiments and response surface methodology (RSM) design. As a result, the optimum cultivation conditions initial pH value, medium volume, inoculum size, temperature, and rotation speed were 7.5, 100 mL/250 mL, 5 %, 28 °C, and 180 rpm, respectively. The optimized media: sucrose 36.23 g?L?1, yeast extract 3.32 g?L?1, sodium chloride 1.13 g?L?1, and calcium chloride 0.20 g?L?1. The maximum predicted yield of EPS was 10.1163 g?L?1 under these conditions. The validation data was 10.012 g?L?1, which could strongly confirm the correlation between the experimental and theoretical values. Gas Chromatography analysis revealed that the polymer was made up of mannose and glucose in the ratio of 1:1. Infrared spectroscopy showed that the polysaccharide had β-D-pyranoid configuration and contained no other substituent. Graded by different multiples of alcohol after specific degradation by enzyme and then detected by LC-ESI-MS, the EPS structure was β-D-Glcp-(1, 4)-β-D-Manp-(1, 4)-β-D-Glcp-(1, 4)-β-D-Manp. The moisture retention ability of the EPS was found to be superior to glycerol and only a little inferior to hyaluronic acid (HA), which presented potential application value in cosmetics and clinical medicine fields.  相似文献

The present research focused on enhancing the production of wedelolactone through cell suspension culture (CSC) in Eclipta alba (L.) Hassk. With an aim of attaining a sustainable CSC, various plant growth regulators, elicitors and agitation speed were examined. Nodal segments of in vitro propagated plantlets induced the maximum percentage (93.47?±?0.61%) of callus inoculated on Murashige and Skoog (MS) medium fortified with picloram (2 mg L?1). The growth kinetics of CSC exhibited a sigmoid pattern with a lag phase (0–6 days), a log phase (6–18 days), a stationary phase (18–24 days) and then death phase thereafter. The highest biomass accumulation in CSC with 7.09?±?0.06 g 50 mL?1 fresh weight, 1.52?±?0.02 g 50 mL?1 dry cell weight, 1.34?±?0.01?×?106 cell mL?1 total cell count and 57.00?±?0.58% packed cell volume was obtained in the liquid MS medium supplemented with 1.5 mg L?1 picloram plus 0.5 mg L?1 kinetin at 120 rpm. High performance thin layer chromatography confirmed that yeast extract (biotic elicitor) at 150 mg L?1 accumulated more CSC biomass with 1.22-fold increase in wedelolactone (288.97?±?1.94 µg g?1 dry weight) content in comparison to the non-elicited CSC (237.78?±?0.04 µg g?1 dry weight) after 120 h of incubation. Contrastingly, methyl jasmonate (abiotic elicitor) did not alter the biomass but increased the wedelolactone content (259.32?±?1.06 µg g?1 dry weight) to an extent of 1.09-fold at 100 µM. Complete plantlet regeneration from CSC was possible on MS medium containing N6-benzyladenine (0.75 mg L?1) and abscisic acid (0.5 mg L?1). Thus, the establishment of protocol for CSC constitutes the bases for future biotechnological improvement studies in this crop.  相似文献

Burkholderia sp. F24, originally isolated from soil, was capable of growth on xylose and removed organic inhibitors present in a hemicellulosic hydrolysate and simultaneously produced poly-3-hydroxybutyrate (P3HB). Using non-detoxified hydrolysate, Burkholderia sp. F24 reached a cell dry weight (CDW) of 6.8 g L?1, containing 48 % of P3HB and exhibited a volumetric productivity (PP3HB) of 0.10 g L?1 h?1. Poly-3-hydroxybutyrate-co-3-hydroxyvalerate copolymers (P3HB-co-3HV) were produced using xylose and levulinic acid (LA) as carbon sources. In shake flask cultures, the 3HV content in the copolymer increased from 9 to 43 mol% by adding LA from 1.0 to 5.0 g L?1. In high cell density cultivation using concentrated hemicellulosic hydrolysate F24 reached 25.04 g L?1 of CDW containing 49 % of P3HB and PP3HB of 0.28 g L?1 h?1. Based on these findings, second-generation ethanol and bioplastics from sugarcane bagasse is proposed.  相似文献

In vitro plantlets of sugarcane cultivar NCo310 were maintained in slow growth conditions at both 18 and 24°C and on four semi-solid media: SG1—Murashige and Skoog (MS) salts and vitamins with 20 g L?1 sucrose, SG2—½ MS with 10 g L?1 sucrose, SG3—MS with 20 g L?1 sucrose and 1 mg L?1 abscisic acid (ABA), and SG4—½ MS with 10 g L?1 sucrose and 1 mg L?1 ABA. After 8, 12, 24, 36, and 48 mo shoot multiplication rates were recorded, shoots were removed from storage and subcultured every 2 wk on SG1 with 0.015 mg L?1 kinetin and 0.1 mg L?1 benzyl aminopurine for 2 mo. At 18°C, all media supported storage for 48 mo with subculturing every 12 mo. Shoot multiplication post-retrieval was significantly higher on the SG2 medium compared with the non-stored control (362 ± 84 and 126 ± 26 shoots per recovered shoot after 2 mo, respectively). In addition, shoots could be maintained for 48 mo on SG2 medium with one subculture without compromising post-storage multiplication ability. At 24°C, storage on all four media supported recovery and multiplication of shoots for 8 mo and only SG2 medium facilitated survival for 12 mo. There was no advantage to incorporating ABA into the storage media, regardless of the temperature and storage time. Cryopreservation of cultivar NCo376 in vitro-derived shoot meristems using the V-cryo-plate method demonstrated that the sucrose concentration in the loading solution (0.8–1.8 M) had no significant effect on survival of the meristems, which ranged from 41.7 ± 4.8 to 69.4 ± 10%.  相似文献

In this study, after the expression of a pyruvate carboxylase gene (PYC) cloned from Meyerozyma guilliermondii in a marine-derived yeast Yarrowia lipolytica SWJ-1b, a transformant PG86 obtained had much higher PYC activity than Y. lipolytica SWJ-1b. At the same time, the PYC gene expression and citric acid (CA) production by the transformant PG86 were also greatly enhanced. When glucose concentration in the medium was 60.0 g L?1, CA concentration formed by the transformant PG86 was 34.02 g L?1, leading to a CA yield of 0.57 g g?1 of glucose. During a 10-L fed-batch fermentation, the final concentration of CA was 101.0 ± 1.3 g L?1, the yield was 0.89 g g?1 of glucose, the productivity was 0.42 g L?1 h?1 and only 5.93 g L?1 reducing sugar was left in the fermented medium within 240 h of the fed-batch fermentation. The present work evaluated biomass productivity, carbon dioxide fixation rate, and biochemical composition of two microalgal species, Phaeodactylum tricornutum (Bacillariophyta) and Tetradesmus obliquus (Chlorophyta), cultivated indoors in high-technology photobioreactors (HT-PBR) and outdoors both in pilot ponds and low-technology photobioreactors in a greenhouse in southern Italy. Microalgae were grown in standard media, under nitrogen starvation, and in two liquid digestates obtained from anaerobic digestion of agro-zootechnical and vegetable biomass. P. tricornutum, cultivated in semi-continuous mode in indoor HT-PBRs with standard medium, showed a biomass productivity of 21.0?±?2.3 g m?2 d?1. Applying nitrogen starvation, the lipid productivity increased from 2.3 up to 4.5?±?0.5 g m?2 d?1, with a 24 % decrease of biomass productivity. For T. obliquus, a biomass productivity of 9.1?±?0.9 g m?2 d?1 in indoor HT-PBR was obtained using standard medium. Applying liquid digestates as fertilizers in open ponds, T. obliquus gave a biomass productivity (10.8?±?2.0 g m?2 d?1) not statistically different from complete medium such as P. tricornutum (6.5?±?2.2 g m?2 d?1). The biochemical data showed that the fatty acid composition of the microalgal biomass was affected by the different cultivation conditions for both microalgae. In conclusion, it was found that the microalgal productivity in standard medium was about doubled in HT-PBR compared to open ponds for P. tricornutum and was about 20 % higher for T. obliquus.  相似文献

In the present paper, we investigated the physiological response of the marine microalga Nannochloropsis sp. to salt stress (13, 27, 54, and 81 g L?1 NaCl). Increasing the sodium chloride concentration caused up to a 70 % decrease in the chlorophyll a concentration, cell growth, and net photosynthesis rate. The chlorophyll a fluorescence measurements indicated a strong reduction in the effective quantum yield of photosystem II (?60 %) and an increase in nonphotochemical quenching when the cells were exposed to NaCl concentrations greater than 27 g L?1 (control). In contrast, the specific lipid content increased up to 80 % when the sodium chloride concentration was increased from 27 to 54–81 g L?1. These results are relevant for the outdoor cultivation of this microalga using open photobioreactors, in which microalgae are subjected to strong changes in salinity concentration caused by water evaporation.  相似文献

Microalgae are considered as a promising feedstock for biomass production. The selection of the most suitable species is based on several key parameters such as lipid and fatty acid productivity. In the present study, the growth of different microalgae strains was examined in freshwater media for photoautotrophs suited for large-scale applications to identify the most suitable medium for each species. In the optimal medium, Scenedesmus obliquus showed the highest biomass productivity measured as increase of cell dry weight (0.25 g cellu dry weight (CDW) L?1 day?1), while Botryococcus braunii showed the highest lipid and total fatty acid content (430 and 270 mg g?1 CDW, respectively) among the tested species. Regarding lipid and total fatty acid productivity, S. obliquus was the most lipid and total fatty acid productive strain with 41 and 18 mg L?1 day?1 during the exponential phase, respectively. Additionally, the proportion of saturated and monounsaturated fatty acids increased with duration of the incubation in S. obliquus, while polyunsaturated fatty acids decreased. These results nominate S. obliquus as a promising microalga in order to serve as a feedstock for renewable energy production.  相似文献

Microalgal starch is a potential feedstock for biofuel production. Nutrient stress is widely used to stimulate starch accumulation in microalgae. Cell growth and starch accumulation in the marine green microalga Tetraselmis subcordiformis were evaluated under extracellular phosphorus deprivation with initial cell densities (ICD) of 1.5, 3.0, 6.0, and 9.0?×?106 cells mL?1. The intracellular stored phosphorus supported cell growth when extracellular phosphorus was absent. The maximum starch content of 44.1 % was achieved in the lowest ICD culture, while the maximum biomass productivity of 0.71 g L?1 day?1, starch concentration of 1.6 g L?1, and starch productivity of 0.30 g L?1 day?1 were all obtained in the culture with the ICD of 3.0?×?106 cells mL?1. Appropriate ICD could be used to regulate the intracellular phosphorus concentration and maintain adequate photosynthetic activity to achieve the highest starch productivity, along with biomass and starch concentration. The recovery of phosphorus-deprived T. subcordiformis in medium containing 0.5, 1.0, or 6.0 mM KH2PO4 was also tested. Cell growth and starch accumulation ability could be recovered completely. A phosphorus pool in T. subcordiformis was shown to manipulate its metabolic activity under different environmental phosphorus availability. Though lower starch productivity and starch content were achieved under phosphorus deprivation compared with nitrogen- or sulfur-deprived conditions, the higher biomass and starch concentration make T. subcordiformis a good candidate for biomass and starch production under extracellular phosphorus deprivation.  相似文献

Microsclerotia (MS) formation was successfully induced in Nomuraea rileyi (Ascomycetes: Hypocreales) in liquid amended medium (AM) culture. To improve MS production, the culture medium was varied. Three quantitative variables, carbon, nitrogen, and basal salt sources, were investigated using the AM. An orthogonal experiment [L9 (3)3] design was applied to optimize the culture medium. Maximum MS production (21.9 × 104 MS ml?1) was obtained using the optimized culture medium (32.0 g l?1 of glucose, 2.0 g l?1 of ammonium citrate, and 0.15 g l?1 of ferrous sulfate). Subsequently, viability, including thermotolerance, shelf-life, and infection, were investigated to evaluate the potential of MS for use as a mycoinsecticide. The MS, collected from the optimized culture medium, exhibited virulence and greater thermotolerance than conidia, and maintained 86.4 % germinability after one-year room temperature storage. These results suggested that the MS obtained through optimization production can be used as a mycoinsecticide.  相似文献

The optimal cultivation conditions ensuring the maximal rate of citric acid (CA) biosynthesis by glycerol-grown mutant Yarrowia lipolytica NG40/UV7 were found to be as follows: growth limitation by inorganic nutrients (nitrogen, phosphorus, or sulfur), 28 °C, pH 5.0, dissolved oxygen concentration (pO2) of 50 % (of air saturation), and pulsed addition of glycerol from 20 to 80 g L?1 depending on the rate of medium titration. Under optimal conditions of fed-batch cultivation, in the medium with pure glycerol, strain Y. lipolytica NG40/UV7 produced 115 g L?1 of CA with the mass yield coefficient of 0.64 g g?1 and isocitric acid (ICA) amounted to 4.6 g L?1; in the medium with raw glycerol, CA production was 112 g L?1 with the mass yield coefficient of 0.90 g g?1 and ICA amounted to 5.3 g L?1. Based on the activities of enzymes involved in the initial stages of raw glycerol assimilation, the tricarboxylic acid cycle and the glyoxylate cycle, the mechanism of increased CA yield from glycerol-containing substrates in Y. lipolytica yeast was explained.  相似文献

A rotational reciprocating plate impeller prototype, designed to improve the mixing homogeneity of viscous non-Newtonian fermentation broth, has been tested in pullulan fermentations. With this new impeller, the operating levels of several factors were investigated to improve pullulan production with Aureobasidium pullulans ATCC 42023 in a 22-L bioreactor using experimental designs. Because both high molecular weight (MW) and high concentration of pullulan were desired; the exopolysaccharide (EPS) concentration and the broth viscosity were used as optimization objective functions to be maximized. A 6-run uniform design was used to investigate five factors. Under the best operating conditions among the six runs, 29.0 g L?1 EPS was produced at 102 h. This condition was used as the starting point for further investigation on the two statistically significant factors, the pH and the agitation speed. An 8-run 3-level custom design that investigates up to second-order effects was used in the second stage. An optimal zone of operating conditions for large quantity of high MW pullulan production was identified. A concentration of 23.3 g L?1 EPS was produced at 78 h. This is equivalent to an EPS productivity of 0.30 g L?1 h?1. The corresponding apparent viscosity of the broth was 0.38 Pa s at the shear rate of 10 s?1.  相似文献

Clostridium beijerinckii optinoii is a Clostridium species that produces butanol, isopropanol and small amounts of ethanol. This study compared the performances of batch and continuous immobilized cell fermentations, investigating how media flow rates and nutritional modification affected solvent yields and productivity. In 96-h batch cultures, with 80 % of the 30 g L?1 glucose consumed in synthetic media, solvent concentration was 9.45 g L?1 with 66.0 % as butanol. In a continuous fermentation using immobilized C. beijerinckii optinoii cells, also with 80 % of 30 g L?1 glucose utilization, solvent productivity increased to 1.03 g L?1 h?1. Solvent concentration reached 12.14 g L?1 with 63.0 % as butanol. Adjusting the dilution rate from 0.085 to 0.050 h?1 to allow extended residence time in column was required when glucose concentration in fresh media was increased from 30 to 50 g L?1. When acetate was used to improve the buffer capacity in media, the solvent concentration reached 12.70 on 50 g L?1 glucose. This continuous fermentation using immobilized cells showed technical feasibility for solvent production.  相似文献

In this study, hypersaline media were used for ocean cultivation of the marine microalga Tetraselmis sp. KCTC12432BP for enhanced biomass and fatty acid (FA) productivity. Hypersaline media (55, 80, and 105 PSU) were prepared without sterilization by addition of NaCl to seawater obtained from Incheon, Korea. The highest biomass productivity was obtained at 55 PSU (0.16 g L?1 day?1) followed by 80 PSU (0.15 g L?1 day?1). Although the specific growth rate of Tetraselmis decreased at salinities higher than 55 PSU, prevention of contamination led to higher biomass productivity at 80 PSU than at 30 PSU (0.03 g L?1 day?1). FA content of algal biomass increased as salinity increased to 80 PSU, above which it declined, and FA productivity was highest at 80 PSU. Ocean cultivation of Tetraselmis was performed using 50-L tubular module photobioreactors and 2.5-kL square basic ponds, closed- and open-type ocean culture systems, respectively. Culturing microalgae in hypersaline medium (80 PSU) improved biomass productivities by 89 and 152% in closed and open cultures, respectively, compared with cultures with regular salinity. FA productivity was greatly improved by 369% in the closed cultures. The efficacy of salinity shift and N-deficiency to enhance FA productivity was also investigated. Lowering salinity to 30 PSU with N-starvation following cultivation at 80 PSU improved FA productivity by 19% in comparison with single-stage culture without N-deficiency at 30 PSU. The results show that salinity manipulation could be an effective strategy to improve biomass and FA productivity in ocean cultivation of Tetraselmis sp.  相似文献

An improved protocol for plant regeneration via somatic embryogenesis was developed using mature macaw palm (Acrocomia aculeata) zygotic embryos as initial explant. For induction of the embryogenic callus (EC), two basic media (BM) were tested consisting of Murashige and Skoog and Eeuwens (Y3) salts with 30 g L?1 sucrose, 0.5 g L?1 glutamine and 2.5 g L?1 Phytagel. The 3,6-dichloro-2-methoxybenzoic acid (dicamba), 4-amino-3,5,6-trichloro-picolinic acid (picloram) and 2,4-dichlorophenoxyacetic acid (2,4-D) auxins were added to the culture media at concentrations of 0, 1.5 or 3.0 mg L?1. After 240 days, the embryogenic calli were transferred to the respective BM media with auxin concentrations reduced to 0.5 or 1.0 mg L?1 in order to differentiate the somatic embryos (SEs). Plant regeneration was performed on the BM media without growth regulators. Embryogenic calli were observed after 180 days of culture and in all treatments with auxin. The Y3 medium showed the best EC formation results (60.8 %). These calli showed yellowish coloration, compact consistency and nodular aspect. After 60 days in differentiation medium, SEs were verified in different stages of development. Histological analysis showed that the SEs were formed from a nodular EC. The SEs generally presented unicellular origin with suspensor formation, and at the end of development, bipolar embryos were observed. The plant regeneration frequency reached levels up to 31.9 % when using induction medium consisting of Y3 associated to 1.5 mg L?1 of 2,4-D and the subsequent auxin reduction to 0.5 mg L?1 in the differentiation stage. Regenerated plants showed normal development, with root and aerial part growth.  相似文献
