Bombesin was microinjected into the ventricular system of a teleost fish, Catostomus commersoni, in order to evaluate its effects on behavioral thermoregulation in a horizontal thermal gradient. General locomotor activity was also examined. Results demonstrated that bombesin decreased the behaviorally selected internal body temperatures by 5.0–7.0°C relative to controls and that the latency of the response was dose-dependent. The peptide did not change locomotor activity. Results are discussed in relation to body temperature setpoint(s) and possible implications for endotherm thermoregulation.  相似文献

Ascorbic acid, at concentrations below that normally present in the brain, inhibited the dopamine-sensitive adenylate cyclase invitro. Ascorbate had no effect on the norepinephrine-sensitive adenylate cyclase. To study the invivo effect of ascorbic acid on central dopaminergic systems, mice (C57 B1/6J) were injected with pharmacological doses (2 g/kg) of ascorbate, which produced a significant elevation in brain ascorbate concentration. Injecting the mice with ascorbate (2 g/kg) blocked the amphetamine-induced (15 mg/kg) increase in stereotype behavior which has been reported to be mediated by dopaminergic neural systems. Ascorbate had no effect on the amphetamine-induced locomotor activity thought to be mediated by norepinephrine systems. Ascorbate (1 g/kg) attenuated apmorphine-induced hypothermia in this same strain of mice. This demonstrates the specific neurochemical, physiological, and behavioral alterations in dopaminergic systems produced by ascorbic acid and suggests possible therapeutic uses for ascorbate in conditions involving functional dopamine excess.  相似文献

Eleven cyclic Hereford heifers were synchronized to estrus with a commercial luteolytic agent. Eight heifers had been inoculated with Anaplasmamarginale. Six of the eight inoculated heifers became anemic, i.e. packed cell volume, red blood cell counts and hemoglobin decreased. White blood cell counts were elevated during severe anemia. Anemic heifers did not show behavioral estrus or ovulate, and during this stage, serum concentrations of progesterone and estradiol-17β were significantly depressed. Mature beef heifers that have acute anaplasmosis suffer from inhibited ovarian function and anestrus.  相似文献

The development of tolerance to caffeine-induced stimulation of locomotor activity was evaluated in rats maintained chronically on average daily doses of 160 mg/kg or more of caffeine by the method of scheduled access to drinking water containing the drug. Dose-response curves were determined for caffeine (6.25–100 mg/kg) and d-amphetamine (0.39–6.4 mg/kg) during chronic drug treatment. In addition, the caffeine curve was redetermined 2–3 weeks after removal of the drug from the drinking water. A control group that had scheduled access to drug-free tap water was also tested. Caffeine produced dose-related increases in the locomotor activity of the controls but failed to modify locomotor activity of the chronic caffeine group. In contrast, d-amphetamine increased locomotor activity of both groups comparably. Spontaneous locomotor of the chronic caffeine group was reduced significantly for 4 days after drug-free tap water was substituted for the caffeine solution. The return of spontaneous locomotor activity to baseline values was associated with restored sensitivity to caffeine-induced stimulation of locomotor activity. Thus, chronic administration of caffeine to rats results in the development of tolerance to caffeine-induced stimulation of locomotor activity that is virtually complete, pharmacologically specific, and fully reversible when drug treatment is stopped. Decreases in spontaneous locomotor activity after abrupt termination of chronic caffeine administration follow a time course consistent with a drug withdrawal syndrome.  相似文献

The transition from embryonic to adult l-glycerol 3-phosphate dehydrogenase (EC 1.1.1.8) was examined in reaggregating cell cultures of mouse brain. Culture conditions were selected that enabled viable aggregates to be produced from neural cells of mice that ranged in age from the 12th day of gestation to 6 days of postnatal age. In all cultures, an increase in the specific and total activity of l-glycerol 3-phosphate dehydrogenase was observed and this was due to an increase in the adult isozyme. The cultures from the older cells, i.e., cerebellar aggregates from mice 6 days of age, expressed approximately a 50-fold increase in activity, whereas cerebellar aggregates from newborn mice exhibited about a three-fold increase in activity. In both cases, the time course for the developmental increase followed the normal temporal sequence. The amount of l-glycerol 3-phosphate dehydrogenase activity in a cerebellar aggregate from a 6-day-old mouse was greater than 1% of the soluble cellular protein, which is about seven times higher than the specific activity determined in the cerebellums of adult mice. Alleles at the structural locus for l-glycerol 3-phosphate dehydrogenase, the Gdc-1 locus, and those alleles that control the activity levels in the cerebellums of mice were expressed in aggregates from BALBcBy and C57BL6J neonates. However, although the expected structural allele was expressed in all cultures, the expression of activity differences in cerebellar aggregates depended on the age of the mouse from which the cerebellar cells were isolated. Acetylcholinesterase (EC 3.1.1.7) was also measured in these aggregates; the specific activities were highest in aggregates from mice in the 16th day of gestation and least in the cerebellar aggregates of neonates, a trend that was opposite to that of l-glycerol 3-phosphate dehydrogenase.  相似文献

There is a bimodal daily rhythm of plasma corticosterone binding activity (CBA) in the white-throated sparrow, Zonotrichiaalbicollis, in the spring migratory condition. Low levels occurred at the beginning (0500) and at the end (2100) of a 16-hour daily photoperiod. Peak CBA occurred at 0900 and 0100, as did peak locomotor activity in this nocturnal migrant. Comparisons of CBA with total plasma corticosteroid concentrations from a previous study of the same group of birds indicate a positive correlation during most of the day but not during the early hours of darkness. The daily rhythm of locomotor activity may account for the rhythm of CBA which, in turn, may be partially responsible for the daily rhythm of plasma corticosteroid concentration.  相似文献

Reaggregating cell cultures of neonatal mouse cerebellar cells express many of the differentiated properties of normal developing cerebellum, including the transition for the embryonic and adult isozymes of l-glycerol 3-phosphate dehydrogenase (EC 1.1.1.8). In order to determine the mechanism leading to increased levels of adult isozyme, aggregates in culture from 2 to 17 days were labeled with radioactive leucine and the relative rate of enzyme synthesis was measured after purification of the enzyme by affinity chromatography on Blue Sepharose 6B. During the course of in vitro differentiation, the relative rate of synthesis increased 100-fold, such that it represented 0.5% of the total protein synthesized in the cytoplasmic fraction of the cell. In vivo, BALBcBy mice have twice the level of enzyme activity in the cerebellum as do C57BL6J mice. Reaggregating cell cultures of cerebellar cells from these strains of mice also express a difference in the activity level, but only when the cerebellar cells are taken from mice 4 days of age or less. When the relative rates of synthesis of l-glycerol 3-phosphate dehydrogenase were measured in cultures expressing the strain-dependent difference in activity, these rates were found to be approximately twofold greater in cultures of BALBcBy cells. In contrast, estimates of the relative rate of enzyme degradation by the double-isotope labeling technique indicate that neither specific enzyme degradation nor degradation of total protein is different in aggregates from the two strains of mice. The results suggest that the genetic mechanisms controlling the levels of l-glycerol 3-phosphate dehydrogenase in the cerebellum during development are intrinsic to the cells and, with the exception of serum factors, are independent of systemic influences.  相似文献

The template activity of isolated rat liver nuclei for DNA synthesis assayed with E.coli DNA polymerase was found to be dependent upon the presence of Ca2+ or Mg2+ in the incubation medium. DNA was prepared from isolated nuclei subjected to conditions which activated the template and centrifuged in an alkaline sucrose gradient. The distribution profile showed that smaller fragments were formed, suggesting enhancement of endonucleolytic activity. When isolated nuclei were incubated with NAD to induce poly(adenosine diphosphate ribose) formation and were subjected to the activation conditions, the template for DNA synthesis remained unchanged. The distribution profile in an alkaline sucrose gradient of DNA prepared from these nuclei and control nuclei was identical. The present findings suggest that the template-activating system for DNA synthesis was blocked when isolated nuclei were treated with NAD invitro.  相似文献

5-Azacytidine, when added to growing E.coli K12, causes a decrease in DNA methylation assayed invitro. This decrease is greater when E.coli DNA is used as substrate than when calf thymus DNA is used. The decrease in activity is not due to the inhibition of protein synthesis caused by this drug, since neither chloramphenicol nor rifampin causes a decrease in enzyme activity. The effect is specific for the DNA(cytosine-5)methylase; the methylation of adenine is not affected. The concentration of drug that inhibits the DNA methylase by 50% is the same concentration that inhibits cell growth by 50%.  相似文献

Peripheral blood mononuclear cells from 48 healthy subjects of ages varying from 20 to 94 years were evaluated for the ability to generate suppressor cell activity following in vitro incubation with concanavalin A. The suppression of proliferative responses by autologous and young, allogeneic lymphocytes to phytohemagglutinin was assessed using suppressor/ responder cell ratios (SR) of 2:1 and 1:1 and by using a summation index. Inducible suppressor cell activity for autologous responder cells was comparable between 24 aged (76.0 ± 10.9 years) and 20 young (26.8 ± 4.6 years) subjects. However, aged subjects exhibited a significant decrease in suppressor cell activity (SR = 1) when allogeneic responder cells were utilized. Our results indicate that autologous inducible suppressor cell activity is preserved in the aged population, whereas allogeneic activity is impaired.  相似文献

O6-Methyl[8-3H]deoxyguanosine in a synthetic DNA polymer, poly(dC, dG, m6dG), is demethylated by cell-free extracts of EscherichiacoliBr adapted by exposure to N-methyl-N′-nitro-N-nitrosoguanidine, as shown by the appearance of 3H-labeled deoxyguanosine in hydrolysates of the recovered DNA. The demethylating activity could not be detected in extracts of nonadapted E. coli. These results provide direct evidence that a previously described inducible repair activity in E. coli acts by demethylating O6-methylguanine at the DNA level.  相似文献

Ewes of three exotic breeds (Florida Native, Barbados Blackbelly and St. Croix) showed no periparturient rise in fecal egg counts (PPR) when housed from late fall through lambing and weaning. Domestic breed ewes (Rambouillet and Finn-Dorset × Rambouillet) showed a pronounced PPR 6–7 weeks post-lambing while St. Croix × domestic ewes showed an intermediate PPR under similar conditions. There was no PPR in non-lambing ewes.In a second experiment, breed differences in the PPR were not as pronounced when ewes were allowed to graze a contaminated pasture after lambing. Florida Native, St. Croix and 34 St. Croix × 14 domestic showed lower PPRs than the remaining breeds (Barbados Blackbelly, F1 hybrids of exotic and domestic breeds and domestic ewes), but the magnitude of the PPR was generally small in all breeds. No relationship was observed between hemoglobin genotype and PPR in either experiment.It was concluded that a low or absent PPR is an important manifestation of breed resistance in exotic ewes and that this may be useful as a marker for breeding parasite-resistant sheep.  相似文献

DNA polymerases induced by Herpes simplex and Vaccinia viruses are inhibited by aphidicolin and this inhibition is probably the basis of its antiviral activity in vivo. Its possible clinical use is however hampered by the concomitant effect on human replicative DNA polymerase α. The inhibition of human α-polymerase is reversible both invitro and in vivo and the changes in the rate of incorporation of thymidine into DNA, following treatment with aphidicolin for a generation time, indicate the likely synchronization of the cells due to this agent. DNA polymerase β, which has recently been shown to carry out repair synthesis of damaged nuclear DNA, is not inhibited by aphidicolin either in vitro on in vivo suggesting that the drug could allow a rapid and simple evaluation of DNA repair synthesis due to DNA polymerase β.  相似文献

The activity of E. coli DNA polymerase I decreases on treatment with γ-rays, methylnitrosourea or dimethyl sulphate. In the case of the first two agents the decrease in activity is accompanied by a decrease in the accuracy of the enzyme in an in vitro assay. TT embryos have been first distinguishable at 8 days post coitum by their gross morphological abnormalities. By quantitative morphometry of histological sections, anomalies in the homozygotes were expressed numerically. At 8 days p.c., morphologically identifiable T-homozygotes had an increased number of ectodermal and a reduced number of mesodermal cells compared to the wild type. At 7 days p.c., embryos with a low mesoderm/ectoderm ratio were found only in litters of T+ × T+ matings at the expected frequency. At 6 days p.c., one-fourth of the embryos in T+ × T+ litters showed a delay in the transition from cuboidal to squamous endoderm. No such embryos were found in the +/+ × +/+ matings. In 6-, 7-, and 8-day mutant embryos, cells proliferated at statistically normal rates. Therefore, it may be said that advanced morphological irregularities of 8-day homozygotes cannot be accounted for by anomalies in cell proliferation. When the total cell number was 5 × 104/embryo (8 days), a sudden change was observed in the regional distribution of mesodermal and ectodermal cells along the anteroposterior axis of TT embryos. Since no regional difference in the cell cycle time was observed, these abnormalities may best be explained by anomalies in cell migration. These results strongly suggest abnormal morphology of TT mutants resulting from defects in morphogenetic movement.  相似文献

Stimulation of transduction in Escherichia coli by ultraviolet irradiation of the transducing phage P1 requires the uvrA-uvrB nuclease but not the uvrC product or DNA polymerase I. 7β,8α-Dihydroxy-9α,10α-epoxy-7,8,9,10-tetrahydrobenzo[a]pyrene (BP diolepoxide, 1) and 1-pyrenyloxirane (2) bind chemically to calf thymus DNA. The fluorescence efficiency of pyrenyl groups in mutagen modified DNA varies appreciably with its conformation and decreases in the order: pyrenees, modified denatured DNA and modified native DNA. A particularly interesting observation is that the fluorescence efficiency of mutagen modified DNA intensifies substantially upon denaturation. Our results suggest that the pyrenyl groups in mutagen modified DNA are intercalated between the base pairs of DNA. Since both 1 and 2 are powerful frame-shifting mutagens for S. typhimurium TA-98, the intercalative covalent binding of these compounds to DNA may provide a molecular basis for their mutagenic activity.  相似文献

The effects of nuclear proteins on DNA synthesis were investigated before and after incubation with radioactive ATP and a crude preparation of nuclear protein kinase. After partial purification by DEAE-cellulose chromatography, the major protein fractions were added separately to DNA polymerase assays. One of the seven protein fractions inhibited DNA synthesis by 50%, whereas three other fractions stimulated DNA polymerase activity 3 to 4-fold. After incubation with ATP, one fraction became inhibitory, and the three stimulatory fractions, which had high levels of radioactivity, were more effective. This stimulation of DNA polymerase activity was proportional to added nuclear protein and was maximum at 6 μg20 μg DNA.  相似文献

In vivo and in vitro studies were carried out to determine the effects of estradiol and other steroid hormones on aryl hydrocarbon hydroxylase-mediated binding of benzo(a)pyrene metabolites to DNA. Injection of female C57B16J mice with 0.2 mg or 2 mg of estradiol 24 hours prior to, during and 24 hours after injection of 3-methylcholanthrene resulted in a significant decrease in the capacity of hepatic microsomes from these animals to mediate the binding of benzo(a)pyrene metabolites to DNA when compared to microsomes from animals receiving 3 methylcholanthrene treatment only. Binding of benzo(a) pyrene metabolites was inhibited between 22 and 50%, depending on the dose of estradiol used. The enzyme and cytochrome components of the aryl hydrocarbon hydroxylase multienzymic complex were not affected by either estradiol treatment. The data suggests that estradiol inhibits aryl hydrocarbon hydroxylase mediated binding of benzo(a)pyrene metabolites to DNA by activity as a non-competitive inhibitor of aryl hydrocarbon hydroxylase activity.  相似文献
