为了确定鸟类免疫器官内有无内分泌细胞的存在,该文应用Grimelius嗜银染色法,MassonFantana亲银染色法,铅苏木精法和5-HT免疫组织化学方法,对鸟胸腺,腔上囊和脾脏进行了观察。结果表明:鸡和虎斑颈槽蛇相同,仅胸腺含有少量嗜银,亲银,铅染和(或)5-免疫反应阳性的细胞,具有一定的内分泌功能,腔上囊虽与胸腺同为中枢免疫器官,但没有此类细胞,这说明二者在内分泌调节等方面且一定差异。  相似文献

The comparative study of the enteropathogenic action of V. cholerae strains of group non-O1, serovar O139, and group O1 with different virulence on the APUD system of the intestine of suckling rabbits after intraenteral infection revealed that V. cholerae of group non-O1 induced inflammatory changes in the intestine and the pronounced toxic lesion of parenchymal organs. This was accompanied by a decrease in the number of apudocytes and an increase in the functional tension of the APUD system. After the infection of the animals with V. cholerae of group O1 changes in the APUD system and internal organs directly depended on the virulence of the microbes and the infective dose.  相似文献

V. cholerae infection with clearly pronounced diarrhea was reproduced in adult rabbits with the use of the RITARD system. The state of the APUD system of the intestine of the animals and morphological changes in internal organs in experimental cholera were described. As noted in this study, the manifestation of changes in the intestine and other organs, as well as the reaction of apudocytes to V. cholerae infection, were linked with the intensity of diarrhea and the time of the death of the animals.  相似文献

Summary Cytochemical techniques, performed sequentially on single sections, have confirmed the identity of human and murine pituitary corticotrophs as APUD cells. These same methods indicate that in both species the acidophil somatotroph, identified as such by immunofluorescence using anti-human GH, possesses APUD qualities not only with respect to amine-handling, but also in terms of enzyme content.Inclusion of the somatotroph in the APUD series of cells which produce polypeptide hormones has far-reaching implications.  相似文献

The complete thyroid and parathyroid gland removal leads to the abrupt reduction of calcitonin, which can be a factor stimulating growth and intensified activity of APUD system cells in the respiratory tract. Thus, neuroendocrine cells in the lungs and trachea in rats after thyroid and parathyroid removal were evaluated. Paraffin specimens of the examined organs were stained with H+E and impregnated with silver. Calcitonin, synaptophysin, somatostatin, and neuronal-specific enolase were detected immunohistochemically by the use of rabbit specific antibodies. Antibodies used in the study immunostained neuroendocrine cells of the examined organs. Rats with removed thyroid and parathyroid glands presented weakened reaction compared to the control group.  相似文献

The morphofunctional state of apudocytes in the gastrointestinal tract and immunocompetent organs (spleen, mesenteric lymph nodes) of mice immunized with chemical bivalent cholera vaccine was studied. The study revealed that the APUD system of the intestine and the argyrophil elements of the immunocompetent organs of white mice gave a response to the oral administration of commercial cholera vaccine. The reaction of the APUD system of the gastrointestinal tract was manifested by a significant increase in the number of apudocytes and their greater synthesizing activity in the immunized animals during the period of maximum immunological transformation of the macroorganism. The immunization of mice with Vibrio cholerae facilitated the maintenance of homeostasis in the macroorganism and prevented appearance of morphological disturbances in its organs and system after subsequent challenge with V. cholerae.  相似文献

Disturbances in renal homeostatic function lead to changes in endocrine cell secretory activity. The aim of this study was the histomorphological estimation of dependence of gastric APUD system cell morphology and function on the time after subtotal nephrectomy in Wistar rats. Fragments of gastric pylorus were collected 1. 2, 4, and 6 weeks after nephrectomy. Paraffin sections were stained with H+E and by silver impregnation. Immunohistochemical reactions with the use of specific antibodies against calcitonin gene-related peptide (CGRP), synaptophysin (SPh). somatostatin (ST), and neuron-specific enolase (NSE) were also performed. Immunoreactivity of the examined substances in the pyloric mucosa in the first week after nephrectomy was lower than in the control group. However, in the following time intervals, endocrine cells showed stronger immunostaining in comparison with the control rats. The results suggest that chronic renal failure can modulate secretory activity of APUD system cells.  相似文献

AgNO3 staining by generally accepted methods revealed the presence of silver-absorptive cells in the liver, particularly in perivascular areas. These cells exhibited positive argyrophilia and argentophilia. Lead haematoxylin staining and Solcia's azo-coupling methods allowed identification of the cells, which can be included among the cells of the APUD system.  相似文献

Many disturbances in electrolyte and hormonal balance in the body induced by functional impairment of renal parenchyma may affect the activity of amine precursor uptake and decarboxylation (APUD) cells, which constitute a very important link in the regulation of homeostasis. The aim of the present study was the morphological, immunohistochemical and ultrastructural estimation of enteroendocrine cells in the stomach of uremic rats. Fragments of gastric pylorus were collected 1, 2 and 4 weeks after nephrectomy. Paraffin embedded sections were stained with H + E and by silver impregnation. For identification of neuroendocrine cells, immunohistochemical reactions were performed using specific antibodies against somatostatin, synaptophysin, neuron-specific enolase and anti-calcitonin gene related peptide. The analysis showed an increased number of APUD cells in the stomach of uremic rats compared to control rats, which may be a morphological expression of their hyperfunction in the functional impairment of renal parenchyma. These results suggest that chronic renal failure can modulate the secretory processes of APUD cells.  相似文献

By means of incubation of slices in 2% solution of glyoxylic, acid adrenergic nervous structures and endocrine cells (APUD cells) have been studied in the ampule wall of the duodenal large papilla and in the lungs in 6 mature male rabbits. Topographic proximity of APUD cells and adrenergic nervous fibers is noted. Contents of serotonin and catecholamines in the structures mentioned have been investigated microfluorometrically. Simultaneous revealing of producers of monoamines make possible to suppose that serotonin and catecholamines get paracrinically from the APUD cells into terminals of the adrenergic nervous fibers.  相似文献

Thirtyfive biologically active peptides are products of the 40 cells of the APUD series, which constitute the Diffuse Neuroendocrine System (DNES). Twentyone of these peptides are found not only in the cells and processes of the DNES but also in the cells and the processes of the nervous system. Hence the appellation common. To the seven original common cytochemical features of the APUD cells it is possible to add the positive results of 3 subsidiary staining techniques (Lead haematoxylin, Argyrophilia, Formaldehyde-Fluorescamine), a singly cytochemical method (Formaldehyde-ozone), and an immunocytochemical molecular marker method (neuron-specific enolase; NSE). Application of this last method demonstrates NSE in the APUD cells and confirms their origin from "neuroendocrine-programmed epiblast."  相似文献

Using specific morphological and ultrastructural methods, the author demonstrated silver-absorptive cells in perisinusoidal areas in the liver parenchyma of guinea pigs. These cells exhibited some features characteristic of the endocrine cells of the diffuse APUD system. In anaphylactic and histamine shock their number decreased. These cells probably play a role in the pathogenesis of experimental bronchial asthma.  相似文献

应用细胞培养技术和四甲基偶氮唑盐(MTT)测定法对高压静电场照射雏鸡血液和免疫器官的T细胞对刀豆蛋白(ConA)的增殖反应及其白细胞介素-2(IL-2)诱生活性的动态变化进行了较全面系统的研究.结果发现,高压正静电场照射雏鸡血液和免疫器官的T细胞增殖功能及其IL-2诱生活性均明显高于高压负静电场照射雏鸡和对照雏鸡;而高压负静电场照射雏鸡血液和免疫器官的上述各项被检指标均不同程度的低于对照雏鸡.表明高压正静电场照射对雏鸡血液和免疫器官的细胞免疫功能及其调节具有促进作用,而高压负静电场照射可使雏鸡血液和免疫器官的细胞免疫功能及其调节减弱或降低.  相似文献

该文应用光镜、电镜和细胞计数技术对胚胎发育期虎斑颈槽蛇胸腺的发育分化进行了研究。在胚胎发育11期,胸腺原基内出现前淋巴细胞。从胚胎发育12期至出生前(16期),淋巴细胞不断增殖分化,小淋巴细胞逐渐增多,而淋巴母细胞和中淋巴细胞逐渐减少。胸腺皮质和髓质形成于16期。巨噬细胞以及肌样细胞和胸腺APUD细胞分别形成于胚胎发育14期和15期,随后数量有所增加,分别分布于胸腺皮质和髓质。  相似文献

Summary AChE activity in pancreas of some amphibians, reptiles and birds was investigated with light microscope using Karnovsky's technique.A nervous and an extranervous localization of the enzyme showing considerable qualitative and quantitative differences depending on the systematic group, could be discerned. Pancreatic peripheral cholinergic autonomic system comprises nerve fibers and perikarya. The latter seem to occur in newt and frogs as Cajal's interstitial cells (CIC) and are undoubtedly present in birds as CIC and as ganglionic cells. In lizard and snakes they were not present.The extranervous localization of AChE activity was observed in islet cells in frogs, lizards and snakes, also in the walls of blood vessels of all investigated poikilotherms.The findings are compared with results previously obtained in mammalian pancreas or in other organs. Some hypotheses on the physiological significance of enzyme presence in endocrine cells, as well as their belonging to APUD cell series are discussed.This work was supported by a fellowship of the Alexander von Humboldt-Stiftung, Bonn-Bad Godesberg and by the Deutsche Forschungsgemeinschaft (La 229-3). Summary The cells of the neural crest have APUD properties at an early stage of devel opment (72 hours in the chick embryo). The FIF procedure provides a cytochemical means for their distinction.Using mouse embryos from mothers injected, intraperitoneally, 1 hr before removal, with l-DOPA (100 mg/kg), the peripheral stream of neural crest cells was clearly identifiable at the 7-somite stage (7–8 days). At the 10-somite stage (8–9 days) the cells were observed to invade the lateral processes of the foregut, and the foregut itself. A particularly high concentration of fluorescent APUD cells was observed in the anterior portion of the IVth pharyngeal pouch, destined to become the ultimobranchial body.At the 14-somite stage (11–12 days) the developing ultimobranchial body still contains fluorescent cells of neural crest origin.The implications of these findings on the question of the origin of the entire APUD series of endocrine polypeptide cells is discussed.  相似文献

Aromatic L-amino acid decarboxylase (AADC) catalyzes the cellular decarboxylation of L-aromatic amino acids and is therefore involved in the synthesis of several biogenic amines. Application of the indirect immunoperoxidase method on human, rat, and mouse tissues using specific antibodies to AADC revealed all AADC-containing cells. Besides mast cells and adrenergic nerve fibers, the following cells were immunostained: neuroendocrine cells in the tracheobronchial epithelium; neuroepithelial bodies in the bronchopulmonary epithelium; Kultschitzky cells in the small intestine and appendix as well as adrenal chromaffin cells. All the latter cells belong to the so-called APUD system, the "D" in the acronym standing for the activity of the enzyme aromatic L-amino acid decarboxylase. Immunocytochemistry for AADC may become an additional tool not only to highlight APUD cells in tissue sections but also to differentiate the sites of cellular amine synthesis from those of amine storage.  相似文献

The authors describe the results of the study of rabbit decalcified temporal bones and guinea-pig membranous cochlea treated according to the Grimelius argyrophil method and immunohistochemical technique with the use of a number of specific antisera. The study revealed the cells of the diffuse endocrine system (APUD system) in various parts of the internal ear: in the main membrane, receptor elements of the spiral chorda, and spiral ligament. Serotonin and melatonin were identified in the cells of the APUD system. The data obtained may form a theoretical basis of a basically new trend in the study of the function of the internal ear in health and disease.  相似文献

Three different cytochemical methods, the formaldehyde-fluorescamine and the ortho-phtalaldehyde fluorescence cytochemical methods as well as immunocytochemistry have been developed for localising the polyamines spermidine and spermine in tissue. All three methods produce identical results and show polyamines to occur inter alia in high concentrations in certain protein- and peptide-secreting cells. Many of these cells also show the capacity to metabolise monoamines and belong to the amine content or amine precursor uptake and decarboxylation (APUD) series. In cell types where fluorescence microscopical resolution allows, most polyamines appear to be localised to secretory granules. Moreover, studies on isolated pancreatic islets reveal active and glucose-dependent polyamine biosynthesis to occur in insulin cells. Possible function of polyamines in secretory granules are discussed in the light of the above findings.  相似文献

Using an immunocytochemical procedure a wide range of immunoreactive vertebrate bioactive peptides (BAPs) has been found in hemocytes of Viviparus ater: bombesin, calcitonin, CCK-8, CCK-39, GH, glucagon, insulin, oxytocin, neurotensin, secretin, serotonin, somatostatin, substance P, vasopressin, and VIP. 2. No immunostaining was observed for antigastrin and antithyroglobulin antibodies. 3. The presence of BAP-like molecules in hemocytes suggests a correlation between hemocyte and APUD cells and is evidence of a relationship between the neuroendocrine and the immune systems.  相似文献
