用基因枪法将DREB基因和诱导型启动子rd29B构建的载体pBAC128F转入烟草悬浮细胞。通过除草剂抗性筛选、PCR和PCR-Southern检测,获得转DREB基因的细胞系。以300mmol·L-1NaCl和15%PEG6000处理14d后,转基因细胞系存活并生长,而野生型则接近死亡。转基因细胞系的脯氨酸含量和超氧物歧化酶活性普遍高于野生型的,而丙二醛含量则相反。  相似文献

以冬小麦品种8901、5-98、99-92和104等品种的幼穗和幼胚为材料,用基因枪转化含逆境诱导转录因子DREB和bar基因的质粒pBAC128F(7024bp)。经筛选与植株再生,共获得70多个转基因小麦植株及其后代株系。转基因株系经PCR分析和RNA点杂交检测,结果表明外源转录因子DREB基因已稳定整合到转基因植株及其后代株系中,并且在部分后代株系中获得了表达。叶片脯氨酸含量测定表明,有16个转基因株系的脯氨酸含量与非转基因对照相比,增加相当显著,其中10个株系的脯氨酸含量在1100μg/g以上,比对照提高了2倍多。室内抗旱模拟实验表明,转基因株系停止浇水15d后,叶片仍然表现绿色,而对照叶片则失绿、枯干;复水10d后,转基因株系恢复活力,对照则死亡。研究表明,利用逆境诱导型启动子(rd29B)来增强外源DREB基因的表达,能显著改良小麦的抗旱性。  相似文献

将携带有AtDREB1A基因, 并以35S或rd29A启动子驱动的植物表达载体转入地被菊花(Dendranthema grandiflorum)的粉色品种‘Fall color’. 与野生型相比, 35S:DREB1A转基因植株表现出严重的生长抑制, 而rd29A:DREB1A植株生长正常. RT-PCR检测表明, 在胁迫条件下, AtDREB1A基因在35S: DREB1A转基因植株中呈现组成型过量表达, 而在rd29A:DREB1A植株中则是受胁迫诱导型过量表达. 这两种启动子驱动的转基因植株对干旱和盐渍胁迫都表现出较强的耐性, 其中rd29A:DREB1A植株耐性显著强于35S:DREB1A植株. rd29A:DREB1A植株中的脯氨酸含量和SOD活性都强烈地被胁迫诱导升高, 且高于35S:DREB1A植株. 这些结果表明, 在地被菊花中表达AtDREB1A基因可以提高植株对干旱和盐渍胁迫的耐性, 同时这些耐性的升高可能与脯氨酸含量和SOD活性的上升有关.  相似文献

根据文献上发表的逆境诱导型启动子rd29A 序列设计并合成了一对引物, 通过PCR 的方法从拟南芥基因组中扩增到rd29A 的启动子序列。根据GenBank 中已发表的转录因子DREB1A基因的cDNA 序列设计并合成了一对引物, 通过RT-PCR 的方法从低温处理的拟南芥总RNA 中扩增出DREB1A 基因的全长cDNA 片段。以植物表达载体pBch 为基础, 构建了由rd29A 调控的DREB1A 基因的植物表达载体pBDR29A, 为利用DREB1A 基因改良植物抗逆性奠定了物质基础。  相似文献

以成熟胚诱导的愈伤组织作为农杆菌转化的受体材料,将诱导型启动子rd29A驱动的拟南芥DREB1A基因导入粳型光温敏核不育系水稻4008S,共获得67株再生苗.再生苗经0.75 mg/L除草剂草铵膦涂布筛选,有62株再生苗表现出对草铵膦抗性.PCR检测抗性苗中DREB1A基因,结果全为阳性.挑选部分进行Southem检测.结果表明目的基因已经整合到水稻基因组中.在干旱胁迫下,转基因水稻当代(T1代)植株的电导率显著低于非转基因对照植株(P<0.05),脯氨酸含量显著高于对照植株(P<0.05),证明DREBIA基因能提高水稻对干旱胁迫的耐受性.  相似文献

脱水应答转录因子CBF1的克隆与转基因小麦的分子检测   总被引:1,自引:0,他引:1  
根据已发表的小麦(T.aestivum)转录因子CBF1基因序列(GenBank Accession No.AF376136),设计引物从小麦品种‘京花1号’叶片中克隆出该基因,用拟南芥RD29B基因为启动子构建含CBF1基因的逆境诱导表达载体pBAC127F(6 967 bp),以‘99-92’、‘5-98’、‘104’和‘轮选987’等冬小麦品种(系)的幼穗和幼胚为材料,基因枪转化该表达载体。经筛选与植株再生,共获得14株转基因植株及其后代株系。这14个株系经PCR分析和点杂交检测,最终确认了5-98-40、5-98-41这2个株系为转基因株系,结果表明拟南芥RD29B启动子调控下的转录因子CBF1基因已稳定整合到转基因植株中。  相似文献

Rd29A启动子驱动AtCDPK1基因转化马铃薯的研究   总被引:1,自引:0,他引:1  
为获得抗旱性强、生长正常的转基因马铃薯植株,以野生拟南芥生态型(Col-0)为材料,利用PCR和DNA重组技术,克隆了拟南芥Rd29A(responsive to dehydration)基因ATG上游+83bp至-1 441bp共1 524bp的启动子区域,其DNA序列与已知拟南芥Rd29A 5'端启动子序列同源性为100%;构建了Rd29A启动子驱动AtCDPK1基因表达的植物表达载体pCHFRd-CDPK1。以马铃薯品种‘费乌瑞它’的试管微型薯为材料,利用农杆菌介导法,将构建成功的pCHFRd-CDPK1载体转入马铃薯中,经筛选与植株再生,获得抗性再生植株。通过PCR和Southern blot检测显示,Rd29A启动子驱动的AtCDPK1基因已整合在马铃薯的基因组中。利用PEG模拟干旱胁迫后,经RT-PCR分析证实,当用20%的PEG胁迫转基因马铃薯植株时,Rd29A启动子驱动AtCDPK1基因表达的转基因马铃薯各个株系中AtCDPK1基因表达量明显增强,而在无胁迫的条件下,植株中AtCDPK1基因基本不表达;同时发现35S控制AtCDPK1转基因植株在PEG胁迫前后,基因转录未见明显差异。形态学观察还表明,在30%PEG胁迫下,转基因植株能正常生长,其长势优于未转基因的对照,且对照植株略有萎焉。该结果可为进一步利用逆境诱导型启动子驱动抗逆基因在农作物中的表达研究及其遗传改良提供依据。  相似文献

Ss DREB是从盐地碱蓬克隆获得的一个DREB2类转录因子,其表达受高盐和干旱诱导,而对ABA和低温处理反应不明显。本研究将Ss DREB与绿色荧光蛋白(GFP)基因构建融合表达载体并在洋葱表皮细胞中进行瞬时表达,结果表明SsDREB编码蛋白定位在细胞核中;酵母转录激活实验证明,Ss DREB能特异性结合DRE顺式作用元件,并激活下游报告基因的表达;采用农杆菌介导法将Ss DREB基因在35S启动子的驱动下转入烟草中,获得的转基因植株对干旱和盐胁迫抗性均显著提高;为研究Ss DREB过量表达提高转基因烟草抗旱、耐盐能力的分子机制,选取烟草中与提高质膜稳定性、消除活性氧和渗透平衡相关的8个逆境胁迫相关基因,以烟草α-tublin基因做为内参,利用半定量RT-PCR方法分析在正常生长条件下这8个基因在转基因烟草和对照烟草中的表达情况,实验结果表明这8个基因都受外源Ss DREB基因的调控,其中6个基因在转基因烟草中的表达量显著高于非转基因植株。  相似文献

将霍乱肠毒素B亚单位(CT-B)基因及内质网引导序列(SEKDEL)克隆到质粒pRTL2和pBI121中,分别构建植物双元表达载体pBI-CTB和pBI-CTBK,CT-B基因由Ca35S启动子控制表达。采用叶盘法经根癌农杆菌介导转化番茄(金丰1号,Jinfeng1)各表达载体得到一批转基因植株。经PCR和Southern blot分析表明CT-B基因整合到了番茄基因组中;ELISA和Western blot分析表明pBI-CTB和pBI-CTBK的转基因植株能够有效表达CT-B多肽,分别占番茄叶片可溶性蛋白的0.055%和0.084%。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
