Ultrastructural study of gonadal muscles in sea star, Asterina pectinifera, showed that myoepithelial cells were located only in the epithelial lining of the genital coelomic sinus. No myoepithelial cells were found in the visceral peritoneal epithelium or within connective tissue layer of the outer sac. Morphology of the myoepithelial cells in gonads of A. pectinifera varies during the reproductive cycle. During the gametogenic phase of the reproductive cycle, the myoepithelial cells get an elongated, spindle-like shape having a length of 20–30 μm. In prespawning gonads, many of the myoepithelial cells form cytoplasmic extensions of 3–5 μm in length, filled with myofilaments and penetrating into the underlying connective tissue of the outer sac or haemal sinus. Besides, myoepithelial cells, simultaneously anchored in the inner and outer sacs, were also observed. These changes result in development of more elaborated musculature and increase in contractility of the gonadal wall in prespawning gonads as compared to that during other stages of the reproductive cycle.  相似文献

Summary The fine structure of the adrenal cortex of the domestic fowl has been studied from the 14th day of embryonic life to 980 days after hatching, using the electron microscope.Many mitochondria and vacuoles, round or oval, are observed in all cortical cells. The cristae mitochondriales are not laminar but villous. In the embryo, the mitochondria, whose cristae are not as well developed as those in chick and hen, are very low in electron density. Cytoplasmic vacuoles, either rough or smooth-surfaced, and with a homogeneous content of low electron density, are enclosed by a similar limiting membrane. Some of them are formed by the outer nuclear membrane and are characterized by many small particles on their outer surfaces. These vacuolated structures are perhaps parts of the endoplasmic reticulum. In the 980 day old hen, a number of osmiophilic droplets, round or irregularly shaped, are seen. Some of these are formed by mitochondria, while others accumulate within or around cytoplasmic vacuoles. In the embryo and in the chick, such droplets are never seen. It is assumed that the droplets are substances related to ageing, and that the lipid implicated in cortical hormone is not osmiophilic in the young chick and the embryo. It is perhaps produced in the cytoplasmic vacuoles with the aid of mitochondria. A perisinusoidal space and interparenchymatous cell spaces similar to those described for mammals by other workers, are observed in the adrenal gland of the domestic fowl. I assume that the secretory substance of the cortical and the medullary cell is first secreted into these spaces and then infiltrates into the capillary lumen.  相似文献

In Proteocephalus thymalli and P. torulosus, a contact of the placental type in uterus was shown to be formed at two different levels. At the first level an interaction occurs between outgrowths of uterine epithelium and thin capsule of embryos closely adjacent to uterine wall. The next level is formation of contact between neighboring egg capsules, which allows distributing nutrients among fetuses present in the uterine cavity. Placental interactions in Clestobothrium acheilognathi are limited in time and space. First, a relatively small number of eggs are involved in interaction of the placental type in the uterine sac, while uterine duct is filled with freely lying eggs. Second, the closest contact is observed in eggs with non-sclerosed egg shell. One of the main evolutionary tendencies in cestodes has been shown to be a modification of uterus for formation of close interrelations with embryonic membranes in the course of transition from the extrauterine to the intrauterine type of embryonic development. Uterus in parasites with a polylecital type of the egg is suggested to serve to the greater extent as a reservoir, whereas in cestodes with oligolecital eggs, uterus performs its direct function—supply of developing embryos with nutrients. As a result, modifications of uterine epithelium are formed: from the appearance of the placental type interactions formed repeatedly in phylogenetically distant groups of cestodes to formation of branched outgrowths separating the uterine space into units or disintegration to actively functioning uterine capsules.  相似文献

To understand the degeneration of unfertilized aposporous embryo sac in an aposporous species Pennisetum squamulatum, the central cell in the unfertilized embryo sac was characterized ultrastructurally . The most prominent sign of degeneration is that the central cell nucleus produced nuclear vacuoles . These nuclear vacuoles can be classified into singleanddouble- layered types . Single- layered nuclear vacuoles are found in the cytoplasm, while the double-layered are inside the nucleus . There are two ways to produce nuclear vacuoles . One is that the outer membrane of the nuclear envelope distends towards the cytoplasm to form nuclear vacuoles ; and the other is the double-layered nuclear envelope derives vacuoles by depressing inwards . Nuclear envelope types are in relation to the ways they are produced . All nuclear vacuoles absorb cytoplasm or nuclear matrix , and trap organelles such as mitochondria .  相似文献

Synopsis Embryos of Latimeria chalumnae develop in well-vascularized compartments in the uterine region of the right oviduct. Compartments conform to the shape of their embryos and yolksacs; they represent a stable, gestation-induced oviductal modification. Late-term pups possess large, flaccid, vascular yolksacs almost devoid of yolk. The sac is in close contact with, but does not adhere to, the lumenal uterine surface. A massive vascular plexus occurs in the wall of the compartment at the site of contact with the yolksac; together they constitute a non-adherent, transposable placenta. The exterior surface of the yolksac is bounded by an attenuated, single-layered, squamous epithelium that surrounds an intercommunicating bed of cortical sinuses. The cortex of the sac is composed mostly of connective tissue stroma. The inner surface is bounded by a layer of yolk-digesting merocytes. Residual yolk occurs as yolk platelets that include yolk crystals. The interior surface of the sac is invested by an uniquely specialized vitelline circulation; no connection seems to exist between the interior of the yolksac and gut. The uterine wall consists of: (1) a lumenal surface composed of an anastomosing network of capillaries with a layer of attenuated, very thin, squamous epithelium, (2) a well-vascularized connective tissue stroma, (3) alternating transverse and longitudinal layers of smooth muscle, also well-vascularized, and (4) an external epithelial layer. Comparison of egg dry weight (184 g) with the estimated dry weights of a late-term pup (171 to 239 g) and a neonate (200 to 280 g) reveals a weight change of – 7 to + 30% and + 9 to + 52%, respectively. This is indicative of matrotrophy. In one female specimen, 19 remarkably large ovulated eggs were found and in another about 30 somewhat smaller ovarian ones. These are many more than ever could be accommodated in the uterine space. During the early and middle phases of development, embryos must be lecithotrophic, using their yolk reserves, with oophagy of fragmented supernumerary eggs as the most probable source of additional nutrients. The well-developed embryonic gut contains brown, amorphous yolk-like material. The limited amount of metachromatic secretory product of the uterine glands can play little or no role in embryonic nutrition.  相似文献

The abdominal fat body of the cockroach Blattella germanica contains three characteristic cell types—trophocytes, bacteriocytes and urate cells—which have been investigated by electron microscopy. The trophocytes are rich in lipid droplets of different sizes; glycogen, rough endoplasmic reticulum and mitochondria are also abundant. In females immediately after eclosion, the trophocytes contain a greater number of lipid droplets, some of which have different electron density; glycogen and cytoplasmic organelles are clearly reduced. The bacteriocytes hold rod-like and spherical bacteroids, which are encapsulated by a vacuolar membrane; they show a thin cytoplasmic membrane and an evident cell wall surrounded by a membrane-like outer envelope. The bacteroids appear to be dividing either by transverse partition or by budding. The urate cells, adjacent to the bacteriocytes, are characterized by complex urate vacuoles delimited by a double layer-structure.  相似文献

The structure of the placental labyrinth, interlobular or "coarse" syncytium, visceral (splanchnopleuric) yolk sac, giant cells and subplacenta of the chinchilla was studied with the electron microscope. The fine structure of the interhemal membrane of the placental labyrinth was found to be hemomonochorial, consisting of a single layer of syncytial trophoblast. In this respect, the placental labyrinth was similar to that of another caviomorph rodent, the guinea pig. The labyrinthine trophoblast had pinocytotic vesicles as well as larger vaculoes and multivesicular bodies. The interlobular syncytium contained granular endoplasmic reticulum, and in one case from early in gestation there were intracisternal granules in the ER. The visceral endodermal cells of the inverted yolk sac placenta had a well-developed system of apical vesicles and tubules as well as larger cytoplasmic vacuoles. Their appearance was similar to that of endodermal cells found in other rodents which are known to absorb proteins and other substances from the uterine lumen. Towards term the giant cells were often vacuolated and contained large deposits of glycogen as well as lipid droplets. The syncytial trophoblast of the subplacenta contained numerous moderately electron-dense granules which may be secretory in function; cytotrophoblastic cells lacked these granules. The subplacental syncytium often surrounded spaces or lacunae which contained an electron-dense granular material.  相似文献

The microanatomy and ultrastructure of the uterus in different stages of ontogenesis were studied in a cyclophyllidean cestode Lineolepis scutigera. In the early stages, developing embryos lie freely in the cavity of a single unbroken uterus, which is later fragmented into separate compartments. As a result, numerous spherical uterine capsules are formed; each of them contains one formed egg. Capsules represent a fragmented but actively functioning uterus. Muscular cells containing numerous lipid inclusions are located around them. In the final stages, placenta-like relationships are formed between eggs and the epithelium of capsules. A comparative morphofunctional analysis of uterine capsules in cestodes is presented. Attention is paid to the formation of close interactions between the uterine epithelium and developing eggs.  相似文献

The ovule is the most important reproductive organ in the pistil of phanerogamae. Camellia oleifera (Theaceae) is an important woody plant producing edible oil in southern China, and its embryo sac structure has a positive effect on seed breeding. In this study, the microstructure, ultrastructure and three‐dimensional structure of the ovule and embryo sac of C. oleifera were observed and described based on a combination of advanced microscopy techniques (SEM, TEM, CLSM). The ovule comprises the inner and outer integument. Large quantities of secretions in the micropylar canal exit and may participate in the guidance of the entry of the pollen tube into the embryo sac. The synergids have a dense cytoplasm, abundant organelles, and strong polarity. Little cytoplasm is present in the egg cell, yet there are many vacuoles. The center of the cell is taken up by a large vacuole, and the cytoplasm is pushed towards the edges to form obvious cytoplasmic cords. The two polar nuclei are large and conspicuous. The antipodal cells degenerate to fulfill a nutritional function.  相似文献

Different developmental stages, fertilized eggs through hatchlings, of the climbing perch,Anabas testudineus, have been studied by scanning electron microscopy. The surface specialization of eggs and hatchlings reveals that while the egg surface is reticulate in appearance, the hatchlings are covered with microridges. Vitelline arteries are seen at the pharyngeal and abdominal regions. They supply nutrients directly from the yolk sac to the developing embryo. Three pairs of such arteries are distinctly seen in the pharyngeal region. Mucous glands are discernible at places over the entire body surface of the embryo before the formation of scales. The skin seems to be helpful in gaseous exchange till the gills and accessory respiratory organs develop and become functional.  相似文献

By electron microscopy of the coxal gills in two species of estuarine amphipod crustaceans, Grandidierella japonica and Melita satifragella, we found a patch-like, specialized tissue area which consisted of unique cells closely resembling the salt-excreting cells in the gill of the brine shrimp and so-called chloride cells in teleost gills. These cells were characterized by an abundance of mitochondria, two kinds of extensive networks of cytoplasmic tubules, well-developed lamellar infoldings of the basal cell membrane, sparse microvillous projections of the apical border, and numerous large vacuoles with several incomplete partitions. The large (60 nm in diameter) and the small (30 nm) cytoplasmic tubular networks were found in the basal and the apical portions of the cell, respectively. The large networks, which were both directly and indirectly (through the lamellar system) continuous with the basal cell membrane, were regarded as extensions of the cell membrane. Both the outer walls and the partition walls of the vacuoles were reinforced with a parallel array of microtubules. The results suggest that this unique tissue plays an important role in the active transport of electrolytes to maintain a constant osmotic pressure of the hemolymph under widely fluctuating salinities of the estuarine environments.  相似文献

Mouse macrophages exposed to 30 µg/ml of chloroquine in vitro develop autophagic vacuoles containing various cytoplasmic components and acid phosphatase. The early toxic vacuoles appear in the perinuclear region within 15 min; on electron microscopy, they show irregular shape, amorphous moderately dense content, apparent double membranes, and in some instances curved thin tubular extensions with a central, dark linear element. Cytoplasmic structures are probably transported into the vacuoles by invagination of the vacuolar membrane. After exposure to chloroquine for 1–4 hr, macrophages display large vacuoles containing degraded cytoplasmic structures, membranous whorls, and amorphous material. When chloroquine is removed by changing the culture medium after 4 hr, the cells survive and 24 hr later they exhibit no abnormality except for large cytoplasmic dense bodies packed with membrane lamellae. During recovery chloroquine disappears from the cells. 24 hr after exposure to chloroquine the macrophages have accumulated less hydrolases than control cells.  相似文献

The addition in vitro of insulin to rat adipose tissue (epididymal) produces marked metabolic changes which may be followed by measurement of the net gas exchange of the tissue. Using this method to monitor the metabolic action of insulin, concomitant observations with the electron microscope on the tissue have been made. These reveal that pronounced morphological changes are induced by insulin. The plasma membranes of the adipose cells become invaginated at many sites to form minute finger-like indentations. Numerous tiny, membrane-bounded vesicles are also present and arranged in relationship to the plasma membrane in such a way as to suggest that their formation occurred when a recessed fold was pinched off. Deeper in the cytoplasm, especially in specimens that had been incubated a longer time, numerous large, smooth, membrane-limited vesicles are seen. Finally, in these incubated specimens the cytoplasmic matrix has lost much of its granular nature, small lipid droplets are frequently found in the cytoplasm and suggestive changes have occurred in mitochondria. In control specimens, incubated without insulin for identical periods of time, indentations and vesicles in the plasma membrane are sparse at best and no vesicles or membrane-bound spaces appear deeper in the cytoplasm. The metabolic and morphologic changes induced by insulin seem to be interdependent events. Both changes appear to be initiated rapidly and concomitantly in the tissue. Both processes are initiated by insulin at concentrations considered to be physiological, 0.004 µg. (100 µunits) per ml. Insulin treated with alkali fails to initiate either process. It is concluded that insulin initiates pinocytosis in rat adipose tissue and the possible significance of this process in the mode of action of insulin is discussed.  相似文献

Electron microscopic examination of the paired coxal glands of the horseshoe crab Limulus polyphemus, focusing on urinary and vascular channels, shows six morphologically distinct regions. Each of four nephridial lobes consists of two cortical layers surrounding a medulla. The outer and inner cortexes contain blood vessels separated by a basement membrane from the urinary space lined by podocytes. Podocyte foot processes are applied to the basement membrane, interdigitate with those from other podocytes, and have a filtration slit diaphragm between them. Cortical morphology demonstrates ultrafiltration of blood, a previously undescribed function of the gland, as well as possible endocytic reabsorption of materials by the podocytes. The medulla drains into the stolon connecting the four lobes. These two areas have urinary tubules of cuboidal epithelium featuring microvillous-like apical projections, cytoplasmic vesicles and vacuoles, elaborate lateral interdigitations with septate junctions, and basal invaginations containing numerous mitochondria. These tubules are closely surrounded by blood channels, lined by a basement membrane containing embedded support cells. The medulla and stolon morphology are suggestive of both ion transport and water movement, in keeping with the gland's role in osmoregulation. The stolon empties into the end sac in the base of the most posterior lobe. It is lined by tall epithelium exhibiting apical overlap, blunt projections into the lumen, apparent endocytic vesicles, and basal plasma membrane infoldings with mitochondria. Clean, nonembryonated Capillaria hepatica eggs recovered from infected liver tissue by physical methods as well as eggs obtained after passage through the mouse gastrointestinal tract were examined with the electron microscope. In eggs collected by physical methods the outer membrane showed defects where it was lacking at various points or disrupted. The pillars of the outer shell were frequently broken or fractured resulting in virtual collapse or separation of the outer shell from the inner shell. No shell matrix was observed in any eggs examined. Even more dramatic effects were observed in eggs recovered following passage through the mouse gastrointestinal tract. Clean, nonembryonated eggs collected by physical methods were suspended at 5 C in phosphate-buffered saline. A large amount of protein was released initially into the medium; the amount released then fell to a low level at which it remained for several weeks. Gel diffusion tests with concentrated protein supernatant and C. hepatica egg-derived antigen were compared using appropriate antisera. Bands of identity were present in both; however, egg antigen contained other proteins not present in egg supernatant. These studies indicate that during physical collection of C. hepatica eggs, sufficient damage occurs to allow for the release of materials into host tissue during experimental egg granuloma formation. An hypothesis is presented concerning the viability of C. hepatica eggs in host liver tissue following host cellular response and the possible modes of action which trigger development of eggs after release from infected liver.  相似文献

Encysting cells of Acanthamoeba castellanii, Neff strain, have been examined with the electron microscope. The wall structure and cytoplasmic changes during encystment are described. The cyst wall is composed of two major layers: a laminar, fibrous exocyst with a variable amount of matrix material, and an endocyst of fine fibrils in a granular matrix. The two layers are normally separated by a space except where they form opercula in the center of ostioles (exits for excysting amebae). An additional amorphous layer is probably present between the wall and the protoplast in the mature cyst. Early in encystment the Golgi complex is enlarged and contains a densely staining material that appears to contribute to wall formation. Vacuoles containing cytoplasmic debris (autolysosomes) are present in encysting cells and the contents of some of the vacuoles are deposited in the developing cyst wall. Lamellate bodies develop in the mitochondria and appear in the cytoplasm. Several changes are associated with the mitochondrial intracristate granule. The nucleus releases small buds into the cytoplasm, and the nucleolus decreases to less than half its original volume. The cytoplasm increases in electron density and its volume is reduced by about 80%. The water expulsion vesicle is the only cellular compartment without dense content in the mature cyst. The volume fractions of lipid droplets, Golgi complex, mitochondria, digestive vacuoles, and autolysosomes have been determined at different stages of encystment by stereological analysis of electron micrographs. By chemical analyses, dry weight, protein, phospholipid, and glycogen are lower and neutral lipid is higher in the mature cyst than in the trophozoite.  相似文献

Ferritin was used as a tracer to study the mechanism by which proteins are segregated into droplets by the visceral epithelium of glomerular capillaries. In glomeruli from both normal and aminonucleoside-nephrotic rats ferritin molecules introduced into the general circulation penetrated the endothelial openings and were seen at various levels in the basement membrane. Striking differences between nephrotic and controls were seen only in the amount of ferritin incorporated into the epithelium. In normal animals, a few ferritin molecules were seen in small invaginations of the cell membrane limiting the foot processes, within minute vesicles in the epithelium, or within occasional large vacuoles and dense bodies. In nephrotics, epithelial pinocytosis was marked, and numerous ferritin molecules were seen within membrane invaginations and in small cytoplasmic vesicles at all time points. After longer intervals, the concentration of ferritin increased in vacuoles and particularly within the dense bodies or within structures with a morphology intermediate between that of vacuoles and dense bodies. In nephrotic animals cleft-like cavities or sinuses were frequently encountered along the epithelial cell surface facing the urinary spaces. Some of these sinuses contained material resembling that filling the dense bodies except that it appeared less compact. The findings suggest that ferritin molecules—and presumably other proteins which penetrate the basement membrane—are picked up by the epithelium in pinocytotic vesicles and transported via the small vesicles to larger vacuoles which are subsequently transformed into dense bodies by progressive condensation. The content of the dense bodies may then undergo partial digestion and be extruded into the urinary spaces where it disperses. The activity of the glomerular epithelium in the incorporation and segregation of protein is similar in normal and nephrotic animals, except that the rate is considerably higher in nephrosis where the permeability of the glomerular basement membrane is greatly increased.  相似文献

In the guinea pig and some other animals, passive immunity is conferred on the developing fetus by passage of immunoglobulin from mother to fetus across the yolk sac. In order to examine the cytological pathway involved in immunoglobulin transport, guinea pig visceral yolk sacs from late in gestation were exposed in vitro to peroxidase-conjugated guinea pig immunoglobulin G (IgG-HRP). Tissue was then fixed, incubated to show the site of localization of peroxidase reaction product and prepared for electron microscopy. The results suggested that the first step in the uptake of IgG-HRP by yolk sac is attachment of the protein to the surface coats of endocytic invaginations at the apical surfaces of the endodermal cells. The endocytic vesicles then appear to pinch off from the surface and move deeper into the cytoplasm. Some of the small endocytic vesicles fuse with large apical vacuoles, which often contain large amounts of reaction product. Other small endocytic vesicles pinch off from the surface, move deeper into the cytoplasm and fuse with the lateral plasmalemma; their protein content is emptied into the intercellular space by exocytosis. From the intercellular spaces the protein presumably diffuses across the basement membrane and connective tissue spaces and enters the vitelline capillary bed. It is postulated that the latter cellular pathway, involving small vesicles and the intercellular spaces, is utilized by those immunoglobulins which are transferred intact across the yolk sac endoderm.  相似文献

The flagellate Crithidia bombi and the neogregarine Apicystis bombi have been found in individuals of Bombus terrestris, a Palaearctic species of bumble bee commercially reared and shipped worldwide for pollination services. B. terrestris has recently entered into the northwestern Patagonia region of Argentina from Chile, where it was introduced in 1998. Prevalence was 21.6% for C. bombi and 3.6% for A. bombi (n = 111). The pathogens were not detected in 441 bumble bees belonging to five of the eight known Argentine native species (Bombus atratus, Bombus morio, Bombus bellicosus, Bombus opifex, Bombus tucumanus) collected elsewhere in the country. Although the absence of natural occurrence of C. bombi and A. bombi in Argentine native bumble bees cannot be ascertained at present due to the limited surveys performed, it is important to report their detection in invasive B. terrestris. The invasion event is relatively recent and the accompanying pathogens are not species specific within the genus Bombus.  相似文献
