Sampling of Xiphinema index for 2 years (1993-95) in two California vineyards indicated that a greater number of nematodes occurred during the winter months. The number of juveniles increased four-fold from December 1993 to January 1994, indicating a high reproductive rate during this time. Extremely high or low soil temperatures corresponded to low nematode numbers. Samples were taken from 0 to 31 cm and 31 to 62 cm deep both within and between the vine rows. Numbers of nematodes were greatest at the 0- to 31-cm depth in one vineyard with a loamy sand soil, and at a depth of 31 to 62 cm in the second vineyard, which had a silt loam soil. In both vineyards, X. index population densities were greater within the vine row. 相似文献

Field experiments were conducted in 1989 and 1990 to examine the population fluctuation patterns of Tylenchorhynchus nudus, Criconemella curvata, and Helicotylenchus cornurus in mixed bentgrass and annual bluegrass putting greens on two golf courses near Chicago, Illinois, to determine if fluctuation patterns could be extrapolated to unsampled greens. Fenamiphos-treated and untreated plots were established on seven putting greens on two golf courses. Greens were sampled intensively five times during the growing season, and statistical comparisons of population levels per gram of root were made among dates for each green. Population levels per gram of root changed significantly on all greens in both years for each of the three nematode populations. Within a putting green in either year, population fluctuation patterns in fenamiphos-treated and untreated plots were similar. Population fluctuation patterns were different between years, however. Within a year, population fluctuation patterns among greens showed similarities indicating that carefully monitoring a few locations may allow extrapolation of population fluctuation data to other locations within that year. 相似文献

The population fluctuation and composition of Xiphinema americanum (sensu stricto) and X. rivesi were studied in a New York apple orchard (only X. americanum present), a Pennsylvania apple orchard (both X. americanum and X. rivesi present), and a Pennsylvania peach orchard (X. americanum, X. rivesi, and X. californicum present). Few clear trends in population fluctuation or composition were observed. The adult female was the predominant stage in most sample periods, and the reproductive period was limited to late spring and early summer. Only a few of the females at any sample period were gravid. All stages were present throughout the year, and all stages overwintered. Eggs in soil were not monitored. In the Pennsylvania apple orchard, X. americanum and X. rivesi were easily separated by morphological characteristics; however, the two species did not display differences in population structure or composition. The predominance of adults, the relatively low reproductive rates, and the association of these species with stable habitats suggest that the life strategies of X. americanum and X. rivesi are K-selected as opposed to r-selected. 相似文献

Though in the past Xiphinema americanum has been the most commonly reported dagger nematode in the eastern United States, our studies revealed the presence in Pennsvlvania of a previously unrecognized and unreported species related to X. americanum, Morphometric data and photomicrographs establish the identity of this form as X. rivesi and show expected variations in populations of this species from various locations. Similar data and illustrations are given for X. americanum populations from Pennsylvania and other areas, showing variations and relationships. Xiphinema rivesi is widely distributed in the fruit producing area of south-central Pennsylvania and is also reported herein from raspberry in Vermont and apple in Maryland and New York. This species is frequently found in fruit growing areas of Pennsylvania associated with tomato ringspot virus-induced diseases and is also found associated with corn, bluegrass sod, and alfalfa. 相似文献

Populations of plant-parasitic nematodes in an Iowa cornfield were studied along north- and west-facing toposequences. Samples were collected monthly during the growing season. The greatest biomass for Xiphinema americanum occurred at the footslope on the north face. Paratylenchus microdorus had its greatest biomass at the summit position, generally more in the west- than in the north-facing slope. Pratylenchus spp. in the roots peaked at the toeslope in the north-facing slope, but at the footslope in the west-facing slope. Helicotylenchus pseudorobustus peaked at the backslope and the toeslope along the north- and west-facing slopes, respectively. Diversity, as computed for each plot by the Shannon-Weiner diversity index, was highest at the backslope in both toposequences. 相似文献

Penetration of cabbage roots by Heterodera schachtii was suppressed 50-77% in loamy sand naturally infested with the nematophagous fungus Hirsutella rhossiliensis. When Heterodera schachtii was incubated in the suppressive soil without plants for 2 days, 40-63% of the juveniles had Hirsutella rhossiliensis spores adhering to their cuticles. Of those with spores, 82-92% were infected. Infected nematodes were killed and filled with hyphae within 2-3 days. Addition of KCl to soil did not increase infection of Heterodera schachtii by Hirsutella rhossiliensis. The percentage of infection was lower when nematodes were touched to two spores and incubated in KCl solution than when nematodes naturally acquired two spores in soil. 相似文献

The use of plant-covers oat (Arena sativa L.), rhodesgrass (Chloris gayana Kunth), soybean (Glycine max [L.] Merr.), and marigold (Tagetes patula L.) during pineapple inter-cycle planting periods was investigated at two sites (Kunia and Whitmore, Oahu, HI) as a potential means to reduce population densities of Rotylenchulus reniformis, Helicotylenchus dihystera, and Paratylenchus spp. Clean fallow and fallow covered with pineapple-plant residues (mulch) were the controls without plant-cover. Regardless of treatments, population densities of R. reniformis declined with time at both sites to low residue levels by the end of the 6-month period. Treatment means of R. reniformis population densities in the plant-cover treatments were lower than the controls'' (P = 0.05). The plant-cover treatments also effected higher rates of R. reniformis population decline at both sites during the period, being 2.0 to 2.2 times that of the mulch control and 1.2 to 1.4 times that of the fallow control. Plant-covers'' effect on H. dihystera during the same period at both sites was variable, resulting in decreased, unchanged, or increased population densities. The change was especially obvious in the oat-cover treatment, where H. dihystera population densities increased 9 to 15-fold at both sites. Population of Paratylenchus spp. was absent or present at low levels at the sites throughout the period. Biological activities antagonistic to R. reniformis at Kunia were estimated at the end of 6 months by comparing the extent of nematode''s reproduction (on cowpea seedlings) in the treatment soils that had been subjected to autoclaving or freezing temperature. Although higher indices of antagonistic activities were observed in soils with prior plant-cover treatments than in soils from the controls, none of the treatments resulted in conferring soils the increased ability to suppress re-introduced R. reniformis populations or enhance subsequent pineapple-plant growth. 相似文献

The host-parasite relationships of asparagus and Meloidogyne spp. were examined under greenhouse and microplot conditions. Meloidogyne species and races differed greatly in their ability to reproduce on asparagus seedlings. Meloidogyne hapla generally failed to reproduce, and M. javanica, M. arenaria race 1, and M. incognita race 3 reproduced poorly, with a reproduction factor (Rf = final population/initial population) usually < 1.0. Only M. arenaria race 2 and M. incognita races 1 and 4 reproduced consistently on all asparagus cultivars tested (Rf typically 1-11). No effect of M. incognita race 4 on host growth was detected. Nematodes found in Ips paraconfusus from ponderosa pine in California were an undescribed species of Parasitaphelenchus, Contortylenchus elongatus, C. reversus, and C. brevicomi. C. elongatus, the most commonly found contortylenchid, was present in 98.2% of the contortylenchid-parasitized beetles. Only one nematode parasite of the gut, a Parasitorhabditis sp., was isolated. Although significant differences in parasitism were observed, they were by collection sites, rather than by elevation or bole sources (slash or standing). Significant changes in parasitism between fall and spring collections were observed but not at every site. Nematode parasitism in the F₁ generation of I. paraconfusus by Parasitaphelenchus, Contortylenchus, or Parasitorhabditis increased or decreased from the parent generation depending upon the experiment.Nematode parasites from I. pini included an undescribed Parasitaphelenchus sp., two undescribed Contortylenchus spp., C. reversus and Parasitylenchus (= Neoparasitylenchus) ovarius from the hemocel, and Parasitorhabditis ipini from the gut. Parasitaphelenchus sp. was found in 99% and 45.3% of the beetles from Idaho and California, respectively. Meloidogyne chitwoodi races 1 and 2 and M. hapla reproduced on 12 cultivars of Brassica napus and two cultivars of B. campestris. The mean reproductive factors (Rf), Rf = Pf at 55 days ÷ 5,000, for the three nematodes were 8.3, 2.2, and 14.3, respectively. All three nematodes reproduced more efficiently (P < 0.05) on B. campestris than on B. napus. Amending M. chitwoodi-infested soil in plastic bags with chopped shoots of Jupiter rapeseed reduced the nematode population more (P < 0.05) than amendment with wheat shoots. Incorporating Jupiter shoots to soil heavily infested with M. chitwoodi in microplots reduced the nematode population more (P < 0.05) than fallow or corn shoot treatments. The greatest reduction in nematode population density was attained by cropping rapeseed for 2 months and incorporating it into the soil as a green manure. 相似文献

Pratylenchus and Xiphinema species have been associated with decline and mortality of brambles (Rubus species) in the Pacific Northwest of the United States. These nematodes cause direct feeding damage and (or) transmit viruses that result in poor fruit quality and plant decline. A nematode management program has been developed by the author to minimize chemical use and nematode-induced damage while optimizing fruit production. Nucleic acid hybridization among root-knot nematode mitochondrial DNAs can be used to identify several Meloidogyne species. Research was initiated to optimize mitochondrial DNA-based molecular diagnostics for the demanding environments likely to be encountered in field isolates. DNA hybridization using reconstituted DNA-soil mixtures revealed a loss of assay sensitivity ranging from 34% to 92% with four agronomic soils tested. This problem was alleviated by the addition of exogenously added DNA. Variation in nematode egg lysis procedures also affected hybridization efficiency, with NaOC1 treatment most effective at disrupting Meloidogyne eggs. These optimized conditions permit detection of mtDNA released from one to five Meloidogyne eggs using standard nucleic acid hybridization procedures. 相似文献

The development of Meloidogyne platani on sycamore was followed for 40 days (22-28 C). Juveniles penetrated the feeder roots behind the root cap and invaded the vascular cylinder within 3 days after inoculation. All subsequent development of the nematodes and host effects occurred only within the stele. The second juvenile molt and sex differentiation occurred by the 17th day. Young females were observed by the 26th day. Eggs were observed inside the roots by the 35th day and were exposed to the surface of galls by the 40th day. In pathogenicity studies, a significant negative correlation was shown to exist between fresh shoot and root weights and inoculum density. Besides sycamore, white ash was the only hardwood species tested to become infected. Of the herbacious plants tested, tobacco was heavily galled, tomato and watermelon moderately galled, and pepper only slightly galled. Research was initiated to physically characterize the mitochondrial genomes of several Meloidogyne spp. and host-races, to address questions regarding their systematics and dispersal, and to assess the possibility of developing molecular diagnostics for these nematodes. Techniques were developed for purification and rapid detection of mitochondrial DNA from root-knot nematodes. Mitochondrial DNAs among Meloidogyne spp. were demonstrated to exhibit extensive divergence. The potential for using the rapidly diverging mitochondrial genomes as a diagnostic assay for M. incognita, M. hapla, M. arenaria, and M. javanica is discussed. 相似文献

Temperature gradient fluctuations that occur naturally as a result of heating and cooling of the soil surface were reproduced within 15-cm-d, 15-cm-long acrylic tubes filled with moist sand. Sunny and rainy periods during the late summer in eastern Texas were simulated. Five ecologically different nematode species were adapted to fluctuating temperatures for 20-36 hours at a simulated depth of 12.5 cm before being injected simultaneously into the centers of tubes at that depth. When heat waves were propagated horizontally to eliminate gravitational effects, the movement of Ditylenchus phyllobius, Steinernema glaseri, and Heterorhabditis bacteriophora relative to the thermal surface was rapid and largely random. However, Rotylenchulus reniformis moved away from and Meloidogyne incognita moved toward the thermal surface. When heat waves were propagated upward or downward, responses to temperature were the same as when propagated horizontally, irrespective of gravity. The initial direction of movement 1.5 hours after introduction to 20-era-long tubes at five depths at five intervals within a 24-hour cycle indicated that M. incognita moved away from and R. reniformis moved toward the temperature to which last exposed. Differences in movement of the five species tested relative to gravity appeared related to body length, with the smallest nematodes moving downward and the largest moving upward. 相似文献

Ten populations of Xiphinema americanum-group nematodes were reared from individual females to evaluate inter- and intraspecific variation under identical host and environmental conditions. Data indicated that morphometric variability of X. americanum was the result of genetic variation rather than phenotypic plasticity and that genetic heterogeneity was greater than previously thought. Morphometrics of single female derived (SFD) populations identified different genotypes present in the field populations. Stylet length was the least variable morphometric character of SFD populations, but collectively stylet measurements of all individuals formed an uninterrupted continuum ranging from 107-148 μm. Range and frequency of stylet measurements of field populations could be accounted for by the relative proportion of different genotypes in the population. Seven field populations of Xiphinema americanum sensu lato from California''s major agronomic areas were tested for their ability to transmit two nepoviruses, including the prune brownline, peach yellow bud, and grapevine yellow vein strains of tomato ringspot virus and the bud blight strain of tobacco ringspot virus. Two field populations transmitted all isolates, one population transmitted all tomato ringspot virus isolates but failed to transmit bud blight strain of tobacco ringspot virus, and the remaining four populations failed to transmit any virus. Only one population, which transmitted all isolates, bad been associated with field spread of a nepovirus. As two California populations of Xiphinema americanum sensu lato were shown to have the ability to vector two different nepoviruses, a nematode taxonomy based on a parsimony of virus-vector relationship is not practical for these populations. Because two California populations of X. americanum were able to vector tobacco ringspot virus, commonly vectored by X. americanum in the eastern United States, these western populations cannot be differentiated from eastern populations by vector capability tests using tobacco ringspot virus. 相似文献
