S ummary . Pelodera chitwoodi was able to distinguish between a Vibrio sp. (food bacterium) and Chromobacterium janthinum (non-food bacterium), and between bacteria and mud. Nematodes migrated preferentially to the Vibrio sp. at various distances, temperatures, and pH values. Migration to food bacteria increased with an increase in bacterial concentration. Resting and dead bacteria failed to attract nematodes. Nematodes were able to detect a vibrio colony separated from them by a plastic barrier, and migrated to areas on agar containing exudate and dialysate from the vibrio. Both exudate and dialyzate contained 4 amino acids and 3 vitamins which attracted nematodes. However, attraction of nematodes to these compounds was not as strong as to metabolizing bacteria, indicating that additional compounds were also involved.  相似文献

A microrespiration device is decribed which uses a Clark electrode to measure the oxygen consumption or production of small and microscopic aquatic organisms in an open flow system. The construction and working principles of the device, which can measure oxygen consumptions as low as 0.5 nl · h−1, are described. The design of the apparatus permits parallel measurements under identical conditions with a single electrode. The device can be matched to various sizes of animal and oxygen consumption rates by means of specimen chambers of different volumes (6 μl, 35 μl, 140 μl) and a variable water flow rate. The microflow respiration device has been used successfully to measure the respiration of zooplankton and meiobenthos organisms as well as protozoans and has also been used successfully on board a research vessel.  相似文献

Dark respiration rates of guard-cell protoplasts of Commelina communis L. were measured over a temperature range (15–30° C) using a Cartesian-diver microrespirometry technique. Measurements were made using a few microliters of suspension medium containing between 400 and 3 700 protoplasts. Respiration rates were approximately linear for at least 1 h at all temperatures. Respiration rates increased rapidly between 20 and 25° C to relatively high levels (6.11·10-6 mol O2 h-1 protoplast-1=1259 mol O2 mg-1 chlorophyll h-1=22.97 mol O2 mg-1 protein h-1) with no further increases above this temperature. Respiration rates were much lower in protoplasts 15–16 h old than in freshly prepared ones indicating considerable deterioration of their viability over this time period.  相似文献

The numerical abundance and biomass of free-living marine nematodes from sandy sediment off the coast of Peru (water depth 40 m) was similar to that reported from other areas. Dispersion chi-square analysis indicated that three species were aggregated, while the remaining species were randomly distributed throughout the samples. The most reasonable hypothesis is that variation in size of individual pore spaces is the mechanism controlling the distribution of the aggregated species.  相似文献

An improved method for the fixation of the third and fourth larval stages and adults of Caenorhabditis clegans has been developed. Worms are placed in a mixture of 1.5% paraformaldehyde and 1.0% glutaraldehyde at pH 7.0 and 70 C and the suspension promptly cooled in a water bath at 20 C for 1 hr. The fixed worms are then immersed in a mixture of 5% glutaraldehyde and hydrogen peroxide at 4 C for 1 hr, stained en bloc in uranyl acetate, and embedded in resin for electron microscopy. The procedure results in superior fixation, particularly of micro filaments and micro tubules. The high temperature of the initial fixation straightens the worms and thus facilitates serial sectioning.  相似文献

This paper describes the purification of yolk proteins, which are important for the reproduction of egg-laying animals, and the structural characterization of two vitellogenins, VT1 and OTI-VIT-6, of the nematode Oscheius tipulae. O. tipulae is an alternative model organism to its relative, the widely used Caenorhabditis elegans, and is a good model to understand reproduction in insect parasitic nematodes of the genus Heterorhabditis. The native purified O. tipulae vitellogenin is composed of three polypeptides (VT1, VT2 and VT3), whereas in C. elegans, vitellogenin is composed of four polypeptides. The gene (Oti-vit-1) encoding yolk polypeptide VT1 has been recently identified in the genome of O. tipulae. Immunoblotting and N-terminal sequencing confirmed that VT1 is indeed coded by Oti-vit-1. Utilizing the same experimental approaches, we showed that the polypeptides VT2 and VT3 are derived from the proteolytic processing of the C- and N-terminal portions of the precursor OTI-VIT-6, respectively. We also showed that the recombinant polypeptide (P40), corresponding to the N-terminal sequence of OTI-VIT-6, preferentially interacts with a 100-kDa polypeptide found in adult worm extracts, as we have previously shown for the native vitellins of O. tipulae. Using the putative nematode vitellogenin amino acid sequences available in the UniProtKB database, we constructed a phylogenetic tree and showed that the O. tipulae vitellogenins characterized in this study are orthologous to those of the Caenorhabditis spp. Together, these results represent the first structural and functional comparative study of nematode yolk proteins outside the Caenorhabditis genus and provide insight into the evolution of these lipoproteins within the Nematode Phylum.  相似文献

An AluI satellite DNA family has been isolated in the genome of the root-knot nematode Meloidogyne chitwoodi. This repeated sequence was shown to be present at approximately 11,400 copies per haploid genome, and represents about 3.5% of the total genomic DNA. Nineteen monomers were cloned and sequenced. Their length ranged from 142 to 180 bp, and their A + T content was high (from 65.7 to 79.1%), with frequent runs of As and Ts. An unexpected heterogeneity in primary structure was observed between monomers, and multiple alignment analysis showed that the 19 repeats could be unambiguously clustered in six subfamilies. A consensus sequence has been deduced for each subfamily, within which the number of positions conserved is very high, ranging from 86.7% to 98.6%. Even though blocks of conserved regions could be observed, multiple alignment of the six consensus sequences did not enable the establishment of a general unambiguous consensus sequence. Screening of the six consensus sequences for evidence of internal repeated subunits revealed a 6-bp motif (AAATTT), present in both direct and inverted orientation. This motif was found up to nine times in the consensus sequences, also with the occurrence of degenerated subrepeats. Along with the meiotic parthenogenetic mode of reproduction of this nematode, such structural features may argue for the evolution of this satellite DNA family either (1) from a common ancestral sequence by amplification followed by mechanisms of sequence divergence, or (2) through independent mutations of the ancestral sequence in isolated amphimictic nematode populations and subsequent hybridization events. Overall, our results suggest the ancient origin of this satellite DNA family, and may reflect for M. chitwoodi a phylogenetic position close to the ancestral amphimictic forms of root-knot nematodes. To understand the roles of nematodes in organic matter (OM) decomposition, experimental setups should include the entire nematode community, the native soil microflora, and their food sources. Yet, published studies are often based on either simplified experimental setups, using only a few selected species of nematode and their respective prey, despite the multitude of species present in natural soil, or on indirect estimation of the mineralization process using O2 consumption and the fresh weight of nematodes. We set up a six-month incubation experiment to quantify the contribution of the entire free living nematode community to carbon (C) mineralization under realistic conditions. The following treatments were compared with and without grass-clover amendment: defaunated soil reinoculated with the entire free living nematode communities (+Nem) and defaunated soil that was not reinoculated (-Nem). We also included untreated fresh soil as a control (CTR). Nematode abundances and diversity in +Nem was comparable to the CTR showing the success of the reinoculation. No significant differences in C mineralization were found between +Nem and -Nem treatments of the amended and unamended samples at the end of incubation. Other related parameters such as microbial biomass C and enzymatic activities did not show significant differences between +Nem and -Nem treatments in both amended and unamended samples. These findings show that the collective contribution of the entire nematode community to C mineralization is small. Previous reports in literature based on simplified experimental setups and indirect estimations are contrasting with the findings of the current study and further investigations are needed to elucidate the extent and the mechanisms of nematode involvement in C mineralization.  相似文献

Two species of free-living nematodes, Panagrellus redivivus and Turbatrix aceti, were cultured axenically at control (20 C) and cold (10 C) temperatures. Oxygen consumption of worms from each population was measured manometrically on days 2 through 8 after exposure to these temperatures. In both species, the slope of the oxygen consumption curve for the controls was greater than that of the worms exposed to the cold on day 2. The slope of the curve of the cold-exposed worms gradually increased until day 7. At this time, the slope of the oxygen consumption curve from the cold-exposed worms exceeded or equaled that of the controls. This is taken as an indication of the onset of the cold-acclimated state in both species of worms by day 7.  相似文献

Aquatic oligochaete species selected on the basis of pollution tolerance or intolerance as determined from field studies were used to determine respiration rates under standard conditions, after exposure to sublethal levels of environmental variables (pH, salinity, temperature) or toxicants (Cd, Hg, NaPCP) alone or in pairs. Respiration rates, critical oxygen levels and degree of regulation were not correlated with field-determined tolerance to organic pollution. Stress factors caused increased or lowered respiration rate, loss of regulation (partial or total) and sometimes shifts in critical oxygen level. Regulation was improved in the face of some stresses. Combined stresses produced additional changes in respiration rates and regulation. These responses were complex, which makes them useful indicators of specific stress factors but also indicate complex mechanisms behind the observed changes.  相似文献

The relation between growth rate traits (height, basal diameter, stem volume and branch diameter) and two measures of respiration rate [metabolic heat rate (q) and CO2 production rate (Rco2)] and their ratio (q/Rco2) was examined on a collection of 192 different genotypes of coast redwoods [Sequoia sempervirens (D.Don) Endl.]. Branch diameter was not correlated with any of the respiratory measures, but the other three growth traits gave highly significant (P < 0.001) correlations with positive slopes. Combining the four growth traits and the three respiratory variables (q, RCo2 and q/Rco2) to give two canonical variates, one representing growth and one representing respiration, gives an even stronger linear correlation (r= 0–85). These data suggest that simultaneous assay of multiple respiratory measures on juvenile trees can be used to predict their longer-term growth rates.  相似文献

Meloidogyne chitwoodi n. sp. is described and illustrated from potato (Solanum tuberosum) originally collected from Quincy, Washington, USA. This new species resembles M. hapla, but its perineal pattern is basically round to oval with distinctive and broken, curled, or twisted striae around and above the anal area. The vulva is in a sunken area devoid of striae. Vesicles or vesicle-like structures are present in the median bulb of females. The larva tail, being short and blunt with a hyaline tail terminal having little or no taper to its rounded terminus, is distinctively different from M. hapla. SEM observations revealed the nature of the perineal pattern and details of the head of larvae and males, and showed the spicules to have dentate tips ventrally. Hosts for M. chitwoodi n. sp. include potato, tomato, corn, and wheat but not strawberry, pepper, or peanut. The latter three crops are excellent hosts for M. hapla. The known distribntion of this new root-knot species presently involves certain areas of Idaho, Washington, and Oregon. The common name "Columbia root-knot nematode" is proposed for M. chitwoodi n. sp.  相似文献

In nonhuman primate groups that utilize crop and food-raiding strategies, the inclusion of human food in the diet can have a pervasive impact on the activity budget. Human food is usually of higher quality and energy per unit than wild food. Thus, metabolic demands may be reached sooner on a human food diet, which in turn leads to improved foraging efficiency. In particular, a reduction in time spent feeding is associated with an increase in time spent resting. We investigated changes in time budget in a group of vervets (Chlorocebus aethiops pygerthrus) living in a tourist and cultivated area of Entebbe, Uganda (Zoo group). Saj collected 123 hr of focal observations on 17 individuals. Human food constituted half of the diet of Zoo group. Access to human food resulted in a high proportion of time resting and a low proportion of time feeding. The group had a small average daily range and a small home range in relation to other reports on unprovisioned groups. The time budget differences suggest that the juveniles spent more time moving and feeding and less time resting than adults did, and that adult females were more social than adult males were. These patterns have been observed in both provisioned and unprovisioned vervet groups. In contrast to a report from an unprovisioned group (Harrison, 1983), lactating females in this provisioned situation were able to maintain feeding and activity levels similar to those of other adults. However, adult females spent significantly less time consuming human food than juveniles did. We suggest that the risk associated with human food consumption may explain the low consumption of human food by females with dependent offspring. The differences may also reflect the different nutritional requirements of lactating females or the tendency for juveniles to be more exploratory than other group members.  相似文献

Recent, parallel developments in the study of freshwater and marine ecosystems have provided evidence that net heterotrophic systems (those in which respiratory organic matter destruction exeeds photosynthetic production) are more prevalent than hitherto believed, including most rivers, oligo- to mesotrophic lakes and some oligotrophic regions of the ocean. In parallel, these aquatic ecosystems have been shown to act as CO2 sources to the atmosphere, as expected from the heterotrophic nature of the communities they contain. The prevalence of net heterotrophic aquatic ecosystems indicates that they must receive significant inputs of organic carbon from adjacent ecosystems, assigning an important role to the lateral exchanges of carbon between land and aquatic ecosystems, between coastal and open ocean ecosystems, as well as internal redistribution within large or complex aquatic ecosystems in determining their metabolic status and the gaseous exchange with the atmosphere. The examination of the carbon budget of ecosystems requires, therefore, an integrative approach that accounts for exchanges between compartments often studied in isolation. These recent findings conform a new paradigm of the functioning of aquatic ecosystems, and the metabolic connectivity between ecosystems in the biosphere.  相似文献

The insect-parasitic female of Sphaerularia bombi everts its uterus and associated reproductive structures into the body cavity of its bumblebee host. This uterine sac then takes over the normal functions of the parasite and leads an independent existence. An examination of this sac shows that the surface of the uterine cells are differentiated into a network of saccular indentations separating off fine cytoplasmic extensions. The folding of adjacent cytoplasmic extensions around portions of the host''s hemolymph results in the formation of pinocytotic vacuoles. Intracellular vacuoles are also formed at the base of the saccular indentations. It appears that the first stage of intracellular digestion in S. bombi initially occurs by pinocytosis in the outer surface of the uterine cells. The inner and outer surfaces of the ovary and oviduct are modified into lobelike projections to increase the absorptive surface area, and electron-dense droplets originating in this tissue were also observed in the developing eggs. The hypothesis is presented that soluble nutrients are passed into the ovary-oviduct tissue where, after being reconstituted into droplets, they enter the developing eggs.  相似文献

A mechanistic model is presented for determination of antimicrobial presence in the environment, using antimicrobial characteristics and Irish-specific usage data. The model simulates release of antimicrobials into the aquatic environment by integrating the effects of antimicrobial use, metabolism, degradation, and dilution. Predicted environmental concentrations (PECs) were ranked in relation to their potential to select for resistant bacteria, toxicity (chronic and acute), and hazard quotient (HQ). The simulated mean PECs of penicillins (PEN), β-lactams (BET), tetracyclines (TET), macrolides (MAC), quinolone/fluoroquinolones (Q/F), and sulphonamides/trimethoprim (S/T) were 1.05, 0.19, 0.06, 0.07, 0.02, and 0.08 mg/m3, respectively. All antimicrobials, excluding Q/F, showed a low HQ (<1) indicating low toxicity. Q/F had a HQ of 1.51 indicating moderate toxicity. All antimicrobial groups expressed varying resistance formation potential with Q/F having the highest. Q/F also exhibited the lowest degradation rate. A sensitivity analysis indicated a possible role for considering metabolism during regulation of new antimicrobials as this can significantly influence PEC values. The observed results suggest that current antimicrobial use can lead to levels in the environment that may increase resistance formation. The results and limitations presented here accentuate the need for further investigation into antimicrobials in the environment and contribution to resistant strains.  相似文献
