Stentor niger collected in the suburbs of Hiroshima contained in its cytoplasm several hundreds of endosymbiotic algae and innumerable brownish pigment granules. The body of the ciliate was dark due to a mixture of the green endosymbiotic algae and brown pigment granules. The algae belonged to the genus Chlorella; each was enclosed in a perialgal vacuole and dispersed uniformly in the host cytoplasm from the myoneme layer inward to the center of the ciliate. The cell wall and plasma membrane of the alga enclosed a nucleus, chloroplast, mitochondrion, Golgi complex, accumulation bodies, myelinated vesicles, and many ribosomes. The chloroplast occupied more than half of the volume of the alga and contained a conspicuous pyrenoid. Algal multiplication occurred by two successive divisions of an alga, leading to four autospores within a perialgal vacuole; the walls of the vacuole invaginated to separate the autospores each into its own vacuole. Three types of pigment granules were scattered uniformly throughout the cytoplasm of the ciliate. The ultrastructure of the membranellar region, somatic cortex, and macro- and micronucleus of the ciliate are also described.  相似文献

The low frequency of nontuberculous mycobacterial infections, nonspecific symptoms for individual mycobacteria, and the lack of specific identification methods could alter correct diagnosis. This study presents a combined microbiology and molecular-based approach for Mycobacterium?marinum detection in four aquarists with cutaneous mycobacterial infection. Simultaneously, ecology screening for M.?marinum presence in the aquarists' fish tanks was performed. A total of 38 mycobacterial isolates originated from four human patients (n?=?20), aquarium animals (n?=?8), and an aquarium environment (n?= 10). Isolate identification was carried out using 16S?rRNA sequence analysis. A microbiology-based approach, followed by 16S rRNA sequence analysis, was successfully used for detection of M.?marinum in all four patients. Animal and environmental samples were simultaneously examined, and a total of seven mycobacterial species were isolated: Mycobacterium chelonae , Mycobacterium fortuitum , Mycobacterium gordonae , Mycobacterium?kansasii , Mycobacterium mantenii , Mycobacterium marinum , and Mycobacterium?peregrinum . The presence of M.?marinum was proven in the aquarium environments of two patients. Although M.?marinum is described as being present in water, it was detected only in fish.  相似文献

The pigment floridorunin was localized to the cuticle of the red alga Lenormandia prolifera (C. Ag.) J. Agardh by x-ray micro-analysis in Ihe transmission electron microscope and by its colour reactions in the light microscope. The pigment was set free from the cell wall by a pectinase. Bromine was also identified in the chloro-plasts. the middle lamellae, the intercellulars and the pore plugs of the alga. The content of bromophenols in the alga increases with increasing age. The cell walls of old plants are stratified and their outer parts are apparently shed. The bromophenols could have a function as regulators of the epiphytes or the shedding of parts of the outer cell wall.  相似文献

Phenotypic switching is characterized as a virulence factor of Candida spp. This study was carried out to evaluate the phenotypic switching ability of C.?krusei ATCC 14243 and to determine its effect on the biological properties, adherence capacity and susceptibility towards chlorhexidine digluconate (CHX). To induce switched generations C.?krusei was cultured under nitrogen-depleted growth conditions by adding phloxine B. These phenotypically switched colonies were designated as the 1st generation. Subsequent sub-culturing was performed to produce the 2nd, 3rd and 4th switched generations. The recovery of the 3rd generation was the highest at 85.7% while that of the 4th generation was lower at 70.8%, and the recovery of the 1st and 2nd generations gradually reduced to 46.6% and 36.4%, respectively. All generations of C.?krusei were susceptible towards CHX. The unswitched C.?krusei was the most susceptible but the least adherent to coated hard surfaces. The 2nd generation was the least susceptible, but with the highest adherent ability. The minimum inhibition concentration and minimal fungicidal concentration of C.?krusei of all generations were determined at 0.4?mg?mL(-1) . These observations suggest that the switching activity of C.?krusei induces changes to its biological properties and susceptibility towards CHX.  相似文献

1.?Many host-pathogen systems show regular seasonal oscillations. 2.?Seasonal variation in mycoplasmal conjunctivitis prevalence in house finches is an example of such oscillations. 3.?An annual pulse of Mycoplasma gallisepticum-na?ve juveniles increasing the number of susceptibles, seasonal changes in flocking behaviour increasing transmission rate and a gradual loss of resistance to reinfection with time are sufficient to model the observed seasonal variation in disease prevalence. Nevertheless, experiments are needed to test the underlying mechanisms. 4.?We carried out an 18-month experiment with small groups of birds in large aviaries to test two hypotheses. 5.?To test the first hypothesis that an influx of na?ve juveniles in a group of recovered adults is sufficient to cause an outbreak, we added eight juveniles to a group of 11 adults that had recovered from an earlier infection. In all, three replicates juveniles became infected, but only after some of the adults relapsed. 6.?To test the second hypothesis that reintroduction of M.?gallisepticum into a multiage group of previously exposed but fully recovered house finches causes a new outbreak, we inoculated two birds in each group in March of the 2nd year. Contrary to what happens in the wild at that time disease prevalence increased rapidly after reintroduction of M.?gallisepticum. 7.?We conclude that asymptomatic, recovered adults can initiate an epidemic and transmit M.?gallisepticum to na?ve house finches and that the reintroduction of M.?gallisepticum is sufficient to cause a new outbreak, even at a time of the year when mycoplasmal conjunctivitis is low in free-living birds. Date, as such, seems to be less important to explain seasonal variation in conjunctivitis than the presence of na?ve juveniles or the introduction on M.?gallisepticum. 8.?Seasonality in outbreaks is most likely tightly linked to seasonal variation in bird movements and behaviour.  相似文献

The unicellular green alga Haematococcus pluvialis Flotow has recently aroused considerable interest due to its capacity to amass large amounts of the ketocarotenoid astaxanthin (3,3'-dihydroxy- β , β -carotene-4,4'-dione), widely used commercially to color flesh of salmon. Astaxanthin accumulation in Haematococcus is induced by a variety of environmental stresses which limit cell growth in the presence of light. This is accompanied by a remarkable morphological and biochemical 'transformation' from green motile cells into inert red cysts. In recent years we have studied this transformation process from several aspects: defining conditions governing pigment accumulation, working out the biosynthetic pathway of astaxanthin accumulation and questioning the possible function of this secondary ketocarotenoid in protecting Haematococcus cells against oxidative damage. Our results suggest that astaxanthin synthesis proceeds via cantaxanthin and that this exceptional stress response is mediated by reactive oxygen species (ROS) through a mechanism which is not yet understood. The results do not support in vivo chemical quenching of ROS by the pigment, although in vitro it was shown to quench radicals very efficiently. The finding that most of the pigment produced is esterified and deposited in lipid globules outside the chloroplast further supports this assumption. We have suggested that astaxanthin is the by-product of a defense mechanism rather than the defending substance itself, although at this stage one cannot rule out other protective mechanisms. Further work is required for complete understanding of this transformation process. It is suggested that Haematococcus may serve as a simple model system to study response to oxidative stress and mechanisms evolved to cope with this harmful situation.  相似文献

The objective of this study was to evaluate the ability of Microthrix parvicella for long-chain fatty acids uptake under anaerobic, anoxic, and aerobic conditions as well as its ability to utilize volatile fatty acids and long-chain fatty acids under anoxic and aerobic conditions. According to the results, a hypothesis on the competition between floc-forming microorganisms and M.?parvicella for long-chain fatty acids uptake under aerobic, anoxic, and anaerobic conditions was formulated. According to this hypothesis, M.?parvicella exhibits similar long-chain fatty acids uptake capacity with floc-forming microorganisms even at relatively high floc loading values that are very often imposed at selector tanks. Following this hypothesis, the failure of selector tanks to provide for an effective M.?parvicella control is evidenced. Based on the experimental results, the ability of M.?parvicella to utilize long-chain fatty acids with rates comparable to those of floc formers, even in anoxic conditions, in conjunction with its lower acetate utilization rates, provides a good explanation regarding its preference to slowly biodegradable organic carbon compounds.  相似文献

More than three-quarters of the Earth's surface is occupied by cold ecosystems, including the ocean depths, and polar and alpine regions. These permanently cold environments have been successfully colonized by a class of extremophilic microorganisms that are known as psychrophiles (which literally means cold-loving). The ability to thrive at temperatures that are close to, or below, the freezing point of water requires a vast array of adaptations to maintain the metabolic rates and sustained growth compatible with life in these severe environmental conditions.  相似文献

An intertidal Chlorella-like alga Mychonastes desiccatus Brown sp. nova, capable of forming achlorophyllous desiccation-resistant cysts, has been grown in unialgal culture. This small alga was first isolated from a dried sample of a well-studied microbial mat. The mat, located at North Pond, Laguna Figueroa, San Quintin, Baja California, Mexico, is a vertically-stratified microbial community which forms laminated sediments. Morphology, pigment composition and G+C content are within the range typical for the genus Chlorella s. 1. Unlike other chlorellae, however, upon desiccation M. desiccatus forms an achlorophyllous, lipid-filled cyst (thick-walled resting stage) in which no plastid is evident. Rewetting leads to chloroplast differentiation, excystment and recovery of the fully green alga. During desiccation, sporopollenin is deposited within a thickening cell wall. Encystment cannot be induced by growth in the dark. The formation of desiccation-induced cysts allows the alga to survive frequent and intermittent periods of dryness. These chlorellae tolerate wide ranges of acidity and temperature; they both grow and form cysts in media in which sodium ions are replaced with potassium. Although the cysts tolerate crystalline salts, the cell grow optimally in concentrations corresponding from three-quarters to full-strength seawater.  相似文献

Because intertidal organisms live close to their environmental tolerances, intertidal species are considered early harbingers of the effects of global change. Using a combination of spatial environmental gradients and long-term datasets, I examine the likely consequences of global warming on the distribution of Mazzaella parksii (formerly Mazzaella cornucopiae ), a turf-forming red alga. On Tatoosh Island, WA, M. parksii is restricted almost entirely to north-facing surfaces. On south-facing slopes, the upper limit of M. parksii , which is set by abiotic stress, occurs much lower on the shore. However, its lower limit, which is set by herbivores, does not vary with aspect. Because its upper and lower limits are independent across the aspect gradient, M. parksii is effectively "squeezed out" of the system as one moves from a north-facing to a south-facing slope. This general pattern is observed at several spatial scales across gradients of wave exposure and air temperature. The vertical distributional patterns of M. parksii also shift across temporal gradients. Over the past 25 years, the upper limit of M. parksii on Tatoosh Island has shifted downshore by approximately 25 cm. Most of this change was driven by three consecutive harsh summers in the mid 1990's. These temporal patterns, combined with the insight gained from spatial patterns, suggest that M. parksii may go locally extinct in many parts of its range as its upper and lower limits converge. Furthermore, because vertical distributional shifts are linked to stochastic climatic effects, temporal change in marine algal populations may be rapid and unexpected.  相似文献

The marine red alga Gracilaria crassa was investigated for its proximate composition, minerals, fatty acids, amino acids, and agar content to decipher its nutritional implications. The growth performance and pigments were studied under different combinations of temperature and salinity. On a dry weight basis the total lipid content was 1.30?±?0.05 %, protein was 5.18?±?0.64 %, carbohydrate was 42.0?±?1.2 %, ash was 43.18?±?1.15 %, and agar content was 21.52?±?0.73 %. Appreciable amounts of macro-, micro-nutrients (K?>?Na, Ca, Mg, and Fe), and essential amino acids (Ileu, His, Thr, Leu, and Lys) were found. Palmitic, stearic acid, and arachidonic acid were major fatty acids detected. The alga showed maximum daily growth rate (DGR %) 5.8?±?0.09 % at 25 °C, 35 ‰ salinity. The highest content of pigment R-phycoerythrin (444.7?±?1.9 μg g?1 fresh weight (FW) basis) was obtained at 25 ‰ salinity at 35 °C while that of R-phycocyanin (476.3?±?2.3 μg g?1 DW) at 30 ‰ salinity at 30 °C. This study revealed that this alga can be utilized as a potential source for food and feed. The data generated on best growth conditions will be very useful for farming of G. crassa in open sea. This alga could be used for production of natural colorants at defined control condition.  相似文献

The extremophilic microalga Chlamydomonas acidophila inhabits very acidic waters (pH 2-3.5), where its growth is often limited by phosphorus (P) or colimited by P and inorganic carbon (CO(2)). Because this alga is a major food source for predators in acidic habitats, we studied its fatty acid content, which reflects their quality as food, grown under a combination of P-limited and different carbon conditions (either mixotrophically with light + glucose or at high or low CO(2), both without glucose). The fatty acid composition largely depended on the cellular P content: stringent P-limited cells had a higher total fatty acid concentration and had a lower percentage of polyunsaturated fatty acids. An additional limitation for CO(2) inhibited this decrease, especially reflected in enhanced concentrations of 18:3(9,12,15) and 16:4(3,7,10,13), resulting in cells relatively rich in polyunsaturated fatty acids under colimiting growth conditions. The percentage of polyunsaturated to total fatty acid content was positively related with maximum photosynthesis under all conditions applied. The two factors, P and CO(2), thus interact in their effect on the fatty acid composition in C. acidophila, and colimited cells P-limited algae can be considered a superior food source for herbivores because of the high total fatty acid content and relative richness in polyunsaturated fatty acids.  相似文献

Sarcocornia fruticosa (L.) A.J. Scott is found in coastal marshes of south-west Spain, growing under a very wide range of interstitial soil salinity from 10 m M up to nearly 1000 m M . A glasshouse experiment was designed to investigate the effect of this range of salinities on the morphology and the photosynthetic apparatus of S. fruticosa by measuring growth rate, photosynthetic and non-photosynthetic area, atrophy of distal branch ends, water status, chlorophyll fluorescence parameters, gas exchange and photosynthetic pigment concentrations. The long-term effects of salinity on the growth of S. fruticosa were mainly determined by the extent of photosynthetic area rather than the variations in net photosynthetic rate. Photosynthetic area was reduced at 1030 m M as a result of a decrease in the length of the photosynthetic portions. This was induced by fewer internodes and, at salinities lower than 510 m M , smaller internode diameter. Net photosynthetic rate increased as the quantum efficiency of photosystem II decreased in the different salinity treatments, which means that the plant could be increasing photorespiration and/or using cyclic electron transport as additional photoprotective mechanisms. The recorded drop in net photosynthetic rate at higher salinities appeared to be due to a reduction in stomatal conductance. The results indicate that S. fruticosa is capable of tolerating very high and continued exposure to salt, showing its greatest growth rate at 510 m M NaCl.  相似文献

Ecological studies were carried out on 19 endangered populations of Meconopsis horridula, M. paniculata, M. simplicfolia, M. sinuata and M. villosa from the Sikkim Himalayas. The populations of M. horridula, M. paniculata and M. sinuata inhabit open alpine slopes, those of M. simplicfolia are restricted to alpine meadows, while those of M. villosa are confined to the interiors of temperate forests and along forest fringes. The habitats of these species remain snow-hound from October to February. Analyses suggest that all the species except M. horridula , have a broad area of distribution, but narrow habitats and niches. All five species are confined to moist, clayey to silty soils with acidic pH, high concentrations of organic matter, nitrate, phosphate, potassium and calcium. M. paniculata is a 'generalist', whereas all other species investigated are 'specialists'.  相似文献

Population estimations are necessary for effective conservation management. In Italy, brown hare populations are commonly censused by spotlighting, but this method does not seem to suit an alpine environment due to its vegetational and orographical complexity. The aim of this study is to evaluate the critical aspects related to spotlight census method in an alpine environment. Spotlight was carried out along two transects of a typical alpine environment. Observed animals were used to define density (number of animal seen/100 ha) and method precision (coefficient of variation (CV) applied to monthly repeated observations). Animal detectability was evaluated using half-normal function with cosine expansion (Distance 5.0®). Animal observability was evaluated by analyzing density estimates related to habitat conformation (unseen areas or full visibility). The exact surface surveyed by spotlighting was evaluated, defining the observation spotlight beam range (OTA) and the land useful sighting (LUS). In the end, LUS was classified in three patches according to hare presence: no hares, occasional hare presence, and constant hare presence. To evaluate habitat influence onto CV, we used a bootstrap simulation. The results show that spotlighting alone is not the most suitable method to apply in the alpine environment because habitat structure highly influences census results. Recommendations to improve spotlight surveys for monitoring European brown hare populations are given.  相似文献

The prasinophycean alga Mantoniella contains, in addition to Chl a and b, at least a third green pigment which is functionally active in the light-harvesting antenna. This third Chl was isolated in order to elucidate its chemical structure. The absorption and fluorescence spectra were measured not only from the purified pigment but also from its pheophytin and its methylpheophorbide. The spectra were compared with those of authentic Chl c-1 and c-2, which were isolated from the diatom Nitzschia sp. and with Mg-DVPP (purified from Rhodobacter). The results show that the pigment from Mantoniella compares best with Chl c-1. In order to clarify the spectral data, Chl c-1 and c-2, Mg-DVPP, and the pigment from Mantoniella were subjected to a chromatographic system that is able to separate these porphyrins. The chromatographic analysis clearly shows that the pigment from Mantoniella co-migrates with Chl c-1 and not with the bacterial pigment. Mantoniella is the first organism which has been demonstrated to contain Chl a, b, and authentic c.  相似文献

Understanding the impact of grazing intensity on grassland production and soil fertility is of fundamental importance for grassland conservation and management. We thus compared three types of alpine steppe management by studying vegetation traits and soil properties in response to three levels of grazing pressure: permanent grazing (M1), seasonal grazing (M2), and grazing exclusion (M3) in the alpine steppe in Xainza County, Tibetan Plateau. The results showed that community biomass allocation did not support the isometric hypothesis under different grassland management types. Plants in M1 had less aboveground biomass but more belowground biomass in the top soil layer than those in M2 and M3, which was largely due to that root/shoot ratios of dominant plants in M1 were far greater than those in M2 and M3. The interramet distance and the tiller size of the dominant clonal plants were greater in M3 than in M1 and M2, while the resprouting from rhizome buds did not differ significantly among the three greezing regimes. Both soil bulk density and soil available nitrogen in M3 were greater than in M1 at the 15–30 cm soil depth (P = 0.05). Soil organic carbon and soil total nitrogen were greater in M3 than in M1 and M2 (P = 0.05). We conclude that the isometric hypothesis is not supported in this study and fencing is a helpful grassland management in terms of plant growth and soil nutrient retention in alpine steppe. The extreme cold, scarce precipitation and short growing period may be the causation of the unique plant and soil responses to different management regimes.  相似文献

Rhizobia are soil bacteria able to develop a nitrogen-fixing symbiosis with legumes. They are taxonomically spread among the alpha and beta subclasses of the Proteobacteria. Mimosa pudica, a tropical invasive weed, has been found to have an affinity for beta-rhizobia, including species within the Burkholderia and Cupriavidus genera. In this study, we describe the diversity of M.?pudica symbionts in the island of New Caledonia, which is characterized by soils with high heavy metal content, especially of Ni. By using a plant-trapping approach on four soils, we isolated 96 strains, the great majority of which belonged to the species Cupriavidus taiwanensis (16S rRNA and recA gene phylogenies). A few Rhizobium strains in the newly described species Rhizobium mesoamericanum were also isolated. The housekeeping and nod gene phylogenies supported the hypothesis of the arrival of the C.?taiwanensis and R.?mesoamericanum strains together with their host at the time of the introduction of M.?pudica in New Caledonia (NC) for its use as a fodder. The C.?taiwanensis strains exhibited various tolerances to Ni, Zn and Cr, suggesting their adaptation to the specific environments in NC. Specific metal tolerance marker genes were found in the genomes of these symbionts, and their origin was investigated by phylogenetic analyses.  相似文献

SUMMARY. Unlike previously studied lakes with prolonged winter ice and snow cover, Lake Paajarvi, southern Finland, has a high humus content and consequently differs in both the quantity and quality of light penetration into its waters. Moreover, the range of temperature fluctuation and the degree of development of thermal stratification are greater in Paajarvi, and this increased environmental heterogeneity apparently stimulates diversity in the phytoplankton community, especially in the seasonal succession of species. Differences in the photosynthetic capacity of algae from different depths in the water column were not great. This is attributed to the extremely shallow euphotic zone, algae circulating freely through the steep light gradient and sedimenting rapidly once they pass through the thermocline into the hypolimnion. It is suggested that 'adaptation' of phytoplankton to the great seasonal changes in irradiance is achieved largely by successive growths of different species in the community, and that the adaptations and vertical migrations by individual algal species, which have been reported from polar and high alpine lakes, may be of secondary importance in Pääjärvi. The species successions in Pääjärvi produce changes in the pigment content of algae similar to those reported from polar and high alpine lakes, confirming that change in pigmentation is an important mechanism in light adaptation, whether at community or individual level. Algal pigment content was particularly high at the end of the long period of winter ice cover, indicating a degree of adaptation to the prolonged low-light conditions, which produced the extremely high photosynthetic capacities measured at this time. However, phytoplankton production at any irradiance was primarily determined by biomass.  相似文献
