Inoculating Proctor barley leaves with Erysiphe graminis decreasedrates of photosynthesis, after an initial lag period, and increasedrespiration. Increasing the area inoculated progressively decreased ratesof photosynthesis, but the effects cannot be attributed to asimple loss of leaf area. When less than 30 per cent of a leafwas inoculated, decreases were equivalent to area losses greaterthan those inoculated; when more than 30 per cent was inoculatedthe photo-synthetic losses were equivalent to area losses lessthan those inoculated. Although the relative effects of E. graminis on photosynthesisand respiration were of the same order, the absolute effectson photosynthesis were greater than those on respiration. Inoculating30 per cent of a leaf decreased photosynthesis by 5–6mg CO2/dm2/hr from 12.9 in the uninoculated controls to 7.3.Respiration increased by 0.6 mg CO2/dm/hr, from 1.7 to 2.3-  相似文献

Resistance was induced in barley by virulent and avirulent Erysiphe graminis f. sp. hordei and by E. graminis f. sp. tritici. The diameters of fluorescent haloes and papillae at the site of the primary germ tube of the inducers were larger until 12 hours after inoculation with E g. f. sp. tritici than in the corresponding periods after inoculation with E. g. f. sp. hordei. Fluorescence at the site of appressorial lobes of the inducers developed, gradually from 10 and 12 hours after inoculation with E. g. f. sp. hordei and E. g. f. sp. tritici, respectively. Penetration success of single infection units of the challenger was reduced in cells with inducer fluorescence, suggesting that resistance is principally localized to cells previously attacked by the inducer. But penetration success was independent of the average distance to the ten nearest inducer fluorescences, and penetration failures did also occur in cells without inducer fluorescence, suggesting that the induced resistance to some extent is translocated to other epidermal cells. The average diameter of the fluorescent papillae at the site of unsuccessful challenger infection units increased as a result of induced resistance.  相似文献

Horsegram yellow mosaic disease was shown to be caused by a geminivirus; horsegram yellow mosaic virus (HYMV). The virus could not be transmitted by mechanical sap inoculation. Leaf dip and purified virus preparations showed geminate virus particles, measuring 15-18 * 30 nm. An antiserum for HYMV was produced and in enzyme-linked immunosorbent assay (ELISA) and immunosorbent electron microscopy (ISEM) tests HYMV was detected in leaf extracts of fieldinfected bambara groundnut, french bean, groundnut, limabean, mungbean, pigeonpea and soybean showing yellow mosaic symptoms. Bemisia tabaci fed on purified HYMV through a parafilm membrane transmitted the virus to all the hosts listed above but not to Ageratum conyzoides, okra, cassava, cowpea, Croton bonplandianus, Lab-lab purpureus, Malvastrum coromandalianum and tomato. No reaction was obtained in ELISA and ISEM tests between HYMV antibodies and extracts of plants diseased by whitefly-transmitted agents in India such as A. conyzoides yellow mosaic, okra yellow vein mosaic, C. bonplandianus, yellow vein mosaic, M. coromandalianum yellow vein mosaic, tomato leaf curl and cassava mosaic. HYMV was also not found to be related serologically to bean golden mosaic, virus.  相似文献

Deposition of Erysiphe graminis Conidia on a Barley Crop   总被引:1,自引:0,他引:1  
Naturally released Erysiphe graminis conidia were trapped (on horizontal slides, on vertical sticky cylinders and in suction traps) in a barley crop infected with powdery mildew and the numbers of single spores and of clumps of different sizes deposited on the traps were counted. The efficiencies of impaction calculated from deposits and wind speed measurements were higher than expected from mean wind speed measurements. The values were consistent with the hypothesis that spores were removedpredominantly in gusts. More than half the conidia were removed in clumps of two or more spores. The measurements suggest that clumps were more effectively deposited than single spores. The measurements demonstrate that spore release mechanisms can influence spore deposition significantly, especially close to the source.  相似文献

Spring barley cultivars and lines were tested for 3 years in field studies for adult plant resistance against Erysiphe graminis f.sp. hordei. The cultivars Osiris and Asse were selected for further detailed cytological studies and compared with the susceptible cultivar Peruvian. Under controlled greenhouse conditions, the percentage of conidia that had formed a functional haustorium and secondary hyphae (infection efficiency) was reduced in fifth leaves of the adult plant resistant cultivars. On fifth and flag leaves of adult plant resistant cultivars, papillae were formed more frequently under primary germ tubes and appressoria, and fungal penetration was prevented more often than on the susceptible cultivar Peruvian. In ultrastructural studies various types of papillae were observed, but could not be strictly correlated with penetration success or failure of the fungus.  相似文献

The peroxidase activities in leaves from resistant and susceptible cultivars of wheat infected and non-infected by Erysiphe graminis DC were studied. In non-infected wheat, soluble and ionic bound peroxidase activity level was found to be higher in the resistant cultivar than that in the one susceptible to Erysiphe graminis DC. After infecting wheat leaves with Erysiphe graminis DC a remarkable increase in the activity of soluble and ionic bound peroxidases was detected 5 days after inoculation only in the resistant cultivar. In the susceptible cultivar a high increase in the activity of the soluble and ionic bound peroxidases occurred only 15 days after inoculation. Using ion exchange chromatography four peroxidase fractions were obtained from infected susceptible and resistant cultivars as from non-infected ones. The fraction II in non-inoculated resistant cultivars was much higher than that in the susceptible one. This fraction increased after inoculation in both cases reaching a higher level in resistant cultivars. Fraction I was higher in the susceptible cultivar. Electrofocusing profiles of peroxidase from the susceptible and resistant cultivar differed from one another. New peroxidase bands after inoculation appeared only in the resistant cultivar.  相似文献

Isolates of powdery mildew were subjected to recurrent selection on partially resistant barley cultivars for 76 inoculation cycles over three years. Previous intermediate tests on up to 32 cycles for adaptation to overcome the host resistance revealed a trend towards adaptation to the cv. 7204 recurrent host in infection frequency data but not towards cv. Golden Promise or 9319, Subsequent cycles demonstrated adaptation to these latter hosts also.  相似文献

The effect of infection by Erysiphe graminis f.sp. hordei onprotein synthesis in a susceptible cultivar of Hordeum vulgarehas been studied using in vivo labelling. Results have indicateda decline in protein synthesis in the chloroplasts at 1, 3 and5 days after inoculation and in the cytoplasm at 3 and 5 daysafter inoculation. The levels of adenosine, the adenine nucleotides AMP, ADP, ATP, as well as NAD, NADP, protein and chlorophyll were determined in young barley plants of which the primary leaves were infected by Erysiphe graminis DC. f. sp. hordei MARCHAL. The largest changes of these metabolite levels, compared to the non-infected control, occurred in the infected leaf and to a lesser degree also in the roots and in healthy younger leaves. The increase in the levels of most metabolites in the primary leaf revealed the sink property of this infected tissue and possible stress or defence reactions of the host, whereas the reductions in the roots showed the impaired supply of this natural sink organ due to the infection. Changes in the healthy leaves were most pronounced in the tertiary leaf and may reflect metabolic stimulation in that healthy organ. The changes of the adenosine pool, a precursor of the adenine nucleotides, were discussed in terms of translocation and its possible role as a precursor for fungal purine nucleotide synthesis.  相似文献

Quantification of resistance induced by avirulent cultures of Erysiphe graminis f. sp. hordei against virulent cultures in barley was attempted. Four mildew cultures and 4 barley varieties with known genes of virulence and resistance respectively were used. Pre, post and simultaneous inoculation of leaves was done with avirulent and virulent cultures. Pre-inoculation with avirulent cultures induced resistance in the host such that the pustule number and spore production by later inoculation of virulent cultures was reduced significantly. Once induced, such resistance was active up to 8 days. There was some indication of induced susceptibility if the inducing culture was characterized by medium virulence. Increase of inceulum density of the avirulent (inducer) culture increased the amount of induced resistance Further studies of the phenomenon of induced resistance are needed in relation to possible applications for disease control through inoculations. varietal mixtures and multilines.  相似文献

Barley leaves were treated with a resistance inducer and inoculated with Erysiphe graminis two days later. 68 hours after inoculation many infection sites with cytoplasmic aggregations with increased number of golgi apparatus and increased endoplasmic reticulum could be observed. The extrahaustorial membrane invaginated and some free pieces of membrane appeared in the matrix. The haustoria vacuolized partially and curly filamentous electron dense material occurred. These changes took place after protective as well as after curative treatment. Due to cytochemical studies the vacuolar electron dense material in the vacuoles of the haustoria contained polymers of mannose or glucose.  相似文献
