Matrix polysaccharide from the brown algae Sargassum turbinarioides collected in the coastal waters of Nosy Be (Madagascar) in the Indian Ocean was isolated and its structure was studied by 1H-NMR spectroscopy, FT-IR, SEC-MALLS and HPAEC. An alginate with a molecular weight of 5.528 × 105 g mol−1 was identified as sole polysaccharide. Values of the M/G ratio, F GG, F MM and F GM (or F GM) blocks were measured at respectively 0.94, 0.39, 0.36 and 0.25 and compared with those of alginates from other Sargassum species. This sodium alginate appeared similar to some of the other Sargassum alginates with M/G < 1, high values of homopolymeric blocks (η < 1) and significant polyguluronic block content.  相似文献

The seasonal growth rates of three Sargassum species were studied along two reef flats of Teluk Kemang, located at Port Dickson, Malaysia from September 2009 to September 2010. Systematic quadrat and line transects were sampled monthly. Nondestructive sampling was conducted, whereby Sargassum plants were tagged and monitored for a 13-month period. The majority of the tagged Sargassum samples belonged to lower length classes (<200 mm), especially in 0–99 mm (Sargassum polycystum, 64.20 %; Sargassum binderi, 68.29 %; Sargassum siliquosum, 56.80 %). Analysis of the monthly mean thallus length (MTL) revealed a bimodal pattern in growth rates, with two periods of high growth rates (January–February 2010 and June–July 2010) and two periods of higher degenerative rates (April 2010 and September 2010). The highest growth rates were recorded in February 2010 (4.08 mm day?1) for S. siliquosum, and in June for S. polycystum (2.54 mm day?1) and S. binderi (1.89 mm day?1). Redundancy analysis (RDA) was employed to test for the overall correlation between monthly variation in MTL and the environmental parameters measured; S. binderi was correlated with ambient temperature (r = 0.5395), while S. siliquosum was correlated with seawater salinity (r = 0.5419) and ammonia (r = ?0.4603). This study reviews the seasonality of Sargassum species on two reefs of Teluk Kemang and their correlation with the selected environmental parameters.  相似文献

Sargassum C. Agardh is one of the most common but little understood genera of Phaeophyta in Malaysia. The difficulty in species delineation is due to morphological plasticity. A combination of morphology and random amplified polymorphic DNA (RAPD) studies of selected Sargassum species was carried out to have a better understanding of the taxonomy. Primer OPA13 was found to be good for discriminating between Sargassum species. Sargassum binderi was shown to be different from S. oligocystum (SD>0.5 = 14.11%), indicating the importance of the vesicle and receptacle in species differentiation. S. baccularia was clearly separated out from S. polycystum and S. stolonifolium using primer OPA13. RAPD analysis showed that the presence of the stolon is an important character for separating S. baccularia (no stolon) from S. polycystum (stolon) and S. stolonifolium (stolon). This study deals with two range-extending brown algae from Tahitian coral reefs, Sargassum mangarevense and Turbinaria ornata; their alginate properties, mannitol and phenolic contents, antioxidant and antimicrobial activities were determined. Turbinaria ornata showed the richest alginate content with the highest extraction yield (19.2 ± 1.3% dw). Their alginates also exhibited the highest viscosity (50 ± 18 mPa.s), but the M:G ratios (mannuronic acid to glucuronic acid) of alginates (1.25–1.42) were similar in both species. Alginate yield displayed spatial variations, but no significant seasonal changes. The highest mannitol content was found in S. mangarevense (12.2 ± 2.1% dw) during the austral winter. With respect to other tropical Fucales, both algae exhibited also a high phenolic content (2.45–2.85% dw) with significant spatio-temporal variations. Furthermore, high antioxidant activity and activity against Staphylococcus aureus were also detected in extracts. According to these preliminary results, these two range-extending algae are of key interest in numerous industrial areas.  相似文献

The kinetics of growth and alginate production from glucose in a nitrogen and phosphate-rich medium by Azotobacter vinelandii DSM576 were studied in a laboratory fermenter at pH 7 and 35°C. Batch fermentations were carried out both without control of dissolved oxygen concentration (DO) and at 1, 2, 5 and 10% DO. Although growth was faster at higher DO, maximum biomass concentration was lower. No alginate was produced at 10% DO. Alginate production was faster at 5 and 2% DO but higher alginate concentrations and yields were obtained without DO control. Alginate production was growth-associated at 5% DO, but significant amounts of alginate were produced after growth had stopped at lower DO values. In fermentations without DO control the molecular weight of the polymer reached a maximum (11–17.6 × 104) when specific growth rate was between 0.02 and 0.04 h−1 and residual concentration of ammoniacal nitrogen was between 0.01 and 0.02 g L−1 and then sharply decreased. Solute mobilities in cuticular membranes of six species (Hedera helix, Malus domestica, Populus alba, Pyrus communis, Stephanotis floribunda, Strophantus gratus) were measured using plant hormones, growth regulators and other organic model compounds varying in molar volumes from 99 to 349 mL · mol−1 The dependence of mobilities (k*) on molar volume (V x ) was exponential and could be described with equations of the type log k*=log k*0 V x . The y-intercepts (log k*0) represent mobilities of a hypothetical solute of zero molar volume. The parameter β′ is a measure of size selectivity of cuticular membranes and no differences among the six species were observed. At 25 °C the average β′ was 0.0095 mol · mL−1. Solute mobility decreased by about a factor of 8.9 when molar volume increased by 100 mL · mol−1 and the mobility of a compound with V x  = 100 mL · mol−1 was about 700-fold higher than the mobility of a compound with V x  = 400 mL · mol−1. Size selectivity decreased with increasing temperatures and for Strophantusβ′-values of 1.6 × 10−2 to 8.0 × 10-4 mol · mL−1 were obtained for 10 and 30 °C, respectively. The-intercepts (log k*0) differed among plant species by 3 orders of magnitude and since size selectivity was the same for all species, solute mobilities for solutes having zero molar volumes were the sole cause for differences among species in solute mobilities and permeabilities. We argue that these differences in k*0 are related to tortuosity of the diffusion path. These results were used to derive an equation which predicts rates of cuticular penetration on the basis of k*0, the average size selectivity of 9.5 × 10−3 mol · mL−1 and the driving forces of penetration. Alginate production by Azotobacter vinelandii was studied in batch and continuous cultures under microaerobic conditions. In batch culture at a pO2 of 2–3% (air saturation) alginate production was enhanced by decreasing the PO3− 4 level in the medium. Alginate yield from biomass (Y P/X) reached the highest value of 0.66 g/g at the lowest phosphate level (100 mg/l), compared to 0.40 g/g and 0.25 g/g at higher phosphate levels (200 mg/l and 400 mg/l, respectively). In contrast, biomass formation behaved differently and the growth yield (Y X/S) decreased with decreasing PO4 3− concentrations. Moreover, the respiratory quotient (RQ) of the culture was dependent on the initial phosphate concentration, especially in the phosphate-limited phase of growth. As the initial phosphate level decreased from 400 mg/l to 100 mg/l, the average RQ value of the culture declined from 1.46 to 0.89. The low RQ value is very close to the theoretical optimum RQ, calculated to be 0.8 on the basis of the stoichiometry of the metabolic pathways for alginate formation from sucrose. This optimum RQ was also confirmed in continuous culture at different dilution rates. Independent of the dilution rate, a pO2 value of 2–5% (air saturation) was found to be optimal for alginate production, the corresponding RQ values being 0.80–0.84. In addition, the molecular mass and composition of alginate were also found to be affected by both phosphate and oxygen concentrations. In conclusion, the RQ appears to be a useful parameter for optimum control of alginate production with this microorganism. Ten species of Sargassum (Sargassaceae, Phaeophyceae) were found along the Gulf of Thailand. Morphological characteristics of Sargassum baccularia (Mertens) C.A. Agardh, S. binderi Sonder, S. cinereum J.G. Agardh, S.crassifolium J.G. Agardh, S. longifructum Tseng et Lu, S. oligocystum Montagne, S. polycystum C.A. Agardh, S. siliquosum J.G. Agardh, S. swartzii (Turner) C.A. Agardh and one unidentified species were examined and are described in detail. The most common species were S. polycystum distributed widely in almost all the study sites, S. crassifolium restricted to Prachuap Khirikhan Province, S. longifructum restricted to Chumphon Province, S. siliquosum restricted to Surat Thani Province and one unidentified species restricted to Songkhla Province. Three species (S. cinereum, S. longifructum and S. swartzii) are new records for the algal flora of Thailand. Five species (S. baccularia, S. cinereum, S. longifructum, S. polycystum and the unidentified species) belong to the section Zygocarpicae (J.G. Agardh) Setchell.  相似文献

The initial responses to cold-water immersion, evoked by stimulation of peripheral cold receptors, include tachycardia, a reflex inspiratory gasp and uncontrollable hyperventilation. When immersed naked, the maximum responses are initiated in water at 10°C, with smaller responses being observed following immersion in water at 15°C. Habituation of the initial responses can be achieved following repeated immersions, but the specificity of this response with regard to water temperature is not known. Thirteen healthy male volunteers were divided into a control (C) group (n = 5) and a habituation (H) group (n = 8). Each subject undertook two 3-min head-out immersions in water at 10°C wearing swimming trunks. These immersions took place at a corresponding time of day with 4 days separating the two immersions. In the intervening period the C group were not exposed to cold water, while the H group undertook another six, 3-min, head-out immersions in water at 15°C. Respiratory rate (f R), inspiratory minute volume ( I) and heart rate (f H) were measured continuously throughout each immersion. Following repeated immersions in water at 15°C, the f R, I and f H responses of the H group over the first 30 s of immersion were reduced (P < 0.01) from 33.3 breaths · min−1, 50.5 l · min−1 and 114 beats · min−1 respectively, to 19.8 breaths · min−1, 26.4 l · min−1 and 98 beats · min−1, respectively. In water at 10°C these responses were reduced (P < 0.01) from 47.3 breaths · min−1, 67.6 l · min−1 and 128 beats · min−1 to 24.0 breaths · min−1, 29.5 l · min−1 and 109 beats · min−1, respectively over a corresponding period of immersion. Similar reductions were observed during the last 2.5 min of immersions. The initial responses of the C group were unchanged. It is concluded that habituation of the cold shock response can be achieved by immersion in warmer water than that for which protection is required. This suggests that repeated submaximal stimulation of the cutaneous cold receptors is sufficient to attenuate the responses to more maximal stimulation. Accepted: 6 February 1998  相似文献

Using the cell pressure probe, the effects of temperature on hydraulic conductivity (Lp; osmotic water permeability), solute permeability (permeability coefficient, Ps), and reflection coefficients (σs) were measured on internodes of Chara corallina, Klein ex Willd., em R.D.W.. For the first time, complete sets of transport coefficients were obtained in the range between 10 and 35 °C which provided evidence about pathways of water and solutes as they move across the plasma membrane (water channel and bilayer arrays). Test solutes used to check for the selectivity of water channels were monohydric alcohols of different molecular size and shape (ethanol, n-propanol, iso-propanol, and tert-butanol) and heavy water (HDO). Within the limits of accuracy, Q10 values for Lp and for the diffusive water permeability (Pd) were identical (Q10 for Lp = 1.29 ± 0.17 (± SD; n = 15 cells) and Q10 for Pd = 1.25 ± 0.16 (n = 5 cells)). The Q10 values were equivalent to activation energies of Ea = 16.8 ± 6.4 and 16.6 ± 10.0 kJ · mol−1, respectively, which is similar to that of self-diffusion or of viscous flow of water. The Q10 values and activation energies for Ps of the alcohols were significantly larger (ethanol: Q10 = 1.68 ± 0.16, Ea = 37.1 ± 5.9 kJ · mol−1; n-propanol: Q10 =  1.75 ± 0.40, Ea = 43.1 ± 15.3 kJ · mol−1; iso-propanol: Q10 = 2.12 ± 0.42, Ea =  52.2 ± 14.6 kJ · mol−1; tert-butanol: Q10 = 2.13 ± 0.56, Ea = 51.6 ± 17.1 kJ · mol−1; ±SD; n = 5 to 6 cells). Effects of temperature on reflection coefficients were most pronounced. With increasing temperature, σs values of the alcohols decreased and those of HDO increased. The data indicate that water and solutes use different pathways when crossing the membrane. Ordinary and isotopic water use water channels and the other test solutes use the bilayer array (composite transport model of membrane). Changes in σs values with temperature were found to be a sensitive measure for the open/closed state of water channels. The decrease of σs with temperature was theoretically predicted from the temperature dependence of Ps and Lp. Differences between predicted and measured values of σs allowed estimation of the bypass flow (slippage) of solutes through water channels which did not completely exclude test solutes. The permeability of channels depended on the structure and size of test solutes. It is concluded that water channels are much less selective than is usually thought. Since water channels represent single-file or no-pass pores, solutes drag along considerable amounts of water as they diffuse across channels. This results in low overall values of σs. The σs of HDO was extremely low. Its response to temperature was opposite to that for the σs of the alcohols. This suggested a stronger effect of temperature on the hydraulic (osmotic) than on the diffusive water flow across individual water channels, i.e. a differential sensitivity of different mechanisms to temperature. A simple procedure is described for the extraction and purification of alginate from the inner stipes of the kelp Laminaria pallida. Alginate yield was about 10–15% of the dry mass, with a 70:30 mannuronic/ guluronic acid ratio. Analysis of the purified alginate revealed a low polyphenol content while proteins were below detection level. The purified alginate was highly viscous, with 10–15 mPa s and 281 mPa s for a 0.1% and 0.5% solution, respectively, indicating a very high molecular mass (larger than 250 kDa). Bead formation occurred in the presence of divalent cations, but also in the presence of artificial serum (FCSIII) without added divalent cations. The biocompatibility of the alginate was tested with the in vitro mice lymphocyte test as well as by implantation of Ba2+ cross-linked beads beneath the kidney capsule of BB/OK rats. There was no evidence for significant mitogenic activity or fibrotic reaction. Biocompatibility of the alginate was also demonstrated by the encapsulation of human chondrocytes into Ca2+ cross-linked alginate beads. A gene, alg7D, from Saccharophagus degradans, coding for a putative alginate lyase belonging to the family of polysaccharide lyase-7, was overexpressed in Escherichia coli. The properties of the recombinant Alg7D were characterized. The enzyme endolytically depolymerized alginate by β-elimination into oligo-alginates with degrees of polymerization of 2–5. Its activity was maximal at 50°C and pH 7 and was slightly increased in the presence of Na+. The K M , V max , k cat , and k cat /K M values were: 3 mg ml−1, 6.2 U mg−1, 1.9 × 10−2 s−1, and 6.3 × 10−3 mg−1 ml s−1, respectively.  相似文献

The emission rates of eight volatile halogenated compounds by three tropical brown seaweed species collected from Cape Rachado, west coast Peninsular Malaysia, under different irradiances have been determined. A purge-and-trap sample preparation system with a gas chromatograph and mass-selective detector was used to measure a suite of halocarbons released by Sargassum binderi Sonder ex J. Agardh, Padina australis Hauck, and Turbinaria conoides (J. Agardh) Kützing. All species are widely distributed in Peninsular Malaysia, with S. binderi a dominant seaweed species at our survey site. Release of few halocarbons was found to be influenced by irradiance. Correlations were also observed between emission of certain halocarbons with photosynthetic activity, especially bromo-and iodinated compounds (0.6?<?r?<0.9; p?<?0.01) suggesting that environmental factors such as light can affect the release of these volatile halogenated compounds by the seaweeds into the atmosphere. Compared with temperate and polar brown seaweeds, tropical species, such as T. conoides, may emit higher levels of bromoform, CHBr3, and other halocarbons. It is therefore important to investigate the contribution of tropical seaweeds towards the local atmospheric composition of halocarbons.  相似文献

The present experiment was designed to study the importance of strength and muscle mass as factors limiting maximal oxygen uptake (O2 max ) in wheelchair subjects. Thirteen paraplegic subjects [mean age 29.8 (8.7) years] were studied during continuous incremental exercises until exhaustion on an arm-cranking ergometer (AC), a wheelchair ergometer (WE) and motor-driven treadmill (TM). Lean arm volume (LAV) was estimated using an anthropometric method based upon the measurement of various circumferences of the arm and forearm. Maximal strength (MVF) was measured while pushing on the rim of the wheelchair for three positions of the hand on the rim (−30°, 0° and +30°). The results indicate that paraplegic subjects reached a similar O2 max [1.23 (0.34) l · min−1, 1.25 (0.38) l · min−1, 1.22 (0.18) l · min−1 for AC, TM and WE, respectively] and O2 max /body mass [19.7 (5.2) ml · min−1 · kg−1, 19.5 (6.14) ml · min−1 · kg−1, 19.18 (4.27) ml · min−1 · kg−1 for AC, TM and WE, respectively on the three ergometers. Maximal heart rate f c max during the last minute of AC (173 (17) beats · min−1], TM [168 (14) beats · min−1], and WE [165 (16) beats · min−1], were correlated, but f c max was significantly higher for AC than for TM (P<0.03). There were significant correlations between MVF and LAV (P<0.001) and between the MVF data obtained at different angles of the hand on the rim [311.9 (90.1) N, 313.2 (81.2) N, 257.1 (71) N, at −30°, 0° and +30°, respectively]. There was no correlation between O2 max and LAV or MVF. The relatively low values of f c max suggest that O2 max was, at least in part, limited by local aerobic factors instead of central cardiovascular factors. On the other hand, the lack of a significant correlation between O2 max and MVF or muscle mass was not in favour of muscle strength being the main factor limiting O2 max in our subjects. The biosynthesis of poly(hydroxyalkanoates) (PHA) by Pseudomonas resinovorans from triglyceride substrates was investigated. Each triglyceride, whether animal fat or vegetable oil, supported cellular growth to relatively high average cell yields (3.3 ± 0.2 g/l). PHA yields ranged from 1.1 g/l to 2.1 g/l, representing approximately 45% of the bacterial cell dry weight. The repeat-unit composition of the polymers was determined by gas chromatography (GC) and GC/mass spectrometry of the β-hydroxyalkanoate methyl esters from the hydrolyzed polymers. With the exception of PHA from soybean oil (PHA-soy), each polyester was composed of β-hydroxyacyl moieties with chain lengths ranging from C4 to C14, with C8 and C10 being the predominant species. PHA-soy contained an additional fraction (2%) of C16 monomers. The alkyl side-chains of the PHA contained varying degrees of unsaturation. PHA from coconut oil was composed entirely of saturated side-chains, whereas PHA-soy contained 4.2 mol% olefinic groups in its side-chains. The increase in the degree of side-chain unsaturation caused decreased melting temperatures, enthalpies of fusion, and glass transition temperatures. The molar masses of the polymers were relatively constant and ranged from 6.5 × 104 to 10.1 × 104 g/mol. Haemoglobin components were analysed for nine species of New Zealand triplefins and their isoelectric points (pI) ranged from 5.1 to 7.0. The number of well-expressed isohaemoglobins was larger in shallow-water and tidal pool species, ranging from four in Grahamina signata to eight in Grahamina capito, and were relatively cathodal. Two strongly anodal isohaemoglobins were expressed in the mid-depth species Ruanoho decemdigitatus and Ruanoho whero, and one in the deeper water species Karalepis stewarti and Forsterygion malcolmi. The red blood cell oxygen-binding properties were determined at 15 °C and 25 °C in the pH range 6.7–7.9 for the shallow-water species G. capito, the shallow to mid-depth species Forsterygion varium, and the deep-water species F. malcolmi. Oxygen affinity was highest for G. capito and the magnitude of the Bohr effect lower (Δlog P 50/ΔpH = −0.37 at 25 °C, where P 50 is the half-saturation coefficient) compared to the two Forsterygion species (Δlog P 50/ΔpH = −0.52 to −0.59). Further, the cooperativity factor, n 50, was lower in G. capito thus maintaining oxygen transport over a wide range of environmental oxygen pressures. Oxygen binding was similarly influenced by temperature in both G. capito and F. malcolmi (maximum heat of oxygenation ΔHmax = −27 kJ mol−1 and −37 kJ mol−1, respectively). Thus, triplefin fishes living in shallow, thermally unstable habitats possess a greater number of cathodally migrating isohaemoglobins, and their red blood cells have a higher oxygen affinity and reduced cooperativity which is less sensitive to changes in pH than do species occurring in more stable, deeper water habitats. Our analysis of an assemblage of closely related species circumvents some of the difficulties inherent in studies where interpretation of experimental results is confounded by phylogeny. The binding of [3H]tyrosyl-PBAN28-33NH2 to pheromone gland membranes of the moth Heliothis peltigera was investigated. The study describes the development of a pheromone biosynthesis-activating neuropeptide (PBAN) radioreceptor assay and demonstrates the presence of a putative PBAN binding site on the pheromone gland. It also describes synthesis of a radioligand and optimization of binding conditions with respect to membrane preparation, number of gland equivalents, kinetics of ligand binding and composition of the binding solution. Binding was found to be optimal when membranes were freshly prepared from frozen glands, incubated at a concentration of one gland equivalent per reaction tube in the presence of 10 mM HCO3 ions. Equilibrium of ligand binding was obtained after 20 min. Presence of other components such as NaCl, KCl or SH reagents did not have any effect on binding. Binding was found to be saturable, with a Kd of 5.73 ± 1.05 × 10−6 M and a Bmax of 1.85 ± 0.22 nmol/mg protein. Binding was effectively displaced by unlabeled PBAN1-33NH2 and PBAN28-33ΝΗ2 with a Ki of 4.3 ± 1.1 × 10−6 M and 4.9 ± 2.6 × 10−6 M, respectively. Accepted: 4 February 1999  相似文献

From November 1992 to February 1995 a quantitative and qualitative phytoplankton study was conducted at a permanent station (Kerfix) southwest off the Kerguelen Islands, in the vicinity of the Polar Front (50°40′S–68°25′E). Phytoplankton populations are low in this area both during summers and winters. They consist, in order of decreasing cell abundance, of pico- and nanoflagellates (1.5–20 μm), coccolithophorids (<10 μm), diatoms (5–80 μm) and dinoflagellates (6–60 μm). Flagellates form the dominant group throughout the year and attain the highest summer average of 3.0 × 105 cells l−1. Next in abundance year-round are coccolithophorids with the dominant Emiliania huxleyi (highest summer 1992 average 1.9 × 105 cells l−1), diatoms (summer 1992 average 1.0 × 105 cells l−1) and dinoflagellates (average 3.8 × 104 cells l−1). Winter mean numbers of flagellates and picoplankton do not exceed 8.4 × 104 cells l−1; those of the three remaining algal groups together attain 2 × 104 cells l−1. Summer peaks of diatoms and dinoflagellates are mainly due to the larger size species (>20 μm). The latter group contributes most to the total cell carbon biomass throughout the year. Dominant diatoms during summer seasons include: Fragilariopsis kerguelensis, Thalassionema nitzschioides, Chaetoceros dichaeta, C. atlanticus, Pseudonitzschia heimii, and P. barkleyi/lineola. This diatom dominance structure changes from summer to summer with only F. kerguelensis and T. nitzschioides retaining their first and second positions. Any one of the co-dominant species might be absent during some summer period. The variable diatom community structure may be due to southward meandering of the Polar Front bringing “warmer” species from the north, and to the mixing of the water masses in this area. The entire community structure characterized both during summer and winters by the dominance of flagellates can be related to deep mixing (ca. 40–200 m) of the water column as the probable controlling factor. Gamma linolenic acid (GLA) degradation in Spirulina followed first-order reaction kinetics. At an accelerated temperature range of 45 to 55°C, the degradation rate constants (k r) of GLA obtained were 4.0 × 10−2 to 8.8 × 10−2 day−1. The energy of activation (E a) was 16.53 kcal mol−1, and the Q10 was 2.22. Based on 20% GLA degradation, the shelf life of sun-dried Spirulina at 30°C is 263 days or 8.6 months using the Arrhenius plot, and 258 days or 8.5 months using the Q 10 approach.
