Monoclonal strains of Chaetoceros species were collected from Chinese warm waters. Vegetative cells and resting spores were examined using light microscopy as well as scanning and transmission electron microscopy. Combined morphological and DNA sequence data from the hypervariable D1–D3 region of the nuclear ribosomal large subunit showed the presence of two new species within the section Compressa, herein described as Chaetoceros bifurcatus sp. nov. and C. millipedarius sp. nov. Both species possessed features typical of the section Compressa, but C. bifurcatus was characterized by the heavy intercalary setae fusing and extending together for a distance before diverging. The heavy setae were not visually contorted, lacking poroids and spines, in contrast with how the section Compressa has been defined. Chaetoceros millipedarius was characterized by a horizontal stagger of ordinary intercalary setae when the chain was seen in broad girdle view, making the chain look like a millipede. The two new species were supported by molecular phylogenetic analyses with C. bifurcatus sister to C. contortus var. ornatus, while C. millipedarius was sister to C. contortus var. contortus. Based on the morphological features exhibited in C. bifurcatus, the diagnosis of the section Compressa was emended to exclude “contorted” heavy setae and compressed valve faces.  相似文献

During different surveys of phytoplankton in the Gulf of California, along the coasts of Baja California, the Gulf of Tehuantepec, and the Mexican Caribbean Sea, new records of diatoms have been made and new species (mainly of the genus Chaetoceros) have been described. This study shows that the planktonic diatom Chaetoceros sumatranus, a rather rare tropical to subtropical taxon, is a typical species of the subgenus Chaetoceros (syn.: Phaeoceros) with robust cells forming chains, thick setae, reduced apertures and small, rounded chloroplasts in cells and setae. Additionally, the species exhibits other singular characteristics, such as heteropolarity of chains, distinct bases of the setae and a pattern of several rimoportulae in every valve of the chain. These features indicate that C. sumatranus is a species closely related to C. coarctatus. Both C. coarctatus and C. sumatranus are considered to be members of the recently proposed section Coarctati (Coarctata), within the subgenus Chaetoceros. The taxonomic relationships of the two species is analyzed, and a formal Latin diagnosis of the section Coarctati is also given.  相似文献

Bacteriastrum furcatum Shadbolt, Chaetoceros bacteriastroides Karsten and C. pseudocurvisetum Mangin are each connected in chains by fusion of silica between setae. The first two have specialized heterovalvate end cells indicating physiological control of the length of chains, but chains of C. pseudocurvisetum have usually been seen broken. There are specialized deeply lobed connecting bands in B. furcatum and C. pseudocurvisetum, allowing the setae of sibling valves to protrude outside the diameter of the chain during their formation. C. bacteriastroides and C. pseudocurvisetum are not identical; differences include presence of small setae, fusion of large setae some distance from the valves, symmetry of apertures between cells, and straight chains of C. bacteriastroides, compared with valve mantle connections with no small setae, large setae joined by crossing over, asymmetry of windows between cells, and curved chains of C. pseudocurvisetum. The evolutionary direction is hypothesized to be from Bacteriastrum to Chaetoceros through C. bacteriastroides.  相似文献

The diatom genus Chaetoceros is one of the most abundant and diverse phytoplankton in marine and brackish waters worldwide. Within this genus, Chaetoceros socialis has been cited as one of the most common species. However, recent studies from different geographic areas have shown the presence of pseudo‐cryptic diversity within the C. socialis complex. Members of this complex are characterized by curved chains (primary colonies) aggregating into globular clusters, where one of the four setae of each cell curves toward the center of the cluster and the other three orient outwards. New light and electron microscopy observations as well as molecular data on marine planktonic diatoms from the coastal waters off Chile revealed the presence of two new species, Chaetoceros sporotruncatus sp. nov. and C. dichatoensis. sp. nov. belonging to the C. socialis complex. The two new species are similar to other members of the complex (i.e., C. socialis and C. gelidus) in the primary and secondary structure of the colony, the orientation pattern of the setae, and the valve ultrastructure. The only morphological characters that can be used to differentiate the species of this complex are aspects related to resting spore morphology. The two newly described species are closely related to each other and form a sister clade to C. gelidus in molecular phylogenies. We also provide a phylogenetic status along with the morphological characterization of C. radicans and C. cintus, which are genetically related to the C. socialis complex.  相似文献

Two new planktonic diatom species of the genusChaetoceros are described herein:Ch. octagonus andCh. rectus, from material of Baja California coasts and the Gulf of California, respectively;Ch. rectus was also found in a sample from Australian waters.Ch. octagonus is a robust species, with a characteristic pyramidal valve shape, heavily silicified valves and setae, and one central rimoportula on every valve in the chain; it is compared with related species, which show a similar morphology, especially by electron microscopy. It belongs into subg.Chaetoceros, sect.Borealia. Ch. rectus is placed in subg.Hyalochaete, and provisionally within sect.Anastomosantia. Its major characters are: chains, cells, and setae are delicate and fragile, cells are joined in the chain by a bridge which connects sibling intercalary setae, valves and setae are weakly silicified, and one rimoportula is present on terminal valves only. Comparison is made with another species of the same section.Ch. rectus shows a different manner of chain-forming within the genus than previously described in the literature.  相似文献

A retrospective study of 20 patients with cryptococcal meningitis and their isolated strains was performed. Cryptococcus neoformans var. neoformans was recovered from 14 (70%) cases, and var. gattii was recovered from six (30%) patients. Twelve patients had AIDS (all carrying var. neoformans), two had other diseases (one with var. neoformans and one var. gattii) and there was no identifiable underlying disease in six (one var. neoformans and five var. gattii). Fourteen patients (11 var. neoformans and three var. gattii) resided in the Mexico City area, where a temperate climate is prevalent, and there were six cases (three var. neoformans and three var. gattii) from states with a tropical/subtropical climate. Although there was no significant statistical difference between the two varieties, the fatal outcome was higher in patients with var. neoformans. The disease caused by var. gattii strains was characterized by a higher opening pressure, more inflamatory changes of CSF and a longer clinical course (delayed clinical and mycological cure). Cryptococcus neoformans var. gattii is a significant cause of cryptococcal meningitis in patients without underlying diseases in Mexico.  相似文献

Numerical studies of morphological data and starch gel electrophoresis have provided a new perspective on important issues pertinent to the origin(s) and subsequent evolution of domesticatedCucurbita pepo. Wild C.texana and/orC. fraterna appear to be the most likely candidates for progenitor(s) of the domesticate. Populations of texana-like plants from beyond Texas share various attributes with Texas populations, suggesting that C. texana once had a more widespread distribution to the northeast. The possibility exists thatC. pepo was domesticated independently in eastern U.S., as well as in Mexico, which is in line with recent archeological findings. Multiple domestications are also supported by allozyme data indicating a substantial divergence within the species. A new classification consisting of C.pepo ssp.pepo (origins in Mexico),C. pepo ssp.ovifera var. ovifera (origins in eastern U.S.), and C.pepo ssp.ovifera var. texana (spontaneous populations in eastern U.S.) is proposed.  相似文献

We document the fine structure of auxospores in a Chaetoceros species isolated from the Acadian coast of New Brunswick, Canada. Auxospore development in this species occurs in a terminal rather than lateral position, a characteristic never before observed in this genus. Our observations suggest that auxosporulation was uniparental, probably an extreme form of autogamy with sister nuclei fusing following meiosis II. Mature auxospores were adze-shaped to sub-globular and contained both scales and transverse perizonia in their walls. The transverse perizonial band structure was similar to longitudinal perizonial bands found in other species of Chaetoceros and differed from the pinnate bands of pennate transverse perizonia, which consisted of a central rib and bilateral fimbria. Instead, the band structure in C. acadianus was more similar to unilateral fimbriate bands in cymatosiroids. We also propose that our diatom represents a species new to science and is a member of the Chaetoceros Section Compressa. We provide its morphological, molecular and reproductive characterization.  相似文献

The effects of temperature, ionic composition, and conductivity on growth rates of ten strains of Chaetoceros muelleri Lemmerman (mostly var. subsalsum Johan. & Rushf.) were studied. Lipid content of stressed and unstressed cells and fatty acid composition were also determined. Considerable physiological variability was observed in the ten strains, although principal components analysis of physiological data indicated that all strains fell into one of two major groups: C. muelleri (var. muelleri and var. subsalsum) and an undescribed Chaetoceros species morphologically close to C. muelleri var. subsalsum. A high degree of agreement was found among morphological, physiological, and biochemical data sets, indicating that physiological and biochemical data may be helpful in making taxonomic decisions in diatoms, particularly in taxa with few morphological characters. We also conclude that nonmorphological characters such as those employed in the present study can be used to test phylogenetic hypotheses formulated from traditional morphological data.  相似文献

Summary Polyhydroxyalkanoates (PHAs) are polyesters of hydroxyalkanoates synthesized by numerous bacteria as intracellular carbon and energy storage compounds and accumulated as granules in the cytoplasm of cells. In this work, we constructed two recombinant plasmids, pBE2C1 and pBE2C1AB. The two plasmids were inserted into Bacillus subtilis DB104 and generated Bacillus subtilis/pBE2C1 and Bacillus subtilis/pBE2C1AB. The two recombinant strains were subjected to fermentation and showed PHA accumulation, the first reported example of medium-chain-length-PHA production in Bacillus subtilis. GC analysis identified the compound produced by Bacillus subtilis/pBE2C1 was a hydroxydecanoate-co-hydroxydodecanoate (HD-co-HDD) polymer while that produced by Bacillus subtilis/pBE2C1AB was a hydroxybutyrate-co-hydroxydecanoate-co-hydroxydodecanoate (HB-HD-HDD) polymer. The results also showed that the recombinant B. subtilis could utilize the malt waste in the medium as a carbon source better than that of glucose and thus could substantially lower the cost of production of PHA.  相似文献

A revision of Brazilian Augusta and general comments on the systematics of this genus are presented. The complex taxonomic history of this taxon has caused confusion over its correct nomenclature. The peculiar geographic distribution of Augusta has stimulated field observations and analysis of the ecology of the Brazilian ecosystems where this taxon occurs, which are here presented. The five Brazilian species of Augusta described by Pohl are here treated as ecotypes of A. longifolia. This species has two distinct varieties: var. Longifolia, widespread throughout the Brazilian cerrado, and var. parvifolia, endemic to the Atlantic forest near Rio de Janeiro. Typification, list of representative specimens, and distribution map of A. longifolia are also included.  相似文献

The juvenile stages and adults of two new Tasm anian crotoniid m ites Holonothrus minus‐culus sp. n. and H. glaesarius sp. n. are described and illustrated. Holonothrus minusculus sp. n. is similar to H. mitis Olszanowski, 1991 in body dimensions and because the openings of the opisthosomal gland are situated close to setae f2. It is similar to H. robus‐tus Olszanowski, 1991 in the length and structure of notogastral setae. H. glaesarius sp. n. is absolutely different from other known species of this genus and that is why it cannot be compared.  相似文献

On the screening of microorganisms which accumulate ultra violet light absorbing substances, some sporogenous bacteria were selected as powerful strains for production of a crystalline substance having a maximum absorption at 267.5 mμ (pH 6.0 in water). These strains were microbiologically examined and named B. subtilis var. thermophilus. Submerged fermentation was carried out for 48 hrs at 37°C in glucose bouillon medium and the isolation of substance was performed by absorption to active carbon and elution with ammonia water.

On the basis of chemical studies of this crystal, it was identified as urocanic acid.  相似文献

Environmental conditions between the outer reef slope (ORS) and lagoon in tropical atolls are significantly different, but the variations of juvenile coral-microbiomes in the two environments and their relationship with coral thermal acclimatization are poorly understood. We explored this issue based on local water conditions and the microbiome of juvenile corals in the ORS and lagoon in the central South China Sea. Coral-symbiotic Symbiodiniaceae showed significant differences among coral species; Pocillopora verrucosa and Pachyseris rugosa in the ORS, and Acropora formosa in the lagoon were dominated by Durusdinium, but other corals were dominated by Cladocopium. Although A. formosa in the ORS were dominated by Cladocopium (C3u), they were dominated by Durusdinium (D1/D1a) and Cladocopium (C50) in the lagoon. Other coral species were both dominated by Cladocopium in the lagoon and ORS. The relative abundance of bacteria in the Deinococcus–Thermus was generally higher in the lagoon corals than in the ORS corals. Our study indicates that P. verrucosa, P. rugosa and Porites lutea may have high thermal tolerance based on the relatively high abundance of heat-tolerant Durusdinium and Thermus scotoductus. Likewise, A. formosa in the lagoon may acclimatize to the thermal environment based on a high relative abundance of heat-tolerant Durusdinium.  相似文献

 Three species of the genus Inocybe are reported as new species or new records from Japan. Inocybe phaeodisca Kühner var. geophylloides Kühner is redescribed by specimens collected in Chiba Prefecture; this is the first record of I. phaeodisca var. geophylloides for Japan. Inocybe pseudoreducta Stangl & Glowinski is also redescribed by materials collected in Hokkaido and Chiba Prefecture, new to Japan. Inocybe subtilis Takahito Kobayashi sp. nov. is proposed for material collected from Tokyo. The sectional position of this species is noted. Summary The cyclomaltodextrin glucanotransferase (CGTase, E.C. 2.4.1.19) gene from an alkalophilic Bacillus circulans var. alkalophilus ATCC21783 was cloned into Escherichia coli and B. subtilis. When cloned from E. coli to B. subtilis, the entire insert containing the CGTase gene was, depending on the plasmid construction, either unstable or the recombinant B. subtilis did not secrete the enzyme in significant amounts. To achieve efficient enzyme production in B. subtilis, the gene was placed under the control of the B. amyloliquefaciens -amylase promoter. In one of the constructions, both the promoter and the signal sequence of the gene were replaced with those of B. amyloliquefaciens, whereas in another construction only the promoter area was exchanged. The recombinant B. subtilis clones transformed with these plasmid constructions secreted CGTase into the culture medium 14 times as much as did the parental strain in shake flask cultures. In fermentor cultures in an industrially feasible medium the enzyme production was substantially higher, yielding 1.2 g/l of CGTase, which is about 33 times the amount of the enzyme produced by the parental strain in corresponding fermentations. Both of the plasmid constructions were stable when grown over 50 generations without antibiotic selection.  相似文献

The arrangement and external morphology of the rodlike setae and associated structures located on the dactylopodites of the walking legs of six species of decapod crustaceans are compared. The dactyls of littoral species, represented by the rock crab, Cancer antennarius, and the spiny lobster, Panulirus interruptus, have dense tufts and bands of rodlike setae, as is typical of many decapods, and additionally only a few small plumed setae. The arrangement of setae on the dactyls of the recently discovered Galapagos vent crab, Bythograea thermydron, closely resembles that of C. antennarius. Rodlike and long plumed setae occur in about equal numbers on the dactyls of the pelagic anomuran, Pleuroncodes planipes. The dactyls having the fewest rodlike setae are those of the terrestrial hermit crab, Coenobita perlatus, and those of the kelp crab, Pugettia producta, where flat setae typical of Majidae have replaced most rodlike setae. The presence and structures of the terminal pores in rodlike setae vary intra- and interspecifically, possibly as a function of molt stage. Variations in some features of rodlike setae, such as tip acuity and presence of microsetae and surface sculpting, appear to be related to development. Serrated setae occur on the dactyls of megalopal P. producta but not in later stages. The topography and typology of setae located on the ambulatory dactyls of decapod crustaceans are considered in light of recent interest in using setal characteristics to determine the sensory functions of sensilla and to clarify the phylogeny of arthropod groups.  相似文献
