The main objective of this study was to determine whether transcutaneous ultrasonography could be used as a diagnostic tool to monitor the reproductive organs of female breeding ostriches. An additional aim was to investigate whether the use of this technique could facilitate prediction of the start or end of the egg production season. A technique for on-farm ultrasound scanning is described and examples of ultrasonographic images of different ovarian structures, developing ova and eggs within the genital tract are presented. These data were obtained by scanning mature female breeding ostriches (n=8). In vivo scanning took place a day prior to slaughter, and immediately after slaughter the reproductive organs were scanned in vitro in a water bath. By comparing ultrasonographic images with post mortem ovarian morphology, it appeared that the following morphological structures can be identified using ultrasonography: ovarian follicles of different sizes (diameter (phi) 1-9 cm), atretic follicles, post-ovulatory follicles (POFs), ova at different stages of development and eggs within the genital tract. Of the number of follicles counted during post mortem investigation, 58% (95% confidence interval 0.41-0.79) had been detected during previous in vivo examination. In the second part of our study, ultrasonographic scans were made at weekly intervals in two farmed female ostriches (n=2) during the breeding season in order to determine the predictive value of the technique. By comparing the images of ovarian activity with individual egg production of these hens, preliminary evidence was obtained that scanning might be of value in predicting egg production, especially at the start and the end of the breeding season. It is concluded that transcutaneous ultrasound scanning in mature female breeding ostriches is an easy, noninvasive technique for the monitoring of ovarian (in)activity, for visualization of different functional ovarian structures, for following the development of individual ova and for visualization of an egg with calciferous shell within the oviduct, and that this technique will be a valuable tool in future research on reproductive physiology and pathology, and the development of more advanced reproductive technologies, such as artificial insemination.  相似文献

Although ultrasound evaluation of the reproductive tract of stallions was introduced to veterinary practice long ago, this examination is not always conducted during routine breeding soundness evaluation. The objective of this study was to investigate the clinical relevance of routine ultrasound evaluation of the stallion's reproductive tract. Breeding soundness evaluation of 113 stallions was performed, including ultrasonography of external and internal genitalia. Various pathological conditions were detected using this technique, with the most frequent being varicocele, thickened vaginal tunics and cystic structures. Varicocele and thickened vaginal tunics were associated with decreased quality of semen, while cysts of the epididymal head, urethra and uterus masculinus were frequently associated with ejaculatory problems. These findings suggested that routine ultrasound evaluation of breeding stallions may be very helpful in detecting pathological conditions that may impact their reproductive career.  相似文献

An infection of any part of female reproductive tract can severely interfere with fertility and reproduction. The fluids and epithelium from the lumen of the female reproductive tract (uterus, oviduct and ovarian follicle) are a known source of antimicrobial action in several species. In this study, we compared the antimicrobial properties of fluids from the reproductive tract of a cow. After removal of small molecules, we demonstrated that there is an antimicrobial activity connected with a fraction of compounds with a molecular mass range between 3500 and 30,000. The most probable candidates responsible for the observed antimicrobial effect were subsequently identified by mass spectroscopy as histones H2A type 2-C, H2B type 1-K, H3.3, and H4. The antimicrobial role of histone H2B was further confirmed by using an antibody against this histone.  相似文献

Ultrasound imaging has been used to elucidate certain aspects of the reproductive biology of wild or endangered species. However, to our knowledge, this tool has not been used for reproductive monitoring of the collared peccary (Tayassu tajacu). In this study, real-time ultrasonography was used in 16 collared peccary females to diagnose early pregnancy status and predict gestational age. Based on the detection of an embryo, the earliest pregnancy diagnosis was made on Day 18 after mating, with the mean time needed for diagnosis being 22 days. Overall accuracies on Days 22, 26 and 28 were 56, 93, and 100%, respectively. On Days 26 and 28, all pregnancy and non-pregnancy diagnoses, respectively, were correct. The fetal measurements that best correlated with gestational age were crown-rump-length (CRL) and the length and diameter of the thorax. CRL was considered the most practical measurement because, contrary to thoracic fetometry, it could be determined when the embryo was first detected. Our findings revealed real-time ultrasound scanning to be a very accurate method for early pregnancy diagnosis and prediction of gestational age in the collared peccary.  相似文献

After 80 years of the commercial application of artificial insemination (AI) in the cow, the method still has numerous benefits over natural insemination including worldwide gene improvement. The efficiency of insemination depends, among many other factors, on the delivery of an appropriate number of normal spermatozoa to the appropriate reproductive tract site at the appropriate time of estrus. The metabolic clearance of steroid hormones and pregnancy associated glycoproteins and the negative effects of different types of stress related to high milk production makes the high-producing dairy cow a good animal model for addressing factors affecting fertility. Nevertheless, extensive studies have shown a positive link between high milk production in an individual cow and high fertility. When a cow becomes pregnant, the effect of pregnancy loss on its reproductive cycle is also a topic of interest. This paper reviews the factors of a noninfectious nature that affect the fertility of lactating dairy cows following AI. Special attention is paid to factors related to the cow and its environment and to estrus confirmation at insemination. Pregnancy maintenance during the late embryonic/early fetal period is discussed as a critical step. Finally, the use of Doppler ultrasonography is described as an available research tool for improving our current understanding of the health of the genital structures and conceptus.  相似文献

The Asian and African elephant populations in North America are not self‐sustaining, and reproductive rates remain low. One problem identified from routine progestagen analyses is that some elephant females do not exhibit normal ovarian cycles. To better understand the extent of this problem, the Elephant TAG/SSP conducted a survey to determine the reproductive status of the captive population based on hormone and ultrasound evaluations. The survey response rates for facilities with Asian and African elephants were 81% and 71%, respectively, for the studbook populations, and nearly 100% for the SSP facilities. Of the elephants surveyed, 49% of Asian and 62% of African elephant females were being monitored for ovarian cyclicity via serum or urinary progestagen analyses on a weekly basis. Of these, 14% of Asian and 29% of African elephants either were not cycling at all or exhibited irregular cycles. For both species, ovarian inactivity was more prevalent in the older age categories (>30 years); however, acyclicity was found in all age groups of African elephants. Fewer elephant females (~30%) had been examined by transrectal ultrasound to assess reproductive‐tract integrity, and corresponding hormonal data were available for about three‐quarters of these females. Within this subset, most (~75%) cycling females had normal reproductive‐tract morphologies, whereas at least 70% of noncycling females exhibited some type of ovarian or uterine pathology. In summary, the survey results suggest that ovarian inactivity is a significant reproductive problem for elephants held in zoos, especially African elephants. To increase the fecundity of captive elephants, females should be bred at a young age, before reproductive pathologies occur. However, a significant number of older Asian elephants are still not being reproductively monitored. More significantly, many prime reproductive‐age (10–30 years) African females are not being monitored. This lack of information makes it difficult to determine what factors affect the reproductive health of elephants, and to develop mitigating treatments to reinitiate reproductive cyclicity. Both in the male and in the female reproductive tract glucose can be converted via either the pentose pathway or the sorbitol pathway. It is shown that a disturbed carbohydrate metabolism can lead to infertility, i.e. in the cow and in the bull semen. Evidence is provided that the serotonin-like indole which occurs in the protein-complex and is liberated, can exert effects on the uterine endometrium comparable to those caused by serotonin. It is suggested that the indole liberated can cause ischaemia, followed by regression of the endometrium. When this occurs in repeat breeder cows, exogenous PGF given in mid-cycle on a suitable day, may restore the endometrium so that the cow can again become pregnant. The possibility is mentioned that in humans the free indole might cause regression of the endometrium and some distress symptoms, but thereafter endogenous PGF does increase the vascular permeability resulting finally in bleeding.  相似文献

The reproductive tracts of 26 estrus synchronized, bred ewes were scanned with a portable 5.0 MHz real-time ultrasound unit within 1 to 6 d postbreeding. Intrarectal scanning was performed on alternate days until Days 28 to 30 and twice weekly until Days 50 of gestation. Transabdominal uterine scans were conducted twice weekly from Days 25 to 65 and continued weekly until parturition. A total of 24 ewes (92%) became pregnant. A nonpregnant ewe was recognized 100% of the time by both methods of ultrasonic screening. Correct identification of a gravid ewe as pregnant was 100% from Days 51 to 150 of gestation using transabdominal real-time ultrasonography. There was a significant association (P < 0.005) between the number of lambs born and the number of fetuses observed using transabdominal real-time ultrasonography after Day 25 of gestation. Accurate differentiation of fetal numbers by transabdominal scanning was 100.0% for ewes carrying one lamb and 97.3% for ewes carrying two lambs at Days 51 to 75 of gestation. Fetal attrition was documented in one ewe at Day 49 of gestation. Hydrops allantois was diagnosed in another ewe at 110 d of gestation. A total of 37 lambs were born to 23 ewes in the project flock. No congenital abnormalities were noted in any of the lambs. Transabdominal real-time ultrasonography is a safe, rapid, accurate and practical method for assessing pregnancy status, fetal number and fetal viability in sheep.  相似文献

This study was carried out to determine the relationship between postpartum intrauterine infections, endocrine patterns and the function of corpora lutea formed following the first postpartum ovulations in dairy cows. Blood samples were collected daily starting from the day of parturition until 30 d after parturition or until the second postpartum estrus, whichever occurred first. Sera were assayed for progesterone (P(4)), prostaglandin F(2alpha) metabolite (PGFM), and luteinizing hormone (LH) concentrations. Palpations per rectum and real-time ultrasound scanning of the reproductive tracts were carried out in all cows once every 4 d for 1 mo, starting from Day 4 after parturition. In addition, endometrial swabs were collected aseptically from each cow once every 4 d during the first month postpartum. The swabs were cultured for aerobic and anaerobic bacteria. Twelve cows (60%) exhibited short estrous cycles (SC; 6 to 14 d long) following first postpartum ovulations. The mean preovulatory LH surges and LH patterns during the first postpartum cycles were similar in both groups, leading us to believe that lack of luteotrophic stimulation was not a factor in the occurrence of SC. Bacterial isolations were frequent in SC cows. The occurrence of moderate to heavy bacterial growth patterns and the repeated isolations of the similar organisms during postpartum suggests the persistence of uterine infections in SC cows. Increases in PGFM concentrations prior to luteolysis in SC cows were associated with moderate to heavy infection. Thus, postpartum uterine infections do not appear to affect ovulations, but prostaglandin (PGF(2alpha)) released in response to uterine infection may contribute to early demise of the corpus luteum formed after the first postpartum ovulation.  相似文献

Over three years all infants in this hospital found to have an abnormality of the urinary tract on antenatal scanning were followed up after delivery with contrast radiography. Disease of the renal tract was confirmed in 17 of 20 infants. Of the 15 survivors, 12 underwent surgery in the first year of life. Abnormality of the fetal urinary tract detected by ultrasound scanning seems to be an important indicator of disease of the renal tract. Before its use is extended, however, further assessment of the benefit of antenatal diagnosis and of the best time to scan is required.  相似文献

The results of a 6-year study of cow reproductive traits in an agro-pastoral system in semi-arid central Mali are reported. Age at first calving was 1505 days (49.5 months). Calving intervals averaged 665 days with a standard deviation of 202 days, but these showed a bimodal distribution centred on 13–16 and 23–26 months. Least squares analysis indicated that parturition number had a significant effect on the calving interval. Some 56% of calves were born in the period April to June, there being a very highly significant correlation between these births and rainfall 9 and 10 months previously. The average number of calves produced per cow in the herd was 2.9. Nutritional stress appears to be the main cause of low reproductive rates and of the seasonal pattern of births and mitigation of this stress would lead to higher calving percentages.  相似文献

We describe herein the sperm morphology of the harvestman Iporangaia pustulosa. Adult males were dissected, the reproductive tract was schematized and the seminal vesicle was processed by light, transmission and scanning electron microscopy. The male reproductive tract is composed of a tubular testis, two deferent ducts, a seminal vesicle, a propulsive organ and a penis, similar to that observed in other Opiliones. The spermatozoa from the seminal vesicle are oval, aflagellate and immotile, presenting a nucleus surrounding an invagination of the cytoplasm, as well as a complex acrosome and projections on the cell surface. In the testis, spermatozoa are devoid of projections. In the seminal vesicle, they gradually acquire the projections with tufts adhering to it. Consequently, spermatozoa in various distinct stages of projection development can be found in the seminal vesicle. We believe that these projections (1) could help transport sperm along the male and perhaps female reproductive tracts; (2) are used to anchor the spermatozoa inside the female spermatheca in order to avoid mechanical displacement by the genitalia of other males and (3) may play a role in oocyte recognition. We propose that the evolution of aflagellarity in Opiliones is related to the unique morphology of the female reproductive tract. Since eggs are fertilized on the tip of the ovipositor just prior to being laid, there is no advantage favoring sperm mobility. Additionally, female sperm receptacles are small and males that produced small spermatozoa would have a higher chance of fertilizing more eggs.  相似文献

The recent development of new ultrasound probes has made real-time intraoperative monitoring of cryosurgery, and thermocouple placement a possibility. It is shown that frozen tissue and thermocouple needles have acoustic characteristics that enable them to be easily visualized by ultrasound examination. Further in vivo animal studies are needed to examine temperature characteristics of visualized cryolesions, to develop scanning techniques, and to correlate ultrasonic findings with histologic changes in tissue.  相似文献

A simple one-way cannula technique for non-surgical recovery of bovine embryos has been described. The collector was very reliable, stable, and easy to use. It could be guided through cervix and without damage to the genital tract into both uterine horns on all attempts. Ninety-six per cent (96%) of the flushing medium was recovered. Bleeding occurred rarely. The post-flushing reproductive performance was satisfactory. An average of 4.1 eggs per dairy cow were recovered yielding a recovery rate of 46%. The method was performed under farm conditions with and without the use of a chute or stanchion. Several general practitioners have used the method with success after a brief training period. Good manipulative skills were a prerequisite for good results.  相似文献

During the preadult development of Drosophila melanogaster, the GLD (glucose dehydrogenase) gene (Gld) is expressed in a variety of tissues, including the immature reproductive tract. At the adult stage the expression of Gld becomes largely restricted to the reproductive tract of males and females. We examined the expression of GLD in the adult reproductive tract of 50 species in the genus Drosophila, as well as in those of a few representative species from four other closely related genera. GLD exhibits considerable organ-specific diversity in the reproductive tract of males and females. Among these species, five male GLD phenotypes and six female GLD phenotypes were found. In contrast, the preadult expression of GLD in representative species from each distinct adult pattern type was determined and found to be highly conserved in both the immature reproductive tract and non-reproductive organs. Moreover, the set of reproductive organs that express GLD during preadult development is equivalent to the sum of the five male and six female adult GLD phenotypes. To initially define the contribution of cis- versus trans-acting factors responsible for differences in adult GLD expression between two of these species--D. melanogaster and D. pseudoobscura--we transferred the D. pseudoobscura Gld to the genome of D. melanogaster and investigated its expression. GLD expression patterns of these transformants displayed characteristics that are unique to both species, suggesting the presence of both cis- and trans-acting differences between these two species.  相似文献

This article is about new reproductive technologies, maternal anxieties, and existential uncertainties. It explores the question of why pregnant women in Vietnam's capital, Hanoi, have become avid consumers of obstetrical ultrasound scanning even while expressing profound doubts regarding the reliability and safety of this new technology of pregnancy. Through a phenomenological analysis of the social production of women's sense of reproductive risks and uncertainties, the article shows how Hanoian women's paradoxical stances toward ultrasound imaging can be explained through a consideration of embodied and historically generated experiences within everyday local worlds. The article argues that the "scientific stories" of fetal well-being and normality that are produced through ultrasonography are challenged by vivid and continual exchanges in everyday lives of stories of the inherent uncertainties of existence in general and of human reproduction in particular.  相似文献

Sonoporation using low-frequency high-pressure ultrasound (US) is a non-viral approach for in vitro and in vivo gene delivery. In this study, we developed a new sonoporation device designed for spatial and temporal control of ultrasound cavitation. The regulation system incorporated in the device allowed a real-time control of the cavitation level during sonoporation. This device was evaluated for the in vitro transfection efficiency of a plasmid coding for Green Fluorescent Protein (pEGFP-C1) in adherent and non-adherent cell lines. The transfection efficiency of the device was compared to those observed with lipofection and nucleofection methods. In both adherent and non-adherent cell lines, the sonoporation device allowed high rate of transfection of pEGFP-C1 (40–80%), as determined by flow cytometry analysis of GFP expression, along with a low rate of mortality assessed by propidium iodide staining. The transfection efficiency and toxicity of sonoporation on the non-adherent cell lines Jurkat and K562 were similar to those of nucleofection, while these two cell lines were resistant to transfection by lipofection. Moreover, sonoporation was used to produce three stably transfected human lymphoma and leukemia lines. Significant transfection efficiency was also observed in two fresh samples of human acute myeloid leukemia cells. In conclusion, we developed a user-friendly and cost-effective ultrasound device, well adapted for routine in vitro high-yield transfection experiments and which does not require the use of any transfection reagent or gas micro-bubbles.  相似文献

Nearly two decades ago, modern wildlife medicine started to gradually use the advantages of the non-invasive ultrasonography, which was already well established in human and classical veterinary medicine. For more than one decade now, the application of imaging ultrasound for reproductive assessments and as a supportive tool during assisted reproduction procedures such as artificial insemination (AI) in elephants and rhinoceroses has dramatically improved the breeding success in captive breeding programmes. The opportunity for identifying potential breeding candidates on the basis of their reproductive health status is widely used for natural mating or for AI, today. The longitudinal sonographic monitoring of pathological processes on the internal female genital tract allowed the identification of pathogenetic causes for the rapid infertility development in older nulliparous females. The factors causing temporary infertility in captive male elephants and rhinoceroses were also identified by the use of ultrasound. Today, ultrasonography is the golden standard for reproductive assessments in megavertebrates such as elephants and rhinoceroses in captive management settings and also in the wild.  相似文献
