Galanin, a recently discovered neuropeptide, was studied in the rat male and female reproductive tracts by immunocytochemistry and in vitro pharmacology. Nerve fibers containing galanin immunoreactivity were most abundant in the female paracervical tissue, where they surrounded non-immunoreactive ganglion cells. Galanin nerves were also found in the uterus and Fallopian tubes, as well as in the vas deferens. When tested in vitro galanin contracted the smooth muscle of both the uterine horn and cervix. Galanin also slightly potentiated the response to electrical field stimulation in preparations from the uterine cervix and vas deferens, but it had no effect on the seminal vesicle. Galanin-(1–10), an N-terminal residue of galanin, also contracted the uterine horn, though higher concentrations were required. The neurally induced contractions were not influenced by galanin-(1–10) in any of the smooth muscle preparations tested. The muscle receptors mediating the direct contractile effects in the uterine horn seem to require the N-terminus of galanin, while the neuromodulatory effects on the electrically induced contractile activity seem to need the C-terminal part or the whole galanin molecule. Galanin may thus function as a neuromediator in the rat male and female genital organs.  相似文献

The influence of chromosome variability on the production of euploid spermatozoa was investigated in a suitable biological model, the Akodon molinae system. This consists of individuals whose chromosome constitution is 2n = 42, 2n = 43, or 2n = 44. The only difference between these three karyotypes occurs through a Robertsonian rearrangement combined with two pericentric inversions. Thus, the animals with 2n = 42 (simple homozygotes or SH) have two large metacentric chromosomes number 1; animals with 2n = 43 (heterozygotes or Ht) have a chromosome 1 and two subterminal chromosomes la and lb homologues of the long and short arms of the chromosome 1, respectively; animals with 2n = 44 (double homozygotes or DH) have a pair of la and a pair of lb chromosomes. The gametic euploidy frequency correlated with each chromosome constitution was evaluated on the basis of the DNA content of spermatozoa, which was determined microdensitometrically after the Feulgen reaction, taking into account the site of the spermatozoa along the male genital tract. A comparative assessment of gametic aneuploidy frequency in caput epididymis versus vas deferens demonstrated (1) a falloff in euploid production in passing from the 2n = 42 to the 2n = 44 chromosome forms, alongside a high degree of intragroup variability, and (2) a lower aneuploidy frequency in the vas deferens than in caput epididymis in all the forms considered. These two features, taken together with similar results in the mouse chromosome variability system, suggest that a selection mechanism is operative against aneuploid spermatozoa in the epididymis. This finding is of interest in a wider perspective, since it might turn out to be valid for many mammals.  相似文献

Natural selection and post‐copulatory sexual selection, including sexual conflict, contribute to genital diversification. Fundamental first steps in understanding how these processes shape the evolution of specific genital traits are to determine their function experimentally and to understand the interactions between female and male genitalia during copulation. Our experimental manipulations of male and female genitalia in red‐sided garter snakes (Thamnophis sirtalis parietalis) reveal that copulation duration and copulatory plug deposition, as well as total and oviductal/vaginal sperm counts, are influenced by the interaction between male and female genital traits and female behaviour during copulation. By mating females with anesthetized cloacae to males with spine‐ablated hemipenes using a fully factorial design, we identified significant female–male copulatory trait interactions and found that females prevent sperm from entering their oviducts by contracting their vaginal pouch. Furthermore, these muscular contractions limit copulatory plug size, whereas the basal spine of the male hemipene aids in sperm and plug transfer. Our results are consistent with a role of sexual conflict in mating interactions and highlight the evolutionary importance of female resistance to reproductive outcomes.  相似文献

The testes, seminal vesicles and spermathecae of 22 species of mosquitoes belonging to the genera Aedes, Anopheles, Culex, Mansonia and Toxorhynchites are investigated under the electron microscope. Modifications of the acrosome and sperm wall occur during the transit of the spermatozoon from the lower region of the testes to the spermathecae throughout the seminal vesicles. The origin and fate of the cell coat and the possible roles of somatic cell layers both in the testes and the seminal vesicles are discussed.  相似文献

Although the frequently large variance in relative male fertilizationsuccess when females are mated by more than 1 male has beenappreciated for some time, the factors that influence relativepaternity are still poorly understood. Recently, experimentalevidence that morphology of male genitalia influences fertilizationsuccess has been documented in 2 water striders, a dung beetle,and a leaf beetle. We explored the role of male genital morphologyin postcopulatory sexual selection in the oriental beetle. Wemated females to 2 males in succession and assessed relativepaternity by the sterile male technique. Morphology of the malegenitalia was found to strongly influence relative paternitybut only for the first male to mate. Male body size influencedrelative fertilization success as well, but again, only forthe first male; surprisingly, smaller males achieved higherpaternity when mating first. We also found suggestive evidencethat copula duration of both the first and second male to mateinfluenced paternity. Other factors, including female size anddegree of asymmetry of hind tibiae length of males had no effecton relative fertilization success. Our results for the orientalbeetle are novel among sperm precedence studies for 2 reasons:1) traits of the first male appear to be more important in influencingpaternity than those of the second, and 2) smaller, not larger,males achieved greater relative paternity. Our results alsocontribute to the growing body of empirical evidence in supportof the hypothesis that male genitalia evolve by postcopulatorysexual selection.  相似文献

The anatomy of the male and female reproductive systems was investigated in the long‐tailed butterfly ray Gymnura poecilura using gross observation and light microscopy. The testes are highly asymmetrical, to the extent that only the left testis is functional and the right testis is completely absent. Both of the male genital ducts are present and symmetrical, although spermatozoa only occur in the left duct. The genital ducts are straight and unconvoluted, with regular incomplete internal partitions throughout. Females do not possess a right ovary, nor do the oviducal glands exhibit distinct club and papillary zones, and the baffle zone lacks baffle plates. In all sections of the gland, the tubules display different secretory activities depending on the proximity to the gland lumen. The gland produces a thin egg membrane that encases each egg individually, while the endometrium is formed into trophonemata.  相似文献

The effects on mouse embryo development in vivo of varying the numbers of spermatozoa used in artificial inseminations was studied. The two criteria used in the evaluation of the progress of embryo development were 1) ability to reach the two-cell stage and 2) success of development from the two-cell stage through implantation. A 44% reduction in the yield of two-cell embryos and a 67% reduction in the number of implants was observed when C3HeB/FeJ females were inseminated with one-twentieth the number of spermatozoa estimated to be present in a typical ejaculate. The reduction in the yield of two-cell embryos was substantially reversed by a second insemination of a large number of heat-inactivated spermatozoa 12 hr after the first insemination. The sperm-dependent reduction in development from the two-cell stage through implantation was prevented only by normal viable (unheated) spermatozoa. These results were rationalized by the hypothesis that in female C3HeB/FeJ mice spermatozoa serve physiological functions beyond the fertilization of ova and that spermatozoa may act to foster early embryo development through modulation of the environments embryos experience as they move through the reproductive tract.  相似文献

It is now widely recognized that sexual selection has been important in the rapid and divergent evolution of male genital morphology. However, distinguishing among putative mechanisms of sexual selection acting on male genital morphology represents a considerable challenge. Although there is growing evidence that variation in the size and/or shape of male genital structures can determine a male's success in gaining fertilizations, our knowledge of the functional morphology of male genitalia remains limited. Here we examine the functional morphology of genital sclerites that are known to influence paternity in the dung beetle Onthophagus taurus . We show that three of the sclerites form a functionally integrated unit that generates the tubular-shaped spermatophore and delivers its opening to the female's spermathecal duct. A fourth sclerite acts as a holdfast device during copulation. Divergent sexual selection within allopatric populations may result in divergent sexual phenotypes, which can act as reproductive barriers between populations upon secondary contact. This hypothesis has been most tested on traits involved in precopulatory sexual selection, with less work focusing on traits that act after copulation and before fertilization (i.e., postcopulatory prezygotic traits), particularly in internally fertilizing vertebrates. However, postcopulatory sexual selection within species can also drive trait divergence, resulting in reduced performance of heterospecific sperm within the female reproductive tract. Such incompatibilities, arising as a by‐product of divergent postcopulatory sexual selection in allopatry, can represent reproductive barriers, analogous to species‐assortative mating preferences. Here, we tested for postcopulatory prezygotic reproductive barriers between three pairs of taxa with diverged sperm phenotypes and moderate‐to‐high opportunity for postcopulatory sexual selection (barn swallows Hirundo rustica versus sand martins Riparia riparia, two subspecies of bluethroats, Luscinia svecica svecica versus L. s. namnetum, and great tits Parus major versus blue tits Cyanistes caeruleus). We tested sperm swimming performance in fluid from the outer reproductive tract of females, because the greatest reduction in sperm number in birds occurs as sperm swim across the vagina. Contrary to our expectations, sperm swam equally well in fluid from conspecific and heterospecific females, suggesting that postcopulatory prezygotic barriers do not act between these taxon pairs, at this stage between copulation and fertilization. We therefore suggest that divergence in sperm phenotypes in allopatry is insufficient to cause widespread postcopulatory prezygotic barriers in the form of impaired sperm swimming performance in passerine birds.  相似文献

Mechanisms were sought through which the control of preimplantation mouse embryo development by spermatozoa might be effected. A potential route for the transmission of sperm-dependent stimuli to C3HeB/FeJ females was uncovered. It was found that within 24–48 hr after artificial insemination with spermatozoa, in which the DNA had been labeled with tritiated thymidine, a minimum of 9% of the radioactivity was transported across the uterine walls. It was deposited among the maternal tissues in a pattern that differed from the patterns of isotope distribution obtained when either free tritiated thymidine or Escherichia coli cells containing DNA labeled with tritiated thymidine were used instead of labeled sper-matozoa. In sperm-treated animals the ovaries, the adrenals, and a mesenteric lymph node exhibited strikingly large accumulations of radioactivity. The heart, spleen, and uterus manifested lesser accumulations of label, but were higher than liver, kidney, lung, brain, muscle, and intestine. The specific activity of the lymph node was found to decrease during the 12–72-hr period following insemination. This result led to the hypothesis that the lymphatic system could serve as a route for the dissemination, to maternal tissues, of radioactivity originally associated with spermatozoa deposited in the uterus. Heat-inactivated spermatozoa, which have the potential for facilitating the first cleavage of fertilized embryos, exhibited a distribution pattern indistinguishable from untreated spermatozoa. Sperm protein kinase was found to survive the heat inactivation of spermatozoa. This stability was interpreted as being compatible with the kinase functioning as an intermediary in the transmission of sperm-dependent stimuli that control preimplantation embryo development in mice.  相似文献

Meiotic metaphases II from archival slides were studied of male house mice caught in a hybrid zone between a population monomorphic for nine centric fusions (2n = 22) and a population with the standard karyotype (2n = 40), near Rome. The frequency of aneuploidy increases, up to 50%, with increasing number of heterozygous centric fusions (1–4). In the coral reef fish Thalassoma bifasciatum , males vary the number of sperm they release in successive spawnings with individual females in accordance with female body and clutch size. The morphology and histological structure of the male genital papilla, sperm duct, oviduct and surrounding musculature were examined in an effort to elucidate the mechanism permitting control of the number of gametes released during mating. In males, urinary and genital ducts pass separately through a common urogenital papilla and are associated with a striated sphincter muscle and a pair of thin, smooth ligament muscles arising from the first proximal anal fin radial and passing laterally around the sperm duct and oviduct. Within the papilla, the sperm duct resembles a narrow funnel whose inner walls contain longitudinal folds or septa protruding into the lumen of the duct. Dorsal to the papilla, the sperm duct enlarges and is divided into numerous, open chambers by irregular, longitudinal trabeculae. The wall of the duct and the trabeculae contain flat epithelium, smooth muscle and loose connective tissue. In females, the oviduct contains no trabeculae and is not divided into chambers. The ligament muscles are more thoroughly embedded in the sphincter muscle of the rectum than in males. Some ways in which these structures might control gamete release are suggested.  相似文献

Mice of the PL/J strain exhibit a high percentage of morphologically abnormal sperm and provide a model for studying the function of abnormal sperm. The ability of such sperm to reach the site of fertilization within the female reproductive tract has been investigated. We have found a decrease in the percentage of structurally abnormal sperm within the population that reaches the oviduct. This observation suggests either that there is an active selection against abnormal sperm or that they are physiologically disadvantaged in reaching the site of fertilization.  相似文献

Sperm competition is a pervasive force. One adaptation is the male ability to displace the rivals' sperm that females have stored from previous copulations. In the damselfly, Calopteryx haemorrhoidalis asturica , males with wider aedeagi displace more spermathecal sperm. The present study documents that the same mechanism operates in another damselfly, Hetaerina americana . However, this genital width in both species decreases along the season, but late-emerging females have more sperm displaced than early-emerging females. Because territorial males mated more and were larger in body and genital size than nonterritorial males, late-season females mated with considerably larger males with respect to female size and this produced higher sperm displacement. Assuming female benefits from storing sperm but that such benefit does not prevail if males displace sperm, it is predicted that, along the season, females will mate less and male harassment (in terms of male mating attempts and oviposition duration) will increase. These predictions were corroborated. In H. americana , it was also tested whether spermathecal sperm became less viable along the season. The results obtained did not corroborate this. The spectacular variability that typically characterizes male genital traits has largely been attributed to the role of sexual selection. Among the evolutionary mechanisms proposed to account for this diversity, two processes in particular have generated considerable interest. On the one hand, females may exploit postcopulatory mechanisms of selection to favour males with preferred genital traits (cryptic female choice; CFC), while on the other hand females may evolve structures or behaviours that mitigate the direct costs imposed by male genitalia (sexual conflict; SC). A critical but rarely explored assumption underlying both processes is that male and female reproductive traits coevolve, either via the classic Fisherian model of preference-trait coevolution (CFC) or through sexually antagonistic selection (SC). Here, we provide evidence for this prediction in the guppy (Poecilia reticulata), a polyandrous livebearing fish in which males transfer sperm internally to females via consensual and forced matings. Our results from a paternal half-sibling breeding design reveal substantial levels of additive genetic variation underlying male genital size and morphology—two traits known to predict mating success during non-consensual matings. Our subsequent finding that physically interacting female genital traits exhibit corresponding levels of genetic (co)variation reveals the potential intersexual coevolutionary dynamics of male and female genitalia, thereby fulfilling a fundamental assumption underlying CFC and SC theory.  相似文献

The males of two mosquito species within the Anopheles gambiae complex, An. gambiae s.s. and An. quadriannulatus, as well as males of An. darlingi, produced sperm of significantly varying lengths, while a sperm polymorphism was absent in Aedes aegypti and other anophelines not suspected of belonging to species complexes. The polymorphic distribution of these sperm lengths was not significantly different in smaller adult males that were reared on a low larval diet. The reproductive tract of the female was more likely to contain larger sperm, but overall sperm retention varied depending on the size of the female and the volume of the spermatheca she contained. The presence of a sperm polymorphism may be a factor that has promoted speciation, as well as providing an indication that females may mate multiply.  相似文献

Theory predicts that costly secondary sexual traits will evolve heightened condition dependence, and many studies have reported strong condition dependence of signal and weapon traits in a variety of species. However, although genital structures often play key roles in intersexual interactions and appear to be subject to sexual or sexually antagonistic selection, few studies have examined the condition dependence of genital structures, especially in both sexes simultaneously. We investigated the responses of male and female genital structures to manipulation of larval diet quality (new versus once‐used mung beans) in the bruchid seed beetle Callosobruchus maculatus. We quantified effects on mean relative size and static allometry of the male aedeagus, aedeagal spines, flap and paramere and the female reproductive tract and bursal spines. None of the male traits showed a significant effect of diet quality. In females, we found that longer bursal spines (relative to body size) were expressed on low‐quality diet. Although the function of bursal spines is poorly understood, we suggest that greater bursal spine length in low‐condition females may represent a sexually antagonistic adaptation. Overall, we found no evidence that genital traits in C. maculatus are expressed to a greater extent when nutrients are more abundant. This suggests that, even though some genital traits appear to function as secondary sexual traits, genital traits do not exhibit heightened condition dependence in this species. When females mate with multiple partners within a single reproductive cycle, sperm from rival males may compete for fertilization of a limited number of ova, and females may bias the fertilization of their ova by particular sperm. Over evolutionary timescales, these two forms of selection shape both male and female reproductive physiology when females mate multiply, yet in monogamous systems, post-copulatory sexual selection is weak or absent. Here, we examine how divergent mating strategies within a genus of closely related mice, Peromyscus, have shaped the evolution of reproductive traits. We show that in promiscuous species, males exhibit traits associated with increased sperm production and sperm swimming performance, and females exhibit traits that are predicted to limit sperm access to their ova including increased oviduct length and a larger cumulus cell mass surrounding the ova, compared to monogamous species. Importantly, we found that across species, oviduct length and cumulus cell density are significantly correlated with sperm velocity, but not sperm count or relative testes size, suggesting that these female traits may have coevolved with increased sperm quality rather than quantity. Taken together, our results highlight how male and female traits evolve in concert and respond to changes in the level of post-copulatory sexual selection.  相似文献

The meiotic segregants of male mice heterozygous for Rb(6.16)24Lub and Rb(16.17)7Bnr were viewed, for the first time, at first cleavage metaphase. Chromosomes were analyzed after G-banding, C-banding, and karyotyping. To study sperm aging effects, chromosomes of 202 one-cell zygotes derived from males mating at intervals of approximately 3,14, and 21 days were examined. At least 89.6% of sperm-derived complements were products of 2:2 segregation; at most, a possible 6.4% were 3:1 segregants. The six expected types of 2:2 segregants, both balanced and unbalanced, were equifrequent in the total zygote population derived from sperm of all ages. When the data were analyzed according to mating frequency, the 3-day sperm population considered most likely to be fresh showed a deficiency of the segregant nullisomic for chromosome 6 and disomic for chromosome 17, when compared with the reciprocal segregant (P < 0.025) as well as to all other 2:2 segregants (P < 0.05). However, these sperm fertilized in greater numbers (P < 0.01) than their reciprocal segregant (disomic for 6 and nullisomic for 17) in the 14-day sperm population. While sperm with chromosomal abnormalities are capable of fertilization, the competence of segregants nullisomic for 6 and disomic for 17 apparently depends on the prior storage period in the male. Further, the results suggest that the effect of aneuploidy on sperm function is dependent on the specific chromosome(s) involved.  相似文献
