We have made an extensive allozyme survey of 21 enzyme and protein loci in populations of the alpine grasshopper Podisma pedestris. This species occurs in two races, differing by a chromosomal fusion which separates the ancestral XO/XX race from a derived neo-XY race. These races also differ in DNA content, and hybrids between them have reduced viability. Electrophoresis reveals that the amount of genetic differentiation between these races is no greater than the variation among populations within each race. Both larger-scale surveys and a detailed survey of an area where the races hybridize, show that the chromosomal change is not correlated with gene frequency changes at any of the 21 loci studied. These findings are consistent with recently developed theory concerning the strength of the barrier to gene flow posed by a hybrid zone with characteristics such as those measured experimentally in Podisma. It is argued that hybrid zones in other species which involve allozymic differences do so because of stronger selection against hybrids rather than through mating isolation.  相似文献

Summary Indian populations divided into a number of endogamous groups consisting of different castes, languages, religions, and tribes provide unique opportunities for examining the extent and nature of genetic differentiation at a microevolutionary stage. The genetic relationships between some of these Indian population groups have been examined using electrophoretic data from several biochemical loci in a gene diversity analysis. Does this type of analysis provide any insight into what causes such gene differentiation? What patterns of genetic variation emerge from these empirical findings? Answers are sought by relating the observed heterozygosity, genetic distance, and allied statistics to a mutation-drift hypothesis. The statistics used are: (1) interlocus mean and variance of heterozygosity, (2) mean and variance of genetic distance, and (3) correlation of heterozygosity and gene identity. The observed relationships between these sets of statistics agree well with the ones predicted by the hypothesis that different alleles at protein loci are selectively equivalent and gene frequency change occurs predominantly due to genetic drift  相似文献

Allozyme genetic variability in three chestnut (Castanea) species was investigated using 19 loci from ten enzyme systems. G-tests of heterogeneity of isozymic allele distribution showed significant differences between the three species at 15 of the 19 loci, and between the 13 C. mollissima populations at 13 of the 19 loci examined. C. mollissima was found to possess a significantly-higher value of mean gene heterozygosity (H=0.3050±0.0419), the percentage of polymorphic loci (P=84.21%) and the average number of alleles per locus (A=2.05), than any other species in the Castanea section Eucastanon. When the genetic variability of populations of C. mollissima from four regions in China was investigated, the population from the Changjiang river region showed a markedly higher mean gene heterozygosity (H=0.3480±0.0436) than populations from the other regions. Genetic relationships among the four regions were assessed by Nei's genetic identity I and standard genetic distance D. An approximately-identical distance between the population from the Changjiang river region and populations from the three other regions was observed, while populations from the latter regions showed almost the same genetic distance from each other. These data, when considered with information existing prior to this study, contribute to an understanding of the possible origin and progenitor of the chestnut species.  相似文献

Mathematical formulae for the sampling variances of average heterozygosity and Nei's genetic distance are developed. These sampling variances are decomposed into their two components, i.e. the inter-locus and intra-locus variances. The relationship between the number of loci and the number of individuals per locus to be examined for estimating average heterozygosity and genetic distance is also discussed. The utility of the inter-locus variance of heterozygosity for studying the mechanism of maintenance of genetic variability in populations is indicated.  相似文献

Genetic diversity within and among six subpopulations of Larix decidua Mill. from two altitudinal transects of Swiss Alps was investigated using 6 enzyme systems coding for 8 loci. Globally, the mean proportion of polymorphic loci was 22.9%, the average number of alleles per locus was 1.3, and the mean expected heterozygosity was 0.095. Only 5.8% of the genetic variation resided among populations. The mean genetic distance was 0.006. Several significant differences of gene frequencies were found between different age classes. Positive values of the species mean fixation index observed in this study suggested a considerable deficit of heterozygotes in the populations of L. decidua of Swiss Alps. At one of the sites (Arpette), the highest subpopulation in elevation gave the lowest level of genetic diversity (as evidenced by the lowest proportion of polymorphic loci and the lowest mean expected heterozygosity) and the largest value of genetic distance when compared to other subpopulations. The genetic differences between the highest subpopulation and the other ones suggest that the founder effect may be an important factor influencing genetic differentiation of L. decidua populations at Arpette transect.  相似文献

Abstract. It has been long recognized that highly polymorphic genetic markers can lead to underestimation of divergence between populations when migration is low. Microsatellite loci, which are characterized by extremely high mutation rates, are particularly likely to be affected. Here, we report genetic differentiation estimates in a contact zone between two chromosome races of the common shrew ( Sorex araneus ), based on 10 autosomal microsatellites, a newly developed Y-chromosome microsatellite, and mitochondrial DNA. These results are compared to previous data on proteins and karyotypes. Estimates of genetic differentiation based on F - and R -statistics are much lower for autosomal microsatellites than for all other genetic markers. We show by simulations that this discrepancy stems mainly from the high mutation rate of microsatellite markers for F -statististics and from deviations from a single-step mutation model for R -statistics. The sex-linked genetic markers show that all gene exchange between races is mediated by females. The absence of male-mediated gene flow most likely results from male hybrid sterility.  相似文献

Two South African populations of Littorina saxatilis were examined by starch-gel electrophoresis at 16 enzyme loci and compared with 13 populations of North Atlantic saxatilis from both American and European coasts, and with six British populations of the closely related species Littorina arcana. The South African animals showed a severely reduced heterozygosity (– 0.052) compared with Atlantic populations of saxatilis ( = 0.181), and the mean genetic distance between the two areas was high ( = 0.203) compared with distances within the North Atlantic saxatilis populations (D = 0.034). In fact, the saxatilis from South Africa were genetically more distant from the North Atlantic samples of L. saxatilis than were the arcana from British shores. The reduced genetic heterozygosity and genetic divergence of the South African populations is attributed to founder effects following a postulated recent introduction by man.  相似文献

Morphological and genetic variation at microsatellite loci of Caucasian shrew Sorex satunini Ogn. is examined and compared with that of the common shrew S. araneus L. Genetic distance at microsatellite loci between the common shrew and Caucasian shrew proved to be threefold higher than between chromosome races of the common shrew. The Caucasian shrew manifested low polymorphism in studies of both microsatellites and morphometric mandibular traits. The heterozygote deficit was also typical. A study of genic variation in natural population of D. melanogaster was undertaken (1) to obtain a better estimate of heterozygosity by sampling a relatively large number of gene loci and (2) to identify different groups of polymorphic loci whose variation patterns might suggest different kinds of selection forces. A total of 117 gene loci (coding for 79 enzymes and 38 abundant proteins) were studied in 15 geographically distant populations originating from different continents. The findings of this study are as follows: (1) of the 117 gene loci studied, 61 are polymorphic and 56 are uniformly monomorphic everywhere. (2) An average population is polymorphic for 43% of its gene loci and an average individual is heterozygous for 10% of its gene loci. These estimates are remarkably similar among populations. (3) The average within-locality heterozygosity (H(S)) for polymorphic loci is uniformly distributed over the range of heterozygosity observed; i.e. , given that a locus has any local variation, it is nearly as likely to have a lot as a little. (4) The distribution of F(ST) (fixation index) is strongly skewed, with a prominent mode at 8-10% and a long tail of high values reaching a maximum of 58%. Two-thirds of all loci fall within the bell-shaped distribution centered on an F(ST) of 8-10%, a result compatible with the notion that they are experiencing a common tendency toward small interlocality differences owing to extensive gene flow among populations. (5) The distribution of total heterozygosity (H(T)) has a prominent bimodal distribution. The lower mode consists of loci with single prominent allele and a few uncommon ones and the upper mode consists of clinally varying loci with a high F(ST ) (e.g., Adh and G6-pd), loci with many alleles in high frequency (e.g., Ao and Xdh) and loci with two alleles in high frequency in all populations but, with little interpopulational differentiation (e.g., Est-6 and alpha-Fuc). The loci in the lower mode are probably under purifying selection; a large proportion of those in the latter mode may be under balancing selection. (6) Comparison of genic variation for loci located inside vs. outside inversions, comparison of F(ST) for inversions and their associated genes, and comparison of F(ST) and map position for pairs of loci all suggest that, while linkage has some influence, it does not seem to constrain the pattern of variation that a locus may develop. (7) Eighteen polymorphic loci show latitudinal variation in allele frequencies which are consistent in populations from different continents. (8) Estimates of Nei genetic distance between population pairs are generally low between populations on the same continent and high between populations on different continents. There are two important exceptions: population pairs for which both localities are in the temperate zone show no relationship to distance, and in cases where both populations are tropical or subtropical, the genetic distance is higher than for the temperate-tropical comparisons and seem even higher than one would expect from the geographic distance separating them. The latter observation suggests that either geographic separation outweighs differences in environment in determining the genetic composition of a population or that all tropical populations are not experiencing the same environment.-The results are discussed in relation to the neutralist-selectionist controversy of genic variation and two important conclusions are drawn: First, there is a negative correlation between the number of loci sampled and the resulting heterozygosity. This means that available estimates of heterozygosity, 85% of which are based on 30 or fewer loci, are high and hence not appropriate for making between-taxa comparisons. Secondly, there is a group of loci, comprising one-third of polymorphic loci (or about 15% of all loci studied), that is distinguishable by different patterns of variation within and among populations. Most of these loci have clinal variation which is consistent with the hypothesis that their genetic variation is maintained by balancing selection.  相似文献

This study investigated a chromosome hybrid zone between two chromosomal races of the common shrew (Sorex araneus). Gene flow and genetic structure of the hybrid zone, located in the northeast of Poland, were studied using seven polymorphic autosomal microsatellite loci (L9, L14, L33, L45, L67, L68, L97) and a Y-linked microsatellite locus (L8Y). Seventy-five animals (46 of the ??gucki M?yn race and 29 of the Popielno race) from nine different localities were examined and the data were analyzed using hierarchical AMOVA and F-statistic. The studied microsatellite loci and races (divided into nine geographical populations) were characterized by observed heterozygosity (H(O)), expected heterozygosities within (H(S)), and between (H(T)) populations, inbreeding coefficient (F(IS)), fixation index (F(ST)), and average allelic richness (A). We found that genetic structuring within and between the two chromosome races were weak and non-significant. This finding and unconstrained gene flow between the races indicates a high level of migration within the ??gucki M?yn/Popielno hybrid zone, suggesting that evolutionarily important genetic structuring does not occur in interracial zones where races which are not genetically distinct come into contact.  相似文献

Vertical slab polyacrylamide gel electrophoresis was used to examine inheritance of four isoenzymes in megagametophytes of Picea koraiensis. Gene diversity and genetic differentiation in four natural populations of P. koraiensis in Northeastern China were analyzed at six loci coding for four enzyme systems. The proportion of loci polymorphism in population was 0.5. The average expected heterozygosity of all samples was 0. 314, and the average observed heterozygosity of all samples was 0. 316. The average number of alleles detected per locus was 2.50. The effective number of alleles per locus was 1. 71. Measurement of gene diversity for the six loci showed a 0. 0059 significance of interpopulation differentiation. More than ninety-nine percent of the total gene diversity resided within population. Every population possesses genetic variations which are achieved through gene mutation, genetic recombination, hybridization, gene duplication etc. These genetic variations provide raw materials for evolutionary forces to create a better surviving species. Genetic polymorphism is reflected at every level in the populations, for example, at phenotypic, chromosomal, protein and DNA levels. Protein or enzyme polymorphisms have been well studied in various organisms including Drosophila and humans. Drosophila has proven to be a good model organism for carrying out polymorphism studies. Among the different species of Drosophila, there is a wide variation in the levels of allozyme polymorphisms and heterozygosities which depends upon species, geographical regions, number and nature of loci in question etc. In Drosophila, the average polymorphic enzyme loci and average heterozygosity ranges from 35 to 70 percent and 10 to 20 percent respectively. The genetic differentiation as observed through allozyme or isozyme variation affords an important parameter in evaluating the phylogenetic relationships between different species of Drosophila and also for discussing the adaptive significance of allozyme polymorphisms. Therefore, this review attempts to compile all studies on allozyme polymorphism in Drosophila that have been undertaken so far.  相似文献

The genetic structure of two brown trout Salmo trutta populations living in Lake Abant in Bolu and Üzüm River in Antalya was determined by examining 15 enzyme coding loci ( AAT, ADH, LDH, MDH, MEP, GPI, PGM and SOD ) using starch gel electrophoresis. Population specific mobilities were observed for the fixed alleles of LDH-B2, mMEP-2 and SOD-1 loci. Polymorphisms in sAA T-4, GPI-B2 loci were observed within the populations. Average heterozygosity of Abant and Antalya populations was 0.0358 and 0.0224 respectively. For LDH-C which is the post glaciation marker locus, the ancestral allele * 105 was found to be fixed in both of the populations. Nei's genetic distance between the two populations was 0.2507 which is the level of genetic distance often found between different species. This difference seems to be due to the presence of unique alleles in the LDH-B2, mMEP-2 and SOD-1 loci of the Abant population, indicating that the conservation of the Abant population and its heterozygosity is of prime importance.  相似文献

The house mouse, Mus musculus domesticus, exhibits a high level of chromosomal polymorphism because of the occurrence and fast fixation of Robertsonian fusions between telocentric chromosomes. For this reason, it has been considered a classical speciation model to analyse the role of the chromosomal changes in reproductive isolation. In this study, we analysed a parapatric contact area between two metacentric races in central Italy, the Cittaducale race (CD: 2n = 22) and the Ancarano race (ACR: 2n = 24), to estimate gene flow at the boundary. Hybrids between these two races show high levels of structural heterozygosity and are expected to be highly infertile. A sample of 88 mice from 14 sites was used. The mice were genotyped by means of eight microsatellite loci mapped in four different autosomal arms. The results show clear genetic differentiation between the CD and ACR races, as revealed by differences in allele frequencies, factorial correspondence analysis and indexes of genetic population (e.g. F(ST) and R(ST)) along the contact zone. The genetic differentiation between the races was further highlighted by assignation and clustering analyses, in which all the individuals were correctly assigned by their genotypes to the source chromosomal race. This result is particularly interesting in view of the absence of any geographical or ecological barrier in the parapatric contact zone, which occurs within a village. In these conditions, the observed genetic separation suggests an absence of gene flow between the races. The CD-ACR contact area is a rare example of a final stage of speciation between chromosomal races of rodents because of their chromosomal incompatibility.  相似文献

Several models of multiple slightly deleterious alleles are reviewed and theoretical consequences of slightly negative selection are discussed in conjunction with evolution and variation at the molecular level. Facts are organized which may be satisfactorily explained by the hypothesis of very slightly deleterious mutations. They are: (1) There appears to be an upper limit in heterozygosity for protein loci as measured by electrophoresis. (2) The excess of rare alleles is more pronounced in Drosophila than in man. (3) Correlation of heterozygosities at a locus among sibling species of the Drosophila willistoni group is too high compared to what is expected by the strict neutral theory, while it is not so among human races and between man and chimpanzee. (4) The rate of protein divergence is exceptionally high in Hawaiian Drosophila.  相似文献

Several studies have reported positive correlations between the degree of enzyme heterozygosity and fitness-related traits. Notable among these are the correlations between heterozygosity and growth rate in marine bivalves. Whether the correlation is the result of intrinsic functional differences between enzyme variants at the electrophoretic loci scored or arises from non-random genotypic associations between these loci and others segregating for deleterious recessive genes (the associative overdominance hypothesis) is a matter of continuing debate. A prediction of the associative overdominance hypothesis, not shared by explanations that treat the enzyme loci as causative agents of the correlation, is that the correlation is not specific to the type of genetic marker used. We have tested this prediction by scoring heterozygosity at single locus nuclear restriction fragment length polymorphisms (RFLPs) in a cohort of juvenile scallops (Placopecten magellanicus) in which growth rate was known to be positively correlated with an individual's degree of allozyme heterozygosity. A total of 222 individuals were scored for their genotypes at seven allozyme loci, two nonspecific protein loci of unknown function and eight nuclear RFLPs detected by anonymous cDNA probes. In contrast to the enzyme loci, no correlation was observed between growth rate and the degree of heterozygosity at the DNA markers. Furthermore, there was no relationship between the magnitude of heterozygote deficiency at a locus and its effect on the correlation. The differences observed between the effects of allozyme and RFLP heterozygosity on growth rate provide evidence against the associative overdominance hypothesis, but a strong case against this explanation must await corroboration from similar studies in different species.  相似文献

Using the stepwise mutation model of Ohta and Kimura (1973), formulas are developed for the correlation of heterozygosity and the variance of genetic distance between two finite populations. Studied in detail is the case where the sizes of the two descendant populations are equal to that of the ancestral population and the mutation rate is the same for all loci. Numerical computations are carried out by using the present formulas and those of Li and Nei (1975Genet. Res.25) for the infinite-allele model. The results are as follows: The correlation of heterozygosity decreases with time faster for the stepwise mutation model than for the infinite-allele model. However, the relationships between the correlation of heterozygosity and the normalized genetic identity for the two models are very similar, if the average heterozygosities of the two populations are around 0.20 or less. On the other hand, the variance of genetic distance for the stepwise mutation model may become considerably smaller than that for the infinite-allele model, if the average heterozygosities of the two populations are larger than 0.05. The ratio of the standard deviation to the mean is, however, very large for the stepwise mutation model as well as the infinite-allele model.  相似文献

The objectives of this study were (1) to investigate the genetic variability; and (2) to determine the phylogenetic relationship in the leguminous tree Faidherbia albida through out its range of distribution in East, West and Southern Africa. A total of 16 populations were subjected to enzyme electrophoresis and 6 enzyme systems (Adh, Mdh, G6pdh, Idh, 6Pgdh, and alpha-Est) encoded by 12 loci were scored. A total of fifty one different alleles were detected, with an average of 2.5 alleles per locus. Forty three percent of the loci were polymorphic at a 95% criterion. The average expected heterozygosity (gene diversity index H(e)) was 0.141. All provenances showed significant deviation from Hardy-Weinberg expectation. The UPGMA cluster analysis, based on Modified Rogers distance revealed close similarities between Eastern and Southern African provenances, except Debre zeit from Ethiopia, which was closest to West African populations than to the East African populations. Also, Bignona from West Africa was peculiarly closer to East African provenances. Differentiation of West African populations from Eastern and Southern African provenances was quite evident, emphasising the clinical pattern of variation in these two major geographical races of F. albida in Africa. Wright's F-statistics showed an overall significant deficit of heterozygotes, a common feature in mixed mating, entomophilous, widespread species such as F. albida. The dendrogram analysis showed wide separation among the three Ethiopian provenances indicating a high level of genetic differentiation and diversity among them.  相似文献
