Summary Gametes and developing larvae of the polychaete Galeolaria caespitosa were examined by scanning electron microscopy. The sperm display a primitive morphology. When treated with 0.33 M CaCl2, they release a branched acrosomal process. At spawning, the polygonal oocytes have a granular surface made up of spherules and the tips of microvilli. The oocyte coat develops a ridged appearance as the oocyte rounds up. At fertilization, the microvilli are withdrawn from the coat surface. Microvilli again appear on the coat surface during the trochophore stage, but the egg coat appears to be retained as the larval cuticle until the demersal stage. The surface of the larva now shows many microvilli. Details of the organization of several ciliary structures are clarified. Moreover, the present study shows rapid, sequential development of paired setal sacs, with the most anterior pair appearing first.  相似文献

To determine when the growing hamster oocyte gains the ability to fuse with the spermatozoon, oocytes at various stages of development were collected from ovaries, and zona-pellucida-free oocytes were inseminated in vitro with acrosome-reacted spermatozoa. Very small primary oocytes were unable to fuse with spermatozoa. Oocytes first became competent to fuse with spermatozoa when they had grown to about 20 microns in diameter. The acquisition of fusibility coincided with the first appearance of zona pellucida material and oolemma microvilli. The fusibility of the oolemma increased as the oocyte grew, reaching a maximum when the oocyte reached the metaphase of the second meiosis. The fusibility of the oolemma was reduced drastically after fertilization, and was lost completely by the 8-cell stage. The appearance and subsequent disappearance of a putative fusion-mediating molecule in the oolemma is proposed. Since this molecule is fairly resistant to proteinase digestion, at least in the hamster, it could be a cryptic protein or a glycolipid.  相似文献

We previously demonstrated that the number and height of oocyte microvilli were reduced in baboon fetuses deprived of estrogen in utero and restored to normal in animals supplemented with estradiol. Phosphorylated ezrin and Na+/H+ exchange regulatory factor 1 (NHERF, now termed SLC9A3R1) link f-actin bundles to the membrane, whereas alpha-actinin cross-links f-actin to form microvilli. Therefore, we determined whether these proteins were expressed in oocytes of the fetal baboon ovary and whether expression and/or localization were altered between mid and late gestation in association with an increase in estrogen and in late gestation in animals in which estrogen was suppressed (>95%) or restored by treatment with an aromatase inhibitor with or without estradiol. Expression of alpha-actinin was low at mid gestation, increased on the surface of oocytes of primordial follicles in late gestation, and was negligible in the ovaries of estrogen-suppressed fetuses and normal in animals treated with estrogen. Ezrin (total and phosphorylated) and SLC9A3R1 expression was localized to the surface of oocytes at mid and late gestation in estrogen-replete baboons and to the cytoplasm in late gestation after estrogen suppression. These results are the first to show that the fetal baboon oocyte expressed ezrin, SLC9A3R1, and alpha-actinin, and that these proteins were localized to the oocyte surface consistent with their role in microvilli development in epithelial cells. The current study also showed that the developmental increase in oocyte expression of alpha-actinin is regulated by estrogen and correlated with the estrogen-dependent increase in oocyte microvilli demonstrated previously. Therefore, we propose that development of oocyte microvilli requires expression of alpha-actinin and that expression of alpha-actinin and localization of ezrin-phosphate and SLC9A3R1 to the oocyte membrane are regulated by estrogen.  相似文献

The present investigation describes the ultrastructural changes which occur at the surface and in the cytoplasm of developing oocytes of the lobster, Homarus americanus, during vitellogenesis. The immature oocytes showed no surface specializations of the oolemma and no pinocytotic activity was observed. Horseradish peroxidase (HRP) tracer studies showed penetration of the tracer into the perivitelline space, but no uptake by the oocytes. The surfaces of oocytes examined during vitellogenesis, when yolk protein accumulation was maximal, exhibited numerous microvilli that projected into the perivitelline space, often appearing to be embedded in the follicular cell mass. In addition, the plasma membrane of vitellogenic oocytes contained many pinocytotic pits frequently situated at the bases of microvilli. The perivitelline space was engorged with electrondense material which appeared similar to that contained in pinocytotic structures of the oocytes. Vitellogenic oocytes incubated in HRP showed uptake of tracer reaction product by the coated pits and vesicles of the oolemma. Aggregation and subsequent fusion of these vesicles into large multivesicular bodies of ingested material were also observed in vitellogenic oocytes. Animals artificially induced to undergo vitellogenesis exhibited modulations of oocyte ultrastructure similar to those of normal vitellogenesis, notably, pinocytotic incorporation of extra-oocytic material and hypertrophy of oocyte surface microvilli. This study supports the hypothesis for a dual source of yolk protein in the American lobster.  相似文献

By the end of previtellogenesis, the oocytes of Glossiphonia heteroclita gradually protrude into the ovary cavity. As a result they lose contact with the ovary cord (which begins to degenerate) and float freely within the hemocoelomic fluid. The oocyte's ooplasm is rich in numerous well-developed Golgi complexes showing high secretory activity, normal and transforming mitochondria, cisternae of rER and vast amounts of ribosomes. The transforming mitochondria become small lipid droplets as vitellogenesis progresses. The oolemma forms microvilli, numerous coated pits and vesicles occur at the base of the microvilli, and the first yolk spheres appear in the peripheral ooplasm. A mixed mechanism of vitellogenesis is suggested. The eggs are covered by a thin vitelline envelope with microvilli projecting through it. The envelope is formed by the oocyte. The vitelline envelope is produced by exocytosis of vesicles containing two kinds of material, one of which is electron-dense and seems not to participate in envelope formation. The cortical ooplasm of fully grown oocytes contains many cytoskeletal elements (F-actin) and numerous membrane-bound vesicles filled with stratified content. Those vesicles probably are cortical granules. The follicle cells surrounding growing oocytes have the following features: (1) they do not lie on a basal lamina; (2) their plasma membrane folds deeply, forming invaginations which eventually seem to form channels throughout their cytoplasm; (3) the plasma membrane facing the ovary lumen is lined with a layer of dense material; and (4) the plasma membrane facing the oocyte forms thin projections which intermingle with the oocyte microvilli. In late oogenesis, the follicle cells detach from the oocytes and degenerate in the ovary lumen.  相似文献

We have developed a coupled Xenopus oocyte expression system for evaluating the functional effects of mutations in known or suspected adhesion molecules, which allows for a very rapid assessment of intercellular adhesion. As a model protein, we first used Protein zero (Po), an adhesion molecule that mediates self-adhesion of the Schwann cell plasma membrane to form compact myelin in the mammalian PNS. A wide variety of mutations in Po cause certain human peripheral neuropathies, such as the Charcot-Marie-Tooth disease (CMT) type 1B and Dejerine-Sottas syndrome (DSS). After wild-type Po mRNA is injected, the protein is synthesized and correctly targeted to the oocyte cell surface. When two oocytes are paired, wild-type Po redistributes and concentrates at the cell-cell apposition region, and by electron microscopy, the oocyte pairs show close cell-cell appositions and are devoid of the microvilli that are observed in uninjected oocyte pairs. These are hallmark features of highly adhesive cell:cell interfaces. Several point mutations in Po were engineered, corresponding to the molecular defects in the CMT type 1B or DSS. The proteins encoded by these mutations reached the cell surface but failed to concentrate at the oocyte interface. Po carrying a point mutation that is found in DSS is not targeted on the plasma membrane and fail to accumulate at the cell-cell contact site.  相似文献

Oocytes of the starfish Pisaster ochraceus exhibit an early response to 1-methyladenine (the maturation-inducing hormone), which is described for the first time. In this response approximately 6,500 spikelike surface projections, much larger than microvilli, emerge transiently from oocytes stripped of their follicle cells and then treated with the hormone in vitro. Each spike contains a prominent bundle of microfilaments, possibly composed of actin. The distribution of spikes when follicle cells are only partially removed and the morphological details of the normal junctional association between follicle cells and oocytes suggest that 1-methyladenine-sensitive sites (receptor sites) can be identified with the approximately 6,500 postjunctional specializations that are part of the oocyte surface. This finding in turn is employed to construct a set of hypotheses concerning the route that 1-methyladenine normally takes from the follicle cells to an oocyte during stimulation of maturation; it is postulated that, for each oocyte, 1-methyladenine is transported along approximately 6,500 thin follicle-cell processes, it is transmitted across the junctional gaps of an equivalent number of junctions between follicle cells and an oocyte, and then interacts with the postjunctional sites where 1- methyladenine receptors are thought to be clustered. Comparative aspects of this mode of intercellular communication are discussed.  相似文献

Pathogenic yeast, Candida albicans, were incubated with hamster and human oocytes for up to 21 hours in order to determine the nature and time course of phagocytosis of these organisms. Aliquotes of the interacting cells were taken at various time intervals for electron microscopic examination. Some specimens had their zona pellucidae enzymatically removed prior to incubation with yeast, and these specimens showed the most extensive interaction and phagocytosis of Candida. The zona pullucida appears to be an effective barrier to yeast, at least over the time span studied. The observations are consistent with the hypothesis of an initial attachment of yeast via a surface component to oocyte microvilli followed by phagocytic uptake into an endosome. There is no compelling evidence of lysosomal degradation of the yeast over the time course of this study; however, the oocytes appear to undergo some degenerative changes at long incubation times.  相似文献

To examine the integration and fate of the sperm plasma membrane following its incorporation into the oocyte plasma membrane, we have fertilized sea urchin (Arbacia punctulata) gametes reciprocally labeled with cationized ferritin. When unlabeled oocytes were inseminated with labeled sperm, cationized ferritin acceptors moved laterally from the sperm plasma membrane into the fertilization cone and surrounding microvilli, mixing with components of the oocyte plasmalemma. Labeled oocytes inseminated with unlabeled sperm produced extremely large fertilization cones, completely devoid of cationized ferritin, while the remainder of the oocyte surface remained heavily labeled. Surface area measurements indicated that if all the sperm plasmalemma were utilized to delimit a fertilization cone it would provide less than 10% of the total surface membrane. Evidence is presented indicating that a principal source of membrane to the expanding fertilization cone of inseminated oocytes is from microvilli, i.e., microvilli are retracted to accommodate fertilization cone formation. Membrane delimiting the fertilization cone has a much lower affinity for agents (cationized ferritin and concanavalin A) that stain negatively charged and carbohydrate moieties compared to other regions of the oocyte surface. These ultrastructural observations indicate that significant rearrangements occur in the oocyte and sperm plasma membranes following gamete fusion which give rise to asymmetries in membrane topography; components of both membranes are redistributed within the bilayer adjacent to and delimiting the fertilization cone.  相似文献

A premature release of cortical granules (CG), found in the cortex of unfertilized oocytes, and the resulting formation of a CG-free domain (CGFD) over the metaphase II spindle are associated with nuclear maturation in the hamster and mouse. The objectives of our study were to characterize and compare the distribution of CG in immature, in vitro-matured, and in vivo-matured domestic cat oocytes while determining if a CGFD is formed that may be useful as a marker for stage and normalcy of oocyte maturation. Immature, intrafollicular oocytes were collected from ovaries obtained from local veterinary clinics, and a portion of these oocytes were matured in vitro. In vivo-matured, metaphase II oocytes were flushed from the oviducts of gonadotropin-treated, ovariohysterectomized cats. CG were visualized by Lens culinaris agglutinin-biotin/Texas red-strepavidin fluorescence, routine transmission electron microscopy, and Lens culinaris agglutinin-biotin/gold-labeled strepavidin transmission electron microscopy. No CGFD was detected in any domestic cat oocyte. Immature, in vitro-matured, and in vivo-matured oocytes had similar, uniform distributions of CG throughout the entire cortical region as measured by fluorescence microscopy. In vivo-matured oocytes were further examined by transmission electron microscopy, which confirmed the lack of a CGFD. All oocytes contained CG having a mean diameter of 0.28 +/- 0.03 micron/granule and a mean density of 51.5 +/- 13.0 CG/100 microns of plasma membrane.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Early in the spawning season female D. excentricus can be induced to spawn oocytes in the late stages of oogenesis. Observations with light microscopy indicate that pigment cells migrate in the jelly coat away from the surface of oocytes coincident with the final growth of the oocyte and its maturation. The pigment cells undergo a series of changes in shape and the oocyte elaborates arrays of long microvilli as the cells elevate from the surface of the oocyte. Transmission electron microscopy (TEM) observations of oocytes fixed within the ovary indicate that features of the sequence observed in the spawned oocytes normally take place within the ovary, prior to spawning. Extracts of the pigment cells induce a low level of germinal vesicle breakdown in asteroid oocytes. It is proposed that these cells are homologous to the follicle cells of other echinoderms and are involved in stimulating the maturation of oocytes.  相似文献

Immature starfish oocytes are surrounded by envelopes consisting of follicular cells. These cells adhere to each other and to the oocyte, immobilizing the latter within the ovary. When isolated oocytes in their follicles are treated with 1-methyladenine (1-MeAde), germinal vesicle breakdown (GVBD) and follicular envelope breakdown (FEBD) occur simultaneously. The 1-MeAde acts on the oocyte surface to produce a maturation-promoting factor (MPF) in the cytoplasm, which brings about GVBD. In the present study, MPF was found to induce FEBD as well as GVBD when injected into immature oocytes with their follicles in Asterina pectinifera. Although GVBD was induced by MPF in the presence of cytochalasin D, this drug prevented MPF-induced FEBD, and each follicular cell remained in situ on the surface of the oocyte. However, desmosomes connecting the processes of the follicle cell with the oocyte surface were disrupted following MPF injection even in the presence of cytochalasin D, and the processes became detached from the oocyte. FEBD occurred in these oocytes when cytochalasin D was removed, resulting in the formation of a small follicular clump by microfilament-mediated contraction of the follicle cells. These results show that FEBD is not brought about by the direct action of 1-MeAde but by the action of MPF. Therefore, in starfish, spawning as well as oocyte maturation is directly triggered by MPF produced under the influence of 1-MeAde.  相似文献

Surface characteristics of the bovine oocyte and its investments before, during, and after maturation, and fertilization in vitro were evaluated by scanning electron microscopy (SEM). Oocyte diameters were also measured during SEM analysis of the oocyte. The cumulus cells manifested a compact structure with minimal intercellular spaces among them in the immature oocytes. These became fully expanded with increased intercellular spaces after maturation in vitro, but contracted again after fertilization. The zona pellucida (ZP) showed a fibrous, open mesh-like structure in the maturing and matured oocytes. The size and number of meshes on the ZP decreased dramatically after fertilization. The vitelline surface of immature oocytes was characterized by distribution of tongue-shaped protrusions (TSPs) varying in density. After 10 and 22 hr of maturation incubation, oocyte surface microvilli (MV) increased to become the predominant surface structure, and TSPs decreased substantially. The vitelline surface of fertilized oocytes (at 6 and 20 hr) was similar to that of the matured oocytes, but unfertilized oocytes had less dense MV than did fertilized oocytes (at 20 hr). The diameter of the oocytes decreased from 99 to 80 μm during maturation and increased to 106 μm after insemination (P < 0.05). Membrane maturation was characterized by surface changes from a TSP-predominant pattern to a MV-predominant pattern. Thus, the bovine oocyte maturation process was found to involve the expansion of cumulus cells and the maturation of the ZP, which changes dramatically upon fertilization. The structure of the developing oocytes in the ovary of unfed and fed femaleArgas (Persicargas) arboreus is described as seen by scanning (SEM) and transmission (TEM) electron microscopy. The unfed female ovary contains small oocytes protruding onto the surface and its epithelium consists of interstitial cells, oogonia and young oocytes. Feeding initiates oocyte growth through the previtellogenic and vitellogenic phases of development. These phases can be observed by SEM in the same ovary.The surface of isolated, growing oocytes is covered by microvilli which closely contact the basal lamina investing the ovarian epithelium and contains a shallow, circular area with cytoplasmic projections and a deep pit, or micropyle, at the epithelium side. In more advanced oocytes the shell is deposited between microvilli and later completely covers the surface.Transmission EM of growing oocytes in the previtellogenic phase reveals nuclear and nucleolar activity in the emission of dense granules passing into the cytoplasm and the formation of surface microvilli. The cell cytoplasm is rich in free ribosomes and polysomes and contains several dictyosomes associated with dense vesicles and mitochondria which undergo morphogenic changes as growth proceeds. Membrane-limited multivesiculate bodies, probably originating from modified mitochondria, dictyosomes and ribosomal aggregates, are also observed. Rough endoplasmic reticulum is in the form of annulate lamellae. During vitellogenesis, proteinaceous yolk bodies are formed by both endogenous and exogenous sources. The former is involved in the formation of multivesicular bodies which become primary yolk bodies, whereas the latter process involves internalization from the haemolymph through micropinocytosis in pits, vesicles and reservoirs. These fuse with the primary yolk bodies forming large yolk spheres. Glycogen and lipid inclusions are found in the cytoplasm between the yolk spheres.  相似文献

We studied the ultrastructural organization of the ovarian follicles in a placentotrophic Andean lizard of the genus Mabuya. The oocyte of the primary follicle is surrounded by a single layer of follicle cells. During the previtellogenic stages, these cells become stratified and differentiated in three cell types: small, intermediate, and large globoid, non pyriform cells. Fluid‐filled spaces arise among follicular cells in late previtellogenic follicles and provide evidence of cell lysis. In vitellogenic follicles, the follicular cells constitute a monolayered granulosa with large lacunar spaces; the content of their cytoplasm is released to the perivitelline space where the zona pellucida is formed. The oolemma of younger oocytes presents incipient short projections; as the oocyte grows, these projections become organized in a microvillar surface. During vitellogenesis, cannaliculi develop from the base of the microvilli and internalize materials by endocytosis. In the juxtanuclear ooplasm of early previtellogenic follicles, the Balbiani's vitelline body is found as an aggregate of organelles and lipid droplets; this complex of organelles disperses in the ooplasm during oocyte growth. In late previtellogenesis, membranous organelles are especially abundant in the peripheral ooplasm, whereas abundant vesicles and granular material occur in the medullar ooplasm. The ooplasm of vitellogenic follicles shows a peripheral band constituted by abundant membranous organelles and numerous vesicular bodies, some of them with a small lipoprotein core. No organized yolk platelets, like in lecithotrophic reptiles, were observed. Toward the medullary ooplasm, electron‐lucent vesicles become larger in size containing remains of cytoplasmic material in dissolution. The results of this study demonstrate structural similarities between the follicles of this species and other Squamata; however, the ooplasm of the mature oocyte of Mabuya is morphologically similar to the ooplasm of mature oocytes of marsupials, suggesting an interesting evolutionary convergence related to the evolution of placentotrophy and of microlecithal eggs. During progesterone-induced nuclear maturation the oocytes of Bufo arenarum undergo a series of nuclear and cytoplasmic changes. The breakdown of heterocellular communications between the follicular cell projections and the oocyte microvilli, and the consequent enlargement of the perivitelline space, were observed at the animal pole. The more evident cytoplasmic feature during nuclear maturation comprised the gathering of glycogen granules in clusters, some phagocytosed by empty vesicles. With respect to the location of these vesicles, some were observed in close proximity to the oolemma and others were freely suspended in the perivitelline space, extruded from the oocyte. Other visible events were the disruption of the annulate lamellae, the formation of an elaborate cortical endoplasmic reticulum and the rearrangement of the cortical granules in a monolayer immediately beneath the oolemma together with aggregates of endoplasmic reticulum cisternae. Our results show that during nuclear maturation the nuclear oocyte changes include a flattening of the spherical oocyte nucleus, its migration towards the surface of the animal pole, the disappearance of the nucleoli and the dissolution of the nuclear envelope.  相似文献

Developing ovarian follicles of Bacillus rossius have been examined ultrastructurally in an attempt to understand how inception of vitel-logenesis is controlled. Early vitellogenic follicles are characterized by a thick cuboidal epithelium that is highly interlocked with the oocyte plasma membrane. Gap junctional contacts are present both at the follicle cell/oocyte interface and in between adjacent follicle cells. In addition, microvilli of follicle cells protrude deeply into the cortical ooplasm of these early vitellogenic oocytes. With the onset of vitellogenesis, wide intercellular spaces appear in the follicle cell epithelium and at the follicle cell/oocyte interface. Gap junctions become progressively reduced both on the follicle cell surface and on the oocyte plasma membrane. Microvilli from the two cell types no longer interlock. From a theoretical standpoint each of the two structural differentiations present at the follicle cell/oocyte interface—gap junctions and follicle cell microvilli—could potentially trigger inception of vitellogenesis. Gap junctions might permit the passage of a regulatory molecule, transferring from follicle cells to oocyte, which would control the assembly of coated pits on the oocyte plasma membrane. Alternatively cell interaction via microvilli might induce the appearance of coated pits, thus creating a membrane focus for vitellogenin receptors. Both possibilities are discussed in relation to current literature.  相似文献

A series of seasonal blood and ovarian tissue samples were taken from five female Atlantic cod Gadus morhua at different times between various batches of eggs and subsequently examined for oocyte size, plasma steroid hormones and ultrastructure of the cortex oocyte cytoplasm and follicle layer. This detailed protocol demonstrated that the spawning cod ovary is vitellogenically very active. Individual oocytes were able to develop at different rates, and at least two cohorts of oocytes with a different developmental status were present simultaneously. The concentration of estradiol-17β varied between batches and decreased over the season. The levels of 17,20β-P and 11-deoxycortisol were generally low. Oocytes of an advanced developing cohort showed numerous coated vesicles and closely apposed oocytic and follicular microvilli located in the chorionic pores. The number of oocytic microvilli was estimated at about 1–8 million cell−1, which increased the oocyte surface area by a factor of 12. Furthermore, the oocytic microvilli were found to stretch during final maturation. This stage-specific mobility of microvilli is discussed in relation to further incorporation of yolk and the relevant actin-based cytoskeleton observed for other classes of animals.  相似文献

Summary We have studied the differentiation of the vitelline coat (VC) of the ascidianCiona intestinalis. In the young previtellogenic oocyte the vitelline coat precursor material (VCPM) makes its first appearance as patches of fibrous material in close apposition to the outer surface of the oocyte. The presence of subcortical vescicles containing a fuzzy electron-dense material and their opening into the oocyte surface parallels the formation of VCPM. Numerous microvillar-like structures emerge from the oocyte surface. When the VCPM completely surrounds the oocyte the microvilli are withdrawn. An overall increase of VCPM parallels the growth of the oocyte. The next step in the differentiation of the vitelline coat consists in the packing of the constituent fibrils in a dense layer at its outer surface, i.e. the one in contact with the follicle cells. At this time the VC is penetrated by microvilli protruding both from the oocyte and follicle cells. The VC reaches its final structure and thickness at the time the test cells are extruded into the perivitelline space.The participation of the follicle cells in VC organization is also discussed.  相似文献

The golden hamster is an attractive model organism for studying reproductive physiology, oncology, genetics and virology. In an effort to establish experimental protocols necessary for cloning golden hamsters, we examined changes in the reciprocal position of the FPB (first polar body) and chromosome set of MII (the second meiotic metaphase) oocytes of golden hamsters. Oocytes were collected under three different conditions: (i) oocyte direct recovery from the oviduct of hormonally treated donor; (ii) oocyte recovery from the oviduct of hormonally treated donor followed by 5 h/10 h in vitro culture; and (iii) oocyte recovery from ovaries of hormonally treated donors and in vitro maturation. Then oocyte recovery was performed from the oviduct of hormonally treated donors, followed by 5 h in vitro culture with colchicine and/or CB (cytochalasin B). Denuded oocytes were stained with Hoechst 33342 and propidium iodide and evaluated under a microscope. Our results demonstrate that the change in FPB position in relation to the MII oocyte chromosome set increases with age of in vivo-matured oocytes. Cumulus cells can protect the FPB of in vitro-cultured oocytes from degeneration but do not significantly affect its repositioning, and in vitro-matured oocytes age slower. The colchicine has a stronger effect on cytoplasmic protrusions of golden hamster oocytes when compared with CB. These results define conditions for changes in FPB position relative to the MII oocyte chromosome set.
