The effect of various growth substances on regeneration and growth of bulblets on excised bulb scale segments of Hyacinthus orientalis L. cv. Pink Pearl was studied. Bulblet formation was promoted by the auxins IAA and IBA but was inhibited by NAA at high concentrations. IAA hardly affected bulblet weight, whereas IBA and NAA at high concentrations decreased it. Cytokinins had hardly any effect on bulblet regeneration and bulblet weight, Gibberellins inhibited both bulblet regeneration and bulblet weight; GA4+7 acted much stronger than GA3. ABA and ethephon decreased bulblet regeneration and bulblet growth.  相似文献

Experiments with Grand Rapids lettuce seeds (Lactuca sativa L.) maintained in darkness or irradiated with red light have shown that the inhibition of germination induced by low concentrations of ABA (2, 4, 6 μM) could be overcome by gibberellins (GA3 or GA4). The same results were obtained, although to a lesser extent, under the influence of two out of the four cytokinins tested (K and BAP) for seeds maintained in darkness. To suppress the block induced by higher concentrations of ABA (for example 8 μM), it was necessary to apply a cytokinin (K, BAP, Z or 2iP) and a gibberellin (GA4 or GA3) simultaneously, or a cytokinin following a red light treatment. Experiments conducted in darkness in which ABA (8 μM) was applied together with a cytokinin (BAP) and a gibberellin (GA4) showed that the gibberellin and the cytokinin played similar roles towards each other and towards ABA.  相似文献

The interrelation between the effects of abscisic acid (ABA) and the effects of cytokinins and gibberellic acid in retarding leaf senescence was investigated. Leaf discs from plants of Taraxacum megallorrhizon, Rumex pulcber and Tropaeolum majus were floated on solutions of cytokinin or GA to which given amounts of ABA were added. After five days, chlorophyll was extracted and the amount estimated spectrophoto-metrically. The interrelation between the effects of abscisic acid and cytokinins differed from that between the effects of ABA and gibberellic acid. Abscisic acid reduced the senescence retarding effect of GA more than that of cytokinins. A high concentration of cytokinins nullified the senescence enhancing effect of low concentrations of ABA. GA did not reverse the effects of ABA.  相似文献

The presence of IAA, ABA and gibberellins in extracts of shoots of Ribes nigrum was demonstrated by gas-liquid chroma-tography (GLC) for both IAA and ABA and by the lettuce hypocotyl assay for gibberellins. Quantitative estimation of the three substances in extracts from upper and lower halves of shoots which had been kept horizontal, and which showed negative geotropic response after 4 h, indicated a redistribution of hormones during the geotropic stimulation. The ratio of the hormones in lower and upper halves was 3.8:1 and 2.8:1 for IAA and giberellins respectively, whereas the ratio of ABA in upper and lower halves was 2.1:1. There is, however, no evidence for the participation of gibberellins and ABA in the early development of negative geotropic curvatures, since shoots of intact Vicia faba seedlings treated with 100 mg/1 solution of GA3, ABA and 10 mg/1 IAA for 30 min prior to geotropic stimulation, developed negative geotropic curvatures, although shoots pretreated with 50 and 100 mg/1 IAA did not develop curvature.  相似文献

The occurrence of gibberellins and abscisic acid (ABA) in extracts of roots of Vicia faba was demonstrated by gas-liquid chromatography (GLC) of the methylated eluates from the relevant zones of thin-layer chromatograms (TLC) of purified extracts. Quantitative determination of the hormone contents in extracts from upper and lower halves of roots which had been kept in the horizontal position for 30 min indicated a redistribution of the hormones during the geotropic stimulation. Gibberellins whose methyl esters appeared at the retention time of methylated gibberellic acid (GA3), used as a standard, occurred in higher concentration in the upper than in the lower halves (ratio 2.08:1), whereas the concentration of ABA was highest in the lower halves (ratio 3.08:1). The ratio of the hormones in right and left halves of vertical roots was close to 1:1. Indoleacetic acid (IAA) and ABA were found to retard the elongation of roots of Vicia faba and Lepidium during the first 24 h. Additional experiments with Lepidium showed that this retardation occurs within the first hour after application. Low concentrations of GA3, when applied to germinating seeds just after the radicles had broken the seed coat, stimulated root elongation in Vicia faba within 24 h and in Lepidium within 36 h. When applied to Lepidium seedlings with 20 mm long roots, GA3 showed a stimulatory trend within the first 2 h, and distinct stimulation in the subsequent hours, particularly at the lowest concentrations, 0.01 and 0.001 mg/1. These results suggest the possibility of a participation of ABA and gibberellins (in addition to IAA) in the development of the positive geotropic curvature.  相似文献

The cytoskeleton of plant and animal cells serves as a transmitter, transducer, and effector of cell signaling mechanisms. In plants, pathways for proliferation, differentiation, intracellular vesicular transport, cell-wall biosynthesis, symbiosis, secretion, and membrane recycling depend on the organization and dynamic properties of actin- and tubulin-based structures that are either associated with the plasma membrane or traverse the cytoplasm. Recently, a new in vivo cytoskeletal assay (cell optical displacement assay) was introduced to measure the tension within subdomains (cortical, transvacuolar, and perinuclear) of the actin network in living plant cells. Cell optical displacement assay measurements within soybean (Glycine max [L.]) root cells previously demonstrated that lipophilic signals, e.g. linoleic acid and arachidonic acid or changes in cytoplasmic pH gradients, could induce significant reductions in the tension within the actin network of transvacuolar strands. In contrast, enhancement of cytoplasmic free Ca2+ resulted in an increase in tension. In the present communication we have used these measurements to show that a similar antipodal pattern of activity exists for auxins and cytokinins (in their ability to modify the tension within the actin network of plant cells). It is suggested that these growth substances exert their effect on the cytoskeleton through the activation of signaling cascades, which result in the production of lipophilic and ionic second messengers, both of which have been demonstrated to directly effect the tension within the actin network of soybean root cells.  相似文献

The banana (Musa spp. AAA) micropropagation shows a high incidence of off-types, among whose variegated plants are very common. Endogenous levels of growth regulators and pigment content were measured in normal and variegated leaves of the micropropagated banana plants growing in a greenhouse. Growth regulators were separated by high pressure liquid chromatography and submitted to enzyme-linked immunosorbent assay for quantification. Pigment content was measured using the colorimetric method. Green leaves contained 1.9 and 10 times more cytokinins compared with green and yellow sectors of variegated leaves, respectively. The levels of indoleacetic acid in normal leaves were significantly higher than those found in green and yellow sectors of variegated leaves; however, the levels of abscisic acid were lower in normal leaves. The lower content of chlorophylls in variegated leaves coincided with decreased endogenous levels of cytokinins, which indicated that variegation in banana leaves may be associated with alterations in the metabolism of this growth regulator. Carbon allocation within grapevines may affect berry growth and development. The plant hormones gibberellins (GAs) and abscisic acid (ABA) control various processes across the plant life and both have been involved in assimilate production and transport in different species. Hence, this work examined the distribution of sugars (sucrose, fructose, and glucose) and starch in grapevines at veraison after foliar applications of GA3, ABA, and an inhibitor of GA biosynthesis, paclobutrazol (PBZ). The results demonstrated that GA3 increased total grapevine mass, with carbon allocated to the whole grapevine (as structural and soluble carbohydrates). Both GA3 and ABA increased monosaccharide (glucose and fructose) levels in berries (up to tenfold) and roots (up to threefold). However, GA3 increased the net carbon fixation whereas ABA did not. PBZ diminished most growth parameters except grapevine mass, and allocated more carbohydrates to roots (up to threefold more sucrose and starch). Such results indicate that GAs promote net carbon fixation and transport, whereas ABA as a stress signal only enhances sugar transport; notwithstanding the two hormones promoted carbon allocation toward roots and berries.  相似文献

Fragments of maize leaves were incubated at controlled temperatureand irradiance either on distilled water or on one of threeconcentrations of cytokinin (10–1, 10–2 and 10–3mol m–3). The effects of zeatin or kinetin on stomatalaperture were determined by stripping abaxial epidermis fromthe fragments after incubation and immediately measuring stomatalapertures under the microscope. At each cytokinin concentrationleaf pieces were incubated at 5 or 350 µmol mol–1CO2 with or without ABA (10–1 mol m–3). At 5.0 µmolmol–1 CO2 increasing the concentrations of zeatin hada negligible effect upon stomatal aperture. When air containing350 umol mol–1 CO2 was bubbled through the incubationsolutions, apertures of stomata incubated on water were morethan halved. Increasing cytokinin concentrations reduced theeffect of CO2 on stomata and incubation on 10–1 mol m–3zeatin completely removed any CO2 response. Four gibberelJins (GAs), GA19, GA20, GA24, and GA53, were identified by gas chromatography-mass spectrometry (GC-MS) from the dormant bulbils of Dioscorea japonica Thunb. ex Murray (Japanese yam), suggesting that two biosynthetic pathways of GAs, early 13- and non-13-hydroxylation pathways, are operating in the bulbils. Seed dormancy is an important economic trait for agricultural production. Abscisic acid (ABA) and Gibberellins (GA) are the primary factors that regulate the transition from dormancy to germination, and they regulate this process antagonistically. The detailed regulatory mechanism involving crosstalk between ABA and GA, which underlies seed dormancy, requires further elucidation. Here, we report that ABI4 positively regulates primary seed dormancy, while negatively regulating cotyledon greening, by mediating the biogenesis of ABA and GA. Seeds of the Arabidopsis abi4 mutant that were subjected to short-term storage (one or two weeks) germinated significantly more quickly than Wild-Type (WT), and abi4 cotyledons greened markedly more quickly than WT, while the rates of germination and greening were comparable when the seeds were subjected to longer-term storage (six months). The ABA content of dry abi4 seeds was remarkably lower than that of WT, but the amounts were comparable after stratification. Consistently, the GA level of abi4 seeds was increased compared to WT. Further analysis showed that abi4 was resistant to treatment with paclobutrazol (PAC), a GA biosynthesis inhibitor, during germination, while OE-ABI4 was sensitive to PAC, and exogenous GA rescued the delayed germination phenotype of OE-ABI4. Analysis by qRT-PCR showed that the expression of genes involved in ABA and GA metabolism in dry and germinating seeds corresponded to hormonal measurements. Moreover, chromatin immunoprecipitation qPCR (ChIP-qPCR) and transient expression analysis showed that ABI4 repressed CYP707A1 and CYP707A2 expression by directly binding to those promoters, and the ABI4 binding elements are essential for this repression. Accordingly, further genetic analysis showed that abi4 recovered the delayed germination phenotype of cyp707a1 and cyp707a2 and further, rescued the non-germinating phenotype of ga1-t. Taken together, this study suggests that ABI4 is a key factor that regulates primary seed dormancy by mediating the balance between ABA and GA biogenesis.  相似文献

Recently there have been breakthroughs on a number of fronts in abscisic acid (ABA) biology research that have advanced the field significantly, including discovery of genes involved in ABA metabolism, along with progress in understanding of ABA signaling (Finkelstein and others 2002; Kushiro and others 2004; Lim and others 2005; Saito and others 2004). At the same time, the chemistry of ABA has advanced. New analytical methods have been developed for profiling ABA and catabolites (Ross and others 2004; Zaharia and others 2005). Novel bioactive catabolites have been discovered from feeding studies with deuterated ABA and catabolites (Zaharia and others 2004; Zhou and others 2004). This review covers recent advances and prospects in natural products chemistry, analysis of ABA catabolism, and applications of ABA analogs for biochemical studies and horticultural uses.  相似文献

Effects of benzyladenine (BA) and abscisic acid (ABA) applied separately or simultaneously on parameters of gas exchange of Phaseolus vulgaris L. leaves were studied. In the first two experimental sets) 100 M ABA and 10 M BA were applied to plants sufficiently supplied with water. Spraying of leaves with ABA decreased stomatal conductance (g s) and in consequence transpiration rate (E) and net photosynthetic rate (P N) already 1 h after application, but 24 h after application the effect almost disappeared. 10 M BA slightly decreased gas exchange parameters, but in simultaneous application with ABA reversed the effect of ABA. Immersion of roots into the same solutions markedly decreased gas exchange parameters and 24 h after ABA application the stomata were completely closed. The effect of ABA was ameliorated by simultaneous BA application, particularly after 1-h treatment. In the third experimental set, plants were pre-treated by immersing roots into water, 1 M BA, or 100 M ABA for 24 h and then the halves of split root system were dipped into different combinations of 1 M BA, 100 M ABA, and water. In plants pre-treated with ABA all gas exchange parameters were small and they did not differ in plants treated with H2O+H2O, H2O+BA, or BA+BA. In plants pre-treated with BA or H2O, markedly lower values of P N were found when both halves of roots were immersed in ABA. Further, the effects of pre-treatment of plants with water, 1 M BA, 100 M ABA, or ABA+BA on the development of water stress induced by cessation of watering and on the recovery after rehydration were followed. ABA markedly decreased gas exchange parameters at the beginning of the experiment, but in its later phase the effect was compensated by delay in development of water stress. BA also delayed development of water stress and increased P N in water-stressed leaves. BA reversed the effect of ABA at mild water stress. Positive effects of BA and ABA pre-treatments were observed also after rehydration.  相似文献

The biomass structure and the contents of (ABA) and cytokinins (zeatin plus zeatin riboside) were studied in flowering plants of ten herbaceous species of the Middle Urals representing stress-tolerant (S) and ruderal (R) ecological strategies. The plants, which explicitly manifested S- and R-strategies, differed in ABA and cytokinin concentrations and phytohormone partitioning in particular organs. Higher ABA accumulation in the leaves and especially in the reproductive organs (here the ABA concentration exceeded by several times that of cytokinins) was characteristic of the R-plants as compared to the S-plants. In addition, the R-plants manifested higher cytokinin contents in the roots, whereas, in the S-plants, cytokinins dominated in the leaves and the reproductive organs (in the latter, the content of cytokinins exceeded that of ABA). The diverse hormonal status of the R- and S-species is discussed in relation to the patterns of plant growth and development and the photosynthate transport and partitioning characteristic of two ecological strategies. The authors conclude that plant ability to maintain the typical hormonal balance is instrumental in acquiring specific ecological strategies.  相似文献

Cytokinins in apices of eight isogenic lines of Mercurialis annua were compared (high performance liquid chromatography-gas chromatography mass spectroscopy-computer system). These apices develop normal staminate or pistillate differentiation processes (sex series lines) or empty (sterile), semiempty (semisterile), and full anthers (restored fertile male) in the sterility series in which a pistillate line was constructed. Both series developed two different cytokinin pathways: trans-cytokinins characterized the sex series, whereas the cis pathway characterized the sterility series. Drastic changes in the trans pathway (0/250 nanograms trans-zeatin and 166/0 nanograms zeatin nucleotide) induced staminate/pistillate differentiations. Less drastic quantitative changes in the cis pathway induced sterility or restored fertility compared to normal fertile anthers (192 or 669 nanograms/traces). The action of the complete cis-pathway was morphologically effective in the sterility series when the ratio of cis to trans pathways was 1:2 or 1:1 instead of 1:3. A final diagram shows the action of each sex or sterility allele on the enzymes controlling specific metabolites in both pathways. The discussion provides insights on the regulation of cytokinin-auxin balances specific for each kind of reproductive differentiation.  相似文献
