The possible involvement of IAA in the effect thatAzospirillum brasilense has on the elongation and morphology ofPanicum miliaceum roots was examined by comparing in a Petri dish system the effects of inoculation with a wild strain (Cd) with those of an IAA-overproducing mutant (FT-326). Both bacterial strains produced IAA in culture in the absence of tryptophan. At the stationary growth phase, production of IAA by FT-326 wasca. 12 times greater than that of Cd. When inoculation was made with bacterial concentrations higher than, 106 colony forming units ml–1 (CFU ml–1), both strains inhibited root elongation to the same extent. At lower concentrations Cd enhanced elongation, by 15–20%, while FT-326 was ineffective. Both strains promoted root-hair development, and root-hairs were produced nearer the root tip the higher the bacterial concentration (e. g. root elongation region was reduced). Effects of FT-326 on root-hair development were greater than those of Cd. Acidified ether extracts of Cd and FT-326 cultures had inhibitory or promoting effects on root elongation depending on the dilution applied. At low dilutions, extracts from FT-326 were more inhibitory for elongation than those from Cd. At higher dilutions root elongation was promoted, but FT-326 extracts had to be more diluted than those from Cd. Dilutions that promoted root elongation contained supra-optimal concentrations of IAA, 1–3 orders of magnitude higher than those required for optimal enhancement by synthetic IAA. It is suggested that the bacteria produce in culture an IAA-antagonist or growth inhibitor that decreases the effectiveness of IAA action. The large variability reported for the effects ofAzospirillum on root elongation could be the result of the opposite effects on root elongation of IAA and other compounds, produced by the bacteria.  相似文献

There is strong evidence that non-leguminous field crops sometimes benefit from associations with diazotrophs. Significantly, the potential benefit from N2 fixation is usually gained from spontaneous associations that can rarely be managed as part of agricultural practice. Particularly for dryland systems, these associations appear to be very unreliable as a means of raising the nitrogen status of plants. However, recent technical advances involving the induction of nodular structures on the roots of cereal crops, such as wheat and rice, offer the prospect that dependable symbioses with free-living diazotrophs, such as the azospirilla, or with rhizobia may eventually be achieved.  相似文献

为探究Zn、Cu与盐复合胁迫对翅碱蓬(Suaeda salsa)萌发生长的影响机理及其调控措施,以翅碱蓬为研究对象,采用水培试验方法,测定Zn、Cu和盐复合胁迫条件下翅碱蓬种子发芽率、萌发速率和幼苗渗透调节物质含量等指标,分析1.5 mg/L吲哚乙酸(IAA)、100 mg/L赤霉素(GA)和0.3%硝酸钾(KNO3)处理对Zn、Cu与盐复合胁迫条件下翅碱蓬萌发与生长的影响。结果表明:(1) Zn、Cu与高盐复合胁迫极显著降低翅碱蓬种子的发芽率,中高浓度的Zn、Cu与盐复合胁迫极显著降低翅碱蓬种子的萌发速率,Zn、Cu和盐复合胁迫对翅碱蓬种子的萌发生长影响表现为低促高抑效应,影响因子间存在明显的协同效应;(2)Zn、Cu和盐复合胁迫条件下,随着盐浓度的升高,翅碱蓬幼苗体内过氧化氢酶(CAT)、过氧化物酶(POD)和超氧化物歧化酶(SOD)3种抗氧化酶活性呈先升高后降低的变化趋势,Zn、Cu和高盐复合胁迫使翅碱蓬幼苗体内丙二醛(MDA)含量增加近2.5倍;(3)1.5 mg/L IAA溶液浸种12 h可显著提高Zn、Cu与高盐复合胁迫条件下翅碱蓬种子的发芽率和萌发速...  相似文献

The torosa-2 tomato mutant is characterized by a strong inhibition of release of axillary shoots, that is not under the control of the main apex and IAA. Microscopic examination indicated that about 70% of leaf axils do not have axillary buds. Of the growth regulators tested, gibberellic acid and cytokinins were able to modify the to-2 phenotype: increasing bud number (GA3 treated) and developing shoots (both substances). Sequential application of growth regulators demonstrated that bud production was only affected by treatments given between sowing time and 32 days after germination. Grafting experiments indicated that endogenous root factors have no essential role in the lateral branching of the genotypes investigated. The control of axillary bud differentiation and the branching pattern in the to-2 appears to be dependent of a complex mechanism involving gibberellins and cytokinins.  相似文献

The transport of radiolabeled indoleacetic acid (IAA), and some of its conjugates, was investigated in nodal stem segments of Phaseolus vulgaris L. Donor agar blocks containing either [2-acetyl-14C]-IAA; [2-acetyl-14C]-indole-3-acetyl-L-aspartate (IAAsp); [2-acetyl-14C]-indole-3-acetyl-L-glycine (IAGly); or [2-acetyl-14C]-indole-3-acetyl-L-alanine (IAAla) were placed on either the apical or basal cut surface of stem segments each bearing an axillary bud at the midline. In some experiments, a receiver block was placed on the end opposite to the donor. After transport was terminated, the segments were divided into five equal sections plus the bud, and the radioactivity of donors, receivers and each part of the stem segment was counted.For all four substances tested, the amount of 14C transported to the axillary bud from the base was the same or greater than that from the apical end. After basipetal transport, the distribution of 14C in the segment declined sharply from apex to base. The inverse was true for acropetal transport. Transport for the three IAA conjugates did not differ substantially from each other.The IAA transport inhibitor, N-1-naphthylphthalamic acid (NPA), inhibited basipetal 14C-IAA transport to the base of the stem segment but did not alter substantially the amount of 14C-IAA recovered from the bud. Transport of 14C-IAA from the apical end to all parts of the stem segment declined when the base of the section was treated with nonradioactive IAA. Taken together with data presented in the accompanying article [Tamas et al. (1989) Plant Growth Regul 8: 165–183], these results suggest that the transport of IAA plays a role in axillary bud growth regulation, but its effect does not depend on the accumulation of IAA in the axillary bud itself.  相似文献

The effect of two Azospirillum strains (SP-7, Dol) was compared on root proton efflux and root enlargement of three wheat cultivars (Ghods, Omid and Roshan). Root colonization varied greatly among strain–plant combinations. Inoculation enhanced proton efflux and root elongation of wheat roots but this effect was directly dependent on the strain–plant combination. Strain SP-7 stimulated the greatest proton efflux and root elongation in cv. Roshan, whereas strain Dol induced the best effect on both these phenomena in cv. Ghods. Based on positive correlation between these two phenomena, it was suggests that proton efflux is related to increasing of root length by Azospirillum inoculation. The number of bacteria of both Azospirillum strains in root of cv. Omid was less than the other cultivars. Proton extrusion and root elongation of cv. Omid failed to respond significantly with these two strains. This may be due to incompatible host-strain combination. Thus compatible strains are necessary for increasing of proton efflux and root extension in wheat cultivars.  相似文献

Recent microscopic evidence acquired using strain-specific monoclonal antibodies and specific gene probes confirms earlier claims that some strains of Azospirillum lipoferum and A. brasilense, but not others, are capable of infecting the interior of wheat roots. The present study was performed to determine whether this strain specificity in the infection of the interior of wheat roots was apparent in the first 24 h of adsorption (`anchoring') of Azospirillum cells to the root surface. Strains of A. brasilense, originally isolated from surface-sterilised wheat roots (Sp 245, Sp 107) or with a proven ability to infect the interior of wheat roots (Sp 245), showed no greater ability to anchor to the roots than other Azospirillum strains isolated from the wheat rhizosphere (Sp 246) or from the rhizosphere or rhizosphere soil of other gramineae (Sp 7, Cd, S 82). The SEM images showed that at the root tip the Azospirillum cells were principally located in cracks between epidermal cells. In the root hair zone the bacteria were more numerous but again principally located in the depressions between epidermal cells. In all zones of the roots mucilage was present, and near the tip this appeared to have been partially digested, forming `halos' around the bacteria and revealing fibril-like strands attached to the bacteria. Subsequent studies were conducted using a technique originally developed for investigating competition of rhizobia for adsorption sites on legume roots. In the adaptation of this technique it was found that the presence of any significant concentration of Ca++ in the incubation medium reduced bacterial adsorption, as did concentrations of (PO4)3- above 50 mM. The influence of the pH of the incubation medium on the adsorption of ten different strains of Azospirillum showed, that with one exception, strains isolated from the roots or rhizosphere of wheat showed optimum adsorption at pH 6.0, and all other strains pH 7.0. Apart from this effect of pH no differences in adsorption were detected between strains with a proven capacity to infect wheat roots and those unable to do so. However, strains varied in their capability to compete for adsorption sites, there being a tendency for strains with a proven capacity to invade the internal tissues of wheat roots to be more competitive for adsorption sites.  相似文献

The tumour-inducing T-DNA gene 4 (T-cyt gene) of the nopaline Ti plasmid pTiC58 was cloned and introduced into tobacco cells by leaf disc transformation using Agrobacterium plasmid vectors. Tobacco shoots exposed to elevated cytokinin levels were unable to develop roots and lacked apical dominance. Using exogenously applied phytohormone manipulations we were able to regenerate morphologically normal transgenic tobacco plants which differed in endogenous cytokinin levels from normal untransformed plants. Although T-cyt gene mRNA levels, as revealed by dot-blot hybridization data, in these rooting plants were only about half those in primary transformed shoots the total amount of cytokinins was much lower than in crown gall tissue or cytokinin-type transformed shoots as reported by others. Nevertheless the cytokinin content in T-cyt plants was about 3 times greater than in control tobacco plants.Elevated cytokinin levels have been shown to change the expression of several plant genes, including some nuclear genes encoding chloroplast proteins. Our results show that the mRNA levels of chloroplast rbcL gene increase in cytokinin-type transgenic tobacco plants as compared with untransformed plants. Data obtained suggest that T-cyt transgenic plants are a good model for studying plant gene activity in different parts of the plant under endogenous cytokinin stress.  相似文献

The effect of cellulase and pectinase on bacterial colonization of wheat was studied by three different experiments. In the first experiment, the root colonization of 3 wheat cultivars (Ghods, Roshan and Omid) by two A. brasilense strains (Sp7 and Dol) was compared using pre-treated roots with cellulase and pectinase, and non-treated with these enzymes (control). Although the root colonization varied greatly among strain-plant combinations in controls, the pre-treatment of roots with polysaccharide degrading enzymes significantly increased the bacterial count in roots, regardless of the strain-plant combination. This might be an indication that cell wall may act as an important factor in plant-Azospirillum interaction. In the second experiment, the root cellulase activity of the same wheat cultivars treated with and without the two Azospirillum brasilense, strains (Sp7 and Dol) was compared. The pre-treatment of wheat roots with Azospirillum enhanced the cellulase activity of wheat root extracts. Thus, the cellulase activity might participate in the initial colonization of wheat roots by Azospirillum. The comparison of the cellulase activity of root extracts within inoculated and non-inoculated seedlings showed that the inoculation had enhanced the cellulase activity in root extracts, but this effect was directly dependent on the strain-plant combination. Strain Sp7 stimulated the highest cellulase activity in cv. Roshan, but strain Dol induced the highest enzyme activity in cv. Ghods. In the third experiment, several growth parameters of those 3 wheat cultivars treated with and without those two bacterial strains (Sp7 and Dol) were compared. The highest magnitude of growth responses caused by Sp7 strain was in the cv Roshan, but Dol strain stimulated the highest growth in cv Ghods. Therefore, effective colonization may contribute to more growth responses.  相似文献

Azosprilla were collected in wheat fields from subtropical and temperate soils of central Nepal at various elevations. Different wheat cultivars responded positively and significantly in grain yield, grain N-yield, and total N-yield in plant shoots to the inoculation with Nepalese isolate Azospirillum 10SW. Nepalese wheat cv. Seto responded significantly better with Azospirillum 10SW than with the Brasilian isolate A. lipoferum Sp 108 st, a strain which was found highly efficient in earlier experiments with German wheat cultivars, especially cv. Turbo. Yield of Turbo was increased by inoculations of both Azospirillum strains too, but it showed no significant differences depending from the inoculum used. The higher efficacy of combining Azospirillum 10SW and Seto, both collected from the same locality, indicates the possibility of improved associations using traditional cultivars and local bacteria. ei]{gnR O D}{fnDixon}  相似文献

Immature zygotic embryos of two wheat (Triticum aestivum L.) genotypes, known for their different ability to generate embryogenic callus, were used as initial explants to establish callus cultures. Embryogenic and non-embryogenic calluses were obtained from the competent genotype (`Combi'), while only non-embryogenic callus was produced by the incompetent one (`Devon'). The morphogenetic competence of each callus type was evaluated by transferring some segments to regeneration conditions. The endogenous hormone concentrations (free indole-3-acetic acid [IAA], abscisic acid [ABA], gibberellins 1, 3 and 20 [GAs], zeatin/zeatin riboside [Z/ZR] and N 6[2-isopentenyl] adenine/ N 6[2-isopentenyl] adenosine; [iP/iPA]) of the initial explants were determined by means of radio-immunoassay and showed that the only difference was the higher concentration of ABA found in the embryos of the most competent genotype; whose embryos showed a reduced rate of precocious germination. When analysing the endogenous hormone concentrations in the various callus types generated in each genotype, it was found that only differences in the free IAA concentrations were associated with variations in the morphogenic properties of the calluses. Higher concentrations of endogenous free IAA were typical of embryogenic callus cultures. It was also observed that a loss in the embryogenic competence of the calluses, due to a prolonged time of culture, occurred concomitantly with a reduction in free IAA concentrations, practically to the concentrations found in the non-embryogenic calluses.  相似文献

Treatment of wheat seedlings with the synthetic auxin, 2,4-dichlorophenoxyacetic acid (2,4-d), induced nodule-like structures or tumours (termed para-nodules) where lateral roots would normally emerge. The formation of these structures promoted increased rates of acetylene reduction at reduced oxygen pressure (0.02–0.04 atm) in seedling inoculated with Azospirillum brasilense, compared to seedlings inoculated without auxin treatment. Fluorescent microscopy, laser scanning confocal microscopy and direct bacterial counts all showed that the 2,4-d treatment stimulated internal colonization of the root system with azospirilla, particularly in the basal region of the nodular structures. Both colonization with azospirilla and acetylene-reducing activity were further stimulated by simultaneous treatment with another synthetic auxin, naphthaleneacetic acid (NAA) and, less reliably, with indoleacetic acid (IAA) and indolebutyric acid (IBA). These auxins produced shortening of many initiated lateral roots, although 20 times the concentration of NAA was required to achieve rounded structures similar to those obtained with 2,4-d. Treatment with NAA, IAA or IBA alone also stimulated colonization with azospirilla and acetylene reduction rates at 0.02 atm oxygen, but less effectively than by treatment with 2,4-d. Such exogenous treatments of wheat seedlings with synthetic growth regulators provide an effective laboratory model for studies on the development of a N2-fixing system in cereals.  相似文献

Influences of plant hormones on photoperiodic flowering in Pharbitis nil, var. Violet was re-evaluated by assaying them with a newly developed perfusion technique which can directly treat mesophyll cells with sample solution. Gibberellin A3 promoted the flowering response and indole-3-acetic acid, trans-zeatin and abscisic acid inhibited it when they were perfused immediately before an inductive dark treatment. The promotion or inhibition of flowering was not or hardly observed when solutions containing these plant hormones were applied by the dropping method to surface of cotyledons or plumules of the assay plants. The detection of clear flower-promoting and -inhibiting effects of the plant hormones may be due to the improved efficiency of incorporation of applied substances into plant tissue in the perfusion technique.  相似文献

Summary Four field experiments were carried out with wheat or sorghum in different regions of Brazil. The aim was to study the establishment of inoculatedAzospirillum strains, marked with resistance to various antibiotics, in the rhizosphere and in roots. The levels of the various antibiotics were chosen according to the resistance of the indigenousAzospirillum population.Azospirillum brasilense strains Sp 107 and Sp 245 could be established in all three wheat experiments and predominated within theAzospirillum population in washed, and especially in surface sterilized, roots. Strains Sp 7 and Cd established poorly in wheat roots.Azospirillum lipoferum Sp S82 represented 72% of the root isolates from sorghum inoculated with this strain. This strain and naturalAzospirillum infection became concentrated in the upper parts of the root system. Improved methods for root surface sterilization in which the absence ofAzospirillum on the root surface was established by pre-incubating roots with paraffin-capped ends in NFb medium confirmed the establishment of inoculatedAzospirillum strains within sorghum roots in the field.  相似文献

Azospirillum-plant association is accompanied by biochemical changes in roots which, in turn, promote plant-growth and tolerance to water stress. To shed light on the possible factors underlying these effects, roots from Azospirillum brasilense Sp245-inoculated Triticum aestivum seedlings growing in darkness under osmotic stress were analyzed for phospholipid (PL) composition, fatty acid (FA) distribution profiles and degree of unsaturation of the major PL classes. Azospirillum inoculation diminished ion leakage and increased 2,3,5-tripheniltetrazolium reducing ability in roots of well irrigated and water-stressed wheat seedlings. Total root PL content remained unaltered in all treatments. Six PL classes were detected, phosphatidylcholine (PC) and phosphatidylethanolamine (PE) comprising over 80% of the total. While water stress increased PC content and diminished that of PE, none of these changes were observed either under Azospirillum inoculation alone or when both treatments were combined. The major FAs found in both PC and PE were 16:0, 18:0, 18:1, 18:2, and 18:3. Higher PC and lower PE unsaturation than in well irrigated controls were observed in roots from Azospirillum-inoculated, water-stressed seedlings. Azospirillum inoculation could contribute to protect wheat seedlings from water stress through changes in the FA distribution profiles of PC and PE major root phospholipids.  相似文献

The effects of 6-benzyladenine (BA), its 3-glucoside (BA-3-G) and 9-glucoside (BA-9-G), and its riboside (BA-R) on the rooting of primary leaf explants of Phaseolus vulgaris L. were compared, alone or in combination with indole-3-butyric acid (IBA). At all except the lowest concentration used (4.4 × 10–7 M), BA and BA-3-G reduced the number of roots and their averge length relative to a distilled water control. Addition of IBA (4.4 × 10–5 M) increased rooting beyond the control, but not to the level of IBA alone. This contrasted with BA-9-G where a response very similar to that of the control was recorded. The riboside of BA was more inhibitory than the free base, its effect extending to the lowest concentration used (4.4 × 10–7 M).  相似文献

Summary The effect of seed inoculation withAzotobacter spp. orAzospirillum spp., and garbage amendment (0.5%), on the growth of wheat was studied in a field experiment under sub-tropical conditions. Two levels of N fertilizer were applied, the usual field rate (150 kg N ha–1) and half this amount. Tillering of plants, dry matter contents and nitrogenase activity were determined 30, 60 and 90 days after sowing. At the end of the experimental period, spore numbers and percentage of mycorrhizal infection were observed in the rhizosphere and root systems of plants. Straw and grain yields were also determined. The results of this study showed that seed inoculation and/or organic amendment stimulated plant growth, nitrogenase activity and mycorrhizal infection. This was more noticeable withAzotobacter than withAzospirillum. Inoculation withAzotobacter together with 1/2 N dose and organic amendment was the most effective application (19.75 and 10.70 t ha–1 were recorded for straw and grain yield, respectively).  相似文献

Summary In this study, auxin (indole-3-acetic acid), gibberellin, cytokinin (zeatin) and abscisic acid production were investigated in the culture medium of the bacteria Proteus mirabilis, P. vulgaris, Klebsiella pneumoniae, Bacillus megaterium, B. cereus, Escherichia coli. To determine the levels of these plant growth regulators, high performance liquid chromatography (HPLC) technique was used. Our findings show that the bacteria used in this study synthesized the plant growth regulators, auxin, gibberellin, cytokinin and abscisic acid.  相似文献

An in vitro propagation system was developed for Echinacea purpureaL. (purple coneflower), a medicinal plant commonly used in the treatment of colds, flu and related ailments. Root organogenesis from Echinacea purpurea hypocotyl explants was effectively induced by indolebutyric acid. Indoleacetic acid was found to be less effective than indolebutyric acid while treatments with naphthaleneacetic acid were ineffective for induction of root organogenesis. The results of this study have established a micropropagation system for Echinacea purpurea that will provide axenic plant material for further investigations into medicinally active biochemicals and the mass production of high-quality Echinacea purpurea root tissues for the commercial market. The hormonal control of axillary bud growth was investigated in cultured stem segments of Phaseolus vulgaris L. When the stem explants were excised and implanted with their apical end in a solid nutrient medium, outgrowth of the axillary buds-located at the midline of the segment-was induced. However, if indoleacetic acid (IAA) or naphthaleneacetic acid (NAA) was included in the medium, bud growth was inhibited. The exposure of the apical end to IAA also caused bud abscission and prevented the appearance of new lateral buds.In contrast to apically inserted segments, those implanted in the control medium with their basal end showed much less bud growth. In these segments, the auxin added to the medium either had no effect or caused a slight stimulation of bud growth.The IAA transport inhibitor N-1-naphthylphthalamic acid (NPA) relieved bud growth inhibition by IAA. This suggests that the effect of IAA applied at the apical end requires the transport of IAA itself rather than a second factor. With the apical end of the segment inserted into the IAA-containing medium, simultaneous basal application of IAA relieved to some extent the inhibitory effect of the apical IAA treatment. These results, together with data presented in a related article [Lim R and Tamas I (1989) Plant Growth Regul 8: 151–164], show that the polarity of IAA transport is a critical factor in the control of axillary bud growth.Of the IAA conjugates tested for their effect on axillary bud growth, indoleacetyl alanine, indoleacetic acid ethyl ester, indoleacetyl-myo-inositol and indoleacetyl glucopyranose were strongly inhibitory when they were applied to the apical end of the stem explants. There was a modest reduction of growth by indoleacetyl glycine and indoleacetyl phenylalanine. Indoleacetyl aspartic acid and indoleglyoxylic acid had no effect.In addition to IAA and its conjugates, a number of other plant growth substances also affected axillary bud growth when applied to the apical end of stem segments. Myo-inositol caused some increase in the rate of growth, but it slightly enhanced the inhibitory effect of IAA when the two substances were added together. Gibberellic acid (GA3) caused some stimulation of bud growth when the explants were from younger, rather than older plants. The presence of abscisic acid (ABA) in the medium had no effect on axillary bud growth. Both kinetin and zeatin caused some inhibition of axillary buds from younger plants but had the opposite effect on buds from older ones. Kinetin also enhanced the inhibitory effect of IAA when the two were applied together.In conclusion, axillary buds of cultured stem segments showed great sensitivity to auxins and certain other substances. Their growth responded to polarity effects and the interaction among different substances. Therefore, the use of cultured stem segments seems to offer a convenient, sensitive and versatile test system for the study of axillary bud growth regulation.  相似文献
