Responses of leaf gas exchange, fluorescence emission, chlorophyll concentration, and morpho-anatomical features to changes in photosynthetic photon flux density (PPFD) were studied in three wild ornamental species of Passiflora L. to select sun and shade species for landscaping projects. Artificial shade was obtained with different shading nylon nets, under field conditions, which allowed the reduction of 25, 50, and 75% of global radiation, along with a control treatment under full sunlight. For Passiflora morifolia the highest mean values of light-saturated net photosynthetic rate (P Nmax) and light compensation point (LCP) were observed at 50 and 25% shade, respectively, while the highest values of dark respiration rate (R D) and apparent quantum yield (α) were observed at 75% shade. For Passiflora suberosa litoralis the highest value of P max was observed at full sunlight. The highest mean values for P max, R D, and LCP for Passiflora palmeri var. sublanceolata were obtained at 25% shade. The highest values of net photosynthetic rate (P N) for P. morifolia, P. palmeri var. sublanceolata, and P. suberosa litoralis were 21.09, 16.15, and 12.36 μmol(CO2) m−2 s−1, observed at 50 and 75% shade and full sunlight, respectively. The values of the minimal chlorophyll fluorescence (F0) were significantly different in P. suberosa litoralis and P. palmeri var. sublanceolata, increasing with the increase of the irradiance. In contrast, the values of maximum photochemical efficiency of PSII (Fv/Fm) were significantly different only in P. suberosa litoralis, being higher at 75%, progressively reducing with the increase of PPFD levels. The total concentration of chlorophyll (Chl) was higher in shaded plants than in the ones cultivated in full sunlight. On the other hand, the values of Chl a/b ratio were reduced in shaded plants. A significant effect of shade levels on leaf area (LA) and specific leaf area (SLA) was found for the three species, whose highest mean values were observed at 75% shade. The thickness of foliar tissues was significantly higher for the three species at full sunlight and 25% shade. These results suggested that P. morifolia and P. palmeri var. sublanceolata appeared to be adapted to moderate shade conditions. P. suberosa litoralis presented higher plasticity to greater variation of the irradiance levels, while the photoinhibition was one of the limiting factors for this species at full sunlight.  相似文献

Panicum coloratum var. makarikariense is a perennial C4 grass native to South Africa with relatively good forage production under limited‐resource conditions. Genetic characterisation and breeding efforts have been scant, thus limiting its use in cattle raising systems. The goal of the present study was to assess the genetic diversity of a collection of P. coloratum var. makarikariense using agro‐morphological traits and molecular markers, in comparison with one accession of var. coloratum and one population of Panicum bergii. Agro‐morphological variability between and within accessions of var. makarikariense in a common garden setting was observed, showing that there is still opportunity for selection. Some accessions performed better than the commercialised material in relation to potential forage production. A total of 117 ISSR bands and 48 SSR alleles allowed the detection of genetic variability between and within accessions. The presence of accession‐specific bands suggested distinctness and limited gene flow. The genetic variability encountered in the commercialised material suggested that it is a stabilised population which has not undergone a strong selection process. Low correlation between agro‐morphologic and molecular variability was observed indicating that both approaches provide complementary information. Both morphological and molecular markers reveal genetic differentiation between varieties and species. This study provides a set of new SSR markers available for diversity assessment and valuable information that can be applied directly in collection management for breeding and conservation programmes.  相似文献

The present study applies RAPD technique and morphometric analysis to study the diversity of some accessions belonging to section Solanum. A total of 252 products were amplified with 23 12-mer arbitrary primer pairs, among which 210 were found to be polymorphic. Sixteen morphological characters were measured and used to compile a dendrogram. Both the morphological and RAPD marker analysis clearly separated the different accessions into similar groups. The results indicate that the analyzed cultivars with unknown origin could be derived from S. retroflexum. We found morphological differences among the S. scabrum subsp. scabrum accession which were not reflected in the molecular data. Presumably these accessions represent cultivated forms selected for their habit, fruit quantity and/or quality and leaf size, respectively.  相似文献

In the past 20 yr, the presence of blood platelet inhibitory activity in plant species from the genus Allium has been confirmed by a range of clinical and in vitro investigations. Although a number of Allium species have been identified as possessing antiplatelet properties, little is known of the variability for this trait among accessions in these species. Experiments were conducted to assess variability in antiplatelet activity of 58 Allium (Alliaceae) accessions. Extracts were prepared from a diverse collection of 16 Allium species accessions, 33 cultivated accessions of Allium cepa including standard cultivars, inbred lines, and open-pollinated populations, and nine Allium cepa plant introductions of diverse origin. Platelet inhibitory activity was determined via a platelet aggregation assay with human platelet-rich plasma. Relative in vitro inhibition of platelet aggregation was measured for each accession and control samples, and inhibition constants (IC50) were calculated. Dose-dependent inhibition was observed and measured for each Allium accession. Significant (P < 0.01) IC50 variability was detected among accessions, with the lowest accession IC50 values exhibiting nearly 50-fold greater inhibition of aggregation than the highest accession IC50 values. IC5() variability among Allium cepa accessions was ≈ 12 times less than among Allium species accessions. Results from this investigation demonstrate substantial variability for efficacy of the antiplatelet factor among Allium accessions.  相似文献

The genusSabazia is expanded to include the genusTricarpha, Calea palmeri, and several newly described taxa,S. tridacoides var.tridacoides and var.latifolia, S. palmeri var.lancifolia, andS. longiradiata. Comparative studies of several morphological features suggest thatSabazia is most clearly related toSelloa andGalinsoga. Karyological data reinforce this observation: inSelloa andGalinsoga x = 8 whereas inSabazia x = 4 andn = 4, 8, 16, or 24. Several species ofCalea are very similar toSabazia in floral features and may be of close phyletic affinity, but most species ofCalea differ conspicuously, suggesting more remote relationships. Other taxa in the Galinsoginae apparently fairly closely allied toSabazia includeOteiza andTridax.

Seventy five accessions belonging to 14 species of the genus Cicer were analysed with PCR-based molecular markers to determine their phylogenetic relationships. Eight of the species were annuals and included the Section Monocicer which contains cultivated chickpea (Cicer arietinum L.). The remaining six species were perennials (five from Section Polycicer and one from Section Acanthocicer). More than one accession per species was analysed in most of the wild species; within C. arietinum, 26 accessions including Kabuli and Desi types, were studied. RAPD analyses using 12 primers gave 234 polymorphic fragments. Variability within species was detected. A dendrogram based on the Jaccard similarity index showed that the distribution pattern of variability between species was related to both growth habit and geographical origin. An accession of Cicer reticulatum was closer to accessions of Cicer echinospermum than to the four remaining of C. reticulatum, suggesting the possibility of gene flow between species. Cluster analysis for cultivated chickpea differentiated Kabuli and Desi types but we did not detect a clear relationship between groups and the geographical origin of the accessions.   • Passiflora edulis f. flavicarpa is the most economically important species in the genus Passiflora. However, the origin of this yellow form of passion fruit remains unclear, being suggested as a hybrid (P. edulis f. edulis × P. ligularis) or wild mutant.
  • Here, the origin and genomic relationships of P. edulis f. flavicarpa with some related species in the genus Passiflora (subgenera Decaloba and Passiflora) were investigated using genomic in situ hybridization (GISH). Genomic DNA of 18 species was used as probe, which was hybridized onto chromosomes of P. edulis f. flavicarpa.
  • Of all genomic DNA probes tested, none allowed us to identify a specific chromosome set in P. edulis f. flavicarpa. Conversely, probes from the subgenus Passiflora, P. edulis f. edulis, P. alata, P. cincinnata, P. coccinea, P. nitida and P. vitifolia, produced intense and uniform hybridizations on all chromosomes of P. edulis f. flavicarpa. Moreover, probes from P. ligularis, P. foetida and P. sublanceolata produced more intense hybridizations in the terminal region of four chromosomes, corresponding to the DNAr 45S locus, and also dispersed, less intense, hybridization across all chromosomes. Probes from the subgenus Decaloba, P. biflora, P. capsularis, P. cervii, P. coriacea, P. micropetala, P. morifolia, P. rubra and P. suberosa, produced hybridizations restricted to the DNAr 45S sites.
  • The hybrid origin of P. edulis f. flavicarpa could not be supported based on the GISH results, and it is suggested that this species is conspecific with P. edulis f. edulis, because the probe with DNA of this form hybridized strongly throughout the target genome. The other putative parent species, P. ligularis, showed only a distant relationship with the target genome. The results also suggest that species of the subgenus Passiflora share many repetitive sequences and that the relationship between subgenera Decaloba and Passiflora is very distant.

The wild potato germplasm of the series Acaulia maintained at the Centre for Genetic Resources, The Netherlands, currently consists of 314 accessions. This collection comprises seed samples of the species Solanum acaule (ssp. acaule, ssp. aemulans, ssp. palmirense and ssp. punae) and Solanum albicans collected from South America. In order to validate taxonomic classification, to investigate the extent of redundancy and to study the distribution of genetic diversity across the collection area, the entire collection was analysed with two AFLP primer pairs on two plants per accession. Within the entire sample a total number of 130 polymorphic bands were scored for the two primer pairs. An UPGMA cluster analysis grouped the majority of plants according to the species and subspecies. A total number of 16 misclassifications were identified, including four cases that did not seem to belong to the series Acaulia. Two accessions were found to consist of plants of different AFLP clusters. AFLP data also allowed the taxonomic classification of the subspecies of 97 accessions that previously were described as S. acaule only. For 126 accessions the two individuals studied displayed identical AFLP profiles. Forty six of these 126 accessions shared their profiles with both or single plants of other accessions. These were all tested for identical profiles for a third primer pair, resulting in 15 duplication groups consisting of a total number of 22 accessions and 14 single plants. Analyses of molecular variance (AMOVA) were performed to examine the distribution of genetic variation. Comparison of geographic distances between the collection site of plants and the number of AFLP polymorphisms revealed no consistent relationship between geographic distance and genetic diversity. AFLP analysis appeared to be an efficient method to verify taxonomic classification and to identify redundancies in the wild germplasm of the series Acaulia. The taxonomic identity of Capsicum species is found to be difficult as it displays variations at morpho-chemical characters. Twenty-two accessions of six Capsicum species, namely, C. annuum, C. baccatum, C. chinense, C. eximium, C. frutescens, and C. luteum were investigated for phenotypic diversity based on flower color and for genetic differences by molecular makers. The genetic cluster analyses of 27 RAPD and eight ISSR primers, respectively, revealed genetic similarities in the ranges of 23–88% and 11–96%. Principal component analysis of the pooled RAPD and ISSR data further supports the genetic similarity and groupings. Different species showed variations in relation to corolla shade of flower. C. annuum accessions formed a single cluster in the molecular analysis as maintaining their flower characteristic. C. chinense accession shared flower features with the accessions of C. frutescens and were found to be closer at genotypic level. C. luteum was found to be rather closer to C. baccatum complex, both phenotypically and genetically. The only accession of C. eximium presenting purple flowers falls apart from the groupings. The floral characteristics and the molecular markers are found to be useful toward the delineation of the species specificity in Capsicum collection and identification of genetic stock.  相似文献

Background and Aims

Recent research on the history of Platanus reveals that hybridization phenomena occurred in the central American species. This study has two goals: to help resolve the evolutive puzzle of central American Platanus, and to test the potential of real-time polymerase chain reaction (PCR) for detecting ancient hybridization.

Methods

Sequencing of a uniparental plastid DNA marker [psbA-trnH(GUG) intergenic spacer] and qualitative and quantitative single nucleotide polymorphism (SNP) genotyping of biparental nuclear ribosomal DNA (nrDNA) markers [LEAFY intron 2 (LFY-i2) and internal transcribed spacer 2 (ITS2)] were used.

Key Results

Based on the SNP genotyping results, several Platanus accessions show the presence of hybridization/introgression, including some accessions of P. rzedowskii and of P. mexicana var. interior and one of P. mexicana var. mexicana from Oaxaca (= P. oaxacana). Based on haplotype analyses of the psbA-trnH spacer, five haplotypes were detected. The most common of these is present in taxa belonging to P. orientalis, P. racemosa sensu lato, some accessions of P. occidentalis sensu stricto (s.s.) from Texas, P. occidentalis var. palmeri, P. mexicana s.s. and P. rzedowskii. This is highly relevant to genetic relationships with the haplotypes present in P. occidentalis s.s. and P. mexicana var. interior.

Conclusions

Hybridization and introgression events between lineages ancestral to modern central and eastern North American Platanus species occurred. Plastid haplotypes and qualitative and quantitative SNP genotyping provide information critical for understanding the complex history of Mexican Platanus. Compared with the usual molecular techniques of sub-cloning, sequencing and genotyping, real-time PCR assay is a quick and sensitive technique for analysing complex evolutionary patterns.  相似文献

Nicotiana benthamiana is a model plant utilised internationally in plant virology because of its apparent hyper-susceptibility to virus infection. Previously, others showed that all laboratory accessions of N. benthamiana have a very narrow genetic basis, probably originating from a single source. It is unknown if responses to virus infection exhibited by the laboratory accession are typical of the species as a whole. To test this, 23 accessions of N. benthamiana were collected from wild populations and challenged with one to four viruses. Additionally, accessions of 21 other Nicotiana species and subspecies from Australia, one from Peru and one from Namibia were tested for susceptibility to the viruses, and for the presence of a mutated RNA-dependent RNA polymerase I allele (Nb-RDR1m) described previously from a laboratory accession of N. benthamiana. All Australian Nicotiana accessions tested were susceptible to virus infections, although there was symptom variability within and between species. The most striking difference was that plants of a laboratory accession of N. benthamiana (RA-4) exhibited hypersensitivity to Yellow tailflower mild mottle tobamovirus infection and died, whereas plants of wild N. benthamiana accessions responded with non-necrotic symptoms. Plants of certain N. occidentalis accessions also exhibited initial hypersensitivity to Yellow tailflower mild mottle virus resembling that of N. benthamiana RA-4 plants, but later recovered. The mutant Nb-RDR1m allele was identified from N. benthamiana RA-4 but not from any of 51 other Nicotiana accessions, including wild accessions of N. benthamiana, demonstrating that the accession of N. benthamiana used widely in laboratories is unusual.  相似文献

We evaluated the ploidy levels and tissue culture responses of 16 Japanese Miscanthus accessions, which are registered and vegetatively maintained in the National Agriculture and Food Research Organization GeneBank, Japan, to screen suitable genotypes for the molecular breeding of Miscanthus species. A ploidy analysis showed that most M. sinensis and M. sinensis var. condensatus (var. condensatus) were putative diploids, but one accession identified as M. sinensis was unexpectedly a putative tetraploid. Additionally, M. sacchariflorus and its hybrid accessions were putative tetraploids. The deoxyribonucleic acid levels in var. condensatus were significantly higher than those in the diploid M. sinensis. Of the accessions, 10, including M. sinensis and var. condensatus, could induce plant regenerable embryogenic calli from apical meristems. We selected three of these M. sinensis accessions for further experiments because their calli growth rates were faster than those of the var. condensatus accessions. Tissue culture experiments with the selected accessions indicated that the frequencies of callus and green shoot formation strongly correlated with genotype. The broad-sense heritabilities of the embryogenic callus and green shoot formation frequencies in the selected accessions were 0.75 and 0.65, respectively, indicating that the cultures’ responses were mainly controlled by genetic factors. Thus, we further selected one accession that had the highest efficiencies in callus and green shoot formation, and we observed that light during callus culturing significantly inhibited calli growth, but promoted plant regeneration from calli in the selected accession.  相似文献

Genotypic and Phenotypic Diversity of Cherry Species Collected in Serbia   总被引:1,自引:0,他引:1  
Genetic diversity of cherry species collected in Serbia has been investigated using 26 simple sequence repeat (SSR) markers developed in Prunus. This material consisted of 77 cherry accessions corresponding to the five following species, Prunus cerasus, Prunus avium, Prunus fruticosa, Prunus mahaleb, and Prunus serrulata. A total of 98 alleles were detected, with an average of 3.7 putative alleles per primer combination. Sixteen unique, species-specific, alleles were detected with nine primer pairs in four species, P. avium, P. cerasus, P. mahaleb, and P. serrulata. The highest number of unique alleles, 8, was observed in P. mahaleb and no species-specific alleles were detected in P. fruticosa. SSR markers generated unique fingerprints for all cherry accessions. Cluster analysis classified accessions into four groups according to their taxonomy, where P. avium and P. cerasus were grouped together, supporting P. avium as one of the progenitors of sour cherry. The highest genetic variability and potential value in rootstock breeding was observed in P. mahaleb and P. serrulata material. Principal component (PC) analysis explained more than 50 % of the total observed phenotypic variability using the first two components. The most important characteristics of PC1 were leaf length and width, fruit taste, color of leaf nectaries, fruit weight, leaf blade margin incisions, petiole length, size of vegetative buds, and length of internode. The most important characteristics of PC2 were shape of leaf blade at base, fruit skin color, and leaf blade length and tip angle. The investigated germplasm proved to be sufficiently genetically diverse for use in breeding programs and development of new cherry cultivars and rootstocks.  相似文献

We examined intraspecific chloroplast (cp) DNA variation within Populus deltoides, P. nigra, and P. maximowiczii by restriction fragment analysis using 16 restriction endonucleases and six heterologous probes of cloned Petunia cpDNA fragments. All three Populus species showed intraspecific cpDNA variation, which was intra- and inter-varietal in P. deltoides, intervarietal in P. nigra, and origin-specific in P. maximowiczii. Two varieties of P. deltoides, var deltoides and var occidentalis, showed distinct cp genomes/DNA. Three distinct cp genomes/DNA, separated by a loss or gain of 1 EcoRV restriction site and/or 1 restriction fragment length polymorphism (RFLP), were observed among the individuals of P. deltoides var deltoides. Within P. nigra, cpDNA of var italica was distinct from that of vars nigra and plantierensis by one RFLP and by a loss or gain of one BamHI restriction site. Populus maximowiczii clones of Chinese origin were separated from those of Japanese origin by a gain or loss of one ClaI restriction site in their cpDNA. The estimate of nucleotide substitutions per site in cpDNA was 0.07% between two varieties of P. deltoides, 0.05% between var italica and var nigra or plantierensis of P. nigra, and 0.01% between Japanese and Chinese accessions of P. maximowiczii.  相似文献

The new combination, Bupleurum candollei Wallich ex de Candolle var. paucefulcrans (C. Y. Wu) X. J. He & C. B. Wang, is proposed. The status of this taxon, endemic to Guizhou, China, has been confused since it was first described in 1963. Morphological and anatomical evidence, based primarily on bractlet number and length, umbel number and length, fruit size and shape, and stem structure indicated that B. hamiltonii N. P. Balakrishnan var. paucefulcrans C. Y. Wu is more closely related to B. candollei than to B. hamiltonii. To evaluate the credibility of the morphological similarity, the phylogenetic relationship of B. candollei var. candollei, B. hamiltonii var. hamiltonii and B. candollei var. paucefulcrans were inferred by Bayesian analysis of nrDNA ITS sequences. The molecular analysis is fully congruent with the morphological data and supports the transfer of the variety to B. candollei.  相似文献

Expressed sequence tags for Persea americana Mill. were investigated to expand upon the number of informative microsatellite markers available for avocado. Seventy informative loci were discovered using 24 P. americana var. americana Mill. accessions. The number of alleles detected ranged from two to 17 and averaged 7.1 alleles per locus. These primers successfully amplified products in different varieties of P. americana, hybrids and a related species, Persea schiedeana. These primers will be useful for characterizing germplasm, determining genetic relationships of cultivated accessions, and for marker‐assisted development of root rot‐tolerant P. americana var. americana rootstock material.  相似文献

拉雅松是广西西北部稀有的乡土用材、用脂树种,有较高的经济价值,但其遗传多样性状况及种间进化关系未知。利用SSR分子标记检测拉雅松群体遗传多样性,希望对该物种保护策略的制定提供参考依据。此外,鉴于该地区自然分布的松属树种仅有拉雅松、细叶云南松与马尾松三个种,试图利用SSR分子标记信息分析拉雅松与细叶云南松、马尾松的种间亲缘关系。结果表明:7对SSR引物在拉雅松群体共检测到14个等位基因。有效等位基因数为1.653,观测杂合度为0.577,期望杂合度为0.374,Shannon信息指数为0.540,Nei多样性指数为0.367,表明拉雅松具有较高的遗传多样性。拉雅松与马尾松遗传距离最近为0.0175,与3个细叶云南松群体距离较远,平均为0.0525。拉雅松与马尾松、细叶云南松平均共祖系数(Θ)分别为0.094、0.066,据此推测拉雅松可能与马尾松存在较近的亲缘关系。讨论了拉雅松的遗传多样性保护策略。  相似文献

Screening of a large number of different diploid Solanum accessions with endosperm balance number (EBN) 1 revealed segregation for strong resistance and sensitivity to Phytophthora infestans in accessions of Solanum mochiquense. Genetic analysis showed that resistance in S. mochiquense accession CGN18263 resides at the distal end of the long arm of chromosome IX, is linked to restriction fragment length polymorphism marker TG328 and is in the neighbourhood of the quantitative trait locus (QTL) Ph-3 conferring resistance to P. infestans in tomato. This is the first genetic study of S. mochiquense, a wild diploid species originating from fog oases in the Peruvian coastal desert.  相似文献
