A strain of Ulocladium botrytis isolated from diseased Orobanche crenata shoots caused disease on the parasitic weed in pathogenicity tests. The potential of the fungus to be developed as a mycoherbicide for Orobanche spp. was further investigated. Although the fungus significantly decreased O. crenata germination in vitro by 80%, it did not generally lead to a decreased number of O. crenata shoots or tubercles in inoculated root chambers or pots. However, the number of diseased or dead tubercles and underground shoots was significantly increased compared to the noninoculated treatments. Postemergence inoculation of O. crenata shoots with a conidial suspension resulted in the death of almost all inoculated plants 14 days after application under greenhouse conditions. In preliminary host-range studies, the pathogen caused disease on Orobanche cumana on sunflower whereas on Orobanche aegyptiaca shoots parasitizing tomato only minimal disease symptoms could be detected after postemergence inoculation. Based on the results of our investigations, we conclude that Ulocladium botrytis has only a limited potential to be used as a biocontrol agent against Orobanche spp.  相似文献

Despite the tremendous economic impact of broomrapes (Orobanche spp.) on agriculture in many countries little is known of the pattern of genetic variation within this group of parasitic weeds. The present paper describes the use of RAPD markers for the study of five Orobanche species in agricultural fields in Israel. Pronounced genetic differentiation was found between the species, and RAPD markers were raised for the identification of each of them. Southern-hybridization patterns of RAPD products of the various species were used to confirm the interpretation. The same markers were valid both for broomrapes collected in agricultural fields and for those collected in natural habitats. The validity of the markers found for O. cumana and O. crenata was confirmed on plants of the same species that were collected in Spain. Parsimony analysis of 86 RAPD characters produced a tree that clearly distinguishes between the five studied Orobanche species, separates the two Orobanche species belonging to sect. Trionychon from those belonging to sect. Osproleon, and supports the separation of O. cumana from O. cernua and of O. aegyptiaca from O. ramosa.  相似文献

列当是一种根寄生种子植物,是我国进境植物检疫对象,同时也是我国的内检对象。新疆是我国列当发生面积最大、危害最重和种类最多的地区,对新疆的主要经济作物,如加工番茄、向日葵、甜瓜等已造成严重危害。在新疆文献记载的列当种类有18种,常见的有埃及列当、大麻列当、向日葵列当和弯管列当,并已广泛分布于新疆各县、市、团场。列当的防治技术主要包括检疫、田间管理、轮作等,但目前的防治效果均不理想。本文通过对新疆列当的相关文献搜集、整理分析,总结了目前新疆列当的种类、分布情况,以及综合防治技术,为今后列当的研究和防治提供参考。  相似文献

Although there are reports of isolation of mycoherbicidal pathogens attacking the widespread broomrapes (Orobanche spp.) that parasitize legumes and vegetables, none is in use or available. This is despite there being no good method of controlling broomrapes in most crops other than by preplant fumigation with methyl bromide. Two highly parasitic fungi, Fusarium arthrosporioides strain E4a (CNCM I-164) and F. oxysporum strain E1d (CNCM I-1622), were isolated from nearly 100 organisms found on diseased, juvenile, emerging Orobanche flower stalks. A near-axenic polyethylene envelope system for culturing broomrape on tomato roots was used to ascertain pathogenicity of these strains. Both organisms fulfilled Koch's postulates for being primary pathogens. Their DNAs were analyzed and fingerprinted by restriction fragment length polymorphism and random amplified polymorphic DNA, showing that they are indeed different from each other and from many other Fusarium spp. and other formae speciales of F. oxysporum including a strain that attacks O. cumana on sunflowers. Both strains infect O. aegyptiaca, O. cernua, and O. ramosa, but not O. cumana. They did not infect any of the vegetable and legume crops tested and thus seem specific to Orobanche. Tomato plant roots dipped into a fungal spore and mycelial suspension and planted in broomrape-infested soil were protected for 6 weeks, as were tomato transplants in pot experiments. About 90% control was also achieved by posttransplant soil drench with fungal suspensions in pot experiments. These pathogens may be effective as seed, transplant, or soil-drench treatments of high-value vegetable and other crops.  相似文献

Summary Propagation and conservation of four pharmaceutically important herbs, Ocimum americanum L. syn. O. canum Sims. (hoary basil); O basilicum L. (swett basil); O. gratissimum L. (shrubby basil); and O. sanctum L. (sacred basil) was attempted using synthetic seed technology. Synthetic seeds were produced by encapsulating axillary vegetative buds harvested from garden-grown plants of these four Ocimum species in calcium alginate gel. The gel contained Murashige and Skoog (MS) nutrients and 1.1-4.4 μM benzyladenine (BA). Shoots emerged from the encapsulated buds on all six planting media tested. However, the highest frequency shoot emergence and maximum number of shoots per bud were recorded on media containing BA. Of the six planting media tested, both shoot and root emergence from the encapsulated buds in a single step was recorded on growth regulator-free MS medium as well as on vermi-compost moistened with halfstrength MS medium. Rooted shoots were retrieved from the encapsulated buds of O. americanum, O. basilicum, and O. sanctum on these two media, whereas shoots of O. gratissimum failed to root. The encapsulated buds could be stored for 60 d at 4°C. Plants retrieved from the encapsulated buds were hardened off and established in soil.  相似文献

Parasitic broomrapes (Orobanche spp.) are majoruncontrolled weeds in the Mediterranean regions of Europe and the NearEast causing major losses to vegetable, grain legume, and sunflowercrops. Selective herbicides alone cannot provide persistent, season-longcontrol of these parasites, and much methyl bromide is used for theircontrol, where affordable. Thus they are excellent targets forbiocontrol. The recent progress by the COST 816 Orobancheworking group in this area is reviewed herein. Natural infestation bythe fly Phytomyza orobanchia of seed capsules of Orobanchecrenata parasitising faba bean halved Orobanche seedproduction while inundative releases of adults reduced it to 5%of viable seeds. The fungi Fusarium arthrosporioides E4a andF. oxysporum E1d, as well as strains of bacteria were isolatedfrom diseased, juvenile, Orobanche flower stalks. They arepathogenic to O. aegyptiaca, O. crenata and O. ramosaon most vegetable crops. A F. oxysporum f. sp.orthoceras was specifically pathogenic to O. cumana onsunflowers. All were used in various experiments with a modicum ofsuccess. Methods were developed to formulate isolated mycelia, whichcould eventually allow the use of transgenic hypervirulent pathogens inasporogenic (deletion) mutants (as a failsafe against spread).Mycotoxins were also isolated from different Fusarium and otherfungal species that kill Orobanche, and are being consideredfor direct use, or to augment other strategies. All threeFusarium spp. used have been transformed with gusand/or gfp genes allowing tracing their movement in theenvironment, and opening the way to future transformations tohypervirulence.  相似文献

Use of sodium chloride (NaCl) as a deicing salt results in high concentrations of ions in roadside soils, which decreases seedling emergence in these areas. Greenhouse experiments performed in soil culture tested the efficacy of three soil fertilizers, gypsum (CaSO4), potash (potassium chloride, KCl) and potassium nitrate (KNO3), in alleviating NaCl stress on seedling emergence of three grass species exhibiting a range of salt tolerance, Poa pratensis (Kentucky bluegrass), Bouteloua gracilis (blue grama), and Puccinellia distans (alkali grass). Two-factor factorial designs were utilized for each species-fertilizer combination. Treatments of 5000 mg/L (0.086 M) NaCl with or without fertilizer, in concentrations that were equal to 0.5, 1, and 2 times the molar equivalent of 5000 mg/L NaCl were applied biweekly. Salt stress on Poa pratensis emergence was alleviated by all fertilizers with CaSO4 having the greatest effect in alleviating NaCl stress and potash and potassium nitrate alleviating stress at lower treatment levels. Emergence of Bouteloua gracilis and Puccinellia distans was in most cases negatively effected by soil amendments.  相似文献

Orobanche cumana Wallr. (sunflower broomrape) is a devastating root parasitic weed, causing enormous crop losses worldwide. The question was whether or not switchgrass has the potential to be a ‘trap crop’ for O. cumana control. To answer this question, the field experiments, pot experiments, and laboratory experiments were conducted in this study. The ability of nine switchgrass (Panicum virgatum L.) cultivars to induce O. cumana seeds germination was tested. Results indicated that root extracts shoot extracts, rhizosphere soil, and root exudates from switchgrass induced O. cumana germination. Ability to induce germination varied significantly among growing stages, with the earlier part of growing season (grown for 2 weeks) generally inducing the highest O. cumana germination rates. The methanol was a more suitable solvent than distilled water for extracting germination stimulants from switchgrass plants. Ten-fold dilutions of the extracts generally induced higher germination rates than either undiluted or 100-fold dilutions. The germination rates of O. cumana seeds in shoot extracts treatments were positively correlated with those in the root extracts (R2 = 0.6397; p < 0.01) and negatively correlated with those in the rhizosphere soil treatments ( R2 = 0.4433; p < 0.05). In conclusion, it is believed that switchgrass is a potential trap crop for the control of root parasitic weed O. cumana.  相似文献

The root holoparasitic angiosperm sunflower broomrape (Orobanche cumana) specifically affects sunflower (Helianthus annuus) growth and causes severe damage all over the world. This investigation was designed to examine the protective effects of salicylic acid (SA) treatment to the seeds of an O. cumana‐susceptible cultivar of sunflower (TK0409). Sunflower seeds were pretreated with different concentrations (0, 0.5 and 1 mM) of SA and inoculated with O. cumana for 4 weeks. O. cumana infection resulted in reduction in plant biomass, endogenous SA level, and the expression of SA‐related genes including pal, chs and NPR1. By contrast, O. cumana infection enhanced the production of reactive oxygen species, activities of antioxidant enzymes as well as contents of phenolics and lignin. Seed treatment with 1 mM SA increased sunflower biomass in terms of plant height, fresh weight and dry weight by 10%, 13% and 26%, respectively, via reducing the number and biomass of established O. cumana. The increase of hydrogen peroxide contents by 14% in the 1 mM SA treated sunflower plants appeared to be because of the inhibition of ascorbate peroxidase and catalase by exogenous SA. The enhanced expression of pathogenesis‐related genes (PR3 and PR12, encoding chitinase and defensin, respectively) after 4 weeks of inoculation indicated that systemic acquired resistance was induced in the SA treated sunflower in which the level of endogenous SA was also elevated in a dose‐dependent manner. The increased expression of a hypersensitive‐responsive (HR) gene hsr indicated that the resistance of sunflowers might be associated with a hypersensitive reaction which was activated by exogenous SA treatment.  相似文献

Nonlinear models were used to estimate first emergence and peak abundance dates for Aphthona lacertosa Rosenhauer and A. nigriscutis Foudras, two flea beetles introduced to control leafy spurge, Euphorbia esula L., in North America. For model development, 26 field sites were sampled for flea beetle abundance at weekly intervals for eight weeks in three western Minnesota counties in 2000, 2001, and 2002. A three-parameter Weibull function, fit to observed cumulative probability distributions, were used to predict accumulated degree-days (ADD) to first emergence. Bias testing indicated the Weibull function provided a useful estimate of first emergence for A. lacertosa (304 ADD, lower developmental threshold 7.5 °C), but failed to produce a useful estimate for A. nigriscutis. A third-order polynomial was used to approximate seasonal abundance and predict peak abundance for each species. Estimated ADD to peak abundance of A. lacertosa was 594 ± 24 (DD > 7.5 °C) and 670 ± 15 (DD > 9.3 °C) for A. nigriscutis. Models were validated with additional data sets from Minnesota, Montana, and North Dakota. Estimated date of peak emergence provided useful predictions of peak emergence for Minnesota and North Dakota, but failed to predict peak emergence in Montana. We speculate that variation in climate and environmental conditions between Midwestern states and Montana were responsible for differing emergence patterns. We conclude that phenology models should be developed regionally to provide useful predictions of peak emergence for land managers. Maps were developed for Minnesota to spatially display predicted dates of peak abundance for A. lacertosa and A. nigriscutis.  相似文献

Significant relationships between the emergence of adult predatory species,Ophyra aenescens (Wiedemann) andMuscina stabulans (Fallen) and prey,Musca domestica L.,Fannia femoralis (Stein) andF. canicularis (L.) existed on 10 poultry ranches in southern California. Predatory species may function in natural control at intervals throughout the year, especially on predominant prey,F. femoralis andF. canicularis. Dipterous species, especiallyO. aenescens andF. femoralis, were only moderately influenced by the surface area of breeding habitat available. Accuracy in the measurement of population trends in Diptere may be enhanced by increasing the number of samples at any given ranch rather than the addition of study ranches. Parasitism, ranging from 22.6–37.8% of collected puparia, involved principally 3 species,Muscidifurax zaraptor Kogan & Legner,M. raptor Girault & Sanders andSpalangia cameroni Perkins.   相似文献

Summary Oviposition by Cactoblastis cactorum on Opuntia ficus-indica and O. aurantiaca was assessed from the positioning of egg sticks on plants in the field. The number of egg sticks laid on O. ficus-indica plants was affected by: (1) plant size; (2) moth emergence near the plant; (3) cladode condition; and (4) plant conspicuousness. These factors contributed towards the clumping of egg sticks on plants. There was no apparent oviposition preference for one of the two host plant species despite the fact that egg predation was higher and fecundity lower on O. aurantiaca. The selection of a site for oviposition on the host plants was influenced by: (1) cladode condition; (2) height above ground; and (3) shelter from wind during oviposition. Succulent cladodes were the favoured sites for oviposition. The evidence suggests that in C. cactorum, oviposition site selection is largely the net result of a compromise between oviposition behaviour selected for increasing the probability of juvenile survival and oviposition behaviour selected for increasing the probability of laying the full complement of eggs. In addition, environmental and physiological factors such as wind and wing-loading, are thought to place constraints on the range of sites available for oviposition.  相似文献

Ostrinia nubilalis is (Lepidoptera: Crambidae) a severe pest ofcorn in the major corn growing areas of theUnited States. The efficacy of a Beauveria bassiana application, for season-long suppression of O. nubilalis was evaluated in 1996 and 1997 at locationsacross Iowa. Beauveria bassiana,Mycotech 726 (Mycotech Corporation, Butte, MT)formulated on corn grit granules (14–20 mesh)at 2.2 × 109 conidia/g and applied with ahand-held applicator at the rate of 0.4 g/plant(8.8 × 108 conidia/plant). Applicationswere made when plants were in the V7 or R1growth stage. The length of larval tunneling,percentage of plants not infested with O.nubilalis, percentage of plants with anendophyte, and yield from treated and controlplots were determined. Whorl-stage applicationof B. bassiana in 1996 resulted in asignificant reduction in centimeters oftunneling (46–55%) and the percentageplants not infested by O. nubilalis. In1997, B. bassiana caused significantreductions in larval tunneling at all locations(20–53%); however, a significant increase inthe percentage of plants not infested with O. nubilalis occurred at only one location. Treatment of plants with B. bassiana in1997 did not significantly increase thepercentage of plants with an endophyte;however, the trend, with the exception of onesite, was for a greater percentage ofendophytic plants in treated versus untreatedplots. A whorl-stage application of a granularformulation of B. bassiana was mostefficacious in reducing O. nubilalis larval damage.  相似文献

The co-evolution of Orobanche spp. and their hosts within the same environment has resulted in a high degree of adaptation and effective parasitism whereby the host releases parasite germination stimulants, which are likely to be unstable in the soil. Our objective was to investigate whether extracts from non-host plants, specifically, Chinese medicinal plants, could stimulate germination of Orobanche spp. Samples of 606 Chinese medicinal herb species were extracted with deionized water and methanol. The extracts were used to induce germination of three Orobanche species; Orobanche minor, Orobanche cumana, and Orobanche aegyptiaca. O. minor exhibited a wide range of germination responses to the various herbal extracts. O. cumana and O. aegyptiaca exhibited an intermediate germination response to the herbal extracts. O. minor, which has a narrow host spectrum, showed higher germination rates in response to different herbal extracts compared with those of O. cumana and O. aegyptiaca, which have a broader host spectrum. Methanolic extracts of many Chinese herbal species effectively stimulated seed germination among the Orobanche spp., even though they were not the typical hosts. The effective herbs represent interesting examples of potential trap crops. Different countries can also screen extracts from indigenous herbaceous plants for their ability to induce germination of Orobanche spp. seeds. The use of such species as trap plants could diminish the global soil seed bank of Orobanche.  相似文献

The chromosome numbers of five species ofOrobanche sect.Orobanche (O. alsatica, O. laserpitii-sileris, O. loricata, O. salviae, O. teucrii) are reported for the first time and previous counts could be verified in ten other species. Now the chromosome numbers of all species of sect.Orobanche occurring in Central Europe are known: they are diploid (2n = 38) with the exception ofO. gracilis (tetra- and hexaploid, aneusomatic).

Normal (nonglutinous) rice plants (Oryza sativa andO. glaberrima) contain more than 18% amylose in endosperm starch, whilewaxy (glutinous) plants lack it in this starch. In contrast, leaf starch contained more than 3.6% amylose even inwaxy plants. SDS-PAGE analysis of proteins bound to endosperm starch granules in the normal plants revealed a single band with aMr of 60 kd, whereaswaxy plants did not exhibit a similar band. The activity of starch synthase (NDP-glucose-starch glucosyltransferase) was completely inhibited by antibody against the 60-kd protein. Thus, we conclude that the 60-kd protein is thewaxy protein encoded by theWx allele, which also plays a role in the synthesis of nonglutinous starch in endosperm tissue. In leaf blades, the proteins bound to starch granules separated into five bands withMr's of 53.6 to 64.9 kd on SDS-PAGE. Analysis of these proteins by immunoblotting using antiserum againstWx protein and inhibition of starch synthase activity by the synthase antibody revealed that none of these proteins was homologous toWx protein. We suggest that the synthesis of amylose in leaf blades is brought about by a protein encoded by a gene(s) different from theWx gene expressed in the endosperm.  相似文献

The staphylinid beetle, Oligota pygmaea (Solier) is an important predator of the red spider mite, Oligonychus coffeae (Nietner) infesting tea. Biology, life table and predatory efficiency of O. pygmaea were studied under laboratory conditions. Duration of developmental stages of O. pygmaea was 3.2, 5.7 and 12.5 d for eggs, larvae and pupae, respectively with an average of 23.0 d from egg to adult emergence. After a mean preovipostion period of 2.9 d, each female laid an average of 400.5 eggs in its life span. Adult O. pygmaea lived for an average of 54.1 d. Adult females lived for a longer period of 58.8 d compared to the longevity of 49.4 d of adult male. Studies revealed that its life table characterized by an intrinsic rate of natural population increase (r) of 0.118 d, net reproductive rates (Ro) of 243.693 eggs/female, gross reproduction rate (Σmx) of 245.313 eggs/female, generation time (T) of 46.575 d, doubling time (DT) of 5.874 d and finite rate of increase (λ) of 1.125 d. Seasonal abundance of O. pygmaea and its prey, O. coffeae was monitored by sampling 25 tea leaves randomly from each experimental block grown under the prevailing field conditions. O. pygmaea showed a typical pattern of population dynamics with a peak during January to March and low incidence during June to September. Peak in the population of O. pygmaea coincided with the abundance of O. coffeae in the tea fields. Weather factors such as high temperature, low relative humidity and low sunshine hours adversely affected the populations of O. pygmaea. The first to third instar larvae of O. pygmaea consumed 31.0–133.2 eggs of mites per day. Third instar larva of O. pygmaea consumed an average of 133.2 eggs, 46.4 hexapod larvae, 39.6 nymphs and 11.4 adults per day. Adult females consumed more number of red spider mites compared to the males.  相似文献

An investigation of pseudocopulation behaviour in species ofOphrys from southern Spain confirms the close relationship betweenCampsoscolia ciliata (Scoliidae) andOphrys speculum, and betweenEucera nigrilabris (Apoidea) andOphrys tenthredinifera. It could be demonstrated thatEucera barbiventris is the pollinator ofOphrys scolopax subsp.scolopax, whereas other species ofEucera andTetralonia which are active at the same time show no interest at all for the flowers of this species. Special attention was paid to the forms of the taxonomically confusedOphrys fusca group:O. fusca s. str.,O. iricolor, O. omegaifera andO. atlantica. WhileO. fusca s. str. is widespread, small-flowered and has late anthesis,O. iricolor has very large flowers and early anthesis. Each of the four members ofO. fusca agg. in S. Spain is pollinated by a different bee, and selective experiments show that each of these four species of bees is specifically attracted only to one of theOphrys species:Andrena flavipes is the pollinator ofO. fusca s. str.,Colletes cunicularius infuscatus ofO. iricolor, Anthophora atroalba ofO. omegaifera, andChalicodoma parietina ofO. atlantica. These four pollinators belong to 4 different bee families (Andrenidae, Colletidae, Anthophoridae, Megachilidae). As this type of pollination represents a very effective pregamic isolation mechanism, these four taxa ofOphrys fusca agg., at least in southern Spain, behave and should be regarded as genuine species.

The infectivity, time to first emergence of infective juveniles (IJs), total number of IJs per insect and IJs body length of the entomopathogenic nematode Heterorhabditis megidis (strain NLH-E87.3) after development in larvae of two insect hosts, Galleria mellonella (greater wax moth) and Otiorhynchus sulcatus (vine weevil) was studied. At a dose of 30 IJs, larvae of G. mellonella show to be significantly more susceptible than O. sulcatus larvae. At a dose of one IJ, vine weevil larvae were more susceptible. The number of invading infective juveniles (IJs) increased with host size while the host mortality at a dose of one IJ decreased with the increase of host size. Time to first emergence was longer at a dose of one IJ per larva and increased with the increase of host size in both insect species. Reproduction of IJs differed between host species, host sizes and doses of nematodes. Generally, the IJs body size increased with an increasing host size. The longest infective juveniles were produced at the lowest IJ doses. Results are discussed in relation to the influence of different host species and their different sizes on the performance of H. megidis (strain NLH-E87.3) as a biological control agent.  相似文献

Patterns of seedling recruitment may have persistent effects on population and community processes. Assuming seed availability is not limiting, the environmental sieve (i.e., the suite of factors influencing seed germination and seedling emergence and survival) determines how many seedlings establish and, most importantly, where they do so. In this study, we identify the spatial structure of some resources and abiotic conditions known to be significant for tree seedling emergence and survival and determine how these environmental factors influence the establishment of Fagus grandifolia, Acer saccharum, Fraxinus americana, and Ostrya virginiana in a deciduous forest of southern Québec (Canada). We expect an increase from Fagus, through Acer and Fraxinus, to Ostrya in the control of environmental variables on seedling emergence and survival, because of differences in the seed size of these species. Density of newly-emerged seedlings of all four species showed positive spatial autocorrelation at distances of up to ca. 10 m. Environmental variables were also structured at the same spatial scale, except for soil moisture. Acer seedling emergence pattern was positively correlated to photosynthetic photon flux density (PPFD), and the pattern of Fraxinus to soil N and moisture. Seedling survival was not spatially autocorrelated for any of the four species, although it was positively density-dependent in Acer and Fagus. In only Ostrya was seedling survival correlated (positively) to one of the environmental variables studied, i.e., PPFD. Overall, environmental variables were spatially less heterogeneous than seedling emergence and survival. Either seed availability was not saturating or factors not considered here, such as competition and predation (the intensity of which often varies with resources and/or abiotic conditions), modified the influence that the physical environment had on patterns of seedling establishment. Our prediction of a greater environmental control on seedling emergence and survival in small-seed species was not totally confirmed.  相似文献
