Data collected from 264 bulls of 13 beef breeds at the Michigan Bull Test Station was evaluated to determine if scrotal circumference (SC) adjusted to 200 d of age could be used to predict scrotal circumference at 1 yr of age. Scrotal circumference of each bull was recorded on arrival at the test station and at the time of breeding soundness examination (BSE) and was adjusted to 200 and 365 d of age, respectively. Bulls with adjusted SC>34.0 cm by 365 d of age averaged a larger SC at 200 d (P < 0.0001) and faster scrotal growth (P < 0.0001) than bulls with a 365 d adjusted SC /= 23.0 cm had a 95% probability of achieving SC > 34.0 cm by one year of age. Calves measuring < 23 cm at 200 d had a 54% probability of achieving > 34.0 cm scrotal size by one year. This information can contribute to the selection of breeding bulls that will achieve desirable scrotal size by one year of age.  相似文献

Dairy heifers were used to compared the effects of two timed AI + controlled internal drug release (CIDR) protocols (5-day vs. a modified 7-day) on: (1) luteal regression to initiate a new ovarian follicular wave; (2) ovarian response to the initial GnRH injection; and (3) pregnancy outcomes. Holstein heifers (N = 543) were assigned randomly to two treatments: (1) 25 mg PGF (im) and a CIDR insert on Day −7 followed by 100 μg of GnRH (GnRH-1) on Day −5 and 25 mg PGF (im) at CIDR insert removal (7-day [7D]) on Day 0; or (2) 100 μg GnRH (GnRH-1) and insertion of a CIDR on Day −5 and 25 mg PGF (im) at CIDR removal (5-day [5D]) on Day 0. Insemination with frozen-thawed conventional or gender-biased semen occurred after detected estrus from Days 0 to 2 or by appointment at 72 h after PGF when a second 100-μg dose of GnRH was given. Blood was collected on Days −7, −5, 0, and 3 to determine concentrations of progesterone and incidence of luteolysis. Ovaries were scanned on Days −5 and 0. Luteolysis in the 7D treatment by 48 h after the initial PGF was greater (P < 0.01) than what occurred spontaneously in the 5D treatment (36.2% vs. 19.7%, respectively). Incidence of ovulation after GnRH-1 on Day −5 was greater (P < 0.05) for 7D than for 5D heifers, but the proportion of heifers with an induced CL on Day 0 did not differ between treatments. Heifers inseminated after detected estrus (166/543, 30.6%) on Days 0, 1, and 2 had greater (P < 0.05) pregnancy per AI (P/AI) at 32 days post AI than after timed AI (38.2% vs. 28.3%) on Day 3. Pregnancy P/AI, however, was greater (P < 0.05) for 7D heifers inseminated at estrus (46.5%) than for 7D heifers receiving the timed AI (26.8%) and differed (P < 0.05) from all 5D heifers regardless of insemination time at estrus (30.5%) or at timed AI at 72 h (29.9%). At the Florida location in which conventional and sexed semen were used during two breeding clusters, P/AI using sexed semen (43.9%, N = 56) did not differ from that of conventional semen (21.2%, N = 50). Remaining replicates of sexed semen produced similar P/AI at the other two locations (sexed = 27.6%, N = 71; and sexed = 31.9%, N = 215). We concluded that the modified 7-day CO-Synch + CIDR program produced more P/AI in heifers inseminated at estrus than a standard 5-day CO-Synch + CIDR program, but when timed AI occurred at 72 h after PGF and CIDR insert removal, P/AI did not differ between programs.  相似文献

Timing of ovulation and changes in plasma progesterone, luteinizing hormone (LH), and prolactin (PRL) during periovulatory stages were determined in Holtzman rats exhibiting regular 4- or 5-day cycles under a daily artificial illumination from 0500 to 1900 h. The 5-day cycling rats ovulated between 0130 and 0930 h on estrus, whereas some of the 4-day cycling animals ovulated as early as about 0130 h and others as late as 1130 h on estrus. Onset time of preovulatory LH and progesterone surges was about 1500 h on proestrus in both the 4- and the 5-day cycling rats. Peak levels of plasma LH and progesterone were measured at 1700 to 1900 h on proestrus, while the first rises and peak values of plasma PRL were evident a few hours earlier than those of plasma LH in the rats with two cycle lengths. Plasma LH levels at 1900 h on proestrus as well as plasma progesterone levels at 1600 and 2300 h on proestrus and at 0130 and 0330 h on estrus were significantly lower in the 5-day cycling rats than in the 4-day cycling animals (p less than 0.05). In contrast, PRL levels from 1500 through 2300 h on proestrus remained consistently higher in 5-day cycling rats than in 4-day cycling rats, and significant differences in PRL levels between these rats were apparent at 1500, 1600, and 2100 h (p less than 0.05-0.01). Thus, these results demonstrate that the 5-day cycling rats exhibit the attenuated magnitude of LH surge accompanied by the augmented preovulatory PRL release, and that plasma progesterone levels reflect the magnitude of LH surge. A tentative working hypothesis concerning the etiology of the 5-day cycle has been proposed.  相似文献

Humans are frequently exposed to combinations of lead (Pb), cadmium (Cd) and Arsenic (As) but there is a paucity of actual data on the molecular effects of these agents at low dose levels. The present factorial design studies were undertaken in rats to examine the effects of these agents at LOEL dose levels on a number of molecular parameters of oxidative stress in hematopoietic and renal organ systems following oral exposure in drinking water at 30, 90 and 180 day time points. Results of these studies demonstrated dynamic, time-dependent alterations in both molecular targets and inducible oxidative stress protective systems in target cell populations. In general, cellular protective systems, which protected against oxidative damage at the 90 day time point, appeared to be finite such that molecular manifestations of oxidative stress became statistically significant at the 180 day time point for several of the combination exposure groups. These data demonstrate the importance of duration of exposure in assessing the toxic potential of Pb, Cd and As mixtures at low dose levels.  相似文献

The administration of 63NiCl2 to 19-day pregnant rats produced a limited transfer of nickel to the foetus. An experimental model based on the incubation of the conceptuses still connected to the maternal organism resulted in an appreciable net transfer of nickel through the amniotic membranes. The placenta retained a significant amount of radioactive nickel as did the lining of the uterine wall. The circulation of nickel through the foetus following the direction maternal blood----placenta----foetus----amniotic membranes----endometrium----maternal venous blood, is postulated. This transfer could provide an additional protection to placental barrier against metal toxicity.  相似文献

Rat embryos (9.5-day-old) were cultured for 48 hours in serum from diet-induced folic-acid-deficient rats. Resultant embryos were frequently abnormal; many were growth retarded and exhibited a defect in the turning mechanism that inverts the embryo from ventrally to dorsally convex. Affected embryos displayed abnormal twisting or kinking of the neural tube. Gross anaemia was also frequently observed, and the protein content of the embryos was markedly less than that of embryos grown in normal rat serum. Supplementation of the deficient serum with folic acid improved growth and greatly reduced the occurrence of deformities. It virtually eliminated the incidence of gross anaemia but only partially restored the protein content of the embryos to the level observed in those grown in normal rat serum. The effects of the folate deficiency could be eliminated by supplementation with normal rat serum. The data indicate that embryos have a requirement for adequate folate in order for normal growth and differentiation to take place; they also suggest that some of the embryopathic effects of maternal folate deficiency are mediated by secondary effects on maternal metabolism. This may take the form of a disturbance in the production of maternally synthesised growth factors necessary for normal embryonic development.  相似文献

Two separate experiments were conducted to determine whether prostaglandin (PG) E2 stimulates the secretion of progesterone by 270- or 200-day Brahman placentas in vitro. Secretion of progesterone, PGF2alpha, pregnancy specific protein B, or estradiol-17beta by 270-day Brahman placentas was not affected (p > or = 0.05) by PGE2, during the 4-h incubation period at the doses tested. Indomethacin or meclofenamic acid decreased (p < or = 0.05) 270-day Brahman placental secretion of PGE and PGF2alpha by 98 and 60%, respectively. However, PGE2 induced (p < or = 0.05) its own secretion, but not the secretion of PGF2alpha (p > or = 0.05), by 270-day Brahman placentas, even in the presence of indomethacin or meclofenamic acid at a dose of 100 ng/mL. Also, secretion of 8-Epi-PGE2 by Day 270 Brahman placentas was increased (p < or = 0.05) by PGE2. Secretion of progesterone, estradiol-17beta, or pregnancy specific protein B by 200-day Brahman placentas was not affected by PGE2, 8-Epi-PGE2, PGF2alpha, estradiol-17beta, or trichosanthin during the 4- or 8-h incubation period (p > or = 0.05). Secretion of estradiol-17beta at 8 h was lower (p < or = 0.05) in all treatment groups and did not differ (p > or = 0.05) among the 8-h incubation treatment groups. Secretion of PGE by 200-day Brahman placentas was reduced (p < 0.05) by indomethacin 72 and 82% and by meclofenamic acid 72 and 96%, respectively, at 4 and 8 h when compared to controls. Secretion of PGF2alpha was reduced (p < or = 0.05) 71 and 86% by indomethacin or 89 and 89% by meclofenamic acid at 4 and 8 h, respectively, and did not differ (p > or = 0.05) between 4 and 8 h of incubation. PGE2 did not (p > or = 0.05) induce secretion of PGE above what was added in any treatment group. PGE in culture media was increased (p < or = 0.05) by 8-Epi-PGE2, pregnancy specific protein B, and the 100 ng/mL PGF2alpha dose (p < or = 0.05), but not by PGE2, progesterone, estradiol-17beta, 8-Epi-PGF2alpha, or trichosanthin. Secretion of PGF2alpha by 200-day Brahman placentas was not affected (p > or = 0.05) by 8-Epi-PGE2, progesterone, or estradiol-17beta, but PGF2alpha secretion was increased (p < or = 0.05) by trichosanthin or PGE2, even in the presence of indomethacin or meclofenamic acid. It is concluded that PGE does not affect secretion of progesterone by 200- or 270-day bovine placentas, but, pregnancy specific protein B may regulate placental secretion of PGE. Also, indomethacin and meclofenamic may affect enzymes converting PGH to PGE rather than acting only on cyclooxygenase because indomethacin and meclofenamic acid lowered PGE secretion by 270-day Brahman placentas more than they lowered PGF2alpha. In addition, it is concluded that PGE2 can induce bovine placental secretion of PGE, but this is dependent upon the stage of gestation.  相似文献

The objectives were to compare follicular dynamics, preovulatory estradiol concentrations, and progesterone concentrations between the 7-day (7CO, n = 15) and 5-day (5CO, n = 13) CO-Synch + controlled internal drug release device (CIDR) program in primiparous suckled beef cows. On Day −7 (7CO) or Day −5 (5CO), GnRH (100 μg) was administered (GnRH-1) and a CIDR was inserted. On Day 0, hour 0, CIDR was removed and cows received PGF (25 mg) at hours 0 and 12. Animals were administered GnRH (100 μg, GnRH-2) at either hour 60 (7CO) or 72 (5CO). Follicular growth and ovulation to both GnRH-1 and GnRH-2 were evaluated using ultrasonography. Concentrations of estradiol were determined in blood samples taken at hours 0, 36, 60, and 72 (5CO). Blood samples were collected on Days 5, 8, and 14 for progesterone quantification. Ovulation rate to GnRH-1 did not differ between the 7CO (11/15) and 5CO (8/13) treatments, and for all dependent variables the statistical model included treatment, ovulation to GnRH-1, and their interaction. Diameter (mm) of the ovulatory follicle did not differ between treatments (13.4 ± 0.3) but was greater (P < 0.05) in cows that responded to GnRH-1 (13.8 ± 0.3) than those did not (12.6 ± 0.6). Maximum estradiol concentrations tended (P = 0.06) to be greater in the 5CO (7.3 ± 0.5 pg/mL) than 7CO (6.1 ± 0.7 pg/mL) treatment and tended to be greater (P = 0.08) in cows that responded to GnRH-1 (7.1 ± 0.5 pg/mL) than those did not (5.6 ± 0.9 pg/mL). Three cows in the 7CO treatment failed to develop a CL after GnRH-2. There was a treatment by response to GnRH-1 interaction (P < 0.05) for progesterone concentrations. In cows that did not respond to GnRH-1 in the 7CO treatment, progesterone concentrations were less (P < 0.05) than in those that responded to GnRH-1 in the 7CO treatment and tended (P = 0.09) to be less than in cows in the 5CO treatment that did not respond to GnRH-1. In conclusion, these findings demonstrate that failure to respond to GnRH-1 is detrimental to estradiol and progesterone concentrations with a 7-day interval between GnRH-1 and PGF but of little consequence when this interval is shortened to 5 days.  相似文献

Cholecystokinin (CCK) activates the myenteric neurons of adult rats. The goal of this work is to determine the ontogeny of this activation by CCK-8 in the myenteric plexus of the duodenum (2 cm immediately following the pyloric sphincter aborally) and compare it with that of the dorsal vagal complex (DVC) - which occurs in 1-day old pups. Despite the existence of both of the CCK receptors, CCK1 and CCK2, in 4, 14, 21 and 35 day old rats, CCK-8 (0, 5, 10, 20 and 40 μg/kg, i.p.) increased Fos-like immunoreactivity (Fos-LI, a marker for neuronal activation) in the myenteric neurons of 21- and 35-day old rats but in the DVC of all age groups. As such, this belated activation of myenteric neurons by CCK-8 compared to the DVC may reflect a delayed role for these neurons in CCK-related functions.  相似文献

Micromanipulation and electrofusion were utilized for nuclear transfer in bovine embryos. Embryonic blastomeres from 5-day (estrus = day 0), 6-day, frozen-thawed 5-day, and first-generation nuclear transfer embryos (embryos were themselves a product of nuclear transfer with the original donor being a 5-day embryo) were transferred into bisected bovine oocytes by electrofusion. The percentage of donor cells fusing with the recipient oocytes was compared between different types of donor embryos. The percentage of embryos developing normally into morula or blastocysts following 6 days culture in the sheep oviduct was also recorded and compared between different donor embryo types. No significant differences were found between donor blastomeres for the percent successfully fused to oocytes: 5-day, 294 of 513 (57.3%); 6-day, 252 of 405 (62.2%); frozen-thawed 5-day, 111 of 144 (77.1%); nuclear transfer, 142 of 223 (63.7%); or the percent developing normally following nuclear transfer: 5-day, 92 of 444 (20.7%); 6-day, 84 of 357 (23.5%); frozen-thawed 5-day, 32 of 127 (25.2%); nuclear transfer, 31 of 199 (15.6%). These data suggest that a variety of donor embryos can successfully be utilized for bovine embryo cloning. Also, development of blastomeres from frozen-thawed 5-day donors and from donors that are themselves the product of nuclear transfer suggest that the production of multiple identical offspring is possible by frozen storage of seed stock and serial recloning.  相似文献

Summary Response to upward selection in 10 replicate lines was linear through 15 generations, had slowed considerably in many lines by generation 23 and continued in other lines through 30 generations. The lines tended to group into those which responsed slowly during early generations and continued to respond through later generations and those which gave rapid early response and very low response later coupled with marked reduction in fitness. Response to downward selection was similar for 3 replicate lines but only 50% as great as that for the high lines. Response declined more rapidly in the low lines due to reduced fitness, declining selection differentials and a marked drop in phenotypic variance.  相似文献

The objective of this research was to estimate the relative magnitude of effects included in contemporary groups (CG) and their interactions with adjusted and actual 120 d and 210 d weights in 72,731 male and female Nelore calves born from 1985 to 2005 in 40 herds from PMGRN (Genetic Improvement Program of Nelore). Ten models with different CG structures were compared. The analyses were done using the general linear models (GLM) procedure run in SAS software. All of the effects included in the CG for each model were significant (p < 0.001) for the four traits analyzed. Inclusion of semester or trimester of birth as part of a CG was more appropriate than its use as an independent effect in the model because it accounted for interactions with the other effects in the CG. Calf sex (CS) and dam age at calving (DAC) had similar effects across the models, which suggested independence from other effects in these models. The corresponding age deviation effect had a larger impact on actual weight at 120 d than any other effect in all of the models tested. The use of actual weights in models with no CS effect in CG provides an alternative that would allow better genetic connectedness among CGs and greater accuracy in genetic evaluations.  相似文献

Mycological analyses were made on specimens obtained from the skin, oral rinse, urine, and feces of three subjects participating in a 56 day Skylab simulation test. Samples were taken over a 175 day period divided into three phases consisting of a prechamber, intrachamber, and postchamber period. A total of 69 species of filamentous fungi and 22 species of yeast and yeast-like fungi were recovered and identified. There was a marked decrease in the isolation incidence of both the filamentous and yeast and yeast-like fungi during the intrachamber period. This, taken with the fact that 71% of the filamentous species and 55% of the yeast and yeast-like species were isolated no more than twice, reflects the transitory nature of the relationship between the human body and most fungi. However, there was a relative increase in the potential pathogens Candida albicans and Candida parapsilosis during the intrachamber period. Over 50% of all fungi recovered were isolated from the nasal and oral cavities. There was no evidence of intercrew transfer of a particular species during the intrachamber period nor was there any evidence of microbial shock during the postchamber period.  相似文献

Meteorotropic associations of heart rate (HR) and HR variability (HRV) were investigated in a clinically healthy 48-year-old man in Kiev. His electrocardiogram (ECG) was determined over 50 days by fitting him with an ambulatorily wearable device; various natural physical environmental variables were also monitored. The mean inter-beat interval, the standard deviation of these intervals, the spectral power in several frequency ranges, the power ratio of the approx. 10.5-s/approx. 3.6-s spectral components and other aspects of HRV were computed over consecutive 14.4-min intervals. Together with ordinary meteorological variables, geomagnetic disturbances (GMD) and fluctuations of atmospheric pressure (FAP) in the range 0.01–0.10 Hz (10–100 s) were measured. The assessable infradian spectra (with frequencies lower than 1 cycle/28 hours) of all HRV parameters showed two major components with periods of about 3.5 and 10 days. Two environmental variables, FAP and wind speed (with which FAP is closely related), revealed both of these rhythms and showed the greatest cross-spectral coherence (0.70–0.98) with corresponding oscillations of HRV. Less specific but statistically significant product-moment correlations with major HRV indices were also found; most of these were with FAP, but correlations with air temperature, humidity, wind speed and geomagnetic disturbances were also found. Long-term ECG recording, essential in the detection of infradian rhythms, proved to be sensitive to physical environmental variables, notably meteorological ones. FAP, usually neglected since its role has not been considered in previous biometeorological studies, or some factor closely related to FAP is probably involved in synchronizing or influencing the approximately 3.5-day HR and HRV rhythms in humans. Effects of 5-day hypoxia on cardiac adrenergic neurotransmission in rats   总被引:1,自引:0,他引:1  
Chronic hypoxiainduces an overall sympathetic hyperactivation associated with amyocardial -receptor desensitization. The mechanismsinvolved in this desensitization were evaluated in 32 male Wistar ratskept in a hypobaric pressure chamber(PO2 = 40 Torr, atmospheric pressure = 450 Torr) for 5 days. In hypoxic compared with normoxic conditions,plasma norepinephrine (NE) levels were higher (2.1 ± 0.7 vs. 0.6 ± 0.2 ng/ml) with no difference in the plasma epinephrine levels(2.2 ± 0.7 vs. 1.8 ± 0.3 ng/ml). In hypoxia neuronal NE uptakemeasured by [3H]NE wasdecreased by 32% in the right ventricle (RV) and by 35% in the leftventricle (LV), and[3H]mazindol in vitrobinding showed a decrease in uptake-1 carrier protein density by 38%in the RV and by 41% in the LV. In vitro binding assays with[3H]CGP-12177 indicate-adrenoceptor density reduced by 40% in the RV and by 32% in theLV, and this was due to reduced1-subtype fraction (competitionbinding experiments with practolol). Hypoxia reduced the production ofcAMP induced by isoproterenol (36% decrease in the RV and 41%decrease in the LV), 5'-guanylylimododiphosphate (40% decreasein the RV and 42% decrease in the LV), and forskolin (39% decrease inthe RV and 41% decrease in the LV) but did not alter the effect ofMnCl2 and NaF. Quantitation ofinhibitory G-protein -subunit by immunochemical analysis showed a46% increase in the cardiac-specific isoformGi2 inhypoxic hearts. The present data demonstrate that in rats 5-day hypoxialeads to changes in pre- and postsynaptic myocardial adrenergicfunction. The myocardial desensitization associated with both areduction in externalized 1-adrenoceptor and an increasein inhibitory G-protein subunit may be caused by increased synaptic NElevels due to impaired uptake-1 system.

The magnetic interactions between dissolved oxygen molecules and nitroxide radical spin probes lead to broadening of electron spin resonance (ESR) lines. Based on this property we described an ESR methodology to measure PO2 values in 4-day chick embryo and adult human blood. The total blood volume required for the measurement was only 9 microliter. Using this method PO2 for adult human mixed venous blood was found to be 36.7 mmHg. This is within the established clinical range of 25-40 mmHg for mixed venous blood PO2. The range of mean PO2 values for 4-day chick embryo blood was 27.3- 35.0 mmHg. This is the first time that PO2 values have been reported for individual chick embryos at such an early stage.  相似文献

A possibility of the cytogenetic sexing of 7-day microsurgically bisected bovine embryos by one of 2-4 parts with the aim of their selection by sex and peculiarities of the karyotype before their transplantation has been studied. Metaphase plates of chromosomes from halves and quarters of late morulae and early blastocysts for the cytogenetic analysis have been obtained.  相似文献
