The occurrence of photorespiration in soybean (Glycine max [L.] Merr.) leaf cells was demonstrated by the presence of an O2-dependent CO2 compensation concentration, a nonlinear time course for photosynthetic 14CO2 uptake at low CO2 and high O2 concentrations, and an O2 stimulation of glycine and serine synthesis which was reversed by high CO2 concentration. The compensation concentration was a linear function of O2 concentration and increased as temperature increased. At atmospheric CO2 concentration, 21% O2 inhibited photosynthesis at 25 C by 27%. Oxygen inhibition of photosynthesis was competitive with respect to CO2 and increased with increasing temperature. The Km (CO2) of photosynthesis was also temperature-dependent, increasing from 12 μm CO2 at 15 C to 38 μm at 35 C. In contrast, the Ki (O2) was similar at all temperatures. Oxygen inhibition of photosynthesis was independent of irradiance except at 10 mm bicarbonate and 100% O2, where inhibition decreased with increasing irradiance up to the point of light saturation of photosynthesis. Concomitant with increasing O2 inhibition of photosynthesis was an increased incorporation of carbon into glycine and serine, intermediates of the photorespiratory pathway, and a decreased incorporation into starch. The effects of CO2 and O2 concentration and temperature on soybean cell photosynthesis and photorespiration provide further evidence that these processes are regulated by the kinetic properties of ribulose-1,5-diphosphate carboxylase with respect to CO2 and O2.  相似文献

The effect of 21% O2 and 3% O2 on the CO2 exchange of detached wheat leaves was measured in a closed system with an infrared carbon dioxide analyzer. Temperature was varied between 2° and 43°, CO2 concentration between 0.000% and 0.050% and light intensity between 40 ft-c and 1000 ft-c. In most conditions, the apparent rate of photosynthesis was inhibited in 21% O2 compared to 3% O2. The degree of inhibition increased with increasing temperature and decreasing CO2 concentration. Light intensity did not alter the effect of O2 except at light intensities or CO2 concentrations near the compensation point. At high CO2 concentrations and low temperature, O2 inhibition of apparent photosynthesis was absent. At 3% O2, wheat resembled tropical grasses in possessing a high rate of photosynthesis, a temperature optimum for photosynthesis above 30°, and a CO2 compensation point of less than 0.0005% CO2. The effect of O2 on apparent photosynthesis could be ascribed to a combination of stimulation of CO2 production during photosynthesis, and inhibition of photosynthesis itself.  相似文献

We grew sunflower (Helianthus annuus L.) plants in nutrient solutions having nutritionally adequate but low or high Mg2+ concentrations and determined whether photosynthesis was effected as leaf water potentials (ψw) decreased. Leaf Mg contents were 3- to 4-fold higher in the plants grown in high Mg2+ concentrations (10 millimolar) than in those grown in low concentrations (0.25 millimolar). These contents were sufficient to support maximum growth, plant dry weight, and photosynthesis, and the plants appeared normal. As low ψw developed, photosynthesis was inhibited but moreso in high Mg leaves than in low Mg leaves. The effect was particularly apparent under conditions of light- and CO2-saturation, indicating that the chloroplast capacity to fix CO2 was altered. The differential inhibition observed in leaves of differing Mg contents was not observed in leaves having differing K contents, suggesting that the effect may have been specific for Mg. Because Mg2+ inhibits photophosphorylation and coupling factor activities at concentrations likely to occur as leaves dehydrate, Mg may play a role in the inhibition of chloroplast reactions at low ψw, especially in leaves such as sunflower that markedly decrease in water content as ψw decreases.  相似文献

The losses in chloroplast capacity to fix CO2 when photosynthesis is reduced at low leaf water potential (ψ1) have been proposed to result from photoinhibition. We investigated this possibility in soil-grown sunflower (Helianthus annuus L. cv IS894) using gas exchange techniques to measure directly the influence of light during dehydration on the in situ chloroplast capacity to fix CO2. The quantum yield for CO2 fixation as well as the rate of light- and CO2-saturated photosynthesis were strongly inhibited at low ψ1. The extent of inhibition was the same whether the leaves were exposed to high or to low light during dehydration. When intercellular partial pressures of CO2 were decreased to the compensation point, which was lower than the partial pressures resulting from stomatal closure, the inhibition of the quantum yield was also unaffected. Photoinhibition could be observed only after high light exposures were imposed under nonphysiological low CO2 and O2 where both photosynthesis and photorespiration were suppressed. The experiments are the first to test whether gas exchange at low ψ1 is affected by potentially photoinhibitory conditions and show that the loss in chloroplast capacity to fix CO2 was entirely the result of a direct effect of water availability on chloroplast function and not photoinhibition.  相似文献

Carbonyl sulfide (COS), a substrate for carbonic anhydrase, inhibited alkalization of the medium, O2 evolution, dissolved inorganic carbon accumulation, and photosynthetic CO2 fixation at pH 7 or higher by five species of unicellular green algae that had been air-adapted for forming a CO2-concentrating process. This COS inhibition can be attributed to inhibition of external HCO3 conversion to CO2 and OH by the carbonic anhydrase component of an active CO2 pump. At a low pH of 5 to 6, COS stimulated O2 evolution during photosynthesis by algae with low CO2 in the media without alkalization of the media. This is attributed to some COS hydrolysis by carbonic anhydrase to CO2. Although COS had less effect on HCO3 accumulation at pH 9 by a HCO3 pump in Scenedesmus, COS reduced O2 evolution probably by inhibiting internal carbonic anhydrases. Because COS is hydrolyzed to CO2 and H2S, its inhibition of the CO2 pump activity and photosynthesis is not accurate, when measured by O2 evolution, by NaH14CO3 accumulation, or by 14CO2 fixation.  相似文献

The nature of the inorganic carbon utilized during photosynthesis by Chlorella pyrenoidosa was investigated using three experimental techniques (open gas analysis system with “artificial leaf” or “aqueous” chambers and O2 electrode system) to measure carbon assimilation. Photosynthesis was studied as a function of pH and CO2 concentration. The CO2 concentration was inadequate to meet the requirements of photosynthesis only when HCO3 was added at high pH. Under all other conditions, the low and constant Km (CO2), in contrast to the highly variable Km (HCO3), suggested that CO2 was the major species utilized.  相似文献

Cyanide and azide inhibit photosynthesis and catalase activity of isolated, intact spinach (Spinacia oleracea) chloroplasts. When chloroplasts are illuminated in the presence of CN or N3, accumulation of H2O2 is observed, parallel to inhibition of photosynthesis. Hybrids have been made between species of Flaveria exhibiting varying levels of C4 photosynthesis. The degree of C4 photosynthesis expressed in four interspecific hybrids (Flaveria trinervia [C4] × F. linearis [C3-C4], F. brownii [C4-like] × F. linearis, and two three-species hybrids from F. trinervia × [F. brownii × F. linearis]) was estimated by inhibiting phosphoenolpyruvate carboxylase in vivo with 3,3-dichloro-2-dihydroxyphosphinoylmethyl-2-propenoate (DCDP). The inhibitor was fed to detached leaves at a concentration of 4 mm, and apparent photosynthesis was measured at atmospheric levels of CO2 and at 20 and 210 mL L−1 of O2. Photosynthesis at 210 mL L−1 of O2 was inhibited 32% by DCDP in F. linearis, by 60% in F. brownii, and by 87% in F. trinervia. Inhibition in the hybrids ranged from 38 to 52%. The inhibition of photosynthesis by 210 mL L−1 of O2 was increased when DCDP was used, except in the C4 species, F. trinervia, in which photosynthesis was insensitive to O2. Except for F. trinervia, control plants with less O2 sensitivity (more C4-like) exhibited a progressively greater change in O2 inhibition of photosynthesis when treated with DCDP. This increased O2 inhibition probably resulted from decreased CO2 concentrations in bundle sheath cells due to inhibition of phosphoenolpyruvate carboxylase. The inhibition of photosynthesis by DCDP is concluded to underestimate the degree of C4 photosynthesis in the interspecific hybrids because increased direct assimilation of atmospheric CO2 by ribulose bisphosphate carboxylase may compensate for inhibition of phosphoenolpyruvate carboxylase.  相似文献

Photosynthesis rates of detached Panicum miliaceum leaves were measured, by either CO2 assimilation or oxygen evolution, over a wide range of CO2 concentrations before and after supplying the phosphoenolpyruvate (PEP) carboxylase inhibitor, 3,3-dichloro-2-(dihydroxyphosphinoyl-methyl)-propenoate (DCDP). At a concentration of CO2 near ambient, net photosynthesis was completely inhibited by DCDP, but could be largely restored by elevating the CO2 concentration to about 0.8% (v/v) and above. Inhibition of isolated PEP carboxylase by DCDP was not competitive with respect to HCO3, indicating that the recovery was not due to reversal of enzyme inhibition. The kinetics of 14C-incorporation from 14CO2 into early labeled products indicated that photosynthesis in DCDP-treated P. miliaceum leaves at 1% (v/v) CO2 occurs predominantly by direct CO2 fixation by ribulose 1,5-bisphosphate carboxylase. From the photosynthesis rates of DCDP-treated leaves at elevated CO2 concentrations, permeability coefficients for CO2 flux into bundle sheath cells were determined for a range of C4 species. These values (6-21 micromoles per minute per milligram chlorophyll per millimolar, or 0.0016-0.0056 centimeter per second) were found to be about 100-fold lower than published values for mesophyll cells of C3 plants. These results support the concept that a CO2 permeability barrier exists to allow the development of high CO2 concentrations in bundle sheath cells during C4 photosynthesis.  相似文献

In well-watered plants of Welwitschia mirabilis, grown in the glass-house under high irradiance conditions, net CO2 assimilation was almost exclusively observed during the daytime. The plants exhibited a very low potential for Crassulacean acid metabolism, which usually resulted in reduced rates of net CO2 loss for several hours during the night. In leaves exposed to the diurnal changes in temperature and humidity typical of the natural habitats, CO2 assimilation rates in the light were markedly depressed under conditions resembling those occurring during midday, when leaf temperatures and the leaf-air vapor pressure differences were high (36°C and 50 millibars bar−1, respectively). Studies on the relationship between CO2 assimilation rate and intercellular CO2 partial pressure at various temperatures and humidities showed that this decrease in CO2 assimilation was largely due to stomatal closure. The increase in the limitation of photosynthesis by CO2 diffusion, which is associated with the strong decline in stomatal conductance in Welwitschia exposed to midday conditions, may significantly contribute to the higher 13C content of Welwitschia compared to the majority of C3 species.  相似文献

The active transport of CO2 in Synechococcus UTEX 625 was measured by mass spectrometry under conditions that preclude HCO3 transport. The substrate concentration required to give one half the maximum rate for whole cell CO2 transport was determined to be 0.4 ± 0.2 micromolar (mean ± standard deviation; n = 7) with a range between 0.2 and 0.66 micromolar. The maximum rates of CO2 transport ranged between 400 and 735 micromoles per milligram of chlorophyll per hour with an average rate of 522 for seven experiments. This rate of transport was about three times greater than the dissolved inorganic carbon saturated rate of photosynthetic O2 evolution observed under these conditions. The initial rate of chlorophyll a fluorescence quenching was highly correlated with the initial rate of CO2 transport (correlation coefficient = 0.98) and could be used as an indirect method to detect CO2 transport and calculate the substrate concentration required to give one half the maximum rate of transport. Little, if any, inhibition of CO2 transport was caused by HCO3 or by Na+-dependent HCO3 transport. However, 12CO2 readily interfered with 13CO2 transport. CO2 transport and Na+-dependent HCO3 transport are separate, independent processes and the high affinity CO2 transporter is not only responsible for the initial transport of CO2 into the cell but also for scavenging any CO2 that may leak from the cell during ongoing photosynthesis.  相似文献

The inhibition of photosynthesis by O2 in air-grown Chlorella pyrenoidosa was investigated using three experimental techniques (artificial leaf, aqueous method, and O2 electrode) to measure carbon assimilation. CO2 response curves were determined under different O2, pH, and temperature conditions. Regardless of the experimental technique and condition, O2 inhibition was not evident until a concentration of 50% was reached; Vmax values were reduced whereas Km (CO2) values were unaffected by the increasing O2 concentration. The response of photosynthesis to O2 was independent of CO2 and HCO3 concentrations as well as temperature. Relative rates of photosynthesis showed a 4 to 5% stimulation in 2% O2, a 12% inhibition in 50% O2, and a 24% inhibition in 100% O2. The inhibition by 50% O2 was still reversible after 20 minutes exposure whereas 100% O2 caused irreversible inhibition after only 4 minutes.  相似文献

Benthic cyanobacterial mats with the filamentous Microcoleus chthonoplastes as the dominant phototroph grow in oxic hypersaline environments such as Solar Lake, Sinai. The cyanobacteria are in situ exposed to chemical variations between 200 μmol of sulfide liter−1 at night and 1 atm pO2 during the day. During experimental H2S to O2 transitions the microbial community was shown to shift from anoxygenic photosynthesis, with H2S as the electron donor, to oxygenic photosynthesis. Microcoleus filaments could carry out both types of photosynthesis concurrently. Anoxygenic photosynthesis dominated at high sulfide levels, 500 μmol liter−1, while the oxygenic reaction became dominant when the sulfide level was reduced below 100 to 300 μmol liter−1 (25 to 75 μmol of H2S liter−1). An increasing inhibition of the oxygenic photosynthesis was observed upon transition to oxic conditions from increasing sulfide concentrations. Oxygen built up within the Microcoleus layer of the mat even under 5 mmol of sulfide liter−1 (500 μmol of H2S liter−1) in the overlying water. The implications of such a localized O2 production in a highly reducing environment are discussed in relation to the evolution of oxygenic photosynthesis during the Proterozoic era.  相似文献

The Na+ requirement for photosynthesis and its relationship to dissolved inorganic carbon (DIC) concentration and Li+ concentration was examined in air-grown cells of the cyanobacterium Synechococcus leopoliensis UTEX 625 at pH 8. Analysis of the rate of photosynthesis (O2 evolution) as a function of Na+ concentration, at fixed DIC concentration, revealed two distinct regions to the response curve, for which half-saturation values for Na+ (K½[Na+]) were calculated. The value of both the low and the high K½(Na+) was dependent upon extracellular DIC concentration. The low K½(Na+) decreased from 1000 micromolar at 5 micromolar DIC to 200 micromolar at 140 micromolar DIC whereas over the same DIC concentration range the high K½(Na+) decreased from 10 millimolar to 1 millimolar. The most significant increases in photosynthesis occurred in the 1 to 20 millimolar range. A fraction of total photosynthesis, however, was independent of added Na+ and this fraction increased with increased DIC concentration. A number of factors were identified as contributing to the complexity of interaction between Na+ and DIC concentration in the photosynthesis of Synechococcus. First, as revealed by transport studies and mass spectrometry, both CO2 and HCO3 transport contributed to the intracellular supply of DIC and hence to photosynthesis. Second, both the CO2 and HCO3 transport systems required Na+, directly or indirectly, for full activity. However, micromolar levels of Na+ were required for CO2 transport while millimolar levels were required for HCO3 transport. These levels corresponded to those found for the low and high K½(Na+) for photosynthesis. Third, the contribution of each transport system to intracellular DIC was dependent on extracellular DIC concentration, where the contribution from CO2 transport increased with increased DIC concentration relative to HCO3 transport. This change was reflected in a decrease in the Na+ concentration required for maximum photosynthesis, in accord with the lower Na+-requirement for CO2 transport. Lithium competitively inhibited Na+-stimulated photosynthesis by blocking the cells' ability to form an intracellular DIC pool through Na+-dependent HCO3 transport. Lithium had little effect on CO2 transport and only a small effect on the size of the pool it generated. Thus, CO2 transport did not require a functional HCO3 transport system for full activity. Based on these observations and the differential requirement for Na+ in the CO2 and HCO3 transport system, it was proposed that CO2 and HCO3 were transported across the membrane by different transport systems.  相似文献

The ability of 21 C3 and C4 monocot and dicot species to rapidly export newly fixed C in the light at both ambient and enriched CO2 levels was compared. Photosynthesis and concurrent export rates were estimated during isotopic equilibrium of the transport sugars using a steady-state 14CO2-labeling procedure. At ambient CO2 photosynthesis and export rates for C3 species were 5 to 15 and 1 to 10 μmol C m−2 s−1, respectively, and 20 to 30 and 15 to 22 μmol C m−2 s−1, respectively, for C4 species. A linear regression plot of export on photosynthesis rate of all species had a correlation coefficient of 0.87. When concurrent export was expressed as a percentage of photosynthesis, several C3 dicots that produced transport sugars other than Suc had high efflux rates relative to photosynthesis, comparable to those of C4 species. At high CO2 photosynthetic and export rates were only slightly altered in C4 species, and photosynthesis increased but export rates did not in all C3 species. The C3 species that had high efflux rates relative to photosynthesis at ambient CO2 exported at rates comparable to those of C4 species on both an absolute basis and as a percentage of photosynthesis. At ambient CO2 there were strong linear relationships between photosynthesis, sugar synthesis, and concurrent export. However, at high CO2 the relationships between photosynthesis and export rate and between sugar synthesis and export rate were not as strong because sugars and starch were accumulated.  相似文献

The effect of purified host-specific toxin from Bipolaris (Helminthosporium) maydis, race T, on dark or light-dependent CO2 fixation was studied with thin (1 × 8 mm) corn (Zea mays L.) leaf slices supplied H14CO3. At 5 to 30 nanograms per milliliter (5 nanomolar), toxin significantly inhibited (20 to 40%) dark CO2 fixation in susceptible (T) corn slices after either dark or light preincubations of 10-20 minutes. The same concentrations were effective to the same degree on photosynthesis, but the effect differed in that significant inhibition occurred after 25 minutes and only with light preincubation. Light preincubation without toxin did not shorten the time required for inhibition of photosynthesis after addition of toxin. Once photosynthetic inhibition was entrained, it was not reversed by subsequent periods of darkness. The results suggest the possibility that race T toxin affects two separate metabolic sites, and the data are discussed in view of currently held concepts of toxin action in susceptible tissue.  相似文献

The metabolism of 14C-glycine (a substrate for photorespiration) was studied in the light and in darkness under natural CO2 concentration (0.03%) in the leaves of ephemeroides Scilla sibirica Haw. and Ficaria verna Huds. at different developmental stages. Using one and the same sample, potential photosynthesis (at 1% CO2), true photosynthesis (at 0.03% CO2), and leaf respiratory capacity were measured by the radiometric and manometric methods, respectively. All measurements were performed at 15°C, an average temperature during ephemer growth. It was found that, in the white zone of the Scilla leaf, the rate of CO2 evolution resulting from metabolization of exogenous 14C-glycine was similar in the light and in darkness. In the green zone of the Scilla leaf and in the green leaf of Ficaria, both 14C-glycine absorption and 14CO2 evolution were lower in the light as compared with darkness, which is explained by CO2 reassimilation. In all treatments of both plant species, a specific inhibitor of glycine decarboxylase complex (GDC), aminoacetonitrile (5 mM) suppressed CO2 evolution by 20–40%. It was concluded that in ephemeroides mitochondrial GDC, responsible for CO2 evolution in photorespiration, is formed at the earliest stage of leaf development. This indicates that photorespiration can occur simultaneously with the development of the leaf photosynthetic activity. On the basis of the assumption that carbon losses in the form of CO2 evolved during photorespiration comprise 25% of true photosynthesis, it was calculated that, in ephemer leaves, the highest rates of photorespiration and photosynthesis were attained during flowering when the leaf area was the largest and the rate of dark respiration was reduced by 1.5–2.0 times. The highest rates of dark respiration were observed in the beginning of growth. In senescing leaves by the end of the plant vegetation, potential photosynthesis and true photosynthesis were reduced, whereas dark respiration remained essentially unchanged. It is concluded that the high rates of potential and true photosynthesis are characteristic of ephemeroides when they complete their short developmental program in early spring (at 15°C); theoretically, photorespiration also occurs at a high rate during this period, when this process provides for a defense against the threat of photoinhibition at low temperature and high insolation.  相似文献

We examined how the freely floating macrophyte, Stratiotes aloides L., sampled from a CO2-supersaturated pond, changes leaf morphology, photosynthesis and inorganic carbon acquisition during its different submerged and emerged life stages in order to evaluate whether S. aloides requires consistently supersaturated CO2 conditions to grow and complete its life cycle. Submerged rosettes formed from over-wintering turions had typical traits of submerged plants with high specific leaf area and low chlorophyll a concentrations. Emergent leaf parts of mature, floating specimens had typical terrestrial traits with stomata, low specific leaf area and high chlorophyll a content, while offsets formed vegetatively and basal, submerged parts of mature plants showed traits in between. All submerged leaf types exhibited some ability to use HCO3 but only rosettes formed from turions had efficient HCO3 use. Rosettes also had the highest CO2 affinity and maximum CO2-saturated photosynthesis in water. Half-saturation constants for CO2 (21–74 μM CO2) were for all submerged leaf parts 5–140 times lower than the concentrations of free CO2 in the pond (350–2800 μM CO2). Emergent leaves were less efficient in water but had significantly higher photosynthesis than submerged, mature leaf parts in air, and rates of photosynthesis of emergent leaves in air were three to five times higher than rates of CO2-saturated photosynthesis of the three submerged leaf types in water. Underwater photosynthetic rates estimated at CO2 concentrations corresponding to air equilibrium were not sufficiently high to support any noticeable growth except for rosettes, in which bicarbonate utilization combined with high CO2 affinity resulted in photosynthetic rates corresponding to almost 34% of maximum rates at high free CO2. We conclude that S. aloides requires consistently high CO2-supersaturation to support high growth and to complete its life cycle, and we infer that this requirement explains why S. aloides mainly grows in ponds, ditches and reed zones that are characterized by strong CO2-supersaturation.  相似文献

Incubating isolated soybean leaf mesophyll cells with glyoxylate increased the rates of CO2 fixation by as much as 150%. In order to cause this stimulation, the glyoxylate must be presented to the cells before the NaHCO3. Significant stimulation was observed 15 seconds after beginning the glyoxylate treatment. The glyoxylate-dependent stimulation was increased by high O2 concentrations and decreased by high CO2 concentrations. Glyoxylate treatment resulted in a 71% inhibition in the rate of CO2 incorporation into glycolate and glycine. Glyoxylate may be stimulating net photosynthesis solely by decreasing photorespiration or it may be increasing the amount of CO2 fixed by both decreasing photorespiration and increasing gross photosynthesis. Ribulose bisphosphate carboxylase, when preactivated and assayed in situ, was unaffected by the glyoxylate treatment.  相似文献

Background and Aims Plants growing under elevated atmospheric CO2 concentrations often have reduced stomatal conductance and subsequently increased leaf temperature. This study therefore tested the hypothesis that under long-term elevated CO2 the temperature optima of photosynthetic processes will shift towards higher temperatures and the thermostability of the photosynthetic apparatus will increase.Methods The hypothesis was tested for saplings of broadleaved Fagus sylvatica and coniferous Picea abies exposed for 4–5 years to either ambient (AC; 385 µmol mol−1) or elevated (EC; 700 µmol mol−1) CO2 concentrations. Temperature response curves of photosynthetic processes were determined by gas-exchange and chlorophyll fluorescence techniques.Key Results Initial assumptions of reduced light-saturated stomatal conductance and increased leaf temperatures for EC plants were confirmed. Temperature response curves revealed stimulation of light-saturated rates of CO2 assimilation (Amax) and a decline in photorespiration (RL) as a result of EC within a wide temperature range. However, these effects were negligible or reduced at low and high temperatures. Higher temperature optima (Topt) of Amax, Rubisco carboxylation rates (VCmax) and RL were found for EC saplings compared with AC saplings. However, the shifts in Topt of Amax were instantaneous, and disappeared when measured at identical CO2 concentrations. Higher values of Topt at elevated CO2 were attributed particularly to reduced photorespiration and prevailing limitation of photosynthesis by ribulose-1,5-bisphosphate (RuBP) regeneration. Temperature response curves of fluorescence parameters suggested a negligible effect of EC on enhancement of thermostability of photosystem II photochemistry.Conclusions Elevated CO2 instantaneously increases temperature optima of Amax due to reduced photorespiration and limitation of photosynthesis by RuBP regeneration. However, this increase disappears when plants are exposed to identical CO2 concentrations. In addition, increased heat-stress tolerance of primary photochemistry in plants grown at elevated CO2 is unlikely. The hypothesis that long-term cultivation at elevated CO2 leads to acclimation of photosynthesis to higher temperatures is therefore rejected. Nevertheless, incorporating acclimation mechanisms into models simulating carbon flux between the atmosphere and vegetation is necessary.  相似文献
