Parameters of the antioxidant defense systems of Brycon amazonicus (matrinx?--a neotropical fish) exposed to phenol for 96 h plus the recovery over 1 and 2 weeks were studied in erythrocytes and liver. Hematocrit increase was observed during phenol exposure and recovery for 1 week. Total superoxide dismutases (SOD), glutathione peroxidase (GPx) and reduced glutathione (GSH) did not change during phenol exposure. Erythrocyte glucose-6-phosphate dehydrogenase (G6PDH) increased during that period while catalase (CAT) activity decreased during phenol exposure and recovery for 2 weeks. In the liver, SOD and CAT did not change, whereas GPx increased in the first week of recovery and decreased after 2 weeks. A late response was observed for G6PDH activity which increased only at the second week. Ascorbate concentration in the brain decreased during phenol exposure and increased over recovery. From our results it appears that the oxidative stress was limited in matrinx? exposed to phenol, but seemed to occur during the recovery period. 相似文献

Phenol and its derivatives are xenobiotics present in many industrial wastewaters and in non-specific pesticides. It is a lipophilic compound and, therefore, accumulates along the trophic chain. Phenol is often found in marine and fresh water environments. The aim of this work was to detect metabolic changes induced by phenol in Brycon cephalus juveniles. Several enzymes activities and metabolites were quantified in the liver, white muscle and plasma. Among the enzymes assayed are alanine and aspartate amino transferases (ALAT and ASAT), lactate dehydrogenase (LDH) and malate dehydrogenase (MDH). Glucose, glycogen, lactate, ammonia and pyruvate were also quantified in tissues and plasma (glycogen in tissues only). The liver was the most responsive organ. The activities of the transaminases increased in muscle and liver, followed by an increase in hepatic ammonia. Correlation between ammonia and transaminases points towards phenol-induced consumption of protein. Hepatic glycogen and glucose contents were lower followed exposure to phenol. The same was observed for muscle glucose, suggesting considerable use of carbohydrate stores. The activity of hepatic lactate dehydrogenase increased with negative correlation with muscle lactate. This suggests that hepatic gluconeogenesis supplies tissues like muscle and brain with glucose. These results indicate that phenol intoxication demands metabolic energy and leads to significant changes of the metabolic profile of the fish, inducing to a certain extent a shift from carbohydrate catabolism to protein catabolism and the activation of gluconeogenesis. 相似文献

This study describes anatomical, histological and histochemical features of the digestive tract mucosal layer of the matrinxã Brycon amazonicus, an omnivorous freshwater fish endemic from the Amazon basin. This species presents short thick oesophagus with longitudinal folds, that allow the passage of large food items. The mucosa is lined with a stratified secretory epithelium rich in goblet cells that secrete neutral and acid mucins. The two mucin types provide different viscosity in anterior and posterior oesophagus related to the protective and lubricant functions, respectively. The stomach is a highly distensible Y-shaped saccular organ. Here, it is proposed that this anatomical shape plays an essential role in food storage when food availability is abundant. The stomach mucosa is composed of epithelial cells with intense neutral mucin secretion to protects against gastric juice. The intestine is slightly coiled and presents internally a complex pattern of transversal folds that increases the absorption surface and the retention time of food. Goblet cells in the intestine secrete acid and neutral mucins that lubricate the epithelium and aid in the digestive processes. In the rectum, an increase in goblet cells population occurs that may be related to better lubrication. 相似文献

Juvenile matrinx? (Brycon amazonicus) were fed with different dietary vitamin C (ascorbic acid or AA) concentrations for 2 months in order to evaluate its effect on the physiological condition of this species. Five treatments were tested: Reference diet-RD (350 mg AA kg(-1) and 45% crude protein commercial feed); C400, C500, C600 and C800 with supplementation of 400, 500, 600 and 800 mg AA kg(-1). Fish fed 800 mg AA kg(-1) supplemented diets presented greater weight gain and survival. Hematocrit, hemoglobin concentrations and erythrocytes showed significant increase (p<0.05) in C600 and C800 relatively to the others. Total leukocyte values were significantly higher in fish fed vitamin C supplemented diets relatively to those fed RD and leukocytes count of C600 and C800 were the highest among the treatments. Monocytes were the only cells of fish defense mechanism that presented variation, and C600 and C800 presented the lowest values (P<0.05). Glucose level was significantly different relatively to RD only in C400 and the plasmatic total protein values for fish fed vitamin C supplemented diets were higher than for fish fed the RD. Fish plasmatic ions (Na(+) and Cl(-)) were significantly lower for C500, C600 and C800 relatively to C400 and RD. The physiological profile and the growth performance of matrinx? suggest that diet supplemented with 800 mg AA kg(-1) can improve its performance and increase the survival rate. 相似文献

Environmental increase in nitrite impairs the function of several aquatic species, including fishes. Nitrite reacts with hemoglobin yielding the non-functional methemoglobin (metHb), and many physiological disturbances can arise. The physiological mechanisms to cope with nitrite are still unclear in fish. Hematological parameters, the role of NADH-methemoglobin reductase system and the electrolytic balance were studied in the freshwater teleost Brycon cephalus (matrinx?) exposed to 0.2, 0.4 and 0.6 mg/L of nitrite N-NO(2) for 24 and 96 h. Hematocrit, total hemoglobin and the red blood cell (RBC) number decreased. Methemoglobin content increased from 1% to 69% for 24 h of exposure and drastically from 5-6% to 90% for 96 h. The activity of NADH-methemoglobin reductase system displayed a tendency of increase in response to nitrite concentration or time of exposure. In the plasma, nitrite was accumulated to values 30-fold higher than the environmental concentration. The plasma K(+) concentration increased only in fish exposed to NO(2) for 24 h. No changes in plasma protein and Na(+) were observed during nitrite exposure but Cl-presented a punctual increase at 0.2 mg/L N-NO(2)-96 h. The hematological data suggest that nitrite caused functional and hemolytic anemia. Furthermore, the electrolytic balance was relatively undisturbed, and the nitrite clearance in matrinx? is likely depending on other factors than NADH-methemoglobin reductase system. 相似文献

Exposure to agrichemicals can have deleterious effects on fish, such as disruption of the hypothalamus-pituitary-inter-renal axis (HPI) that could impair the ability of fish to respond to stressors. In this study, fingerlings of the teleost jundiá (Rhamdia quelen) were used to investigate the effects of the commonly used agrichemicals on the fish response to stress. Five common agrichemicals were tested: the fungicide - tebuconazole, the insecticide - methyl-parathion, and the herbicides - atrazine, atrazine+simazine, and glyphosate. Control fishes were not exposed to agrichemicals and standard stressors. In treatments 2-4, the fishes were exposed to sub-lethal concentrations (16.6%, 33.3%, and 50% of the LC(50)) of each agrichemical for 96 h, and at the end of this period, were subjected to an acute stress-handling stimulus by chasing them with a pen net. In treatments 5-7 (16.6%, 33.3%, and 50% of the LC(50)), the fishes were exposed to the same concentrations of the agrichemicals without stress stimulus. Treatment 8 consisted of jundiás not exposed to agrichemicals, but was subjected to an acute stress-handling stimulus. Jundiás exposed to methyl-parathion, atrazine+simazine, and glyphosate presented a decreased capacity in exhibiting an adequate response to cope with stress and in maintaining the homeostasis, with cortisol level lower than that in the control fish (P<0.01). In conclusion, the results of this study clearly demonstrate that the acute exposure to sub-lethal concentrations of methyl-parathion, atrazine+simazine, and glyphosate exert a deleterious effect on the cortisol response to an additional acute stressor in the jundiá fingerlings. 相似文献

This study describes at ultrastructural level the germ cells in the testis of matrinx? (Brycon cephalus) raised in captivity. The specimens 'matrinx?' were maintained in four breeding tanks of 200 m(2), at the Aquaculture Research Center at Vale do Ribeira-CEPAR, from Fishery Institute, in Pariquera-A?u City, S?o Paulo, Brazil. The samples were collected from March 1994 to February 1996. Methyl parathion (MP), an organophosphate widely applied in agriculture and aquaculture, induces oxidative stress due to free radical generation and changes in the antioxidant defense system. The antioxidant roles of selenium (Se) were evaluated in Brycon cephalus exposed to 2 mg L(-1) of Folisuper 600 BR (MP commercial formulation - MPc, 600 g L(-1)) for 96 h. Catalase (CAT), glutathione peroxidase (GPx), superoxide dismutase (SOD), glutathione S-transferase (GST), reduced glutathione (GSH) and lipid peroxidation (LPO) levels in the gills, white muscle and liver were evaluated in fish fed on diets containing 0 or 1.5 mg Se kg(-1) for 8 weeks. In fish treated with a Se-free diet, the MPc exposure increased SOD and CAT activities in all tissues. However, the GPx activity decreased in white muscle and gills whereas no alterations were observed in the liver. MPc also increased GST activity in all tissues with a concurrent decrease in GSH levels. LPO values increased in white muscle and gills and did not change in liver after MPc exposure. A Se-supplemented diet reversed these findings, preventing increases in LPO levels and concurrent decreases in GPx activity in gills and white muscle. Similarly, GSH levels were maintained in all tissue after MPc exposure. These results suggest that dietary Se supplementation protects cells against MPc-induced oxidative stress. 相似文献

Phospholipids and phospholipases play important roles in several cellular processes and responses to adverse growth conditions. However, the mechanism of action of phosphatidylinositol-specific phospholipase C (PI-PLC), and its resulting minor lipid function in response to pollutant-induced stress, remains to be elucidated. In this work, we studied the effects of phenol treatment on the PLC pathway, using two lines of hairy root cultures (HRs) from Nicotiana tabacum as plant model systems: wild-type (WT) and double-transgenic (DT) HRs. We quantified several product formations such as PIP, PIP2, DGPP, and PA, which are mainly synthesized by specific lipid kinases belonging to the PLC pathway. In both HRs, phenol treatment significantly increased the formation of these compounds in a differential manner. In WT HRs, PA formation was twofold higher than in control. PIP2 and PIP levels were about onefold higher than those of the controls while DGPP levels increased by 50%. In DT HRs, PIP levels were onefold higher than those of the controls while PIP2, DGPP, and PA levels increased by 120%. Phenol treatment also upregulated the PLC4 gene expression mainly in the first hours of exposure in both HRs, while the DGK1 gene expression was only upregulated in WT HRs after 24 h of treatment. These results show an active participation of the PLC pathway under phenol treatment suggesting that this signal pathway could be important in plant cell responses to phenol-induced stress. 相似文献

We analyzed the diet and seasonal variation in diet composition of Brycon orthotaenia, an endangered and endemic species of the São Francisco River Basin, in the middle stretch of the São Francisco River. Brycon orthotaenia had an herbivorous diet, with occasional consumption of animals, mainly in the dry season. Variation among dry and wet seasons in diet composition may be related to the species’ reproduction and energy allocation, since some items ( e.g. insects, fishes) may support gonadal development. 相似文献

Plant vacuoles play several roles in controlling development, pathogen defence, and stress response. γVPE is a vacuolarlocalised cysteine protease with a caspase-1 like activity involved in the activation and maturation of downstream vacuolar hydrolytic enzymes that trigger hypersensitive cell death and tissue senescence. This work provides evidence that γVPE is strongly expressed in Arabidopsis guard cells and is involved in water stress response. The γvpe knock-out mutants showed reduced stomatal opening and an increased resistance to desiccation suggesting a new role of γVPE in control of stomatal movements. 相似文献

The study investigated the changes in individual molecular species in PE and the effects of a variety of dietary fats with varying proportions of saturated and unsaturated fatty acids on membrane composition, eicosanoid production and cytokine production in thioglycollate-elicited rat macrophages.The data obtained indicates that the greatest degree of modulation by dietary fats on cytokine production was observed after 8 weeks feeding and at this time, the total diacyl species containing linoleic acid (18:2 n-6) and arachidonic acid (20:4 n-6) at the sn-2 position related in a curvilinear fashion to total 18:2 n-6 intake and that IL1 and IL6 production related in a curvilinear fashion to the total diacyl species with 20:4 and 18:2 at the sn-2 position.After 4 weeks of feeding, fish and olive oils enhanced production of IL6 and LTB 4, however, while IL1 production, after 8 weeks of dietary treatment, was greatest from macrophages of animals fed corn and olive oils, PGE 2 production was greatest in the former group and LTB 4 production in the latter. Thus an eicosanoid effect may explain the modulatory influence of olive oil and IL1 production but, cannot explain the effect of corn oil on production of the cytokine. The data from the present study provides some insight into how dietary fats could provide therapy for conditions in which inflammatory cytokines are implicated. 相似文献

The stress sensitivity, determined in copper exposureexperiments and in survival in air tests, and thegenetic structure, measured by means of isoenzymeelectrophoresis, were assessed in populations of theBaltic clam Macoma balthica (L.) from itssouthern to its northern distribution limit, in orderto test the hypotheses that near the distributionlimit the clams would be more stress sensitive andwould have a lower genetic variability. Thepopulations in west and north Europe show a stronggenetic resemblance. The populations in the sub-ArcticWhite Sea are genetically slightly different, and showa low stress sensitivity. The populations in theArctic Pechora Sea are genetically very distant fromthe other populations, and show the lowest stresssensitivity. Near the southern distribution limit, inagreement with the hypotheses, genetic variability islow and stress sensitivity high. On the other hand, incontrast to expectation, near the northerndistribution limit, in the populations of the PechoraSea, the genetic variability was higher, thus notreduced, and the stress sensitivity was low comparedto all other populations. ABSTRACTBackground: Stomatal response functions of dominant plant species can provide insights into the behaviour of ecosystems under environmental stress, and provide tools for modelling their response to climate change. However, they remain little studied in tropical Alpine ecosystems. Aims: Our objective was to formulate and compare stomatal response functions for two dominant páramo species with different adaptive strategies to drought, the stress-tolerant shrub Hypericum laricifolium, and the stress avoiding giant rosette Espeletia schultzii and thus enable making projections as to their future fitness in a changing climate. Methods: A reanalysis of data found in the literature and new ecophysiological and micrometeorological measurements were used to fit and test new stomatal response functions to environmental variables for these two species. Results: The response functions of vapour pressure differences between leaf and air showed an exponential decrease for both species, while for photosynthetically active radiation (PAR), peak-form response functions provided the best fit. The response function for leaf water potential was linear for the drought-tolerant shrub and decreased exponentially for the stress avoiding giant rosette. Several thresholds prior to stomatal closure were also included in the functions. Conclusions: Although stress-avoiding and stress-tolerant strategies are both successful in the Andean páramo, the response functions suggest that the tolerant shrub could be more resistant to more intense drought. 相似文献

An integrated population monitoring study was initiated to investigate whether occupational exposure to current low levels of butadiene is mutagenic to workers. Ten exposed workers (mean production area concentration of 3.5 ppm) and 10 matched plant controls(mean exposure to 0.03 ppm) were selected and blood samples were collected for our study. The standard cytogenetic assay was used to determine chromosome aberration frequencies. In addition, a challenge assay was used to determine response to γ-rays as an indication of DNA repair deficiencies. In the latter assay, cells were exposed to γ-rays at the G1 phase of the cell cycle in vitro and the frequencies of chromosome aberrations in the first post-irradiation metaphase cells were quantitated. Based on results of the cytogenetic assay, the exposed group had a higher frequency of cells with chromosome aberrations and higher chromatid breaks per 100 cells compared with the control. However, the difference was not significant ( p > 0.1). With the challenge assay, the exposed group had a higher frequency of aberrant cells ( p < 0.04), chromatid breaks ( p < 0.05), deletions ( p < 0.07), and dicentrics ( p < 0.02) than the controls. In addition, the dicentric frequencies from workers were significantly correlated with the presence of a butadiene metabolite [1,2-dihydroxy-4-( N-acetyl-cysteinyl- S)butane] in urine with a correlation of coefficient of 0.6 ( p < 0.01). Two outliers were identified and our interpretation of their responses will be discussed. This study indicates that the workers had exposure-induced mutagenic effects. Together with the observation of gene mutation in a subset of the present population, this study indicates that the current occupational exposure to butadiene may not be safe to workers. 相似文献

Lactobacillus salivarius, a probiotic bacterium, encounters acidic conditions in its passage through the gastrointestinal tract of human and animal hosts. We studied the effect of a rapid downshift in extracellular pH from 6.5 to 4 on cell growth. The maximum growth rate was higher in low pH medium with glutathione supplementation than without. Cells developed a GSH-mediated acid-tolerance response and, when grown with 0.5 mM GSH, reached a higher final density than with other conditions. These findings suggest that the increased growth rate is caused by uptake of GSH which acts as a nutrient source as well as having protective functions, allowing for continued growth. 相似文献

Citrinin is a type of mycotoxin that negatively affects Monascus products. Application of a low-frequency magnetic field (LF-MF) decreased citrinin production by M. purpureus in liquid-state fermentation during shake flask culture. Under 30 °C incubation, six different magnetic field induction intensity (MF-II), four different exposure times and five exposure time periods were tested to discover optimal treatment conditions. The cultures were exposure to a MF-II of 1.6 mT from 0 to 2 d of incubation time. With LF-MF treatment, peak citrinin production decreased by 46.7% while yellow, red, orange pigments and monacolin K production increased by 31.3%, 40.3%, 41.7% and 29.3%, respectively, compared with control groups at 12 d of incubation. Moreover, the relative expression levels of the citrinin biosynthesis genes pksCT and ctnA were 0.46 and 0.43 times lower, respectively, than the control values relative to the GAPDH reference gene. This study provides evidence that LF-MF is a preferable way to alter M. purpureus metabolism to reduce citrinin production and to increase pigments and monacolin K production without affecting cell growth. Therefore, LF-MF may be used as a tool to process Monascus products to obtain important functional food additive while reducing the adverse effects of citrinin. 相似文献
